Abstract:
In this study the utilization of neuro-fuzzy techniques is investigated for the realisation of intelligent control. Neuro-fuzzy systems combine the learning capabilities of neural networks with the rule based system description offered by fuzzy logic. Techniques are evaluated by means of a simple two-dimensional simulation of a three-segment robotic manipulator. The inverse kinematics and path planning required in such a system provides all the complexity needed for testing and evaluation. In complex systems, obtaining sufficient training examples also prove to be a problem. To address this problem an automated process of 'action evolution' was implemented, through which a genetic algorithm is used to collect examples as training data. A generic modular controller architecture is developed in order to simplify the comparison of different neural and neuro-fuzzy controllers. This architecture unifies numerous controller architectures into a single controller, capable of representing and combining classical, adaptive, intelligent and reinforcement learning controllers and it exposes the presence of various cognitive attributes. Neural and neuro-fuzzy systems are implemented and evaluated for local trajectory tracking and for global path planning. A serious problem of contradicting solutions encountered in the examples produced by the genetic algorithm, is solved through reinforcement learning. A modified fuzzy clustering algorithm is used to estimate the system's state values and control commands are derived by optimising this value. Modifications included negative reinforcement of prohibited states and concepts borrowed from ant algorithms for establishing solution paths. This study highlights the effect of ill-posed problems on the training of intelligent controllers. It shows how the problem can be simplified by basing the control policy on a value function and it implements neuro-fuzzy techniques to rapidly construct such a function. Proposals are made on how memory based search algorithms can be used to improve training data integrity and how evolving self-organising maps might prevent erroneous policy interpolation. This study contributes valuable conclusions on the implementation of intelligent controllers for the control of complex non-linear systems.<br>Thesis (M.Ing. Abstract:
The sole electricity supplier in South Africa, Eskom, currently has an electricity supply crisis. The supplier requires additional available electricity urgently, particularly during the evening peak period between 18:00 and 20:00. This electricity shortage is due to a steady increase in the demand for electricity, which exceeded the increase in supply capacity, and the inefficient utilisation of electricity. In order to address this problem, Eskom introduced a Demand Side Management (DSM) programme. The aim of DSM projects is to reduce the load of consumers without negatively affecting consumers. Demand Side Management is beneficial for both Eskom and the client. The client benefits from a lower electricity bill and new equipment, while Eskom benefits from a reduced power demand. Various DSM strategies are implemented in different sectors, such as mining and residential. These projects are managed by Energy Service Companies (ESCo). The ESCo is responsible for the identifying, implementing, and maintaining the DSM project. Any identified DSM project is presented to Eskom, which agrees to fund the project depending on the proposed power saving. The mining industry, which has been selected as a candidate for DSM projects, as it is a major consumer of energy with numerous DSM opportunities, is examined in this dissertation. Because compressors are major consumers of electricity on the mines, significant DSM opportunities exist on compressed air systems. The purpose of this research project is to investigate and implement sustainable DSM projects on the compressed air systems of the mining industry. The focus is on automatically controlling the underground demand for compressed air. Reducing the demand for compressed air will result in lower power consumption by the compressors.<br>Thesis (M.Ing. Abstract:
One hundred wheat samples, with moisture ranging from 14.42 %to 7.66 %, were used to test the moisture loss from grinding, the accuracy of moisture content from two different electric moisture meters, and the accuracy and precision of Near Infrared Reflectance Spectroscopy whole wheat moisture determination. Fifty samples were ground through three grinders, Wiley, Cemotec , and UDY Cyclone grinder. Moisture content was determined from these ground samples, and compared with whole wheat moisture. The Cemotec grinder caused the least amount of moisture loss, followed by Wiley and then UDY Cyclone. Fifty whole wheat samples were used to calibrate the Tecator GP6010 moisture meter. Another 50 whole wheat samples with known moisture values, were used to test the accuracy of the Tecator GP6010 moisture meter. Moisture and compared to moisture readings from a Motomco meter. Tecator moisture value had a better correlation coefficient to whole wheat moisture than Motomco moisture values (0.935 and 0.917, respectively). Three sample sets from different grinding preparation methods were scanned by NIRS . Eight moisture values were entered into the computer, and separate multiple regression equations developed. Sixteen wheat samples with known moisture content were used to test the equations. For .whole wheat moisture equation, SEA and r 2 were 0.159 and 0.991, 0.223 and 0.972, 0 .223 and 0 .973, 0.322 and 0.888, 0.218 and 0.968, 0.248 and 0.969 , 0.208 and 0.978 , 0.249 and 0.970 for whole wheat, Cemotec, Wiley, UDY, C3 , C5, C6, and Motomco moisture, respectively. For Cemotec moisture equation, SEA and r2 were 0.226 and 0.944, 0.208 and 0.927, 2.267 and 0.071, 0.208 and 0.853, 0.272 and 0.876, 2.296 and 0.088, 0.291 and 0.896, 0.248 and 0.927, for whole wheat, Cemotec, Wiley, Udy, C3, CS , C6, and Motomco moisture, respectively. For UDY moisture equation, SEA and r2 were 0 .402 and 0.909, 0.364 and 0.900, 0.353 and 0.907, 0.119 and 0 .973, 0.407 and 0.858, 0.484 and 0.856, 0.514 and 0.835, 0.403 and 0.897, for whole wheat, Cemotec, Wiley, UDY, Tecator C3, CS, C6, and Motomco moisture, respectively. Many desirable features make the whole wheat method a good direction for Near Infrared Reflectance Spectroscopy.

The aims of the study were: 1. to determine the different types of goal orientation and the interactive relationships between goal orientation, subject choice and academic achievement, from a literature review; 2. to empirically determine the relationship between goal orientation, subject choice and the achievement of standard 7 students; 3. to empirically determine the relationship between the goal orientation of standard 7 students and the school variables, and 4. to empirically determine the relationship between goal orientation and the age of standard 7 students. From the literature view it was evident that there are different orientations which influence subject choice and academic achievement. Students who are mastery/learning-oriented want to develop their competence, choose challenging tasks and perform better. Ego/performanceoriented students are interested in demonstrating to others that they are capable, choose easy tasks and they do not perform as well. A strong relationship was also evident between goal orientation and the attributional style of students. Mastery-oriented students attribute their success to internal, stable variables like ability or effort (an unstable but controllable cause), and experience high levels of self-efficacy and pride. Ego-oriented students attribute both failure and success to stable but uncontrol1able causes (such as ability or teaching methods), and experience shame and decreased self-efficacy in the event of failure. A relationship was also evident between goal orientation and the school variables (such as the teacher's selfefficacy, experience and educational/training level). The empirical study led to the following conclusions: • there is no relationship between the goal orientation and subject choice of the students of this study, in both mathematics and history; • there is a relationShip between the goal orientation and academic achievement of the students who were used in this study in mathematics, but not in history; • there is also a relationship between the goal orientation of the students of this study and the school variable, yet • there is no relationship between goal orientation and the age of the studens who were used in this study.<br>Skripsie (MEd (Psigo-opvoedkunde))--PU vir CHO, 1995

These interpretation are found in the works Literal Commentary on Genesis, concerning Genesis Against the Manicheans, Literal Commentary on Genesis, Unfinished, Books Xi-XIII of the Confessions, as well a brief exposition in Books XI-XIV of the City of God. Exposition of these Augustinian Commentaries seeks to demonstrate various interpretations made by author in one group of texts revealing a hermeneutics centered in the interpreter and not in established interpretive rules. In sequence there is described succinctly the evolution of textual hermeneutics during the modern period up to the reflections conducted by Martin Heidegger in the first half of the twenties. Based in Heidegger s existential commentaries on the Pauline Epistles (Galatians and I-II Thessalonians) and on the tenth Book of the Confessions, there is shown a return to a interpreter-based hermeneutics, such as practiced by Augustine. 碩士<br>國立臺灣大學<br>生物化學暨分子生物學研究所<br>93<br>Hepatitis delta virus (HDV) is a satellite virus of hepatitis B virus and requires hepatitis B virus surface antigens for the production of virus particles. HDV contains a circular RNA genome. Inside HDV particles, there are two distinct delta antigens, the small form (HDAg-S; 195 a.a.) and the large form (HDAg-L; 214 a.a.). Both HDAgs are identical in the N-terminal region, but the large form has extra 19 amino acid residues at the C-terminus. HDAg-S is essential for the viral RNA replication, whereas HDAg-L is required for virus assembly. HDV replicates in the nucleus. In the late stage of the viral life cycle, the RNA genome is associated with delta antigens as ribonucleoprotein (RNP) complexes that are exported from the nucleus to the cytoplasm for assembly. Our laboratory has previously identified a proline-rich nuclear export signal (NES) located at the C-terminus of HDAg-L, designated NES(HDAg-L). The NES(HDAg-L) mediates the nuclear export of HDAg-L via a chromosome region maintenance 1 (CRM1)-independent pathway. In addition, a cellular factor NES(HDAg-L)-interacting protein (NESI) was identified to be involved in the nuclear export of HDAg-L and the assembly of HDV. Sequence analysis of NESI revealed a putative actin-binding site from amino acid residues 4 to 13 and a bipartite nuclear localization signal from amino acid residues 193 to 209. In this study, the specific interaction between NESI and actin was demonstrated by GST pull-down assay. Deletion analysis indicated that the N-terminal 185 amino acid residues of NESI were sufficient to interact with HDAg-L. In addition, antibodies specific to the N-terminal 185 amino acid residues of NESI were generated. Indirect immunofluorescence staining with the NESI-specific antibodies demonstrated that endogenous NESI is localized in the nucleus of mammalian culture cells. These results suggest that the actin-binding activity of the nucleus-localized NESI protein may be involved in the CRM1-independent nuclear export pathway.

HDV causes liver damage by co-infection or superinfection with HBV. HDV is a spherical RNA virus that contains a single-stranded circular RNA genome of 1.7 kb in length. Hepatitis delta antigen (HDAg) is the only known protein of HDV. There are two forms of delta antigen, the small HDAg (HDAg-S, 195 a. a., 24 kDa) and the large HDAg (HDAg-L, 214 a. a., 27 kDa). The HDAg-S facilitates the replication of HDV RNA, whereas the HDAg-L is required for the viral assembly. Previous studies from our laboratory identified a novel proline-rich nuclear export signal at the C-terminus of HDAg-L, designated NES(HDAg-L). The NES(HDAg-L) mediates the nuclear export of HDAg-L via a chromosome region maintenance 1 -independent pathway. In addition, a cellular factor NES(HDAg-L)-interacting protein (NESI) was identified to be involved in the nuclear export of HDAg-L and the assembly of HDV. NESI consists of 467 amino acid residues. Sequence analysis of NESI revealed a putative actin-binding site from amino acid residues 4 to 13 and a bipartite nuclear localization signal from amino acid residues 193 to 209. The role of NESI involved in nuclear export is not fully understood. In this study, mechanisms involved in the NESI-mediated nuclear export and possible interactions between NESI and nuclear envelope proteins were investigated. By performing immunoprecipitation followed by LC-MS/MS analysis, lamin A/C was identified to interact with NESI. The specific interaction between NESI and lamin A/C was further confirmed by Western blot analysis following immunoprecipitation. In addition, colocalization of NESI and lamin A/C on the nuclear envelope was demonstrated by confocal microscopy. GST pull-down assay demonstrated that the N-terminal 31 to 116 amino acid residues are required for NESI to interact with lamin A/C. Sucrose gradient centrifugation analysis implicated that NESI, lamin A/C and the phenylalanine-glycine nucleoporins may be present in the same complex on nuclear envelope. To investigate the functional role of NESI involved in HDAg-L-associated nuclear export, shRNA to NESI or lamin A/C applied in an HDV assembly culture system. The results demonstrated that both shRNAs to NESI and lamin A/C specifically inhibited HDAg-L nuclear export and HDV assembly, but had no effect on the assembly of the small surface antigen of HBV. Taken together, these results indicate that NESI may form complexes with lamin A/C and the phenylalanine-glycine nucleoporins for nuclear export. 碩士<br>臺灣大學<br>生物化學暨分子生物學研究所<br>95<br>Hepatitis delta virus (HDV) is a spherical RNA virus, which is enveloped by the surface antigens of the hepatitis B virus. Inside the viral particles, HDV contains a single-stranded, negative sense, circular RNA genome of 1.7 kb and its only known products, small delta antigen (HDAg-S) and large delta antigen (HDAg-L). Previous studies from our laboratory identified a novel proline-rich nuclear export signal at the C-terminus of HDAg-L, designated NES(HDAg-L). In addition, host protein NESI which specifically interacts with NES(HDAg-L) participated in nuclear export of HDV RNP complex and virus assembly. NESI consists of 467 amino acid residues. It bears a putative actin binding site and a bipartite nuclear localization signal. Association of NESI(1-185) and actin was demonstrated in our laboratory. In addition, In NESI-knockdown cells, the cell mobility was increased in the scratch assay. However, regulations of the putative actin-binding domain from amino acid residues 4 to 13 in the cell mobility are unclear. In this study, the specific interaction between NESI and actin was further confirmed by co-immuoprecipitation analysis. GST pull-down assay indicates that the N-terminal 30 amino acid residues are required for NESI to interact with actin. The actin-binding ability of NESI protein was lost when mutations were introduced into the actin binding domain. These results indicate that the consensus sequence of the actinin-type actin binding domain is important for NESI-actin complex formation. Immunofluorescence staining demonstrated that NESI localized in the nucleus and the perinuclear region. The cytoplasmic NESI co-localized with F-actin. However, no co-sedimentation of NESI(1-30) peptide with purified F-actin was detected, and no significant changes in the equilibrium between G-actin and F-actin was observed in a in vitro binding assay. These results suggest that, unlike alpha-actinin, NESI may not function as F-actin bundling.

<div><div><div><div>Mass spectrometry is a widely used tool for efficient chemical characterization and identification. The development of electrospray ionization as a soft ionization method enables mass spectrometry for large biomolecule investigation. Protein as one of the most important classes of biomolecules, its structural changes including folding, unfolding, aggregation, degradation and post-translational modification all influence protein bioactivity. Protein characterization and identification are important for protein behavior mechanism understanding, which may further contribute to disease treatment development. Protein conformation changes are normally very fast, and the initial stages, which significantly influence the conformation changing pathway, normally occur in milliseconds or shorter time scale. Such a fast structural change is hard to be monitored using traditional bulk solution manipulations, and fast sample preparation methods are required. </div><div><br></div><div>In this thesis project, theta tips are applied as a microreactor and nESI-MS emitter to perform fast protein manipulation immediately before MS analysis. Theta tips can be operated in two modes. The first mode is for submillisecond time scale reactions. Proteins and reagents are loaded into different channels and directly sprayed out simultaneously. Proteins and reagents mix and react in the Taylor cone and subsequent droplets for submillisecond time scale. Through this method, pH induced protein folding is investigated and protein folding intermediates were captured. The second mode is for milliseconds or longer reactions. Differential voltages can be applied to each channel before ionization and spray. The electric field between the two channels induces in-tip electroosmosis, which lead to an in-tip mixing and reaction. In this mode, the reaction time is not limited by the droplet lifetime as in the first mode, but is controlled by electroosmosis time. By changing the electroosmosis square wave frequency and cycles, the mixing time can be elongated to milliseconds or longer, which is suitable for slower reaction study. </div><div>Joule heating is discovered during theta tip electroosmosis when samples are dissolved in buffer. The Joule heating effect is high enough to heat up the aqueous solution to at least 75 oC based on Raman thermometry measurement, while the actual peak temperature could be higher. The Joule heating effect in theta tip electroosmosis can be easily controlled by electroosmosis voltage, time, buffer concentration etc.. Proteins are thermally denatured by the Joule heating effect, and the denaturation extent correlates with Joule heating parameters. </div><div>With this results in hand, we are developing a protein melting temperature measurement method using theta tip Joule heating effect and mass spectrometry. This new melting temperature measurement method measures changes in protein mass and charge state distributions. Therefore, it could sensitively detect ligand loss and protein tertiary structural changes, which is an important compensation to current protein melting temperature measurement techniques like CD or DSC. Since the heating time is short and protein concentration for MS is low, protein aggregation and thermal fragmentation are highly avoided so a complete protein thermal unfolding process is monitored. Theta tip electroosmosis combining MS characterized protein thermal denaturation behavior from a new aspect.</div><div><br></div><div>Besides single protein folding and unfolding, protein identification and post-translational modification are important for proteomics study. The traditional bottom-up, top-down and middle-down methods are not able to both preserve intact protein mass and efficiently generate enough fragment peaks easily without performing gas phase dissociation. In this thesis, we also developed a new way to identify proteins combining limited trypsin digestion and mass spectrometry. Intact protein mass was preserved for protein size and PTM identification. Enough tryptic peptides were also generated for protein identification through database search.</div></div></div></div><div><br></div>

博士<br>國立臺灣大學<br>生物化學暨分子生物學研究所<br>93<br>Heaptitis virus (HDV) is the causative agent of acute and chronic liver diseases including fulminant hepatitis and accelerated liver cirrhosis. HDV particles contain a single-stranded circular RNA genome of 1.7 kilobases. The HDV antigenomic RNA contains an open reading frame which encodes delta antigens( HDAgs). There are two forms of HDAgs, the small HDAg (HDAg-S, 24 kDa) and the large HDAg (HDAg-L, 27 kDa). The two HDAgs are identical in the amino acid sequence, except that the HDAg-L contains an additional 19 amino acids at it C terminus. Nevertheless, functions of two forms of HDAgs are quite different. The HDAg-S is required for the HDV replication, whereas the HDAg-L has negative effects on genome replication is required for HDV assembly. The HDV genome RNA is associated with delta antigen to form ribonucleoprotein. Previous results have shown that a cellular factor, GAPDH interacted with HDV RNA. In addition, nuclear export signal that located in the C-terminus of HDAg-L from amino acid residues 198-210, designated NES(HDAg-L), has been identified. To investigate the biological role of GAPDH in HDV life cycle, cells were transfected with plasmid pSVD2 representing the dimeric HDV cDNA and proceeded to fluorescence in situ hybridization/immunofluorescence staining. In the presence of HDV replication, GAPDH relocalized from the cytoplasm to the nucleus. The biological significance of the interaction between GAPDH and HDV RNA was further elucidate by examing the possible role of GAPDH on the catalysis of HDV ribozyme. This cis-cleavage activity of the HDV anigenomic RNA increase from 34 to 62 % following a 40-min incubation with 50 ng/ l of GAPDH. The enhancing effect of GAPDH on the catalysis of HDV ribozyme was reproducible and was not observed with bovine serum albumin (BSA). These results suggest a possible role of GAPDH involved in HDV multiplication. The process of host factor-mediated nucleocytoplasmic transport is critical for diverse cellular events in eukaryotes and the life cycle of viruses. A previously identified chromosome region maintenance 1 (CRM1)-independent nuclear export signal (NES) at the C-terminus of the large form of hepatitis delta antigen (HDAg), designated NES(HDAg-L) is required for the assembly of hepatitis delta virus (HDV) To look for interacting proteins of the NES(HDAg-L), yeast two-hybrid screening was applied using GAL4 binding domain fused to NES(HDAg-L) as bait. Among the positive clones, one encodes a protein, designated NESI (NES-interacting protein) that specifically interacted with the wild-type NES(HDAg-L), but not with the export/package-defective HDAg-L mutant, NES*(HDAg-L) in which Pro-205 has been replaced by Ala. Northern blot analysis revealed NESI as the gene product of a 1.9 kb endogenous mRNA transcript that is present predominantly in human liver tissue. NESI consists of 467 amino acid residues and bears a putative actin-binding site and a bipartite nuclear localization signal. Specific interaction between HDAg-L and NESI was further confirmed by coimmunoprecipitation and immunofluorescence staining. Overexpression of an antisense NESI RNA abolished HDAg-L-mediated assembly of HDV genomic RNA into virus-like particles. These data indicate a critical role of NESI, through the interaction of HDAg-L, involved in the assembly of HDV.

Catholic Church museums have their own very particular characteristics that distinguish, or should distinguish them, from other museological institutions managed by other entities, even if they share collections with similar characteristics. These elements of differentiation may be summarized and centralized on the intrinsic pastoral mission of any ecclesiastical museum, so that this very dimension should shape the strategic definition of their mission, vocation, goals and objectives, and, consequently, direct and influence the museological functions, based on indispensable quality standards. Based on the large and creative potentialities of ecclesiastical museums serving the fundamental Church mission – evangelization purposes –, this work aims to analyze the way this pastoral dimension is developed in the museological context of the Patriarchate of Lisbon, investigating the obstacles and needs that arise in the operationalization of that important vector. As a way of answering the investigation problem, we will focus, in an evolutional way and in an initial approach, on some of the problems associated with the cultural goods and museums of Church, trying to theoretically and conceptually frame these themes, connecting them with Church mission and action, and also with some aspects of museological theory and practice. On a second approach we will perform a diagnosis of the museological reality of the Patriarchate of Lisbon, identifying the existing museological units, making an individualized critical analysis as well as analyzing the entire diocesan tissue. At last, and considering identified obstacles and needs, it will be formulated a sector qualification proposal, consubstantiated through the construction of a ‘Network of Museums and visitable Collections of the Patriarchate of Lisbon’
