Sleep is well-established to become more superficial and fragmented as we age, with deficits in cognitive processing also commonly observed. While effects have been identified in both humans and mice (used in this thesis), there are important species differences in these findings and importantly, very little is known about the neural dynamics underlying these changes. By integrating several state-of-the-art approaches from putative single unit electrophysiological recordings to behavioural and pharmacological assessments, this thesis aimed to provide novel insights into the neural mechanisms i

Protein with Tau-like repeats (PTL-1) is the sole Tau /MAP2/MAP4 homolog in Caenorhabditis elegans. Dysregulation of Tau is a pathological hallmark of neurodegenerative diseases such as Alzheimer’s disease. Therefore, reducing Tau levels has been suggested as a therapeutic strategy. We used PTL-1 to model the biological functions of a Tau-like protein without the complication of functional redundancy. Firstly, our data indicate that PTL-1 in the nervous system mediates the oxidative stress response in a pathway that may involve the C. elegans homolog of the Nrf2 transcription factor, SKN-1. I

This thesis examined the age-related impairments in the function of motor neurones, the connection between our spinal cord and the muscles, and the use of exercise as a therapy to counteract this dysfunction. Older adults have reduced function of the motor neurones, which could contribute to muscle weakness and physical function impairments observed in this population. Exercise can be used as medicine to restore motor neurone function, contributing to improvements in muscular strength and general physical function in older adults.

Senescence is not a simple growth arrest, but is accompanied by a host of phenotypic changes, including the generation of pro-inflammatory molecules, and is maintained by a network of auto- and paracrine reinforcement. Senescence is now also understood to occur in post-mitotic cells, including neurons - contrary to the former definition of senescence occurring exclusively in proliferating cells. Chemotaxis is a powerful paradigm to study how sensory stimulations drive orientation behaviors in an organism. Drosophila larvae navigate odor gradients by controlling the duration of runs and the direction of turns. A turn is preceded by lateral head sweeps (“casts”) that sample the stimulus from the surroundings. In addition, larvae correct their course towards the odor source during runs, a phenomenon called “weathervaning”. Peristaltic waves that propagate along the body axis drive forward run events.

Abstract:
Ensuite,
