<p> A neuron&rsquo;s unique physiological waveform arises from its palette of ion channels and receptors, as superimposed on its geometrical structure. The crustacean stomatogastric ganglion (STG), a small rhythmic motor circuit, exhibits fourteen identified neuron types with highly-conserved physiological waveforms and complex morphologies. In this thesis, I examine how morphology shapes neuronal physiology in the STG. Using high-resolution neuronal reconstructions and a suite of computational tools, I quantify numerous morphological features of four STG neuron types. This work revealed remar

Connexins are transmembrane proteins that form hemichannels allowing the exchange of molecules between the extracellular space and the cell interior. Two hemichannels from adjacent cells dock and form a continuous gap junction pore, thereby permitting direct intercellular communication. Connexin 36 (Cx36), expressed primarily in neurons, is involved in the synchronous activity of neurons and may play a role in aberrant synchronous firing, as seen in seizures. To understand the reciprocal interactions between Cx36 and seizure-like neural activity, we examined three questions: a) does Cx36 defic

In the larval stages of Drosophila, CA and prothoracic gland (PG) are located in a structure called ring gland. Previous report indicates that some neurosecretory cells near the mushroom bodies extent their axonal processes into CA of the ring gland. However, the developmental changes of CA and the upstream regulator of those CA-innervated neurons are still unclear. In this thesis, I used confocal analysis

Abstract:
We have evaluated the spatial relationship between local blood flow and the structural integrity of layer 5 cortical neurons in YFP-H transgenic mice 2-10 h after stroke. Fortuitously, ischemic zones could be finely mapped with respect to dysmorphic YFP labelled axons and dendrites using histology since texas-red dextran used to measure blood flow in vivo was trapped within fixed ischemic vessels. Ischemic damage to layer 5 neurons located at the border of ischemia was contained within apical tuft dendrites and did not propagate to deeper dendrites despite the potential for diffusion of releas

Next generation sequencing technologies are changing the way scientific experiments and diseases diagnostics are performed and thus will allow what is called personalized medicine. The sense of presented thesis is to make survey of new approaches to DNA sequencing and demonstrate usage and constraints of bioinformatic analytical tools available to day. Discussed techniques are then applied to the case study of finding molecular basis for rare hereditary disease. Abstract:
Imaging methods for visualizing synaptic contacts within neuronal circuits are under constant development and could contribute to the generation of a connectome of the striatum, which will be crucial to decode how striatal cells integrate motor information1–4. We attempted to optimize the mammalian GFP reconstitution across synaptic partners (mGRASP) technique to better understand how spiny projection neurons (SPNs) integrate input information from cortical intratelencephalic (IT) and pyramidal tract (PT) neurons. In order to do that, we set out to map corticostriatal inputs to SPNs with

Few of these systems provide function that renders only two or more volume data at the same time. In this thesis, we build a texture-based multi-volume visualization system that renders up to six channel volume image. The system is applied to render volume images of neurons. To enhance the neuron in each volume, our system supports many “real-time” effects such as depth enhancement, NPR, lighting, transfer function, and pre-integrated volume rendering. These

The neuronal and molecular mechanisms underlying behavioral responses triggered by fear have received wide interest in the last few years, and various preclinical studies have addressed potential treatments for fear-related disorders. The central histaminergic system is an important modulator of memory related to adverse events and the integrity of the brain histamine system is necessary for the consolidation of this type of memory. Therefore, the use of antihistaminic drugs could be useful for the treatments of conditions such as obsessive-compulsive disorders, post-traumatic stress disorders
