Abstract:

In this paper we discuss the inverse problem of the identification of mechanical stresses by measuring the deformation of an electric potential field in a so called differential strain gauge (D-DMS). We derive a mathematical model, where the forward operator is given in terms of an elliptic boundary value problem. Derivatives of the forward operator are considered and the solution of the inverse problem via a least-squares minimization is introduced. Here, the discretized problem is solved with the Gauss-Newton method. Numerical studies of practical interest are presented.

DNA damage in spermatozoa is a crucial contributor to spontaneous abortion, severe genetic disease in the offspring and infertility. The chromatin of spermatozoa is highly compacted, transcriptionally and translationally silent, hence lacking DNA damage response (DDR). A technology for optimization of potential parameters from condensed phase simulations (POP) is discussed and illustrated. It is based on direct calculations of the derivatives of macroscopic observables with respect to the potential parameters. The derivatives are used in a local minimization scheme, comparing simulated and experimental data. In particular, we show that the Newton Trust-Region protocol allows for accurate and robust optimization.
