Abstract:
Zinc finger nucleases (ZFNs) induce double-strand DNA breaks at specific recognition sites. ZFNs can dramatically increase the efficiency of incorporating desired insertions, deletions, or substitutions in living cells. These tools have revolutionized the field of genome engineering in several model organisms and cell types including zebrafish, rats, and human pluripotent stem cells. There have been numerous advances in ZFN engineering and characterization strategies, some of which are detailed in this work. The central theme of this dissertation is improving the activity and specificity of engineered zinc finger nucleases with the ultimate goal of increasing the safety and efficacy of these tools for human therapy. As a first step, I undertook a large-scale effort to demonstrate that the modular assembly method of ZFN synthesis has a significantly higher failure rate than previously reported in the literature. This strongly suggested that engineering of ZFNs should better account for context-dependent effects among zinc fingers. The second advance reported in this dissertation is a method for biasing repair of zinc finger protein-induced DNA breaks toward homology-driven rather than error-prone repair in the presence of a donor template. Catalytically inactivating one monomer of a ZFN dimer results in a zinc finger nickase (ZFNickase) whose cleavage preference is directed at only one DNA strand. In human cell reporter assays, these ZFNickases exhibit a higher likelihood of repair by homology-driven processes, albeit with reduced absolute rates of correction. With further optimization, zinc finger nickases could provide a safer alternative to ZFNs in the context of gene correction therapies. Third, realizing there was no robust method for determining off-target cleavage sites of ZFNs in a genome-wide manner, I validated a collaborator’s novel in vitro selection system in human cells by identifying eight new potential off-target cleavage sites for a ZFN pair currently being used in clinical trials. Although it is unlikely these low-frequency mutations would be deleterious to patients, these results demonstrated that ZFNs induced more off-target effects than had been appreciated by previous work in the field. Collectively, the findings of this dissertation have contributed to more robust strategies for designing and evaluating ZFNs.

Ah! This show is a physical explanation of discovery - an attempt to better understand the role of contemporary hand thrown pottery and what it means to make it. A hand thrown pot can contain nourishment as well as it can provide it. Pottery can infuse artful practice and a direct relationship to the producer into everyday objects. It is the potter's responsibility to continue to push form and surface to make the best pots possible. Well crafted handmade objects support the argument for locality and autonomy, which give them greater value in a contemporary context rooted in ambiguous commodity consumption. It is our responsibility to craft the things we consume.

Although F. Scott Fitzgerald's The Great Gatsby has received extensive critical attention since the middle of the century, there remains an unaddressed and unanswered question that demands further exploration: what makes Gatsby "great?" It seems that the source of Gatsby's greatness, for narrator Nick Carraway, is that Gatsby has a quality that sets him apart from others: it is not a "flabby impressionability," but a "heightened sensitivity to the promises of life" and "an extraordinary gift for hope" that Nick has never seen before, nor does he expect to see again (6). I contend that what Nick sees as Gatsby's belief and hope in the possibilities of life are embodied in what Kierkegaard discusses in his works Either/Or and Fear and Trembling as choosing to live an ethical existence free from the pain of the material world. Gatsby makes this choice (of living ethically) when the young James Gatz chooses to become Jay Gatsby and free himself from the pain of losing Daisy. Through this choice, according to Kierkegaard, the ethical individual is inducted into the knighthood as a knight of infinity. If the knight makes one more movement, he becomes a knight of faith who believes, "Nevertheless I have faith that I will get her--that is, by virtue of the absurd, by virtue of the fact that for God all things are possible" (Fear and Trembling 46). Gatsby is a "son of God" that "sprang from his Platonic conception of himself"; he is a Kierkegaardian knight who has chosen an ethical existence; he is a knight who has the ability to look impossibility in the eye and still have faith to the point of absurdity, even if a reunion with his love (Daisy) is not possible. This is Gatsby's "extraordinary gift for hope," which Kierkegaard attributes to "the only great one," the knight of faith. Thus, Nick's narrative is not only a canonization of his "great" knight, but an imaginative recollection that traces the movements of his knight, Gatsby, down the same path Kierkegaard imaginatively follows and observes his great knight of faith in Fear and Trembling.

Abstract:
I denna uppsats undersöks hur de sex professionella idrottsutövarna, Nicklas Bäckström, Alexandr Legkov, Abeba Aregawi, Marija Sjarapova, Nicklas Axelsson och Justin Gatlin framställs i två av Sveriges största morgon- respektive kvällstidningar: Dagens Nyheter och Aftonbladet. Detta efter att de alla sex idrottsutövarna, blivit avslöjade som fuskare och gett ett positivt resultat under en eller flera dopingkontroller. Observerade skillnader och likheter i rapporteringen av de idrottsutövande har analyseras utifrån två relevanta teorier om objektivitet och nationalism: Westerståhls objektivitetsmodell och Billigs teori gällande nationalism. De i studien utvalda texterna har granskats huruvida de utmålar värderingar om ”Vi” och ”dom andra”. Texterna har analyserat med komparativ kvalitativ textanalys med utgångspunkt i de tre frågeställningarna, Hur stor plats i medierna får de individuella fallen? Är det någon skillnad i hur utövarna och deras dopingfall framställs i två av Sveriges största tidningsmedier och om så är fallet, vilka är de? Dessutom undersöks huruvida nationaliteten på utövarna påverkar hur de återges i svenska medier.   I analysen ingår 65 artiklar där resultaten visar klart och tydligt att vissa idrottsutövare fick en mer harmlös behandling där de nästintill sågs som offer medan andra fick utstå hård kritik där också högre grad av negativt värdeladdade ord tenderade att förekomma. Utrymmet de individuella idrottsutövarna fick i de två utvalda tidningarna, Dagens Nyheter och Aftonbladet tenderade att skifta rejält. Nationalitet och etnicitet spelade stor roll när journalisterna rapporterade om dopingfallen. Tydligt var att journalisterna strävade, eller omedvetet stod för ett skapande av ”Vi” och ”dom andra”, där ett återkommande tema var att Ryssland utmålades som fienden medan vi svenskar minsann aldrig fuskar. Analysen visar att Dagens Nyheter och Aftonbladet tenderar att växla i sin attityd när de rapporterar om händelserna kring de sex dopingfallen. I den svenska ishockeyspelaren Nicklas Bäckströms fall var det tydligt att de två tidningarna snarare ventilerade starka åsikter istället för att rapportera om händelseförloppet vilket journalisterna gjorde tydligare i de övriga fem dopingfallen.

Abstract:

Individuals diagnosed with autism face multiple challenges especially when aging out of the public school system, such as finding a job or a place to stay while their parents are at work. A day program is a service, which can provide support for individuals with autism. The purpose of this project was to write a grant proposal for the implementation of a professional staff training program at Nick's Community, which is a nonprofit day program. The major components of the proposed training program are conducting new and ongoing training to the staff at Nick's Community through workshops, small group activities, and hands-on participation. The submission for funding of this grant was not required for the successful completion of this thesis grant-writing project. Implications for social work research and practice are included.

Abstract:
Surface damage can be introduced into rotor disc components during assembly, maintenance and overhaul operations for aircraft engines. This is known as handling damage and is often in the form of surface scratches, which are known to reduce total component fatigue life. This work provides a holistic understanding of the effect of artificial scratch damage on the fatigue performance of a nickel-based superalloy used for compressor and turbine disc applications. Following extensive analysis of results from previous test programmes completed at Rolls Royce plc., a series of low cycle fatigue tests were performed on Alloy 720Li specimens. A test matrix was designed to isolate and identify every factor that may contribute to the fatigue properties of scratch damaged components. The geometry of a scratch, which causes a local stress concentration and increases the ‘effective Kt’ at the scratch root, is the most significant factor in controlling fatigue performance. Scratches cause a decrease in crack initiation life, thereby reducing total component fatigue life. Compressive residual stresses induced by shot peening and the scratching process itself are beneficial to component life. Varying levels of scratch damage were fully characterised by scanning electron microscopy, microhardness testing and electron backscatter diffraction to show the severe microstructural modification and increased hardness caused by the scratching process. Computational modelling was carried out to determine geometrical effects of scratches on local stress and strain fields. The ‘effective Kt’ values under scratches were calculated, which helps to predict fatigue performance. This type of comprehensive investigation is important for damage tolerance lifing approaches and helps to minimise premature component retirement.

Com a constante evolução tecnológica nas áreas aeronáutica, espacial, automobilística, dentre outras, o uso de materiais com elevadas resistência mecânica e à corrosão em altas temperaturas tem sido cada vez mais necessário. Dentre os materiais que atendem a estes requisitos em temperaturas até 815 ºC está a liga à base de níquel Pyromet® 31V (N07032). Esta liga é utilizada na confecção de válvulas de exaustão em motores diesel de alta potência nas quais é exigida, além de resistência a altas temperaturas, uma grande resistência à sulfetação, à corrosão e à fluência. Entretanto, devido à sua grande resistência mecânica e baixa condutividade térmica, ocorre uma concentração de calor principalmente na interface cavaco-ferramenta dificultando assim a sua usinabilidade. Este trabalho teve como objetivo estudar a usinabilidade da liga Pyromet® 31V nas condições laminada à quente (42 HRC) e seguida de solubilização e envelhecimento (37 HRC) utilizando-se ferramentas de metal duro com e sem recobrimento, com lubrificação abundante e com a técnica de MQF. Em termos de vida da ferramenta, os melhores resultados para as ferramentas recobertas foram para avanços de 0,15 (laminadas) e 0,12 mm/rot. (solubilizadas). Devido à imprevisibilidade na formação do desgaste de entalhe e APC, não houve uma tendência clara de elevação ou queda na vida e/ou volume de material removido em função do avanço. Os mecanismos de desgaste atuantes nas ferramentas recobertas foram abrasão, attrition e martelamento e nas sem recobrimento foram difusão e abrasão. Na usinagem com MQF obteve-se bom acabamento superficial, vida razoável e reduzido volume de material removido. Já a ausência de recobrimento ocasionou uma redução de 70% no volume removido. Os cavacos gerados foram diversificados, mas predominantemente longos, destacando-se também a formação de cavacos duplos

However, due to its high mechanical strength and low thermal conductivity, it occurs concentration of heat in the workpiece-tool and tool-chip interfaces, resulting in low machinability. This work aimed to study the machinability, using the cemented carbide coated and uncoated tools, of the hot-rolled (42 HRC) and solubilizing-aging (37 HRC) conditions of the Pyromet® 31V alloy, using two conditions, abundant fluid and MQF techniques. The best results for tool life with coated tools were with 0.15 (hotrolled) and 0.12 mm/rev. (solubilizing-aging) feed rates. Due to the unpredictability in the formation of the notch wear and built-up edge (BUE) it wasn’t observed a noticed tendency to increase or decrease in tool life and/or volume of removed material due to the feed rates. The wear mechanisms in coated tools were abrasion, attrition and hamming and in the uncoated tools were diffusion and abrasion. In the turning of this alloy with MQF, good surface finish, reasonable tool life and reduced volume of material removed were obtained. However, the absence of coated caused a 70% reduction in volume removed. The chips generated were of several types, but predominantly long, highlighting also the doubles chips formation

Resumo: Uma prática comum entre pessoas que frequentam um mesmo espaço (como estudantes, por exemplo) é o ato de atribuir apelidos para os colegas, entretanto esse gesto determina e é determinado pela posição-sujeito assumida por ele no discurso. Esta pesquisa teve como objetivo analisar esse deslocamento que o sujeito sofre ao receber um apelido a partir do enunciado "Dadinho é o caralho, meu nome agora é Zé Pequeno, porra" produzido por um dos personagens do filme Cidade de Deus, de Fernando Meirelles. Esta escolha foi determinada primeiramente pelo trabalho que este filme tem com a linguagem, principalmente no que diz respeito a nomes e apelidos. Outro fator determinante para esta escolha foi a relevância, nos cenários nacional e internacional desse filme, bem como a grande circulação do enunciado analisado, de modo que ele se tornou emblemático quando se fala do filme em questão. Para nortear a análise, nos baseamos no referencial teórico da Análise de Discurso materialista, bem como da Semântica da enunciação, trabalhando, desse modo, com o sujeito que "é atravessado pela linguagem e pela história, sob o modo imaginário" de modo que ele "só tem acesso a parte do que diz. (...). Ele é sujeito à língua e à história, pois para se constituir, para (se) produzir sentidos ele é afetado por elas" (ORLANDI, 1999:49). A partir disso, vimos que os nomes/apelidos que o sujeito assume durante a sua vida têm uma historicidade e que essa passa a constituir o sujeito nomeado/apelidado. Do mesmo modo, retomamos o trabalho de Hashiguti (2008) sobre corpo de memória para mostrar os sentidos que o corpo faz circular e como ele entra nesse processo de atribuição de apelidos. Por fim, mostramos o deslocamento que o personagem Dadinho sofre ao trocar o seu apelido para Zé Pequeno, mostrando que o apelido determina e é determinado pela posição-sujeito que o sujeito ocupa no discurso
Abstract: A common practice among people that attend the same place (students, for instance) is the act of attribute nicknames for their mates, however this action determines and is determined by the subject-position which is assumed by it in the discourse. This research's goal was to analyze the displacement that the individual suffers when he receives a nickname from the enunciation "Li'l Dice my ass, my name is now Li'l Zé now" produced by one of the characters from the movie City of God, by Fernando Meirelles. This choice was made, first, because of the way this movie deals with the language, mainly considering the names and nicknames. Another important factor for this choice was the relevance, in the national and international scenarios of this movie and because of the big spread of the analyzed enunciation as well, in a way that it became emblematic when people talk about this movie. To lead the analysis, we were based on the theoretical of the materialist Discourse Analysis and on the Semantics of enunciation as well, working with the individual that "is crossed by the language and by the history, in the imaginary way" and he "only has access to part of what he says. (¿). He is subjected to the language and to the history, because to constitute and to produce senses he is affected by them" (ORLANDI, 1999:49). From this, we saw that the names/nicknames assumed by the individual during his life have a historicity and that it starts to constitute the named/nicknamed individual. In the same way, we resumed the project of Hashiguti (2008) about the body of memory to show the senses that the body makes circulate and how it begins this process of nicknames' attribution. Finally, we showed the displacement suffered by the character Li'l Dice when he changes his nickname to Li'l Zé, showing that the nickname determines and is determined by the subject-position that the individual occupies in the discourse

Abstract:
Studies syfte är att undersöka samt problematisera hur företaget Nick’s medierar hälsa genom deras förpackningsdesign. Detta analyseras utifrån tre förpackningar och tre instagrambilder av företagets produkter. Analysen av materialet utförs i två steg. Inledningsvis via en semiotisk analys med målet att identifiera relevanta visuella attribut i förhållande till studiens syfte och frågeställning. Som sedan analyseras mer djupgående via en diskursanalys med utgång i teorier kring hälsa och förpackningsdesign. Resultatet visar på att Nick´s genom sin förpackningsdesign framställer hälsa genom en övergripande association till ett naturligt ursprung samtidigt som de patologiserar ohälsa och åberopar ett individuellt hälsoansvar. Avslutningsvis framställs hälsa även som en social tillhörighet som kan uppnås via konsumtion av företagets produkter.
