Příspěvek vychází z projektu „Stárnutí, věk a diskriminace – nové souvislosti pro českou společnost“ a zamýšlí se nad pojetím a tematizací věkových norem v českém socio-kulturním prostředí. Stať je uvedena přehledem teorie věkových norem, jejich definic a měření a následně mapuje základní diskusi v oboru. V empirické části využíváme data z 31 kvalitativních, polo-strukturovaných rozhovorů, které se zaměřovaly na reflexi věku, věkových norem a věkově ukotvených událostí v průběhu životního cyklu komunikačních partnerů v různém (chronologickém) věku, připadajícím různým kohortám a generacím. Na třech typech věkových norem – implicitních, formálních a reprodukčních – ilustrujeme variabilitu sankcí provázejících jejich porušení a nabízíme k diskusi otázku využití kohortního přístupu k dalšímu zkoumání věkových norem jako významných prvků sociální struktury.

Abstract Biological processes (like microbial growth & physiological response) are usually dynamic and require the monitoring of metabolic variation at different time-points. Moreover, there is clear shift from case-control (N=2) study to multi-class (N>2) problem in current metabolomics, which is crucial for revealing the mechanisms underlying certain physiological process, disease metastasis, etc. These time-course and multi-class metabolomics have attracted great attention, and data normalization is essential for removing unwanted biological/experimental variations in these studies. However, no tool (including NOREVA 1.0 focusing only on case-control studies) is available for effectively assessing the performance of normalization method on time-course/multi-class metabolomic data. Thus, NOREVA was updated to version 2.0 by (i) realizing normalization and evaluation of both time-course and multi-class metabolomic data, (ii) integrating 144 normalization methods of a recently proposed combination strategy and (iii) identifying the well-performing methods by comprehensively assessing the largest set of normalizations (168 in total, significantly larger than those 24 in NOREVA 1.0). The significance of this update was extensively validated by case studies on benchmark datasets. All in all, NOREVA 2.0 is distinguished for its capability in identifying well-performing normalization method(s) for time-course and multi-class metabolomics, which makes it an indispensable complement to other available tools. The article deals with the polemical discourse of A.S. Norov, who was the hero of the war 1812, which he leads with Leo Tolstoy from the point of view of the direct participant in the battles with Napoleon. Analyzing then description of the events of the Patriotic War of 1812 in the novel of Leo Tolstoy “War and Peace”, A.S. Norov opposes them to a description which was made by the hero Yermolov of the war of 1812. The authors speculate that the polemics of A.S. Norov with the Leo Tolstoys’ point of view to the driving forces in the war of 1812 would not be so acute if it is considered in the categories of literary criticism, as a polemical discourse of the work with artistic fiction (the novel by Leo Tolstoy) with a work of documentary where fiction is excluded (wroks by A.S. Norov).

ABSTRACT Elevated levels of fluoroquinolone resistance are frequently found among Escherichia coli clinical isolates. This study investigated the antibiotic resistance mechanisms of strain NorE5, derived in vitro by exposing an E. coli clinical isolate, PS5, to two selection steps with increasing concentrations of norfloxacin. In addition to the amino acid substitution in GyrA (S83L) present in PS5, NorE5 has an amino acid change in ParC (S80R). Furthermore, we now find by Western blotting that NorE5 has a multidrug resistance phenotype resulting from the overexpression of the antibiotic resistance efflux pump AcrAB-TolC. Microarray and gene fusion analyses revealed significantly increased expression in NorE5 of soxS, a transcriptional activator of acrAB and tolC. The high soxS activity is attributable to a frameshift mutation that truncates SoxR, rendering it a constitutive transcriptional activator of soxS. Furthermore, microarray and reverse transcription-PCR analyses showed that mdtG (yceE), encoding a putative efflux pump, is overexpressed in the resistant strain. SoxS, MarA, and Rob activated an mdtG::lacZ fusion, and SoxS was shown to bind to the mdtG promoter, showing that mdtG is a member of the marA-soxS-rob regulon. The mdtG marbox sequence is in the backward or class I orientation within the promoter, and its disruption resulted in a loss of inducibility by MarA, SoxS, and Rob. Thus, chromosomal mutations in parC and soxR are responsible for the increased antibiotic resistance of NorE5.

One of the notable personalities during the reign of the Emperor Nicholas I was Abraham Sergeyevich Norov who occupied the post of the Minister of People’s Education from 1853 to 1858. In domestic historical literature the opinion has been rooted that he was a model of a conservative intellectual, a weak-willed person who fell under the influence of other people. The paper attempts to find out how A. S. Norov was treated by professors at Russian universities who often communicated with him and what they thought about him. On the basis of an analysis of their memoirs and letters the author comes to the conclusion that A. S. Norov evoked respect from the entire teaching staff community, enjoyed well-deserved authority and trust.

ABSTRACT Analysis of the Rhodobacter sphaeroides 2.4.3 genome revealed four previously unidentified sequences similar to the binding site of the transcriptional regulator NnrR. Expression studies demonstrated that three of these sequences are within the promoters of genes, designated paz, norEF, and cdgA, in the NnrR regulon, while the status of the fourth sequence, within the tat operon promoter, remains uncertain. nnrV, under control of a previously identified NnrR site, was also identified. paz encodes a pseudoazurin that is a donor of electrons to nitrite reductase. paz inactivation did not decrease nitrite reductase activity, but loss of pseudoazurin and cytochrome c2 together reduced nitrite reduction. Inactivation of norEF reduced nitrite and nitric oxide reductase activity and increased the sensitivity to nitrite in a taxis assay. This suggests that loss of norEF increases NO production as a result of decreased nitric oxide reductase activity. 2.4.3 is the only strain of R. sphaeroides with norEF, even though all four of the strains whose genomes have been sequenced have the norCBQD operon and nnrR. norEF was shown to provide resistance to nitrite when it was mobilized into R. sphaeroides strain 2.4.1 containing nirK. Inactivation of the other identified genes did not reveal any detectable denitrification-related phenotype. The distribution of members of the NnrR regulon in R. sphaeroides revealed patterns of coselection of structural genes with the ancillary genes identified here. The strong coselection of these genes indicates their functional importance under real-world conditions, even though inactivation of the majority of them does not impact denitrification under laboratory conditions.

Hypotézy norem složené z více podmínek jsou v právním řádu natolik rozsáhlé, že jejich sestavení je časově i intelektuálně náročné, a tudíž v řadě případů může jejich praktickou aplikaci podpora automatizovaného systému významně usnadnit, zlevnit a zkvalitnit. Takový systém ovšem vyžaduje nejprve model právních norem. V tomto článku explicitně formuluji hypotézy některých právních norem z oblasti práva na informace, a to konkrétně k hmotněprávním důvodům poskytování informací, poskytování utajovaných informací, poskytování informací o platech z veřejných prostředků a vydání rozhodnutí o odmítnutí žádosti o informace. Metodu v úvodu zasazuji do kontextu a dále ukazuji, jakou mají právní normy strukturu, definuji jejich funkční a strukturní vzorce na úrovni vět a navrhuji postup pro sestavení řetězce vyplývání a jeho pokrácení. Popisuji, jak se individualizací, prokázáním skutkového stavu a výběrem z alternativ tvoří z normy argument.

Renewal is defined as the recovery of an extinguished response if extinction and retrieval contexts differ. The context dependency of extinction, as demonstrated by renewal, has important implications for extinction-based therapies. Persons showing renewal (REN) exhibit higher hippocampal activation during extinction in associative learning than those without renewal (NOREN), demonstrating hippocampal context processing, and recruit ventromedial pFC in retrieval. Apart from these findings, brain processes generating renewal remain largely unknown. Conceivably, processing differences in task-relevant brain regions that ultimately lead to renewal may occur already in initial acquisition of associations. Therefore, in two fMRI studies, we investigated overall brain activation and hippocampal activation in REN and NOREN during acquisition of an associative learning task in response to presentation of a context alone or combined with a cue. Results of two studies demonstrated significant activation differences between the groups: In Study 1, a support vector machine classifier correctly assigned participants' brain activation patterns to REN and NOREN groups, respectively. In Study 2, REN and NOREN showed similar hippocampal involvement during context-only presentation, suggesting processing of novelty, whereas overall hippocampal activation to the context–cue compound, suggesting compound encoding, was higher in REN. Positive correlations between hippocampal activation and renewal level indicated more prominent hippocampal processing in REN. Results suggest that hippocampal processing of the context–cue compound rather than of context only during initial learning is related to a subsequent renewal effect. Presumably, REN participants use distinct encoding strategies during acquisition of context-related tasks, which reflect in their brain activation patterns and contribute to a renewal effect.

I Noreg tar dette form av eit 50 timars kurs i samfunnskunnskap for innvandrarar frå utanfor EØS-området. I denne samanhengen er det spenningar mellom einsidig vektlegging på arbeidslivet og på eit breiare fokus på samfunnsdeltaking. Det er også spenningar mellom myndiggjerande ambisjonar på den eine sida og ambisjonar om tilpassing til norske verdiar og tradisjonar på den andre. Denne artikkelen vil drøfta den medborgaridentiteten som vert konstruert for innvandrarar i samfunnskunnskap i dette spenningsfeltet. Det empiriske materialet er læremiddel som er i bruk i opplæring av innvandrarar i samfunnskunnskap.

Chaperone therapy is a newly developed molecular approach to lysosomal diseases, a group of human genetic diseases causing severe brain damage. We found two valienamine derivatives, N-octyl-4-epi-β-valienamine (NOEV) and N-octyl-β-valienamine (NOV), as promising therapeutic agents for human β-galactosidase deficiency disorders (mainly GM1-gangliosidosis) and β-glucosidase deficiency disorders (Gaucher disease), respectively. We briefly reviewed the historical background of research in carbasugar glycosidase inhibitors. Originally NOEV and NOV had been discovered as competitive inhibitors, and then their paradoxical bioactivities as chaperones were confirmed in cultured fibroblasts from patients with these disorders. Subsequently GM1-gangliosidosis model mice were developed and useful for experimental studies. Orally administered NOEV entered the brain through the blood-brain barrier, enhanced β-galactosidase activity, reduced substrate storage, and improved neurological deterioration clinically. Furthermore, we executed computational analysis for prediction of molecular interactions between β-galactosidase and NOEV. Some preliminary results of computational analysis of molecular interaction mechanism are presented in this article. NOV also showed the chaperone effect toward several β-glucosidase gene mutations in Gaucher disease. We hope chaperone therapy will become available for some patients with GM1-gangliosidosis, Gaucher disease, and potentially other lysosomal storage diseases with central nervous system involvement.

Abstract:
Abnormally elevated postprandial glucose and triacylglycerol (TAG) concentrations are risk factors for cardiovascular disease in type 2 diabetes. The most effective time to exercise to lower postprandial glucose and TAG concentrations is unknown. Thus the aim of this study was to determine what time is more effective, either pre- or postdinner resistance exercise (RE), at improving postprandial risk factors in patients with type 2 diabetes. Thirteen obese patients with type 2 diabetes completed three trials in a random order in which they consumed a dinner meal with 1) no RE (NoRE), 2) predinner RE (RE → M), and 3) postdinner RE beginning 45 min after dinner (M → RE). Clinical outcome measures included postprandial glucose and TAG concentrations. In addition, postprandial acetaminophen (gastric emptying), endocrine responses, free fatty acids, and β-cell function (mathematical modeling) were measured to determine whether these factors were related to changes in glucose and TAG. The TAG incremental area under the curve (iAUC) was ∼92% lower ( P ≤ 0.02) during M → RE compared with NoRE and RE → M, an effect due in part to lower very-low-density lipoprotein-1 TAG concentrations. The glucose iAUC was reduced ( P = 0.02) by ∼18 and 30% during the RE → M and M → RE trials, respectively, compared with NoRE, with no difference between RE trials. RE → M and M → RE reduced the insulin iAUC by 35 and 48%, respectively, compared with NoRE ( P < 0.01). The glucagon-like peptide-1 iAUC was ∼50% lower ( P ≤ 0.02) during M → RE compared with NoRE and RE → M. Given that predinner RE only improves postprandial glucose concentrations, whereas postdinner RE improves both postprandial glucose and TAG concentrations, postdinner RE may lower the risk of cardiovascular disease more effectively.

In 1968, an antisemitic campaign, launched by the Polish government, caused around 13,000 Jews to leave Poland. About 2500 of these refugees came to Denmark, while only about 25 ended up in Norway. The migration to Norway could potentially have reached low hundreds, but as oral-history sources indicate, the Jewish congregation in Oslo turned down a government initiative in 1969. Based on written and oral sources, and secondary literature, I argue that there was an equally important factor differentiating the two countries. Comparing the Danish and Norwegian refugee reception policies, the article finds that Danish authorities and their NGO partners at decisive stages in the process were more proactive than their Norwegian counterparts in their efforts to persuade Polish Jews to come. The most critical point was in June 1969, when Denmark’s embassy in Warsaw started issuing Jews with automatic visas, while Norway retained its existing application process.

Modern scholarship on the textual history of Church Slavonic biblical translation recognizes two distinct revisions of the Church Slavonic Psalter from the early fourteenth century, Redaction III (sometimes called the ‘Athonite’ redaction) and Redaction IV, known only in the Norov psalter manuscript. Although they are both attested from the same period and in manuscripts of similar Bulgarian provenance, these two redactions are in some respects systematically different in their linguistic character, their approach to translational issues and their Greek textual basis. In the light of A.A. Turilov’s observation that the Mihanović Psalter, possibly the earliest witness to Redaction III, is written in the same hand as the greater part of the Norov Psalter, this paper examines the textual antecedents of the two redactions and the importance of the Mihanović Psalter as a link between them.

ABSTRACT In addition to causing diarrhea, Escherichia coli O157:H7 infection can lead to hemolytic-uremic syndrome (HUS), a severe disease characterized by hemolysis and renal failure. Differences in HUS frequency among E. coli O157:H7 outbreaks have been noted, but our understanding of bacterial factors that promote HUS is incomplete. In 2006, in an outbreak of E. coli O157:H7 caused by consumption of contaminated spinach, there was a notably high frequency of HUS. We sequenced the genome of the strain responsible (TW14359) with the goal of identifying candidate genetic factors that contribute to an enhanced ability to cause HUS. The TW14359 genome contains 70 kb of DNA segments not present in either of the two reference O157:H7 genomes. We identified seven putative virulence determinants, including two putative type III secretion system effector proteins, candidate genes that could result in increased pathogenicity or, alternatively, adaptation to plants, and an intact anaerobic nitric oxide reductase gene, norV. We surveyed 100 O157:H7 isolates for the presence of these putative virulence determinants. A norV deletion was found in over one-half of the strains surveyed and correlated strikingly with the absence of stx 1. The other putative virulence factors were found in 8 to 35% of the O157:H7 isolates surveyed, and their presence also correlated with the presence of norV and the absence of stx 1, indicating that the presence of norV may serve as a marker of a greater propensity for HUS, similar to the correlation between the absence of stx 1 and a propensity for HUS.

Salmonella possesses multiple enzymes that utilize NO as a substrate, and could therefore contribute to the organism's ability to resist nitrosative killing by macrophages. Flavorubredoxin is an oxygen-sensitive enzyme that reduces NO to nitrous oxide. The Salmonella enterica serovar Typhimurium norV gene encoding flavorubredoxin was disrupted and the NO sensitivity of the mutant was determined. The norV mutant showed a greater sensitivity to NO than wild-type S. Typhimurium, but did recover growth after a transient inhibition. The mutant phenotype suggests that multiple enzymes are employed by S. Typhimurium to detoxify NO under anaerobic conditions, one of which is flavorubredoxin.

Normy profesní etiky často stanoví advokátům při výkonu jejich profese odlišné standardy chování, než jaké by v totožné situaci platily pro obyčejného člověka. Článek se zabývá otázkou, jak můžeme tyto rozdíly ospravedlnit prostřednictvím různých etických teorií. Autor v něm nejprve charakterizuje profesní etiku advokátů jako soubor právních a morálních norem. Následně analyzuje samotný problém její justifikace. Pozornost je věnována nejprve různým deontologickým přístupům (např. Charles Fried, David Luban). Autor rozebírá jejich nedostatky a dospívá k závěru, že musí primárně zdůrazňovat význam morálního a racionálního aktérství, pokud mají být věrohodné. Vstřícně se pak staví k utilitaristickému ospravedlnění. Také to je podle něj v kombinaci s některými pragmatickými hledisky schopno profesní etiku advokátů věrohodně justifikovat. Obě skupiny přístupů lze tedy považovat za relevantní a není proto vhodné z debat o výše uvedené etice některý z nich vylučovat.

Abstract The results of experimental investigations on the removal of NOx from gases applying ozone as the oxidizing agent and the absorption of higher nitrogen oxides in the sodium hydroxide solutions are presented. The experiment was conducted using a pilot plant installation with the air flow rate 200 m3/h, being a prototype of a boiler flue gas duct and a FGD scrubber. It was shown that in the range of [NOref] = 50 ÷ 250 ppm the mechanism of NO ozonation depends on the molar ratio X = O3/NOref: for X ≤ 1.0 oxidation of NO to NO2 predominates and NO2 is poorly absorbed, for X >> 1.0 NO2 undergoes further oxidation to N2O5, which is efficiently absorbed in the scrubber. The stoichiometric molar ratio of complete conversion of NO into N2O5 is X = 1.5, in these studies to reach the effectiveness η ≥ 90% the molar ratio X was much higher (2.75).
