Full textThe aim of this work concerns the synthesis of selective NTS2 neurotensin analogues as potent antinociceptive agents followed by biological activities studies. Poor bioavailability of neurotensin is one of the major obstacles for therapeutic effects. Indeed, this tridecapeptide has a short half-life time due to enzymatic degradation and is too hydrophilic to cross blood-brain barrier. First, we developed different methods to obtain hydrophobic unnatural amino acids like (trimethyl)silylalanine, N-substituted glycines and unsatured α-amino acids. All these peptide building blocks combine lipophilic properties facilitating membrane permeability and enzymatic unrecognition advantages. Some of them have been incorporated in the C-terminal active fragment NT[8-13] in order to evaluate their impact on affinity and/or biological activities. Thus, a series of new peptides and pseudopeptides was prepared and in vivo behavior was studied using a pain model designed in Sherbrooke university. Full textThe Delta expendable launch vehicle has been flying onboard video cameras. The camera is an NTSC analog camera that directly modulates an FM transmitter. A standard FM deviation is used to maximize link performance while minimizing transmitted bandwidth. Pre-emphasis per CCIR recommendation 405 is used to improve the video signal-to-noise ratio. The camera and transmitter obtain power from either a separate battery or the vehicle power system. Lighting is provided by sunlight, or a light may be added when sunlight is unavailable. Multiple cameras are accommodated by either using multiple transmitters or by switching the individual cameras in flight. IRIG-B timing is used to correlate the video with other vehicle telemetry.

We will explore some of the tradeoffs and benefits between direct video insertion and data insertion into the SDI metadata. We will explore the impact transmission infrastructure has on time accuracy, positional control and data recovery when transporting HD-SDI video from source to destination. Further some the side effects of compression tools used (e.g. JPEG 2000, MPEG, H.264) to accomplish transport will be discussed.

Full textThe need exists to encode NTSC composite video into a serial digital bit stream for encryption prior to transmission. Further, this need exists in places where power and volume are at a premium. This paper describes a simple solution using the Continuously Variable Slope Delta Modulation technique of encoding all lines and fields in real time and is usable with clock rates from 5 to 25 MHz. The circuits presented use only a 5-volt power supply and two active devices: a comparator and either a dual flip-flop or serial shift register.

Full textIn our first installment, analog and HD video terms were compared. It was shown that resolution is three-dimensional in HD. High HD data rates force the use of video compression in order to transport video. Compression compromises video detail and introduces latency. Latency affects the overlay of time critical measurements. It is therefore important to time stamp at the source. In this volume, the focus is on the key regions of the HD video frame and metadata. SMPTE specifications are the foundation on which MISB builds its use of metadata. It will be shown that only two KLV packets can hold all TSPI and calibration data with frame-by-frame updates. This capacity is new in HD. Metadata is permanently merged with images and the time that both were collected. We show how employing the KLV metadata packet can result in a single video record where picture taking are all in lockstep. Using KLV metadata enables one to record clean video while retaining the ability to place crosshairs and data during playback.

Full textChronic lymphocytic leukemia (CLL) is a malignant hemopathy characterized by the accumulation of apoptosis resistant mature B lymphocytes in peripheral blood and secondary lymphoid organs. In these secondary lymphoid organs, the tumor microenvironment, notably Nurse-like Cells (NLCs), plays a major role in leukemic cells survival and proliferation promotion. In this study, an overexpression of neurotensin receptor NTSR2, a G-protein coupled receptor, was identified. NTSR2 is constitutively activated in circulating leukemic cells and its activation depends on its interaction with tyrosine kinase activity receptor TrkB upon binding of its ligand, BDNF. Activation of NTSR2-TrkB by BDNF induces survival signaling by Src and Akt pathways, and in term anti-apoptotic proteins Bcl-2 and Bcl-XL overexpression. Inhibition of NTSR2 in those cells impacts their viability. In the presence of NLCs, expressions of NTSR2, TrkB, BDNF, and sortilin, a neurotrophin and Trk receptor transport regulator, are enhanced. NLCs produce BDNF, stimulate Src activation, and their protective role on leukemic cells is BDNF-dependent. Taken together, this study highlights a key role of NTSR2-TrkB-BDNF in leukemic cells survival signaling, both in the circulation or in the tumor microenvironment, and might thus constitute a potential new therapeutic target

Full textChronic lymphocytic leukemia (CLL) is characterized by the abnormal accumulation of mature B lymphocytes. Current therapies for CLL rely on using kinase inhibitors targeting B-cell receptor (BCR) pathways, but they are limited by the high level of apoptosis-resistant B-CLL cells, which results in a high frequency of patient relapse. Because current therapies fail to eradicate these apoptosis-resistant cells, it is essential to identify alternative survival pathways as novel targets for anticancer therapies. Overexpression of cell-surface G protein-coupled receptors (GPCRs) drives cell transformation, and thus plays a critical role in malignancies. In this study, we show that neurotensin receptor 2 (NTSR2), a G-protein-coupled receptor, is an essential driver of apoptosis resistance in B-CLL. NTSR2 was highly expressed and constitutively active in B-CLL cells, and its activation depended on its interaction with the tropomyosin-related kinase B receptor (TrkB) and the recruitment of Gi proteins, instead of its interaction with its natural ligand, neurotensin (NTS). The NTSR2-TrkB interaction acted as a conditional oncogenic driver requiring the TrkB ligand BDNF (Brain-Derived Neurotrophic Factor), which is highly expressed in B-CLL cells, unlike its natural ligand NTS. The NTSR2-TrkB interaction activates survival signaling pathways, including the Src and AKT kinase pathways, as well as expression of the anti-apoptotic proteins Bcl-2 (B-cell lymphoma-2) and Bcl-xL (B-cell lymphoma-extra large). TrkB failed to protect B-CLL cells from a drastic decrease in viability via typical apoptotic cell death when NTSR2 was down-regulated. Taken together, the results suggest that the NTSR2-TrKB interaction and the sustained activation of signaling pathways reliant on this interaction constitute an essential driving force for apoptosis evasion of B-CLL cells. Targeting NTSR2 could represent a promising strategy for treating B-CLL malignancy

Full textTargeted Radionuclide Therapy (TRT) uses radionuclides coupled to carrier molecules (radiopharmaceuticals) capable of targeting receptors overexpressed by tumor cells or their microenvironment. Among different strategies used in TRT, the one based on analogs of radiolabeled peptides, such as [177Lu]Lu-DOTATATE or [177Lu]Lu-PSMA, is in full expansion. These treatments are part of a personalized theranostic approach, where a companion molecule labeled with a positron emitter such as 68Ga is used for PET imaging as a pre-requisite to TRT. This PhD work aimed at exploring the potential offered by the targeting of two neuropeptide receptors, the neurotensin receptor NTS1 and the neuropeptide Y receptor Y1R, known to be overexpressed in several cancers.Regarding the targeting of NTS1, the design of peptide radiopharmaceuticals is challenging due to the short plasmatic half-life of the endogenous neurotensin peptide. In the first part of this work, we explored the impact of unnatural amino acids in neurotensin analogs, radiolabelled with 68Ga. Insertion of a silylated alanine in the N-terminal position stabilized the [68Ga]Ga-JMV6659 peptide while maintaining a good affinity and selectivity profile. In vivo, the analog [68Ga]Ga-JMV6659 showed excellent tumor uptake in nude mice carrying an HT-29 xenograft making this compound a good candidate for PET imaging. Work is ongoing to investigate the therapeutic potential of this compoundFurther increasing the dose deposited in tumor cells would enhance the efficacy of TRT. An emerging strategy, little explored today, consists of the nuclear targeting with therapeutic short-range emitters such as terbium-161, rich in Auger-electrons. With this in mind, we explored the feasibility of nuclear addressing via the Y1R receptor of an agonist analogue of a modified neuropeptide Y. The modified analog uses a cleavage sequence that is handled by endosomal cathepsin B, subsequently releasing a nuclear localization sequence (NLS) supported by cytoplasmic importins providing transport to the nucleus of tumor cells. Thus, the compound Pb12 was studied after radiolabelling with 111In, as a substitute for 161Tb. Results obtained on MCF7 breast cancer cells expressing Y1R showed that this multi-step nuclear addressing technology works, opening promising prospects with other short-range therapeutic emitters such as alpha emitters

Background: In this day and age, the media is omnipresent and a part of our daily life. The phenomenon of media convergence can be traced back to the birth of the internet, and today all of the major newspapers have a website of their own. As a result of multichannel publishing, consumers have a wide range of tv channels to choose from, but there is still uncertainty as to whether more channels automatically equates to more and better news. There is no doubt about the fact that multi channel publishing is a necessary survival skill for media conglomerates today, but effects such as news recycling and the concentration of ownership are in conflict with the consumers’ availability demands.
Purpose: The purpose of this thesis is to, by means of a case study, describe how NT’s multi channel publishing is an expression of media convergence, as well as to study how the news are affected by multi channel publishing.
Research procedure: This study is based on three different research methods, observations, text analyses and qualitative interviews. Interviews were conducted with three employees at NT, which was supplemented by an observational study of the work carried out by the NT employees. Moreover, a text analysis of three news articles that were published in multiple channels at NT was carried out.
Results: The case study shows that NT is more converged in theory than in practice, and that NT is currently in a development stage of convergence. The results also clarify that NT has an open work environment and advocates that employees work in different publishing channels. It is also possible to find traces of cross promotion at NT. The study indicates that the desire to publish the news in multiple channels gives rise to news recycling and as well as news homogenization, which may impair the quality of the news.

Resumo: O gênero Engystomops apresenta ampla distribuição geográfica e constitui um interessante grupo de anuros para estudos cariotípicos. As populações de Engystomops encontradas na Amazônia têm sua identificação taxonômica ainda controversa. Análises genéticas e citogenéticas apoiam hipóteses que sugerem a existência de um complexo de espécies crípticas e especiação incipiente. Muitas vezes a variação citogenética observada entre diferentes populações estudadas dificultou o reconhecimento de homeologias cromossômicas entre os cariótipos. Uma caracterização cromossômica mais detalhada poderia auxiliar no possível reconhecimento de homeologias cromossômicas e, dessa forma, contribuir para o estudo dos processos envolvidos na divergência desses anuros. Já que o gene do DNAr 5S tem sido importante marcador genético e citogenético para estudos evolutivos e para a identificação e comparação de espécies em diversos grupos, no presente trabalho o DNAr 5S de Engystomops freibergi e de exemplares de Engystomops petersi de duas localidades Equatorianas (Puyo e Yasuní) foi estudado. Em todos os casos, dois tipos de DNAr 5S, facilmente diferenciados pelo tamanho e composição da sequência do seu espaçador não transcrito, foram isolados. A provável região promotora do gene do RNAr 5S (ICR) foi localizada nos dois tipos de sequências de DNAr 5S e a presença de possíveis sequências regulatórias adicionais foi discutida. No cariótipo de E. freibergi, sonda contendo a unidade repetitiva do DNAr 5S tipo I hibridou na região pericentromérica do braço curto dos cromossomos do par 3, e o DNAr 5S tipo II foi mapeado na região distal do braço longo dos cromossomos do par 6. A sonda formada somente pela região de NTS do DNAr 5S tipo I claramente detectou a região pericentromérica de 3p nos cariótipos de E. freibergi e E. petersi (Puyo) e de 5p no cariótipo de E. petersi (Yasuní), porém nenhum sinal distal ou intersticial foi observado. A sonda formada pela região de NTS do DNAr 5S tipo II detectou apenas a região distal de 6q nos três cariótipos estudados, corroborando a distribuição diferencial dos dois tipos de DNAr 5S nesses cariótipos. Tais sítios de DNAr 5S constituem novos marcadores cromossômicos, os quais permitem sugerir a homeologia entre o cromossomo 6 dos cariótipos de E. freibergi e de E. petersi, e entre o cromossomo 5 do cariótipo de E. petersi de Yasuní e o cromossomo 3 dos cariótipos de E freibergi e de E. petersi de Puyo. Já que os dois tipos de DNAr 5S encontrados em Engystomops são relacionados àqueles de Physalaemus tanto quanto à composição nucleotídica quanto à localização cromossômica, é ainda possível inferir que a origem desses dois tipos de sequências tenha antecedido a divergência evolutiva desses gêneros
Abstract: The Engystomops genus is widely distributed geographically and constitutes an interesting group for karyotypic studies. The taxonomic identifications of Amazonian populations of Engystomops is still controversial. Genetic and cytogenetic analyses suggest the existence of a complex of cryptic species and incipient speciation. The cytogenetic variations found among some populations prevent the recognition of chromosomal homeologies between the described karyotypes. A more detailed chromosomal characterization could help in the recognition of chromosome homeologies and, therefore, could contribute to the study of the processes involved in the divergence of these anurans. Since the 5S rDNA gene has been an important genetic and cytogenetic marker for evolutionary studies and even for the identification and comparison of species in diverse groups, in the present work the 5S rDNA of Engystomops freibergi and exemplars of Engystomops petersi from two Ecuadorian locations (Puyo and Yasuní) was studied. In all cases, two types of 5S rDNA, easily differed by size and compositon of their non-transcribed spacer, were isolated. A putative promoting region of the 5S rRNA gene (ICR) was recognized in the two types of 5S rDNA sequences and the presence of possible additional regulatory sequences was discussed. In the E. freibergi karyotypes, the entire type I 5S rDNA repeating unit hybridized to the pericentromeric region of 3p, whereas the entire type II 5S rDNA repeating unit mapped to the distal region of 6q, suggesting a differential localization of these sequences. The type I NTS probe clearly detected the 3p pericentromeric region in the karyotypes of E. freibergi and E. petersi from Puyo and the 5p pericentromeric region in the karyotype of E. petersi from Yasuní, but no distal or interstitial signals were observed. Interestingly, this probe also detected many centromeric regions in the three karyotypes, suggesting the presence of a satellite DNA family derived from 5S rDNA. The type II NTS probe detected only distal 6q regions in the three karyotypes, corroborating the differential distribution of the two types of 5S rDNA. Because the 5S rDNA types found in Engystomops are related to those of Physalaemus with respect to their nucleotide sequences and chromosomal locations, their origin likely preceded the evolutionary divergence of these genera. In addition, our data indicated homeology between chromosome 5 in E. petersi from Yasuní and chromosomes 3 in E. freibergi and E. petersi from Puyo. In addition, the chromosomal location of the type II 5S rDNA corroborates the hypothesis that the chromosomes 6 of E. petersi and E. freibergi are homeologous despite the great differences observed between the karyotypes of the Yasuní specimens and the others

Full textAThe chemistry of N-acyliminiums ions is one important area of research in modern organic chemistry. Lewis superacid catalysis has contributed in the improvement of many synthetic processes. We have developed some direct α-amidoalkylation reactions starting from unmodified ketones and aldehydes. The reactions were catalyzed by the Lewis superacid tin IV triflimidate, Sn(NTf2)4, at a very low catalyst loading(0.5-2 mol%), providing access to a set of very interesting carbonylated structures in good yields. The influence of different parameters governing the reactivity and the scope of the reaction were studied. Thereafter, the synthetic potential of the obtained products was evaluated. Two intramolecular approaches from suitably functionalized alkylated products were tested using gold and metathesis catalysts, allowed access to some polycyclic structures

Full textThis thesis is dedicated to the study of groups and fields whose definable sets do not admit certain combinatorial patterns. Given a group G, one particular problem we are interested in is to find definable envelopes for arbitrary abelian, nilpotent or solvable subgroups of G which admit the same algebraic properties. Such evelopes exists if G is stable and even if G is merely dependent but sufficiently saturated, with the additional hypothesis of normality in the solvable case. In groups with a simple theory, one obtains definable envelopes up to finite index.We introduce the notion of an almost centralizer and establish some of its basic properties. This enables us to extend the aforementioned results to Mc~ groups, i. e. groups in which any definable section satisfies a chain condition on centralizers up to finite index. These include any definable group in a rosy and in particular in a simple theory. Furthermore, inspired from the proof in dependent theories as well as using techniques developed for almost centralizers in this thesis, we are able to find definable envelopes up to finite index for abelian, nilpotent and normal solvable subgroups of any enough saturated NTP2 group. Moreover, using envelopes for nilpotent subgroups of Mc~ groups and the chain condition on centralizer up to finite index, we show additionally that the Fitting subgroup of any Mc~ group is nilpotent and that its almost Fitting subgroup is virtually solvable.The second part of this thesis focuses on the study of n-dependent fields. We prove that any n-dependent field is Artin-Schreier closed and that non separably closed PAC fields are not n-dependent for any natural number n

Full textThe Comoros is an archipelago of four islands in the Indian Ocean at the northern entrance of the Mozambique Channel. It was a French colony until 1975 when it gained its independence. The mix of applicable legal systems seems to be the origin of the legal and institutional stagnation gripping the country since its independence. Indeed, both Muslim and French legal systems have been superimposed local traditional law. Initially, these systems were merged into local practices, but will thereafter be considered that these customs can not coexist with the institutions of a modern state. But recent history shows that ignorance or contempt for tradition and its system of social regulation undermines social reforms introduced by the prosecution because it has no support from society. Comorian land law is also facing this legal pluralism. Itis the origin of land conflicts that plague society. The villagers, families and even the state are at the center of these conflicts. The judiciary is supposed to adjust paralyzed by its complex organization, its lack of resources, but also by widespread corruption. The lack of a unified positive law contributes to legal uncertainty. Several failed attempts to stabilize since the colonial era, but recent reforms, such as the OHADA legislation and that relating to the Judicature (Act No. 05-016 / AU), have a sign of hope
