Darbe yra apžvelgiamos NURBS paviršių atvaizdavimo realizavimo problemos mobiliesiems įrenginiams. Pagrindinis darbo tikslas yra įsitikinti tokios realizacijos galimumu, bei surasti kiek įmanoma efektyvesnį būdą, išsprendžiantį mobiliųjų platformų realizacijų problemas. Dar vienas tikslas, sukurti prototipinį sprendimą, padengiantį NURBS standartą. Analizei pasirinkta M3G mobiliosios grafikos standartas, kuris šiuo metu yra labiausiai paplitęs. Realizacijų palyginimui buvo pasirinkti du algoritmai: Oslo ir tiesioginis. Atlikus veikimo analizę nustatyta, jog tiesioginis algoritmas yra efektyvesnis, kurio pagrindu buvo išbaigtas NURBS standartas, realizuojant Trimmed NURBS modelius.
In the thesis it is analyzed the problems related with NURBS rendering implementation for mobile devices. The main goal of the thesis is to make find out if such implementation is possible at all and to find most effective way to implement it. One more goal is to create a prototype implementation for NURBS standard including Trimmed NURBS. M3G mobile graphics platform was chosen for analysis, which is mostly adopted across the vendors nowadays. There was chosen two rendering algorithms for analysis purposes: Oslo and directional. During analysis it was discovered, that Directional algorithm is more effective than Oslo, and it was adapted to prototype solution – Trimmed NURBS.

Abstract:
Radiostereometric analysis (RSA) is a method used for determining migration and wear of orthopaedic implants through measurements performed in radiographs. There are two approaches of RSA: marker-based and model-based (MBRSA). Marker-based RSA is the conventional and most accurate method. However, it requires markers to be inserted or attached to the implants. MBRSA avoids using markers by employing 3D geometric models of the implants. State-of-the-art methods for model-based RSA use either elementary geometric shapes (EGS) or triangulated surface (TS) models. EGS models are smooth but not generic. They can only model implants that have simple canonical shapes. In contrast, TS models are generic but not smooth. They can model both basic and free-form shapes but not as smooth as EGS models. The degree of smoothness depends on the number of triangles used. In this thesis, we introduce Non-uniform Rational B-Splines (NURBS) models that are both generic and smooth. Two algorithms were developed: 1) A fitting algorithm that fits multi-patch NURBS models to unorganized point clouds representing the surface of the implants. 2) An RSA algorithm that fits the NURBS models to points measured in radiographs. To evaluate the algorithms, NURBS models of a femoral head and an acetabular component were generated by the fitting algorithm. The femoral head penetration into the acetabular component was computed by the NURBS-based RSA algorithm using clinical double examinations for a single hip replacement patient. The precision of the NURBS-based RSA algorithm for computing the femoral head penetrationis comparable to the reported precision of a state-of-the-art EGS based RSA algorithm for solving the same problem. This result suggests that NURBS models can be a good alternative to the current models used in MBRSA.

O método isogeométrico foi introduzido em 2005 por Hughes [1], quando foi colocado como um novo método para a análise de problemas descritos por Equações Diferenciais Parciais (EDP). O método tem muitas características em comum com o FEM [2] e é geometricamente baseado em NURBS [3], teoria bastante utilizada em Computação Gráfica para modelagem com excelente precisão de objetos tais como curvas e superfícies. Apesar da ideia do método ser simples a prática é complicada. Não é automática a transição da representação no contexto de CAGD (Computer Aided Geometric Design) para o de FEM. Vuong fez uma abordagem nesse sentido para o caso bidimensional [4]. Representar a resolução de EDP por base de funções NURBS é um desafio de interação entre a análise de elementos finitos e a tecnologia geométrica computacional, que antes trabalhavam com os mesmos objetos mas de forma independente. Os principais resultados mais recentes, sob uma perspectiva da análise numérica [5], e também com perspectiva de aplicações [6], mostram que essa interação tem impulsionado o desenvolvimento do método Isogeométrico. Nesta dissertação, a aluna irá desenvolver a teoria do método Isogométrico de maneira matematicamente consistente e irá obter resultados computacionalmente robustos com aplicações de um algoritmo também desenvolvido pela aluna em problemas clássicos que envolvem EDP.

Diploma thesis deals with image interpolation. The aim of this work is to study theoretically and then describe the nature of the various methods of image interpolation and some of them implemented in the program MATLAB. The introductory part of this work theoretically closer to important terms that are closely related to this topic of digital image processing sufficient to understand the principle. In the following of the thesis will be discussed all of today's commonly used method of image interpolation. Will hear all about the method of image interpolation using nearest neightbor interpolation and image help of polynimals such as (bi)linear, (bi)quadratic and (bi)kubic method. Then work theoretically analyzes the theory of individual species curves and splines. More specifically, coming to their most frequently used variants of B-spline curves and ther generalizations called NURBS, with addressing the problem of interpolating these curves. The final chapter consists of the results achieved in the program MATLAB.

O objetivo primordial desse trabalho está concentrado no estudo de Curvas NURBS (B-spline Racional N˜ao-Uniforme). A literatura em português sobre NURBS é escassa, pouco difundida e os textos e artigos existentes tendem a ser rigorosos, longos e teóricos. Assim, o presente estudo está direcionado para os conceitos matemáticos de NURBS, para o qual foi utilizado uma ferramenta chamada DesignMentor com a finalidade de testar os algoritmos desses conceitos. NURBS são funções paramétricas que podem representar qualquer tipo de curva. NURBS são usadas em computação gráfica na indústria de CAD/CAM e estão sendo consideradas um padrão para criar e representar objetos complexos (indústria automobilística, aviação e embarcação). As ferramentas de criação gráfica mais sofisticadas provêem uma interface para usar NURBS, que são flexíveis suficiente para projetar uma grande variedade de formas. Hoje é possível verificar o uso expandido de NURBS, modelando objetos para as artes visuais, arte e escultura; também estão sendo usados para modelar cenas para aplicações de realidade virtual. NURBS trabalha bem em modelagem 3D, permitindo facilidade para manipular e controlar vértices, controlar curvatura e suavidade de contornos. NURBS provêm uma base matemática, unificada para representar formas analíticas e livres além de manter exatidão e independência de resolução matemática.

Aquesta tesi proposa una millora del clàssic mètode dels elements finits (finite element method, FEM) per a un tractament eficient de dominis amb contorns corbs: el denominat NURBS-enhanced finite element method (NEFEM). Aquesta millora permet descriure de manera exacta la geometría mitjançant la seva representació del contorn CAD amb non-uniform rational B-splines (NURBS), mentre que la solució s'aproxima amb la interpolació polinòmica estàndard. Per tant, en la major part del domini, la interpolació i la integració numèrica són estàndard, retenint les propietats de convergència clàssiques del FEM i facilitant l'acoblament amb els elements interiors. Només es requereixen estratègies específiques per realitzar la interpolació i la integració numèrica en elements afectats per la descripció del contorn mitjançant NURBS.

La implementació i aplicació de NEFEM a problemes que requereixen una descripció acurada del contorn són, també, objectius prioritaris d'aquesta tesi. Per exemple, la solució numèrica de les equacions de Maxwell és molt sensible a la descripció geomètrica. Es presenta l'aplicació de NEFEM a problemes d'scattering d'ones electromagnètiques amb una formulació de Galerkin discontinu. S'investiga l'habilitat de NEFEM per obtenir solucions precises amb malles grolleres i aproximacions d'alt ordre, i s'exploren les possibilitats de les anomenades malles NEFEM, amb elements que contenen singularitats dintre d'una cara o aresta d'un element. Utilitzant NEFEM, la mida de la malla no està controlada per la complexitat de la geometria. Això implica una dràstica diferència en la mida dels elements i, per tant, suposa un gran estalvi tant des del punt de vista de requeriments de memòria com de cost computacional. Per tant, NEFEM és una eina poderosa per la simulació de problemes tridimensionals a gran escala amb geometries complexes. D'altra banda, la simulació de problemes d'scattering d'ones electromagnètiques requereix mecanismes per aconseguir una absorció eficient de les ones scattered. En aquesta tesi es discuteixen, optimitzen i comparen dues tècniques en el context de mètodes de Galerkin discontinu amb aproximacions d'alt ordre.

La resolució numèrica de les equacions d'Euler de la dinàmica de gasos és també molt sensible a la representació geomètrica. Quan es considera una formulació de Galerkin discontinu i elements isoparamètrics lineals, una producció espúria d'entropia pot evitar la convergència cap a la solució correcta. Amb NEFEM, l'acurada imposició de la condició de contorn en contorns impenetrables proporciona resultats precisos inclús amb una aproximació lineal de la solució. A més, la representació exacta del contorn permet una imposició adequada de les condicions de contorn amb malles grolleres i graus d'interpolació alts. Una propietat atractiva de la implementació proposada és que moltes de les rutines usuals en un codi d'elements finits poden ser aprofitades, per exemple rutines per realitzar el càlcul de les matrius elementals, assemblatge, etc. Només és necessari implementar noves rutines per calcular les quadratures numèriques en elements corbs i emmagatzemar el valor de les funciones de forma en els punts d'integració. S'han proposat vàries tècniques d'elements finits corbs a la literatura. En aquesta tesi, es compara NEFEM amb altres tècniques populars d'elements finits corbs (isoparamètics, cartesians i p-FEM), des de tres punts de vista diferents: aspectes teòrics, implementació i eficiència numèrica. En els exemples numèrics, NEFEM és, com a mínim, un ordre de magnitud més precís comparat amb altres tècniques. A més, per una precisió desitjada NEFEM és també més eficient: necessita un 50% dels graus de llibertat que fan servir els elements isoparamètrics o p-FEM per aconseguir la mateixa precisió. Per tant, l'ús de NEFEM és altament recomanable en presència de contorns corbs i/o quan el contorn té detalls geomètrics complexes.
This thesis proposes an improvement of the classical finite element method (FEM) for an efficient treatment of curved boundaries: the NURBSenhanced FEM (NEFEM). It is able to exactly represent the geometry by means of the usual CAD boundary representation with non-uniform rational Bsplines (NURBS), while the solution is approximated with a standard piecewise polynomial interpolation. Therefore, in the vast majority of the domain, interpolation and numerical integration are standard, preserving the classical finite element (FE) convergence properties, and allowing a seamless coupling with standard FEs on the domain interior. Specifically designed polynomial interpolation and numerical integration are designed only for those elements affected by the NURBS boundary representation.

The implementation and application of NEFEM to problems demanding an accurate boundary representation are also primary goals of this thesis. For instance, the numerical solution of Maxwell's equations is highly sensitive to geometry description. The application of NEFEM to electromagnetic scattering problems using a discontinuous Galerkin formulation is presented. The ability of NEFEM to compute an accurate solution with coarse meshes and high-order approximations is investigated, and the possibilities of NEFEM meshes, with elements containing edge or corner singularities, are explored. With NEFEM, the mesh size is no longer subsidiary to geometry complexity, and depends only on the accuracy requirements on the solution, whereas standard FEs require mesh refinement to properly capture the geometry. This implies a drastic difference in mesh size that results in drastic memory savings, and also important savings in computational cost. Thus, NEFEM is a powerful tool for large-scale scattering simulations with complex geometries in three dimensions. Another key issue in the numerical solution of electromagnetic scattering problems is using a mechanism to perform the absorption of outgoing waves. Two perfectly matched layers are discussed, optimized and compared in a high-order discontinuous Galerkin framework.

The numerical solution of Euler equations of gas dynamics is also very sensitive to geometry description. Using a discontinuous Galerkin formulation and linear isoparametric elements, a spurious entropy production may prevent convergence to the correct solution. With NEFEM, the exact imposition of the solid wall boundary condition provides accurate results even with a linear approximation of the solution. Furthermore, the exact boundary representation allows using coarse meshes, but ensuring the proper implementation of the solid wall boundary condition. An attractive feature of the proposed implementation is that the usual routines of a standard FE code can be directly used, namely routines for the computation of elemental matrices and vectors, assembly, etc. It is only necessary to implement new routines for the computation of numerical quadratures in curved elements and to store the value of shape functions at integration points.

Several curved FE techniques have been proposed in the literature. In this thesis, NEFEM is compared with some popular curved FE techniques (namely isoparametric FEs, cartesian FEs and p-FEM), from three different perspectives: theoretical aspects, implementation and performance. In every example shown, NEFEM is at least one order of magnitude more accurate compared to other techniques. Moreover, for a desired accuracy NEFEM is also computationally more efficient. In some examples, NEFEM needs only 50% of the number of degrees of freedom required by isoparametric FEs or p-FEM. Thus, the use of NEFEM is strongly recommended in the presence of curved boundaries and/or when the boundary of the domain has complex geometric details.

Vor dem Hintergrund der Ausspracheabweichungen, die chinesische Muttersprachler beim Erwerb der deutschen Aussprache produzieren bzw. aufweisen, widmet sich die vorliegende Arbeit einer intensiven kontrastiven phonetisch-phonologischen Analyse des Deutschen (Standardbinnendeutsch) und des Chinesischen (Putonghua) auf der segmentalphonetischen und -phonologischen, der phonotaktischen und der suprasegmentalen Ebene, um hierauf aufbauend internetbasierte Lehr- und Lernmaterialien zur deutschen Aussprache für chinesische Muttersprachler zu entwickeln. Exemplarisch für diese Konzeption stehen zwei zweieinhalb- bis dreiminütige selbst entwickelte und professionell ausgearbeitete Lernvideos (zu den deutschen Kurz- und Langvokalen sowie zu Konsonantenhäufungen).
Considering pronunciation deviations which are produced by Chinese native speakers in the learning of German pronunciation, this dissertation conducts an intensive contrastive phonetic-phonological analysis of German (Standard internal German) and Chinese (Putonghua) on the segmental, the phonotactic and the suprasegmental levels, in order to further develop internet-based teaching and learning materials for German pronunciation targeting Chinese native speakers. Two educational videos about German pronunciation for Chinese native speakers based on this contrast are developed as a result of this analysis, with one focusing on German short and long vowels and the other on consonant clusters.

Viene sviluppata in XCSurf, un pacchetto di XCModel, una struttura dati chiamata B-Rep il cui scopo è quello di poter accogliere sia geometrie mesh che nurbs. La struttura B-Rep è stata progettata nel lavoro di tesi di F.Pelosi a seguito del riscontro di diverse analogie fra la struttura winged-edge (per mesh) e la struttura B-Rep (per nurbs). In questa tesi viene sviluppata ed integrata ulteriormente. Il punto di arrivo è la possibilità di attaccare due modelli qualsiasi (Nurbs + Nurbs, Mesh + Nurbs, Mesh + Mesh), deformando opportunamente le parti da attaccare, ma mantenendo tutte le informazioni in un'unica struttura.

Abstract:
Le complexe de remodelage des nucléosomes et de désacétylation des histones (NuRD) est l'un des principaux régulateurs épigénétiques du génome. Il contribue à la formation et au maintien de l'hétérochromatine, une structure très dense d'ADN et de protéines réprimant la transcription. La NuRD joue un rôle central dans les processus biologiques pertinents tels que la régulation de la pluripotence ou la tumorigenèse. Malgré cela, sa structure et son mécanisme d'action restent largement inconnus. Cela est dû en grande partie à l'hétérogénéité inhérente de composition et de conformation de NuRD. Dans ce travail de doctorat, nous avons essayé de surmonter ces défis en utilisant une approche multidisciplinaire. Nous avons combiné la purification par affinité de complexes et de sous-unités NuRD endogènes et recombinants, la réticulation, la cryo-microscopie électronique et la spectrométrie de masse. Grâce à cela, nous avons pu identifier un sous-complexe d'histone désacétylase stable à activité indépendante (PMMR) et résoudre sa structure à 16,6 Å. Nous avons également obtenu des structures à faible résolution de plusieurs sous-complexes plus petits. The nucleosome remodeling and histone deacetylase (NuRD) complex is one of the main epigenetic regulators of the genome. It contributes to the formation and maintenance of the heterochromatin, a tightly packed structure of DNA and proteins that represses transcription. NuRD plays a central role in relevant biological processes such as pluripotency regulation or tumorigenesis. Despite that, its structure and action mechanism remain unknown. This is largely due to the inherent compositional and conformational hetesrogeneity of NuRD. In this PhD work we tried to overcome these challenges using a multidisciplinary approach. We combined affinity purification of endogenous and recombinant NuRD complexes and subunits, cross-linking, electron cryo-microscopy and mass spectrometry. Thanks to that we were able to identify a stable histone deacetylase subcomplex with independent activity (PMMR) and solve its structure at 16.6 Å. We also obtained low resolution structures of several smaller subcomplexes. Studying these subcomplexes allowed us to propose a Holo-NuRD assembly pathway, leading to the publication of a scientific paper.Key words: NuRD, Epigenetic regulation, Chromatin remodeling, Histone deacetylase, Cryo-EM

Ces travaux de thèse sont dédiés à la mesure de formes 3D, de champs cinématiques 3D surfaciques et à l'identification de paramètres mécaniques à partir de mesures par corrélation d'images. Cette méthode optique se base sur l'utilisation de caméras comme support de la mesure. Pour cette raison, une étude des différents modèles de caméra classiquement utilisés est effectuée, puis le passage d'un formalisme continu pour la description des images à une approche discrète utilisant la notion de pixel est décrit. Une attention particulière est portée aux distorsions optiques et une approche utilisant la corrélation d'images pour les évaluer est développée. Une nouvelle méthode de mesure de forme 3D et de champs de déplacements 3D surfaciques par stéréo-corrélation d'images est introduite. Elle utilise un modèle numérique de l'objet observé comme support pour la corrélation. Cette méthode mène à une formulation globale de la stéréo-corrélation. Elle peut être écrite dans le cas général et particularisée à des surfaces NURBS (non-uniform rational B-splines). Le champ de déplacement est également exprimé dans un formalisme NURBS complètement cohérent avec le modèle géométrique utilisé pour définir la forme observée. Des mesures sont validées en utilisant des mouvements imposés à un carreau de Bézier usiné. La faisabilité de la technique dans plusieurs cas industriels est également étudiée avec, par exemple, la mesure des déplacements d'une pièce composite de train d'atterrissage chargée mécaniquement. Enfin, à partir de cette formulation isogéométrique de la mesure de champs, une étude de la caractérisation de propriétés élastiques est effectuée. Deux méthodes inspirées d'approches existantes sont ainsi proposées, utilisant des mesures de champs et des calculs numériques dans un formalisme isogéométrique pour identifier des paramètres de loi de comportement élastique isotrope sur un cas test numérique et pour un essai de traction uniaxiale
This thesis is dedicated to measurement of 3D shapes, 3D kinematic fields on surfaces and identification of mechanical properties from digital image correlation measurements. This optical method uses cameras as measurement tools. For this reason, a study of the classical camera models used is performed and the description of the digitalization of an image from a continuous to a discrete formalism using the pixel is described. A specific work is dedicated to optical distortions and a method based on digital image correlation to evaluate these distortions is developped.A new method for 3D shapes and 3D displacement fields on surfaces using stereo-correlation is introduced. A numerical description of the observed object is used as a support to perform the correlation. This method lead to a global approach to stereo-correlation. It can be rewrite in a generic case or in particular to be applied to NURBS (Non-Uniform Rational B-Splines) surfaces. The displacement fields is therefore expressed in a NURBS formalism which is completely consistent with the geometrical model used to described the observed shape. Measurements are validated using prescribed motions on a Bezier patch. The feasibility of such a technique in several industrial cases is then studied with for example the measurement of the displacement of a composite part of a landing gear under mechanical loading.Finally, from this isogeometric formulation of full-field measurement, a study of the identification of elastic properties is performed. Two methods inspired from existing approaches are proposed, using full-field measurement and numerical simulations in a common isogeometric formalism to identify parameters of an isotropic linear elastic constitutive law on a both a numerical test case and a uniaxial tensile test

The production of cell diversity in multicellular organisms depends on asymmetric cell divisions, in which the mother cell segregates cell fate determinants asymmetrically into only one of the two daughter cells, thereby instructing different fates. In the developing retina, the cell fate determinant Numb, a Notch signalling inhibitor, is asymmetrically localized in mitotic retinal progenitor cells (RPCs) and depending on the orientation of cell division, inherited symmetrically or asymmetrically by the daughter cells. Previous studies have suggested correlation between asymmetric inheritance of Numb and asymmetry in cell fate choice, but the precise role of Numb in vertebrate neural cell fate decisions remains unclear. Numb function was mainly studied in the developing mouse cortex, where it seems to promote the progenitor fate. However, most studies addressed the role of Numb during early stages of neurogenesis and its specific role at later stages remains unknown. Here we used the mouse retina as a model system to study Numb function at different stages of neural development. We first used the Cre-loxP system to conditionally inactivate numb and its homolog numblike in vivo in RPCs from E 9.5. Analysis of these conditional double knockout (cdKO) mice revealed laminar disorganization and defects in neuronal and glial production. We further show that this phenotype is due to depletion in the progenitor pool at early stages of retinal development. Next, to determine whether the role of Numb homologs changes during development, we used retroviral vectors expressing Cre-recombinase and a reporter gene to infect Numb flox/flox; Numblike flox/flox mouse retinas at early and late stages of retinal development. Analysis of cell composition and size of the resulting knockout clones showed that early inactivation of numb and numblike decreases RPCs cell cycle progression but did not change the mode of cell divi
Au cours du développement des organismes multicellulaires, la diversité cellulaire est en partie issue de divisions asymétriques, au cours desquelles une cellule mère produit deux cellules filles différentes à la naissance. Cette différence d'identité résulte souvent de la répartition inégale de déterminants cellulaires durant la mitose. Ce processus de répartition inégale requiert que la cellule mère soit polarisée avant sa division. En réponse à cette polarisation, les déterminants cellulaires sont accumulés à un pôle de la cellule et le fuseau mitotique s'oriente le long d'un axe passant par ce pôle, permettant ainsi la distribution inégale de ces déterminants entre les deux cellules filles. Abstract:
The primary focus of this thesis is to determine the best methods for controlling the interpolation of NURBS curves and surfaces. The various factors that affect the quality of the interpolant are described, and existing methods for controlling them are reviewed. Improved methods are presented for calculating the parameter values, derivative magnitudes, data point spacing and twist vectors, with the aim of producing high quality interpolants with minimal data requirements. A new technique for obtaining the parameter values and derivative magnitudes is evaluated, which constructs a C\(^1\) cubic spline with orthogonal first and second derivatives at specified parametric locations. When this data is used to create a C\(^2\) spline, the resulting interpolant is superior to those constructed using existing parameterisation and derivative magnitude estimation methods. Consideration is given to the spacing of data points, which has a significant impact on the quality of the interpolant. Existing methods are shown to produce poor results with curves that are not circles. Three new methods are proposed that significantly reduce the positional error between the interpolant and original geometry. For constrained surface interpolation, twist vectors must be estimated. A method is proposed that builds on the Adini method, and is shown to have improved error characteristics. In numerical tests, the new method consistently outperforms Adini. Interpolated surfaces are often required to join together smoothly along their boundaries. The constraints for joining surfaces with parametric and geometric continuity are discussed, and the problem of joining \(N\) patches to form an \(N\)-sided region is considered. It is shown that regions with odd \(N\) can be joined with G\(^1\) continuity, but those with even \(N\) or requiring G\(^2\) continuity can only be obtained for specific geometries.

Deformable image registration (DIR) is an essential tool in medical image processing. It provides a means to combine image datasets, allowing for intra-subject, inter-subject, multi-modality, and multi-instance analysis, as well as motion detection and compensation. One of the most popular DIR algorithms models the displacement vector field (DVF) as B-splines, a sum of piecewise polynomials with coefficients that enable local shape control. B-splines have many advantageous properties in the context of DIR, but they often struggle to adequately model steep local gradients and discontinuities. This dissertation addresses that limitation by proposing the replacement of conventional B-splines with a generalized formulation known as a Non-Uniform Rational B-Splines (NURBS). Beginning with the 1D fitting, heuristic rules are developed to determine the values of the additional free parameters introduced by NURBS. These rules are subsequently modified and extended to the 2D and 3D fitting of anonymized and publicly available patient DVFs. Based on the lessons learned from these increasingly complex test cases, a 2D DIR scheme is developed and tested on slices from a thoracic computed tomography (CT) scan. Finally, an automatic, non-uniform scheme is presented, and its registration performance is compared to the conventional uniform methods.

This report treats the implementation of collision detection algorithms for Boundary representations (BREPs) consisting of connected trimmed surfaces, mainly Non Uniform Rational Basis Spline (NURBS) surfaces. Using the OpenNurbs software package, complicated geometries created in CAD program Space Claim were imported to the physics engine AgX, where dynamic simulations were carried out. Collision detection algorithms for the geometry pairs BREP-line, BREP-plane and BREP-sphere have been developed and investigated. In the case of BREP-sphere collision detection, experiments have been carried out which show that BREP-shape representation exceeds trimesh-shape representation both in computational performance and in collision accuracy. The conclusion is that BREP representation has the potential to replace trimesh representation for some complex geometries with higher computational performance and more accurate simulations as a result.

Le chapitre deux donne un nouveau theoreme sur les courbes nurbs, qui montre que le resultat du reparametrage d'une courbe nurbs par une fonction homogrpahique est encore une courbe nurbs. Le troisieme chapitre s'interesse au probleme de l'interpolation ou de l'approximation de donnees par une courbe nurbs, et plus particulierement du parametrage a priori des donnees et de l'utilisation des poids rationnels. Le chapitre quatre decrit une methode de manipulation directe de courbes et surfaces nurbs, grace a l'utilisation de matrices pseudo-inverses. Le chapitre cinq contient les definitions des lignes de decoupe et des surfaces connectees, qui sont necessaires a la comprehension de certains aspects du dernier chapitre, lequel apprehende le probleme du calcul des courbes d'intersection entre deux surfaces nurbs. CRMP5 appartient à la famille des Collapsin Response Mediator Protein. Ces protéines sont très exprimées dans le cerveau en développement et les zones de neurogénèse chez l'adulte. Dans un contexte tumoral, l'expression des messagers de CRMP5 émergent dans un cluster de gènes associés à la plus faible survie des 20 patients suivis, et à la prolifération (Liang et al., 2005). Nous avons confirmé ces résultats dans une série rétrospective de 183 GBM où la forte expression protéique de CRMP5 est corrélée à une plus faible survie des patients (7.14 mois vs 10 mois) ; de plus les tumeurs exprimant fortement CRMP5 présentent un index mitotique 2 fois plus important (p = 0.0009) que les tumeurs exprimant faiblement CRMP5. Dans des cultures primaires ou lignées cellulaires de GBM nous montrons que la prolifération des glioblastomes est dépendante de l'expression de CRMP5 et de la voie de signalisation Notch. Des analyses en western blot démontrent que CRMP5 protège les récepteurs Notch de la dégradation lysosomale. Nous avons approfondi ce mécanisme et montré une nouvelle voie de régulation de Notch par CRMP5 qui par une interaction protéique avec Numb, l'inhibiteur de Notch empêche la dégradation du récepteur. Parallèlement, l'analyse en immunohistochimie sur les biopsies de GBM montrent une forte expression Notch et sa cible Hes1 dans les tumeurs exprimant fortement CRMP5. Ces résultats montrent la corrélation entre l'expression de CRMP5 dans les GBM, l'activation de la voie Notch et la faible survie des patients. Le ciblage de l'interaction CRMP5-Numb est une stratégie potentielle pour un traitement ciblé des glioblastomes

Epigenetic regulation of gene expression is critical for lineage determination during development and the stable transmission of cellular identity through cell growth and division. Chromatin modifiers are a varied group of enzymes that regulate patterns of gene expression through the modification of DNA and histones. Epigenetic modifications alter the physical structure of chromatin or recruit effectors to promote or inhibit transcription. In this study, I examined the role of pre-existing histone modifications on the activity of Polycomb and Trithorax chromatin modifying enzymes. Additionally, identification of a novel H3K36me3-reader PWWP2A may constitute a new histone crosstalk pathway between H3K36me3 and histone deacetylation. Polycomb and Trithorax proteins are two important families of transcriptional regulators that promote gene repression and activation respectively. In this study, I find that histone crosstalk between Polycomb complexes PRC1 and PRC2 promote one another's activity while inhibiting Trithorax function. Recombinant nucleosomes carrying the PRC2 modification H3K27me3 stimulates the activity of CBX-PRC1, and nucleosomes with the PRC1 modification H2AK119u1 stimulates the activity of JARID2-PRC2. H2AK119u1 negatively regulates Trithorax activity and inhibits both the H3K4 methyltransferases MLL1/4 and the H3K36 methyltransferase NSD2. Histone crosstalk appears to be an important mechanism in the regulation of the recruitment and activity of chromatin modifiers. I identified a novel H3K36me3 binding protein PWWP2A in an unbiased screen for readers of H3K27me3 and H3K36me3. Biochemical characterisation of PWWP2A in HeLa cells and mouse embryonic stem cells (mESCs) showed that PWWP2A interacts with NuRD subunits MTA1-3, HDAC1/2, and RBBP4/7. Genome-wide mapping of PWWP2A targets in mESCs shows that it is enriched over the gene bodies of highly transcribed genes sharing many gene targets with HDAC1. PWWP2A localisation correlates highly with H3K36me3, and its localisation at gene bodies is dependent on the putative H3K36me3-binding PWWP domain. In budding yeast, H3K36me3-mediated recruitment of the yeast HDAC complex Rpd3S is important for maintaining transcriptional integrity over coding regions. PWWP2A-NuRD may represents an analogous pathway of transcriptional regulation in mammalian organisms.

A pesquisa realizada teve como principal propósito, apresentar uma metodologia para representação de estruturas anatômicas em três dimensões, utilizando um modelo de representação baseado em curvas NURBS, proporcionando a visualização das imagens através de técnicas de Realidade Virtual. Foi desenvolvido posteriormente, um protótipo, contemplando o método aqui apresentado, que tomando como entrada um conjunto de pontos representando um objeto 3D em modelo poligonal, gera a representação do objeto em modelo baseado em curvas (NURBS) e sua representação em linguagem de modelagem em Realidade Virtual (VRML).

Abstract:
Non-uniform rational basis spline (NURBS) is a geometric language which has been commonly used in computer graphics and Computer Aided Design over years, due to its great flexibility and precision for handling modeled shapes. In computer-aided design, NURBS has become part of industry standards. In most CAD programs every solid model is described by NURBS and stored in NURBS data structure. Recently, the geometric basis function of NURBS is directly used in analysis, leading to the birth of a new area of computational mechanics called isogeometric analysis (IGA). Compared with the traditional finite element analysis, IGA eliminates the disconnection between analysis and design at the geometric level, showing superiority on efficiency and accuracy over the traditional finite element method. This thesis focuses on the development of an IGA preprocessor for converting an original CAD model (2D-NURBS model) to an ASG (analysis suited geometry) model. ASG means models with mesh quality suitable for analysis. Core technologies utilized during the development include the well-developed NURBS algorithm of knot insertion and knot removal, and a self-developed technology of constructing and utilizing “multi-patch tree”. The developed IGA preprocessor is utilized to process a body armour model consisting of multiple NURBS patches with overly dense meshes. After the processing, the output model has a relatively reasonable elements distribution; the uniformity of the mesh across presumably contact patch boundaries is rebuilt.

The nucleosome remodeling factor (NURF) is a mutli-protein complex that plays a role in the regulation of gene expression through its ability to remodel nucleosomes. The largest subunit of this complex, Bptf (Bromodomain PHD Finger Transcription Factor) is important for many cellular processes as a transcriptional regulator and improper function results in disease or malignancy. To further understand the genome-wide recruitment of the NURF complex, the interaction partner for the N-terminal PHD finger domain of Bptf was investigated through pull down assays followed by mass spectrometry. It was determined that this domain does not recognize histones; instead it recognizes a nonhistone protein, Thoc4 or Hmgb1. The expression of a cDNA corresponding to Bptf was also tested for expression in mouse ES cells after the addition of two exons found to be missing in the original cDNA. Addition of this sequence did not allow for exogenous Bptf expression in ES cells.

Abstract:
The role of NUMB, a protein important for cellular differentiation and endocytosis in non-placental cells, was investigated in syncytiotrophoblast development and function in the human placenta. The BeWo choriocarcinoma cell line was used as a model for villous cytotrophoblast cells and syncytiotrophoblast to investigate NUMB’s involvement in differentiation and epidermal growth factor receptor (EGFR) endocytosis. NUMB isoforms 1 and 3 were found to be the predominant isoforms and were upregulated following forskolin-induced differentiation. Overexpression of NUMB isoforms 1 and 3 did not mediate differentiation or EGFR signaling. Immunofluorescence analysis revealed that NUMB colocalized with EGFR at perinuclear late endosomes and lysosomes following EGF stimulation. We have demonstrated for the first time that NUMB isoforms 1 and 3 are expressed in BeWo cells, are upregulated in forskolin-differentiated BeWo cells and are involved in ligand-dependent EGFR endocytosis in BeWo cells.

Der Artikel stellt eine musikdidaktische Perspektive dar, die ihren Ausgang bei der Frage nimmt, ob und wie Musik im Musikunterricht erscheint, sofern das Phänomen Musik nur in erfüllter Praxis erscheint. Erfüllte Praxis wird verstanden als eine gelingende Konstellation verschiedener Situationsmerkmale wie Wahrnehmungsinteressen, Themen, Vollzugs-, Interaktions- und Kommunikationsweisen der beteiligten Personen. Nach einer erfahrungsbezogenen Einleitung wird im ersten Abschnitt der Praxisbegriff als Dreh- und Angelpunkt der musikdidaktischen Theorie und auch Videographie dargestellt. Im zweiten und dritten Abschnitt werden ästhetische Praxis und kulturelle Bedeutung als allgemeine Form und inhaltliche Konkretisierung erfüllter Praxis dargestellt. Zweimal vier Kriterien für die Analyse und Gestaltung von erfüllter Praxis im Musikunterricht (=SchulMusik) werden ausgewiesen und mit den gemeinsamen Videos des Symposions und des Buchs veranschaulicht. Über den deskriptiven Kern des Artikels hinaus, der auf normative Setzungen verzichtet, wird in einem Exkurs ein musikdidaktisches Modell „Musikpraxen erfahren und vergleichen“ entworfen
The article constitutes an approach on music education, which takes its starting point from a question: Does music appear in music lessons and how, assuming the phenomenon music appears in fulfilling praxis only. Fulfilling praxis is defined as a meaningful constellation of different characteristics, such as the attitude of perception, the topic, the ways of action, interaction and communication of the participating people. The introduction leads from observations in school settings to the keypoints of music didactics, focusing on three different scientific parts of the related issues. Part one portrays “praxis” as the key issue connecting different theories ranging from epistemology to videography. Part two and three depict aesthetical praxis as general form and culture-related meaning as specification of fulfilling musical praxis. Two times four criteria identify aesthetical and cultural aspects of fulfilling musical praxis in the classroom (=SchoolMusic) and are illustrated by the joint music lessons on DVD, which are appended to the book. While the main emphasis of the article is descriptive and does not aim for normative purposes, an excursus drafts a model of music education called “Experiencing and comparing different kinds of musical praxis”
