Abstract:
AbstractFreeform curves are commonly used in contemporary design practices, especially with digital modeling tools. We investigate facilitating shape subtraction and addition with two-dimensional (planar) non-uniform rational basis-spline (NURB) curves with the codes and conventions of modeling while preserving the visual continuity of curved shapes. Our proposed tool, developed in a common digital modeling environment, automates the adjustment of parameters for tangential continuity of curves in shape rule applications. When the user designates a curve range to subtract from an initial shape and provides a new curved shape to add to it, the tool splits the initial shape, scales and aligns the curve to be added to fit into this range, introduces additional control points at the joining ends of the new curve to preserve continuity and redraws the new curve. We present a sample set of design variations produced using this practical approach which can be utilized as a method or become part of an automated NURB curve manipulation tool for designers.

ABSTRACT Chromatin boundaries facilitate independent gene regulation by insulating genes from the effects of enhancers or organized chromatin. However, the mechanisms of boundary action are not well understood. To investigate whether boundary function depends on a higher order of chromatin organization, we examined the function of several Drosophila melanogaster insulators in cells with reduced chromatin-remodeling activities. We found that knockdown of NURF301 and ISWI, key components of the nucleosome-remodeling factor (NURF), synergistically disrupted the enhancer-blocking function of Fab7 and SF1 and augmented the function of Fab8. Mutations in Nurf301/Ebx and Iswi also affected the function of these boundaries in vivo. We further show that ISWI was localized on the endogenous Fab7 and Fab8 insulators and that NURF knockdown resulted in a marked increase in the nucleosome occupancy at these insulator sites. In contrast to the effect of NURF knockdown, reduction in dMi-2, the ATPase component of the Drosophila nucleosome-remodeling and deacetylation (NuRD) complex, augmented Fab7 and suppressed Fab8. Our results provide the first evidence that higher-order chromatin organization influences the enhancer-blocking activity of chromatin boundaries. In particular, the NURF and NuRD nucleosome-remodeling complexes may regulate Hox expression by modulating the function of boundaries in these complexes. The unique responses by different classes of boundaries to changes in the chromatin environment may be indicative of their distinct mechanisms of action, which may influence their placement in the genome and selection during evolution.

Abstract:
The use of fusel oil as an ‘alternative’ carbon source for denitrification in the mainstream and sidestream treatment processes was studied. Research comprised two kinds of batch experiments as well as acclimation of process biomass to external carbon sources. In the conventional nitrate utilization rate (NUR) measurements (one-phase experiments with non-acclimated biomass), the NUR with fusel oil was 1.4–1.7 g N/(kg VSS·h which was comparable to NUR with ethanol and with slowly biodegradable fraction of the settled wastewater. When fusel oil was added at the beginning of the anoxic phase, preceded by an anaerobic phase (in two-phase experiments with non-acclimated biomass), the NURs of 2.5–2.9 g N/(kg VSS·h) were comparable to the tests without the addition of any external carbon sources. The addition of fusel oil and ethanol resulted in a significant enhancement of the denitrification efficiency in lab-scale sequencing batch reactors treating sludge reject water. The NURs continuously increased from below 1 g N/(kg VSS·h) to over 10 g N/(kg VSS·h) over the entire 4-week operational period, indicating gradual acclimation to the substrate. The overall total N removal efficiency reached ∼90%.

Abstract:
Turfgrass cultivars that have superior nitrate uptake ability are needed for the protection of ground water from pollution by excess nitrate. Information on temporal variation of nitrate absorption is also needed to enhance the environmental safety of turfgrass N fertilization programs. Our objectives were to evaluate Kentucky bluegrass (Poa pratensis L.) cultivars for their differences in nitrate uptake rate (NUR) and temporal variation in NUR. Six cultivars (Barzan, Blacksburg, Connie, Dawn, Eclipse, and Gnome) were propagated from individual tillers and six plants of each cultivar were generated from one mother plant. Plants were grown in silica sand, mowed weekly, and watered daily with half-strength modified Hoagland's nutrient solution containing 1 mM nitrate. When 5 months old, the plants were excavated, the roots were washed to remove sand, and the plants were transferred to 120-mL black bottles. After 24 hours in tap water, the plants were supplied with half-strength nutrient solution containing 0.5 mM nitrate, and the solutions were replaced daily for 8 days. NURs expressed as micromoles per plant per hour were calculated from solution nitrate depletion data. Significant genotypic differences in NUR were found: `Blacksburg' > `Connie' > `Dawn' > `Barzan' = `Eclipse' > `Gnome'. Significant temporal variation in NUR was also found, with NUR on the second day more than the first day after tap water. A significant interaction was noted between genotype and time. Temporal variation was greatest in `Blacksburg', while none noted in `Connie' and `Eclipse'. In `Barzan' and `Gnome', NUR on the last day was higher than the first day.
