AbstractNumerical simulations offer a wide range of benefits, therefore they are widely used in research and development. One of the biggest benefits is the possibility of automated parameter variation. This allow testing different scenarios in a very short period of time. Nevertheless, physical experiments in the laboratory or on a test rig are still necessary and will still be necessary in the future. The physical experiments offer benefits e.g. for very complex and/or nonlinear systems and are needed for the validation of numerical models.Fraunhofer LBF has developed hardware solutions to bring the benefit of rapid and automated parameter variation to experimental environments. These solutions allow the tuning and emulation of the mechanical properties, like stiffness, damping and eigenfrequencies of structures.The work presents two approaches: First a stiffness tunable mount, which has been used in laboratory tests in the field of semi-active load path redistribution. It allowed the researcher to test the semi-active system under different mechanical boundary conditions in a short period of time. Second, a mechanical Hardware-in-the-loop (mHIL) approach for the NVH development of vehicles components is presented. Here a mHIL-system is used to emulate the mechanical characteristics of a vehicle’s body in white in a wide frequency range. This allows the experimental NVH optimization of vehicle components under realistic boundary conditions, without actually needing a (prototype) body in white.

AbstractCompetitive industrial transmission systems must perform most efficiently with reference to complex requirements and conflicting key performance indicators. This design challenge translates into a high-dimensional multi-objective optimization problem that requires complex algorithms and evaluation of computationally expensive simulations to predict physical system behavior and design robustness. Crucial for the design decision-making process is the characterization, ranking, and quantification of relevant sources of uncertainties. However, due to the strict time limits of product development loops, the overall computational burden of uncertainty quantification (UQ) may even drive state-of-the-art parallel computing resources to their limits. Efficient machine learning (ML) tools and techniques emphasizing high-fidelity simulation data-driven training will play a fundamental role in enabling UQ in the early-stage development phase.This investigation surveys UQ methods with a focus on noise, vibration, and harshness (NVH) characteristics of transmission systems. Quasi-static 3D contact dynamic simulations are performed to evaluate the static transmission error (TE) of meshing gear pairs under different loading and boundary conditions. TE indicates NVH excitation and is typically used as an objective function in the early-stage design process. The limited system size allows large-scale design of experiments (DoE) and enables numerical studies of various UQ sampling and modeling techniques where the design parameters are treated as random variables associated with tolerances from manufacturing and assembly processes. The model accuracy of generalized polynomial chaos expansion (gPC) and Gaussian process regression (GPR) is evaluated and compared. The results of the methods are discussed to conclude efficient and scalable solution procedures for robust design optimization.

Night active insects inspired the development of image enhancement methods that uncover the information contained in dim images or movies. Here, I describe a novel bionic night vision (NV) algorithm that operates in the spatial domain to remove noise from static images. The parameters of this NV algorithm can be automatically derived from global image statistics and a primitive type of noise estimate. In a first step, luminance values were ln-transformed, and then adaptive local means’ calculations were executed to remove the remaining noise without degrading fine image details and object contours. Its performance is comparable with several popular denoising methods and can be applied to grey-scale and color images. This novel algorithm can be executed in parallel at the level of pixels on programmable hardware.

This chapter discusses the genealogy of construction materials testing at acoustic laboratories founded in the late 1920s and 1930s, through the example of Norges Tekniske Høgskole (NTH) in Trondheim, Norway. Whereas architectural acoustics entailed mainly fieldwork, in the form of on-site research and consulting, the standardized testing of materials required idealized laboratory conditions where acoustic behavior could be isolated and reduced to the parameters of absorption, transmission, and reflection. With the rise of the acoustic laboratory, “objective” measurement systems replaced musical listening as a crucial skill for acousticians. Electroacoustic media technologies quickly spread on a global scale, yet acoustic laboratories still catered to the needs of local markets. One of these requirements was resource optimization, and the objective of standardized testing generally was less to improve the quality of construction than to optimize its cost. Traditional materials and buildings often exhibited better acoustic properties than the new constructions based on scientific principles.

Repurposing or repositioning means validating and application of previously approved drugs in the treatment of another disease that might be relevant or irrelevant to existing use in disease based on the principle of polypharmacology. Repurposed drugs are already well documented for pharmacokinetic, pharmacodynamic, drug interaction, and toxicity parameters. In 1962, thalidomide treatment in pregnant women led to phocomelia in their newborn but while repurposed based on anti-angiogenesis property, it showed efficacy in hematologic malignancies like multiple myeloma. The repurposing is becoming an essential tool in the anti-cancer drug development due to existing drugs are not effective, high cost of treatment, therapy may degrade the quality of life, improvement of survival after treatment is not guaranteed, relapse may occur, and drug resistance may develop due to tumor heterogeneity. Repurposing can be addressed well with the help of literature-based discovery, high throughput technology, bioinformatics multi-omics approaches, side effects, and phenotypes. Many regulatory bodies like EML, NIH, and FDA promote repurposing programs that support the identification of alternative uses of existing medicines. Cancer becomes the major health issue, and the need to discover promising anti-cancer drugs through repurposing remains very high due to decline in FDA approval since 1990, huge expenses incurred in the drug development and prediction of dangerous future burden.

Abstract.—A water filtration and ozonation system was recently installed to treat creek water used to culture species of concern at the U.S. Fish and Wildlife Service’s Northeast Fishery Center, Lamar National Fish Hatchery (NFH). Past experience with fish culture indicates that the following bacterial pathogens are endemic to the creek water supply: <em>Aeromonas salmonicida</em>, <em>Yersinia ruckeri</em>, <em>Flavobacterium columnaris</em>, and <em>Flavobacterium psychrophilia</em>. Water samples were collected from sites located before and after filtration and ozonation and examined for culturable bacteria. Variable operation of the filtration/ozonation system was used to examine (1) the effect of microscreen filtration (i.e., using drum filters containing 37-μm sieve panels) on ozone inactivation of bacterial microorganisms, (2) the effect of dissolved ozone contact times on inactivation of bacterial microorganisms, and (3) the effect of water quality fluctuations on the dissolved ozone demand measured during the course of these tests. Inactivation exceeded 98% for all bacteria when ozone <em>C*t </em>values were about 1.0 and reached 100% at 21.3, regardless of water quality parameters or implementation of microscreen filtration. These results indicate that the use of ozonation to treat surface water supplies used for fish culture facilities will effectively inactivate the majority of bacteria entering the system and will likely serve to prevent introduction of bacteria that can be pathogenic to fish.

Past experience with fish culture indicates that the following bacterial pathogens are endemic to the creek water supply: <em>Aeromonas salmonicida</em>, <em>Yersinia ruckeri</em>, <em>Flavobacterium columnaris</em>, and <em>Flavobacterium psychrophilia</em>. Water samples were collected from sites located before and after filtration and ozonation and examined for culturable bacteria. Variable operation of the filtration/ozonation system was used to examine (1) the effect of microscreen filtration (i.e., using drum filters containing 37-μm sieve panels) on ozone inactivation of bacterial microorganisms, (2) the effect of dissolved ozone contact times on inactivation of bacterial microorganisms, and (3) the effect of water quality fluctuations on the dissolved ozone demand measured during the course of these tests. Inactivation exceeded 98% for all bacteria when ozone <em>C*t </em>values were about 1.0 and reached 100% at 21.3, regardless of water quality parameters or implementation of microscreen filtration. These results indicate that the use of ozonation to treat surface water supplies used for fish culture facilities will effectively inactivate the majority of bacteria entering the system and will likely serve to prevent introduction of bacteria that can be pathogenic to fish.
