ABSTRACT To examine the mode of JC virus (JCV) transmission, we collected urine samples from second- and third-generation Japanese-Americans in Los Angeles, Calif., whose parents and grandparents were all Japanese. From the urine samples of these Japanese-Americans, we mainly detected two subtypes (CY and MY) of JCV that are predominantly found among native Japanese. This finding provides support for the hypothesis that JCV is transmitted mainly within the family through long-term cohabitation.

Abstract. Transportable biomass conversion facilities producing biochar, briquettes, and torrefied wood are modeled and optimized for five different sub-regions within the Pacific Northwest. Subregional case studies in Quincy, California; Lakeview, Oregon; Oakridge, Oregon; Port Angeles, Washington; and Warm Springs, Oregon, are evaluated to characterize the potential economic viability of these novel transportable designs. A mixed integer program is used to characterize the supply chain from residue extraction to market optimizing transportation, production, and plant mobility in order to minimize the supply chain costs. Regional variations including log specifications, energy rates, trucking, and logistic capacities are considered within the model and supporting analyses to differentiate regional costs and market viabilities. It was found that the optimal transportable design included facility movement on a 1 to 2.5 year frequency depending on product and region with biochar being the most likely to be economically viable. Regional feedstock composition and availability was the biggest indicator of lower cost production. Supply chain costs varied by 5%-10% depending on product and region being produced. Transportation and mobilization were found to account for 15%-30% of the overall supply chain cost. Quincy, California, and torrefied wood were found to have the lowest of these costs due to low move frequency and high wood availability while Port Angeles, Washington, with briquettes was the highest. With regards to fuel price sensitivity, torrefied wood was the most sensitive as its conversion process was most energy intensive (±12%-13%) and biochar least sensitive (±3-5%).Transportation accounted for 5% to 30% of the fuel price variation due to diesel prices depending on product and region. When including grid-connectivity, cost reductions were approximately 6%-7% for biochar, 27%-29% for briquettes and 33%-38% for torrefied wood. These findings indicate biochar as the most likely candidate for a transportable conversion system given its relatively low power consumption, high allowable moisture content, and low product transportation cost. Quincy, California, was found to be the most desirable sub region with the lowest overall production costs attributed to its high input quality feedstock and relative accessibility; its higher grid-connected power cost also makes transportable options relatively more attractive than other off-grid locations. Port Angeles, WA had the highest production costs and lowest grid-energy costs. Our results indicate that a rise in diesel price, while incentivizing transportable conversion facilities due to more cost effective transportation, would be more than offset by the higher cost energy consumption during the conversion process when compared with grid-power with the potential exception of biochar. Overall, we see a transportable operation with grid-power would likely be the difference between an economically viable supply chain and one that is not. ABSTRACT Helaeomyia petrolei larvae isolated from the asphalt seeps of Rancho La Brea in Los Angeles, Calif., were examined for microbial gut contents. Standard counts on Luria-Bertani, MacConkey, and blood agar plates indicated ca. 2 × 105 heterotrophic bacteria per larva. The culturable bacteria represented 15 to 20% of the total population as determined by acridine orange staining. The gut itself contained large amounts of the oil, had no observable ceca, and maintained a slightly acidic pH of 6.3 to 6.5. Despite the ingestion of large amounts of potentially toxic asphalt by the larvae, their guts sustained the growth of 100 to 1,000 times more bacteria than did free oil. All of the bacteria isolated were nonsporeformers and gram negative. Fourteen isolates were chosen based on representative colony morphologies and were identified by using the Enterotube II and API 20E systems and fatty acid analysis. Of the 14 isolates, 9 were identified as Providencia rettgeriand 3 were likely Acinetobacter isolates. No evidence was found that the isolates grew on or derived nutrients from the asphalt itself or that they played an essential role in insect development. Regardless, any bacteria found in the oil fly larval gut are likely to exhibit pronounced solvent tolerance and may be a future source of industrially useful, solvent-tolerant enzymes.

ABSTRACT The ubiquity of fecal indicator bacteria such as Escherichia coli and Enterococcus spp. in urban environments makes tracking of fecal contamination extremely challenging. A multitiered approach was used to assess sources of fecal pollution in Ballona Creek, an urban watershed that drains to the Santa Monica Bay (SMB) near Los Angeles, Calif. A mass-based design at six main-stem sites and four major tributaries over a 6-h period was used (i) to assess the flux of Enterococcus spp. and E. coli by using culture-based methods (tier 1); (ii) to assess levels of Enterococcus spp. by using quantitative PCR and to detect and/or quantify additional markers of human fecal contamination, including a human-specific Bacteroides sp. marker and enterovirus, using quantitative reverse transcriptase PCR (tier 2); and (iii) to assess the specific types of enterovirus genomes found via sequence analysis (tier 3). Sources of fecal indicator bacteria were ubiquitous, and concentrations were high, throughout Ballona Creek, with no single tributary dominating fecal inputs. The flux of Enterococcus spp. and E. coli averaged 109 to 1010 cells h−1 and was as high at the head of the watershed as at the mouth prior to discharge into the SMB. In addition, a signal for the human-specific Bacteroides marker was consistently detected: 86% of the samples taken over the extent during the study period tested positive. Enteroviruses were quantifiable in 14 of 36 samples (39%), with the highest concentrations at the site furthest upstream (Cochran). These results indicated the power of using multiple approaches to assess and quantify fecal contamination in freshwater conduits to high-use, high-priority recreational swimming areas.

ABSTRACT Helaeomyia petrolei (oil fly) larvae inhabit the asphalt seeps of Rancho La Brea in Los Angeles, Calif. The culturable microbial gut contents of larvae collected from the viscous oil were recently examined, and the majority (9 of 14) of the strains were identified as Providencia spp. Subsequently, 12 of the bacterial strains isolated were tested for their resistance or sensitivity to 23 commonly used antibiotics. All nine strains classified as Providencia rettgeri exhibited dramatic resistance to tetracycline, vancomycin, bacitracin, erythromycin, novobiocin, polymyxin, colistin, and nitrofurantoin. Eight of nineProvidencia strains showed resistance to spectinomycin, six of nine showed resistance to chloramphenicol, and five of nine showed resistance to neomycin. All 12 isolates were sensitive to nalidixic acid, streptomycin, norfloxacin, aztreonam, cipericillin, pipericillin, and cefotaxime, and all but OF008 (Morganella morganii) were sensitive to ampicillin and cefoxitin. The oil fly bacteria were not resistant to multiple antibiotics due to an elevated mutation rate. For each bacterium, the number of resistant mutants per 108cells was determined separately on rifampin, nalidixic acid, and spectinomycin. In each case, the average frequencies of resistant colonies were at least 50-fold lower than those established for known mutator strain ECOR 48. In addition, the oil fly bacteria do not appear to excrete antimicrobial agents. When tested, none of the oil fly bacteria produced detectable zones of inhibition on Pseudomonas aeruginosa, Escherichia coli, Staphylococcus aureus, or Candida albicans cultures. Furthermore, the resistance properties of oil fly bacteria extended to organic solvents as well as antibiotics. When pre-exposed to 20 μg of tetracycline per ml, seven of nine oil fly bacteria tolerated overlays of 100% cyclohexane, six of nine tolerated 10% xylene, benzene, or toluene (10:90 in cyclohexane), and three of nine (OF007, OF010, and OF011) tolerated overlays of 50% xylene–50% cyclohexane. The observed correlation between antibiotic resistance and organic solvent tolerance is likely explained by an active efflux pump that is maintained in oil fly bacteria by the constant selective pressure of La Brea's solvent-rich environment. We suggest that the oil fly bacteria and their genes for solvent tolerance may provide a microbial reservoir of antibiotic resistance genes.
