Full textThe Finite Volumes Method adopted by the commercial software Star-CCM+ is applied to the simulation of the steady state regime of incompressible and compressible turbulent flows over selected airfoil’s sections. The physical model used with the incompressible flow case is a NACA 0012 airfoil section at zero angle of attack. The ONERA’s OAT15A supercritical airfoil section at small angle of attack applies to the compressible flow case. The computational domains are discretized by structured and unstructured meshes for different topologies and far field configurations. The structured meshes are of the quadrilateral type with hyperbolic node distribution whilst the unstructured meshes use polyhedral cells. The grids are generated by applying a methodology where estimates of the flow variables are used as input for the grid’s constructive parameters like the near wall cell thickness. Grid dependency studies are carried out in order to verify the grid’s quality and suitability to represent the physical phenomena. The grid’s asymptotic convergence index GCI and its observed order of convergence are evaluated for three refinement levels and far field position for the compressible flow cases. The objective is to select the most suitable grid taking into account the accuracy requirements and the computational resources available. The one-equation Spalart-Allmaras turbulence model and the two-equation k-ω/SST turbulence models are used. The numerical results are discussed from the physical point of view and compared with the experimental ones available in literature.

Herb-drug interaction and Chinese hebal medicine induced nephrotoxicity (CHN) raise great concern since the broad use of Chinese herbal medicine (CHM) in Taiwan. Human ortholog organic anion transporter 1 (hOAT1) located in the kidney basolateral membrane is reported to play an important role in drug-drug interaction and drug induced nephrotoxicty. Therefore, the purpose of this study is to estimate the functional modulation effect of CHM on hOAT1 and to evaluate the possibility of the candidates induced herb-drug interaction and CHN. hOAT1 transfected Madin-Darby canine kidney type II (MDCK II-hOAT1) cell line and original MDCK II cell line were used to complete the in vitro CHM screening and cytotoxicity test. The potential candidates were selected to further in vivo renal functional study, which was performed by continuous infusion of p-aminohippuric acid (PAH) and inulin mixture to Wistar rat. CHM induced kidney injury was then evaluated by kidney biopsy. The results of in vitro screening showed that seven Chinese herbal formulae (Gui-Zhi-Fu-Ling-Wan, Liu-Wei-Di-Huang-Wan, Chia-Wei-Hsiao-Yao-San, Chi-Chu-Di-Huang-Wan, Chih-Po-Di-Huang-Wan, Hsin-I-Ching-Fei-Tang and Lung-Tang-Hsieh-Kan-Tang; by inhibition order) effectively inhibited more than 50% of hOAT1 mediated transport; where two single herbs (Huang-Chin and Huang-Lian; by inhibition order) achieved 30 – 50% inhibition. All of the above mentioned CHM showed concentration-dependent manner of hOAT1 inhibition ability, which revealed that dosage might be related to the effect of CHM on renal function. In the cytotoxicity test, Huang-Chin and Huang-Lian dramatically reduced cell viability of MDCK II-hOAT1 and MDCK II cells, respectively. The reason of the opposite cytotoxicity results in Huang-Chin and Huang-Lian remained unclear and need to be evaluated by further study. In order to estimate the effect of potential CHM candidates on aminal renal function, performed by evaluating PAH and inulin clearance in Wistar rat, a novel analytical method that could simultaneously determine PAH and inulin in rat plasma by LC-MS/MS was developed and well validated. The analytical method was applied to the in vivo study. The results of the in vivo study showed that Gui-Zhi-Fu-Ling-Wan and Chia-Wei-Hsiao-Yao-San significantly decreased PAH secretion clearance (PAHsec) (1.89 ± 0.16/1.96 ± 0.09; control: 2.34 ± 0.05 mL/min/100 mg BW); where Chi-Chu-Di-Huang-Huang, Hsin-I-Ching-Fei-Tang and Lung-Tan-Hsieh-Kan-Tang significantly reduced glomerular filtration rate (GFR) (0.88 ± 0.04/0.80 ± 0.03/0.83 ± 0.04; control: 0.99 ± 0.03 mL/min/100 mg BW). High dose of Gui-Zhi-Fu-Ling-Wan and Lung-Tan-Hsieh-Kan-Tang resulted a more obivious suppression of PAHsec (1.79 ± 0.10/1.76 ± 0.14 mL/min/100 mg BW) rather than GFR (0.92 ± 0.03/0.82 ± 0.05 mL/min/100 mg BW). Biopsy of these two high dose CHM groups showed severe tubular damage. In conclusion, Gui-Zhi-Fu-Ling-Wan, Chia-Wei-Hsiao-Yao-San, Chi-Chu-Di-Huang-Huang, Hsin-I-Ching-Fei-Tang and Lung-Tan-Hsieh-Kan-Tang might be related to herb-drug interaction and CHN.
