Abstract:
Purpose. To investigate how multipotent retinal stem cells (RSCs) isolated from mice respond to the proinflammatory signaling molecules, IL-1β, IL-18, and IL-17A.Materials and Methods. RSCs were cultured in a specific culture medium and were treated with these cytokines. Cell viability was detected by MTT assay; ultrastructure was evaluated by transmission electron microscopy; expression of IL-17rc and proapoptotic proteins was detected by immunocytochemistry and expression ofIl-6andIl-17awas detected by quantitative RT-PCR. As a comparison, primary mouse retinal pigment epithelium (RPE) cells were also treated with IL-1β, IL-18, or IL-17A and analyzed for the expression ofIl-6andIl-17rc.Results. Treatment with IL-1β, IL-18, or IL-17A decreased RSC viability in a dose-dependent fashion and led to damage in cellular ultrastructure including pyroptotic and/or necroptotic cells. IL-1βand IL-18 could induce proapoptotic protein expression. All treatments induced significantly higher expression ofIl-6andIl-17rcin both cells. However, neither IL-1βnor IL-18 could induceIl-17aexpression in RSCs.Conclusions. IL-1β, IL-18, and IL-17A induce retinal cell death via pyroptosis/necroptosis and apoptosis. They also provoke proinflammatory responses in RSCs. Though IL-1βand IL-18 could not induceIl-17aexpression in RSCs, they both increaseIl-17rcexpression, which may mediate the effect ofIl-17a.

Abstract:
In this article, we propose and study a new three-parameter distribution, called the odd Fréchet inverse Lomax (OFIL) distribution, derived by combining the odd Fréchet-G family and the inverse Lomax distribution. Since Fréchet is a continuous distribution with wide applicability in extreme value theory, the new model contains these properties as well as the characteristics of the inverse Lomax distribution which make it more flexible and provide a good alternative for some well-known lifetime distributions. We initially present a linear representation of its functions and discussion on density and hazard rate function. Then, we study its various mathematical properties. Different estimation methods are used to estimate parameters of OFIL. The Monte Carlo simulation study is carried out to compare the efficiencies of different methods of estimation. The maximum likelihood estimation (MLE) method is used to estimate the OFIL parameters by considering three practical data applications. We show that the related model is the best in comparisons based on Akaike information criterion (AIC), Bayesian information criterion (BIC), and other goodness-of-fit measures.

DNA samples from 339 Crohn's disease (CD) and 407 randomly selected controls from the Auckland (New Zealand) IBD project, were genotyped for five common single nucleotide polymorphisms inIL-23R(rs11805303, rs7517847, rs1343151, rs11209026, and rs10889677) and two inIL-12B(rs1363670 and rs6887695). While theIL-12Bvariants did not show an overall association and otherIL23Rvariants led to minor changes in the risk of CD, rs1343151 and/or rs7517847 variants in theIL-23Rgene strongly reduced the risk of developing CD at both allelic and genotype levels. A significantly decreased risk of first diagnosis of childhood CD was observed in individuals carrying the A allele of rs1343151, or between 17–40 y in individuals carrying the G allele in rs7517847 ofIL-23R. A significantly decreased risk of ileocolonic or structuring disease was observed in individuals carrying the A allele in either rs11209026 or rs1343151, or the G allele in rs7517847 ofIL-23R, and when such individuals did develop the disease, they were unlikely to require a bowel resection. Certain haplotypes very strongly modified risk. There was evidence for interactions ofIL-23Rvariants with theNOD2wild-type (d/d) genotype. Down-regulating the function of theIL-23Rgene may decrease CD risk in the normal population.

Inflammation has been demonstrated to be involved in Parkinson’s disease (PD) pathogenesis. There were evidences that the disturbance of the protective function ofIL-10gene contributed to PD. In our study, haplotype analyses were conducted ofIL-10rs1800871 and rs1800872 on 371 PD patients. Because the two SNPs exposed significant linkage disequilibrium demonstrated by Haploview software, we included 177 carriers of both rs1800871 and rs1800872 and 190 noncarriers in clinical phenotype analyses. As to nonmotor symptoms, the score of the gastrointestinal dysfunction domain in Nonmotor Symptom Scale (NMSS) was lower in the carrier group of both SNPs than in the noncarrier group in PD patients (SC: −0.198,p:0.023). Other nonmotor symptoms reflected by relevant rating scales showed negative results. As to comorbidity, no significant statistical significance was observed between the two SNPs and Charlson Comorbidity Index (CCI). In conclusion, we found less severe gastrointestinal dysfunctions of bothIL-10rs1800871 and rs1800872 carriers than noncarriers in PD.
