Abstract:
The aim of this study was to evaluate the effects of temperature and storage time on the formation of biogenic amines (BAs) in the traditional Czech curd cheese (Olomoucké tvarůžky). Samples were stored for 7 weeks at 5 °C and 20 °C. BAs were studied as dansyl derivatives by the RP-HPLC method with fluorescence detection, histamine was determined using a photodiode array detector. Physical and chemical properties were analyzed as specified by the Czech National Standard, as were the sensory characteristics (colour, odour, texture and flavour). The major amines found were cadaverine (124–2413 mg kg-1) and tyramine (117–1058 mg kg-1), followed by putrescine (75–767 mg kg-1) and histamine (74–411 mg kg-1). Low concentrations of tryptamine, spermine and spermidine were present. Total concentrations of BAs significantly increased with storage time (P < 0.01), depending significantly on temperature (P < 0.01). Total BAs in cheese stored at 20 °C compared to 5 °C were more than three times higher, reaching 4600 mg kg-1 at the end of storage. The toxicologically critical value of 900 mg kg-1 for the sum of histamine + tyramine + putrescine + cadaverine was reached 17 days later in the cheese stored at 5 °C compared to 20 °C. When stored at 5 °C, the samples retained adequate sensory characteristics for the entire safe storage time. The effects of storage conditions on BAs formation are relevant to reducing the risk associated with consumption of cheese high in BAs.

Abstract:
Olomouc cake of cheese (Olomoucké tvarůžky) is smear-ripened cheese, which is produced from sour industrial curd. Brevibacterium linens, which are added during the production process, are reproducing and make gold-yellow smear cover. The aim of this work was to assess the chemical analysis of the quality of Olomouc cake of cheese. Changes in chemical composition were evaluated during different stages of production and at the same time it was detected whether changes in chemical composition during the manufacturing process are same in spring as well as in summer, without statistically significant differences. Dry matter of Olomouc cake of cheese was ranged from 35 % to 39 %. The increase of dry matter during production is evident, but these changes were in the most cases not statistically signifiant (P > 0.05). The value of titratable acidity of the cheese considerably changes during the manufacturing process, it has a decreasing tendency. Titratable acidity of cheese after shaping was 106.64 (136.12) SH and at the end of life it was 49.91 (65.06) SH. These changes were very highly statistically significant (P < 0.001). Increasing the dry matter of cheese is also proportionatelly increase content of salt in cheese, although this changes are not statistically significant (P > 0.05) in cheese from summer period. Content of salt is increased from 5.30 % to 5.98 %, respectively 6.10 %. In spring period the oposite changes in most cases occured (P < 0.001), it is increased from 4.27 % to 6.20 %, respectively 6.94 %. When the chemical composition of the cheese in spring and in summer period is compared, there are no significant changes (P > 0.05).

Abstract The collection of ancient coins of the Palacký University in Olomouc is recently preserved in the department of history there. It is very likely that the collection was founded sometimes between the second half of the 19th century and the beginning of the 20th century – no details are known about it and the collection became the university property evidently after 1946. The core of the Olomouc collection is represented by denarii dating back to 154–74 BC (cat. nos. 3–69). Only individual pieces of the earlier issues (cat. nos. 1–2) and the later issues (cat. nos. 70–72) are present here. Publication of this collection could be accepted as a small contribution to documentation of the physically verified material of the mentioned period. Parallelly, it is an interesting testimony of the collecting activities in Olomouc at the break of the 19th and 20th centuries.

Cooperation differently oriented public institutions in order to develop leisure activities of people with disabilities is crucial in relation to the implementation of a series of legislative standards (UN Convention on the Rights of Persons with Disabilities, Act on the promotion of sports, etc.). The city of Olomouc is one of the most active operators in the long term municipal level, which addresses support for people with disabilities or physical handicaps comprehensively. In 2012, the APA Center began an intense collaboration (Department APA, FTK UP in Olomouc) with the Department of Social Affairs to address accessibility of cultural and sports facilities, public spaces, playgrounds, etc for people woth special needs. As part of efforts to unify methodologies and sharing information we intend to use in the monitoring of barrier-free collaboration with Kazuist company. APA Center was invited to a group of community planning in the city of Olomouc. In addressing the use of specific professional works contracts by the city for the development of leisure activities, promotion of tourism and social services.

SummaryVaccination againstNeisseria meningitidisis not part of routine immunization schemes in any country; instead, targeted vaccination of groups at the highest risk is recommended during outbreaks and epidemics. After a long period of sporadic occurrence of meningococcal invasive disease, a new clone ofNeisseria meningitidisC: 2a: P1.2, ET-15/37, occurred in the Czech Republic, and caused local outbreaks in two neighbouring districts, Olomouc and Bruntal, in spring 1993. In Olomouc, a mass campaign was conducted during which 6191 students were vaccinated (5·6 % of the total population of this locality and 96% of all students in the age group 15–19) within 2 weeks in June 1993. In Bruntal district, no such campaign was organized. In Olomouc, the incidence of invasive disease caused byNeisseria meningitidisC in the age group 10–24 decreased from 57 to 0 per 100000 (P < 0·001) during the post-vaccination period (July 1993–August 1994), but no such decrease was observed in Bruntal. Although other factors can affect the frequency of disease, these results support the current recommendations of targeted vaccination in outbreaks of meningococcal disease.

In 1830, a new theater building was opened in the Olomouc Upper square. The stable theatrical life enriched enormously the cultural life of the city and encouraged the development of publishing activities in the field of music journalism and publishing. The public debates on the artistic value of theater performances, on abilities of particular artists and on other subjects gained new quality after the 1860 October diploma because Czechs living in and around the traditional German town put pressure on theater directors and demanded Czech plays on the stage. The fights for the national repertoire on the stage of the Olomouc Provincial Theater are demonstrated in this essay in two contrary ways: at first, the introduction of Czech dramas into the German scene during the 1860s is discussed, then the intensive promotion of German operas during the 1880s and 1890s when internationally played Slavonic operas were performed in all theaters. The director Carl König (1862–1868) offered a contract to many artists who were able to speak both German and Czech, so he could open an independent subscription for the Czech public. The relatively tolerant atmosphere allowed König’s company to give performances in both languages and connect the Olomouc theatrical life to the Prague Provisional Theater. However, Czech nationalism was getting stronger during the 1870s and provoked competitive and unfriendly reactions on German side. The arguments for refusal of Smetana’s and Tchaikovsky’s operas by the directors of the Olomouc theaters are discussed on the basis of archival sources as well as articles published in contemporary periodicals.

Abstract The service sector is quite broad: it includes basic services for the population as well as highly sophisticated services, public as well as commercial services, etc. Commuting to services is one of the fundamental regional processes and together with work commuting it creates an entry basis for the construction of socalled catchment, or nodal regions which should be crucial in the construction of an administrative division of a country. The main objective of this paper is to identify a selected group of spatial interaction in the daily urban system of Olomouc, i.e. in a region dominated by work commuting to the city of Olomouc. More specifically, the paper deals with the interaction of commuting to a wide range of services of non-commercial (social) as well as of commercial character in two hierarchical levels - local and micro-regional. Based on the data obtained by the method of questionnaire research from local leaders (mayors) we analysed the mobility of people to primary and secondary schools, hospitals, shops, culture and sport. As far as the retail network is concerned, the analysis of spatial differentiation of the catchment area to three large department stores was made for villages in Olomouc hinterland. Individual catchment regions for specific services were constructed by a verified method based on commuting flows. Synthetic evaluation contains an identification of so-called complex service regions in the daily urban system of Olomouc. In comparison with our previous studies we tried to describe basic trends in service infrastructure and changes in travelling to services.

Abstract:
This article deals with the phenomenon of invalid voting which has appeared in regional elections in the Olomouc region in the Czech Republic. First, contemporary theories of invalid voting are introduced. Second, the given hypotheses, which include both institutional and socio-economic factors that, according to the theories, contribute to higher levels of invalid voting, are tested. Statistical analysis such as linear regression is employed in the research. The dataset is comprised of municipalities (cases) in which regional elections were held. The results show that higher levels of invalid voting are associated with concurrent elections (elections to the regional assembly and elections to the upper house (the Senate) of the Czech Parliament) in those municipalities in which residents are older and less well-educated. If regional elections are held without concurring elections, socio-economic variables have a smaller effect on invalid voting. Therefore, the state authorities should consider a revision of respective electoral law and simplify the electoral process involved in concurrent elections.

Abstract:
<p>In the Czech Republic, clinical legal education is not very developed. It does not fit into the traditional view of legal education, which itself is based mainly on an Austrian and German legal tradition which emphasizes legal positivism, legal history and a lecture style of lessons. The university environment in the Czech Republic is very conservative. Open minded teachers (mostly young assistants) have very little space for their own creativity during the lessons. Most of the Czech law teachers do not have any training in teaching methodology. Moreover the general attitude is that there is no special need for practical courses because “the students will receive their practical training once they are in practice after finishing the law school”. These attitudes result in very sceptical approach to the methodology of teaching, if we can speak about any methodology at all. To put it simply, the content of the lecture is seen as more important than the way the content is taught.</p>

Abstract:
This paper attempts to give a survey of the origin of the present collection of some forty works of Grotius in the present Scientific Library of Olomouc. After a short introduction about education in the Czech lands and especially in Olomouc, the present works of Grotius are discussed in connection with their origin. Most works were added to the collection due to the Josephine abolition of monasteries in the 1780s. Premonstratensian and Cistercian monasteries were the most important former possessors. A couple of Grotiana were donated by noblemen. A look is given to some course books that have been preserved. A complete list of all works of Grotius printed before 1800, present in the Scientific Library with an identification of their owners, is given in the annex.

Abstract:
The objective of the paper was to present the concept of undergraduate preparation of future primary school teachers at the Faculty of Education in Olomouc. This course has had a long tradition in Olomouc, and the current concept of the course is based on an overview of the system of primary education presented in this paper. In addition to this concept, the authors reflect on the needs of primary schools and try to implement these needs within the educational context. The concept of the course focuses not only on students’ theoretical knowledge but primarily on the application sphere. The paper is a partial outcome of the project Development of the concept of undergraduate training of primary school teachers with an emphasis on communication competences.

Abstract:
The manuscript of the "Introduction to the Knowledge on the Hereditary Markgraviate of Moravia" as an appendix to the lecture of political science at the Olomouc lyceum written in 1797 by Prof. Kryštof Passy deals also in several paragraphs with the description of the climate of Moravia. The author, departing from the meteorological observations by Josef Gaar in Olomouc, mentions the description of air pressure, temperature and moisture, evaporation and wind. Besides the description of regional peculiarities of the Moravian climate, Passy tries to explain their causes and deals in detail with the effect of eight basic wind directions on changes in air temperature, air moisture and the course of weather from January to July. Passy's description is verified with respect to the results of modern measurements and the present-day knowledge on Moravian climate.

Abstract:
Olomouc, nowadays a city with 100,000 inhabitants, has always been considered as one of the most prominent Czech cities. It is a social and economical centre, which history started just about the 11th century. The present appearance of the city has its roots in the 18th century, when the city was almost razed to the ground after the Thirty years’ war and a great fire in 1709. After that, the city was rebuilt to a baroque military fortress against Prussia army. At the beginning of the 20th century the majority of the fortress was demolished. Character of the town is dominated by the large number of churches, burgher’s houses and other architecturally significant buildings, like a Holy Trinity Column, a UNESCO World Heritage Site. Aim of this project was to state the most suitable methods of visualization of spatial-temporal change in historical build-up area from the tourist’s point of view, and to design and evaluate possibilities of spatial data acquisition. There are many methods of 2D and 3D visualization which are suitable for depiction of historical and contemporary situation. In the article four approaches are discussed comparison of historical and recent pictures or photos, overlaying historical maps over the orthophoto, enhanced visualization of historical map in large scale using the third dimension and photorealistic 3D models of the same area in different ages. All mentioned methods were geolocalizated using the Google Earth environment and multimedia features were added to enhance the impression of perception. Possibilities of visualization, which were outlined above, were realized on a case study of the Olomouc city. As a source of historical data were used rapport plans of the bastion fortress from the 17th century. The accuracy of historical maps was confirmed by cartometric methods with use of the MapAnalyst software. Registration of the spatial-temporal changes information has a great potential in urban planning or realization of reconstruction and particularly in the propagation of the region and increasing the knowledge of citizens about the history of Olomouc.
