Abstract:
Biodesulfurization (BDS) is an alternative to hydrodesulfurization (HDS) as a method to remove sulfur from crude oil. Dibenzothiophene (DBT) was chosen as a model compound for the forms of thiophenic sulfur found in fossil fuels; up to 70% of the sulfur in petroleum is found as DBT and substituted DBTs; these compounds are however particularly recalcitrant to hydrodesulfurization, the current standard industrial method. My thesis deals with enhancing BDS through novel strains and through nanotechnology. Chapter highlights are: Chapter 2. My first aim was to isolate novel aerobic, mesophilic ba

A method was developed to access the interior of light-guiding structures in order to exploit the enhanced sensing potential of the highly confined electromagnetic field distributions, located within the core of a waveguide. The work presented in this thesis explores therefore the possibilities of optical waveguide spectroscopy utilising transparent mesoporous thin-film waveguides deposited on top of athin gold layer. These multi-layer assemblies are employed in a prism-coupling attenuated total internal reflection (ATR) configuration. The angular read-out of the reflected light intensity allo

Abstract:
A broadband, multichannel, single mode, planar waveguide based ATR spectrometer was developed. A sensitivity enhancement of three orders of magnitude compared to the conventional transmission spectroscopic technique has been experimentally achieved. Applications to protein submonolayers adsorbed on glass surfaces provided the first results on the spectral characterization of those molecular films. The increased information content and the higher sensitivity response allowed us to experimentally determine the molecular orientation of organic films over a broad spectral range. The work developed

This dissertation reports the application of nonlinear optical effects for the investigation of vibrational and electronic spectroscopy of molecules adsorbed on thin film metal oxide surfaces and metal oxide surfaces. The main emphasis of the experiments cited here is to introduce the recently developed multi-photon technique, Waveguide Surface Coherent anti-Stokes Raman Scattering Spectroscopy (WSCARS), to the scientific community. Planar optical waveguides have been utilized to generate large optical field enhancements on metal oxide surfaces. Guided waves have been employed to obtain the su

Le régime de couplage fort lumière-matière et l’effet laser à polaritons ont pu être observés de 5 K à 300 K pour les deux microcavités. Abstract:
Nisin, an amphiphilic, antimicrobial peptide, has been shown to integrate into the hydrophobic inner region of poly(ethylene oxide) (PEO) brush layers; however, the presence of integrated nisin may compromise the protein repulsive character of the PEO layer. In particular, the introduction of fibrinogen to nisin-loaded brush layers has been observed to cause changes consistent with partial elution of nisin and/or location of fibrinogen at the interface. Questions surrounding the possibility of fibrinogen adsorption warrant further investigation, as the location of procoagulant proteins at a pe

A more quantitative understanding of peptide loading and release from polyethylene oxide (PEO) brush layers will provide direction for development of new strategies for drug storage and delivery. The antimicrobial peptide nisin shows potent activity against Gram-positive bacteria including the most prevalent implant-associated pathogens, its mechanism of action minimizes the opportunity for the rise of resistant bacteria and it does not appear to be toxic to humans, suggesting good potential for its use in antibacterial coatings for selected medical devices. Abstract:
Cavity ring-down spectroscopy has been used over the last twenty years as a highly sensitive absorption spectroscopic technique to measure light attenuation in gases, liquids, and solid samples. An optical cavity is used as a multi-pass cell, and the decay time of the light intensity in the cavity is measured, thereby rendering the techniques insensitive to light intensity fluctuations. Optical waveguides are used to build the optical cavities presented in this work. The geometries of such waveguides permit the use of very small liquid sample volumes while retaining the advantages of cavity ri
