Meiothermus pingtungensis sp. nov. I40 (I40) isolated from hot spring is a newly identified bacterium, and is able to degrade the feather keratin. The objective of this study is to determine the optimal medium compositions and incubation condition, for the production of keratinase maximal using Plackett-Burman design and response surface methodology (RSM). The degradation rates of feather by I40, was almost disintegrated under the 6 days incubation of I40 at 55˚С. The crude keratinase was isolated from the centrifuged and used in the following studies. The optimum pH for crude keratinolytic activity was at 8.5; optimum temperature at 70˚С. The crude enzyme was stable under 70˚С for 1 h, and the enzyme showed 18% activity at 80˚С for 1 h. Storage of the crude keratinase for 10 days, the enzyme exhibited more than 80% activity at -20 or -80˚С, whereas 60 % activity at 4˚С. Moreover, of the enzyme showed an apparent molecular mass at 243 and 172 KDa on SDS-PAGE and activity staining analysis. Nine medium compositions were used to study their effect on I40 using Plackett-Burman design. The production of biomass for I40 was affected in order of yeast extract, tryptone, glucose, and production of protease affected in order of feather, tryptone and yeast extract. According to the above results, feather, tryptone, yeast extract as well as incubation time and temperature were selected for further studies by RSM. Data obtained showed the maximal biomass production and keratinase activity occurred at above 96 h. From the commercial consideration, the incubation time was fixed at 3 days and reinvestigated by RSM. The maximal keratinase activity was obtained from the following condition: tryptone 0.16%, yeast extract 0.27%, feather 0.08%, incubation temperature 51.7˚С. The keratinase activity 20 U/mL obtained from RSM study was 13.3 folds higher than that from the original incubation condition, and the incubation time shorten from 5 days to 2 days.

abstract The production processes of A356 Al-wheels in a specific foundry was evaluated in this study. The process standardization was proposed for that specific foundry based upon the evaluation results. A rotary impeller degasser is currently used for the degassing treatment in the foundry. The degassed melt contained commercially accepted low gas level. However, the gas content of the melt increased gradually with increasing holding time, and the gas content reached an unacceptable level after holding for over 90 minutes. The microstructures of the Al-wheels currently produced in the foundry were thoroughly analyzed with the results indicating that substantial shrinkage porosity was present in the central portion of the rim section of the Al-wheels. Failing to maintain a directional solidification pattern and/or high enough temperature gradients were found to be responsible for the formation of the porosity defect. Modifying the ingate design and employing forced cooling at the central rim section were suggested for the elimination of this porosity defect. Furthermore, the feasibility of reducing the section thickness of the as-cast Al-wheels was evaluated in this study by using tensile and impact properties as the criteria. The results show that the original thickness of rim section of 10- 15㎜ can be reduced to around 9㎜. Also, accompanying with the reduction of section thickness, the T6 solution time at 540℃ can be shortened to 2-4hr from original 6hr and still obtains improved properties.

The aim of this research is to establish the optimal processing parameters which are required for manufacturingsound EPC A356 brake-oil-pump housing castings. The processing parameters studied include the gating system design, the pattern material, the coating thickness, the GFN of molding sand , the application of a vacuum and the pouring temperature. The optimal conditions have been respectively established for various gating systems ranging from one layer-one casting system (Type A) to tow-layer-six casting system (Type E). The mold filling behavior of the gating system Type E has been thoroughly analyzed, with the results indicating that though the metal flow follows essentially a sequential pattern, the majority of the first incoming melt (approximately 70%) enters the lower layer. This is due primarily to the relatively large metallostatic head and the cross sectional area of the sprue used in this study. In addition, the previously derived EPC Al flow-solidification equation has been slighly to modified to calculate theoreticlly the flow length of a pipe-shape casting, similar to the pump housing casting studied herein. The calculated flow lengths are in agreement with the experimental results. The relationship between the %porosity and the processing parameters has also been investigated. The results show that raising the pouring temperature increases the porostiy content. Also, employing PMMA pattern material results in less porosity than adopting EPS pattern material. Howwver, changes in gating system design and GFN of the molding sand have little effect on porosity content. Microstructural analyses show that the as-cast eutectic Si exhibits a partial modification structure (3-4 modification degree). This is due to the presence of about 0.026%Sr in the A356 ingots used and the relatively thin section of the casting studied. The SDAS is affected mainly by the local solidification cooling rate of the castings. Two principal types of porosity porosity defects were observed in the castings, namely the pyrolysis porosity defects caused by the decomposing pattern and the microshrinkage defect.
