Abstract Purpose To determine the exposure–response (ER) relationships between atezolizumab exposure and efficacy or safety in patients with advanced non-small cell lung cancer (NSCLC) or urothelial carcinoma (UC) and to identify alternative dosing regimens. Methods ER analyses were conducted using pooled NSCLC and UC data from phase 1 and 3 studies (PCD4989g, OAK, IMvigor211; ClinicalTrials.gov IDs, NCT01375842, NCT02008227, and NCT02302807, respectively). Objective response rate, overall survival, and adverse events were evaluated vs pharmacokinetic (PK) metrics. Population PK-simulated exposures for regimens of 840 mg every 2 weeks (q2w) and 1680 mg every 4 weeks (q4w) were compared with the approved regimen of 1200 mg every 3 weeks (q3w) and the maximum assessed dose (MAD; 20 mg/kg q3w). Phase 3 IMpassion130 (NCT02425891) data were used to validate the PK simulations for 840 mg q2w. Observed safety data were evaluated by exposure and body weight subgroups. Results No significant ER relationships were observed for safety or efficacy. Predicted exposures for 840 mg q2w and 1680 mg q4w were comparable to 1200 mg q3w and the MAD and consistent with observed PK data from IMpassion130. Observed safety was similar between patients with a Cmax above and below the predicted Cmax for 1680 mg q4w and between patients in the lowest and upper 3 body weight quartiles. Conclusion Atezolizumab regimens of 840 mg q2w and 1680 mg q4w are expected to have comparable efficacy and safety as the approved regimen of 1200 mg q3w, supporting their interchangeable use and offering patients greater flexibility.

Introduction: Bitter tasting of drugs leads to non-compliance especially in the case of pediatric patients due to their inability to swal-low medication.&nbsp; Aim: In this study, we aimed to mask the bitter taste of acetaminophen (APAP) particles through coating.&nbsp; Materials and methods: A pH independent water insoluble ethylcellulose polymer was used to coat the APAP. The coating of water insoluble ethylcellulose on APAP can have a significant impact on the dissolution profile. Various grades of APAP were used for coating; fine grade, Compap L90% having wide particle size distribution (PSD), and a special granular (SG) APAP 1680 having narrow PSD. Coating was performed using top spray (Vector) for Compap L90% and SG APAP 1680 grade of APAP.&nbsp; Results: Bitter taste of SG APAP was masked after spraying dispersion equivalent to a weight gain of 10% compared to 35% used for Compap L90%. Using bottom spray (Wurster coater, GPCP 2.0), coating was performed on SG APAP 1680 grade of APAP by spraying aqueous dispersion of ethylcellulose (Surelease) equivalent to a weight gain of 10%. The scalability of the top spray process was also evaluated in GPCG 30 and bitter taste was masked by using Surelease dispersion equivalent to a weight gain of 6%. Coated APAP was examined for particle size (PS), particle size distribution (PSD), flowability, and drug release profile. Dissolution was performed using USP apparatus 2 and 4 in phosphate buffer and evaluated for mechanism of drug release. Particle size obtained for coated SG APAP 1680 via top and bottom spray process was 404 &micro;m d(90) and 487 &micro;m d(90) respectively.&nbsp; Conclusions: The results of the study demonstrated the potential of Surelease dispersion in taste masking. The use of SG APAP 1680 having narrow PSD allowed taste masking to achieve at low weight gain without greatly affecting the dissolution profile.

Abstract:
The objective of the present work was to study the serological status and epidemiological situation of porcine teschovirus 1 (PTV-1) on swine farms in different Lithuanian regions by using virus neutralization test and virus isolation. Clinical, epidemiological, serological and virological examinations were performed for the diagnosis of Teschen disease (porcine enterovirus encephalomyelitis, TE). Swine blood serum samples (n = 1680) were collected on 28 farms (60 samples per farm) of 8 regions in 2003. For virus isolation 29 samples of pathological material were taken from the brains of pigs (2 - 4 months of age) that died suddenly on seven swine farms in five regions. Epidemiological and clinical examination revealed no signs of porcine enterovirus encephalomyelitis on 32 Lithuanian swine farms. PTV-1 was not isolated and identified in the PK-15 cell culture. Antibodies against PTV-1, detected by using VNT, were found on all investigated farms and in all age groups. Negative serum (2.3%, 39 from 1680) samples were found in 15.5% (16 from 103) of 2-4-month-old pigs and in 4.7% (23 from 494) of 4-6-month-old ones. The positive serological and negative epidemiological, clinical and virological results suggest that less virulent or avirulent PTV-1 strains were spread on Lithuanian swine farms.

AbstractThis study throws new light on the composition of Boyle's Free Enquiry into the Vulgarly Receiv'd Notion of Nature (1686); it also draws more general conclusions about Boyle's methods as an author and his links with his context. Its basis is a careful study of the extant manuscript drafts for the work, and their relationship with the published editions. Section 2 describes Boyle's characteristic method of composition from the late 1650s onwards, involving the dictation of discrete sections of text to amanuenses; it also assesses the effect this had on the structure and presentation of Boyle's writings. Section 3 considers the published text section by section and indicates which parts were written when; it also surveys unpublished draft material relating to the work. Section 4 places the work in context, considering the intellectual threats that Boyle sought to confront in it, both when he initially composed it in the 1660s and when he rewrote it c. 1680. It thus anchors him more precisely than hitherto in the intellectual debates of his day.

Field studies were conducted in Kansas and Maryland to compare the safety and efficacy of halosulfuron-methyl (HM) and bentazon for topkill of yellow nutsedge (Cyperus esculentus L.). Kentucky bluegrass (Poa pratensis L.) and creeping bentgrass (Agrostis palustris Huds.) treated with single (in Kansas) or sequential (in Maryland) HM (35 to 140 g·ha–1) or bentazon (1120 or 1680 g·ha–1) applications exhibited little injury, and treated turf had acceptable quality in all studies. Bentazon caused an unacceptable reduction in perennial ryegrass (Lolium perenne L.) quality at ≥5 weeks after treatment in four of five tests. Perennial ryegrass quality declined linearly with increasing HM rates (between 35 and 140 g·ha–1). In Maryland, HM (≥70 g·ha–1) elicited unacceptable perennial ryegrass quality for 2 or 3 weeks; however, in Kansas, quality was unacceptable for ≈1 week. In Kansas, yellow nutsedge topkill by HM (70 kg·ha–1) ranged from 52% to 97%. A single HM application (35, 70, or 140 kg·ha–1) provided > 97% topkill in Maryland. Yellow nutsedge topkill by bentazon (1680 g·ha–1) generally was inferior to that by HM (70 g·ha–1). ABSTRACT Objective: To compare the initial failure rate (≤4 months) for extra-alveolar mandibular buccal shelf (MBS) miniscrews placed in movable mucosa (MM) or attached gingiva (AG). Materials and Methods: A total of 1680 consecutive stainless steel (SS) 2 × 12-mm MBS miniscrews were placed in 840 patients (405 males and 435 females; mean age, 16 ± 5 years). All screws were placed lateral to the alveolar process and buccal to the lower first and second molar roots. The screw heads were at least 5 mm superior to the soft tissue. Loads from 8 oz–14 oz (227 g–397 g, 231–405 cN) were used to retract the mandibular buccal segments for at least 4 months. Results: Overall, 121 miniscrews out of 1680 (7.2%) failed: 7.31% were in MM and 6.85% were in AG (statistically insignificant difference). Failures were unilateral in 89 patients and bilateral in 16. Left side (9.29%) failures was significantly greater (P &lt; .001) compared with those on the right (5.12%). Average age for failure patients was 14 ± 3 years. Conclusion: MBS miniscrews were highly successful (approximately 93%), but there was no significant difference between placement in MM or AG. Failures were more common on the patient's left side and in younger adolescent patients. Having 16 patients with bilateral failures suggests that a small fraction of patients (1.9%) are predisposed to failure with this method.

Abstract:
Thrombin-catalyzed activation of factor (F)VIII by proteolytic cleavages at Arg372, Arg740, and Arg1689 is essential for the propagation phase of blood coagulation cascade. Activated FVIII (FVIIIa) forms the tenase complex and markedly amplifies FX activation as a cofactor of FIXa. We previously reported that thrombin interacts with FVIII through the A2 domain (residues 392-394 and 484-509) and C2 domain, and these interactions governed the cleavages at Arg740, Arg372 and Arg1689, respectively (Nogami, JBC 2000, 2005, BJH 2008). A previous report suggested, however, that FVIII lacking the C2 domain retained >50% cofactor activity (Wakabayashi, JBC 2010), supportive of the presence of other thrombin-binding site responsible for cleavage at Arg1689 within the A3-C1 domain. Recently, we focused on similar sequence in A3 acidic region to hirugen residues 54-65, and demonstrated that two region of residues 1659-1669 and 1675-1685 might contained the thrombin-binding site(s) responsible for cleavage at Arg1689 by functional and binding experiments using synthetic peptide (Minami. ASH 2015). In this study, to identify the crucial residues, seven acidic clustered residues and two sulfated Tyr residues in these regions as a series of rFVIII mutants were converted to Ala, and 9 single mutants for D1663A, Y1664A, D1665A, D1666A, Y1680A, D1681A, E1682A, D1683A, E1684A and 2 double mutants for D1665A/D1666A, D1683A/E1684A, were prepared by using a BHK system. Specific activities of FVIII in all of 5 mutants (D1663A, Y1664A, D1665A, D1666A, D1665/D1664) in the former region and 4 mutants (Y1680A, D1683A, E1684A, D1683A/E1684A) in the latter region, assessed by a one-stage clotting assay were 40-70% and 50-70% of wild type (WT), respectively. These mutants exhibited the assay discrepancy for FVIII activity between one-stage clotting assay and FVIIIa-dependent FXa generation assay (one-stage < FXa generation assay), guessing possible association with these mutated residues for thrombin reaction. Next, FVIII mutants (10 nM) were examined for activation by thrombin (0.4 nM) in a one-stage clotting assay. Regards FVIII mutants in the 1663-1666 region, FVIII activation by thrombin in all mutants were not significant difference from that in WT. We further examined thrombin-catalyzed cleavage at Arg1689 of these mutants by using SDS-PAGE and Western blot using an anti-FVIII monoclonal antibody recognizing the A3 acidic region for detection. The initial velocity of thrombin cleavage at Arg1689 in these mutants showed no significant difference from that in WT. In addition, the rate of thrombin cleavage at Arg372 in them was also almost similar to that in WT. These results indicated that the 1663-1666 region appeared unlikely to participate in the functional association between FVIII and thrombin. On the other hand, regards the 1680-1684 region, peak activity in FVIII activation by thrombin for D1683A, E1684A and D1683A/E1684A mutants were modestly reduced with an ~60% level of WT, and that for Y1680A mutant was significantly diminished with peak activity of ~30% of WT. FVIII activation by thrombin in D1681A and E1682A mutants was not significant difference from WT, however. Evaluated by SDS-PAGE, the initial velocities of cleavage at Arg1689 of D1681A and E1682A were comparable to WT, whilst those of D1683A, E1684A and D1683A/E1684A mutants were ~60% level of WT. It was note that Y1680A mutant showed a ~10% of velocity rates of WT on the cleavage at Arg1689, supportive of the results obtained from thrombin-catalyzed activation of FVIII. However, the rate of thrombin cleavage at Arg372 in these mutants was almost similar to that in WT. These results suggested that three residues were involved with the cleavage at Arg1689. In conclusion, we for the first time identified that that 1680-1684 residues in A3 acidic region, in particular sulfated Tyr1680, played a key role in thrombin-interactive sites responsible for cleavage at Arg1689. [VCl2(15-crown-5)]+[VOCl4]- has been prepared by the reaction of VCl4 with 15-crown-5 in acetonitrile solution in the presence of water, forming black-brown single crystals. They were characterized by IR spectroscopy as well as by an X-ray structure determination. Space group Pnma, Z = 4, 1530 observed unique reflexions, R = 0.052. Lattice dimensions at -120°C: a = 1128.5(4), b = 1063.3(3), c = 1680(1) pm. The compound forms ions, in which the cation contains vanadium(+III), which is seven coordinate by the five oxygen atoms of the crown ether molecule and by two chlorine atoms in axial positions of a pentagonal bipyramide. With four-probe and laser light scattering techniques, we have studied the dependencies of the conductivity and the particle sizes of p-toluene sulfonic acid doped polypyrrole on the mechanical grinding time. As the chemically synthesized polypyrrole was mechanically grounded in a mortar from 0 to 8 hours, its conductivity decreased from 607 to 10 S/m while and its particle size decreased from 27 to 4 m. If the mechanical grinding continued, the conductivity of the polymer was found to be up to 1680 S/m. Our results have demonstrated that mechanical grinding is an effective method to control the conductivity of the p-toluene sulfonic acid doped polypyrrole.

Nonsingular projective 3-folds $V$ of general type can be naturally classified into 18 families according to the pluricanonical section index${\it\delta}(V):=\text{min}\{m\mid P_{m}\geqslant 2\}$ since $1\leqslant {\it\delta}(V)\leqslant 18$ due to our previous series (I, II). Based on our further classification to 3-folds with ${\it\delta}(V)\geqslant 13$ and an intensive geometrical investigation to those with ${\it\delta}(V)\leqslant 12$, we prove that $\text{Vol}(V)\geqslant \frac{1}{1680}$ and that the pluricanonical map ${\rm\Phi}_{m}$ is birational for all $m\geqslant 61$, which greatly improves known results. An optimal birationality of ${\rm\Phi}_{m}$ for the case ${\it\delta}(V)=2$ is obtained. As an effective application, we study projective 4-folds of general type with $p_{g}\geqslant 2$ in the last section.

Iznošenje argumenata koji ukazuju na potrebu ukidanja ispita iz matematike na osnovnoj razini koji se rješava na državnoj maturi. S maturantima se u 12 godina obrazovanja ostvaruje 1120 sati nastave matematike u osnovnoj školi i 420 ili 560 sati u srednjoj školi (ovisno je li se sati odnose na 3 ili 4 sata nastave matematike tjedno), dakle ukupno 1540 ili 1680 sati nastave matematike. Nema potrebe da se elementarnim zadatcima, većinom iz programa matematike osnovne škole ispituje znanje maturanata. Predlaže se jedinstveni ispit matematike na državnoj maturi. Posebno se ukazuje na problem ocjenjivanja ispita iz matematike na državnoj maturi, niske pragove za pojedinu ocjenu (odstupanje od normalne raspodjele). Predlaže se uz ocjenu i evidentiranje postotne rješivosti zadataka. Zabrinjavajuće su ocjene učenika na ispitu iz matematike ostvarene na državnoj maturi školske godine 2017./2018. u ljetnom roku.

Perubahan sumber energi matahari dengan memanfaatkan panel surya menjadi sumber energi listrik yang terbarukan merupakan upaya untuk mengurangi konsumsi bahan bakar fosil, panel surya memiliki kelebihan yaitu mampu mengurangi polusi dan hemat energi jika diaplikasikan pada mobil listrik. Penelitian ini bertujuan untuk mendapatkan hasil pengukuran arus dan tegangan dari panel surya serta baterai sebagai sumber listrik untuk pengisian tenaga pada panel surya. Sistem panel surya mencakup 4 panel surya 50 Wp, 1 battray charge unit, 4 baterai 35 Ah, dan 1 motor listrik yang terpasang dalam mobil listrik. Dari penelitian yang dilakukan perancangan panel surya pada mobil listrik dengan kemiringan 45 agar nantinya panel surya mendapat cahaya matahari yang lebih besar. Maka dapat diperoleh hasil yaitu pengukuran dari pukul 07:00 – 17:00 dan nilai output panel surya ± 60.00 Volt dan Arus ± 23.00 A (fluktuasi nilai tegangan dan arus akan berubah berdasarkan waktu, cuaca dan lingkungan sekitar) sehingga daya yang dihasilkan pada ± 10720,1 Watt, lama waktu pemakaian beban dari motor litrik daya yang dihasilakan dibagi dengan beban mobil listrik 10720,1 watt / 1680 watt maka akan mendapat 6,4 jam pemakaian mobil listrik.

The following problems are solved. The pressure profiles at the sample-soft barrier interface in the spall experiments with flat one-dimensional loading are measured. The equations of state of three polymorphic modifications of hafnium are constructed in the pressure range up to 100 GPa. The thermodynamic states of hafnium under the conditions of the performed experiments were calculated. These calculations, together with the results of measurements of pressure profiles, made it possible to determine the spall strength of hafnium heated in a shock compression-unloading cycle. The spall strength of hafnium was -4 (1) GPa at a temperature of 1680 (380) K. Mathematical modeling of spall experiments in the framework of a one-dimensional hydrocode was performed.

Abstract Following exposure to cytotoxic agents, acute myeloid leukemia (AML) blasts elevate cellular cholesterol in a defensive adaptation that increases chemoresistance, but blockade of HMG-CoA reductase with statins restores chemosensitivity in vitro. This phase 1 study evaluated adding pravastatin (PV) (40-1680 mg/day, days 1-8) to idarubicin (Ida) ([12 mg/(M2 · day), days 4-6]) + high-dose cytarabine (Ara-C; HDAC) [1.5 g/(M2 · day) by CI, days 4-7] in 15 newly diagnosed and 22 salvage patients with unfavorable (n = 26) or intermediate (n = 10) prognosis cytogenetics. Compared with historical experience with Ida-HDAC, the duration of neutropenia and throbmbocytopenia and the toxicity profile were unaffected by the addition of PV. During PV loading (day 0-4) serum triglyceride and total and LDL cholesterol levels decreased in nearly all patients. Pharmacokinetic studies demonstrated higher and more sustained serum PV levels with PV doses above 1280 mg/day. CR/CRp was obtained in 11 of 15 new patients, including 8 of 10 with unfavorable cytogenetics, and 9 of 22 salvage patients. An MTD for PV + Ida-HDAC was not reached. Addition of PV to Ida-HDAC was safe, and the encouraging response rates support conducting further trials evaluating the effect of cholesterol modulation on response in AML.
