Este projeto teve como principal objetivo o estudo de aspectos químicos, biológicos e biossintéticos dos metabólitos secundários de duas espécies de Celastraceae, Maytenus ilicifolia e Salacia campestris. O estudo fitoquímico das cascas das raízes das duas espécies selecionadas resultou no isolamento de oito substâncias pertencentes a três diferentes classes de metabólitos secundários, incluindo os triterpenos quinonametídeos, maitenina e pristimerina; os alcalóides piridínicos sesquiterpênicos, aquifoliunina E1, maiteina, wilforina, sendo dois inéditos, maiteinol e campestrina; além de uma catequina, ourateacatequina. Os extratos, frações e substâncias puras foram avaliados quanto a atividade antifúngica, frente aos patógenos humanos Candida albicans, C. kruseii, C. parapsilosis e Cryptococcus neoformans; tripanocida, frente a cepa Y da forma epimastigota do Trypanossoma cruzi; leishmanicida, frente a formas promastigotas de L. amazonensis e L. chagasis e antitumoral avaliado por testes de viabilidade metabólica usando três tipos de células tumorais, incluindo B16 (melanoma), HeLA (colo uterino) e U343 (astrocitoma cerebral). Em todas as matrizes biológicas avaliadas, os triterpenos quinonametídeos, maitenina e pristimerina, se mostraram os mais potentes, evidenciando atividade biológica superior aos controles positivos utilizados. Além dos triterpenos quinonametídicos, os alcalóides piridínicos sesquiterpênicos, aquifoliunina E1 e maiteinol se mostraram ativos nos testes leishmanicida e tripanocida, respectivamente, mas menos potentes que os triterpenos quinonametídeos. Experimentos para avaliar a biossíntese das etapas de oxidação que ocorrem no anel E dos triperpenos quinonametídeos foram baseados em protocolos para a extração da enzima oxidoredutase do tipo citocromo P450, supostamente envolvida nas referidas etapas de oxidação...
The project was addressed to investigate chemical, biological and biosynthetic aspects of major secondary metabolites from two Celastraceae species, Maytenus ilicifolia (espinheira-Santa) and Salacia campestris. The phytochemical study of the bark roots resulted in the isolation of eight compounds belonging to three different classes of natural products including the quinonemethides triterpenes, maytenin and pristimerin; the alkaloids pyridine sesquiterpenes, aquifoliunine E1, mayteine, wilforine, including two novel, mayteinol and campestrine; in addition to one catechin, ourateacatechin. The localization of quinonemethides triterpenes in roots of M. ilicifolia was achieved by histochemical analysis based on complexation reactions using some specific reagents to identify groups or organic functions in the interested molecule. The crude extracts, fractions and pure compounds had their antifungal properties assayed against the human pathogens Candida albicans, C. kruseii, C. parapsilosis and Cryptococcus neoformans. Additionally, their anti-parasite activity were assessed against Trypanossoma cruzi (epimastigote form of Y strain), L. amazonensis and L. chagasis (promastigote forms) while the antitumoral activity was based on metabolic viability metabolic using two kinds of tumor cells, B16 (melanoma), HeLA (uterus) e U343 (cerebral astrocytoma). The quinonemethides triterpenes, maytenin and pristimerin displayed considerable potential since were more active than controls. The alkaloids pyridine sesquiterpenes, aquifoliunine E1 and mayteinol revealed to have some potential as leishmanicidal and trypanocide, respectively, but not comparable to quinonemethides triterpenes. As the biosynthesis of quinonemethides triterpenes is concerned, the oxidation step at ring E was investigated considering the participation of cytochrome P450 oxidoreductase besides enzymatic assays using pristimerin... Full textA chamada modernização da agricultura representou um período de intensificação do processo de degradação ambiental e modificação profunda nas relações de trabalho.Um dos movimentos de resistência a esse novo padrão de produção verticalizada é a agricultura orgânica. No Estado de Santa Catarina, essas experiências têm se desenvolvido amplamente, assumindo características próprias e gerando uma diversidade de relações sociais que tem deflagrado novos movimentos territoriais. No caso chapecoense, a agricultura orgânica desenvolve-se amparada por agricultores, ONG s e Cooperativas, criando uma rede territorial que assume ritmos e tempos diferentes, mas que tem construído um novo território nesse município. Novo justamente porque pensa e organiza os elementos do espaço geográfico seguindo um padrão próprio, construído coletivamente e produto da diversidade cultural, política, econômica e ambiental local. Novas redes e fluxos foram ativados inserindo esses atores sociais de uma forma singular no contexto local. Buscando compreender essas relações lançou-se mão de revisão bibliográfica, revisão de sítios especializados, entrevistas, trabalhos de campo, aplicações de questionários, entre outros, que possibilitaram a coleta de dados para posterior compilação e apresentação em forma de mapas temáticos, tabelas, gráficos e conclusões teóricas a respeito da agricultura orgânica enquanto movimento territorial no município de Chapecó, Oeste catarinense.
The so-called modernization of agriculture exemplified a period of intensification of the process of environmental degradation and profound changes in work relations. One of the resistance movements to this new standard of verticalized production is organic agriculture. In the state of Santa Catarina, these experiments have developed widely taking on their own characteristics producing a variety of social relations that have evolved into new territorial movements. In the case of Chapecó, organic agriculture has developed supported by agriculturalists, NGOs and cooperatives, creating a territorial network that has taken on different times and rhythms, but which has built a new territory in this municipality. It is new exactly because it thinks and organizes the elements of geographic space following its own standard, building collectively, and is a product of cultural, political, economical and local environmental diversity. New networks and currents were activated, inserting these social actors in a singular manner in the local context. Seeking to comprehend these relations, bibliographic revision, specialized farm revisions, interviews, field work, use of questionnaires, among others were employed, which allowed for information gathering for later compilation and presentation in the form of thematic maps, tables, graphics and theoretical conclusions regarding organic agriculture as a territorial movement in the municipality of Chapecó in the western region of Santa Catarina.

Três espécies de Piper foram escolhidas como modelos para estudos biossintéticos, transcriptômicos e proteômicos: P. tuberculatum, P. nigrum e P. crassinervium. As duas primeiras espécies acumulam principalmente metabólitos secundários da classe das amidas e essas substâncias foram alvo de experimentos biossintéticos. Foram monitoradas as atividades de diversas enzimas envolvidas na via fenilpropanoídica, dentre elas a fenilalanina amônia liase (PAL) e a cinamato 4-hidroxilase (C4H). Observou-se também atividade de uma enzima que catalisa a conversão do ácido ferúlico ao ácido 3,4-metilenodioxicinâmico, denominada 3,4- metilenodioxicinâmico sintase. Esta é provavelmente uma enzima monooxigenase-dependente do citocromo P450. A principal questão biossintética está centrada na sequência na qual as reações ocorrem: o grupo metilenodioxílico pode ser formado antes ou depois da porção amídica. Para elucidar esta questão, sintetizou-se dois possíveis precursores contendo o grupo 4-hidróxi-3- metóxifenílico: o ácido ferupérico e a feruperina. No entanto, experimentos de atividade enzimática in vitro utilizando estes dois precursores não foram esclarecedores, devido a existência de piperina endógena no extrato enzimático proveniente de folhas de P. nigrum, o que impediu observar a formação de produto. Foi avaliado também a participação de ácido malônico e L-valina na biossintese destas amidas: incorporou-se precursores isotopicamente marcados em plântulas de P. tuberculatum: ácido 2-[13C]-malônico, ácido malônico-U-2H e 15N-L-valina). Após análise por CLAEEM, conclui-se que o ácido malônico participa da reação de condensação com fenilpropanóides, gerando os derivados C6-C5 e L-valina é precursor do grupo isobutilamina. Three Piper species were chosen as a models for biosynthetic, transcriptomic and proteomics studies: P. tuberculatum, P. nigrum and P. crassinervium. The first two species accumulate mainly amides as secondary metabolites. This class of compounds was targeted by biosynthetic experiments. It was monitored the enzymatic activities of several enzymes involved in the phenylpropanoid pathway, including phenylalanine ammonia-lyase and cinnamate 4-hydroxylase. It was also observed the activity of an enzyme that catalyzes the conversion of ferulic acid to 3,4- methylenedioxycinnamic acid, named 3,4 methylenedioxycinnamic acid synthase. The main question was centered on the biosynthetic sequence: whether the methylenedioxy group can be formed before or after the amide portion. In order to elucidate this question, two possible precursors containing the group 4-hydroxy-3-methoxyphenyl were synthesized: feruperic acid and feruperine. However, experiments of in vitro enzymatic activity using these two precursors were not enlightening, due to the existence of piperine as endogenous in the enzymatic extract from leaves of P. nigrum, which prevented observe the formation of product. It was also evaluate the participation of malonic acid and L-valine in the biosynthesis of these amides, by incorporating isotopically labeled precursors in plants of P. tuberculatum: 2-[13C]-malonic acid, malonic-U-2H acid and 15N-Lvaline. After analysis by HPLC-MS, it was concluded that the malonic acid participates in the condensation reaction with phenylpropanoids, generating the C6-C5 derivatives and L-valine is the putative precursor of the isobutylamine group. Full textA abóbora (Cucurbita moschata Dusch), planta de origem americana, é cultivada em diversos estados do Brasil, principalmente por agricultores familiares que, muitas vezes, produzem suas próprias sementes. Porém, estes agricultores nem sempre aproveitam a variabilidade existente para selecionar as melhores plantas. O objetivo desta pesquisa foi avaliar a produção e a qualidade dos frutos de populações de abóbora obtida a partir de melhoramento participativo e método da seleção massal estratificada em três ciclos de seleção, em uma propriedade biodinâmica familiar no município de Botucatu-SP. Apenas no segundo e terceiro ciclo foi realizada emasculação das plantas fora do padrão comercial através de seleção antes do florescimento. Foram realizados dois experimentos comparando a população original e três ciclos de seleção, com semeaduras em setembro/2009 e agosto/2010. Os ensaios foram realizados em delineamento experimental de blocos casualizados, com sete repetições. Foram avaliadas as seguintes características: número de frutos por planta (total, comercial e não comercial), percentagem de frutos comerciais e não comerciais, massa média por fruto, massa dos frutos por planta (total, comercial e não comercial), comprimento do fruto (total, pescoço e bojo), diâmetro do fruto (pescoço e bojo), relação entre comprimento e diâmetro do bojo e do pescoço, comprimento total médio/diâmetro médio, comprimento do pescoço/comprimento do bojo e diâmetro do pescoço/diâmetro do bojo. The squash (Cucurbita moschata Dusch), plant of American origin, is cultivated in many states in Brazil, mainly by familiar farmers. There is a great biodiversity of genetic resource that is not select by this farmers. The aim of this work was to evaluate the yield and the quality of fruits of squash population obtained with the participatory breeding using stratifeid mass selection method in three generations cycles, in a biodynamic familiar farm in the Botucatu city in São Paulo state-Brazil. Only at the second and third cycles was done plant emasculation of plants with no commercial fruits, through the selection before flowering. There were two experiments comparing the original population and three cycles of selection, one in September (2009) and another in November (2009). These assays were conducted in randomized blocks experimental design, with seven replications. It was evaluated the following traits: number of fruits per plant (total, commercial and not commercial); percentage of commercial fruits and not commercial fruits per plant; average weight of fruits; weight of fruits per plant (total. commercial and not commercial ); total length of the neck and the bulge, diameter of the neck and the bulge; ratio of length to diameter of the neck and the bulge; ratio of total length with an average diameter; ratio of the length of the neck to the length of the bulge and ratio of the diameter of the neck to the diameter of the bulge. The results indicate an increase in the frequency of plants with commercial fruits, and show that the practice of participatory breeding and stratified mass selection is feasible for these conditions.
