Abstract:
O objetivo deste trabalho foi a seleção de bactérias capazes de biodegradar os pesticidas organoclorados dieldrin, DDD e PCP. Inicialmente, foram realizados os ensaios de tolerância visando à seleção das bactérias degradadoras; posteriormente foram realizados os ensaios de biodegradação em meio liquido utilizando a bactéria selecionada. Dentre as 14 linhagens bacterianas isoladas testadas, selecionou-se a linhagem Pseudomonas aeruginosa L2-1 por apresentar maior tolerância a todos os pesticidas. Os ensaios de biodegradação foram realizados em diferentes meios de cultura, variando-se a concentração de glicose, a fonte de nitrogênio e a presença de ramnolipídeo. Os ensaios de biodegradação foram realizados determinando-se a concentração de pesticida, a concentração de glicose, o número de células viáveis, e o pH. O meio de cultura que mais favoreceu a biodegradação dos três pesticidas foi o meio com nitrato de sódio e 0,5% de glicose, obtendo-se biodegradação de aproximadamente 50% para cada pesticida após três dias de incubação. Na ausência de glicose, o meio com nitrato de amônio e 0,1% de ramnolipídeo, favoreceu a biodegradação, obtendo-se após 14 dias de incubação 36,8% de biodegradação de dieldrin; 33,7% de DDD e 42,8% de PCP. De uma forma geral, as taxas de biodegradação pela P. aeruginosa L2-1 foram maiores em menores concentrações de glicose e na presença de ramnolipídeo. Ao alterar a fonte de nitrogênio foram observados resultados diversos sobre a taxa de biodegradação: na ausência de glicose, o nitrato de sódio favoreceu a biodegradação de PCP, enquanto o nitrato de amônio favoreceu a biodegradação de dieldrin, na presença de glicose observou-se o inverso. A taxa de biodegradação do DDD não foi significativamente alterada ao variar a fonte de nitrogênio. A bactéria selecionada P. aeruginosa L2-1 apresentou potencial para biodegradação de pesticidas organoclorados, sendo que as condições nutricionais do meio influenciaram diretamente a biodegradação.
The objective of this work was the selection of bacteria capable of biodegrading the organochloride pesticides dieldrin, DDD and PCP. Initially tolerance tests were conducted in order to select degrading bacteria subsequently, biodegradation tests were carried out in liquid medium using the selected bacteria. Among the 14 bacterial isolated strains, Pseudomonas aeruginosa L2-1 was selected due to its greater tolerance to all pesticides. The biodegradation tests were conducted on different culture media, varying the concentrations of glucose, nitrogen source and presence of rhamnolipid. Biodegradation studies were performed by measuring the concentration of pesticide, the concentration of glucose, the number of viable cell and pH during time. The best medium for the biodegradation of all three pesticides was composed of sodium nitrate and 0.5% glucose, giving approximately 50% yield after three days of incubation. In the absence of glucose, the medium containing ammonium nitrate and 0.1% rhamnolipid improved biodegradation yielding, after 14 days of incubation, 36.8% biodegradation of dieldrin; 33.7% DDD and 42.8% of PCP. In general, the biodegradation rates of pesticides by P. aeruginosa L2-1 were greater at lower concentrations of glucose and in the presence of rhamnolipid. Nitrogen source had different effects on the rate of biodegradation: in the absence of glucose, sodium nitrate favored the biodegradation of PCP, whereas ammonium nitrate favored the biodegradation of dieldrin; and in the presence of glucose, it was observed the opposite. The biodegradation rate of the DDD was not significantly altered by the nitrogen source tested. The selected bacteria, P. aeruginosa L2-1, showed potential for the biodegradation of organochloride pesticides demonstrating that nutritional conditions has a direct effect on degradation yields.

Abstract:
Os compostos organoclorados causam grande impacto na natureza devido a três características básicas: persistência ambiental, bioacumulação e alta toxicidade. Os mamíferos marinhos estão entre os organismos mais vulneráveis à toxicidade crônica desses contaminantes porque, além de concentrá-los em grande quantidade, a fêmea transfere parte de sua carga ao filhote durante a gestação e a lactação. Assim, o presente trabalho teve como objetivos otimizar uma metodologia para determinação de organoclorados (pesticidas e PCBs) em matrizes gordurosas e verificar a ocorrência dos mesmos na gordura subcutânea de mamíferos marinhos amostrados na costa de São Paulo (Brasil) e na Ilha Rei George (Antártica). No protocolo metodológico otimizado, a extração foi realizada em extrator Soxhlet (8 h) com uma mistura de n-hexano e diclorometano. A etapa de purificação foi feita através de tratamento ácido e o extrato final analisado em cromatógrafo a gás equipado com detector de captura de elétrons (GC-ECD). A performance do método foi avaliada com material de referência certificado, enquadrando-se dentro de padrões internacionais de controle de qualidade. O limite de detecção do método foi em média 2 ng g-1. As análises apontaram DDTs e PCBs como os grupos que mais causam impacto nos cetáceos da costa de São Paulo. Isso refletiu o histórico de ambos no Brasil, tanto em indústria como em agricultura e saúde pública. Em contrapartida, HCHs e HCB não apresentaram concentrações elevadas, fato que pode ser atribuído à volatilidade dos mesmos em regiões de clima tropical. Da mesma maneira, a-clordano, g-clordano e mirex não foram detectados em níveis significativos. A foca de Weddell (Leptonychotes weddelli), habitante do continente antártico, evidenciou as menores cargas de contaminante entre os animais estudados. As toninhas (Pontoporia blainvillei) e o golfinho-nariz-de-garrafa (Tursiops truncatus) também apresentaram baixos níveis de organoclorados. Os botos-cinza (Sotalia fluviatilis) revelaram concentrações de DDT iguais ou superiores a cetáceos da Índia, país onde esse pesticida ainda não está proibido. Já o golfinho-de-dentes-rugosos (Steno bredanensis) mostrou a maior contaminação entre os animais analisados, comparável a espécies estudadas em águas costeiras de países desenvolvidos (onde os organoclorados foram muito utilizados).
Organochlorine compounds cause strong impact on the nature, as a consequence of three basic characteristics: environmental persistence, bioaccumulation, and high toxicity. The marine mammals are one of the most vulnerable organisms to the chronic toxicity of these contaminants. Besides the high concentration in the body, the female transfers part of her load to the offspring during gestation and lactation. The aim of this study was (1) the optimization of a methodology for determining chlorinated hydrocarbons (pesticides and PCBs) in fatty biological matrices and (2) the analysis of organochlorines in marine mammals blubber sampled along São Paulo coast (Brazil) and King George Island (Antarctica). According to the optimized methodology, the extraction was carried out in Soxhlet apparatus (8 h) with a mixture of n-hexane and dichloromethane. The clean-up was carried out with acid treatment and the resulting extract injected into gas chromatography coupled to electron capture detector (GC-ECD). The method performance was evaluated with certified reference material and fitted for international standards of control quality. The mean method detection limit was 2 ng g-1. DDTs and PCBs were the most concentrated organochlorines in the cetaceans from São Paulo coast. These findings reflected their past usage in Brazil by industry, agriculture, and public health. On the other hand, both HCHs and HCB were not found in high concentration likely due to their volatility in tropical climate areas. Mirex, a-chlordane and g-chlordane were not detected in elevated levels. The Weddell seal (Leptonychotes weddelli), from Antarctic continent, presented the smallest load among the studied animals. As the same way, the franciscanas (Pontoporia blainvillei) and the bottlenose dolphin (Tursiops truncatus) presented low organochlorine levels. The marine tucuxi (Sotalia fluviatilis) showed equal or higher DDT concentration than Indian cetaceans where that pesticide is still in use. The rough-toothed dolphin (Steno bredanensis) revealed the greatest contamination among the analyzed animals, comparable to species studied in the coastal waters of developed countries (where organochlorines were extensively used).

O estudo objetivou desenvolver e validar um método para identificar e quantificar poluentes orgânicos persistentes, conhecidos como POPs, em solo de regiões industriais dos municípios de Caieiras e Franco da Rocha SP, via cromatografia a gás acoplada a Espectrometria de Massas (GC-MS) e Detector por Captura de Elétrons (ECD). Em observância ao Tratado de Estocolmo, patrocinado pela Organização das Nações Unidas ONU, que prevê o banimento de pelo menos doze dos POPs, e no qual o Brasil é um dos 113 países signatários, o trabalho visou colaborar positivamente com essa questão ambiental tão importante. Estes compostos são tóxicos e altamente estáveis no ambiente e em organismos vivos, dentre os quais, são abordados nesse trabalho o clordano cis/trans (C10H6Cl8), o heptacloro (C10H5Cl7), o heptacloro epóxido cis/trans (C10H5Cl7) e os isômeros α-, β-, γ- e δ-BHC (C6Cl6). Para garantir a confiabilidade das análises realizadas, foram realizados ensaios de Validação da metodologia, com base nas diretrizes do INMETRO. A técnica de extração utilizada foi o QuEChERS, obtendo resultados de recuperação na faixa de 70 a 120% para a maioria dos compostos estudados, considerados aceitáveis para matrizes complexas. Os limites de detecção e quantificação do método compreenderam a faixa de 0,0002 e 0,01 μg.g1, respectivamente. As amostras analisadas apresentaram contaminação pelos compostos hexaclorobenzeno α-, β-, γ- e δ-, muitas das quais estão acima dos limites máximos permissíveis, de acordo com as legislações nacionais e internacionais vigentes.
The study aimed to develop and validate a method to identify and quantify persistent organic pollutants, known as POPs in soil of industrial regions Caieiras and Franco da Rocha municipalities in São Paulo, via gas chromatography coupled with Mass Spectrometry (GC-MS) and Electron Capture Detector (ECD). In compliance with the Treaty of Stockholm, sponsored by the United Nations - UN, which provides for a ban of at least twelve POPs, in which Brazil is one of the 113 signatory countries, the work aims to contribute positively to the environmental issue as important . These compounds are highly toxic and stable in the environment and living organisms, among which are address in this work chlordane cis / trans (C10H6Cl8), heptachlor (C10H5Cl7), heptachlor epoxide the cis / trans (C10H5Cl7) and the isomers α-, β-, γ- and δ-BHC (C6Cl6). To ensure reliability of the analysis carried out, tests were carried out validation method, based on the guidelines of INMETRO. The extraction technique was used QuEChERS, achieving recovery in the range 70 to 120% for the most of compounds, acceptable for complex matrices. The limits of detection and quantification of the method comprises the range of 0.0002 and 0.01 μg.g-1, respectively. The samples analyzed were contaminated by compounds hexachlorobenzene α-, β-, γ- and δ-, many of which are above the maximum allowable in accordance with national legislation and international law.

Abstract:
The focus of this thesis was to investigate developmental neurotoxic effects of different persistent and non-persistent environmental pollutants, alone or in binary mixtures, when exposure occurs during a critical period of brain development, in mice. The compounds investigated included a perfluorinated compound, perfluorohexane sulphonate (PFHxS), and four different pesticides, endosulfan, cypermethrin, chlorpyrifos and carbaryl. Both persistent and non-persistent pollutants are detected in the environment and in humans, which shows that exposure to these compounds is occurring in real life. Humans can therefore be exposed to various pollutants during their whole lifetime, starting from the gestational period to adulthood. Furthermore, exposure to environmental pollutants is rarely exclusive to a single compound, but rather occurs through combinations of various pollutants present in the environment. Exposure to environmental pollutants during human brain development have been suggested to be a possible cause for neuropsychiatric disorders, such as autism spectrum disorder (ASD) and attention deficit hyperactivity disorder (ADHD). Previous studies have shown that chemicals can induce irreversible disorders in brain function when exposure to these chemicals occurs during a critical defined period of the brain development known as the brain growth spurt (BGS). The BGS is characterized by a rapid growth and development of the immature brain. In humans, and mice, this period also overlaps the lactation period indicating that newborns and toddlers can be exposed via mothers’ milk as well. This thesis has shown that a single oral exposure to PFHxS, endosulfan, cypermethrin, chlorpyrifos or carbaryl can induce developmental neurotoxic effects in mice, when exposure occurs during a critical period of brain development. These effects are manifested as persistent altered adult spontaneous behavior in a novel home environment, modified habituation, altered susceptibility of the cholinergic system and changed levels of neuroproteins in the mouse brain. Furthermore, a single neonatal co-exposure to a binary mixture of carbaryl/chlorpyrifos or PFHxS/endosulfan can interact and exacerbate the adult behavioral effects. These effects were seen at dosages were the single compound did not elicit a response or induced a much weaker behavioral effect. This indicates that risk assessments conducted on single compounds might underestimate interaction effects of mixtures when co-exposed.

En Afrique, la contamination des zones humides par les métaux et les pesticides va se poursuivre durablement. Pour autant, le statut toxicologique de ces milieux reste trop peu documente. Le but du présent travail est d’estimer la valeur du varan du Nil (Varanus niloticus) en tant qu’espèce sentinelle pour la contamination des zones humides continentales d’Afrique sub-Saharienne. Plomb, cadmium, et pesticides organochlorés et organophosphorés ont été quantifies, par spectrométrie d’absorption atomique et chromatographie gazeuse (respectivement), dans plusieurs tissus provenant de 71 spécimens issus de quatre sites (au Mali et au Niger) juges a priori inégalement contamines. Bien que des différences claires apparaissent, la contamination environnementale s’avère modérée sur les quatre sites, et ne semble pas constituer un risque notable pour les varans ni pour les humains qui s’en nourrissent occasionnellement. Toutefois la variabilité interindividuelle est importante. Les organotropismes des polluants détectés sont cohérents avec ceux préalablement décrits. Si l’on n’a pu mettre en évidence de différence liée au sexe en ce qui concerne les pesticides, les femelles présentaient des charges en métaux supérieures. La relation entre d’autres facteurs (taille, proportion de graisse) et les concentrations tissulaires a également été considérée. Les varans sont susceptibles de révéler des différences subtiles de contamination environnementale entre sites, et la résolution spatiale de l’outil semble très fine. La possibilité pratique d’utiliser cet indicateur se trouve donc validée. Un travail expérimental sur des varans captifs a par ailleurs été mené pour approfondir l’étude
In Africa, metal and pesticide contamination of wetlands is supposed to continue on a permanent basis. However, the ecotoxicological status of these ecosystems remains poorly documented. The aim of the present work is to assess the value of the Nile monitor (Varanus niloticus) as a sentinel species for the environmental contamination of continental wetlands in sub-Saharan Africa. Lead and cadmium on the one hand, and organochlorine and organophosphate pesticides on the other, have been quantified in several tissues by atomic absorption spectrophotometry and gas chromatography, respectively. Samples come from 71 specimens originating from four sites considered unequally contaminated (in Mali and Niger). Although clear differences appear between sites, the environmental contamination turns out to be moderate at the four sites, and does not seem to represent a significant risk neither for the monitors themselves, nor for occasional human consumers. However, the interindividual variability is important. The organotropisms relative to the detected pollutants are consistent with those described in previous studies. Concerning the pesticides, no gender effect has been found, whereas females were more contaminated by metals. The relation between other factors (size, proportion of fat) and tissue concentrations has been considered too. Nile monitors can reveal subtle differences in local pollution and the spatial resolution of this tool seems to be very sharp. Its practical relevance is thus validated. Additionally, an experimental work has been carried out on captive monitors to go into the subject in greater depth

Algumas ilhas oceânicas como o Arquipélago de São Pedro e São Paulo podem ser consideradas áreas remotas e preservadas devido a sua distância do continente. Entretanto, essas regiões não estão isentas da influência de agentes antrópicos oriundos das regiões costeiras, como é o caso dos poluentes orgânicos persistentes (POPs). A presente proposta visou determinar a ocorrência e distribuição de POPs na biota marinha, do Arquipélago de São Pedro e São Paulo. As extrações das amostras foram realizadas em micro-ondas e os pesticidas organoclorados e os PCBs/PBDEs foram analisados respectivamente por cromatografia a gás com detecção por captura de elétrons e espectrometria de massas. Os compostos predominantes foram os PCBs e os DDTs que apresentaram, respectivamente, as concentrações máximas de: 127,7 e 34,37 ng g-1 para Exocoetus volitans (Peixe-voador), 98,15 e 8,89 ng g-1 para Sula leucogaster (Atobá-marrom) e 90,5 e 18,86 ng g-1 Grapsus grapsus (Aratu). Os baixos valores dos contaminantes sugerem um relativo grau de isolamento e preservação, porém a ocorrência e o perfil de distribuição dos PCBs sustenta a hipótese de que a principal fonte de contaminação em áreas remotas é o transporte atmosférico de longa distância, e demonstra a ubiquidade desses poluentes no ambiente marinho.
Some oceanic islands, such as São Pedro e São Paulo Archipelago may be considered remote areas and preserved due to its distance from the mainland. However, these areas are not exempt from the influence of anthropogenic agents from coastal regions, such as persistent organic pollutants (POPs). This proposal aimed to determine the occurrence and distribution of POPs in marine biota of São Pedro e São Paulo Archipelago. Samples extractions were performed with microwave and organochlorine pesticides and PCBs/PBDES was respectively analysed by gas chromatography with electron capture detection and mass spectrometry. The predominant compounds were PCBs and DDTs that presented, respectively, the maximum concentrations 127.7 and 34.37 ng g-1 to Exocoetus volitans (Flying fish), 98.15 and 8.89 ng g-1 to Sula leucogaster (Brown Booby) and 90.5 and 18.86 ng g-1 to Grapsus grapsus (Sally Lightfoot crab). The low levels of contaminants suggest a relative degree of isolation and preservation, but the occurrence and distribution profiles of PCBs supports the hypothesis that the main source of contamination in remote areas is long range atmospheric transport, and demonstrates the ubiquity of those pollutants in the marine environment.

Numerosos químicos de origem industrial e natural estão a aumentar a nível global, enquanto os seus efeitos de longo-termo permanecem desconhecidos. A coruja-da-torres é um bom bioindicador de poluição ambiental e as suas penas podem ser utilizadas como ferramentas de biomonitorização minimamente invasivas. A variação intra-individual das concentrações de mercúrio pode ser minimizada através do cálculo de uma média a partir de várias penas do indivíduo, independentemente do tipo de pena. Várias penas do mesmo ninho podem ser combinadas para obter uma melhor estimativa da contaminação no território. Penas de indivíduos atropelados são representativas da contaminação ao nível regional. Menor bioacumulação está relacionada com culturas irrigadas e paisagens agrícolas heterogéneas. O habitat parece mediar a transferências de mercúrio do solo para as presas, e da cadeia trófica até à coruja-das-torres. As penas podem ser particularmente úteis para detetar pesticidas organoclorados em desuso, que geralmente ocorrem em concentrações residuais, embora a contaminação externa deva ser avaliada; Summary: Numerous industrial and natural chemicals are increasing worldwide, while their long-term effects on wildlife and human health remain unknown. Barn owls are good biondicators of environmental pollution and their feathers may be used as minimally invasive monitoring tools. Intra-individual variation in mercury concentrations can be minimized by calculating an average from several feathers from an individual, regardless of feather type. Several feathers from the same nest may be pooled to better represent the average mercury contamination in the territory. Feathers from road-killed barn owls are representative of contamination at a regional level. Lower bioaccumulation in the barn owl is linked with irrigated crops and heterogeneous agricultural landscapes. Habitat seems to mediate the transfer of mercury from the soil to the prey, and along the food web to the barn owl. Feathers may be particularly suitable to detect legacy organochlorine pesticides which generally occur in residual concentrations, but external contamination should be assessed.

Based on the persistence and the lipophilicity of OCPs, these chemicals are ubiquitous in the environment and can be accumulated in fatty tissues of animals through the food chain. Even being restricted for years, OCPs are still detected in human bodies. In this thesis, analytical methods for the determination of OCPs were developed and applied for the analysis of cord serum samples. The evaluation of prenatal exposure to OCPs and its effects on birth outcomes as well as the postnatal growth were investigated. Due to the toxicology and carcinogenesis, biomonitoring of the OCP exposure to human is needed. Therefore, an analytical method with high sensitivity and specificity is required to detect OCPs at trace levels in serum. We compared two data acquisition modes of mass spectrometry (MS), namely selected ion monitoring (SIM) and multiple reaction monitoring (MRM). Higher sensitivity and selectivity were achieved by MRM because the background noise was reduced by lowering the matrix effects. Different ionization techniques, including electron ionization (EI), chemical ionization (CI) and atmospheric pressure chemical ionization (APCI) were evaluated. The EI source is a universal ionization technique available with the MS library for the compound identification. The negative chemical ionization (NCI) is more suitable to analyze compounds with high electronegativity. The novel ionization technique APCI was coupled to gas chromatography-tandem mass spectrometry (GC-MS/MS). The APCI source was evaluated by terms of ionization and fragmentation performance. APCI was a soft ionization technique generating molecular ions with high intensity. The selectivity and sensitivity of APCI were comparable or better than the EI source. As one of the largest consumers and producers of OCPs, China has suffered severe OCP pollution. Previous monitoring studies reported detectable levels of OCPs in human bodies. However, studies concerning the prenatal exposure to OCPs in China are limited. Due to the vulnerability of fetuses, the effects of prenatal exposure to OCPs could be more severe than those of adults. We collected cord serum samples during the delivery period in Wuhan, China and measured the OCP concentrations to assess the prenatal exposure by using GC-MS/MS. Compared with other areas in China, the OCP levels in Wuhan were comparable in this population. The identified predominant OCPs were β-HCH and p,p'- DDE, with geometric means of 8.67 and 33.9 ng/g lipid, respectively. Slight positive associations were found between α-HCH and β-HCH, and between o,p′- DDT and p,p′-DDT, which indicated similar exposure source of these chemicals. The obtained results showed that HCH levels were associated with maternal age, body mass index (BMI) before pregnancy, education levels, and passive smoking. Associations between the prenatal exposure of OCPs and birth outcomes were investigated. The sex-specific relationships between the OCP exposure and birth size were indicated. Concentrations of β-HCH were inversely associated with birth weight and ponderal index for boys, while for girls these associations were not significant. Our results suggested that the prenatal exposure to OCPs exerted negative effects on the fetal growth, and precautions should be taken even though the OCP levels were relatively low.

Bifenilos policlorados (PCBs) e pesticidas organoclorados são compostos que não ocorrem naturalmente no ambiente e não são facilmente degradados química ou microbiologicamente. Seu estudo no ambiente é importante devido à sua persistência, toxicidade, lipossolubilidade e conseqüente biomagnificação. Por isso, representam a maioria dos poluentes orgânicos persistentes (POPs) considerados prioritários pela UNEP (United Nations Environmental Programme) e banidos ou restritos pela Convenção de Estocolmo, de maio de 2001. Tais poluentes podem ser facilmente emitidos para a atmosfera, atingindo áreas remotas como a Antártida, integrando um processo cíclico de contaminação conhecido como destilação global. No presente trabalho otimizou-se uma metodologia analítica capaz de detectá-los em ovos gorados de pingüins e indivíduos de krill coletados na Baía do Almirantado, Ilha Rei Jorge, Antártida. Tal metodologia foi avaliada e enquadrou-se em critérios internacionais de controle de qualidade. Os compostos mais presentes foram, de um modo geral, os PCBs, DDTs e o HCB e a ocorrência pareceu ser espécie-específica dentro do gênero Pygoscelis. Em todos os casos, o teor dos compostos não foi superior ao de aves árticas em nível trófico semelhante. A análise do krill permitiu estimar a biomagnificação dos compostos encontrados nos ovos, cuja única fonte de contaminação é a transferência fêmea-filhote.
Polychlorinated biphenyls (PCBs) and organochlorine pesticides are compounds that do not occur naturally in the environment and are not easily degraded by chemical or microbiological action. Their study in the environment is important due to persistence, toxicity, liposolubility and consequent biomagnification. For these reasons, they represent the majority of the persistent organic pollutants (POPs), considered to have priority by the UNEP (United Nations Environmental Programme) and banished and/or restricted by the Stockholm Convention of May, 2001. Such pollutants can be easily ejected into the atmosphere and reach areas as remote as Antarctica, integrating a cyclical contamination process known as ?global distillation?. In the present work an analytical methodology capable of detecting such compounds in unhatched penguin eggs and whole krill was optimized. The samples were collected in Admiralty Bay, King George Island, Antarctica. This methodology was evaluated and fitted international quality control criteria. The compounds found in higher levels were, in most of the samples, the PCBs, DDTs and HCB and the occurrence seemed to be species-specific for the Pygoscelis genus. In all of the cases, the levels found were not higher than the ones in arctic birds in a similar trophic level.The krill samples analysis made it possible to estimate the biomagnification of the compounds found in eggs, whose only source of contamination is the female-offspring transfer.

Bifenilos policlorados (PCBs) e pesticidas organoclorados são poluentes resistentes à degradação e têm sido identificados em vários compartimentos do ambiente marinho. O objetivo deste trabalho foi otimizar e validar a metodologia, e avaliar a contaminação por pesticidas organoclorados e PCBs em algas e sedimentos costeiros no Estuário e Baía de Santos-São Vicente e no Complexo Estuarino-Lagunar de Iguape e Cananéia (SP). Foram coletadas nessas regiões um total de 37 amostras de sedimento superficial, 3 testemunhos com 27 sub-amostras e 10 amostras de alga, no período de 1998-2001. O método, que incluiu a extração e purificação com solventes orgânicos e análise por GC-ECD, mostrou-se satisfatório dentro dos padrões internacionais. Na região de Cananéia-Iguape predominaram os DDTs (sedimentos: 0,78 a 14,52 ng.g-1 peso seco) e as maiores concentrações ocorreram nas proximidades da Cidade de Cananéia e Baía de Trapandé. Na região de Santos predominaram os PCBs (sedimentos: 4,08 a 133,42 ng.g-1 peso seco, testemunhos de sedimentos: 3, 70 a 44,65 ng.g-1 peso seco e algas: 15,83 a 43,21 ng.g-1 peso seco) e os maiores valores foram encontrados no emissário submarino, no Estuário de Santos, em locais próximos as indústrias, e no Estuário de São Vicente. De maneira geral, as concentrações tanto de PCBs quanto de pesticidas organoclorados em Santos e Cananéia-Iguape foram baixas. Santos apresentou maior incidência de PCBs, provavelmente devido à industrialização, enquanto que em Cananéia-Iguape predominaram os pesticidas organoclorados, que podem ser atribuídos às antigas campanhas de saúde pública ou aplicações nas lavouras.
Polychlorinated biphenyls and chlorinated pesticides are degradation resistant pollutants. They have been detected in several compartments of marine environment. The objective of this study was the optimization of the methodology and the evaluation of the contamination of chlorinated pesticides and PCBs em algae and coastal sediments in the Estuary and Bay of Santos-São Vicente and in the Lagunar Complex of Iguape and Cananéia (SP). An amount of 37 samples of superficial sediment, 3 cores with 27 subsamples and 10 samples of algae was collected in the period of 1998-2001. The methodology was considered satisfactory within international standards, that includes extraction and clean up with organic solvents and analysis with GC-ECD. In the Cananéia-Iguape region, DDTs were predominant and the highest levels were found around the city of Cananéia and in Trapandé Bay. In the Santos area, the PCBs were predominant and the highest values were found around the Santos Bay outfall and in the Estuary of Santos-São Vicente. In general, either the concentrations of PCBs or chlorinated pesticides in Santos and Cananéia-Iguape were low. Santos presented higher incidence of PCBs, probably due to industrialization, while in Cananéia-Iguape the chlorinated pesticides predominated, and that can be attributed to past public health campaigns or to agricultural uses.

The Trinity River passes through the Dallas-Fort Worth metroplex receiving point and non-point source contaminant loadings. Lepomis spp. were collected at twelve sampling locations in the Trinity River in August 1987 and September 1988 and analyzed for organochlorine pesticides and heavy metals. Results from the study were compared to existing U.S. FDA action and tolerance levels, LC50s, and historical data. Various longitudinal trends and some concentration patterns were observed. Continual study of pesticide and metal body burdens in fish allow testing for trends, and thereby, lead to a better understanding of the distribution of contaminants in the Trinity River.

Os compostos organoclorados (OCs), como pesticidas e bifenilos policlorados (PCBs), são persistentes, tóxicos e amplamente distribuídos através do transporte atmosférico e correntes oceânicas. Poucos são os estudos realizados de OCs em tartarugas, sendo que nenhum foi realizado na costa brasileira. A Chelonia mydas é a maior tartaruga marinha de carapaça dura, que se distribue por todos os oceanos, nas zonas de águas tropicais e subtropicais. O presente trabalho visa verificar a ocorrência de organoclorados na espécie C. mydas, bem como sua possível relação com algum tipo de alteração morfológica ou fisiológica. Foram coletadas amostras de gordura, fígado, rim e músculo em 27 espécimes juvenis do litoral de Ubatuba, São Paulo. Os organoclorados foram extraídos com solventes orgânicos e o extrato foi purificado com ácido concentrado. A identificação e quantificação dos PCBs e pesticidas foram realizadas em cromatógrafo a gás com espectrômetro de massas e com detector de captura de elétrons, respectivamente. Os pesticidas organoclorados não foram detectados em nenhuma amostra. As concentrações de PCBs totais em peso úmido foram de: <1,6 48,9 ng.g-1 em gordura, <1,6 17,4 ng.g-1 no fígado e <1,6 9,2 ng.g-1 no rim. Os baixos níveis encontrados estão relacionados principalmente a sua dieta alimentar, uma vez que a C. mydas é basicamente herbívora, e ao local de coleta, que é uma região não industrializada.
Organochlorine compounds, such as pesticides and polychlorinated biphenyls (PCBs), are persistent, toxics and widely distributed due to the atmospheric transport and oceanic currents. Few studies have reported the occurrence of these compounds in turtles, but none from the Brazilian Coast. Chelonia mydas is the biggest marine turtle with hard carapace that extends throughout tropical and subtropical seas around the world. The present study aims to assess the concentrations of organochlorines in the C. mydas as well as any relationship with morphological or physiological variations. Fat, liver and kidney samples were collected from 27 juvenile specimens found dead in Ubatuba Coast, São Paulo State. Organochlorine were extracted with organic solvents and the extracts were purified with concentrated acid. PCBs and pesticides were identified and quantified using a gas chromatograph with mass spectrometer and with electron capture detector, respectively. Pesticides were not detected in any sample analyzed. PCBs concentrations in wet weight were: <1.6 48.9 ng.g-1 in fat, <1.6 17.4 ng.g-1 in liver and <1.6 9.2 ng.g-1 in kidney. The low levels detected are related, especially, to the feeding habits of the species since C. mydas is mainly herbivore, and to a non industrialized sampling area.

Abstract:
This thesis presents an evaluation of the dispersive solid-phase extraction (d-SPE) method referred to as the quick, easy, cheap, effective, rugged and safe (QuEChERS) method for the determination of four organochlorine pesticide residues in fish samples. The pesticides investigated in this study were o, p′-DDT, p, p′-DDE, aldrin and dieldrin. The combined use of Gas Chromatography with an Electron Capture Detector (GC-ECD) and sensitive Time of Flight (TOF) mass detector facilitated the identification of the target analytes. In the absence of certified reference material, the overall analytical procedure was validated by systematic recovery experiments on spiked samples at three levels of 2, 5 and 10 ng/g. The targeted compounds were successfully extracted and their recovery ranged from 76 to 96% with relative standard deviations of less than 13%. The optimum QuEChERS conditions were 2g of fish powder, 10 ml acetonitrile and 1 min shaking time. The optimal conditions were applied to assess the levels of chlorinated pesticides in blunt-tooth catfish (Clarias ngamensis), tigerfish (Hydrocynus vittatus), Oreochromis andersonii and red-breasted tilapia (Tilapia rendalli) from the Okavango Delta, Botswana. Dieldrin, p, p′-DDE and aldrin were detected in all the analysed samples with a concentration range of 0.04 – 0.29, 0.07 – 0.33, 0.04 – 0.28 and 0.03 – 0.24 ng/g per dry weight in O. andersonii, C. ngamensis, T. rendalli and H. vittatus respectively. These concentrations were below the US-EPA 0.1 μg/g allowable limit in edible fish and the Australian Maximum Residue Limit (MRL) of 50 - 1000 ng/g in fresh fish. DDT was not detected in all the fish species investigated. The mean lipid content recorded in the fish samples were 1.24, 2.16, 2.18 and 4.21% for H. vittatus, T. rendalli, O. andersonni and C. ngamensis respectively. No systematic trend was observed between fish age and pesticide levels in fish. Acetylcholinesterase (AChE) activity assays were performed to assess the effects of organochlorine pesticides in Clarias ngamensis. The enzyme activity recorded in Clarias ngamensis from the Okavango Delta and the reference site was 12.31 μmol of acetylcholine iodide hydrolysed/min/g brain tissue. The enzyme activity remained the same indicating no enzyme inhibition. The conclusions drawn from this study are that the QuEChERS method is applicable for the determination of organochlorine pesticide residues in fish matrices. The fish from the Okavango Delta are safe for human consumption.

Foram feitas coletas de peixes das espécies Paralonchurus brasiliensis, Trichiurus lepturus e Cathorops spixii em três diferentes localidades. Depois de sub-amostradas em músculo, fígado, brânquias e gônadas, as amostras foram extraídas em aparelhagem Soxhlet e analisadas por técnicas cromatográficas. O DDT apresentou-se em 66 % das amostras com concentrações variando entre 18,92 e 690,29 ng.g-1 lipídio. O HCH foi detectado em 34 % das amostras com concentração variando de 8,92 a 103,04 ng.g-1 lipídio, sendo a região da Ilha da Moela a mais afetada por esse composto. O Mirex apresentou-se em 28 % das amostras e alcançou valores entre 5,90 e 222,17 ng.g-1 lipídio. O HCB foi o inseticida menos encontrado neste trabalho, com 16 % das amostras apresentando contaminação variando de 7,75 a 94,47 ng.g-1 lipídio, todas na espécie bentônica Cathorops spixii. Os PCBs estiveram presentes em todas as localidades analisadas, sendo os congêneres com número entre quatro e sete cloros os que mais bioacumularam. A bioacumulação de organoclorados nos diferentes tecidos não ocorreu de forma igual para todas as espécies analisadas. Aparentemente, a maior contribuição para poluição orgânica dos peixes da Baía de Santos e áreas próximas origina-se do estuário ou do emissário.
Industrial and sewers pollution have being affected seriously the region of Baixada Santista, in the central shore of São Paulo State, Brazil. The great development of Cubatão city caused diverse health problems in the 70´s and 80´s. This present study shows organochlorine distribution in fishes collected in the Santos Bay and Moela Island. Paralonchurus brasiliensis, Trichiurs lepturus and Cathorops spixii had been captured in three different locations, sub-sampled in muscle, liver, gills and gonads. The samples was extracted in Soxhlet apparatus and analyzed by chromatographic techniques. DDT was presented in 66 % of the samples and concentrations varied between 18,92 and 690,29 ng.g-1 lipid. The HCH was detected in 34 % of the samples with concentration ranged from 8,92 to 103,04 ng.g-1 lipid. The region of the Moela Island was the most affected region by HCH pollution. The Mirex was presented in 28 % of the samples and reached values between 5,90 and 222,17 ng.g-1 lipid mostly in the Moela Island and Santos Bay. The HCB was the least found insecticide in this work and occurred in 16 % of the samples that presented concentration that varied from 7,75 to 94,47 ng.g-1 lipid. The muscles and gills of the bentic specie Cathorops spixii appeared to be an important tissues to accumulation of HCB. The PCBs were present in every studied locations. Congeners with number of chlorine between four and seven were the most bioacumulated ones. The bioaccumulation of different tissues did not occurred in the same way. Apparently, the greatest contribution for organic pollution to the fishes of Santos Bay and surrounding areas originates from the estuary or the emissary.

L'activité humaine est responsable de pollutions diffuses des sols entrainant des dommages sur l'écosystème. Afin d'éviter des déséquilibres importants comme la diminution de la biodiversité ou bien la contamination des chaînes trophiques par des polluants, il est nécessaire de restaurer les écosystèmes. Les différentes techniques utilisées à l'heure actuelle sont couteuses, diminuent la fertilité du sol et ont un effet négatif sur la fonctionnalité des écosystèmes. A contrario, la phytoremédiation ou dépollution grâce aux plantes, apparaît comme une solution digne d'intérêt. Néanmoins, elle nécessite d'être améliorée puisque de nombreux mécanismes restent inexpliqués. L'originalité de notre travail est d'étudier la fonctionnalité de la rhizosphère du maïs (Zea mays) exposée à des pesticides organochlorés (OCPs), le lindane et la chlordécone, interdits respectivement en 1998 et 1993 et persistants dans l'environnement. Cette étude a été réalisée en présence ou absence de microorganismes afin de préciser les mécanismes mis en jeux par les microorganismes et les matrices végétales. Ce travail a mis en évidence les mécanismes impliqués dans la phytotoxicité des deux organochlorés sur les cellules racinaires. Les réponses cellulaires sont dose-dépendantes et montrent un effet des OCPs sur le cycle cellulaire (induction de l'endoréplication et de la ploïdie) et la mort cellulaire (induction de ROS, du calcium cytosolique, des caspase-3-like, de l'apoptose) lors d'expositions extrêmes (correspondant à des cas particuliers comme les friches industrielles). A de plus faibles expositions ayant une réalité environnementale, les racines exposées montrent une perturbation importante de leur métabolome primaire. Ces modifications mesurées sur la production des sucres, des acides organiques, des acides aminées et des lipides peuvent être induites par le stress oxydant produit par les OCPs. Par ailleurs, de tels changements dans le métabolome peuvent provoquer une modification de la composition des exsudats racinaires, jouant sur la relation plante/microorganismes de la rhizosphère. Nos résultats montrent que la fonctionnalité globale de la rhizosphère (allocation des ressources en carbone et azote, catabolisme des microorganismes) n'est pas modifiée sur le long terme en présence des OCPs. Enfin, nous avons démontré l'implication de la rhizosphère (rétention/métabolisation des OCPs) sur l'absorption des OCPs par Zea mays. Le lindane semble être en partie minéralisé par la microflore rhizosphèrique puis les métabolites libérés sont absorbés par les racines, où ils sont majoritairement bioconcentrés. Ces connaissances fondamentales mettent en évidence l'adaptation de la rhizosphère à la pression chimique des OCPs et montrent que la rhizosphère reste fonctionnelle en présence de matrices contaminées par les OCPs
Human activities lead to diffuse pollution causing damages to the ecosystem. To avoid major disturbances such as biodiversity loss or contamination of trophic chains by pollutants it is necessary to restore these ecosystems. The classical techniques used in depollution are expensive, reduce soil fertility and have a negative effect on ecosystem functionality. In contrast, the use of plants to clean up soils called phytoremediation seems to be a more effective solution. However, it needs to be improved because many biological mechanisms remain unexplained. The originality of our work is to study the functionality of the rhizosphere of maize (Zea mays) exposed to organochlorine pesticides (OCPs), lindane and chlordecone, banned in 1998 and 1993 respectively and persistent in the environment. This study will be conducted in the presence or absence of microorganisms to clarify the relationships between microorganisms and plant matrices. In this study, the mechanisms involved in the phytotoxicity of OCPs in root cells were analyzed. The cytotoxicity was dose-dependent and showed effects on the cell cycle (induction of endoreduplication and level of ploidy) and on cell death (induction of ROS, cytosolic Ca2+, caspase-3-like, apoptosis) under higher doses of exposure (particular case of former industrial sites). Lower exposures usually detected in the environment showed great disturbances in the root metabolome. These changes in sugars, organic acids, amino acids and lipids syntheses could be induced by oxidative stress provided by OCPs. Moreover, metabolome changes might cause modifications in root exudate composition which is involved in plant-microbe relationships. Ours results demonstrate that the overall rhizosphere functionality (resource allocation, carbon and nitrogen contents and microorganism's catabolism) was not disturbed by OCPs long-term exposure. Finally, we demonstrated rhizosphere positive impact on uptake of OCPs by Zea mays. Lindane seems to be partially mineralized by rhizospheric microorganisms. Then, these produced metabolites were uptaken by roots where they were bioconcentrated. These findings highlighted the rhizosphere adaptation to chemical pressure by OCPs and showed the good rhizosphere functionality despite of the OCPs contamination

Des études épidémiologiques ont montré que l’exposition à certains xénobiotiques est associée à une augmentation de la prévalence des maladies métaboliques. L’Homme est exposé à des mélanges de xénobiotiques de manière chronique et inévitable. Nous avons étudié les effets de l’interaction de deux xénobiotiques sur le métabolisme du foie, organe majeur de détoxification de l’organisme. Nous avons choisi deux perturbateurs endocriniens et polluants organiques persistants, qui activent des voies de signalisation différentes: la 2, 3, 7, 8 tétrachlorodibenzo-p-dioxine (TCDD), agissant via le récepteur aux hydrocarbures aromatiques (AhR), et l’α-endosulfan, un pesticide organochloré, qui peut agir via la voie du récepteur aux oestrogènes (ER) ou du récepteur X aux pregnanes (PXR). Notre objectif est de déterminer l’effet du mélange de ces polluants par rapport à chaque polluant isolé sur la régulation de certaines voies du métabolisme hépatique in vitro dans la lignée hépatocytaire humaine, HepaRG. Dans une première publication, une étude du transcriptome de cellules HepaRG différenciées a été effectuée. Ces cellules ont été exposées pendant 30 heures à 25nM de TCDD, 10μM d’α-endosulfan, ou au mélange. Nous avons observé que le mélange inhibe fortement l’expression de certains gènes impliqués dans le métabolisme glucidique et dans celui des alcools. Dans une seconde étude, nous avons donc étudié le mécanisme d’action du mélange sur le métabolisme glucidique. L’expression de deux gènes de la néoglucogenèse hépatique, le transporteur de glucose 2 (Glut2) et la glucose 6 phosphatase (G6Pc), est réduite de plus de 80% par le mélange. L’expression d’autres gènes du métabolisme glucidique (pyruvate kinase, glycogène synthase, glycogène phosphorylase, pyruvate déhydrogénase 2) est aussi diminuée, suggérant que le mélange peut affecter ce métabolisme de manière significative. De plus, la production de glucose diminue de 80% avec le mélange dans des conditions néoglucogéniques. En condition glycolytique, l’oxydation du glucose en CO2 diminue de 30% après 72 heures d’exposition au mélange. Un traitement à plus long terme (8 jours) avec des doses plus faibles des polluants (0.2 à 5nM de TCDD, 3μM d’α-endosulfan) diminue aussi l’expression de la G6Pc et de Glut2. Nous avons montré que la TCDD active bien la voie du AhR, et que le ER est impliqué dans l’action de l’α-endosulfan. Dans la troisième partie de cette thèse, nous avons étudié la régulation de plusieurs enzymes impliquées dans le métabolisme de l’alcool (alcool déshydrogénases, ADHs, cytochrome P450 2E1, CYP2E1) après l’activation du AhR. Les agonistes du AhR entrainent la diminution de l’expression des ARNm des ADH1, 4, 6 et du CYP2E1 et des protéines correspondantes. Nous avons montré que cette régulation utilise la voie génomique du AhR. De plus, cet effet est également observé après traitement de 8 jours par de faibles doses de TCDD. L’exposition chronique de l’Homme à de faibles doses de xénobiotiques en mélange pourrait affecter le métabolisme glucidique hépatique et contribuer, en partie, au développement du syndrome métabolique
Epidemiological studies have shown that exposure to certain xenobiotics is associated with an increased prevalence of metabolic diseases. Humans are exposed to mixtures of xenobiotics in a chronic and inevitable way. We studied the effects of the interaction of two xenobiotics on metabolism in the liver, the major organ for detoxification in the body. We chose two endocrine disruptors and persistent organic pollutants which activate different signaling pathways: 2,3,7,8 tetrachlorodibenzo-p-dioxin (TCDD), which uses the AhR (Aryl hydrocarbon receptor) pathway, and α-endosulfan, an organochlorine pesticide, which acts via the PXR (pregnane X receptor) and/or the ER (estrogen receptor) pathway. Our aim was to determine the effects of this pollutant mixture, as compared to each pollutant alone, on the regulation in vitro of some hepatic metabolism pathways in the human hepatic cell line, HepaRG. In the first publication, a transcriptomic study of differentiated HepaRG cells was performed. The cells were exposed for 30h to 25nM TCDD, to 10 µM α-endosulfan or to the mixture. We observed that the mixture strongly inhibited the expression of some genes involved in the metabolism of glucose and alcohol. In the second study, we studied the mechanism of action of the mixture of pollutants on the metabolism of glucose. The expression of two genes involved in hepatic gluconeogenesis, glucose transporter 2 (GLUT2) and glucose-6-phosphatase (G6Pc), were reduced 80% by the mixture. The expression of other glucose metabolism genes (pyruvate kinase, glycogen synthase, glycogen phosphorylase, pyruvate dehydrogenase 2) also was decreased suggesting that the mixture might impact markedly carbohydrate metabolism. Furthermore, glucose production decreased 40% with the mixture under gluconeogenic conditions. Under glycolytic conditions, the oxidation of glucose into CO2 decreased 30% after 72h of exposure of the cells to the mixture. Long-term treatment (8 days) with lower doses (0.2 to 5 nM TCDD, 3 µM α-endosulfan) similarly decreased G6Pc and GLUT2 expression. We showed that TCDD activated the AhR pathway, and that ER was partly involved in the α-endosulfan effect. In the third part of this thesis, we studied the regulation of several enzymes involved in the metabolism of alcohol (alcohol dehydrogenase, ADH, cytochrome P450 2E1, CYP2E1) after activation of AhR. AhR agonists led to a decrease in the amounts of mRNAs for ADH1, 4, 6 and CYP2E1 and the corresponding proteins. We showed that this regulation uses the AhR genomic pathway. Furthermore, this effect was also observed after 8 days of treatment with lower doses of TCDD. Chronic exposure of individuals to low doses of xenobiotics in mixtures might significantly affect hepatic carbohydrate metabolism and be a contributing factor for the development of the metabolic syndrome

Organochlorine (OCs) pesticides are persistent, bioaccumulative substances in the food chain, with high mobility in the environment. This is of concern, because vry often these compounds are detected in surface and groundwater, affecting thus quality of water resources and biodiversity. Because of the serious contamination of aquatic ecosystems by OCs, it is necessary to develop new and eficiente adsorbent materials for remediation of already contaminated areas. The objective of this work was to prepare and to characterize organophilic clays chemically treated with the hexadecyltrimethyl-ammonium bromide salt (HDTMA-Br), aiming their use as an adsorbent material for the removal of residues of OCs in the environment. In order to analyze the OCs, was optimized a methodology based on solid phase extraction followed by gas chromatography with electron capture detector (SPE-GC/ECD). The characterization of the material was carried out by determination of the cation exchange capacity (CTC), as well as X-ray diffraction (XRD), infrared spectroscopy (FTIR) and electron microscopy of scanning (SEM). The adsorptive capacity of the organophilic clay was determined by kinetic adsorption tests and adsorption isotherms. Chromatographic method, besides high selectivity, showed adequate linearity (arounding 0.25 to 20.0 µg.L-1), sensitivity (LODs varying from 0.03 to 0.79 µg.L-1 and LOQs varying from 0.13 to 2.63 µg.L-1) and precision for trace analysis (CVs varying between 2.16 to 26.69 µg.L-1). The natural clay presented adequate cation exchange capacity (CTC) for organophilization and the results of the characterization indicated the intercalation of HDTMA salt cations between the intermediate layers of the modified clays. The kinetic adsorption assays performed revealed 125% COC as the best adsorbent, exhibiting affinity for aldrin and 4,4'-DDT pesticides. The experimental data were better fitted to the pseudo-first-order and pseudo-second order model for the 4,4'-DDT and aldrin compounds, respectively. The Langmuir isotherm model described better the adsorption of the 4,4'-DDT compound, whereas the Freundlich model represented better the adsorption of the aldrin compound, indicating a very favorable adsorption condition.
Pesticidas organoclorados (POCs) são substâncias persistentes, bioacumulativos na cadeia alimentar, com grande mobilidade no ambiente e frequentemente detectados em águas superficiais e subterrâneas, causando preocupação com relação à qualidade dos recursos hídricos e à biodiversidade. Devido à contaminação dos ecossistemas aquáticos por POCs, são necessários que sejam desenvolvidos novos e eficientes adsorventes para remediação das áreas já contaminadas. O objetivo deste trabalho foi preparar e caracterizar argilas organofílicas quimicamente tratadas com o sal brometo de hexadeciltrimetilamônio (HDTMA-Br), objetivando seu uso como material adsorvente para remoção de resíduos de POCs no ambiente. Para análise dos POCs, otimizou-se uma metodologia baseada na extração em fase sólida seguida por cromatografia gasosa com detector por captura de elétrons (SPE-GC/ECD). A caracterização das argilas foi realizada mediante determinação da capacidade de troca de cátions (CTC), bem como por análises por difração de raios-x (DRX), espectroscopia no infravermelho (FTIR) e microscopia eletrônica de varredura (MEV). A capacidade adsortiva das argilas organofílicas foi determinada por ensaios cinéticos e isotermas de adsorção. O método cromatográfico, além de elevada seletividade, apresentou adequadas linearidade (0,25 a 20 µg.L-1), sensibilidade (LDs variando de 0,03 a 0,79 µg.L-1 e LQs variando de 0,13 a 2,63 µg.L-1) e precisão para análises de traços (CVs variando entre 2,16 e 26,69 % entre medidas realizadas no mesmo dia). A argila natural apresentou capacidade de troca catiônica (CTC) adequada para a organofilização e os resultados da caracterização indicaram a intercalação de cátions do sal HDTMA entre as camadas intermediarias das argilas modificadas. Os ensaios cinéticos de adsorção realizados revelaram a argila organofílica 125% CTC como melhor adsorvente, apresentando afinidade para os pesticidas aldrin e 4,4’-DDT. Os dados experimentais ajustaram-se melhor ao modelo de pseudo-primeira ordem e pseudo-segunda ordem para os compostos 4,4’-DDT e aldrin, respectivamente. O modelo de isoterma de Langmuir descreveu melhor a adsorção do composto 4,4’-DDT, enquanto que o modelo de Freundlich representou melhor a adsorção do composto aldrin, indicando condição de adsorção bastante favorável.

The aim of this study is to assess the biological accumulation of pesticide residues in aquatic organisms in Sai Gon - Dong Nai (SG-DN) estuary. Fish and mollusks were collected directly at the Soai Rap and Long Tau estuary of the SG-DN river system, washed and separated for taking the tissue. The organochlorine compounds from the tissue were then extracted and analyzed by gas chromatography system. The results showed that, the concentration of OCPs in Tegillarca granosa, Meretrix lyrata, Margaritifera auricularia and Bostrychus sinensis varied from 6.4 to 59.9 μg/kg, 7.2 to 322 μg/kg, 4.5 to 62.1 μg/kg and 2.9 to 114.3 μg/kg fresh weight, respectively. In general, molluscs species that accumulate more heptachlor, aldrin, endrin or dieldrin tend to accumulate less DDT (dichlorodiphenyltrichloroethane). Endosulfan was the most commonly found in three bivalve mollusks while DDTs (1.5–75.2 μg/kg, averaging 8.7 μg/kg weight) was the most popular OCPs in the fish (Bostrychus sinensis) samples. In DDT group, the p,p’-DDT metabolite accounted for the largest percentage, reaching 50% of total DDTs. In HCH (Hexachlorocyclohexane) group, β-HCH isomer was predominant in almost samples.

Abstract:
Altered immune responses have been observed following occupational, inadvertent, or therapeutic exposure to xenobiotics. Many pesticides are known to cause immunotoxicity. Exposure to mixtures of pesticides, either concurrently or sequentially, may result in potentiating this effect partly because one can effect the metabolism of the other. The objective of this study was to determine the effect of the insecticides endosulfan, permethrin and their mixtures on C57/BL6 male mice thymocytes in vitro and to ascertain the mechanism by which these effects take place. Permethrin, a broad-spectrum synthetic pyrethroid, is a widely used insecticide in agriculture and public health. Endosulfan is a highly toxic chlorinated hydrocarbon insecticide used worldwide. We examined the immunotoxic potential of these pesticides using a flow cytometric technique in combination with 7-Amino Actinomycin D (7AAD) to distinguish live, early apoptotic, and late apoptotic/necrotic cells. DNA ladder assay, a hallmark of apoptosis , was also used to determine the occurrence of apoptosis. Both endosulfan and permethrin were found to cause significant apoptotic death of thymocytes in a dose- and time- dependent manner. Thus, permethrin at 50, 100 or 300 µM was found to cause 5.5, 11.5 and 26.1% increases in early apoptotic cell death relative to control, respectively. Endosulfan at 25, 50 or 250 µM was found to cause 11.9, 15.7 and 68.0% early apoptotic cell death, respectively. For the mixture study, concentrations of 100 µM permethrin and 50 µM endosulfan were selected and found to cause 27.1% apoptosis. Thus, these pesticides in mixture have an additive immunotoxic effect. Increases in late-apoptotic/necrotic cells were found at these concentrations for either pesticide when exposed for 12 hours. DNA ladder assay confirmed the presence of DNA fragments and therefore the presence of significant apoptotic cell death. Apoptosis is a morphologically distinct form of cell death that can be mediated by a variety of pathological and physiological stimuli. Because permethrin and endosulfan were found to induce apoptosis in C57/BL6 mice thymocytes in vitro, the objective of the second half of this study was to elucidate the potential mechanism by which these pesticides regulate apoptosis in immune cells. Caspases are a family of cystine-dependent, aspartate-directed proteases that have an integral role in apoptotic cell death. Caspases, which are normally inactive in healthy cells, are activated during apoptosis and form an irreversible cascade. There are two subsets of caspases, initiator caspases (i.e. caspase 8 and 9) and effector caspases (i.e. caspases 3 and 6). Caspase 3, a downstream effector of apoptosis, is activated by many different pathways and is an apoptotic marker in cells. Caspase 8 is the apical caspase in the extrinsic pathway. Caspase 9 is the apical caspase in the intrinsic pathway, therefore we investigated mechanisms of pesticide induced apoptosis involving the thymocyte caspase system. Thymocytes from C57/BL6 mice were incubated with varying concentrations of pesticides for varying amounts of time. Active caspase 3 was then measured using EnzCheck Caspase 3 Assay Kit. Relative fluorescence for permethrin exposed cells after 12 hours incubation in the presence of pesticides at 150, 100, and 50 µM and 40 minutes in the presence of AFC-substrate was found to be 387, 386, and 297, respectively. Relative fluorescence for endosulfan exposed cells at 150, 100 and 50 µM was 188, 177, and 294. Caspase 3 activity increased as permethrin concentrations increased and decreased as endosulfan concentrations were increased. Then the extrinsic and intrinsic pathways of apoptosis were further investigated. Active caspase 8 was measured using the ApoAlert Caspase Fluorescent Assay Kit. Relative fluorescence for permethrin exposed cells after 7 hours incubation in the presence of pesticides at 100, 150, and 200 µM was found to be 35.5, 10.5, and 0, respectively. Relative fluorescence for endosulfan exposed cells after 7 hours incubation at 25, 50, 100 and 150 µM was found to be 32.8, 63.8, 69.5, and 55.5, respectively. A mixture study was then performed using endosulfan (50, 100, 150 µM) combined with permethrin (100 µM). All combinations were found to have more than an additive effect, therefore the extrinsic pathway seems to be involved. Caspase 9 activity was measured using Caspase 9/Mch6 Fluorometric Protease Assay Kit. Relative fluorescence for endosulfan exposed cells after 7 hours incubation at 25, 50, 100 and 150 µM was found to be 43, 73, 78.9, and 5.12, respectively. Relative Fluorescence for permethrin exposed cells at 100, 150 and 200 µM was found to be 34.5, 39, and 55.5, respectively. A mixture study was then performed using endosulfan (25, 50 µM) combined with permethrin (100 µM). Both combinations were found to have less than an additive effect. These results suggest that apoptosis caused by both endosulfan and permethrin exert their effects via the caspase pathway. The results also show that mixtures of pesticides have a less than additive effect on caspase 9 activation and more than an additive effect on caspase 8 activation, therefore the extrinsic pathway is predominantly involved in thymocyte apoptosis caused by mixtures of permethrin and endosulfan.

Os contaminantes ambientais de origem química, como metais pesados, pesticidas, hidrocarbonetos, PCB, organoclorados e dioxinas estão causando um forte impacto na saúde de animais marinhos, incluindo tartarugas marinhas, e a bioacumulação desses poluentes em tecidos e órgãos influenciam o crescimento e desenvolvimento de populações naturais de tartarugas marinhas em todo o mundo, podendo causar mortalidade em vários estágios de seu desenvolvimento. Uma vez que estão em perigo é extremamente importante para a compreensão das respostas às medidas de impacto e conservação à longo prazo nas populações de tartarugas marinhas. Este trabalho é uma visão geral dos estudos sobre a quantificação de contaminantes em ovos e sangue de tartarugas marinhas, buscando relações entre o tamanho da carapaça e a atividade de enzimas antioxidantes usando técnicas não-letais. A concentração de pesticidas organoclorados (OCPs) no plasma das fêmeas adultas e os ovos das tartarugas de pente (Eretmochelys imbricata) e tartaruga verde (Chelonia mydas) que desovam na área costeira de Campeche serão analisados. O objetivo principal desta tese é avaliar o potencial das populações naturais como bioindicadores de contaminação química. Para tal, foram analisados os valores hematológicos, contaminação de OCPs no plasma e ovos de duas espécies de tartarugas marinhas com hábitos alimentares diferentes e uma possível transferência materna na espécie de tartaruga de pente. Por fim, a atividade de biomarcadores enzimáticos foram correlacionadas com a contaminação por OCPs. Os resultados mostraram que foram encontrados OCPs em todos os ovos analisados, mostrando maiores concentrações de ΣHCH e ΣDrines nas duas espécies selecionadas, sendo encontradas diferenças significativas entre as espécies e os anos analisados. Foram encontras diferenças significativas entre as concentrações de OCPs (ΣHCHs, ΣDrines e ΣChlordanes) em relação ao sangue e ovos, indicaram que esses contaminantes químicos estão sendo transferidos maternalmente. Nenhuma enzima antioxidante teve relação significativa com os OCPs mais encontrados no sangue (ΣDDTs, ΣHCHs e ΣDrines).
Sea turtles have been suffering strong population impact due to anthropogenic activities, such as those caused by the increase in commercial and industrial exploitation of coastal areas, habitat alteration, ingestion of solid waste, incidental capture in fishing activities and contamination of chemical pollutants. Of the seven species of marine turtles existing distributed by almost all oceans (Caretta caretta, Chelonia mydas, Dermochelys coriacea, Eretmochelys imbricata, Lepidochelys olivacea, Natator depressus and Lepidochelys kempii), except the Natator depressus, all other six are listed as vulnerable, endangered or critically endangered on the IUCN list. Environmental contaminants of chemical origin such as heavy metals, pesticides, hydrocarbons, PCBs, organochlorine, and dioxins are causing major health impacts of marine animals, including sea turtles, and bioaccumulation of these pollutants in tissues and organs influence the growth and development of natural populations of sea turtles worldwide, may cause mortality in various stages of development. Because they are endangered is extremely important to the understanding of responses to impact and conservation measures in the long term in the populations of sea turtles in developing. This work is an overview of studies on the quantification of contaminants in blood and sea turtle eggs, searching for relationships between, carapace size and the activity of antioxidant enzymes using non-lethal techniques. The concentration of organochlorine pesticides (OCPs) in plasma of adult females and eggs of hawksbill turtles (Eretmochelys imbricata) and green turtle (Chelonia mydas) that nest in the coastal area of Campeche will be analysed. The main objective of this thesis is to evaluate the potential of natural populations as bioindicators of chemical contamination. For this, the hematological values, contamination of OCPs in the plasma and eggs of two species of sea turtles with different feeding habits and a possible maternal transfer in the hawksbill turtles were analysed. Finally, the activity of enzymatic biomarkers was correlated with the contamination by OCPs. The results showed that OCPs were found in all analysed eggs, showing higher concentrations of ΣHCH and ΣDrines in the two species selected, being found significant differences between the species and the studied years. We find significant differences between the concentrations of OCPs (ΣHCHs, ΣDrines and ΣChlordanes) of blood to eggs showed that these chemical contaminants being transferred maternally. No antioxidant enzyme had significant relationship with COPs main found in the blood (ΣDDTs, ΣHCHs and ΣDrines).

Previous monitoring studies by federal agencies such as the United States Geological Survey have shown that environmental contaminants rarely occur as single compounds but, rather, as mixtures. In aquatic ecosystems, mixtures of these compounds are often complex, sometimes containing dozens of compounds across a number of different chemical classes. Non-target aquatic organisms are frequently exposed to varying levels of contaminants based upon the physical properties of the chemicals, such as water solubility, and life-cycle habits of the individual organisms. In addition to this, past research has indicated that the presence of one class of contaminant may have an influence on the toxicities of other chemical classes. Water-only toxicity testing has historically provided a means by which researchers can rapidly determine the toxic effects of water-soluble compounds such as triazine herbicides and organophosphate insecticides. However, many legacy pesticides, such as organochlorine, and some current-use pesticides, such as pyrethroids, are strongly hydrophobic, and suspended or bedded sediments, rather than water, would generally be more appropriate matrices for monitoring. Yet sampling of sediments and quantification of residues of these pesticides is often lacking. Similarly, there have been few studies examining the toxicity of mixtures of these compounds in sediment. The first goal of this research was to examine the effects of select triazine herbicides on organophosphate insecticide toxicity utilizing water-only toxicity test with the aquatic amphipod Hyalella azteca. The second goal was to analyze an existing database of chemical concentrations using a toxicity-based screening approach in order to estimate the environmental hazard posed by mixtures of pyrethroid, organochlorine, and organophosphate insecticides in sediment to H. azteca. The third goal of this research was to examine the toxic effects of mixtures of different pyrethroid insecticides to H. azteca using compounds identified as most relevant from the screening phase of the study. The fourth goal of this research was to examine how pyrethroid and organochlorine insecticides partition between different size fractions within sediment and detritus, as well as between sediments with differing organic carbon content, and the resulting effects to compound toxicity and bioavailability. The final goal of this research was to examine potential modifications to bifenthrin sediment partitioning, toxicity, and bioaccessibility resulting from various dissolved salt concentrations in overlying water using H. azteca and Chironomus dilutus as reference organisms. Together, the individual objectives of this study provide a thorough and multi-tiered approach to determining the occurrence, environmental fate, biological effects, and bioavailability of frequently detected and co-occurring environmental contaminants in both agricultural and urban landscapes.

Os albatrozes e petréis (Procellariiformes) são aves oceânicas e migratórias de grande interesse conservacionista. Neste trabalho foram estudadas duas classes de poluentes bastante conhecidos por afetarem negativamente as aves marinhas: os poluentes orgânicos persistentes e os plásticos. Bifenilos policlorados (PCBs) e pesticidas organoclorados foram detectados no tecido adiposo, fígado e músculo de oito espécies de Procellariiformes. Apesar da grande variabilidade intraespecífica nas concentrações, os perfis de PCBs e pesticidas organoclorados foram semelhantes entre os indíviduos, com predôminância de PCBs penta, hexa e heptaclorados e do p´p-DDE. A condição corporal se mostrou um fator importante na variação e redistribuição dos organoclorados nos tecidos das aves. As análises de isótopos estáveis de carbono e nitrogênio no fígado e músculo das aves mostraram que a dieta não foi suficiente para explicar as concentrações de organoclorados nas espécies estudadas e reforçaram a influência de fatores como idade, distribuição e especifidade da dieta na contaminação por estes compostos em aves marinhas. PCBs e pesticidas organoclorados foram detectados em pellets e fragmentos plásticos encontrados no trato digestório das aves estudadas, com perfis semelhantes aos encontrados nos tecidos dos Procellariiformes. A ocorrência de poluentes orgânicos em plásticos evidencia a capacidade destes de adsorver e transportar estes compostos e reforça o potencial dos plásticos como uma fonte adicional de contaminação para os animais que os ingerem, como as aves marinhas.
Albatrosses and petrels (Procellariiformes) are migratory oceanic birds of considerable conservational interest. The aim of the present study was to evaluate two classes of pollutants that negatively affect seabirds: persistent organic pollutants and plastics. Polychlorinated biphenyls (PCBs) and organochlorine pesticides (OCPs) were detected in the adipose tissue, liver and muscle of eight species of Procellariiformes. Although organochlorine concentrations exhibited a high degree of intra-species variability, the profiles of PCBs and OCPs were similar among the individuals, with predominance of penta, hexa and heptachlorobiphenyls and p´p-DDE. Body condition was an important factor in the variation and redistribution of organochlorine compounds in the tissues of the birds. Stable isotope analysis of carbon and nitrogen in the liver and muscle revealed that diet alone was insufficient to explain the organochlorine concentrations in the species studied, suggesting the influence of factors such as age, distribution and diet specificity regarding organochlorine contamination in seabirds. PCBs and OCPs were detected in plastic fragments and pellets found in the digestive tract of Procellariiformes, with profiles very similar to those found in the tissues of the birds. The occurrence of organic pollutants in plastics demonstrates their ability to adsorb and transport these compounds and underscores the potential of plastics as an additional source of contamination in organisms that ingest these products, such as seabirds.

Ce travail s’intéresse à évaluer l’efficacité de procédés d’oxydation avancée pour l’élimination de pesticides dans l’eau, plus particulièrement l’acide 2,4-dichlorophénoxyacétique (2,4-D), un herbicide largement répandu, qui a été récemment classé comme cancérogène possible pour l’homme. Ces procédés utilisent différents types de rayonnement – UV/visible, ultrasons, rayons gamma –, seuls ou en combinaison avec des oxydants et/ou catalyseurs (ozone, peroxyde d’hydrogène, réactif de Fenton). L'influence du pH, de la dose d'oxydant, du type et de la concentration du catalyseur, du spectre d'irradiation lumineuse, de la dose de rayons gamma et de la fréquence ultrasonore est également analysée, de façon à déterminer par plans d’expériences les plages de fonctionnement optimales pour la conversion et la minéralisation du polluant. Parmi les procédés individuels, seules l’ozonation et l’oxydation Fenton homogène d’une part, la photolyse et l’irradiation gamma d’autre part permettent d’éliminer plus de 25% du carbone organique total en 1 heure. La sonolyse (à haute fréquence) apparaît comme le traitement moins performant, avec une dégradation du 2,4-D inférieure à 15% sur la même durée. Par ailleurs, des effets synergiques marqués sont mis en évidence en associant les différents types de rayonnement avec H2O2 ou le réactif de Fenton. Dans le dernier cas, le polluant est décomposé en moins de 10 minutes, tandis que le rendement de minéralisation est plus que doublé par rapport aux procédés séparés. Il en est de même pour le procédé couplé UV/O3 par rapport à l’ozonation et la photolyse seules. Ramenés à leur consommation énergétique, les traitements les plus efficaces sont, respectivement en termes de conversion et de minéralisation, l’oxydation radio-Fenton et l’oxydation photo-Fenton utilisant une lampe UV à basse pression de mercure. Par ailleurs, contrairement à la photolyse, ce dernier procédé est également activé par une lampe à arc Xenon, dont le spectre d’émission est proche de celui de la lumière du soleil. Sur la base de ces résultats, un photo-réacteur solaire à recirculation est mis en oeuvre pour traiter par oxydation photo-Fenton homogène des solutions de 2,4-D, préparées dans l’eau du robinet ou une eau résiduaire en entrée de station d’épuration. Dans les deux cas, la conversion du pesticide dépasse 95% en 1 heure et sa minéralisation 75% en 5 heures. Par ailleurs, l’eau ainsi traitée respecte tous les paramètres de qualité établis par la norme cubaine de rejet des eaux usées, en incluant une étape successive de neutralisation avec Ca(OH)2 et de filtration. Enfin, plusieurs parmi les meilleurs procédés sont évalués pour éliminer deux autres pesticides organochlorés, particulièrement toxiques et persistants dans l’environnement : le chlordécone et le bêta-hexachlorocyclohexane. Des résultats encourageants sont obtenus, avec une dégradation pratiquement totale des polluants en cinq heures, ce qui démontre l’intérêt des méthodes étudiées pour cette problématique
This work investigates the efficacy of advanced oxidation processes for the remediation of pesticidesin water, more particularly 2,4-dichlorophenoxyacetic acid (2,4-D), a widely used herbicide that hasrecently been classified as a possible carcinogen to humans. These methods use different types ofradiation - UV / visible, ultrasound, gamma rays - alone or in combination with oxidants and / orcatalysts (ozone, hydrogen peroxide, Fenton’s reagent). The influence of pH, oxidant dose, catalysttype and concentration, light irradiation spectrum, gamma radiation dose and ultrasound frequencyis also analyzed, in order to determine by experimental design the optimal operating ranges for theconversion and mineralization of the pollutant. Of individual processes, only ozonation andhomogeneous Fenton oxidation on the one hand, photolysis and gamma irradiation on the otherhand, achieve the abatement of more than 25% of total organic carbon in one hour. (High frequency)sonolysis appears as the less efficient treatment, resulting in less than 15% of 2,4-D being degradedover the same duration.In addition, marked synergistic effects are demonstrated by combining the different types of radiationwith H2O2 or Fenton’s reagent. In the latter case, the pollutant is decomposed in less than 10 minutes,while the mineralization yield more than doubles compared to the separate processes. It is the samefor UV/O3 coupled process with respect to ozonation and photolysis alone. When accounting for theirenergy consumption, the most effective treatments are, respectively in terms of conversion andmineralization, radio-Fenton oxidation and photo-Fenton oxidation using a low-pressure mercuryvaporUV lamp. Moreover, unlike photolysis, the latter method is also activated by a Xenon arc lamp,whose emission spectrum is close to that of sunlight. Bases on these results, a solar photoreactoroperating in closed loop is used to treat 2,4-D solutions, prepared with tap water or inlet stream ofwastewater treatment plant, by homogeneous photo-Fenton oxidation. In both cases, the conversionof the pesticide exceeds 95% in 1 hour and its mineralization 75% in 5 hours. Moreover, the treatedwater complies with all the quality parameters established by the Cuban norm for wastewaterdischarge, when including a subsequent step of neutralization with Ca(OH)2 and filtration.Finally, several of the best processes are evaluated to eliminate two other organochlorine pesticides,particularly toxic and persistent in the environment: chlordecone and beta-hexachlorocyclohexane.Encouraging results are obtained, with an almost complete degradation of the pollutants in fivehours, which demonstrates the value of the investigated methods for this issue
En este trabajo se estudió la factibilidad de diferentes procesos avanzados de oxidación para laeliminación de plaguicidas en aguas, específicamente el ácido 2,4-diclorofenoxiacético (2,4-D), unode los herbicidas más utilizados a nivel mundial y que recientemente ha sido clasificado comocancerígeno para el hombre. Estos procesos utilizan diferentes tipos de irradiación – UV/Visible,ultrasonido, radiación gamma – solos o en combinación con oxidantes y/o catalizadores (ozono,peróxido de hidrógeno, reactivo de Fenton). Con el objetivo de determinar las condiciones óptimaspara la degradación y mineralización del contaminante, se analizó la influencia del pH, la dosis deoxidante, el tipo y la concentración del catalizador, el espectro de irradiación luminosa, la dosis deirradiación gamma y la frecuencia y potencia ultrasónicas. Entre los procesos individuales,solamente la oxidación Fenton homogénea, la ozonización, la fotólisis y la radiación gamma lograroneliminar más de un 25% del carbono orgánico total en una hora. La sonólisis (a alta frecuencia)resultó el proceso menos eficiente, con una degradación del 2,4-D inferior al 15% en el mismotiempo.Al asociar los diferentes tipos de irradiación con el H2O2 o el reactivo de Fenton, se observaronefectos sinérgicos importantes. Para este último, el contaminante se degradó totalmente en menosde 10 minutos, mientras que la mineralización fue superior al doble de la obtenida para los procesospor separado. Lo mismo ocurrió para el proceso combinado UV/O3, respecto a los procesos defotólisis y ozonización solos. Teniendo en cuenta el consumo energético asociado a la degradacióny mineralización respectivamente, los tratamientos más eficientes fueron el radio-Fenton y el foto-Fenton utilizando una lámpara de baja presión de mercurio. Por otra parte, al contrario de la fotólisis,este último proceso se activó con una lámpara de arco de Xenon, la cual posee un espectro deemisión cercano a la luz solar. Teniendo en cuenta estos resultados, se realizó un grupo deexperimentos a escala de banco utilizando un foto-reactor solar, donde se trataron por oxidaciónfoto-Fenton homogénea disoluciones de 2,4-D preparadas con agua técnica y agua residualobtenida a la entrada de una planta de tratamiento de aguas. En los dos casos, la degradación delpesticida supera el 95% en 1 hora y su mineralización en 75% en 5 horas. Además, al incluir unaetapa de neutralización con Ca(OH)2, el agua residual tratada logra cumplir las normas de calidadestablecidas por la norma cubana de vertimiento de aguas residuales.Finalmente, algunos de los procesos con mejores resultados se evaluaron para la eliminación otrosdos pesticidas organoclorados de mayor toxicidad y persistencia en el medio ambiente: laclordecona y el beta hexaclorociclohexano. Al aplicar las técnicas de tratamiento a estoscompuestos se lograron buenos resultados, cercanos a la degradación total de ambos plaguicidasen cinco horas, lo que demuestra el interés de los métodos estudiados en esta tesis

Abstract:
Introduction Epidemiologic data supporting the role of organochlorine pesticides in pubertal development are limited. Methods Using a nested case-control design, serum collected during pregnancy from mothers of 218 girls who reported menarche before 11.5 years of age (cases) and 230 girls who reported menarche at or after 11.5 years of age (controls) was analyzed for 9 organochlorines and metabolites. We analyzed the association between in utero organochlorine concentrations and early menarche using multivariate logistic regression controlling for mother's age at menarche, or mother's prenatal BMI. Results We did not observe an association between in utero exposure to HCB, β-HCH, ϒ-HCH, p,p′-DDT, p,p′-DDE, oxychlordane or trans-nonachlor and early menarche. Conclusions This study is the first to examine the association between in utero exposure to HCB, β-HCH, ϒ-HCH, oxychlordane or trans-nonachlor and early menarche. In utero exposure to organochlorine pesticides does not appear to have a role in the timing of menarche in this study.

A maior parte dos pesticidas organoclorados (OCPs) possuem comercialização controlada ou proibida a décadas, no entanto são ainda encontrados em todos os locais do planeta. A determinação destes compostos em matrizes complexas como tecido de peixe é alvo constante de pesquisas, especialmente com o objetivo de minimizar consumo de reagentes e tempo. Este estudo objetivou desenvolver e validar um método QuECheRS com ultrassom e d-SPE para determinação por CG/ECD/MS de p,p’-DDE, p,p’-DDT, α-endosulfan, β-endosulfan, Aldrin e Dieldrin em tecidos de peixe, aplicar em amostras de tecidos de peixe comercializadas e em tecidos coletados em ambiente natural e realizar uma avaliação de risco das concentrações de OCPs detectadas nos tecidos. Três planejamentos fatoriais foram empregados para otimizar a etapa de preparo da amostra. O método desenvolvido foi validado para verificar seu desempenho e um possível efeito matriz, sendo posteriormente aplicado na extração de OCPs de músculo de Tilápia e Lambari comercializados em supermercados de Ponta Grossa e em músculo, ovas e vísceras de Lambari coletado da represa de Alagados, em Ponta Grossa. Ao final realizou-se uma avaliação de risco relacionando o consumo diário local, com massa corporal média da população e contaminação por OCPs nos tecidos analisados. Na validação o método apresentou recuperações entre 77,3% e 110,8%, com RSD inferior a 13% e os limites de detecção estiveram entre 0,24 μgkg-1 e 2,88μgkg-1, mostrando boa sensibilidade e precisão. Resíduos dos OCPs estudados foram detectados nos tecidos analisados em concentrações que variaram entre 3,4 μgkg-1 e 417,6 μgkg-1, verificando-se que os poluentes possuem uma tendência a bioacumular em maior proporção nas vísceras, do que em ovas e no músculo, a presença destes compostos nas ovas, demonstra que indivíduos da espécie analisada entram em contato com os OCPs desde a fase de embrião. Na avaliação de risco verificou-se que ao se consumir esses peixes, esses OCPs podem ao longo do tempo gerar efeitos adversos para a saúde dos consumidores. O método proposto mostrou-se efetivo na determinação de baixas concentrações de OCPs em tecidos de peixe, utilizando pequena massa de amostra (0,5 gramas), viabilizando a análise sem a necessidade de agrupamento (pool), ao mesmo tempo possibilitou a análise de tecidos, de peixe de pequena massa corporal total (Lambari), avaliando ainda a bioacumulação dos OCPs em diferentes tecidos. O método é aplicável para análise de OCPs aplicado à área toxicológica.
Most organochlorine pesticides (OCPs) are controlled or banned for decades, but they are still found everywhere in the world. The determination of these compounds in complex matrices such as fish tissue is a constant goal of research, especially with the aim of minimizing reagent consumption and time. The aim of this study was to develop and validate a Quechers method with ultrasound and d-SPE for determination of p, p'-DDE, p, p'-DDT, α-endosulfan, β-endosulfan, Aldrin and Dieldrin in fish tissues, apply on samples of commercially available fish tissues and on tissues collected in the natural environment and carry out a risk assessment of the concentrations of OCPs detected in the tissues. Three factorial designs were used to optimize the preparation stage of the sample. The method developed was validated through figures of merit and matrix effect, and it was later applied in the extraction of OCPs from Tilapia and Lambari muscle marketed in supermarkets in Ponta Grossa and Lambari muscle, eggs and viscera collected from the Alagados dam in Ponta Thick. At the end of the study, a risk assessment was performed relating local daily consumption, mean body mass of the population and OCP contamination in the tissues analyzed. In the validation, the method presented recoveries between 77.3% and 110.8%, with RSD lower than 13% and detection limits between 0.24 μg kg -1 and 2.88 μg kg -1, showing good sensitivity and precision. Residues of the OCPs studied were detected in the tissues analyzed at concentrations ranging from 3.4 μgkg-1 to 417.6 μgkg-1, with pollutants having a tendency to bioaccumulate more in the viscera than in roe and in muscle, the presence of these compounds in the eggs, demonstrates that individuals of the analyzed species come into contact with OCPs from the embryo stage. In the risk assessment it was found that there is a risk when consuming these fish, these OCPs may over time generate adverse effects on the health of consumers. The proposed method was effective in the determination of low concentrations of OCPs in fish tissues, using a small sample mass (0.5 grams), making the analysis possible without the necessity of pooling, at the same time making possible the analysis of tissues, of fish of small total body mass (Lambari), also evaluating the bioaccumulation of OCPs in different tissues. The method is promising for OCP analysis applied to the toxicological area.

Este trabalho apresenta a caracterização geoambiental de solos de uma área contaminada, localizada no município de Santo André, Estado de São Paulo, Brasil. O objetivo foi determinar parâmetros experimentais para permitir o estudo de alguns aspectos do transporte do pesticida organoclorado Hexaclorociclohexano (HCH). A área de estudo contém células com cerca de 30.000 toneladas de um material contendo HCH em elevadas concentrações. As investigações de campo demonstram que há concentrações de HCH na água subterrânea. Em 2011, foram coletadas amostras deformadas e indeformadas de solo em quatro diferentes pontos superficiais da área contaminada, e posteriormente uma amostra composta da camada de solo de alteração de rocha (SAR), a fim de responder a questionamentos quanto à mobilidade do contaminante na área. A caracterização geoambiental, baseada em ensaios laboratoriais, foi realizada por meio das seguintes análises: análise granulométrica, limites de Atterberg, densidade específica, porosidade, umidade do solo, curva de retenção de água no solo, condutividade hidráulica, ensaio de adsorção, pH, condutividade elétrica, capacidade de troca catiônica (CTC), teor de matéria orgânica e teor de voláteis. O solo superficial da área é heterogêneo e a capacidade de adsorção dos solos analisados foi menor do que o esperado. Esta caracterização subsidiou o estudo de fluxo e transporte do HCH, por meio de simulação numérica, utilizando o programa Hydrus 1D. As simulações tiveram dois objetivos diferentes: no primeiro foi avaliado os resultados da simulação em comparação à situação atual no campo; no segundo foi avaliada a sensibilidade do transporte em função da alteração dos parâmetros obtidos experimentalmente Ks; Kd; s e (Van Genuchten); e dos parâmetros obtidos na literatura. Os resultados permitiram concluir que o modelo Hydrus 1D consegue representar a contaminação do HCH no solo. O parâmetro de degradação na água foi essencial para chegar em ordens de grandeza de concentrações próximas das identificadas em campo. Os parâmetros de degradação, difusividade, dispersividade e condutividade hidráulica são os que mais afetam os resultados finais de concentração do HCH na água subterrânea e, portanto, merecem atenção maior durante as futuras investigações.
This paper presents the geoenvironmental characterization of soils from a contaminated site, located in Santo André, São Paulo State, Brazil. The objective was to determine experimental parameters to allow the study of some aspects of transport of the organochlorine pesticide hexachlorocyclohexane (HCH). The study site contains trenches with about 30,000 tons of a material containing high concentrations of HCH. The site investigations show that there are HCH concentrations in groundwater. In 2011, disturbed and undisturbed soil samples were collected at four different surface points of the contaminated site, and then a sample of soil from rock alteration layer (SAR) was also composed for characterization in order to answer questions concerning the mobility of contaminant in the site. The environmental characterization based on laboratory tests, was performed by the following analyzes: grain size distribution, Atterberg limits, specific gravity, porosity, moisture content, water retention curve, hydraulic conductivity, pH, electrical conductivity, cation exchange capacity (CEC), organic matter content and volatile content. The surface soil of the area is heterogeneous and the adsorption capacity of the soils analyzed were lower than expected. This characterization allowed the study of flow and transport of HCH through numerical simulation using the Hydrus 1D software. The simulations had two different goals, the first was evaluated the simulation results compared to the current situation in the field. In a second step the sensitivity of transport due to the change of the parameters obtained experimentally Ks; Kd; s and (Van Genuchten); and parameters obtained from the literature. The results showed that the Hydrus 1D model simulates HCH contamination in soil. Water degradation parameter was essential to get orders of magnitude similar to those identified in field concentrations. The degradation, diffusivity, hydraulic conductivity and dispersivity parameters are those that most affect the final results of concentration of HCH in groundwater and therefore deserve more attention in future investigations.

Apesar da pequena influência antrópica direta em seu território, a Antártida não pode ser considerada livre do alcance de poluentes. No presente estudo, avaliou-se a distribuição e transferência de poluentes orgânicos (PCBs, pesticidas organoclorados e PBDEs) em compartimentos abióticos (neve e água de degelo) e bióticos (liquens, musgos, invertebrados, peixes, ovos de aves e tecidos de aves e pinípedes, para os quais também foi aplicada análise isotópica de nitrogênio-15 e carbono-13) do ecossistema da Baía do Almirantado. Os compartimentos abióticos tiveram concentrações entre 1,4 e 156 pg L-1, apenas para grupos de organoclorados, com variação intra-sazonal importante e compatibilidade com níveis tróficos inferiores e amostras de vegetação, que por sua vez refletiram a importância de fontes secundárias de poluentes. Nos animais, grosso modo, os compostos prevalecentes (em ng g-1 peso úmido) foram PCBs, (6,82-1821), HCB (0,060-136) e DDTs, (0,410-524), níveis de duas a três ordens de grandeza superiores aos encontrados em vegetação. PBDEs ocorreram, de modo geral, em níveis uma ou duas ordens de grandeza abaixo dos organoclorados. A análise isotópica mostrou-se útil como ferramenta indicadora de nível trófico e origem de matéria orgânica consumida, correlacionando-se quantitativa e qualitativamente com os poluentes orgânicos pela indicação de fatores como stress nutricional e mudança de área de forrageio e/ou de dieta. Como alternativa, foi proposto um novo indicador de biomagnificação baseado no peso molecular dos PCBs, que apresentou resultados estatisticamente superiores ao ?15N na maioria dos casos.
Despite the small antropic direct influence on its territory, Antarctica cannot be considered out of pollutants\' reach. The present study evaluated the distribution and transfer of organic pollutants (PCBs, organochlorine pesticides and PBDEs) in both abiotic (snow and melting water) and biotic (lichens, mosses, invertebrates, fishes, bird eggs and bird and pinnipeds\' tissues, for which was also applied carbon-13 and nitrogen-15 stable isotopes analyses) compartments of the ecosystem from Admiralty Bay. Abiotic compartments showed concentrations between 1,4 and 156 pg L-1, only for organochlorine groups, with considerable intraseasonal variation and compatibility with lower trophic levels and vegetation samples, which in turn reflected the importance of secondary pollutants sources. In animal samples, grosso modo, the prevailing compounds were (in ng g-1 wet weight) PCBs (6,82-1821), HCB (0,060-136) and DDTs, (0,410-524), levels from two to three orders of magnitude superior to the ones found in vegetations. PBDEs occurred, in a general way, in levels from one to two orders of magnitude lower than the organochlorines. Stable isotope analyses proved useful as an indicator tool for trophic level and consumed organic matter, correlating itself in a qualitative and quantitative way with the organic pollutants by indication of factors as nutritional strees and variation in foraging areas and/or diet. As an alternative, a new biomagnifications indicator based on PCBs molecular weight was proposed, showing results statistically superior to the ?15N in the majority of the cases.

The organochlorine pesticide endosulfan has been shown to be highly toxic to fish and there is some evidence to support that it may act as an endocrine disrupting chemical. Juvenile rainbow trout (Oncorhynchus mykiss) were caged at 4 sites in Oshawa Creek during the fall and spring of 2008 and 2009 while another group was intra-peritoneal injected in the laboratory with varying concentrations (ppm) of endosulfan. Plasma vitellogenin (VTG) levels, liver ethoxyresorufin-O-deethylase (EROD), citrate synthase (CS), lactate dehydrogenase (LDH), and brain acetylcholine esterase (AChE) (caged fish only) enzymatic activities were measured. Trout injected with endosulfan experienced an increase of the anaerobic (LDH activity) and a decrease of the aerobic (CS activity) metabolic pathways, while male VTG levels increased. Since it was a singular injection, VTG results have to be confirmed. Fall caged trout showed increased EROD activity and inhibited AChE activity while those caged in the spring experienced an unexpected exposure to the lampricide 3-Trifluoro-Methyl-4-Nitro-Phenol (TFM) which disrupted metabolic parameters (inhibited CS and increased LDH activity). Both fall and spring caged trout experienced no induction of VTG activity. Further research is needed since the spring exposure was altered due to the unplanned TFM treatment and thus did not represent a valid temporal replicate.

Abstract:
Dans ce travail de thèse, nous avons analysé les résidus des plusieurs contaminants anthropiques dans des prédateurs de la chaîne alimentaire aquatique et des proies de la chaîne terrestre. En effet, la loutre, le grand cormoran et le silure sont prédateurs piscivores au sommet de la chaîne alimentaire, les pesticides organochlorés, les PCBs et les éléments traces métalliques (plomb, cadmium, cuivre et mercure) ont été détectés dans tous les foies de ces prédateurs prélevés dans les bassins de la Loire en France. Les PCBs sont les organochlorés les plus répandus dans les trois espèces, suivi des DDTs. Les valeurs maximales de PCBs ont atteint 1,61, 1,96 et 0,28 mg/kg de poids frais pour les cormorans, les loutres et les silures, respectivement. La concentration du mercure a dépassé la limite toxique pour plus de 40 % des cormorans analysés. L‘effet « cocktail » de ces contaminants sur les trois espèces n‘est pas étudié dans cette thèse. Cependant, Les concentrations des organochlorés et des éléments traces dans ces prédateurs sont généralement modestes ou faibles. Ainsi, Il nous semble que ces contaminants ne constituent pas une menace pour la conservation de ces espèces à court ou moyen terme. Par ailleurs, dans le réseau trophique terrestre, nous avons trouvé que trois rodenticides anticoagulants la chlorophacinone, la bromadiolone et le brodifacoum s‘accumulent dans les limaces de la manière rapide et stable au cours de temps, par contre aucune mortalité n‘a été observée chez les limaces. Le brodifacoum a été détecté dans plus de 90 % des limaces prélevées sur le terrain, et nous avons constaté que ces limaces représentaient un risque élevé d'empoisonnement secondaire pour trois de leurs prédateurs, le hérisson européen, la musaraigne commune et l'étourneau sansonnet. On pourrait donc confirmer que les invertébrés constituent une source non négligeable de rodenticides pour leurs prédateurs.L‘objectif de cette thèse est de montrer l‘influence du régime alimentaire sur la contamination des proies, des prédateurs et d‘évaluer les risques toxicologiques pour ces espèces
In this work, we analyzed the residues of several anthropogenic contaminants in predators of the aquatic food chain and prey of the terrestrial chain. The otter, the great cormorant and the catfish are piscivorous predators at the top of the food chain. Organochlorine pesticides, PCBs and trace elements (lead, cadmium, copper and mercury) were detected in all these predators collected from the basins of the Loire in France. Residue level of PCBs in these predators was the highest followed by DDTs and maximal PCB values reached 1.61, 1.96, and 0.28 mg/kg wet weight for cormorants, otters, and catfish, respectively. The concentration of mercury exceeded the toxic thresholds for more than 40% of the cormorants analyzed. The possible combined effect of these contaminants on the three species has not been studied in this thesis. However, the concentrations of organochlorines and trace elements are generally modest or low. Thus, it seems that these contaminants do not constitute a major threat to the conservation of these species for the short or medium term. Furthermore, in the terrestrial food chain, we found that three anticoagulant rodenticides chlorophacinone, bromadiolone and brodifacoum accumulate in slugs in a stable way over the time, whereas no mortality was observed in slugs. Brodifacoum was detected in more than 90% of the slugs collected in the field, and we found that these slugs represented a high risk of secondary poisoning for three of their predators, European hedgehog, common shrew and European starling. Therefore, it could be confirmed that invertebrates constitute a non-negligible source of rodenticides for their predators.The objective of this thesis is to show the influence of the diet on the contamination of prey, predators and to evaluate the toxicological risks for these species

O aumento das intervenções antropogênicas, nos âmbitos marinho e costeiro representa grande risco aos ecossistemas de suporte de numerosas populações de animais como as tartarugas marinhas. Desequilíbrios no balanço ecológico desses habitats afetam negativamente a sobrevivência de quelônios de longa vida, como a Chelonia mydas, seriamente ameaçada de extinção. As ameaças à espécie incluem captura incidental por pesca, doenças e degradação dos habitats de alimentação e nidificação. Uma das principais doenças é a fibropapilomatose (FP) ou Green Turtle Fibropapiloma Disease (GTFD), afecção tumoral e de caráter infeccioso que acomete predominantemente espécimes juvenis. Numerosas hipóteses têm surgido a respeito da etiologia da doença, mas ainda nada é conclusivo. Contudo existe o consenso de que a etiologia da mesma seja \"multifatorial\" envolvendo um α-herpesvírus como agente etiológico primário e a participação de diversos co-fatores genéticos, nutricionais e ambientais. No Brasil a prevalência média de FP en C. mydas varia conforme a região, existindo localidades nas quais não há registro da doença. O aumento da prevalência da FP tem sido implicada à degradação dos habitats marinhos, relacionado por sua vez a contaminantes ambientais como os organoclorados e seus metabólitos. Estes compostos caracterizam-se por serem altamente persistentes, bioacumulando e biomagnificando no ambiente marinho através do tempo. Além disso, são conhecidos por causarem inúmeros efeitos na saúde, interferindo na imunidade do indivíduo e favorecendo a manifestação de doenças, inclusive sendo apontados como agentes carcinogênicos. No presente trabalho foi desenvolvida uma técnica de extração e purificação baseada no método QuEChERS (Quick, Easy, Cheap, Effective, Rugged and Safe) para monitoramento pela cromatografia gasosa com captura de elétrons (GC-μECD) dos agrotóxicos clorados: α-BHC, β-BHC, Heptacloro, Dicofol, α-Endosulfan, β-endosulfan, Sulfato endosulfan, pp\' -DDD, op\' -DDD, pp\' -DDE, op\' -DDE e Mirex nos tecidos adiposo e hepático de 64 tartarugas-verdes juvenis (Cumprimento Curvo da Carapaça - CCC = 38,68±5,41cm) acometidas ou não pela FP (24 com FP vs. 40 sem FP), espécimes que vieram a óbito por captura incidental em redes de pesca ou foram encontradas encalhadas nas praias de Ubatuba/SP, Praia Grande/SP e Vitória/ES. Objetivou-se esclarecer possíveis correlações das suas concentrações com variáveis como sexo, tamanho (CCC), índice de condição corporal (ICC), severidade; índice de fibropapilomatose (FPI) e escore de fibropapilomatose (FPS) e ocorrência da doença em algumas localidades da região sudeste do país com diferentes taxas de prevalência da FP, na tentativa de inferir o papel dos mesmos na patogenia desta afecção. Foi observada correlação significativa entre a porcentagem lipídica entre as amostras dos tecidos adiposo e hepático, assim como entre a concentração total de organoclorados (ΣOCS) nos tecidos adiposo e hepático. Embora observadas algumas diferenças na presença ou ausência de alguns contaminantes, estas foram dispersas e não determinantes, ou seja, não permitiram estabelecer um padrão em relação aos diferentes fatores do histórico individual. Não foram encontradas diferenças significativas entre a ΣOCS para ambos os tecidos, em relação ao sexo do animal ou localidade, que permitissem inferir alguma relação entre o grau de contaminação e a variação regional da prevalência da FP. Também não foi possível identificar correlações significativas das concentrações dos poluentes em relação à presença da FP nem entre o FPI e o FPS e a ΣOCS nos tecidos adiposo e hepático. Tampouco foi possível identificar correlações significativas entre o ICC ou CCC e a ΣOCS. Os resultados deste trabalho apresentam um registro temporal do perfil de contaminação nas tartarugas-verdes de vida livre da costa sudeste, confirmando a biodisponibilidade de OCs nas diferentes regiões avaliadas. Embora não tenha sido encontrada uma correlação direta entre a concentração de OCs e a Fibropapilomatose, este estudo demonstra que as tartarugas verdes oriundas da costa sudeste do Brasil estão expostas a níveis significativos de pesticidas organoclorados os quais poderiam ter algum papel na etiologia multifatorial da doença. Estes contaminantes ambientais podem agir afetando a taxa de prevalência de FP, agindo possivelmente como agentes imunossupresores ou através da genotoxicidade e promoção tumoral. Estudos futuros serão desenvolvidos para melhor caracterizar a ação dessas substâncias e seus efeitos nas tartarugas marinhas e para esclarecer como outros fatores podem influenciar esses resultados.
The increase of anthropogenic impacts on both marine and coastal environments is threatening the ecosystems of numerous animal populations, including sea turtles. Disturbances in the ecological balance of these habitats negatively affect the survival of longlived chelonids such as green turtles (Chelonia mydas), which are already considered endangered. Threats to this species include incidental capture by fisheries, habitat degradation of nesting and feeding areas, and diseases. One of the most relevant diseases is fibropapillomatosis (FP) or Green Turtle Fibropapilloma Disease (GTFD), an infectious tumoral disease that predominantly affects juveniles. Numerous hypotheses have been proposed to explain this diseases aetiology, and while none has been conclusively demonstrated there is a consensus that it is multifactorial and involves a -herpesvirus as the primary etiologic agent along with genetic, nutritional and environmental cofactors. The prevalence of FP in C. mydas in Brazil varies among regions, with locations in which the disease remains unreported. Prevalence has been correlated to the degree of degradation of marine habitats, which in turn is related to the presence of environmental contaminants such as organochlorines and their metabolites. These compounds are difficult to break down and persist, bioaccumulate and biomagnify in marine environments over long periods of time. They are also known to produce numerous health effects in the organisms in which they accumulate, hampering immunity and favouring the manifestation of opportunistic disease, with some of these compounds being also carcinogens. In this study we developed extraction and purification technique based on the QuEChERS method (Quick, Easy, Cheap, Effective, Rugged and Safe) and used gas chromatography with electron capture (GC-μECD) to detect and quantify organochlorine (α-BHC, β-BHC, Heptachlor, Dicofol, α-endosulfan, β-endosulfan, Endosulfan sulphate, pp\' -DDD, op\' -DDD, pp\' -DDE, op\' -DDE, and Mirex) in adipose and hepatic tissue samples from 64 juvenile green sea turtles [Curved Carapace Length - CCL = 38.68 ± 5.41 cm] affected or not by FP (24 with FP vs. 40 without FP). These specimens died due to incidental capture by fishing nets or were found dead on beaches of Southeast Brazil: Ubatuba / SP, Praia Grande / SP and Vitória / ES. Aiming to clarify the role of the organochlorines in the pathogenesis of FP, we attempted to establish correlations between the concentration of these compounds and variables such as sex, size (CCL), Body Condition Index (BCI) and disease severity [fibropapillomatosis index (FPI) and fibropapillomatosis score (FPS)], and local FP prevalence. A significant correlation was observed between the lipid percentage of adipose and hepatic samples, as well as between adipose and hepatic organochlorine total concentrations (ΣOCS). Although some differences were observed regarding the presence or absence of contaminants in relation to characteristics of the individual history, it was not possible to establish patterns that reflected biological significance. For both adipose and hepatic tissues, no significant differences were found in the ΣOCS with regards to individual sex or capture locality that would allow inferring relationships between the individual contamination levels and the regional variation on the prevalence of FP. It was also not possible to identify significant relationships between each pollutant concentration in the adipose or hepatic tissue and the presence of FP. No correlation was observed between ΣOCS in adipose or hepatic tissues and FPI, FPS, BCI and CCL. The results provide a contamination profile of free-ranging green turtles of the southeast coast during the study period, confirming the bioavailability of OCs in the different locations evaluated. Although a direct correlation between the concentration of OCs and fibropapillomatosis was not found, this study demonstrates that green turtles from the southeast coast of Brazil are exposed to significant levels of organochlorine pesticides that could play a role in the multifactorial etiology of the disease. These contaminants may affect the prevalence of FP, possibly acting as immunosuppressive agents or through genotoxicity and tumor promotion. Future studies will be conducted to better characterize the role of these substances and their effects on sea turtles and to clarify how other factors that may influence their impacts.
