Abstract:
Performance can be interpreted as an extra role such as cooperation, helping colleagues and superiors, and contributing to organizational goals. Culture in an organization can improve performance, because it can create positive motivation to give the best ability to the organization. This study is looking at the influence of organizational culture and work motivation on performance. Organizational culture is defined as the way a person behaves in an organization, and work motivation is the achievement that someone produces because of a drive to complete work in the most possible effective and efficient time to get better results and be able to adjust to the work climate. The purpose of this study is to determine the effect of organizational culture and work motivation on the performance of the members of Bhabinkamtibmas (a police sergeant responsible for establishing and maintaining security and public order in a village) of Resort Police. This research uses quantitative approach with survey method. T he popul ation in this study are 53 members of Bhabinkamtibmas of Bungo Resort Police , using total sampling. Th e data collection technique uses questionnaire consisting of 45 - statement items, which are from 9 indicators, and the measurement uses Likert scale with the answer ran ge of 1 to 5. The results show that organizational culture has a significant effect o f 93.7% on performance variable and w ork motivation has a significant effect of 87.4% o n the performance variable . The re fore, it can be explained that the organizational culture variable (X1) has a greater influence on the performance variable (Y) than the motivation variable (X2). Organizational culture and motivation simultaneously provide a significant influence o f 94.1% on the p erformance. Performance can be improved if the organ izational culture is healthy, thus can increase the motivation of members. Key words: corruption, culture, motivation, performance

Abstract:
The actin cytoskeleton plays a fundamental role in all eukaryotic cells—it is a major determinant of cell morphology and polarity and the assembly and disassembly of filamentous actin structures provides a driving force for dynamic processes such as cell motility, phagocytosis, growth cone guidance and cytokinesis. The ability to reorganize actin filaments is a fundamental property of embryonic cells during development; the shape changes accompanying gastrulation and dorsal closure, for example, are dependent on the plasticity of the actin cytoskeleton, while the ability of cells or cell extensions, such as axons, to migrate within the developing embryo requires rapid and spatially organized changes to the actin cytoskeleton in response to the external environment. W ork in mammalian cells over the last decade has demonstrated the central role played by the highly conserved Rho family of small GTPases in signal transduction pathways that link plasma membrane receptors to the organization of the actin cytoskeleton.

Abstract:
The purpose of this study as to determine the effect of the implementation of the bid value policy below 80 % 0n the quality of construction work in Bandung Regency. The method used in this study uses descriptive analysis techniques that are causal associative, which describes the effect of the independent variable on the dependent variable using statistical analysis t-test and f-test. The sample in this study used a probability sampling technique to the Commitment Making Officer (PPK) of construction service activities. The findings show that the implementation of policies in term of organization, interpretation, and application has a significant effect in a positive direction, meaning that even if the policy implementation is improved, and the variable dimensions of the organization are improved, the interpretation is improved, and the application is improved, it will affect the quality of construction ork for the better. Where the percentage value of the coefficient of determination (Adjusted R-Square) is obtained at 87,7% while the remaining (1-R2)=12,3% is explained by other causes outside the model, with the magnitude of the influence of each variable for the organization 18,4%, interpretation 46,6%, and application 23,5%.

Abstract:
Background: Organizational readiness for implementation (ORI) is a barrier to disseminating recovery-oriented evidence-based practices for people with severe mental illnesses. However, there is a knowledge gap regarding implementation strategies and specific actions related to those strategies that may enhance ORI across organizations. The Transtheoretical Model (TTM) is a well-established stage-based model used to support organizational change. In TTM, the first three stages—Pre-contemplation, Contemplation, and Preparation—represent the pre-implementation phase, during which ORI should be developed. This study explored ORI enhancement strategies reported as useful by different stakeholders in the field of community mental health (CMH) and tested the applicability of the TTM readiness stages as an organizing framework to direct ORI development. Methods: The study included the following two phases: (1) a qualitative exploration of ORI enhancement actions reported by various stakeholders in the CMH field ( N = 16) based on their experience in successful implementation projects and (2) a consolidation process performed by the research team to create coherent groups of actions and their overarching implementation strategies per readiness stage. Results: A comprehensive list of strategies and related actions was created. The strategies and actions correspond with each of the first three stages of the TTM. Stakeholders differed in their methods of facilitating engagement across the organization and in their level of involvement in developing ORI. Conclusions: This study is a first step in conceptualizing a systematic process for developing ORI using the TTM as a theoretical framework. Future studies should test the transferability and effectiveness of this repository to other implementation efforts, expend the lists per stage, and further explore differences between stakeholders’ role in ORI development. Plain language abstract: Interventions that were proven effective in supporting the recovery of people with mental illnesses are not sufficiently available in the field of community mental health. One barrier to getting those interventions implemented is the lack of willingness and preparedness of organizations to change their practice, also referred to as organizational readiness for implementation (ORI). Little is known about how to enhance ORI to increase the availability of interventions. This study explored actions used to enhance ORI by people in different roles who successfully implemented new recovery-oriented interventions in community mental health settings. The actions were organized into three stages of readiness development based on the Transtheoretical Model (TTM) of behavioral change. Differences were found between administrators, consultants, supervisors, and providers in the type and number of strategies they described. The results show the applicability of the TTM as an organizing framework for ORI development and provide sets of strategies and specific actions to support different readiness needs across the organization.

Abstract:
As globalization and IT application facilitate organizations to develop without boundary in the era of the Industrial Revolution, innovations and changes have become the new normal of organizations. In this background, psychological boundaries can help employees enhance their sense of identity and sense of belonging to the organization, which is an effective way to improve the organizational effectiveness and the most essential source for org-improvement. This paper teases out the domestic and foreign research results pertaining to the connotation, measurement, and influencing factors of psychological boundaries. The perspective of the psychological boundaries provides new ideas for the managers in the era of the Industrial Revolution to establish a long-term “Heart-Central” management mechanism which reveals the essence of management problems and leads management back to its origin.

Statutory funds have been created in India to provide welfare facilities with a view to improve the quality of life of unorganized and scattered workers like those in.mica, iron ore, and manganese ore mines and the beedi industry. These funds are administered by country-wide welfare organizations with several zonal offices. The paper presents case studies of organization and management of sectoral programmes like health, water supply, housing, and education. These suggest a mis-match between the structure and administrative practices and the goals of the programme~ The welfare activities were also not in accordance with the national health policy. The study suggests the need for organizational changes, bureaucratic reorientation, training of personnel, and changes in the approach and content of the various welfare programmes.

Abstract:
Purpose The purpose of this paper is to present a degendered organizational resilience model challenging current and dominant conceptualizations of organizational resilience by exploring how gendered organizational power structures, language and practices of everyday organizational life interplay and limit inclusive constructions of organizational resilience. Design/methodology/approach The degendered organizational resilience model was developed using Acker’s (1990) model of gendered organizations, Martin’s (2003) gendering practices, Lorber’s (2000) degendering and other feminist research on gendered organizations. The purpose of the model is to explore power structures, practices and language within the organizational context during conditions requiring organizational resilience. Findings A conceptual model for analyzing the theoretical development of organizational resilience is presented. The model analyzes the following three different aspects of organizations: power structure, to identify which resilient practices receive status based on established gendered organizational hierarchies and roles; actions, to identify how resilience is enacted through practices and practicing of gender; and language, to identify how and what people speak reinforces collective practices of gendering that become embedded in the organization’s story and culture. Practical implications The degendered organizational resilience model offers a process for researchers, managers and organizational leaders to analyze and reveal power imbalances that hinder inclusive theoretical development and practices of organizational resilience. Social implications The degendered organizational resilience model can be used to reveal power structures, gendered practices and language favoring normative masculine organizational practices, which restrict the systemic implementation of inclusive democratic practices that incorporate and benefit women, men and other groups subject to organizational subordination. Originality/value This paper offers an original perspective on the theoretical development of organizational resilience by proposing a degendering model for analysis. A feminist perspective is used to reveal the gendered power structures, practices and language suppressing the full range of resilient qualities by restricting what is valued and who gives voice to resilient processes that lead to resilient organizations.

Abstract:
This paper is a theoretical description of workforce diversity (WD), specifically highlighting its significance in a wide array of organizational outcomes. The paper conceptualizes diversity comprising the workforce in terms of similarities and dissimilarities regarding age, gender, ethnicity, sexual orientation, and religion. Workforce diversity was observed as a phenomenon whose adoption transcends the compelling forces of government legislations but remains indispensable as a result of the incontestable complications associated with it. This paper proposes that it is imperative for organizations to recognize and foster an enabling environment that is inclusive and appreciative of individual uniqueness. Such an environment increases employee output and ultimately encourages activeness and peaceful work relations across the organization.

Abstract:
A virtual organization is a community of people who interact together socially on a technical platform. These kinds of communities are built on a common interest, a common problem or a common task of its members that is pursued on the basis of implicit and explicit codes of behavior. The six dimensions that are normally used to analyze virtual organizations are the use of technologies, sense of belonging, success factors, level of trust from members, virtual community management, and contents of the virtual community. The virtual organizations defined in the literature are not defined separately for non-profit research virtual organization. Here we present analysis of non-profit research virtual organization, European Working Group on Operational Research for Development (EGW ORD). This paper provides a summary of achievements and challenges faced in building a virtual organization. This kind of analysis plays a vital role in establishing new non-profit virtual organizations to serve the research community in their field of interest. It is also helpful to the group in broadening its presence and involving more researchers, practitioners and students in the field of operational research.

A selective replicative pressure occurs during the evolution of simian virus 40 variants. When the replication origin is duplicated as an inverted repeat, there is a dramatic enhancement of replication. Having regulatory sequences located between the inverted repeat of ori magnifies their enhancing effect on replication. A passage 20 variant and a passage 45 variant containing three pairs of an inverted repeat of ori replicated more efficiently than a passage 13 variant containing nine copies of ori arranged in tandem. A 69-base-pair cellular sequence inserted between inverted repeats of ori of both passage 40 and 45 variants enhanced simian virus 40 DNA replication. Differences in replication efficiencies became greater as the total number of replicating species was increased in the transfection mixture, under conditions where T antigen is limiting. In a competitive environment, sequences flanking the replication origin may be inhibitory to replication.

Abstract:
Purpose The aim of this study is to analyze the capitals moved in decisions about sustainability in narratives from and referring to Samarco Minerações, S.A. under a perspective guided by Bourdieusian sociology. Design/methodology/approach Oriented by historicist ontology and historical epistemology, this research is classified as qualitative, descriptive and documentary, with narrative analysis and case study. The selected organization-case was Samarco Minerações, S.A. The documentary sources considered were sustainability reports, social networks channels and news published in Brazilian newspapers of high circulation. The collected information was submitted to the narrative analysis method. Findings Samarco Minerações, S.A. maintained the sustainability posture before and after the ore tailings dam rupture. The decision models adopted (decentralized) and the moved capitals (economic and technological) after the ecocide revealed a change in the organizational practices in front of a new instability scenario, and the organization’s attempt to reach acknowledgment, legitimacy and power. Social implications The organization was selected because of its involvement in an ecocide. The crime generated economic (suspension of tax collection caused by the organizational inactivity), social (unemployment and deaths) and environmental (iron ore tailings contaminated the region’s ecosystem) impacts. Originality/value The greatest value and contribution this paper offers is an alternative intermediary methodological approach using Bourdieusian micro-sociology to analyze narratives based on capitals dynamics and doxa. This theoretical and methodological approach can prove fruitful for further research in sustainability studies on other topics, and even in other fields.

Abstract:
New World Wide Web (web) mapping techniques may inform and ultimately facilitate meaningful participation in current science and technology debates. The technique described here “landscapes” a debate by displaying key “webby” relationships between organizations. “Debate-scaping” plots two organizational positionings—the organizations' inter-hyperlinking as well as their discursive affinities. The underlying claim is that hyperlinking and discursive maps provide a semblance of given socio-epistemic networks on the web. The climate change debate on the web in November 1998 serves as a test case. Three findings are reported. First, distinctive .com, .gov and .org linking styles were found. Second, organizations take care in making hyperlinks, leading to the premise that the hyperlinks (and the “missing links”) reveal which issue and debate framings organizations acknowledge, and find acceptable and unacceptable. Finally, it was learned that organizations take substantive positions and address other organizations' positions. Thus, we found the makings of a “debate” that may be mapped. Scenarios of use to support new public participation techniques and experiments are discussed by way of conclusion.

Abstract:
Some rules on gene recognition and ORF organization in theSaccharomyces cerevisiaegenome are demonstrated by statistical analyses of sequence data. This study includes: (a) The random frame rule—that the six reading frames W1, W2, W3, C1, C2 and C3 in the double-stranded genome are randomly occupied by ORFs (related phenomena on ORF overlapping are also discussed). (b) The inhomogeneity rule—coding and non-coding ORFs differ in inhomogeneity of base composition in the three codon positions. By use of the inhomogeneity index (IHI), one can make a distinction between coding (IHI > 14) and non-coding (IHI ≤ 14) ORFs at 95% accuracy. We find that ‘spurious’ ORFs (with IHI ≤ 14) are distributed mainly in three classes of ORFs, namely, those with ‘similarity to unknown proteins’, those with ‘no similarity’, or ‘questionable ORFs’. The total number of spurious ORFs (which are unlikely to be regarded as coding ORFs) is estimated to be 470. (c) The evaluation of ORF length distribution shows that below 200 amino acids the occurrence of ATG initiator ORFs is close to random.

Abstract:
Purpose The purpose of this paper is to understand the crisis communication strategies used by the Caribbean medical tourism industry in the 2017 hurricane season, and also evaluate the quality of the disaster communication messages delivered via digital mediums. Design/methodology/approach This study includes a comprehensive, qualitative content analysis of 149 risk and crisis messages from 51 healthcare organizations distributed through digital media. The medical tourism providers (MTPs) include hospitals, medical tourism facilitators, practitioners/private physicians, specialty clinics, and dental and cosmetic providers. Findings Nearly half of the MTPs included in the data set delivered no post-disaster information to external audiences. The most prominent post-disaster message strategy utilized was conveying operational messages. Furthermore, an unexpected finding was the sheer magnitude of unrelated health-oriented and promotional destination marketing content disseminated before, during and after these events. Research limitations/implications This analysis excludes internal organizational channels of communication which may have been used to communicate risk and crisis messages during these events (i.e. employee e-mails, announcements made through intercom systems, etc.). Our analysis does not include content disseminated through medical tourism forums (i.e. Realself.com, Health Traveler’s Forum, FlyerTalk Forum). Practical implications Small-scale MTPs can improve on any weaknesses through proactive planning and preparation by creating organizational goals to complete basic crisis communication training courses and in doing so support the applied professional development of disaster and crisis responders in the Caribbean region. Second, MTPs exposed to similar risks of natural disasters may use these findings for comparative analysis purposes to support their own organizational planning. Finally, this study supports the continued utility of the National Center for Food Protection & Defense guidelines for analyzing and evaluating organizational performance. Originality/value Currently much of the academic scholarship of applied disaster communication narrowly focuses on the response strategies of one organization, or analyzes one social media platform at a time (i.e. Twitter). A strength of this analysis is the inclusion of an organizational sector (i.e. Caribbean medical tourism providers) and the range of platforms from which the content was captured (e.g. websites, org. blogs and social media networks).

Abstract:
This study aimed at an investigation of the structure, ecology and mapping of mixed communities with the participation of spruce, pine and broad-leave trees in one of the regions of broad-leave–coniferous zone. Despite the long history of the nature use of the study area, including forestry practices (Kurnayev, 1968; Rysin, Saveliyeva, 2007; Arkhipova, 2014; Belyaeva, Popov, 2016), the communities kept the main features of the indigenous forests of the broad-leave–coniferous zone — the tree species polydominance of the stands, the multilayer structure of communities and the high species diversity. In the course of field works in the southwestern part of the Moscow Region (2000–2016) 120 relevés were made. Spatial structure, species composition as well as cover values (%) of all vascular plants and bryophytes were recorded in each stand. The relevés were analysed following the ecology-phytocenotic classification approach and methods of multivariate statistical analysis that allowed correctly to differentiate communities according the broad-leave species participation. The accuracy of the classification based on the results of discriminant analysis was 95.8 %. Evaluation of the similarity of the selected units was carried out with the help of cluster analysis (Fig. 12). Clustering into groups is performed according to the activity index of species (A) (Malyshev, 1973) within the allocated syntaxon using Euclidean distance and Ward’s method. The classification results are corrected by DCA ordination in PC-ORD 5.0 (McCune, Mefford, 2006) (Fig. 1). Spatial mapping of forest cover was carried out on the basis of ground data, Landsat satellite images (Landsat 5 TM, 7 ETM +, 8 OLI_TIRS), digital elevation (DEM) and statistical methods (Puzachenko et al., 2014; Chernenkova et al., 2015) (Fig. 13 а, б). The obtained data and the developed classification refine the existing understanding of the phytocenotic structure of the forest cover of the broad-leave–coniferous zone. Three forest formation groups with different shares of broad-leave species in the canopy with seven groups of associations were described: a) coniferous forests with broad-leave species (small- and broad-herb spruce forests with oak and lime (1)); broad-herb spruce forests with oak and lime (2); small- and broad-herb pine forests with spruce, lime, oak and hazel (3); broad-herb pine forests with lime, oak and hazel (4)), b) broad-leave–coniferous forests (broad-herb spruce–broad-leave forests (5)), and c) broad-leave forests (broad-herb oak forests (6), broad-herb lime forests (7)). In the row of discussed syntaxa from 1 to 7 group, the change in the ratio of coniferous and broad-leave species of the tree layer (A) reflects regular decrease in the participation of spruce in the plant cover (from 66 to 6 %; Fig. 3 A1, A2) and an increase in oak and lime more than threefold (from 15 to 65 %; Fig. 4 a). Nemoral species predominate in the composition of ground layers, the coverage of which increases (from 40 to 80 %) in the range from 1 to 7 group, the coverage of the boreal group varies from 55 to 8 % (Fig. 11) while maintaining the presence of these species, even in nemoral lime and oak forests. In forests with equal share of broad-leave and coniferous trees (group 5) the nemoral species predominate in herb layer. In oak forests (group 6) the species of the nitro group are maximally represented, which is natural for oak forests occurring on rich soils, and also having abundant undergrowth of hazel. Practically in all studied groups the presence of both coniferous (in particular, spruce) and broad-leave trees in undergrowth (B) and ground layer (C) were present in equal proportions (Fig. 3). This does not confirm the unambiguity of the enrichment with nemoral species and increase in their cover in complex spruce and pine forests in connection with the climate warming in this region, but rather indicates on natural change of the main tree species in the cenopopulations. Further development of the stand and the formation of coniferous or broad-leave communities is conditioned by landscape. It is proved that the distribution of different types of communities is statistically significant due to the relief. According to the results of the analysis of remote information, the distribution areas of coniferous forests with broad-leave species, mixed and broad-leave forest areas for the study region are represented equally. The largest massifs of broad-leave–coniferous forests are located in the central and western parts of the study area, while in the eastern one the broad-leave forests predominate, that is a confirmation of the zonal ecotone (along the Pakhra River: Petrov, Kuzenkova, 1968) from broad-leave–coniferous forests to broad-leave forests.

Abstract:
The genome of Grapevine leafroll-associated virus-5 (GLRaV-5) was cloned, and the sequence of 4766 nt was determined. Degenerate oligonucleotide primers designed from the conserved closterovirus heat shock 70 protein (HSP 70) homologue were used to obtain viral-specific sequences to anchor the cloning of the viral RNA with a genomic walking approach. The partial nucleotide (nt) sequence of GLRaV-5 showed the presence of four open reading frames (ORF A through D), potentially coding for the HSP 70 homologue (ORF A); a 51-kDa protein of unknown function with similarity to GLRaV-3 p55 (ORF B); the viral capsid protein (ORF C); and a diverged viral duplicate capsid protein (ORF D). The ORF C was identified as GLRaV-5 viral capsid protein based on sequence analyses and the reactivity of the recombinant protein to GLRaV-5 specific antibodies by western blot analyses. The antiserum produced with the in vitro-expressed GLRaV-5 ORF C protein product specifically reacted with a 36-kDa polypeptide from GLRaV-5 infected vines but did not react with protein extracts from vines infected with other GLRaVs or uninfected vines. Furthermore, specific primers were designed for the sensitive detection of GLRaV-1 and GLRaV-5 by polymerase chain reaction.

Abstract:
The complete nucleotide sequence of the genomic RNA of an aphid-infecting virus, Aphid lethal paralysis virus (ALPV), has been determined. The genome is 9812 nt in length and contains two long open reading frames (ORFs), which are separated by an intergenic region of 163 nt. The first ORF (5′ ORF) is preceded by an untranslated leader sequence of 506 nt, while an untranslated region of 571 nt follows the second ORF (3′ ORF). The deduced amino acid sequences of the 5′ ORF and 3′ ORF products respectively showed similarity to the non-structural and structural proteins of members of the newly recognized genus Cripavirus (family Dicistroviridae). On the basis of the observed sequence similarities and identical genome organization, it is proposed that ALPV belongs to this genus. Phylogenetic analysis showed that ALPV is most closely related to Rhopalosiphum padi virus, and groups in a cluster with Drosophila C virus and Cricket paralysis virus, while the other members of this genus are more distantly related. Infectivity experiments showed that ALPV can not only infect aphid species but is also able to infect the whitefly Trialeurodes vaporariorum, extending its host range to another family of the order Hemiptera.

Abstract:
This article discusses the role and involvement of the Financial Services Authority (OJK) in overseeing the Baitul Maal wa Tamwil (BMT) institution, especially in relation to the protection of illegal BMT customers. After the legislation and enactment of Law Number 1 of 2013 about Microfinance Institutions (UULKM), the establishment, guidance, regulation, and supervision of BMTs have changed. BMT organizations that originally could have been in the form of cooperative, foundation, limited liability companie, and even non-governmental organization, now must be in the form of cooperative or limited liability companie. However, in terms of guidance, OJK still needs to coordinate with ministries that carry out cooperative and domestic affairs, and therefore LKM guidance are actually under three authorities: OJK, the Ministry of Cooperatives and SMEs, and the Ministry of Home Affairs. Whereas in terms of regulation and supervision, authority is absolutely in OJK. The validity of UULKM also requires BMT to obtain a business license from OJK, and if not, the status means illegal. Taking the BMT (illegal) Global Insani case as an example, this article shows the role of OJK in BMT supervision has been expanded, from the institutional to the consumer protection aspects. Consumer protection efforts that can be done by OJK are through the mechanism of complaint, investigation, reporting, and prosecution.

Abstract:
A model of the process of metrological control of measuring instruments is presented. The model includes 6 or9 stages, depending on the results of metrological control. Based on the list of measuring instruments, a contract is concludedfor metrological control. After the conclusion of the contract, the collection of documents necessary for verification iscollected. Further, the measuring instruments are transferred to an accredited organization. An accredited organization carriesout verification. If, according to the results of verification, the tool is deemed suitable for use, then the stage of obtainingconfirmation of the verification is carried out. The output at this stage will be a verification certificate. If, according to theresults of verification, the measuring instrument is not suitable for operation, then the stage of canceling the verificationcertificate is carried out. The next steps in this case are to receive notice of unsuitability for use and write-off of the measuringinstrument. At this process is considered complete. The target indicators (indicators) of the process are: the measurementerror of specific measuring instruments within the established limits. Monitoring (control) of the process is carried out bya metrology engineer of a medical organization. Control methods are: control of the calibration intervals of measuringinstruments. The model is recommended as a model for use in medical organizations. The use of this model in the presenceof a target indicator (indicator) of the process of metrological control will ensure the necessary quality of metrological workin a medical organization within the framework of the internal quality control system.

Abstract:
The article discusses the issues of creating an organizational model of selection based on cytogenetic and mor-phological markers for reforestation in the conditions of the central forest-steppe. The results of the development of an organizational model are presented, presented by a complex of context diagrams, created in the software product Busi-ness Studio 4.0. The decomposition of selection processes based on cytogenetic and morphological markers in the form of context diagrams made it possible to visualize the identification process of pedigree genotypes of pedunculate oak and to build in space and time a sequential process of using additional selection markers in conjunction with classical breeding approaches, selection of methods and means of achieving breeding goals. Providing the visibility of the pe-dunculate oak breeding process based on cytogenetic and morphological markers allows, due to the technological de-scription of the composition of the processes, the characteristics of the consumed resources (material, labor and finan-cial), to get an idea of the process under study and quickly draw a logical conclusion from the large amount of data obtained. The model organization of the process of creating forest plantations of fast-growing species was carried out with the aim of the most efficient use of technology and an increase in labor productivity, which determines its promising use as a selection tool and makes it possible to accelerate the selection process of Quercus robur L. offspring when creating forest crops

Abstract:
The efficiency of simian virus 40 (SV40) DNA replication is dependent on the structural organization of the regulatory region. The enhancing effect of the G + C-rich 21-base-pair (bp) repeats on SV40 DNA replication is position and dose dependent and to some extent orientation dependent. The inverted orientation is about 50% as effective as the normal orientation of the 21-bp repeat region. Movement of the 21-bp repeat region 180 or 370 bp upstream of the ori sequence abolishes its enhancing effect, whereas no replication is detected if the 21-bp repeat region is placed downstream of the ori sequence. The dose-dependent enhancement of the 21-bp repeat of SV40 DNA replication as first described in single transfection by Bergsma et al. (D. J. Bergsma, D. M. Olive, S. W. Hartzell, and K. N. Subramanian, Proc. Natl. Acad. Sci. USA 79:381-385, 1982) is dramatically amplified in mixed transfection. In the presence of the 21-bp repeat region, the 72-bp repeat region can enhance SV40 DNA replication. In the presence of the 21-bp repeats and a competitive environment, the 72-bp repeat region exhibits a cis-acting inhibitory effect on SV40 DNA replication.

On June 21, 2019, the International Law Organization (ILO) adopted the groundbreaking Convention No. 190 concerning the Elimination of Violence and Harassment in the World of Work (C190) and the accompanying Recommendation on the same topic (R206). ILO conventions are legally binding instruments, once ratified by states, while recommendations provide advice and guidance. The result of two years of negotiations by ILO members—governments, workers’ representatives, and employers’ organizations—is an international treaty setting out standards that are aimed at ending violence and harassment in the world of work. In its “Women, Business and the Law 2018” study, the World Bank reports that there is a lack of legal protection against harassment at work in 70 percent of the countries in the Middle East and North Africa, half of the economies in East Asia and the Pacific, and one-third of those in Latin America and the Caribbean, affecting an estimated 500 million working-age women.

Abstract:
ABSTRACT Plasmid cp8.3 of Borrelia afzelii IP21 carries several open reading frames (ORFs) and a 184-bp inverted repeat (IR) element. It has been speculated that this plasmid may encode factors involved in virulence or infectivity. In this report, we have characterized the distribution, molecular variability, and organization of ORFs 1, 2, and 4 and the IR elements among isolates of the Borrelia burgdorferi sensu lato complex. ORFs 1 and 2 are contained within a segment of cp8.3 that is bordered by the IR elements, while ORF 4 resides just outside of the IR-bordered region. By PCR, ORF 4 was amplified from most isolates while ORFs 1 and 2 were amplified from only some B. afzelii isolates. However, Southern hybridization analyses with ORF 1, 2, and 4 probes detected related sequences even in some isolates that were PCR negative. The ORF restriction fragment length polymorphism patterns varied widely even among isolates of the same species. Two-dimensional contour-clamped homogeneous electric field–pulsed-field gel electrophoresis and Southern hybridization detected ORF 1-, 2-, and 4-related sequences on linear and circular plasmids. In addition, an ORF 4-related sequence was detected on a previously uncharacterized, circular plasmid that is greater than 70 kb in size. The IR elements originally identified on plasmid cp8.3 of B. afzelii IP21 were also analyzed by Southern hybridization. Related sequences were detected in some but not all B. burgdorferi sensu lato isolates. These sequences are carried on plasmids in addition to cp8.3 in some isolates. Single-primer PCR analyses demonstrated that in some isolates these sequences exist with IR orientation. The data presented here demonstrate that the IR elements and the ORF 1-, 2-, and 4-related sequences are multicopy and are variable in organization and in genomic location among isolates of the B. burgdorferi sensu lato complex. These analyses provide additional evidence for the highly variable organization of the plasmid component of the B. burgdorferi sensu lato genome.

Abstract:
The complete nucleotide sequence of Grapevine fleck virus (GFkV) genomic RNA was determined. The genome is 7564 nt in size, excluding the 3′-terminal poly(A) tail, is characterized by an extremely high cytosine content (ca. 50%), and contains four putative open reading frames and untranslated regions of 291 and 35 nt at the 5′ and 3′ ends, respectively. ORF 1 potentially encodes a 215·4 kDa polypeptide (p215), which has the conserved motifs of replication-associated proteins of positive-strand RNA viruses. ORF 2 encodes a 24·3 kDa polypeptide (p24) identified as the coat protein. ORFs 3 and 4 are located at the extreme 3′ end of the viral genome and encode proline-rich proteins of 31·4 kDa (p31) and 15·9 kDa (p16), respectively, of unknown function. Phylogenetic analysis of the viral replicase and coat protein genes showed that GFkV is related to members of the Tymovirus and Marafivirus genera. Two subgenomic RNAs were present in the GFkV preparations as ascertained by molecular hybridization. The genome organization of GFkV resembles to some extent that of tymoviruses and marafiviruses. However, differences in the biological and epidemiological behaviour, cytopathology and molecular properties (i.e. size of genomic RNA and coat protein, and number of ORFs) support the notion that GFkV is a separate virus belonging in a new genus.

Abstract:
ABSTRACT A detailed map of the transcriptional organization of the CELO virus genome was produced. Recent computer analysis of CELO virus has indicated the presence of 38 putative open reading frames (ORFs). This study, based on analysis of the transcriptional products of CELO in vitro, confirmed the presence of RNAs for 26 of these 38 ORFs. All of the results were obtained by cDNA isolation or specific reverse transcriptase PCR. Observation of ORF transcription kinetics postinfection revealed the existence of early and late expression, with the earliest starting at 2 h postinfection. The 5′ untranslated regions of some RNAs were also studied, and this revealed the existence of a bipartite leader sequence, comparable in structure to the tripartite leader of mastadenovirus. The leader most probably involved in transcriptional activity was observed in most of the structural protein genes of the CELO virus genome. This suggests some homology in transcriptional organization between the avian adenovirus CELO and known mastadenoviruses such as human adenovirus.

ABSTRACT Of 30 baculovirus genomes that have been sequenced to date, the only nonlepidopteran baculoviruses include the dipteran Culex nigripalpus nucleopolyhedrovirus and two hymenopteran nucleopolyhedroviruses that infect the sawflies Neodiprion lecontei (NeleNPV) and Neodiprion sertifer (NeseNPV). This study provides a complete sequence and genome analysis of the nucleopolyhedrovirus that infects the balsam fir sawfly Neodiprion abietis (Hymenoptera, Symphyta, Diprionidae). The N. abietis nucleopolyhedrovirus (NeabNPV) is 84,264 bp in size, with a G+C content of 33.5%, and contains 93 predicted open reading frames (ORFs). Eleven predicted ORFs are unique to this baculovirus, 10 ORFs have a putative sequence homologue in the NeleNPV genome but not the NeseNPV genome, and 1 ORF (neab53) has a putative sequence homologue in the NeseNPV genome but not the NeleNPV genome. Specific repeat sequences are coincident with major genome rearrangements that distinguish NeabNPV and NeleNPV. Genes associated with these repeat regions encode a common amino acid motif, suggesting that they are a family of repeated contiguous gene clusters. Lepidopteran baculoviruses, similarly, have a family of repeated genes called the bro gene family. However, there is no significant sequence similarity between the NeabNPV and bro genes. Homologues of early-expressed genes such as ie-1 and lef-3 were absent in NeabNPV, as they are in the previously sequenced hymenopteran baculoviruses. Analyses of ORF upstream sequences identified potential temporally distinct genes on the basis of putative promoter elements.

Abstract:
The nucleotide sequence of a novel icosahedral DNA virus infecting Dendrolimus punctatus has been determined. The genome is 5039 nt long and includes inverted terminal repeats of 200 nt containing 131 nt long J-shaped terminal hairpins. The ‘plus' strand of the genome contains three large open reading frames (ORFs), the left and the mid-ORFs (within the left ORF) in the left-half encoding the non-structural proteins and the right ORF in the right-half encoding viral capsid proteins. NS1 protein contains conserved replication initiation and DNA-dependent ATPase/helicase domains. VP1 protein contains a conserved PGY and phospholipase A2 motifs and shows high identities with VPs of Casphalia extranea densovirus and Bombyx mori densovirus-1 belonging to the genus Iteravirus. Phylogenetic analysis also revealed that this virus is most closely related to Casphalia extranea densovirus and Bombyx mori densovirus-1. Consequently, this virus was considered as a new third member of the genus Iteravirus of the subfamily Densovirinae, and designated Dendrolimus punctatus densovirus.

Abstract:
Drosophila virilis genomic DNA corresponding to the D. melanogaster embryonic lethal abnormal visual system (elav) locus was cloned. DNA sequence analysis of a 3.8-kb genomic piece allowed identification of (i) an open reading frame (ORF) with striking homology to the previously identified D. melanogaster ORF and (ii) conserved sequence elements of possible regulatory relevance within and flanking the second intron. Conceptual translation of the D. virilis ORF predicts a 519-amino-acid-long ribonucleoprotein consensus sequence-type protein. Similar to D. melanogaster ELAV protein, it contains three tandem RNA-binding domains and an alanine/glutamine-rich amino-terminal region. The sequence throughout the RNA-binding domains, comprising the carboxy-terminal 346 amino acids, shows an extraordinary 100% identity at the amino acid level, indicating a strong structural constraint for this functional domain. The amino-terminal region is 36 amino acids longer in D. virilis, and the conservation is 66%. In in vivo functional tests, the D. virilis ORF was indistinguishable from the D. melanogaster ORF. Furthermore, a D. melanogaster ORF encoding an ELAV protein with a 40-amino-acid deletion within the alanine/glutamine-rich region was also able to supply elav function in vivo. Thus, the divergence of the amino-terminal region of the ELAV protein reflects lowered functional constraint rather than species-specific functional specification.

Abstract:
The study shows the importance of innovation and investment in economics, management and business. Some key theoretical aspects of the essence of the methodology of formation of the mechanism of innovation and investment activity and development of iron ore enterprises in modern economic conditions are determined. It is established that the lack of funding for iron ore enterprises requires effective management and rational organization of both individual stages of capital investment in their innovation activities and the investment process as a whole. At the same time, the most significant and important for the organization of investment in innovative business processes of iron ore enterprises is the application of a systematic approach. In the context of improving the mechanism of innovation and investment development of the iron ore enterprise, it is proposed to carry out a multi-criteria assessment of the level of stability of sustainable innovation and investment development, taking into account the risk. The implementation of the proposed recommendations for improving the management mechanisms of innovation and investment development of mining and processing enterprises based on the theory of potential and a systematic approach to the organization of investment in innovation processes in periods characterized by the most characteristic states of the economic environment in terms of market transformations. «Stagnation» and «recession», where weight point estimates that reflect higher priority for retrospective periods, for which less contributions were made to the total value of the evaluation function of innovation and investment development are calculated on the basis of quantitative estimates of the risk matrix. Summarizing the research of theoretical and methodological aspects of assessing the level of stability of sustainable innovation and investment development of the iron ore enterprise, a system of conditions for controlling the level of stability of sustainable innovation and investment development (RZSSIIR) has been formed. The formed system of conditions of control of RZSSIIR of the iron ore enterprise is directed on maintenance of effective strategic management at the expense of strengthening of a role of investments in innovation management for realization of process of acceptance of long-term decisions.

Abstract:
ABSTRACT Members of the Roseobacter lineage, an ecologically important marine clade within the class α-Proteobacteria, harbor genes for the protocatechuate branch of the β-ketoadipate pathway, a major catabolic route for lignin-related aromatic compounds. The genes of this pathway are typically clustered, although gene order varies among organisms. Here we characterize genes linked to pcaH and -G, which encode protocatechuate 3,4-dioxygenase, in eight closely related members of the Roseobacter lineage (pairwise 16S rRNA gene sequence identities, 92 to 99%). Sequence analysis of genomic fragments revealed five unique pca gene arrangements. Identical gene organization was found for isolates demonstrating species-level identity (i.e., >99% 16S rRNA gene similarity). In one isolate, six functionally related genes were clustered: pcaQ, pobA, pcaD, pcaC, pcaH, and pcaG. The remaining seven isolates lacked at least one of these genes in their clusters, although the relative order of the remaining genes was preserved. Three genes (pcaC, -H, and -G) were physically linked in all isolates. A highly conserved open reading frame (ORF) was found immediately downstream of pcaG in all eight isolates. Reverse transcription-PCR analysis of RNA from one isolate, Silicibacter pomeroyi DSS-3, provides evidence that this ORF is coexpressed with upstream pca genes. The absence of this ORF in similar bacterial pca gene clusters from diverse microbes suggests a niche-specific role for its protein product in Roseobacter group members. Collectively, these comparisons of bacterial pca gene organization illuminate a complex evolutionary history and underscore the widespread ecological importance of the encoded β-ketoadipate pathway.
