Abstract:
OSBP (oxysterol-binding protein) homologues, ORPs (OSBP-related proteins), constitute a 12-member family in mammals. We employed an in vitro [3H]25OH (25-hydroxycholesterol)-binding assay with purified recombinant proteins as well as live cell photo-cross-linking with [3H]photo-25OH and [3H]photoCH (photo-cholesterol), to investigate sterol binding by the mammalian ORPs. ORP1 and ORP2 [a short ORP consisting of an ORD (OSBP-related ligand-binding domain) only] were in vitro shown to bind 25OH. GST (glutathione S-transferase) fusions of the ORP1L [long variant with an N-terminal extension that carries ankyrin repeats and a PH domain (pleckstrin homology domain)] and ORP1S (short variant consisting of an ORD only) variants bound 25OH with similar affinity (ORP1L, Kd=9.7×10−8 M; ORP1S, Kd=8.4 ×10−8 M), while the affinity of GST–ORP2 for 25OH was lower (Kd=3.9×10−6 M). Molecular modelling suggested that ORP2 has a sterol-binding pocket similar to that of Saccharomyces cerevisiae Osh4p. This was confirmed by site-directed mutagenesis of residues in proximity of the bound sterol in the structural model. Substitution of Ile249 by tryptophan or Lys150 by alanine markedly inhibited 25OH binding by ORP2. In agreement with the in vitro data, ORP1L, ORP1S, and ORP2 were cross-linked with photo-25OH in live COS7 cells. Furthermore, in experiments with either truncated cDNAs encoding the OSBP-related ligand-binding domains of the ORPs or the full-length proteins, photo-25OH was bound to OSBP, ORP3, ORP4, ORP5, ORP6, ORP7, ORP8, ORP10 and ORP11. In addition, the ORP1L variant and ORP3, ORP5, and ORP8 were cross-linked with photoCH. The present study identifies ORP1 and ORP2 as OSBPs and suggests that most of the mammalian ORPs are able to bind sterols.

Abstract:
This paper presents the design and application of an efficient hybrid algorithm for solving the Optimal Reactive Power Flow (ORPF) problem. The ORPF is formulated as a nonlinear constrained optimization problem where the active power losses must be minimized. The proposed approach is based on the hybridization of Particle Swarm Optimization (PSO) and Tabu-Search (TS) technique. The proposed PSO-TS approach is used to find the settings of the control variables (i.e. generation bus voltages, transformer taps, and shunt capacitor sizes) which minimize transmission active power losses. The bus locations of the shunt capacitors are identified according to sensitive buses. To show the effectiveness of the proposed method, it is applied to the IEEE 30 bus benchmark test system and is compared with PSO and TS without hybridization, along with some other published approaches. The obtained results reveal the effectiveness of the proposed method in dealing with the highly nonlinear constrained nature of the ORPF problem.

Abstract:
In this paper, stochastic fractal search method (SFS) is employed for solving the optimal reactive power flow (ORPF) problem with a target of optimizing total active power losses (TPL), voltage deviation (VD), and voltage stability index (VSI). SFS is an effective metaheuristic algorithm consisting of diffusion process and two update processes. ORPF is a complex problem giving challenges to applied algorithms by taking into account many complex constraints such as operating voltage from generators and loads, active and reactive power generation of generators, limit of capacitors, apparent power limit from branches, and tap setting of transformers. For verifying the performance, solutions of IEEE 30 and 118-bus system with TPL, VD, and VSI objectives are found by the SFS method with different control parameter settings. Result comparisons indicate that SFS is more favorable than other methods about finding effective solutions and having faster speed. As a result, it is suggested that SFS should be used for ORPF problem, and modifications performed on SFS are encouraged for better results.

Abstract:
Aims Open reduction and plate fixation (ORPF) for displaced proximal humerus fractures can achieve reliably good long-term outcomes. However, a minority of patients have persistent pain and stiffness after surgery and may benefit from open arthrolysis, subacromial decompression, and removal of metalwork (ADROM). The long-term results of ADROM remain unknown; we aimed to assess outcomes of patients undergoing this procedure for stiffness following ORPF, and assess predictors of poor outcome. Methods Between 1998 and 2018, 424 consecutive patients were treated with primary ORPF for proximal humerus fracture. ADROM was offered to symptomatic patients with a healed fracture at six months postoperatively. Patients were followed up retrospectively with demographic data, fracture characteristics, and complications recorded. Active range of motion (aROM), Oxford Shoulder Score (OSS), and EuroQol five-dimension three-level questionnaire (EQ-5D-3L) were recorded preoperatively and postoperatively. Results A total of 138 patients underwent ADROM; 111 patients were available for long-term follow-up at a mean of 10.9 years (range 1 to 20). Mean age was 50.8 years (18 to 75);79 (57.2%) were female. Mean time from primary ORPF to ADROM was 11.9 months (6 to 19). Five patients developed superficial wound infection; ten developed symptomatic osteonecrosis/post-traumatic arthrosis (ON/PTA); four underwent revision arthrolysis. Median OSS improved from 17 (interquartile range (IQR) 12.0 to 22.0) preoperatively to 40.0 (IQR 31.5 to 48.0) postoperatively, and 39.0 (IQR 31.5 to 46.5) at long-term follow-up (p < 0.001). Median EQ-5D-3L improved from 0.079 (IQR -0.057 to 0.215) to 0.691 (IQR 0.441 to 0.941) postoperatively, and 0.701 (IQR 0.570 to 0.832) at long-term follow-up (p < 0.001). We found that aROM improved in all planes (p < 0.001). Among the variables assessed on multivariable analysis, a manual occupation, worsening Charlson Comorbidity Index and increasing socioeconomic deprivation were most consistently predictive of worse patient-reported outcome scores. Patients who subsequently developed ON/PTA reported significantly worse one-year and late OSS. Conclusion ADROM in patients with persistent symptomatic stiffness following ORPF can achieve excellent short- and long-term outcomes. More deprived patients, those in a manual occupation, and those with worsening comorbidities have worse outcomes following ADROM. Cite this article: Bone Joint J 2022;104-B(1):157–167.

Abstract:
This research deals with the problem regarding the optimal siting and sizing of distribution static compensators (D-STATCOMs) via the application of a master–slave optimization technique. The master stage determines the nodes where the D-STATCOMs must be located and their nominal rates by applying the generalized normal distribution optimizer (GNDO) with a discrete–continuous codification. In the slave stage, the successive approximations power flow method is implemented in order to establish the technical feasibility of the solution provided by the master stage, i.e., voltage regulation and device capabilities, among other features. The main goal of the proposed master–slave optimizer is to minimize the expected annual operating costs of the distribution grid, which includes the energy loss and investment costs of the D-STATCOMs. With the purpose of improving the effectiveness of reactive power compensation during the daily operation of the distribution grid, an optimal reactive power flow (ORPF) approach is used that considers the nodes where D-STATCOMs are located as inputs in order to obtain their daily expected dynamical behavior with regard to reactive power injection to obtain additional net profits. The GNDO approach and the power flow method are implemented in the MATLAB programming environment, and the ORPF approach is implemented in the GAMS software using a test feeder comprising 33 nodes with both radial and meshed configurations. A complete comparative analysis with the Salp Swarm Algorithm is presented in order to demonstrate the effectiveness of the proposed two-stage optimization approach in the fixed operation scenario regarding the final objective function values. In addition, different tests considering the possibility of hourly power injection using D-STATCOMs through the ORPF solution demonstrate that additional gains can be obtained in the expected annual operative costs of the grid.

Large-aperture towed linear hydrophone array has been widely used for beamforming-based signal enhancement in passive sonar systems; however, its performance can drastically decrease due to the array distortion caused by rapid tactical maneuvers of the towed platform, oceanic currents, hydrodynamic effects, etc. In this paper, an enhanced data-driven shape array estimation scheme is provided in the passive underwater acoustic data, and a novel nonlinear outlier-robust particle filter (ORPF) method is proposed to acquire enhanced estimates of time delays in the presence of distorted hydrophone array. A conventional beamforming technique based on a hypothetical array is first used, and the detection of the narrow-band components is sequentially carried out so that the corresponding amplitudes and phases at these narrow-band components can be acquired. We convert the towed array estimation problem into a nonlinear discrete-time filtering problem with the joint estimates of amplitudes and time-delay differences, and then propose the ORPF method to acquire enhanced estimates of the time delays by exploiting the underlying properties of slowly changing time-delay differences across sensors. The proposed scheme fully exploits directional radiated noise targets as sources of opportunity for online array shape estimation, and thus it requires neither the number nor direction of sources to be known in advance. Both simulations and real experimental data show the effectiveness of the proposed method.

Abstract:
ORPs are lipid-transport proteins belonging to the oxysterol-binding protein family. They facilitate the transfer of lipids between different intracellular membranes, such as the ER and plasma membrane. We have solved the crystal structure of the ORP8 lipid transport domain (ORD8). The ORD8 exhibited a β-barrel fold composed of anti-parallel β-strands, with three α-helices replacing β-strands on one side. This mixed alpha–beta structure was consistent with previously solved structures of ORP2 and ORP3. A large cavity (≈1860 Å3) within the barrel was identified as the lipid-binding site. Although we were not able to obtain a lipid-bound structure, we used computer simulations based on our crystal structure to dock PS and PI4P molecules into the putative lipid-binding site of the ORD8. Comparative experiments between the short ORD8ΔLid (used for crystallography) and the full-length ORD8 (lid containing) revealed the lid’s importance for stable lipid binding. Fluorescence assays revealed different transport efficiencies for PS and PI4P, with the lid slowing down transport and stabilizing cargo. Coarse-grained simulations highlighted surface-exposed regions and hydrophobic interactions facilitating lipid bilayer insertion. These findings enhance our comprehension of ORD8, its structure, and lipid transport mechanisms, as well as provide a structural basis for the design of potential inhibitors.

Mitochondria is no longer viewed as merely a powerhouse of the cell. It is now apparentthat mitochondria play a central role in signaling, maintaining cellular homeostasis and cell fate.Mitochondrial dysfunction has been linked to many human diseases caused by cellular metabolicderegulation, such as obesity, diabetes, neurodegenerative disease, cardiovascular disease andcancer. Eukaryotic organisms have evolved an efficient way in sensing, communicating andresponding to cellular stress and regulating mitochondrial activity correspondingly through acomplex network of intercommunicating protein kinases and their downstream effectors. Thisdissertation focuses on the interplay of two of the master metabolic regulators in the cell: AMPKand PASK, and characterization of the functions of their downstream substrates: OSBP andMED13. AMPK is an energy sensing kinase that maintains energy homeostasis in the cell,whereas PASK is a nutrient sensing kinase that regulates glucose partitioning and respiration inthe cell. Both kinases play important roles in mitochondrial function and regulation, anddeficiency in either kinase has been found to associate with various human pathologies. Furthercharacterization of the cross-talk and molecular mechanisms of both kinases in controllingmitochondrial health and function may aid in the identification of new targets for treatingmetabolic diseases.

Since 2012, it has been notified WSSV occurrence in the state of Rio Grande do Norte - RN, one of most important shrimp production region. Despite its importance, there is a lack of information about the origins and biodiversity of circulating virus. The aim of investigation was to detect WSSV in Litopenaeus vannamei from farms in the RN as well as perform genotyping of the isolates. A total of five farms were surveyed, from which 10 samples were collected each one. We selected two ponds of each of the farms, chosen because clinical suspicion of WSSV. Genomic DNA was extracted to detect the virus by polymerase chain reaction (PCR). Subsequently, was performed by PCR genotyping of the positive DNA samples analysing variable number of tandem repeats - VNTR in three molecular markers (ORFs 75, 94 and 125). The results revealed the presence of virus in only one of the 5 analyzed farms. The loci ORF 75 and ORF 94 were identified in isolates from positive farm and no amplification failure for ORF 125. The number of repeating units (RUs) was calculated, and found to 9 repetitions ORF 75 and 10 RUs to ORF 94, a pattern that had not been described, stressing the need for more strain typing for screening and surveillance of the disease<br>A síndrome da mancha branca é uma doença notificável cujo agente etiológico é o Vírus da Síndrome da Mancha Branca (White Spot Syndrome Virus - WSSV) responsável por graves perdas econômicas na carcinicultura mundial. Desde 2012, têm sido verificados registros oficiais da presença do vírus no estado do Rio Grande do Norte - RN , região de extensa produção camaroneira. Apesar da importância, pouca informação tem sido produzida quanto às origens e biodiversidade dos virus circulantes. Diante disso, objetivou-se investigar a presença do vírus em criações de Litopenaues vannamei oriundos de fazendas do RN, bem como realizar a genotipagem dos isolados. Um total de cinco fazendas foram submetidas ao estudo, das quais foram coletadas 10 amostras de cada. Foram selecionados dois viveiros de cada uma das fazendas, escolhidos mediante suspeita clínica de WSSV. O DNA genômico foi extraído para investigação da presença ou ausência do vírus através da reação em cadeia da polimerase (PCR). Posteriormente, foi realizada por PCR a genotipagem dos isolados utilizando as repetições em tandem de número variável – VNTR (ORFs 75, 94 e 125). Os resultados revelaram a presença do vírus em apenas uma das 5 fazendas analisadas. As regiões (ORF 75 e ORF 94) foram identificadas nos isolados da fazenda positiva e não houve amplificação para o ORF 125. O número de unidades repetidas (RUs) foi calculado, sendo verificado 9 repetições para o ORF 75 e 10 para o ORF 94, um padrão que ainda não havia sido descrito, alertando para a necessidade de mais isolados caracterizados geneticamente para o rastreamento e vigilância epidemiológica da doença<br>2017-03-23

Art-Glam Post-Orf es un título similar al de una revista, la idea es ironizar de un modo postmoderno con las revistas de arte y asimilar una revista de arte con una revista más pop. Las obras se caracterizan por ser técnicas mixtas que nacen a partir de la orfebrería y se van alejando de ella paulatinamente.

Master of Music<br>Department of Music, Theatre, and Dance<br>Frederick Burrack<br>Prominent elementary music methods like Orff-Schulwerk, Kodaly, Suzuki, Music Learning Theory, and Dalcroze Eurhythmics share a belief in a sound-before-symbol approach: delaying notation instruction in order to first develop audiation, musical vocabularies, and concrete musical experiences. Unfortunately, piano pedagogy has not taken the same journey, with method books continuing to center on reading from the earliest. While piano pedagogy has made great strides as a professional music community, now it must adopt the same sound-before symbol approach of its general music colleagues, making room for experiential and creative activities as a core component of instruction. Through the integration of movement, exploration, and improvisation activities like the included examples, teachers can move beyond strict adherence to the method book, and change their focus from notational literacy to authentic musicianship.

TITLE: Comparison of the Orff-Schulwerk Music Education Approach in the United States and Taiwan MAJOR PROFESSOR: Dr. Pamela Stover The Orff-Schulwerk approach is a common and popular approach in the United States and Taiwan. In this paper, this paper compares the music classes in the United States and Taiwan. If the music teachers can see the differences in music education between the two countries then they can think about their teaching and learn new strategies with what works in other countries. In addition, music educators can consider using the Orff-Schulwerk approach in the future. This study is a two-pronged approach to discover more about the implementation and attitudes toward the Orff-Schulwerk (music and movement education) approach of teaching in the United States and Taiwan. What are the teachers' and students' attitudes and thoughts about the Orff-Schulwerk approach? First, surveys for teachers and students using quantitative and qualitative response was given. Orff-Schulwerk teachers in Taiwan and America were given questionnaires by e-mail or in person. The questions were multiple choice, with space provided for additional comments. From these surveys, selected music teachers who were highly trained in the Orff-Schulwerk approach were viedo-taped while they were teaching grades 2-6. Target teachers in Taiwan and in the United States have studied at the Orff Institute or have additional Orff-Schulwerk teacher training. Teachers were asked about their preference for various Orff-Schulwerk, their attitudes in class and what they learned. Second, selected classes were video taped to observe the students' activities in Orff-Schulwerk music classes in both Taiwan and the United States. After the classes were video-taped, the students were given a survey in person. This was a paper and pencil Likert-scale short questionaire about their attitudes of different the Orff-Schulwerk elements. The results show that the Orff-Schulwerk teachers in America and Taiwan were similar in their teaching techniques. However, the students were different. Taiwanese students did not perform much or answer questions. In the fifth grade, Taiwanese students did more music composition, even though they liked composition less than playing musical games. American students were more active in class and liked to perform in class. The American classrooms that observed focused on playing the Orff instruments. Although both the Orff-Schulwerk was implimented in both countries, the differences observed were mostly cultureal differences and not a result of teaching techniques.

Abstract:
This thesis studies concurrency control and composition of transactions in computing environments with long living transactions where local data autonomy of transactions is indispensable. This kind of computing architecture is referred to as a Disconnected System where reads are segregated -disconnected- from writes enabling local data autonomy. Disconnecting reads from writes is inspired by Bertrand Meyer's "Command Query Separation" pattern. This thesis provides a simple yet precise definition for a Disconnected System with a focus on transaction management. Concerning concurrency control, transaction management frameworks implement a 'one concurrency control mechanism fits all needs strategy'. This strategy, however, does not consider specific characteristics of data access. The thesis shows the limitations of this strategy if transaction load increases, transactions are long lived, local data autonomy is required, and serializability is aimed at isolation level. For example, in optimistic mechanisms the number of aborts suddenly increases if load increases. In pessimistic mechanisms locking causes long blocking times and is prone to deadlocks. These findings are not new and a common solution used by database vendors is to reduce the isolation. This thesis proposes the usage of a novel approach. It suggests choosing the concurrency control mechanism according to the semantics of data access of a certain data item. As a result a transaction may execute under several concurrency control mechanisms. The idea is to introduce lanes similar to a motorway where each lane is dedicated to a certain class of vehicle with the same characteristics. Whereas disconnecting reads and writes sets the traffic's direction, the semantics of data access defines the lanes. This thesis introduces four concurrency control classes capturing the semantics of data access and each of them has an associated tailored concurrency control mechanism. Class O (the optimistic class) implements a first-committer-wins strategy, class R (the reconciliation class) implements a first-n-committers-win strategy, class P (the pessimistic class) implements a first-reader-wins strategy, and class E (the escrow class) implements a first-n-readers-win strategy. In contrast to solutions that adapt the concurrency control mechanism during runtime, the idea is to classify data during the design phase of the application and adapt the classification only in certain cases at runtime. The result of the thesis is a transaction management framework called O|R|P|E. A performance study based on the TPC-C benchmark shows that O|R|P|E has a better performance and a considerably higher commit rate than other solutions. Moreover, the thesis shows that in O|R|P|E aborts are due to application specific limitations, i.e., constraint violations and not due to serialization conflicts. Abstract This paper discusses an innovative technique employed on the 2017 AETC Corrosion Control Study conducted on 19 NASA wind tunnel facilities located at three NASA Centers: Langley, Ames and Glenn Research Centers. The technique adds structural and prioritization context to corrosion and coating assessments which yields an Overall Risk Priority (ORP) score for each inspection item. The ORP score is comprised of (1) the coatings grade from field inspections and sample data, (2) the functional purpose of the facility and (3) the risk to facility availability (mission) if the asset were to fail due to corrosion. The ORP score is used to assist facility engineers in developing a Structural Integrity and Service Life Opinion (SISLO) that provides a qualitative characterization of each wind tunnel. With some wind tunnels exceeding 70 years of age, the 2017 Corrosion Control Study is a vital tool used by NASA to properly integrate and prioritize maintenance planning for each of these important national assets.

Abstract Oxidizing biocides, as their name implies, make use of their oxidizing ability to control microorganisms and prevent the deposition of corrosion causing microbiological deposits. This oxidizing ability can also promote corrosion in systems if the oxidizing biocide is accidently overfed or if an incompatible corrosion inhibitor system is used. Proper control of feed rates of oxidizing biocides is necessary not only from a corrosion prevention standpoint, but an economical one as well. The use of oxidation reduction potential to control oxidizing biocide feed systems is explored in this work and shown to be an effective means of control.

Abstract Experimental tests were carried out on two commercially manufactured devices reputed to prevent the formation of scale, remove preexisting scale and reduce corrosion of carbon steel. One device operates by applying a high voltage, low alternating current electrostatic field to the flowing solution. The second device contains two opposing permanent magnets and a special geometric flow channel through which the solution flows. To determine the effect of the devices, with regard to the claims made by the manufacturers, an enclosed recirculating test loop system was fabricated. A brine solution formulated such that it would not form significant scale at room temperature, but would at elevated temperature was circulated in the system. The corrosion rate, amount of calcium carbonate scale deposition, pH, oxidizing reducing potential (ORP) of the water, water chemistry, and crystalline form of the carbonate scale were the monitored parameters. The results of the investigation showed that neither tested device had any measurable beneficial effect on either corrosion rate or scale deposition.

At the aim of alleviating the computational burden of complicated engineering optimization problems, metamodels have been widely employed to approximate the expensive blackbox functions. Among the popular metamodeling methods RBF metamodel well balances the global approximation accuracy, computational cost and implementation difficulty. However, the approximation accuracy of RBF metamodel is heavily influenced by the width factors of kernel functions, which are hard to determine and actually depend on the numerical behavior of expensive functions and distribution of samples. The main contribution of this paper is to propose an optimized RBF (ORBF) metamodel for the purpose of improving the global approximation capability with an affordable extra computational cost. Several numerical problems are used to compare the global approximation performance of the proposed ORBF metamodeling methods to determine the promising optimization approach. And the proposed ORBF is also adopted in adaptive metamodel-based optimization method. Two numerical benchmark examples and an I-beam optimization design are used to validate the adaptive metamodel-based optimization method using ORBF metamodel. It is demonstrated that ORBF metamodeling is beneficial to improving the optimization efficiency and global convergence capability for expensive engineering optimization problems.

Abstract Open Reduction and Internal Fixation (ORIF) of scapula fractures have increased in numbers recently. This is due to better functional outcomes achieved post-ORIF than non-operative management techniques. In South Africa, there is only one available supplier for anatomical contoured scapula plates used in the ORIF. This study examines the fit of these plates on the bony topology of fifty healthy scapula. It was observed that the short medial body plate performed the best in adhering to the bone topology followed by the short acromion plate. The glenoid plate and the long acromion and body plates were not adequately designed to fit their intended regions on the scapula. In conclusion, this study highlights the drawbacks in design of current commercial plates available for ORIF of scapula. Further studies are needed in order to evaluate the quantitative-fit performance of these plates on fracture scapula surfaces.

Rugose wood is a complex disease of grapevines, characterized by modification of the woody cylinder of affected vines. The control of rugose wood is based on the production of healthy propagation material. Detection of rugose wood in grapevines is difficult and expensive: budwood from tested plants is grafted onto sensitive Vitis indicators and the appearance of symptoms is monitored for 3 years. The etiology of rugose wood is complex and has not yet been elucidated. Several elongated clostero-like viruses are consistently found in affected vines; one of them, grapevine virus A (GVA), is closely associated with Kober stem grooving, a component of the rugose wood complex. GVA has a single-stranded RNA genome of 7349 nucleotides, excluding a polyA tail at the 3' terminus. The GVA genome includes five open reading frames (ORFs 1-5). ORF 4, which encodes for the coat protein of GVA, is the only ORF for which the function was determined experimentally. The original objectives of this research were: 1- To produce antisera to the structural and non-structural proteins of GVA and GVB and to use these antibodies to establish an effective detection method. 2- Develop full length infectious cDNA clones of GVA and GVB. 3- Study the roll of GVA and GVB in the etiology of the grapevine rugose wood disease. 4- Determine the function of Trichovirus (now called Vitivirus) encoded genes in the virus life cycle. Each of the ORFs 2, 3, 4 and 5 genes of GVA were cloned and expressed in E. coli and used to produce antisera. Both the CP (ORF 4) and the putative MP (ORF 3) were detected with their corresponding antisera in-GVA infected N. benthamiana and grapevine. The MP was first detected at an early stage of the infection, 6-12 h after inoculation, and the CP 2-3 days after inoculation. The MP could be detected in GVA-infected grapevines that tested negative for CP, both with CP antiserum and with a commercially available ELISA kit. Antisera to ORF 2 and 5 encoded proteins could react with the recombinant proteins but failed to detect both proteins in GVA infected plants. A full-length cDNA clone of grapevine virus A (GVA) was constructed downstream from the bacteriophage T7 RNA polymerase promoter. Capped in vitro transcribed RNA was infectious in N. benthamiana and N. clevelandii plants. Symptoms induced by the RNA transcripts or by the parental virus were indistinguishable. The infectivity of the in vitro-transcribed RNA was confirmed by serological detection of the virus coat and movement proteins and by observation of virions by electron microscopy. The full-length clone was modified to include a gus reporter gene and gus activity was detected in inoculated and systemic leaves of infected plants. Studies of GVA mutants suggests that the coat protein (ORF 4) is essential for cell to cell movement, the putative movement protein (ORF 3) indeed functions as a movement protein and that ORF 2 is not required for virus replication, cell to cell or systemic movement. Attempts to infect grapevines by in-vitro transcripts, by inoculation of cDNA construct in which the virus is derived by the CaMV 35S promoter or by approach grafting with infected N. benthamiana, have so far failed. Studies of the subcellular distribution of GFP fusion with each of ORF 2, 3 and 4 encoded protein showed that the CP fusion protein accumulated as a soluble cytoplasmatic protein. The ORF 2 fusion protein accumulated in cytoplasmatic aggregates. The MP-GFP fusion protein accumulated in a large number of small aggregates in the cytoplasm and could not move from cell to cell. However, in conditions that allowed movement of the fusion protein from cell to cell (expression by a PVX vector or in young immature leaves) the protein did not form cytoplasmatic aggregates but accumulated in the plasmodesmata.

Review question / Objective: We seek to conduct a meta-analysis of relevant studies to evaluate and compare functional outcomes and complication rates between locked fibula intramedullary nail fixation and plate fixation for treatment of ankle fractures. Condition being studied: Ankle fractures, with an incidence rate of 4.22/10, 000 person-years in the United States, are one of the most common lower extremity fractures. Currently, the standard surgical treatment approaches for unstable ankle fractures involves open reduction and internal fixation (ORIF) with plates and screws. However, ORIF has resulted in little efficacy during treatment of fractures since the 1960s, while plate and screw fixation has also been associated with several complications. Previous studies have shown that closed reduction and internal fixation (CRIF) with fibula intramedullary nail (IMN) has achieved satisfactory efficacy in treatment of ankle fractures, and is associated with low complication rates. Additionally, a systematic review showed that a locked intramedullary nail (LIMN) device provides better stability and rotation control, thereby reducing the risk of nail migration and loss of fixation, compared to unlocked nails. Therefore, a meta-analysis is imperative to provide evidence on whether LIMN can replace PF for treatment of ankle fractures, owing to an increase in related studies that have been published in recent years.
