Mitochondria is no longer viewed as merely a powerhouse of the cell. It is now apparentthat mitochondria play a central role in signaling, maintaining cellular homeostasis and cell fate.Mitochondrial dysfunction has been linked to many human diseases caused by cellular metabolicderegulation, such as obesity, diabetes, neurodegenerative disease, cardiovascular disease andcancer. Eukaryotic organisms have evolved an efficient way in sensing, communicating andresponding to cellular stress and regulating mitochondrial activity correspondingly through acomplex network of intercommunicating protein kinases and their downstream effectors. Thisdissertation focuses on the interplay of two of the master metabolic regulators in the cell: AMPKand PASK, and characterization of the functions of their downstream substrates: OSBP andMED13. AMPK is an energy sensing kinase that maintains energy homeostasis in the cell,whereas PASK is a nutrient sensing kinase that regulates glucose partitioning and respiration inthe cell. Both kinases play important roles in mitochondrial function and regulation, anddeficiency in either kinase has been found to associate with various human pathologies. Furthercharacterization of the cross-talk and molecular mechanisms of both kinases in controllingmitochondrial health and function may aid in the identification of new targets for treatingmetabolic diseases.

Since 2012, it has been notified WSSV occurrence in the state of Rio Grande do Norte - RN, one of most important shrimp production region. Despite its importance, there is a lack of information about the origins and biodiversity of circulating virus. The aim of investigation was to detect WSSV in Litopenaeus vannamei from farms in the RN as well as perform genotyping of the isolates. A total of five farms were surveyed, from which 10 samples were collected each one. We selected two ponds of each of the farms, chosen because clinical suspicion of WSSV. Genomic DNA was extracted to detect the virus by polymerase chain reaction (PCR). Subsequently, was performed by PCR genotyping of the positive DNA samples analysing variable number of tandem repeats - VNTR in three molecular markers (ORFs 75, 94 and 125). The results revealed the presence of virus in only one of the 5 analyzed farms. The loci ORF 75 and ORF 94 were identified in isolates from positive farm and no amplification failure for ORF 125. The number of repeating units (RUs) was calculated, and found to 9 repetitions ORF 75 and 10 RUs to ORF 94, a pattern that had not been described, stressing the need for more strain typing for screening and surveillance of the disease<br>A síndrome da mancha branca é uma doença notificável cujo agente etiológico é o Vírus da Síndrome da Mancha Branca (White Spot Syndrome Virus - WSSV) responsável por graves perdas econômicas na carcinicultura mundial. Desde 2012, têm sido verificados registros oficiais da presença do vírus no estado do Rio Grande do Norte - RN , região de extensa produção camaroneira. Apesar da importância, pouca informação tem sido produzida quanto às origens e biodiversidade dos virus circulantes. Diante disso, objetivou-se investigar a presença do vírus em criações de Litopenaues vannamei oriundos de fazendas do RN, bem como realizar a genotipagem dos isolados. Um total de cinco fazendas foram submetidas ao estudo, das quais foram coletadas 10 amostras de cada. Foram selecionados dois viveiros de cada uma das fazendas, escolhidos mediante suspeita clínica de WSSV. O DNA genômico foi extraído para investigação da presença ou ausência do vírus através da reação em cadeia da polimerase (PCR). Posteriormente, foi realizada por PCR a genotipagem dos isolados utilizando as repetições em tandem de número variável – VNTR (ORFs 75, 94 e 125). Os resultados revelaram a presença do vírus em apenas uma das 5 fazendas analisadas. As regiões (ORF 75 e ORF 94) foram identificadas nos isolados da fazenda positiva e não houve amplificação para o ORF 125. O número de unidades repetidas (RUs) foi calculado, sendo verificado 9 repetições para o ORF 75 e 10 para o ORF 94, um padrão que ainda não havia sido descrito, alertando para a necessidade de mais isolados caracterizados geneticamente para o rastreamento e vigilância epidemiológica da doença<br>2017-03-23

Master of Music<br>Department of Music, Theatre, and Dance<br>Frederick Burrack<br>Prominent elementary music methods like Orff-Schulwerk, Kodaly, Suzuki, Music Learning Theory, and Dalcroze Eurhythmics share a belief in a sound-before-symbol approach: delaying notation instruction in order to first develop audiation, musical vocabularies, and concrete musical experiences. Unfortunately, piano pedagogy has not taken the same journey, with method books continuing to center on reading from the earliest. While piano pedagogy has made great strides as a professional music community, now it must adopt the same sound-before symbol approach of its general music colleagues, making room for experiential and creative activities as a core component of instruction. Through the integration of movement, exploration, and improvisation activities like the included examples, teachers can move beyond strict adherence to the method book, and change their focus from notational literacy to authentic musicianship.

TITLE: Comparison of the Orff-Schulwerk Music Education Approach in the United States and Taiwan MAJOR PROFESSOR: Dr. Pamela Stover The Orff-Schulwerk approach is a common and popular approach in the United States and Taiwan. In this paper, this paper compares the music classes in the United States and Taiwan. If the music teachers can see the differences in music education between the two countries then they can think about their teaching and learn new strategies with what works in other countries. In addition, music educators can consider using the Orff-Schulwerk approach in the future. This study is a two-pronged approach to discover more about the implementation and attitudes toward the Orff-Schulwerk (music and movement education) approach of teaching in the United States and Taiwan. What are the teachers' and students' attitudes and thoughts about the Orff-Schulwerk approach? First, surveys for teachers and students using quantitative and qualitative response was given. Orff-Schulwerk teachers in Taiwan and America were given questionnaires by e-mail or in person. The questions were multiple choice, with space provided for additional comments. From these surveys, selected music teachers who were highly trained in the Orff-Schulwerk approach were viedo-taped while they were teaching grades 2-6. Target teachers in Taiwan and in the United States have studied at the Orff Institute or have additional Orff-Schulwerk teacher training. Teachers were asked about their preference for various Orff-Schulwerk, their attitudes in class and what they learned. Second, selected classes were video taped to observe the students' activities in Orff-Schulwerk music classes in both Taiwan and the United States. After the classes were video-taped, the students were given a survey in person. This was a paper and pencil Likert-scale short questionaire about their attitudes of different the Orff-Schulwerk elements. The results show that the Orff-Schulwerk teachers in America and Taiwan were similar in their teaching techniques. However, the students were different. Taiwanese students did not perform much or answer questions. In the fifth grade, Taiwanese students did more music composition, even though they liked composition less than playing musical games. American students were more active in class and liked to perform in class. The American classrooms that observed focused on playing the Orff instruments. Although both the Orff-Schulwerk was implimented in both countries, the differences observed were mostly cultureal differences and not a result of teaching techniques.

Abstract:
This thesis studies concurrency control and composition of transactions in computing environments with long living transactions where local data autonomy of transactions is indispensable. This kind of computing architecture is referred to as a Disconnected System where reads are segregated -disconnected- from writes enabling local data autonomy. Disconnecting reads from writes is inspired by Bertrand Meyer's "Command Query Separation" pattern. This thesis provides a simple yet precise definition for a Disconnected System with a focus on transaction management. Concerning concurrency control, transaction management frameworks implement a 'one concurrency control mechanism fits all needs strategy'. This strategy, however, does not consider specific characteristics of data access. The thesis shows the limitations of this strategy if transaction load increases, transactions are long lived, local data autonomy is required, and serializability is aimed at isolation level. For example, in optimistic mechanisms the number of aborts suddenly increases if load increases. In pessimistic mechanisms locking causes long blocking times and is prone to deadlocks. These findings are not new and a common solution used by database vendors is to reduce the isolation. This thesis proposes the usage of a novel approach. It suggests choosing the concurrency control mechanism according to the semantics of data access of a certain data item. As a result a transaction may execute under several concurrency control mechanisms. The idea is to introduce lanes similar to a motorway where each lane is dedicated to a certain class of vehicle with the same characteristics. Whereas disconnecting reads and writes sets the traffic's direction, the semantics of data access defines the lanes. This thesis introduces four concurrency control classes capturing the semantics of data access and each of them has an associated tailored concurrency control mechanism. Class O (the optimistic class) implements a first-committer-wins strategy, class R (the reconciliation class) implements a first-n-committers-win strategy, class P (the pessimistic class) implements a first-reader-wins strategy, and class E (the escrow class) implements a first-n-readers-win strategy. In contrast to solutions that adapt the concurrency control mechanism during runtime, the idea is to classify data during the design phase of the application and adapt the classification only in certain cases at runtime. The result of the thesis is a transaction management framework called O|R|P|E. A performance study based on the TPC-C benchmark shows that O|R|P|E has a better performance and a considerably higher commit rate than other solutions. Moreover, the thesis shows that in O|R|P|E aborts are due to application specific limitations, i.e., constraint violations and not due to serialization conflicts. In the literature review is made an approach to rhythm and movement in relation to its importance in learning and development of musical and motor skills, and its influence on the instrument practice. The research presented in this paper compares the strategy of learning instrumental Orff, reducing the movement practice, with the strategy of application of body movement, reducing the practice of Orff instruments.

A metodologia utilizada foi o estudo de caso de índole exploratória. Foi efectuada uma revisão da literatura que incidiu essencialmente sobre o e-learning e o IMS – Learning Design, seguindo-se a criação das estratégias didácticas e da planificação de toda a Unidade de Aprendizagem sobre Instrumentos Orff. O passo seguinte foi a criação e desenvolvimento da Unidade de Aprendizagem, que culminou com a implementação dos pacotes IMS/SCORM finais do aluno e do professor. A avaliação dos materiais desenvolvidos foi efectuada através de questionários a que responderam um grupo de alunos e de professores. Como forma de conclusão deste trabalho de investigação, foi realizada uma reflexão sobre o modelo IMS – Learning Design e as actuais ferramentas de apoio à implementação. ABSTRACT: The purpose of this dissertation is the exploration of the model IMS – Learning Design and supporting tools applicability, through the creation and development of a learning unit on Orff Instruments based on the model for 1st Basic Teaching Cycle. Method used was an exploratory case study. A complete overview of relevant literature has been done, essentially on elearning and IMS – Learning Design, followed by the building of learning and planning strategies for the whole Learning Unit on Orff Instruments. The following step was the creation and the development of the Learning Unit, that leaded to the final version IMS/SCORM of student’s and teacher’s final package. The evaluation of the materials developed has been done through an inquest targeted to teachers and students. As a conclusion of this investigation, a complete reflection and overview of the IMS Model-Learning Design and its Current supporting tools to it implementation is presented.

Uma das razões que fazem de Saccharomyces cerevisiae um organismo modelo é o grau de conservação dos mecanismos celulares que existe entre esta levedura e eucariotos superiores. Porém, mesmo após 21 anos do seqüenciamento do seu genoma, ainda existem mais de 600 ORFs com função desconhecida. Neste trabalho, selecionamos quatro delas para o estudo detalhado. MRPL34 (YDR115w) está presente na subunidade maior do ribossomo mitocondrial de levedura e apresenta similaridade com o gene L34 de E. coli e MRP-L34 de humanos. O mutante &#916;mrpl34 apresenta DNA mitocondrial (mtDNA) instável e para estudá-lo foram gerados alelos sensíveis à temperatura (ts). Com os ensaios de síntese protéica mitocondrial in vivo foi possível identificar clara diminuição da síntese de proteínas do mutante condicional. Mrpl34p foi identificada no extrato ribossomal, conforme esperado. A desestruturação da subunidade maior do ribossomo mitocondrial, utilizando os mutantes ts, nos forneceu indícios sobre intermediários existentes no seu processo de montagem. Verificamos que a porção N-terminal da proteína é responsável pelo endereçamento à mitocôndria. YPR116w também apresenta alta instabilidade do DNA mitocondrial, desta forma, mutantes termossensíveis foram utilizados nos experimentos. Uma das estratégias utilizadas visou a busca de parceiros genéticos. Verificamos que ylr091wp aumenta a estabilidade do mtDNA de ts- ypr116w, sugerindo atividade supressora. Também averiguamos que o alelo ts-ypr16w apresenta menor quantidade de tRNA mitocondrial, através de ensaios de Northen blot. Duas das ORFs escolhidas (YDL119c e YOR022c) tiveram sua caracterização inicial publicada em 2016, refletindo a importância deste tipo de pesquisa. Vimos que a proteína codificada por YDL119c está localizada na membrana interna da mitocôndria e que o mutante &#916;yor022c apresenta quantidades reduzidas de cardiolipina, quando crescido à 37ºC.<br>One of the reasons that turn Saccharomyces cerevisiae a model organism is the degree of conservation of cellular mechanisms that exist between this yeast and higher eukaryotes. However, even after 21 years of sequencing their genome, there are still more than 600 ORFs with unknown function. In this work, we selected four of them for the detailed study. MRPL34 (YDR115w) is present in the major subunit of the yeast mitochondrial ribosome and bears similarity to the L34 gene of E. coli and MRP-L34 from humans. The &#916; mrpl34 mutant shows unstable mitochondrial DNA (mtDNA) and to study it, temperature sensitive alleles (ts) were generated. With the mitochondrial protein synthesis assays in vivo, it was possible to identify a clear decrease in the protein synthesis of the conditional mutant. Mrpl34p was identified in the ribosomal extract as expected. The disassembly of the major subunit of the mitochondrial ribosome, using the ts mutants, provided us some clues about intermediates in its assembly process. We have verified that the N-terminal portion of the protein is responsible for addressing the mitochondria. YPR116w also shows high mitochondrial DNA instability, in this way, thermosensitive mutants were used in the experiments. One of the strategies used was the search for genetic partners. We verified that ylr091wp increases the stability of ts-ypr116w mtDNA, suggesting suppressor activity. We also found that the ts-ypr16w allele has a smaller amount of mitochondrial tRNA, through Northen blot assays. Two of the chosen ORFs (YDL119c and YOR022c) had their initial characterization published in 2016, reflecting the importance of this type of research. We have seen that the protein encoded by YDL119c is located on the inner membrane of the mitochondria and that the &#916;yor022c mutant presents reduced amounts of cardiolipin when grown at 37 ºC.

Abstract:
Apoptosis plays important roles in host defences against virus infection. It is therefore not surprising that viruses have developed a vast array of modulators that block this process at different stages within the apoptotic pathways. Intrestingly, Orf virus (ORFV), a member of the Parapoxvirus genus, did not reveal any of the known poxviral inhibitors of apoptosis, but was found to express a unique anti-apoptotic protein, ORFV125. The aim of this PhD project was to determine the subcellular localisation of this protein and to further characterise its anti-apoptotic activity. This included exploring its ability to inhibit early, intermediate and late events of apoptosis and identifying the mechanism by which this viral protein functions to prevent cell death. Experiments revealed that ORFV125 was localised to the mitochondria through a C-terminal mitochondrial-targeting motif, and this specific location was necessary for the protein�s anti-apoptotic function. Furthermore, the viral protein inhibited UV-induced apoptotic events at and downstream of the mitochondria such as cytochrome c release, caspase activation and DNA fragmentation. However, it was not able to prevent UV-induced activation of the c-Jun-NH₂ kinase (JNK), an event occurring upstream of the mitochondria, consistent with its localisation to this organelle. The ability to prevent apoptosis was comparable with that of the cellular anti-apoptotic protein Bcl-2, which belongs to a family of mitochondrial regulators of apoptosis. Although standard BLAST analysis failed to detect homology to anti-apoptotic members of the Bcl-2 family, a manual alignment of the primary sequence of ORFV125 with these proteins revealed characteristic residues of Bcl-2 homology (BH) domains within ORFV125. These motifs are conserved within the Bcl-2 proteins and important for their structure and function. In addition, mutating amino acids within the ORFV125 BH domains led to a loss of the anti-apoptotic function of the mutated proteins, indicating the functional importance of these residues for the viral protein. These observations suggest that ORFV125 might be classified as a viral Bcl-2-like protein. To provide evidence for this hypothesis, it was investigated if ORFV125 acts in a Bcl-2-like manner to inhibit apoptosis. The viral protein was able to entirely block the activation of the pro-apoptotic Bcl-2 family members Bak and Bax, although it did not directly bind to these proteins. Instead, ORFV125 interacted with a subset of the pro-apoptotic BH3-only proteins, which can trigger the activation of Bax and Bak. Furthermore, this study demonstrated that ORFV125 could inhibit apoptosis induced by BH3-only proteins to which the viral protein could bind. On the other hand, ORFV125 was not able to prevent the activity of pro-apoptotic proteins that it failed to interact with. This shows that ORFV125�s mechanism of action is to inhibit the activity of BH3-only proteins by binding and neutralising their function. Overall, these results provided evidence that ORFV125 is potent anti-apoptotic protein that can prevent UV-induced cell death without the participation of other ORFV proteins. Furthermore, the viral protein shared primary sequence and secondary structure similarities to Bcl-2 family members and acted in a Bcl-2-like manner to inhibit apoptosis.

Recombination events in bacteriophages frequently occur by illegitimate exchange at short tracts of sequence homology, enabling these viruses to acquire novel genes and serve as vehicles for horizontal gene transfer. The emergence of new pathogenic organisms due to the acquisition of virulence determinants from bacterial viruses has stimulated considerable interest in the mechanisms of phage recombination. Bacteriophage λ encodes its own recombination system, consisting of Exo, β and γ proteins. An additional λ recombinase, Orf, participates in the early stages of exchange, supplying a function equivalent to the Escherichia coli RecFOR complex. The host enzyme complex promotes the loading of the RecA strand exchange protein onto SSB-coated ssDNA. This thesis describes the purification and biochemical analysis of the λ Orf protein, in parallel with two distantly related homologs from E. coli cryptic prophage DLP12 and Staphylococcus aureus phage ɸETA.X Orf was found to belong to a family of proteins originating from diverse lambdoid phage and prophage sources. Members of this family reside within a conserved genetic module located between phage replication and cell lysis functions. Orf exists as a homodimer, arranged as a toroid with a shallow cleft running perpendicular to the central cavity. K binds preferentially to DNA containing single- stranded regions, and associates with E. coli SSB protein in the presence of ssDNA. The Orf homolog from E. coli DLP12 displayed similar properties. This work suggests that members of the Orf family function as recombination mediator proteins, stimulating the assembly of strand exchange proteins onto ssDNA, and highlights the importance of overcoming the barrier presented by SSB proteins during lambdoid phage recombination.

The aim of this study was to functionally characterise the A1HV-1 ORF 57 gene product. The studies carried out have shown that A1HV-1 ORF 57 is expressed as an immediate early gene. It has also been demonstrated that the ORF 57 gene product is a transactivator protein with post-transcriptional regulatory functions. In transient transfection assays A1HV-1 ORF 57 gene product has been shown to activate transcription from only the A1HV-1 ORF 50 promoter. The ORF57 gene product up-regulated the activity of an unspliced reporter gene driven by ORF 50 promoter to 50 fold and of a spliced reporter gene driven by ORF 50 promoter to 6 fold: RT-PCR analysis using RNAs extracted from BHK cells transfected with spliced and unspliced 50 promoter reporter constructs with or without addition of ORF 57 expression construct have demonstrated that the increase in the amount of protein, produced was not correlated with a similar increase in the mRNA level. This indicated that the ORF 57 gene product acts at a post-transcriptional level. Transient transfection assays with full length ORF 57 and truncations of ORF 57 have demonstrated that the activation domain of ORF 57 lies at the C-terminus. When ORF 57 was fused to the enhanced green fluorescent protein (EGFP), the fusion protein exhibited a punctate nuclear distribution in transfected BHK cells. Immunofluorescence studies have shown that ORF 57 protein co-localises with the nucleolar phosphoprotein C23. It was also observed that EGFP-ORF 57 shuttles from the nucleus to the cytoplasm in the presence of Actinomycin D. The functional domain responsible for the nuclear localisation of ORF57 protein was found to reside at the N-terminus. Thus, this study has demonstrated that A1HV-1 ORF 57 gene product shares some of the functional properties with other gammaherpesvirus homologues.

Abstract:
Venous thromboembolism (VTE) encompasses both deep vein thrombosis (DVT) and pulmonary embolism (PE).1,2 Factors such as venous stasis, vascular injury, hypercoagulability and major orthopaedic surgery such as hip/ knee replacement or hip fracture surgery increases the risk for thrombosis. Currently there is limited information on the incidence and risk of VTE following ankle fracture surgery and the economic impact on the use of prophylaxis for the prevention of VTE in these patients. Chapter one of this thesis provides a comprehensive literature review highlighting the epidemiology, risk factors, prophylactic modalities used and complications in both medical and surgical patients for the prevention of VTE. Chapter two is a retrospective study that examines this area, in particular, practice patterns in VTE prevention both in-hospital and beyond hospitalisation following ankle fracture surgery. These results showed that most of these patients received low molecular weight heparin (LMWH) and that the overall in-hospital VTE incidence was 2.9% (95% CI: 1.3 - 4.4). There was a 1.5-fold increase risk of VTE at three months post hospitalisation (4.3% (95% CI: 2.3 - 6.2), bleeding rates were low and there were no major complications. This led to a systematic literature review for chapter three evaluating the economic impact with use of pharmacological prophylaxis in major orthopaedic surgery for VTE prevention as well as complications and treatment costs. These results highlighted economic benefits with the use of LMWH compared to difference in healthcare systems. The first two chapters emphasise the importance of immobility for the risk of thrombosis. This is discussed in chapter four together with a summary of the main findings, gaps and future directions. The need to explore the importance of immobility on DVT risk led to a protocol development and ethics approval for the evaluation of the rates of DVT in diabetic foot ulcer patients with total contact casts.

The anaphase promoting complex (APC/C) is an ubiquitin ligase that is an essential regulator of multiple steps in the cell cycle. The complex consists of at least 12 subunits with a catalytic core formed by a scaffold protein, APC2, and a RING-H2 protein, APC11. The Parapoxvirus, Orf virus (OV), encodes a RING-H2 protein, B5L, with clear sequence similarities to APC11. The disruption of APC/C function leads to pre-mature entry into S phase and a delayed M phase exit and, potentially, apoptosis. This investigation explored the functional significance of the similarity between B5L and APC11 and specifically sought to determine if B5L manipulates cell cycle regulation by targeting APC/C function. Co-immunoprecipitation experiments from lysates of cells expressing a range of constructs revealed an interaction between B5L and APC2 in the same manner as seen with APC11. Furthermore, B5L was found to associate with endogenous APC/C. However, although APC11 promoted the formation of polyubiquitin chains in substrate-independent in vitro assays, B5L was inactive in this assay. Bioinformatics comparisons of APC11 and other known RING ubiquitin ligases with B5L and its poxviral homologues revealed some subtle differences. In particular a domain of APC11 (amino acids 61-74), that is essential for its ubiquitin ligase activity is not conserved in B5L or its homologues. When this APC11 domain was incorporated in place of the corresponding region of B5L (amino acids 59-67), the mutated B5L acquired ubiquitin ligase activity. On the other hand, APC11 protein in which the domain was replaced with that of B5L lost ubiquitin ligase activity. Stable cell lines expressing B5L showed an increased number of cells in G2/M phase (30�4%) compared with cell lines expressing APC11 (11�2%, n=3, p<0.05, ANOVA, Tukey�s), consistent with impaired APC/C function. APC/C substrates such as cyclin A, cyclin B and the thymidine kinase were stablized in B5L-expressing cells compared with control cells. Furthermore, transient hyper-expression of B5L induced apoptosis in 25�2% (n=3, p<0.05) of the cell population compared with only 6�1% apoptotic cells when APC11 was hyper-expressed. Analysis of the DNA content of OV-infected cells revealed enhanced DNA synthesis compared with cells infected with a B5L knockout OV. These observations indicate that B5L is a non-functional mimic of APC11. It associates with APC/C, but lacks ubiquitin ligase activity, and hence disrupts APC/C function. These abilities may enable OV to induce a cellular environment that enhances viral replication.

The purpose of this research was to investigate the relationship between the oxidation-reduction potential (ORP) measurement and volatile fatty acid (VFA) production in the acid-phase anaerobic digestion process under different conditions of temperature and residence time. Two identical anaerobic digesters were operated while VFAs, SCOD, VSS, alkalinity, ORP and pH were measured. In digester 1, VFA production of 5,556 mg/L was generated with an ORP of -315 mv at a 10 day SRT; while 5,400 mg/L of VFA with an ORP of -389 mv was recorded in digester 2. The SRT was adjusted at 5, 8, 10, 12 and 15 days and the optimum SRT was 10 days in both digesters. The results of this study indicate there were no tight relationship between VFA production and ORP values, thus ORP by itself is not a good predictor of the amount of VFAs generated. However, ORP combined with temperature had good linear relationship with VFA production. An ORP range of -315 to -390 mv was desirable for maximizing VFA production in both anaerobic digesters. Different temperatures (14, 29 and 37 ℃) were trialed and the results indicate that the conditions at 29 ℃ and 37 ℃ were not significantly different in terms of VFA production, however, less VFAs were generated at the lowest temperature of 14 ℃.

Analysis of the proteins of capripoxvirus (KS-1) revealed a 32kd protein that is one of the major structural proteins of the virus and is localised in the virus envelope. Monospecific serum prepared against the 32kd envelope protein neutralised the virus indicating that this protein contains neutralising epitopes. Lymphocyte proliferation studies, using the 32kd protein and peripheral blood mononuclear cells from capripoxvirus (KS-i) vaccinated sheep, showed that this protein strongly induced cellmediated immune responses. The 32kd protein is capripoxvirus specific and induced antibodies in early stages of capripoxvirus infections. Immunoblot analysis of antibody responses against this protein has provided a basis for the differential diagnosis of capripoxvirus and orf virus infections. The 32kd protein bound to the surface of cultured lamb testis cells. The binding of the 32kd protein was completely inhibited by prior incubation of cells with purified capripoxvirus (KS-1) but not by bovine serum albumin. Trypsin treatment of capripoxvirus (KS-1) degraded the majority of the 32kd protein with a minimal effect on a few other virus proteins. Trypsin removed an external 10kd fragment from the 32kd protein, leaving a 22kd fragment associated with the virus. In addition, the trypsin treatment reduced the virus infectivity by at least ten fold, suggesting that the cell surface binding domain of the 32kd protein is located within the external 10kd fragment. The monospecific serum to the 32kd protein had no effect of the infectivity titre of the trypsin treated virus further supporting the concept that the external 10kd fragment of the 32kd protein is involved in binding of the virus particle to the cell surface. A degenerate oligonucleotide probe, based on an internal amino acid sequence obtained from V8 protease cleavage products of the 32kd protein, was used to identify the gene encoding the 32kd protein. The gene encoding the 32kd protein was identified within the 2.8kb HindI1l Q1 fragment of the capripoxvirus (KS-1) genome. The nucleotide sequence analysis of the Hindu Q1 fragment revealed five open reading frames (Q11L, Q12R, Q13L, Q14R and Q15L), one of these open reading frames, Q13L, is capable of encoding a 30.6kd protein and contains the complete internal amino acid sequence obtained from the V8 protease cleavage products of the 32kd protein, indicating that the Q13L encodes the 32kd envelope protein of capripoxvirus (KS-1). The deduced amino acid sequence of the Q13L shows a 34.1% identity and 61.3% similarity with that of H3L open reading frame of vaccinia virus.

L'etude de leur expression d'une part et de la fonction des proteines codees d'autre part, a ete entreprise. Les cinetiques d'expression des transcrits au cours du cycle viral dans les cellules sf21, ainsi que les sites d'initiation et de terminaison de la transcription ont ete determines. Ces donnees transcriptionnelles, cumulees avec une analyse de l'activite des promoteurs en cellules non infectees, nous ont permis de montrer que pnk/pnl etait un gene precoce immediat, alors que lef-4 et lef-5 etaient du type precoce retarde. La suppression du gene pnk/pnl du genome d'acmnpv c6 (knock-out) a permis de montrer que ce gene n'etait pas indispensable pour l'execution du cycle viral. Par ailleurs, l'analyse de populations virales a permis de montrer que ce gene etait absent de certaines souches d'acmnpv et d'autres virus comme bmnpv et opmnpv. L'obtention d'un anticorps dirige contre des peptides internes a lef-4, a permis d'une part, d'etudier la cinetique d'expression de cette proteine, et d'autre part de determiner la localisation sub-cellulaire par des techniques immunologiques couplees a l'observation en microscopie confocale et electronique. Differents systemes d'expression ont ete utilises, afin de produire les proteines en quantite importante. En systeme bacterien, des problemes d'insolubilite ont ete rencontres. En complement, le systeme baculovirus a ete utilise pour l'hyper-production de lef-4 et lef-5. Deux virus recombinants, acoverlef-4 et acoverlef-5 ont ete construits. Le virus acoverlef-4 nous a permis de realiser les premieres experiences concernant la determination des domaines fonctionnels de lef-4.

Thesis (D.M.A.)--Boston University<br>This study examines teacher training in Orff Schulwerk in the United States and how the training system currently in practice was initially developed. Since the first Orff Schulwerk teacher training, conducted by Gunild Keetman at the Mozarteum in Salzburg, Austria in 1953, teachers have been seeking ways to adapt the Schulwerk approach to classrooms. By the mid-1950s teachers were traveling to Austria from throughout the world to learn how to use these ideas and publications. By the early 1960s interest was growing in the United States as evidenced by the founding of the national organization, now the American Orff-Schulwerk Association (AOSA), in Muncie, Indiana in 1968. Teacher training courses sprang up like "wildflowers," a term often used in the movement, in universities and local school systems during this period. The history of teacher training in the Orff Schulwerk approach in the United States is chronicled here, including why and how it evolved from free-form workshops exploring the creativity of Orff Schulwerk into a three-level sequential curriculum. The time line begins with those offered in the late 1950s through the establishment of the first AOSA Course Guidelines for Levels I, II, and III in 1980. Special focus is given to the development of teacher training programs listed as certification courses by the AOSA Higher Education Committee in The Orff Echo (Spring 1977). Responding to the need for some level of common practice in the burgeoning training programs that sprang up throughout the nation, members ofthis AOSA committee selected three models and organized the Level II and III guidelines into three curricular tracks, based on common practice in the curriculum outlines from the more than thirty programs surveyed. Common goals were determined, but differences arose over the length of courses and content sequence in Levels II and III. The compromise decided upon allowed for creative license when designing how to organize the material. This information was compiled from interviews with early teachers of Orff Schulwerk training courses in the USA, a study of archived materials from individuals and universities, a review of journal articles, dissertations, and theses, and AOSA video-recorded interviews.

The emergence of novel pathogenic organisms due to the acquisition of virulence determinants from bacteriophages has generated significant interest in the pathways responsible for genomic rearrangements. Phageλ encodes its own recombination system, the Red system, comprising Exo, β and γ proteins. In addition,λ encodes another recombinase, Orf, which participates in the initial stages of genetic exchange and supplies a frmction equivalent to that of the Escherichia coli RecFOR proteins. This thesis focuses on determining the function of Orf in phage and bacterial recombination pathways by analysing its impact on recombinases encoded by λ and E. coli. Experiments revealed that Orf interacts with bacterial and phage recombination proteins in the initial exchange step of recombination, modulating the activities of both Exo and RecA. Orf, along with β, attenuates the 5'-3' exonuclease activity of Exo, a feature that depends largely on the ability of Orf to bind DNA. Orf also facilitates loading of RecA onto ssDNA pre-coat SSB but only if a ssDNA:dsDNA intersection is incorporated in the substrate. A motif similar to that found at the BRCA2-Rad51 interface may be responsible for Orf mimicking a RECA monomer to initiate nucleoprotein filament formation. Significantly, this would direct recombination down the bacterial RecA pathway of break restoration rather than the phage Red pathway with potentially important consequences for the outcome of the exchange reaction.

A doença atinge todas as variedades comerciais de citros e, o entendimento da relação fitopatogênica permitirá conhecer formas mais eficientes de controle da bactéria. Com o sequenciamento e análise do genoma e transcriptoma da Xac, foram identificadas ORFs que, provavelmente, estão envolvidas em processos de ataque e colonização da bactéria. Para iniciar a caracterização de uma destas ORFs foram utilizadas, neste estudo, as estratégias de mutação sítio-dirigida de PCR por sobreposição e extensão e modelagem molecular. A ORF XAC0239, é predita como proteína de membrana plasmática, e por homologia, possivelmente, pode fazer parte de uma proteína transportadora de zinco, além disso, há indícios de que é regulada por um fator sigma. A mutação teve efeito negativo sobre a ação de celulases e motilidade swimming e efeito positivo sobre a formação de biofilme e a multiplicação bacteriana in vivo. Aqui é sugerido que na falta desta proteína, houve pouca absorção de zinco, necessário para a formação de biofilme e consequentemente, para a patogenicidade. Além disso, sugestiona-se que, como subterfúgio, a bactéria superexpressou genes para produção e/ou ação de celulases, tentando atacar o hospedeiro de forma alternativa.<br>Xanthomonas citri subsp. citri (Xac) causes the citrus canker disease. The disease reaches all commercial varieties of citrus and, the understanding of the phytopathogenic relationship allow knowing the ways of control of bacteria. With sequencing of the Xac genome and the realization of a transcriptome, ORFs have been identified that are probably involved in bacterial attack and colonization processes. To initiate the characterization of one of these ORFs, in this study, the strategies of site-directed mutagenesis by overlap extension PCR and molecular modeling were used. The ORF XAC0239 is predicted as plasma membrane protein, and by homology may possibly be part of a zinc transporter protein, moreover, there are indications that it is regulated by a sigma factor. The mutation had a negative effect at the cellulases action and swimming motility and a positive effect on biofilm formation and in vivo multiplication. Here it is suggested that in the absence of this protein, there was little absorption of zinc, necessary for biofilm formation and consequently for pathogenicity. In addition, it is suggested that, as a subterfuge, the bacteria overexpressed genes for production and / or cellulases action, trying to attack the host on alternative way.<br>Mestre

Given the change in federal legislation allowing the consideration of a Response-To-Intervention (RTI) approach to making eligibility decisions for specific learning disabilities (IDEIA, 2004), it is crucial to establish sufficient evidence for using curriculum-based measurements (CBM) or CBM-like measures (e.g., Dynamic Indicators of Basic Early Literacy Skills; DIBELS) for monitoring students' responses to interventions as part of the identification process for Specific Learning Disabilities. One critical component in an effective RTI model is the availability of parallel measures suited for frequent progress monitoring to index student performance over time. The current study investigated the equivalence of the twenty first grade DIBELS Oral Reading Fluency (DORF) progress monitoring probes' using a one-way ANOVA for repeated-measures. Participants were 68 first grade students (36 females and 32 males) from two Midwestern elementary schools. No selection criteria other than parental consent were used. The DORF probes were given in a pseudo-counterbalanced order across four days at the end of the school year to avoid instruction and practice effects. The standard error of measurement (SEM) of the twenty first grade DORF progress monitoring probes was calculated using alternate form reliability coefficients. Results suggested that the equivalence assumption of the first grade DORF was not held. Most of the probes were significantly different in their difficulty level from each other and did not yield consistent mean levels of performance. The findings also suggested a significant magnitude of score fluctuation at the individual student level. The average SEM value was about 9 (ranging from 8 to 10), similar to previous research findings using test-retest reliability for calculation. Limitations, educational implications for CBM ORF users and researchers, and directions for future research are presented.

O relato e respectiva reflexão crítica focar-se-ão na observação directa de aulas bem como na prática lectiva referente a dois níveis de ensino: o quinto e o sétimo ano de escolaridades, respectivamente segundo e terceiro ciclos do ensino básico. A componente prática está subordinada ao tema: Arranjo Instrumental na Aula de Educação Musical – Uma reflexão sobre contributos pedagógicos da metodologia Orff no contexto do Currículo do Ensino Básico. O arranjo instrumental pedagógico constitui uma prática recorrente do dia-a-dia do professor de educação musical, sendo genericamente utilizado para promover, de forma rápida e eficiente, o contacto dos alunos com a realização musical e instrumental. Estando comprometido com alguns pressupostos didácticos, como a facilitação e adequação ao nível e processo de aprendizagem musical dos alunos, exige portanto o conhecimento e aplicação de princípios, muitos dos quais já consagrados por músicos e pedagogos, como Carl Orff. Apesar de existir algum material didáctico nesta área, relativamente pouca fundamentação pedagógica parece existir, nomeadamente em Portugal. Por outro lado, parece continuar a ser necessário produzir e criar mais e diferentes recursos pelas seguintes razões: tratamento de novas linguagens (Pop e Rock), adequação de material musical aos recursos instrumentais existentes na sala de aula, integração de processos específicos de aprendizagem, etc. O objectivo deste trabalho é fazer uma breve análise pedagógica do que pode constituir e caracterizar princípios e técnicas de arranjo e harmonização implementadas pela metodologia Orff, tendo em vista a sua aplicação na aula de Educação Musical, e, apresentar alguns exemplos implementados nas sessões realizadas sugestivos quanto à integração, naquele tipo de instrumentos, de estilos como o Pop e o Rock e músicas do mundo nomeadamente a música africana. Para finalizar, e na sequência dos contributos apresentados, será relatada uma descrição e respectiva análise crítica do trabalho prático que realizei, bem como um registo vídeo onde se poderá observar o resultado prático nas turmas com que trabalhei, nomeadamente o 6ºH e 7ºD da Escola EB 2/3 Pedro Jacques de Magalhães.

Orf is a highly contagious, eruptive skin disease of sheep and goats caused by a parapox virus. The virus enters through abrasions in the skin, where it replicates in the regenerating epidermal keratinocytes. Despite the generation of a specific antiviral response, orf virus reinfections can be established easily, although the lesions are milder and generally regress more rapidly than after primary exposure. The cutaneous response to orf involves the formation of a dense network of MHC class II<SUP>+</SUP> dendritic cells at the lesion. The primary aim of this project was to characterise these dendritic cells and to identify the cytokines produced by orf infected keratinocytes <I>in vitro</I>, that might be involved in the accumulation of the dendritic cells <I>in vivo</I>. <I>In vivo</I> studies of primary and secondary orf virus lesions identified the class II<SUP>+</SUP> dendritic cells to be a population of CD1<SUP>-</SUP> cells, which are also found within the dermis of normal ovine skin. A subpopulation of these cells also expressed the antigen, coagulation factor XIIIa. Factor XIIIa<SUP>+</SUP> dendritic cells comprised over half the dendritic cells seen in the network of a primary orf lesion but were only observed in small numbers, transiently, in the secondary orf lesion. All the dendritic cells lacked the expression of ovine macrophage markers. The proliferative response of the primary and secondary orf lesions also differed. High proliferative activity was observed in the epidermis and dermis in the primary response to orf but not in the secondary response. A few of the proliferating cells were identified as dendritic cells but it would appear that the dendritic cell network in both primary and secondary orf lesions does not arise by local cell division.

碩士<br>國立臺北藝術大學<br>音樂學系碩士在職專班<br>94<br>Carl Orff (1895-1982) is one of the famous composers of the 20th century, and his master piece “Carmina Burana” has been performed all over the world until now. His contribution to music education is just as important as composition . Started with the three basic directions-- “Music,” “Movement”, and “Speech”, Orff then added different elements of music, dance, poetry, literature, and drama. With the emphasis of “humaninity”, Orff Method is a comprehensive art education, which served various functions. When he established the school for gymnastics, musicand dance, with Dorothee Günther, Orff composed the Klavier Übung, which was published in 1933 with two volumes, for the reuired piano lessons and basic music skill training. Orff then established the unique Drone method to train the polyphonic and improvising skills, with two or more students playing one or two pianos. And these scores were recorded from memory in the third book of his totally eight books of working diary. This study is based on the two scores of the composer and other related reference sources. This paper is divided into five parts: 1) Introduction of Carl Orff’s life, experience, and thought. 2) Explanation of how did Orff Method started and evolved, with detailed information of each stage and important people. 3) Analysis of the piano practice, and demonstration of forty short pieces with guidelines including melody, rhythm and chord. 4) Uniqueness of Orff Method’s piano practice with references of fifteen practice pieces. 5) Conclusion. The purpose of this study is not to find a new set of teaching method and material, but to bring out the important elements of Orff Method with intense study, to reorganize and identify the parts, and to strengthen the educators’ understanding, both theoretically and practically, of the Orff Method. The most amazing result is to see how the Orff Method’s ideal is being spread out and applied well to the real teaching.
