Abstract:
Oil palm is a very significant industrial plant in producing vegetable oils that require high levels of nutrients in its cultivation. Cultivation of oil palm yield can produce industrial waste, the oil palm empty trunks (TKKS) which have the potential to supply nutrients to oil palm land. The placement of empty trunks at various depths is expected can give effect to the population of Oryctes rhinoceros L pre-adult stage and chemical soil properties. This research was conducted on oil palm plantation in PT. Perkebunan Nusantara V Sei Galuh Tapung, Kampar, Riau from July to October 2017. The aim of this research is to examine the effect of empty trunks placement against the Oryctes rhinoceros L population development and chemical soil properties. This research was done using a randomized experiment design complete with the treatment of the depth empty trunks placement (placement of oil palm empty trunks on the ground at the depth of 10 cm, 20 cm, and 30 cm) which is repeated four times. The observed parameters include the population of infestation of Oryctes rhinoceros L (population of eggs, larvae and pupae) and soil chemical properties (soil pH, C-organic, N-total, P-total and K-total). The results showed that empty trunks placed below the surface at various depths affected the Oryctes rhinoceros L population. Placement of TKKS depth in the soil cannot be infested by Oryctes rhinoceros L. Placement of empty trunks at the depth of the soil tends to increase pH (from very acidic categories to acidic categories) and total K (from the low category to very high category), is not effected on C-organic and N-total and can reduce P-total (from the very high category to the high category) if compared to the placement of empty trunks on the ground.Key words : Chemical Soil properties, Oryctes rhinoceros L., Oil palm Empty trunks, Soil depth

Rhinoceros beetle (Oryctes rhinoceros) is one of the important pests of coconut tree. One of eco-friendly control applied for this pest is by using entomopathogenic fungiMetarhizium anisopliae. There is not much information about the variability and virulence ofM. anisopliae toward O. rhinoceros. M. anisopliae isolates obtained from Biological Control Laboratory, Faculty of Agriculture, Universitas Gadjah Mada were cultured on PDA medium.M. anisopliae isolates was isolated from O. rhinoceros larvae (MaOr), Lepidiota stigma larvae (MaLs), Brontispa longissima beetle (MaBl).O. rhinoceros beetles were obtained from Kulon Progo, DIY. This study used molecular test, and virulence test toward 3rd stadium of O. rhinoceros larvae by using dipping method. Molecular test by sequence and phylogenetic analysis, showed that MaOr was located at different group (out group) with MaLs and MaBr. Belum banyak diketahui mengenai keragaman dan juga virulensi dari M. anisopliae terhadap O. rhinoceros. Tujuan dari penelitian ini adalah untuk mengetahui keragaman genetik M. anisopliae dan virulensinya pada larvakumbang badak. Isolat yang digunakan berasal dari Laboratorium Pengendalian Hayati, Fakultas Pertanian, Universitas Gadjah Mada dalam bentuk kultur murni pada medium PDA. Isolat yang gunakan diisolasi dari larvaOryctes rhinoceros (MaOr), larva Lepidiota stigma (MaLs), dan kumbang Brontispa longissima (MaBl). Serangga yang diuji berasal dari daerah Kulon Progo, DIY. Pengujian secara molekuler dengan analisis sekuensing dan filogenetik, menunjukkan bahwa isolat MaOr terletak pada grup yang berbeda dengan MaLs dan MaBl berdasarkan pada urutan basa DNA. Abstract:
ABSTRACTOryctes rhinoceros is one of the most serious pests in coconut palm tree. Biological control for controlling the pest is done by applying fungal entomopathogen Metarhizium anisopliae on its breeding sites to infect the larvae. Recent development for controlling Oryctes beetle was including the use of pheromone trap baited with ethyl-4-methyl octanoic which attract both male and female of the Oryctes beetle. This research was aimed to determine the effect of combination of both entomopathogen and pheromone application on the population dynamics of rhinoceros beetle, and the intensity of leaf damage on coconut tree. For this purpose, a research was conducted in local farmer coconut tree in the Bojong Village, Panjatan District, Kulon Progo from June 2009−January 2010. Observation including leaf damage intensity before and after application, the number of adult beetle trapped by pheromone, and the number infected larvae in the breeding site. The result showed that there were significant differences among all treatments in term of intensity of leaf damage, the number of trapped adult beetle, and the number of larvae at the breeding site. Leaf damage on control, pheromone application, and combined treatment were: 4.73%; 1.08% and 0.65%. The number of trapped Rhinoceros beetle by ferotrap was 101; in combined treatment was 52. The number of M. anisopliae infected grub were 265 out of 281 total observed grub.Keywords: Metarhizium anisopliae, Oryctes rhinoceros, pheromoneINTISARIKerusakan tanaman kelapa akibat serangan Oryctes rhinoceros terjadi mulai pada tanaman muda. Mengingat besarnya kerugian yang ditimbulkan, maka perlu diupayakan cara pengendalian yang efisien, efektif dan aman bagi sumber daya alam dan lingkungan. Salah satu cara pengendalian secara hayati adalah dengan menggunakan cendawan patogenik Metarhizium anisopliae. Selain menggunakan cendawan, upaya terkini dalam mengendalikan kumbang badak adalah dengan menggunakan perangkap berferomon. Feromon dengan bahan aktif Etil-4-metil oktanoat dapat memikat kumbang Oryctes jantan maupun betina. Penelitian ini bertujuan untuk mengetahui pengaruh gabungan feromon dan Metarhizium anisopliae terhadap dinamika populasi O. rhinoceros dan intensitas kerusakan pada tanaman kelapa. Penelitian ini dilaksanakan di kebun kelapa rakyat di Desa Bojong, Kecamatan Panjatan, Kabupaten Kulon Progo, dari bulan Juni 2009 sampai dengan Januari 2010. Parameter yang diamati adalah intensitas kerusakan sebelum dan setelah perlakuan, jumlah imago yang terperangkap oleh feromon, dan jumlah larva yang berada di breeding site. Analisis data menggunakan analisis varian dan dilanjutkan uji DMRT. Hasil penelitian menunjukkan terdapat beda nyata pada intensitas serangan, jumlah imago dan jumlah larva pada semua perlakuan. Intensitas serangan baru, yakni serangan yang terjadi setelah dilakukan pengendalian, secara berurut dari yang tertinggi adalah perlakuan kontrol, perlakuan perangkap berferomon, dan perlakuan gabungan yakni sebesar 4,73%; 1,08%; dan 0,65%. Jumlah imago yang terperangkap sebesar 101 ekor pada perlakuan ferotrap dan 52 ekor pada perlakuan gabungan. ABSTRACTEffectiveness trial of pheromone traps against Oryctes rhinoceros L. and its damage intensity on coconut tree at Batulahat Village, Nusaniwe District, Ambon IslandThe research aimed to determine the efficacy of traps containing pheromone in catching the coconut pest, coconut rhinoceros beetle (Oryctes rhinoceros L.) and its effect on reducing the damage severity caused by the pest. The experiment was conducted by observing the damage intensity before treatment and continued with the efficacy testing of the traps containing pheromone (Ethyl 4-methyloctanoate) against the population of the coconut rhinoceros beetle. The damage intensity was measured following the traps treatment. The result demonstrated that the average of damage intensity before and after treatment were 13.33% and 9.61%, respectively. This indicated the effectiveness of traps in catching the coconut rhinoceros beetle. The number of O. rhinoceros caught during the experiment was analysed using quantitative analysis model. The highest number of coconut rhinoceros beetle caught was 9 bettles/2 months at traps containing pheromone. Whilst, the number of coconut rhinoceros beetle caught at traps containing pheromone with lamp demonstrated lower number of coconut rhinoceros beetle. Furthermore, the lower damage intensity at the experimental location was categorized as low. Penelitian dilakukan dengan menghitung Intensitas Kerusakan (IK) yang disebabkan oleh hama O. rhinoceros dan dilanjutkan dengan menguji efektivitas perangkap feromon (Ethyl 4-methyloctanoate) terhadap perkembangan populasi hama tersebut. Intensitas kerusakan dihitung dengan menggunakan formula IK untuk tanaman yang terserang dan yang tidak terserang O. rhinoceros sebelum dan sesudah aplikasi feromon. Hasil penelitian menunjukkan rerata IK sebelum dan sesudah aplikasi feromon masing-masing sebesar 13,33% dan 9,61%. Terjadi penurunan IK karena efektivitas feromon dapat memerangkap hama O.rhinoceros. Jumlah tangkapan O. rhinoceros tiap perlakuan dianalisa dengan model analisis kuantitatif sederhana. Jumlah tangkapan O. rhinoceros terbanyak dengan rerata tangkapan 9 ekor/2 bulan atau sama dengan 4,75 ekor/bulan pada perangkap berferomon saja. Sementara perangkap feromon dan lampu menghasilkan jumlah tangkapan hama O. rhinoceros yang lebih rendah. Lebih lanjut, rerata IK hama O. rhinoceros di desa Latuhalat tergolong kategori ringan karena sistem kultur teknik dari petani setempat sudah dilakukan dengan baik sehingga areal pertanaman kelapa cukup terawat.Kata Kunci: Kelapa, Feromon, Intensitas kerusakan

Oryctes rhinoceros beetle is one of the pests that attacksimmature oil palm plants.This pest attacks oil palm plants in larvae or imago phase. This pest is usually damaging the respiratory roots, trunk bark, dry petiole,or periphery crown of oil palm leaves which can interferes the leaf growth and finally reduce the production even collapse the tree. Chemically pest control have the positive impact with faster pest death beside the negative impacts such as resistance, resurgence, disturbing human health and also environmental and ecosystem pollution. Biological control is one alternative pest control that is safe for the environment and can reduce chemical residues in agricultural products and environment. This study was conducted in Medan Denai district on July – Agustus 2019. This study aimed to determine the effect of the use of Beauveria bassiana and palm oil mill effluent to control Oryctes rhinoceros larvae. The method used in this study is a two factorial randomized block design (RBD), with 2 replications. The results of this study showed that the best density of Beauveria bassiana spores in controlling Oryctes rhinoceros larvae was 109 spores / ml and 50ml LCPKS (I2Q2) with death time of 40 hours, daily mortality 60%, total mortality 100%.

Abstract:
El género Oryctes cuenta con 42 especies conocidas en todo el mundo, y en su mayoría corresponden a plagas de la palma. Oryctes rhinoceros es uno de los insectos más dañinos para el cocotero (Cocos nucifera) y para la palma aceitera africana (Elaeis guineensis) en el sur, sureste de Asia y las islas del oeste del Pacífico, aunque puede atacar otras palmas ornamentales. En el presente trabajo se documenta el primer registro de intercepción del Oryctes rhinoceros (escarabajo rinoceronte del cocotero) en el puerto de Manzanillo, Colima; uno de los puntos de ingreso de mercancías más importantes de México. Se colectó un solo espécimen adulto y vivo en muebles de palma, procedentes de Indonesia, cuyo destino final era el estado de Jalisco. La muestra se envió al Laboratorio de Análisis y Referencia en Sanidad Forestal de la Secretaría del Medio Ambiente y Recursos Naturales, para su identificación, posteriormente, se establecieron medidas fitosanitarias para impedir que este insecto, plaga exótica; ingresara al país. Asimismo, se exponen algunas consideraciones acerca de las implicaciones económicas que tendría para México, si O. rhinoceros se estableciera en el territorio nacional.

Abstract:
Experimental evidence from previous study has demonstrated the hypolipidemic effects of Oryctes rhinoceros oil (ORO) when fed as a supplement to a cholesterol-based diet. Due to renew interest in the consumption of insect derived oil, the present study was designed to elucidate the effect of Oryctes rhinoceros oil (ORO) supplementation in comparison to vitamin E on oxidative status in some tissues of rats fed a cholesterol-based diet. Forty (40) Swiss albino rats were divided into 4 groups (n = 10) and maintained on a basal diet (cholesterol free as control), a cholesterol-based diet (5% cholesterol as cholesterol), a cholesterol-based diet supplemented with ORO (cholesterol + ORO) and a cholesterol-based diet supplemented with vitamin E (Cholesterol + vit E) for 10 weeks. Animals in the cholesterol group had a significantly (p <0.05) higher malondialdehyde (MDA), conjugated diene and nitric oxide concentrations in the serum, liver, heart, kidney and lung compared to control, cholesterol + ORO and cholesterol + vit E groups. Tissue glutathione (GSH) concentration was significantly (p <0.05) higher in rats fed cholesterol-based diet supplemented with ORO and vitamin E compared to those fed cholesterol-based diet alone. Xanthine oxidase activity was significantly (p <0.05) reduced in tissues of rats fed ORO and vitamin E supplemented diets compared to cholesterol rat group. In addition, catalase and superoxide dismutase activities in the various tissues examined were significantly (p <0.05) higher in both ORO and vitamin E supplemented groups compared to the cholesterol group. No significant difference was observed between animals fed ORO and vitamin E supplemented diets. These results showed that Oryctes rhinoceros larva oil exhibited similar protective effects to vitamin E against diet-induced oxidative stress in rats. In addition, data from this study showed that Oryctes rhinoceros larva oil possessed antioxidant property. Overall, the potential nutritional benefit of Oryctes rhincoceros larva oil consumption on cardiovascular health could possibly involve its ability to upregulation of cellular antioxidant defense mechanisms.

Abstract:
The coconut rhinoceros beetle, an economically important pest of coconut and oil palms, is effectively managed by application of its natural pathogen, the Oryctes nudivirus (OrNV), which act as a bioinsecticide. While this approach offers an environment-friendly alternative to chemical pesticides, the current method of production in infected larvae suffers from inconsistencies in virus productivity and purity. While the anchorage-dependent DSIR-HA-1179 insect cell line has been identified as a susceptible and permissive host for OrNV and therefore would be suitable for the in vitro mass production of the virus, no attempts have been made toward the mass production of the virus, because of the technological challenges that working with DSIR-HA-1179 cells represent. Thus, the main objective of this research was to develop processes for the in vitro production of OrNV in the DSIR-HA-1179 cell line. Knowledge of the growth kinetics and metabolic properties of the host cell line in a chosen culture medium, as well as the selection of an appropriate infection strategy, form the basis for the rational development of bioreactor-based virus production processes. However, characterization of these properties in the DSIR-HA-1179 cell line has been virtually precluded, due to its strongly adherent growth characteristics and the lack of a reliable method to accurately dissociate and count cells grown in monolayers. Using TrypLE™ Express enzyme, a technique allowing the precise counting of cells was developed. The cell line was adapted to grow in four serum-supplemented culture media: TC-100, IPL-41, Sf-900 II and Sf-900 III, which were then individually screened for cell growth and virus production in 25 cm2 attached T-flask cultures. TC-100 supplemented with 10% fetal bovine serum was chosen as a suitable culture medium, based on its capacity for achieving a high cell yield and OrNV production. The cell line metabolism was characterized with respect to nutrient consumption and metabolites production in this culture medium. Glucose, along with glutamine were found to be the nutrients that were consumed faster and to a greater extent, while other amino acids were not consumed to a significant degree. The production of metabolites was characterized by non-production of lactate and ammonia, and production of alanine, as a non-toxic alternative to ammonia. The influence of cell density (CD) at time of infection (TOI) and multiplicity of infection (MOI) on OrNV production was evaluated in T-flask cultures that were infected at different CDs at the TOI and a range of MOIs. The CD at TOI was found to significantly influence OrNV yields, while MOI influenced the dynamics of infection. The cell density effect was found to exist for the DSIR-HA-1179/OrNV system with the progressive decline in cell-specific yield beginning at low cell densities. It was found that in order to maximize OrNV volumetric yield, a combination of MOI and CD at TOI should be selected that allows to keep the maximum cell density reached by the infected culture within a range between 5.0 and 7.0 x 105 viable cells/ml. The roller bottle system was evaluated for its potential to scale-up DSIR-HA-1179 cell growth and OrNV production, and culture parameters were optimized for the improvement of cell and virus yields. An inoculum density of 3.3 x 105 cells/ml and culture volume of 60 ml resulted in the highest cell yield of 1.5 x 106 cells/ml, in 490 cm2 roller bottles. It was found that an optimal infection strategy for roller bottle cultures, which represented the most efficient use of viral inoculum, involved infecting cells at a density of 5.0 x 105 cells/ml and at a MOI of 1. The resulting OrNV volumetric yield of 2.5 x108 TCID50/ml, improved significantly the viral yields obtained in attached T-flask cultures infected under similar conditions (6.8 x 107 TCID50/ml). The microcarrier system was also evaluated for culturing DSIR-HA-1179 cells and producing OrNV in spinner flask bioreactors. Three types of microcarriers (Cytodex-1, Cytodex-3 and Cultispher-G microcarriers) were screened for their ability to support DSIR-HA-1179 growth. Cells attached to Cytodex-1 and 3, but failed to attach to Cultispher-G microcarriers. The final cell density reached in microcarrier culture was dependent on bead type and concentration, and the cell to bead ratio. At an optimal bead concentration of 1 mg/ml and cell to bead ratio of 30, cells grew to a maximum density of 1.7 x 106 cells/ml on Cytodex-1, but only to 1.3 x 106 cells/ml on Cytodex-3 microcarriers. Since it supported higher cell yields, Cytodex-1 was chosen to study the kinetics of OrNV production in this system. Microcarrier cultures infected at a cell density of 5.0 x 105 cells/ml and a MOI of 1, produced OrNV at 1.4 x 108 TCID50/ml, which was higher than the yield obtained in T-flask cultures infected under similar conditions. A framework of knowledge on the physiology, metabolism and growth kinetics of the DSIR-HA-1179 insect cell line has been developed in this thesis. In addition, the feasibility of using roller bottles and microcarrier systems for the in vitro production of the virus has been ascertained.
