The processes leading to the formation of 2-methyl-2-(methylthio)propanal oxime, known industrially as aldicarb oxime, have been studied. The three stages of the synthesis, viz., chlorination, thiomethylation and oximation have been thoroughly investigated, with the aim of optimising the yield and purity of aldicarb oxime. 碩士<br>國立清華大學<br>原子科學研究所<br>82<br>99mTcO-d,l-HMPAO是目前使用最廣泛的核子醫學腦造影劑之主要成份，其臨床醫學的研究仍如火如荼進行中，而其藥劑來自英國Amersham公司所提供商業化注射小瓶(commercial kit)，價格極其昂貴，因此對基礎科學研究造成不便與負擔，有鑑於此，本研究採用單一步驟合成法，不僅合成出 HMPAO非對映立體異構物更進一步合成出CBPAO非對映立體異構物，其產率分別為 31.0 ± 2% 與 17.0 ± 4.4% ；接著以乙酸乙酯進行部份再結晶 (fractional recrystallization)，有效分離出d,l-form和meso- form 兩種光學異構物，其中d,l-HMPAO和meso-HMPAO之回收率分別為6 ±4%及56± 2%，而d,l-CBPAO和meso-CBPAO之回收率分別為5.5±3.7% 及52±2.8%，經熔點測定、核磁共振光譜儀、元素分析以及質譜儀確認無誤。

This thesis describes the synthesis of hetrocylic oxime esters derived from anthraquinone and heterocyclic acyl chlorides with the hope to get better DNA-cleaving agents. Deprotonation of oxime 2 with sodium hydride followed by treatment with heterocyclic acyl chlorides gave ten substitution products 3a-b﹑4a-b﹑5a-c﹑6a-c, which are being tested for their DNA-cleaving efficiency by Professor J. These compounds were synthesized via alkylation of hydroxyl precursors followed by the reaction with NH2OH . The preliminary assays indicated that (Z)-7-(2-(4-fluorophenyl)-2-(h ydroxyimino)ethoxy)-3,4-dihydroquinolin-2(1H)-one (13c) was the most active against U46619 induced platelet aggregation with an IC50 value of 3.51 μM.
