Abstract:
Chrome (VI) has a high level of toxicity. Formaldehyde is potential to be developed as a substitute of chrome tanning agent. The aims of this study are to explain the amount of formaldehyde bound, shrinkage temperature, tensile strength, and tear strength on Mondol stingray (Himantura gerrardi) leather. Pickled mondol stingray has been used as raw material. The study was divided into 4 treatments, that is P.I (formaldehyde 2%), P.II (formaldehyde 4%), P.III (formaldehyde 6%), and P.IV (formaldehyde 8%). The method used is drum tanning. Shrinkage temperatures, tensile strength and tear strength of P.I and P.II are significantly different from P.III and P.IV, but P.I is not significantly different from P.II, whereas P.III is not significantly different from P.IV. P.III is shown to improve the quality of leather stingray more than P.I and P.II, and has fulfilled the requirements of leather stingray according to SNI. 06-6121-1999. It can be concluded that the optimal level of formaldehyde tanning agent that can be used as a chrome tanning agent substitute in tanning Mondol stingray skin is formaldehyde 8% (P.III).

Abstract:
The neisserial porin P.I is a GTP binding protein that forms a voltage-gated channel that translocates into mammalian cell membranes and modulates host cell signaling events. Here, we report that P.I confers invasion of the bacterial pathogen Neisseria gonorrhoeae into Chang epithelial cells and that this event is controlled by GTP, as well as other phosphorus-containing compounds. Bacterial invasion was observed only for strains carrying the P.IA subtype of porin, which is typically associated with the development of disseminated neisserial disease, and did not require opacity outer membrane proteins, previously recognized as gonococcal invasins. Allelic replacement studies showed that bacterial invasiveness cotransferred with the P.IA (por1A) gene. Mutation of the P.I-associated protein Rmp did not alter the invasive properties. Cross-linking of labeled GTP to the porin revealed more efficient GTP binding to the P.IA than P.IB porin subtype. GTP binding was inhibited by an excess of unlabeled GTP, ATP, and GDP, as well as inorganic phosphate, but not by UTP or beta-glycerophosphate, fully in line with the respective invasion-inhibitory activities observed for these compounds. The P.IA-mediated cellular invasion may explain the more invasive behavior of P.IA strains in the natural infection and may broaden the basis for the development of a P.I-based gonococcal vaccine.

In 2019, 220 years since the birth of the great Russian national poet A.S. Pushkin and 190 years since the birth of P.I. Bartenev. The right to immortality is justified by people's memory and love. And A.S. Pushkin, and PI Bartenev belonged to the oldest noble families. P.I. Bartenev, following A.S. Pushkin, "a proud ancient man of his kind" and zealously serving the Fatherland, preserves rare documents of the era, spreading them accessible to the Russian public. Interest in the preservation of Pushkin's heritage was not accidental. As a student at Moscow University P.I. Bartenev could get carried away by the personality of A.S. Pushkin, listening to the lectures of Professor SP Shevyreva and M.P. Pogodin, contemporaries of the poet. A subsequent acquaintance with the book. P.A. Vyazemsky, A.S. Khomyakov, the Aksakovs family - deeply honored the name of A.S. Pushkin encouraged him to search for unpublished, unknown materials about the poet. Studies on A.S. Pushkin is represented in the scientific and educational activities of P.I. Bartenev.

AbstractThe effect of albendazole therapy on the reduction of drugsusceptible and drug-resistant strains of Haemonchus contortus larvae on day 10 post infection (p.i.), distribution and the relative numbers of innate immunity cells — eosinophils/neutrophils and mast cells in the stomach wall of immunosupressed Mongolian gerbils on days 4/1, 7/4, 10/7 and 14/11 post infection/post therapy (p.i./p.t.) were investigated in the present study. The efficacy of albendazole was significantly lower on benzimidazole (BZ) resistant larvae (L3 and L4 stages) (58.92 %) than the efficacy on susceptible strain of larvae (94.15 %). H. contortus infection elicited strong inflammation in mucosal and submucosal layers of the stomach, where mucosal mast cells MMC) were in the highest numbers in the lamina propria mucosae on day 7/4 p.i./p.t. Reduction of larval numbers following treatment resulted in a gradual decrease of MMC and connective tissue mast cells (CTMC). The lower counts of CTMC in the submucosa were seen in gerbils infected with BZ-susceptible strain during the whole period post therapy. In case of infection with BZ-resistant strain, peroxidase containig cells (eosinophils) peaked on day 7/4 p.i./p.t., whereas infection with BZ-susceptible strain elicited massive accumulation of these cells on day 4/1 p.i./p.t., particularly in the submucosa. No marked differences in eosinophils localisation were observed between both groups after the therapy. Goblet cells were found only in the proximal parts of glandulae gastricae close to the mucosal surface and no differences in the distribution in the stomach wall of both groups of animals were observed. After therapy the higher larval counts in case of BZ-resistant strain were in the correlation with the lower decline of CTMC and eosinophils, but MMC numbers were not significantly different between both treated groups. Present data indicate that in early stage post infection, the distribution of individual innate immunity cells might be directly affected by the larvae, and that the genetic and consequently biological differences related to the resistance to benzimidazoles probably had the impact on the interactions of larvae with the different immune cells in their niche.

BACKGROUND: Oral infections associate statistically with cancer. OBJECTIVE: We hypothesized that certain periodontal microorganisms might specifically link to malignancies in general and set out to investigate this in our ongoing cohort study. METHODS: A sample of 99 clinically examined patients from our cohort of 1676 subjects was used to statistically investigate the associations between harboring periodontal microorganisms Aggregatibacter actinomycetemcomitans (A.a), Porphyromonas gingivalis (P.g), Prevotella intermedia (P.i), Tannerella forsythia (T.f) and Treponema denticola (T.d). We used oral infection indexes and the incidence figures of malignancies as registered in 2008–2016 in the Swedish National Cancer Register. RESULTS: The pathogen A.a showed strong association with malignancy in 32 out of the 99 patients while P.g and P.i were more prevalent among patients without malignancy. In principal component analyses, A.a appeared in the strongest component while the second strongest component consisted of a combination of T.f and T.d. The third component consisted of a combination of P.g and P.i, respectively. Of basic and oral health variables, gingival index appeared to be the strongest expression of inflammation (Eigen value 4.11 and Explained Variance 68.44 percent). CONCLUSIONS: The results partly confirmed our hypothesis by showing that harboring certain periodontal bacteria might link to malignancy. However, the associations are statistical and no conclusions can be drawn about causality.

Increasing attention has been paid to the possible link between periodontal disease and atherosclerosis over the past decade. The aim of this study is to investigate the presence of five periopathogens: Porphyromonas gingivalis (P.g.), Aggregatibacter actinomycetemcomitans (A.a.), Tannerella forsythia (T.f.), Treponema denticola (T.d.), and Prevotella intermedia (P.i.) in atheromatous plaques obtained from the carotid and coronary arteries in patients who underwent coronary artery bypass graft surgery and carotid endarterectomy. Group I (carotid arteries) consisted of 30 patients (mean age: 54.5 ± 14.8 ), and group II (coronary arteries) consisted of 28 patients (mean age: 63 ± 12.1 ). Clinical periodontal examinations consisted of plaque index, gingival index, sulcus bleeding index, and periodontal probing depth and were performed on the day of vascular surgery. The presence of periopathogens in periodontal pockets and atherosclerotic vessels was detected using polymerase chain reaction. In both subgingival plaque and atherosclerotic plaque of carotid arteries, P.g., A.a., T.f., T.d., and P.i. were detected in 26.7%, 6.7%, 66.7%, 10.0%, and 20.0%, respectively, while for coronary arteries, P.g. was detected in 39.3%, A.a. in 25%, T.f. in 46.4%, T.d. in 7.1%, and P.i. in 35.7%. The presence of five periopathogens in carotid and coronary atherosclerotic vessels showed correlation in regard to the degree of periodontal inflammation. The present study suggests the relationship between periodontal pathogenic bacteria and atherogenesis. Further studies are necessary in relation to the prevention or treatment of periodontal disease that would result in reduced mortality and morbidity associated with atherosclerosis.

AbstractThe immunological, haematological and enzymatic responses to the inoculation in pigs of 100,000 embryonated eggs of Toxocara canis were studied. Fifteen females were inoculated and three remained as controls. Haematological values were analysed from day 7 p.i. until day 126 p.i. In the inoculated group, white blood cells were raised on day 14 p.i. and eosinophil values on days 7, 14, 21, 35 and 49 p.i. showing significant differences compared with controls (P<0.05). Absolute eosinophil counts (per ml) presented two rises, the first on days 7, 14 and 21 p.i. and the second on days 35 and 49 p.i. Blood biochemistry was maintained within normal values. Serological examination by ELISA to determine antibody levels against Toxocara canis L2/L3 excretory–secretory (ES) antigens showed values higher than the positive cut-off (1:32) from day 7 p.i. and until the end of the study on day 126 p.i., presenting two peaks: one on day 28 p.i. and the second covering days 49 to 56 p.i. Western blots of sera of inoculated animals presented, from day 7 p.i., two polypeptide bands of 55 and 70 kDa MW and, from day 56 p.i., an additional band of 120 kDa MW, all of which persisted until the end of the study. Immunological responses were sustained over time. No direct correlation was observed between the rise in eosinophils and antibody titres. To validate the conclusions, more studies are required on the polypeptide bands.

The genital primordia in excysted metacercariae were differentiated into those of metraterm, Mehlis' gland, ovary and cirrus pouch, with the primordial testes appearing on day 1 p.i. The vitellaria and eggs in the uterus were present in flukes on days 2 and 4 p.i., respectively. Mature flukes were established in the lower part of the small intestine on day 5 p.i., with the peak of egg production occurring on day 14 p.i. Growth of the flukes continued up to day 28 p.i.

AbstractThe bird nasal schistosome Trichobilharzia regenti is a new agent of cercarial dermatitis. Cercariae are able to penetrate the skin of birds and mammals including man. The parasite then attacks the central nervous system. The present study has shown that schistosomula avoid penetration of blood capillaries and enter the peripheral nerves of the legs of mice and ducks as early as 1 day post-infection (p.i.) and 1.5 days p.i., respectively. These peripheral nerves are used as a route to the spinal cord. In the specific host (duck) schistosomula were found in the spinal cord from 2 days p.i. until 15 days p.i. and in the brain from 12 days p.i. until 18 days p.i. In non-specific hosts (mice; inbred strains BALB/c, hr/hr, SCID) living schistosomula were found in the spinal cord from 2 days p.i. until 21 or 24 days p.i. (depending on the mouse strain) and in the brain of two (BALB/c, SCID) of three inbred strains from 3 days p.i. until 24 days p.i. No correlation was found between the infection dose and clinical status of the experimental hosts. A high affinity of schistosomula for the peripheral nerves was also proved in vitro, suggesting a new type of migratory behaviour in schistosomatids.

Des nymphes ont été observées jusqu'au 23ème jour après l'infestation et des adultes ont été vus à partir du 15ème jour p.i. Les tiques mâles se sont montrées actives à partir du 17ème jour p.i. et le détachement des femelles gorgées a commencé entre le 22ème jour et le 24ème jour p.i. Un détachement massif est apparu à partir du 28ème jour p.i. Les sites préférentiels des stades parasitaires étaient les oreilles et le cou (parois latérales).

While healthcare industry is striving to achieve e-health systems for improvements in healthcare quality, cost, and access, privacy and security about medical records should be considered carefully. This paper makes a deep study of Public Key Infrastructures (PKIs) and Privilege Management Infrastructures (PMIs) and how they can secure e-health systems. To access resources, e.g. patient records, both authentication and authorization are needed, so public key certificates and attribute certificates are both required to protect healthcare information. From a typical medical scenario, we see not only static but also dynamic permissions are required. Dynamic authorization maybe the most complex problem in e-health systems.

We present a search for physics beyond the standard model (SM) through a measurement of the violation of the charge-parity (CP) symmetry in two decays of the Bs meson using data collected by the Collider Detector at Fermilab (CDF) in proton-antiproton collisions at the center-of-mass energy of 1.96 Tev. We exploit the decays Bs->Jpsi (->mu mu) phi(-> KK) and Bs -> phi(-> KK) phi(-> KK), for which the SM accurately predicts very small or vanishing CP violation; both decay modes are very sensitive to new sources of CP violation expected in a broad class of SM extensions. We analyze the time-dependent CP asymmetry of the Bs->Jpsi phi decays collected in the full CDF Run II dataset for providing the final measurement of the Bs-antiBs mixing phase, 2*betas, and we present the first measurement of CP violation in Bs -> Phi Phi decays, through the determination of two time-integrated CP asymmetries, Av and Au, using an original method developed in this work. We find: -0.06< betas<0.30 at the 68% confidence level; Av=(-12.0 +/-6.4(stat) +/- 1.6(syst))%; and Au=(-0.7 +/-6.4(stat) +/- 1.8(syst))%. In addition, we provide measurements of the decay width difference between the light and heavy mass eigenstates of the Bs meson, DeltaGamma_s=0.068 +/- 0.026(stat) +/- 0.009(syst) ps^{-1}; and of their mean lifetime, tau_s=1.528 +/- 0.019(stat) +/- 0.009(syst) ps. All results are among the most precise determinations from a single experiment and exhibit an excellent agreement with the SM predictions. They have been published in two letters in Physical Review.

In this dissertation, we present the results of a time-dependent angular analysis of Bs -+ J/,0 decays performed with the use of initial-state flavor tagging. CP violation is observed in this mode through the interference of decay without net mixing and decay with net mixing, that is, Bs -- J/0 and Bs -+ B, -+ J/q0. The time-dependent angular analysis is used to extract the decay widths of the heavy and light B, eigenstates and the difference between these decay widths AF, - FL - FH. Initial-state flavor tagging is used to determine the matter-antimatter content of the B. mesons at production time. We combine flavor tagging with the angular analysis, which statistically determines the contributions of the CP-even and CP- odd components at decay time, to measure the CP-violating phase 0,. The phase 3, is expressed in terms of elements of the Cabibbo-Kobayashi-Maskawa matrix as s - arg (-VtsV4/VcsV*b), and is predicted by the Standard Model to be close to zero, O3SM = 0.02. In the measurement of AF,, we use a dataset corresponding to 1.7 fb- of luminosity, collected at the CDF experiment from proton-antiproton collisions at a center of mass energy f = 1.96 TeV. In the measurement of Ps, we use a dataset corresponding to 1.3 fb-1 of collected luminosity. We measure AF, = (0.071+0.064 ± 0.007) ps-1 using the time-dependent angular analysis. Combining the angular analysis with flavor-tagging, we find that assuming the Standard Model predictions of p/ and AF,, the probability of a deviation as large as the level of the observed data is 33%. We obtain a suite of associated results which are discussed in detail in this dissertation alongside the main results.

Recherche de nouvelle physique dans le canal B0s --> J/psi phi auprès de l'expérience LHCb: Dans le Modèle Standard, la différence de phase apparaissant dans la désintégration B0s --> J/psi phi est prèdite avec une grande précision. Cette observable est une sonde pour mettre en évidence de la Nouvelle Physique car l'oscillation B0s -B0sbar s'effectue via un diagramme en boucles sensible à la nouvelles particules. Nous avons développé une sélection simplifiée pour les données de 2010. Elle évite tous biais sur la distribution en temps propre afin de réduire l'incertitude systématique. De plus, nous contrôlons les performances d'étiquetage pour les événements B0s --> J/psi phi en utilisant les canaux similaires B0d--> J/psi K*0 et B+ --> J/psi K+. Avec les données de 2010, nous obtenons 570 événements de signal avec une luminosité intégré de 36 pb−1, une puissance de d'étiquetage de (2, 2 ± 0, 4)% et une résolution temporelle de 50 fs. Nous avons étudié une sélection alternative, qui maximise la sensibilité à la phase phis en utilisant des coupures biasant le temp propre. Nous avons proposé une méthode pour corriger la déformation de temps propre à partir des données. Nous avons développé un programme d'ajustement pour déterminer la phase phis. Avec les données 2010, la valeur touveé est phis = [−2, 7,−0, 5] rad à 68% de confiance. Ce résultat est compatible avec la prédiction du Modèle Standard.

A pavement management system (PMS) is a tool that supports decision-making specialists to maintain the pavement at an optimal service level. The absence of a PMS could lead to inadequate decision-making, a disorganized road inventory, poor selection in road maintenance techniques and inefficient technical support to justify the execution of maintenance activities. For these reasons, this research proposes the application of a PMS, with the objective of improving and standardizing the processes for evaluating the condition of roads that are under the jurisdiction of low-budget government organizations, guaranteeing the selection of the most appropriate type of maintenance. As a case of study, a 1.1 km local road located in one of the districts of Lima was evaluated, which was divided into 11 sections. With this evaluation, it was obtained that 2,605.43 m2 of the total road is in poor condition, due to the presence of functional failures. For the identification of failures, the pavement condition index (PCI), the international roughness index (IRI) and the present serviceability index (PSI) were used to determine the condition, roughness and serviceability of the pavement. In conclusion, it was identified that the proposed PMS is adequately adapted to the way of working and available budget of a local administration, since if it is applied it would only spend 20.55% of the fund that would be destined to a total reconstruction of road.

La mesure de la phase phis violant la symétrie CP dans les désintégrations Bs -> J/psi phi est une des analyses phares de l'expérience LHCb. Le Modèle Standard prédit cette observable avec une faible incertitude théorique. La présence de Nouvelle Physique notamment dans les diagrammes en boucle de l'oscillation Bs - anti Bs peut significativement modifier la mesure de cette phase. Durant cette thèse, nous avons participé à différents aspects de l'analyse des désintégrations Bs -> J/psi phi. Nous avons proposé une sélection multi-variables, basée sur un algorithme d'arbres de décision boostés. Cette nouvelle sélection augmente le nombre de signal Bs -> J/psi phi de 19% mais nécessite une correction temporelle. Une étude portant sur l'origine des distorsions angulaires caractéristiques des désintégrations Bs -> J/psi phi a aussi été menée. Il s'avère que la principale source de distorsion est la géométrie du détecteur LHCb. Nous avons aussi contribué à un autre élément clef de cette analyse : l'étiquetage de la saveur. Afin de connaître la saveur initiale des mésons B, un algorithme d'étiquetage a été développé. Il exploite les traces chargées provenant du hadron B de côté opposé. Nous avons participé à son optimisation ainsi qu'à son étalonnage en étudiant les désintégrations Bd -> J/psi K*. En utilisant l'intégralité des données enregistrées en 2011, soit 1 fb-1, après optimisation et calibration de cet algorithme, la puissance d'étiquetage dans le canal Bs -> J/psi phi est : (2.29 +- 0.07 +- 0.26) %. La valeur de phis mesurée est alors : \phis = -0.001 +- 0.101 +- 0.027 rad dans les désintégrations Bs -> J/psi phi. L'analyse combinée de Bs -> J/psi phi avec les désintégrations Bs -> J/psi \pi+ \pi- donne la meilleure mesure mondiale de \phis = -0.002 +- 0.083 +- 0.027 rad. Cette mesure est compatible avec le Modèle Standard, mais ses incertitudes laissent possible la présence de Nouvelle Physique.

Introdução: Lectinas são moléculas bioativas presentes em diferentes organismos, que atuam em processos biológicos relevantes, incluindo atividade antimicrobiana, antifúngica, antioxidante, antitumoral, bem como na defesa vegetal. Estudos recentes relatam que a família de Kinases Receptoras de Lectinas do tipo L (LecRK) possui papel no desenvolvimento vegetal e na resposta a estresses ambientais, sendo diferencialmente expressas durante o crescimento vegetal e em resposta a diferentes estímulos. O feijão-caupi (Vigna unguiculata L.) é uma espécie de grande interesse social e econômico para o Brasil, que tem sido alvo de diversos estudos genéticos nas últimas décadas. No entanto, ainda se conhece pouco sobre o papel das LecRKs nesta espécie, especialmente em condições de estresses abióticos e bióticos. Objetivo: Este trabalho teve como objetivo realizar uma prospecção das LecRKs presentes no genoma do feijão-caupi. Material e Métodos: Dessa forma, foi realizada uma busca por sequências sonda no banco de dados Uniprot. A ferramenta HMMER3 foi utilizada para a análise de homologia das sequências contra o genoma de referência do feijão-caupi, depositado no banco de dados Phytozome. A busca por domínios completos foi realizada no Batch CD-search do NCBI, assim como foram preditos o ponto isoelétrico (p.I) e peso molecular (p.M) pelo JVirGel 2.0. Para a verificação da localização subcelular utilizou-se o Cell-Ploc 2.0. Adicionalmente, foi avaliada a presença de peptídeo sinal no SignalP e pontes dissulfeto no DiaNNA. Resultados: Foram obtidas 27 sequências candidatas que continham o domínio LecRK, com p.I variando de 3.93 a 9.05 e p.M. de 26,27 a 76,14, e todas as sequências foram localizadas no núcleo, corroborando ao verificado na literatura. Desse total, dez sequências apresentaram peptídeo sinal e 11 exibiram padrões distintos de pontes dissulfeto, indicando possíveis mudanças na estrutura tridimensional de proteínas da família LecRK. Conclusão: Os resultados contribuíram para a seleção de LecRKs promissoras para avaliação do perfil de expressão in silico em transcriptomas disponíveis no nosso grupo de pesquisa e posterior validação por PCR quantitativa em tempo real (qPCR).

Abstract:
At the beginning of this project, it was clear that R. albus adhered tightly to cellulose and its efficient degradation of this polysaccharide was dependent on micromolar concentrations of phenylacetic acid (PAA) and phenylpropionic acid (PPA). The objectives for our research were: i) to identify how many different kinds of cellulose binding proteins are produced by Ruminococcus albus; ii) to isolate and clone the genes encoding some of these proteins from the same bacterium; iii) to determine where these various proteins were located and; iv) quantify the relative importance of these proteins in affecting the rate and extent to which the bacterium becomes attached to cellulose. BARD support has facilitated a number of breakthroughs relevant to our fundamental understanding of the adhesion process. First, R. albus possesses multiple mechanisms for adhesion to cellulose. The P.I.'s laboratory has discovered a novel cellulose-binding protein (CbpC) that belongs to the Pil-protein family, and in particular, the type 4 fimbrial proteins. We have also obtained genetic and biochemical evidence demonstrating that, in addition to CbpC-mediated adhesion, R. albus also produces a cellulosome-like complex for adhesion. These breakthroughs resulted from the isolation (in Israel and the US) of spontaneously arising mutants of R. albus strains SY3 and 8, which were completely or partially defective in adhesion to cellulose, respectively. While the SY3 mutant strain was incapable of growth with cellulose as the sole carbon source, the strain 8 mutants showed varying abilities to degrade and grow with cellulose. Biochemical and gene cloning experiments have been used in Israel and the US, respectively, to identify what are believed to be key components of a cellulosome. This combination of cellulose adhesion mechanisms has not been identified previously in any bacterium. Second, differential display, reverse transcription polymerase chain reaction (DD RT-PCR) has been developed for use with R. albus. A major limitation to cellulose research has been the intractability of cellulolytic bacteria to genetic manipulation by techniques such as transposon mutagenesis and gene displacement. The P.I.'s successfully developed DD RT- PCR, which expanded the scope of our research beyond the original objectives of the project, and a subset of the transcripts conditionally expressed in response to PAA and PPA have been identified and characterized. Third, proteins immunochemically related to the CbpC protein of R. albus 8 are present in other R. albus strains and F. intestinalis, Western immunoblots have been used to examine additional strains of R. albus, as well as other cellulolytic bacteria of ruminant origin, for production of proteins immunochemically related to the CbpC protein. The results of these experiments showed that R. albus strains SY3, 7 and B199 all possess a protein of ~25 kDa which cross-reacts with polyclonal anti-CbpC antiserum. Several strains of Butyrivibrio fibrisolvens, Ruminococcus flavefaciens strains C- 94 and FD-1, and Fibrobacter succinogenes S85 produced no proteins that cross-react with the same antiserum. Surprisingly though, F. intestinalis strain DR7 does possess a protein(s) of relatively large molecular mass (~200 kDa) that was strongly cross-reactive with the anti- CbpC antiserum. Scientifically, our studies have helped expand the scope of our fundamental understanding of adhesion mechanisms in cellulose-degrading bacteria, and validated the use of RNA-based techniques to examine physiological responses in bacteria that are nor amenable to genetic manipulations. Because efficient fiber hydrolysis by many anaerobic bacteria requires both tight adhesion to substrate and a stable cellulosome, we believe our findings are also the first step in providing the resources needed to achieve our long-term goal of increasing fiber digestibility in animals.
