Abstract:
ABSTRACT The zinc finger transcription factor PacC undergoes two-step proteolytic activation in response to alkaline ambient pH. PalA is a component of the fungal ambient pH signal transduction pathway. Its mammalian homologue AIP1/Alix interacts with the apoptosis-linked protein ALG-2. We show that both PalA and AIP1/Alix recognize a protein-protein binding motif that we denote YPXL/I, where Tyr, Pro, and Leu/Ile are crucial for its interactive properties. Two such motifs flanking the signaling protease cleavage site mediate direct binding of PalA to PacC, required for the first and only pH-regulated cleavage of this transcription factor. PalA can bind the “closed” (i.e., wild-type full-length) conformer of PacC, suggesting that PalA binding constitutes the first stage in the two-step proteolytic cascade, recruiting or facilitating access of the signaling protease, presumably PalB. In addition to recognizing YPXL/I motifs, both PalA and AIP1/Alix interact with the Aspergillus class E Vps protein Vps32 homologue, a member of a protein complex involved in the early steps of the multivesicular body pathway, suggesting that this interaction is an additional feature of proteins of the PalA/AIP1/Alix family.

Abstract:
In June 2009, wilted hop bines were observed in a yard in Marion County, OR. The wilt was associated with a stem rot that occurred ~1 m from the ground near the point where bines are tied together for horticultural purposes. Samples of affected stems were submitted to the Oregon State University Plant Clinic. White hyphae and large, black sclerotia were present on the stems, with a clear delineation between healthy and diseased tissue. The pathogen was identified as Sclerotinia sclerotiorum based on morphological characters. In June 2011, bine wilting was observed on the same farm but in a different hop yard (cv. Nugget) ~10 km from the 2009 occurrence. Affected plants had upward curled leaves with necrotic margins or wilted bines that were severed at the soil line. Wilted bines tended to have smaller diameters than bines with foliar symptoms only. Of 100 plants examined, 75% displayed some foliar symptoms and 66% had at least one bine that was wilted. Yield loss was estimated at 10 to 20% due to bine wilting before cone development. Unlike the 2009 occurrence, wilted bines did not display aerial signs of S. sclerotiorum. Rather, water-soaked lesions covered in white, cottony mycelium were apparent on affected stems 2.5 to 5 cm below the soil surface, some bearing large, irregularly shaped sclerotia. Isolations made onto potato dextrose agar yielded isolates with rapid growth rates and morphological characters consistent with S. sclerotiorum (1). DNA was extracted (2) and pathogen identity was confirmed by PCR amplification and sequencing of the internal transcribed spacer regions from isolates SS001 and SS002 as described before (4). The amplicons were sequenced bidirectionally and consensus sequences were 100% similar to S. sclerotiorum (GenBank No. AAGT01000678.1). Two nucleotide polymorphisms were present that differentiated the sequences from those of 12 S. trifoliorum accessions in GenBank that could be aligned (2). Greenhouse assays utilizing a toothpick inoculation procedure (3) were conducted to fulfill Koch's postulates. Stems of five 4-week-old hop plants of cv. Agate were pierced with a toothpick colonized with S. sclerotiorum. Five control plants were similarly inoculated with toothpicks without the fungus. Inoculated plants developed symptoms similar to those observed in the field within 11 days; four of five plants inoculated with isolate SS001 and two of five plants inoculated with isolate SS002 completely wilted. S. sclerotiorum was reisolated from all inoculated plants but not the control plants. To our knowledge, this is the first report of Sclerotinia wilt on hop in Oregon or the Pacific Northwest (1), where nearly all commercial hop production occurs in the United States. The disease appears to be localized to a limited number of yards, although given the widespread distribution and host range of S. sclerotiorum, it is plausible that the disease may occur in other yards. Recurrent outbreaks and spread of the disease among yards on the affected farm suggests that Sclerotinia wilt has the potential to become a perennial problem on hop and efforts to limit the introduction of S. sclerotiorum into other yards are warranted. Clinical introductionA man in his late 40s presented with severe chest pain and progressive dyspnoea after hitting a tree at high speed during a bike ride in the woods. On admission, the patient appeared agitated, pale and sweaty. Core temperature was 35.5°C, respiratory rate 35/min and blood pressure 90/50 mm Hg with a regular pulse at 110 beats/min. A second species of the previously monotypic snake genus Fimbrios is described from the Truong Son Mountain Range, Vietnam. Fimbrios smithi sp. n. is included in this genus on the basis of the combination of the following characters: rostral, mental and first three to four labials with raised, erected edges; a horizontal ridge of tissues above the rostral; a very large loreal. It is distinguished from F. klossi by having the suture between the internasals longer than that between the prefrontals; two suboculars; 193 ventrals, and 72 unpaired subcaudal scales; total length of at least 440 mm in males (with a tail length of 94 mm, and a tail / total length ratio of 0.214); dorsum greyish brown, with a paler flank area, and pale blotches and stripes in the neck region. Fimbrios smithi sp. n. is the seventh snake species that has been described as new from the Phong Nha—Ke Bang National Park, Quang Binh Province, central Vietnam, in the last decade; it is the forty-fourth colubrid snake species known from that region, which now comprises 60 snake species in general.

Para ello analizamos el patrón exportador de Argentina, Brasil, Chile, Cuba, Perú, Uruguay y Venezuela centrándonos en los productos primarios y las manufacturas basadas en recursos naturales. Distinguimos el comercio con destino a China y el del resto del mundo; para luego estudiar las consecuencias del cambio en el patrón exportador sobre las estructuras económica nacionales. Las principales conclusiones del trabajo serían que, si bien para la mayoría de los países estudiados, se aprecia un proceso de reprimarización de sus exportaciones; solo Brasil y Perú presentan indicios de una reciente dinámica reprimarizadora de su estructura interna, de aún muy corto recorrido.

Abstract:
Ce travail a développé une nouvelle méthode pour l'identification et la quantification des peptides, par l'optimisation de certaines stratégies disponibles appropriées pour le marquage des peptides avec des métaux lanthanide, une séparation par nano-HPLC et détection UV, et suivi par MALDI MS. Tout d'abord, les peptides ont été marqués avec les trois métaux lanthanides différents et un réactif fonctionnel - DOTA. Les résultats montrent que la réaction de transformation en dérivé à l'aide du réactif chélateur DOTA-NHS-ester a été efficace pour des peptides individuels et des mélanges de peptides, vérifiées à partir de la relation m/z obtenue par MALDI MS. L'application optimisée d’un complexe (Cytochrome C digest) a montré des résultats comparables à ceux obtenus avec des peptides modèles. En parallèle, nous avons effectué l’optimisation pour la purification de métalloprotéine dans la bile de poisson, qui est signalée entant que biomarqueurs de contamination métallique de l'environnement. Des procédures différentes (différents moments de centrifugation et différentes températures de traitement thermique) et les agents (DTT, β-mercaptoéthanol et TCEP) réduisant ont été apliqués pour purifier les MT isolées de la bile et du foie des poissons (Oreochromis niloticus). Des analyses spectrophotométriques ont été utilisées pour quantifier les échantillons de MT, et le gel SDS-PAGE a été utilisé pour évaluer qualitativement les différents résultats de la procédure. Chaque procédure a en suíte été évaluée statistiquement, une méhtode des surfaces de réponse a été appliquée. Les MT de la bile semblent être plus adéquate pour la surveillance de l'environnement en ce qui concerne l'exposition récente à des xénobiotiques qui peuvent influer sur l'expression protéomique et metalloproteomique de cette matrice biologique. Une procédure d’exposition à des métaux dans le laboratoire a montré que les métaux étaient significativement importante pour l’évaluation de la contamination à partir de la quantification de MT, selon le traitement de données par une techinique de réseau neural
This work developed a new method for the identification and quantification of peptides, by optimizing some of the available strategies suitable for labeling peptides with lanthanide metals with subsequent separation by nano-HPLC with UV detection, matrix-assisted laser desorption ionization-mass spectrometry (MALDI MS). First, peptides were labeled with the three different lanthanide metals using a functional DOTA-based reagent. The results demonstrate that the derivatization reaction using the chelating reagent DOTA-NHS-ester was effective for single peptides and peptide mixtures, verified from the m/z relation obtained by MALDI MS. The application of the optimized method in a more complex matrix (Cytochrome C digest) showed results comparable to those obtained with model peptides. In parallel, environmental analyses were conducted, by performing the standardization of metalloprotein purification in fish bile, since this matrix has been reported as a biomarker for environmental metal contamination. Different procedures (varying centrifugation times and heat-treatment temperatures) and reducing agents (DTT, β-mercaptoethanol and TCEP) were applied to purify MT isolated from fish (Oreochromis niloticus) bile and liver. Spectrophotometrical analyses were used to quantify the resulting MT samples, and SDS-PAGE gels were used to qualitatively assess the different procedure results. Each procedure was then statistically evaluated. A response surface methodology was applied for bile samples, in order to further evaluate the responses for this matrix. In an environmental context, biliary MT was lower than liver MT, and, bile MT seems to be more adequate in environmental monitoring scopes regarding recent exposure to xenobiotics that may affect the proteomic and metalloproteomic expression of this biological matrix. A procedure for exposure to metals in the laboratory showed that some metals are significantly important for the assessment of contamination from the quantification of MT, according to the data processing by atifical neural network (ANN)

Abstract:
Enterohepatic Helicobacter species (EHS) are emerging infectious disease agents. Infection of the enterohepatobiliary tract of several mammals by this group of bacteria results in various pathological disorders. The availability of the Helicobacter hepaticus sequenced and annotated genome, allowed molecular characterisation of the responses of H. hepaticus to host factors such as bile. The adaptation/responses of the bacterium to bovine, porcine and human bile were investigated using proteomics and transcriptomics. Ninety-one different proteins were identified in the responses of H. hepaticus response to the three types of bile. These proteins participate in several key cellular processes including DNA replication; protein transcription, translation and folding; oxidative stress response; motility; virulence; and metabolism. In particular, the bacteria deployed several strategies such as inhibition of the TCA cycle and the electron transport chain as well as iron sequestration to ensure control of the levels of hydroxyl radicals. The results of this study revealed also the modulation by bile of the expression of H. hepaticus genes involved in response to oxidative stress and virulence. The responses of human HEp-2 and Huh7-derived cell-lines to H. hepaticus and Helicobacter bilis, respectively, were investigated employing proteomics and transcriptomics. One-hundred and twenty different proteins were differentially expressed in the responses of the human cells to the presence of Helicobacter spp. in the cell cultures. These proteins are involved in regulation of cell proliferation and structure; metabolism; protein transcription, translation and modification; stress response; and tumour induction. For example, in co-cultures of Huh7-derived cells and H. bilis, the activation of several mitochondrial and endoplasmic reticulum stress-related proteins and the dysregulation of several apoptosis effectors were suggested as mechanisms that could result in the death of the liver cells. Importantly, the differential expression of several tumour-related proteins by the Huh7 cells supported a possible role for Helicobacter spp. in liver cancer.

Dans cette thèse, nous traitons de l’apport des théories du courant de pensée de Palo Alto pour aborder la transformation des situations de doubles contraintes au sein des organisations de travail, afin de faciliter le développement de l’apprentissage organisationnel. Après une présentation des concepts d’apprentissage organisationnel et de double contrainte, notre démarche de recherche prend place dans une épistémologie interprétativiste procédant par recherche-action. Nous nous appuyons sur trois études de cas d’accompagnement, individuel et collectif, de responsables confrontés à des difficultés de management. Nous mettons en évidence de quelle manière la théorie de la double contrainte constitue une grille d’intelligibilité des conduites d’évitement. Les envisageant comme seules possibilités de se comporter dans une situation où toutes les autres possibilités d’agir sont bloquées, nous expliquons l’intérêt d’un changement de paradigme de compréhension de leur formation pour renouveler les perspectives d’intervention. L’acceptation des conduites d’évitement comme émergeant d’une situation de double contrainte permet de les aborder en tant que dysfonctionnement organisationnel plutôt que comme la conséquence d’une carence individuelle. Les perceptions de doubles contraintes apparaissent lorsqu’une impossibilité de métacommuniquer est ressentie, au sujet d’un paradoxe exigeant un choix impossible, dans un contexte professionnel dont il n’est pas envisageable de s’échapper.Notre principal apport est de montrer, au travers d’une revue de littérature et des trois études de cas, les forces et les limites de l’approche interactionnelle issue du courant de Palo Alto, qui, même si elle est connue dans le monde de la gestion, reste largement inappliquée dans les organisations. Nous positionnons le processus de métacommunication par un tiers intervenant en le mettant au service de changements. Nous précisons le rôle qu’il peut avoir pour aider à passer d’un cercle vicieux à un cercle vertueux, dans une situation managériale, et l’effet de levier qu’il joue dans la transformation d’une dynamique d’apprentissage organisationnel défensif en une dynamique menant au progrès
In this thesis, we discuss the contribution of current theories of thought in Palo Alto to tackle the transformation of situations of double bind within working organizations in order to improve the organizational learning development. After a presentation of the concepts of organizational learning and double bind our research process takes place in an interpretivist epistemology conducting by action research. We rely on three case studies accompanying individual and group leaders facing with difficulties in management.We underline how the theory of the double bind is a grid of intelligibility avoidance behavior. Considering them only as possibilities to behave in a situation where all other possibilities to act are blocked, we explain the interest of a paradigm shift in understanding of their training opportunities in order to renew intervention prospect.Acceptance of avoidance behavior as emerging from a double bind situation can address them as organizational dysfunction rather than as a consequence of individual failure. Perceptions of double bind appear when unable metacommunication is felt, about a paradox requiring an impossible choice, in a professional context inconceivable to escape from.Our main contribution is to show, through a literature review and three case studies, the strengths and limitations of the interactional approach of Palo Alto, which, although it is known in the world management remains largely unimplemented in organizations. We position the metacommunication process by a third party involved in the service changes. We point out the role he may have to help them move from a vicious to a virtuous circle, in a managerial situation, and leverage in the transformation of a dynamic organizational learning defensive into a dynamic leading to progress

El Paleolítico medio abarca unos 300.000 años (MIS 10 a MIS 3). Durante este tiempo los comportamientos humanos se han visto afectados por diferentes factores que han producido un mosaico de respuestas que han marcado la diversidad cultural de las poblaciones Neandertales. La variabilidad de los comportamientos técnicos líticos durante este periodo es una de las temáticas que ha creado más debate científico en la Arqueología prehistórica. A través del estudio de los comportamientos técnicos líticos y en extensión de la puesta en común de sus resultados con los obtenidos de las disciplinas que estudian el resto de las evidencias arqueológicas, se pretende aportar datos sobre la variabilidad de los conjuntos líticos. Para ello se han elegido los conjuntos líticos de los yacimientos de Payre (MIS 5e), el Abric Romaní (MIS 3) y de la Grotte Tournal (final MIS 3). El estudio de de la tecnología lítica utilizada en cada uno de ellos, el análisis comparativo de los resultados obtenidos y su contextualización en el panorama del Paleolítico medio del suroeste de Europa permitirá aportar datos sobre la variabilidad de los conjuntos líticos y las posibles causas que los han producido, intentando ver que relación existe entre la composición del registro lítico, los actividades de subsistencia llevadas a cabo y el tipo de ocupaciones.

The Middle Palaeolithic covers around 300,000 years (MIS 10 to MIS 3). During this time, human behavior has been affected by various factors leading to a mosaic of responses which characterize the cultural diversity of Neanderthal populations. Through the study of lithic technical behavior and the comparison of these results with those obtained from other disciplines that study archaeological evidence, the intention here is to provide data on the variability of lithic assemblages. For that purpose, the lithic assemblages from Payre, Abric Romaní and Grotte Tournal were selected. The study of lithic technology at each site as well as a comparative analysis of the results and their contextualization in the Middle Palaeolithic panorama of south-western Europe, will provide data on the variability of lithic assemblages and the possible and will add to the understanding of the relationship between the composition of the lithic record, subsistence activities, and the type of occupations.

Le Paléolithique Moyen à une durée d'environ 300.000 ans (MIS 10 à MIS 3). Pendant cette période, les comportements humains ont été affectés par divers facteurs qui ont produit une mosaïque de réponses et généré la diversité culturelle des populations néandertaliennes. A travers l'étude des comportements techniques lithiques, et la comparaison des résultats avec ceux obtenus par les disciplines qui étudient les autres évidences archéologiques, l'objectif est d'apporter de nouvelles données sur la variabilité des assemblages lithiques. Pour cela, les assemblages lithiques des gisements suivants ont été sélectionnés: Payre, l'Abric Romani et la Grotte Tournal. L'étude de la technologie lithique, l'analyse comparative des résultats et leur place dans le panorama du Paléolithique Moyen du sud-ouest de l'Europe fournira des données sur la variabilité des assemblages lithiques et ses causes.

Abstract:
This chapter considers the question of whether daily news over the past century has gone along with the modern trend of shorter news. When the occupation of journalist first emerged in the nineteenth century, realist news was mainly short, and everything in the modern world has seemed to go only faster for more than a century. First radio picked up the pace and then television followed, requiring shorter attention spans. Along came faxes, then electronic mail, and now video messaging. MTV made images move faster, television commercials got shorter, and online ads shrank to a few seconds. Critics call it sound-bite society or McDonaldization, reducing information to nuggets. However, studies show that news has been getting longer, moving away from brief realist descriptions of stand-alone events and aligning with modern impulses toward big-picture explanation. The trend occurred across legacy news media: newspaper reporters writing longer, television reporters speaking more, and even reporters on public radio, the home of extended news, talking more in longer stories.

With more than 5 million college students taking at least one online or blended college course, (Seaman et al, 2018) and 5 percent of K-12 students taking an online or blended course (The Evergreen Education Group), research regarding student success in blended and online classes has begun to emerge and shed light on potential trouble spots. This chapter uses an action research model to further examine binge studying in two, beginning university Korean classes in relation to success on exams, overall course grades, language recall after a four-week break, and continued enrollment in Korean. Findings indicate a correlation between overall page views and course grade, between overall page views in 101 to retention in 102 and a correlation was also evident between binge studying and 102 assessment. A negative correlation was seen between binge studying and 101 final exam scores. The strongest correlation appeared to be between non-binge studiers, or those which dispersed their study throughout the semester, and language retention in Korean 102.

Abstract:
The narrative setting for chapter 2 is the Veterans Administration Hospital in Palo Alto, California, in the 1950s, where Bateson lead the double-bind research group investigating paradox and disorder in family relations. The chapter traces the early development of the double-bind theory of schizophrenia and its source in Russellian logical paradox. It discusses the double bind as a resonant and empirically rich example of similar constructs that distilled the modern predicament as an impossible dilemma. Other examples include Joseph Heller's catch-22, Reinhold Niebuhr's reformulation of original sin and his Serenity Prayer; Albert Camus's concept of the absurd and his novel The Stranger; and constructions in Kurt Vonnegut, Jr.'s Slaughterhouse Five. Double-bind equivalents as reactions to the atom bomb are described in works such as Joanna Greenberg's I Never Promised You a Rose Garden, Allen Ginsberg's Howl, and the film Rebel Without a Cause. Meanwhile, social critics such as Lewis Mumford used psychiatric and systems theory language and paradoxical constructions to talk about the failure of "pragmatic liberalism," the arms race, and policies of nuclear deterrence.
