Sequencing and phylogenetic analysis was used to characterise a number of Scottish IPNV isolates. The majority of isolates from Scotland were genetically closely related to the Sp strain of IPNV. There appears to be a link between the IPN status of a farm and the presence of IPNV in the environment but this is short lived as IPNV does not typically persist in natural reservoirs at sufficiently high levels to allow re-infection to occur. The persistence of infectious pancreatic necrosis virus (IPNV) in Atlantic salmon was examined with particular emphasis on the association of IPNV with leucocytes. Atlantic salmon were infected with IPNV via the water and via the feed. Despite relatively low mortalities, a high prevalence of IPNV was demonstrated particularly when the virus was introduced via the water. No virus-specific pathology or reduction in the growth performance of infected populations was evident. A rainbow trout (Slamo gairdneri Richardson) population of IPN virus carriers was studied over a one-year period using both homogenisatian and co-cultivation for virus isolation. The percentage of virus-yielding fish was high between March and June but then declined. Three closely related isolates belonging to the A��� serotype of infectious pancreatic necrosis virus (IPNV) were selected for comparison, to provide insight into the nature of variation in the virulence of IPN viruses. Brook trout fry (Salvelinus fontinalis) were experimentally infected with the three isolates by immersion. Cumulative mortalities over a 62 day period for the three isolates were 67%, 78%, and 93%. The A segment of infectious pancreatic necrosis virus (IPNV) is expressed as a polyprotein encoding three primary gene products, VP2, NS and VP3, from a large open reading frame. The nucleotide sequence for the A segment of the Sp isolate of IPNV was determined. The NS protein is the putative autocatalytic proteinase responsible for the cleavage of the polyprotein. The functional boundaries of the NS proteinase were mapped by plasmid deletion analysis and examined in an La vitro, translation system. The NS proteolytic activity was determined to lie within the EcoRI and Nsil restriction sites.

碩士<br>輔仁大學<br>生物學研究所<br>75<br>IPN 病毒類,HB-1文蛤病毒分離株在高濃度下經4 個月繼代培養,發現病毒效價有波 動情形,其波動度高於以低濃度感染之病毒,因其教價會降低,且皆低於以低濃度接 種之繼代病毒,顯示有病毒干擾現象。取經40代高濃度繼代之高毒與正常低濃度感染 之病毒,於同時間內分別純化,取得多DI病毒及多正常病毒之收集管。將兩者之收隻 管加以分析,多DI管感染效價最高在第7 收集管,干擾效價最高在第9 收集管,正常 病毒之感染效價最高在第7 收集管,且其所有收集管均測不到干擾現象,觀測其蛋白 質電泳分析圖譜,發現兩者之α、β蛋白質群有差異,γ蛋白質群則其相同分子量之 蛋白質,多成熟DI管之α蛋白質分子量分別為94Kd ,105Kd,成熟正常病毒為94Kd, 多成熟DI管之β蛋白質分子量分別為50Kd ,51Kd,成熟正常病毒為52.3Kd ,推測分 子量50Kd及105Kd 者為成熟DI特有之蛋白質,應和干擾現象有關。至於以10%聚烯醯 胺分析基因

The characteristics of an inhibitor of infectious pancreatic necrosis virus (IPNV) found in normal rainbow trout serum (RTS) were studied. The serum inhibitor had a molecular weight of approximately 150 kDa and was dependent on divalent cations, either Ca����� or Mg�����. It was stable at temperatures up to 50��C and at a pH range between 4-10. The inhibitor directly inactivated the virus and the inhibition level was dependent on cell densities and on the time at which virus was exposed to RTS. The level of virus inhibition by RTS was altered by the cell line in which virus was produced. IPNV

博士<br>國立臺灣大學<br>動物學系<br>81<br>SB細胞株源自青魚鰾組織,在初級細胞培養時,即表現出喪失停泊依賴性 ,喪失接觸抑制性,血清依賴性降低及高群落形成率等腫瘤細胞特性,以 倒立顯微鏡觀察,SB細胞以纖維母細胞為主,其間夾雜許多巨大細胞。以 Acridine orange 染色,並以螢光顯微鏡觀察,可觀察到在這些巨大細胞 細胞質的位置有黃綠色包涵體,應為雙股核酸病毒的包涵體。在掃描式電 子顯微鏡下,SB細胞表現鋪展不良,富於長微絨毛等強轉型細胞特性。SB 細胞的培養液接種於CCT及TO-2細胞,導致嚴重細胞病變。這些結果顯示 ,SB細胞源自腫瘤組織,並有雙股核醣核酸病毒的持續性感染。以穿透式 電子顯微鏡觀察此尾青魚之組織,在前腎可觀察到細胞質空泡化,細胞核 膜增生等病變,並可觀察到二十面體病毒的存在。 SB細胞亦有類似細胞 病變,但病變的程度嚴重並且在細胞質可觀察到大量病毒及病毒質的戌b ,病毒顆粒之直徑約 55-70nm。分析病毒來源
