Nitrous oxide (N2O) is an important greenhouse gas that is also responsible for stratospheric ozone depletion [1, 2]. Human activity is the main source of N2O due to the use of fertilisers in agriculture. Nitrous oxide reductase (N2OR) is the only enzyme to destroy N2O as part of a biological process termed denitrification. The effect of nitrite stress induced in Paracoccus denitrificans PD1222 was examined using additions of sodium nitrite to an aerobic bacterial culture. Nitrite generates a strong stress response in P. denitrificans, causing growth inhibition. This is dependent on both the concentration of nitrite present and the pH. The pH dependent effect of nitrite growth inhibition is likely a result of nitrite and free nitrous acid (FNA; pKa = 3.16) and subsequent reactive nitrogen oxides, generated from the intracellular passage of FNA into P. denitrificans. A study of the electron transfers associated with the cytochrome c peroxidase of <I>Paracoccus denitrificans</I> has been made. The peroxidase is similar to the well-studied enzyme from <I>Pseudomonas aeruginosa</I>, although significant differences do exist. Like the <I>Pseudomonas</I> enzyme, the <I>Paracoccus</I> peroxidase contains two haem c groups, one high potential (E<SUB>m</SUB>=+226mV) and one low potential (E<SUB>m</SUB>≈-100mV). Ziel der vorliegenden Arbeit war die Charakterisierung des Wachstumsverhaltens von Mikroorganismen auf Alkanen am Beispiel des Bodenbakteriums Rhodococcus opacus 1CP. Unter Einsatz kalorimetrischer Methoden konnte das Wachstumsverhalten von 1CP auf Glucose und Alkanen mit sehr guter Reproduzierbarkeit aufgezeichnet werden. In Abhängigkeit der Substrateigenschaften wurden Änderungen der Wachstumskinetik registriert, welche auf die Aggregation der Zellkultur zurückgeführt werden konnten. O fungo termodimórfico Paracoccidioides brasiliensis (P. brasiliensis) é o agente causador da paracoccidioidomicose (PCM), doença endêmica na América Latina. A partir do reconhecimento de componentes fúngicos, macrófagos são ativados e adquirem propriedades que favorecem a eliminação do patógeno. Essas células produzem citocinas responsáveis pelo direcionamento da resposta adaptativa, sendo que o perfil Th1 é associado a proteção. Paracoccina (PCN) é um constituinte de Paracoccidioides brasiliensis, um patógeno humano que causa a paracoccidioidomicose, micose sistêmica mais prevalente na América Latina. A PCN é uma proteína de função dual, com domínios de atividade lectínica e de N-acetilglicosaminidase.
