Este artigo, no qual analisamos a novela de Hilda Hilst Com meus olhos de cão (1986), tem como objetivo principal apontar os aspectos paratópicos na obra selecionada. Paratopia é um conceito formulado por Dominique Maingueneau e trata, basicamente, do não-lugar criado e ocupado por algumas literaturas e seus autores. Para isso, estudaremos a linguagem literária desenvolvida pela autora na obra, como também os personagens, o espaço e o tempo da narrativa, os quais acabam por deslocá-la do lugar mais comum do cânone literário.Palavras-chave: Paratopia. AbstractThis article, in which we analyze Hilda Hilst’s novella Com meus olhos de cão [With my eyes of Dog] (1986), has as its main objective to point out the paratopyc aspects in the selected work. ‘Paratopy’ is a concept formulated by Dominique Maingueneau and it basically deals with the non-place created and occupied by some literatures and their authors. Abstract:
The research we will present aims to further develop a discussion started in the master’s degree research entitled Publishing mediation in scientific communication: a study of two human sciences journals, focusing on a fundamental issue of the current times: the constitution of authorship in scientific communication. In order to investigate that issue, we aim to examine the defining conditions of authorship in the production of scientific papers in different areas of knowledge. Therefore, the corpus is composed by four important journals nationwide, Geousp: espaço e tempo, from the Postgraduate Program in Geography of the FFLCH-USP, Cerâmica Industrial, from the Brazilian Ceramic Association, Cadernos Brasileiros de Terapia Ocupacional, from the Occupational Therapy Department of UFSCar, and Revista do Instituto de Estudos Brasileiros (Rieb), from the Brazilian Studies Institute of USP. Having the French discourse analysis as theoretical framework, we take into account the relationship between the author and other co-enunciators in the editorial genetic rites (Salgado, 2011), also considering the place of the author as a creative paratopy (Maingueneau, 2014). At the event, we will discuss more precisely the current stage of the research, focusing on the methodology of data collection, in which we used interviews with different professionals involved in the publishing of the mentioned journals.

ABSTRACT Combinatorial cloning and expression library analysis were used to determine the expressed human antibody repertoire specific for the capsular polysaccharide (PS) of Streptococcus pneumoniae serotype 6B. Sequence analysis of 55 6B-specific antibody Fab fragments isolated from six vaccinated donors reveal that different individuals used a variety of heavy and light chain germ line variable (V) region genes to form pneumococcal capsular PS (PPS) 6B-specific paratopes. Within each donor, however, the response was more restricted, with five of the six donors using at most one or two gene pairs to form combining sites. Analysis also indicated that although the response in each donor was oligoclonal in terms of variable gene usage, the combination of extensive somatic hypermutation, deletion of germ line-encoded residues, insertion of non-germ line-encoded residues, and intraclonal isotype switching generated a surprising degree of paratope diversity within the individuals analyzed. In contrast to previously studied PS-specific responses, we find that the PPS 6B repertoire makes use of a diverse collection of heavy-chain and light-chain V region gene products to form specific paratopes, with no apparent tendency for conservation of immunoglobulin gene usage between individuals.

Abstract:
Crovalimab (RO7112689) is a novel anti-human C5 antibody engineered with Sequential Monoclonal Antibody Recycling Technology (SMART Ig)(Fukuzawa et al., Sci Rep. (2017) 7(1):1080), resulting in significant half-life extension and enabling infrequent SC dosing using small volumes (1mL - 4mL) in C5 mediated diseases. To establish pharmacokinetics (PK), pharmacodynamics (PD), safety, efficacy, and optimal dose of crovalimab, we conducted a four-part adaptive clinical trial (Figure 1) in healthy volunteers (Part 1), treatment naïve PNH patients (Part 2, Part 4) and in eculizumab pre-treated patients (Part 3, Part 4). The conventional PK metrics to determine optimal target inhibition is drug concentration, however this has some limitations when considering an abundant fluctuating soluble target such as C5. Therefore, we proposed to characterize C5 inhibition by quantifying the level of crovalimab free paratopes (i.e. the concentration of free crovalimab antigen-binding sites not bound to C5) and to illustrate how it can be used to estimate the available binding capacity reserve of crovalimab. Total crovalimab concentration, free C5 and total C5 protein levels (measuring free C5, C5 bound to one crovalimab Fab arm, or two C5s bound to the two crovalimab Fab arms) were measured using validated assays. In assuming steady state conditions at every measurement time for the binding of crovalimab with C5 (due to the rapid binding of C5 with crovalimab), a mathematical model based on law mass action principle was used to describe the equilibrium between free C5, free crovalimab and crovalimab bound to one or two C5 molecules. Available crovalimab free paratope was estimated from the model in using total C5 and total crovalimab concentrations collected in the COMPOSER trial. Paratope level was expressed in C5-binding ability concentration equivalent (i.e. how much additional C5 could be bound). Relationships between total concentration time course, free C5 and available free epitopes were evaluated using graphical analysis. To complement these analyses and using the mathematical model, a sensitivity analysis was performed to identify key antibody properties driving the level of available free crovalimab paratopes. In COMPOSER Part 2, the longitudinal time course of crovalimab free paratope concentration (Figure 2) shows that 170mg SC weekly provided a median reserve of free paratopes that allows binding around 120µg/mL of additional C5 corresponding approximatively to two times the baseline C5 levels observed in Part 2 (i.e. 129µg/mL). Therefore, if the level of C5 in the circulation doubled from baseline, e.g. during infection, there would be a sufficient binding capacity to block the activity of these new C5 molecules at least for 75% of the patients (Figure 2). In COMPOSER Part 3, the observed median reserve of free paratopes was lower during the first 60 days after crovalimab initiation (Figure 2). This is driven by the remaining presence of eculizumab and the binding of both crovalimab and eculizumab to C5. Eculizumab and crovalimab bind different C5 epitopes and switching patients from eculizumab to crovalimab induces formation of drug target drug complexes (DTDC). DTDCs clearance was estimated to be ten times faster than crovalimab-C5 complexes clearance resulting in a drop of free paratope level during the first 60 days. Washout from eculizumab is not feasible in PNH patients. To increase the availability of crovalimab free paratopes after switching, crovalimab dose and regimen was optimized and is currently tested in COMPOSER Part 4 (Figure 1).The sensitivity analysis demonstrated that the main driver of free paratopes availability were the recycling efficiency of free crovalimab after endocytosis and the clearance of crovalimab while the affinity of crovalimab to C5 had little effect. Free paratope concentration enables characterization of available binding reserve of crovalimab. The capacity to adequately control C5 increase due to the SMART-Ig engineering is expected to result in a better control of breakthrough hemolysis in PNH patients compared with an antibody without this technology. For soluble targets, this approach provides more stringent criteria than antibody concentration as it defines the capacity of the drug to bind free target at any time. Abstract:
A primary goal of HIV-1 vaccine development is the consistent elicitation of protective, neutralizing antibodies. While highly similar neutralizing antibodies (nAbs) have been isolated from multiple HIV-infected individuals, it is unclear whether vaccination can consistently elicit highly similar nAbs in genetically diverse primates. Here, we show in three outbred rhesus macaques that immunization with Env elicits a genotypically and phenotypically conserved nAb response. From these vaccinated macaques, we isolated four antibody lineages that had commonalities in immunoglobulin variable, diversity, and joining gene segment usage. Atomic-level structures of the antigen binding fragments of the two most similar antibodies showed nearly identical paratopes. The Env binding modes of each of the four vaccine-induced nAbs were distinct from previously known monoclonal HIV-1 neutralizing antibodies, but were nearly identical to each other. The similarities of these antibodies show that the immune system in outbred primates can respond to HIV-1 Env vaccination with a similar structural and genotypic solution for recognizing a particular neutralizing epitope. These results support rational vaccine design for HIV-1 that aims to reproducibly elicit, in genetically diverse primates, nAbs with specific paratope structures capable of binding conserved epitopes.

Abstract:
Proteases are frequent pharmacological targets, and their inhibitors are valuable drugs in multiple pathologies. The catalytic mechanism and the active-site fold, however, are largely conserved among the protease classes, making the development of the selective inhibitors exceedingly challenging. In our departure from the conventional strategies, we reviewed the structure of known camelid inhibitory antibodies, which block enzyme activities via their unusually long, convex-shaped paratopes. We synthesized the human Fab antibody library (over 1.25 × 109 individual variants) that carried the extended, 23- to 27-residue, complementarity-determining region (CDR)–H3 segments. As a proof of principle, we used the catalytic domain of matrix metalloproteinase-14 (MMP-14), a promalignant protease and a drug target in cancer, as bait. In our screens, we identified 20 binders, of which 14 performed as potent and selective inhibitors of MMP-14 rather than as broad-specificity antagonists. Specifically, Fab 3A2 bound to MMP-14 in the vicinity of the active pocket with a high 4.8 nM affinity and was similarly efficient (9.7 nM) in inhibiting the protease cleavage activity. We suggest that the convex paratope antibody libraries described here could be readily generalized to facilitate the design of the antibody inhibitors to many additional enzymes.

A hybridoma antibody (11E7-1) was isolated from a myeloma fusion with nu/nu BALB/c immunized against the T15 idiotype. This IgM antibody exhibited a dual specificity, binding both to PC and to anti-PC antibodies from two idiotype families. Binding to PC and anti-PC antibodies are completely inhibited by PC analogs. Furthermore, the hybridoma antibody binds to itself. Self-binding is also inhibited by PC analogs. From these data, we suggest that 11E7-1 hybridoma antibody has a PC-specific paratope site, and at same time expresses the internal PC antigen idiotope. The term autobody is proposed to signify its self-binding and potential role in autoimmunity. Autobodies may have a unique role in the network of immune system. Furthermore, it may be a model for designing idiotype vaccines.

Background: B-cell epitope prediction is a computational approach originally developed to support the design of peptide-based vaccines for inducing protective antibody-mediated immunity, as exemplified by neutralization of biological activity (e.g., pathogen infectivity). Said approach is benchmarked against experimentally obtained data on paratope-epitope binding; but such data are curated primarily on the basis of immune-complex structure, obscuring the role of antigen conformational disorder in the underlying immune recognition process. Objective: This work aimed to critically analyze the curation of epitope-paratope binding data that are relevant to B-cell epitope prediction for peptide-based vaccine design. Methods: Database records on neutralizing monoclonal antipeptide antibody immune-complex structure were retrieved from the Immune Epitope Database (IEDB) and analyzed in relation to other data from both IEDB and external sources including the Protein Data Bank (PDB) and published literature, with special attention to data on conformational disorder among paratope-bound and unbound peptidic antigens. Results: Data analysis revealed key examples of antipeptide antibodies that recognize conformationally disordered B-cell epitopes and thereby neutralize the biological activity of cognate targets (e.g., proteins and pathogens), with inconsistency noted in the mapping of some epitopes due to reliance on immune-complex structural details, which vary even among experiments utilizing the same paratope-epitope combination (e.g., with the epitope forming part of a peptide or a protein). Conclusion: The results suggest an alternative approach to curating paratope-epitope binding data based on neutralization of biological activity by polyclonal antipeptide antibodies, with reference to immunogenic peptide sequences and their conformational disorder in unbound antigen structures.

It also aims to evaluate the role of paratopia in the testimony and documental proxemics characterization. It is important subject, since the paratopia related to literary discourse as well as the discourse of blackness are issues still little explored academically, serving as general category for analysis in the literary context and of the black condition in Brazilian history. Our hypothesis is that the ideal of blackness presented in literary discourse embodied in testimony and documentary because it is constructed and legitimized from paratopies and atopies displacement intertwined with discursive historicity. We also assume that the testimony and documentary discourse presents this displacement movement to legitimize a discourse or an ideal that constitutes a kind of "stylistic or semiotic design" of testimonial speech, which we call discursive proxemics . In order to achieve our goals, we have mainly used as theoretical support the studies by Discourse Analysis from Maingueneau (2010, 2008a, 2008b, 2008c, 2007, 2006, 2001, 2000, 1996, 1992 and 1983), Charraudeau (2004, 2005 and 2012) and Foucault (1992, 2004, 2005a and 2005b). We have also used concepts of Literary Theory, Philosophy, Sociology and Anthropology extracted from Lucáks (2000), Bakhtin (2003), Benjamin (1985), De Marco (2004), Seligman -Silva (2006) and Bosi (1995), Fernandes (2006, 2008 and 2005-6), Munanga (2004, 2006, 2007 and 2009), Hall (2013), Fanon (2008), and Bhabha (1998). All of them have been taken as elements for understanding the discursive testimonial aspect and to support our vision of the socio-historical conditions of production. We have proved our hypothesis that the documentary evidence is given of paratopic shape and we have also extended the analysis of this category to other possibilities we call paratopies: authorship, aphasic, investigative, documentary, testimonial and testimonial-documentary<br>Este trabalho insere-se nos princípios teórico-metodológicos da Análise do Discurso de linha Francesa com o objetivo de analisar e compreender as estratégias discursivas presentes em Vencidos e Degenerados de José do Nascimento Moraes que compõem o discurso da negritude que emana das cenas de enunciação e avaliar o papel da paratopia, numa possível definição ou caracterização proxêmica testemunho-documental. Trata-se de tema relevante, uma vez que a paratopia relacionada ao discurso literário, bem como o discurso da negritude, são temas ainda pouco explorados academicamente, servindo como categorias para análises gerais no âmbito literário e para análise da condição do negro na História do Brasil. Partimos da hipótese de que o ideal de negritude que se apresenta no discurso literário consubstancia-se em testemunho-documental porque se constrói e é legitimado, a partir do deslocamento de paratopias e atopias entrelaçadas com a historicidade discursiva, supomos que o discurso testemunho-documental se apresenta nesse movimento de deslocamento para legitimação de um discurso ou de um ideal, que configura uma espécie de desenho estilístico ou semiótico do discurso testemunhal, que chamaremos de proxêmica discursiva. Abstract:
La coprésence de langues dans le roman antillais contemporain. Le roman francophone est souvent considéré comme le lieu d’enjeux stratégiques concernant la coprésence d’usages de langue(s). À cet égard, les Antilles présentent une situation tout à fait originale dans laquelle une “cacophonie” pourrait être envisagée comme un moyen d’expression des différentes tensions (narratives, énonciatives ou linguistiques) qui habitent le texte, avec toutes les conséquences que cela peut entraîner pour les lecteurs potentiels. Il s’agit pour les rédacteurs de L’Éloge de la créolité d’aller à l’encontre des attentes de clarté du lecteur pour préserver sans altération aucune une identité multiple. Cependant, d’autres auteurs antillais francophones tels que Simone Schwarz-Bart, Maryse Condé et Daniel Maximin, ne partagent pas le point de vue des signataires de l’Éloge. Bien que leur écriture soit marquée par une certaine présence créole, ils considèrent que l’identité linguistique antillaise ne peut pas se réduire à une confrontation du créole et du français. Pour ces auteurs, il ne s’agit pas de reconquérir le français en le créolisant. Par conséquent, ce travail de thèse consistera à analyser la structure linguistique du roman antillais francophone en prenant autant en compte les différents partis pris des auteurs ainsi que la réception et les transpositions tentées par les traducteurs. Cette étude propose une mise en perspective de l’écriture en coprésence de langues en mettant en relation les œuvres des auteurs antillais contemporains avec des tentatives antérieures de superposition de langues et des écritures créolisées issues d’autres sphères linguistiques. Ces recherches permettront de saisir les influences et la portée de l’écriture en coprésence de langues des romanciers antillais contemporains<br>The Pluri-language Writing in the Contemporary West Indian NovelThe francophone novel is often regarded as field of strategic issues as to the pluri-language writing. In this respect, West Indies offer a very peculiar situation in which “cacophony” could be considered as a way for various strains (narrative, enunciative and linguistic) to express themselves within the textual frames, with many consequences for the potential readers. For the writers of In Praise of Creoleness, it means deceiving the reader’s expectations of clarity to preserve unaltered a multiple identity.Nevertheless, other West Indian francophone writers such as Simone Schwarz-Bart, Maryse Condé and Daniel Maximin, do not share this point of view. Although their writing is marked by a certain Creole presence, they assert that West Indian linguistic identity can not be summarised in the confrontation of Creole and French. According to them, the point is not to reconquer French through creolization.This thesis thus aims to analyze the linguistic structure of West Indian francophone novel with respect both to its different writers’ stances, its reception and the transpositions tempted by the translators.This study proposes a contextualization of the plurilingual texts through a confrontation of the works of the contemporary West Indian authors with the previous overlapping languages attempts and creolized writings stemming of the other linguistic spheres.This research will allow to seize the influences and impacts of the pluri-language writing of the contemporary West Indian novelists

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior<br>The aim of this study is to analyze, from the theoretical perspective of discourse analysis, especially from the theoretical notions proposed by Dominique Maingueneau in Literary Discourse (2006), aspects of the discursive practices of Brazilian singer and composer Raul Seixas, so as to investigate the way in which paratopy is constituted in his work. As understood by Maingueneau, paratopy is a characteristic of the self-constituting discourses (religious, philosophical, scientific and literary) proposed as discourses of origin and validated by an enunciation scene that authorizes them. For the author, paratopy comes from a paradoxical locality that does not concern the lack of a specific "place" for the discourse, but refers itself to the difficult negotiations between the place and the non-place, a process that emerges from the very impossibility of this discourse to stabilize itself. Such a process falls upon three dimensions Maingueneau classifies as paratopic engines: the scenography, the language code, and the ethos. In order to demonstrate the way in which Raul Seixas paratopic place is constituted, this study comprises a corpus of discursive and intersemiotic practices of the composer: i) songs; ii) album covers; iii) a presentation video at a festival; iv) and a comic-manifesto which was distributed in some of his shows. The chosen corpus and analysis carried out were outlined based on the paratopic engines, which we suppose that work as drivers to the creation of the work under study. Thus, in this perspective, they build their paratopy, which, in the specific case of Raul Seixas, emerges from the requirement and/or the announcement of a transcendent and metaphysical time and space beyond the enunciation itself.<br>Nesta dissertação, objetivamos analisar, sob a perspectiva teórica da Análise do Discurso, sobretudo a partir das noções teóricas propostas por Dominique Maingueneau em Discurso Literário (2006), aspectos das práticas discursivas do cantor e compositor baiano Raul Seixas, a fim de verificar o modo de constituição da paratopia em sua obra. Conforme compreendido por Maingueneau, a paratopia é característica dos discursos constituintes (o religioso, o filosófico, o científico, o literário), os quais se propõem como discursos de Origem, que são validados por uma cena de enunciação que os autoriza a si mesmos. Para o autor, a paratopia advém de uma localidade paradoxal, que não diz respeito à falta de um lugar próprio ao discurso, mas refere-se à difícil negociação entre o lugar e o não-lugar, processo que emerge da própria impossibilidade desse discurso estabilizar-se. Tal processo incide sobre três dimensões classificadas por Maingueneau como embreantes paratópicos: a cenografia, o código de linguagem e o ethos. Com o intuito de demonstrar o modo de constituição do lugar paratópico de Raul Seixas, esta dissertação se debruça sobre um corpus de análise composto por prática discursiva intersemióticas do compositor: i) canções; ii) capas de álbuns; iii) vídeo de apresentação em um festival; iv) e um gibi-manifesto que foi distribuído em alguns shows do cantor. A escolha desse corpus e as análises realizadas foram delimitadas em função dos embreantes paratópicos que funcionam como motores que impulsionam a criação da obra em estudo e, nessa perspectiva, constroem a sua paratopia que, no caso específico de Raul Seixas, advém da reivindicação e/ou anunciação de um tempo e um espaço transcendentes e metafísicos, que estão além da própria enunciação.<br>Mestre em Estudos Linguísticos

Le roman négro-africain de dénonciation que nous appelons " roman subversif " et que d'aucuns insèrent dans la catégorie dite roman de la rupture, en tant que macro-acte de langage, comporte bien une visée illocutoire subversive. Cela est vrai même si le discours littéraire négro-africain d'après les indépendances, à l'instar de tout discours, reste contraint, c'est-à-dire en grande partie déterminé par le contexte sociopolitique, mais également le champ littéraire francophone où entrent en concurrence divers " positionnements " et " postures " d'auteurs. Nous montrons dans ce travail que cette visée qui est une entreprise de dévoilement des dérives des nouveaux régimes politiques et/ou religieux, résultante de l'intrication du contexte non verbal négro-africain et de l'intérieur des romans, et qui se traduit par la construction littéraire de divers ethos individuels et collectifs, peut s'étudier, entre autres, au moyen de la méthode d'analyse du discours. En reliant extérieu r et intérieur du texte littéraire, ce qui écarte l'immanence structuraliste, nous abordons le roman négro-africain d'après les indépendance comme un dispositif d'énonciation dont le centre déictique et modal est le garant du discours, à savoir le narrateur principal qui est le plus souvent " homodiégétique " dans notre corpus à l'exception de Perpétue où il est " extradiégétique ". Ainsi, la thèse ruine la conception romantique qui distingue le moi social de l'écrivain et le moi créateur. Nous considérons donc que les romans de notre corpus sont des activités sociales s'insérant dans les pratiques discursives d'une société, ce qui consacre définitivement la relation texte et société en mettant en branle des notions de la problématique de l'énonciation à grande portée socioculturelle comme la " scénographie ", la " scène générique ", la " scène validée ", la " paratopie ", etc. Nous confirmons donc la possibilité d'un enrichissement des approches de la littéraire négro-afric aine considérant l'histoire littéraire composée de trois entités séparées (l' " homme ", l' " uvre " et le " milieu ") et qui sont restées plus ou moins classiques, c'est-à-dire thématiques, souvent sociologiques. En recourant systématiquement aux outils de la linguistique de l'énonciation, de la pragmatique, de la linguistique textuelle, de l'argumentation, de la linguistique interactionniste, etc., nous appliquons à quelques romans négro-africains d'après les indépendances et à plusieurs séquences textuelles que nous avons sélectionnées et tirées de ce corpus, la méthode de l'analyse du discours telle qu'elle est théorisée dans la sphère européenne par des chercheurs tels que Dominique Maingueneau et Patrick Chareaudeau, mais également d'autres qui ont développé des problématiques linguistiques proches ou similaires : Jean Michel Adam, Ruth Amossy, Emile Benveniste, Catherine Kerbrat-Orecchioni, Oswald Ducrot, etc., pour ne citer que ceux-là. L'étude de la polyphonie pour la détermination des voix en présence dans les romans nous conduit à l'étude, d'une part, de la double énonciation qui se traduit par les dialogues en tant que modalité narrative où plusieurs énonciateurs sont mis en scène, de l'autre, de tous les types de discours rapporté, mais également de la manifestation verbale du peuple négro-africain. Ces voix définissent des identités énonciatives, celles des camps opposés, à savoir le pouvoir politique et/ou religieux et les opposants qui sont sans cesse en conflit dans les romans. Bannissant l' " authenticité " qui était la visée des conceptions identitaires négro-africaines comme la Négritude, les auteurs du corpus utilisent le discours rapporté et la double énonciation non pour restituer la réalité crue, comme dans le roman à thèse, mais pour dénoncer en dictant, en creux, au lecteur modèle ce qu'il faut penser ou croire. La tourmente politique et/ou religieuse est décriée par la présentation au lecteur de " patrons discursif " et d'un code langagier qui s'insérèrent dans l' " interdiscours " et qui montrent différents ethos populaires ou individuels fonctionnant comme des repoussoirs utilisés pour la dénonciation. Le lecteur modèle arrive à produire l'effet discursif attaché aux textes par l'activité d'" incorporation " de ces ethos qui se manifestent par une certaine corporalité et une vocalité précise.Mots clés :Énonciation, polyphonie, " scène d'énonciation ", genre, dialogue, interaction, argumentation, " posture ", " paratopie ", ethos, discours, texte, contexte.

Cette thèse a pour ambition d’interroger les positions inévitablement instables que ne cesse de briguer l’écrivain Germano Almeida au sein de la société capverdienne. Partant du principe que l’énonciation littéraire est constitutivement tenue de réfléchir dans l’univers fictionnel qu’elle construit les conditions de sa propre existence, cette étude vise d’abord à montrer comment Germano Almeida met en place des scénographies qui lui permettent de négocier l’inscription de ses œuvres tant dans le champ du pouvoir que dans le champ littéraire du Cap-Vert. Dans la première partie, l’analyse porte sur les rapports spécifiques qu’entretiennent littérature et politique dans un espace d’abord colonial puis post-colonial, l’introduction de l’humour et de l’ironie dans la littérature « savante », la tension entre une oralité revendiquée et une écriture effective qui prend une dimension particulière dans un espace linguistique diglossique et finalement les écarts entre la réception planifiée et la réception effective de son œuvre. La deuxième partie met en évidence la volonté chez Germano Almeida de participer à la configuration de l’espace social capverdien. Si les premières fictions almeidiennes ont systématiquement déconstruit les représentations traditionnellement rurales et intégrées de la société créole, les plus récentes dessinent en définitive les contours d’une société plutôt urbaine qui ne cesse de faire face à la contingence. La troisième partie fait justement ressortir une volonté constante d’inscrire les diverses expériences créoles dans un continuum temporel qui permette non seulement de donner sens au passé mais également d’avoir une meilleure maîtrise de l’avenir et d’échapper ainsi au danger de la répétition carnavalesque de l’histoire. L’étude de ces configurations spatio-temporelles met finalement en évidence le fait que leur énonciateur se conçoit non seulement comme un élément à part entière de la société capverdienne mais également son observateur distancié et donc averti<br>This thesis intends to examine the inevitably precarious stances that the writer Germano Almeida constantly seeks out within Cape Verdean society. Starting from the premise that by definition literary expression constitutes a reflection on its own existence within the fictional universe it creates, this study intends, first, to illustrate how Germano Almeida, through his scenography, manages to position his work both within the power structure and on the literary scene of Cape Verde. The first part of the analysis focuses on the special relationship between literature and politics in both colonial and post-colonial society, the inclusion of humor and irony in scholarly literature, the conflict that arises between a well-established oral tradition and actual writing with the latter playing a singular role within a highly differentiated bilingual society. Lastly, the gap between the expected and the real reactions to Almeida’s work is examined. The second part of the analysis illustrates Germano Almeida’s desire to participate actively in shaping Cape Verdean society. If Almeida’s first fictions systematically deconstruct traditional representations of a rural and cohesive Creole society, more recent works definitely depict the traits of a primarily urban society that is forced to constantly deal with contingencies. The third part of this study highlights Almeida’s constant desire to place the diversity of Creole experiences on a time continuum that not only gives meaning to the past but also provides greater command of the future and the possibility to thus avoid the danger of grotesquely repeating history. In the end, the study of these time-space configurations makes it obvious that their author sees himself not only as a full member of Cape Verdean society but also as an objective, and therefore, informed observer

L’oeuvre de Sergio Chejfec (Buenos Aires, 1956), composée de romans et de nouvelles,de nombreux essais et de deux recueils poétiques, figure parmi les propositions les plus singulières de la littérature argentine contemporaine. Entamée à la fin des années 1980, dans le contexte d’intense questionnement sur les moyens et les fins de la littérature nationale qui caractérise la période post-dictatoriale, cette oeuvre se poursuit encore aujourd’hui, désormais depuis New York. L’analyse transversale de l’oeuvre que nous proposons par la présente thèse s’appuie sur un corpus primaire de sept romans et s’organise autour des notions d’étrangeté et d’étrangèreté.La première partie s’attache à montrer que les altérations qui viennent semer le trouble dans la relation du sujet au réel ont partie liée avec la disparition de l’alter ego, qu’elles peuvent se concevoir à la lumière de la notion freudienne d’Unheimliche (« l’étrange familier »), et que s’y articule notamment la figure spectrale, en lien avec un passé – intime ou national – qui ne passe pas. L’étrangèreté géographique, sujet de la deuxième et la troisième partie, est définie comme une impossible territorialisation et se manifeste par les motifs récurrents de la marge et de l’errance. Au leitmotiv marginal – lieux périphériques, personnages paratopiques –s’associe également une figure d’auteur instable dans le champ littéraire argentin. Le motif de l’errance, quant à lui, réactive à la fois le mythe du peuple juif errant, par lequel l’auteurs se réapproprie ses origines, et le topos littéraire argentin du « désert ». L’errance, enfin, est aussi celle du lecteur, que Chejfec ne cesse de désorienter par un ensemble de procédés d’« étrangisation » (ostranenie) de la forme de ses textes.C’est enfin l’étrangèreté à soi et le brouillage des identités qui nous occupera dans un quatrième temps : il s’agira d’analyser la déconstruction du personnage romanesque, mais aussi les phénomènes de dédoublements, d’aliénations ou d’hybridités des sujets qui sont à l’oeuvre chez Chejfec. Les figures troublées de l’auteur, la question de la filiation et des origines ainsi que le rapport d’étrangèreté à la langue seront par la suite les axes principaux de cette dernière partie<br>Sergio Chejfec was born in Buenos Aires, in 1956, and wrote novels, short stories, numerous essays and two collections of poems. His work is listed among the most unusual propositions in contemporary Argentine literature. Sergio Chejfec started writing at the end of the 1980s, in a context of intense questioning of the means and ends of the national literature which characterizes the post-dictatorship period, and now goes on with his work from New York City. This thesis intends to offer a cross-disciplinary analysis, relies on a corpus of seven novels, and is organized around the notions of strangeness and strangerness.The first part endeavors to show that the distortions that trigger a trouble some relationship between the subject and the world are hand in glove with the disappearance ofthe alter ego; that they can be conceived of in the light of the Freudian notion of the uncanny (das Unheimliche); and that they articulate a spectral figure that is linked with either an intimate or national past, and yet does not pass. Geographical strangerness is the main focus of the second part and is defined as impossible territorializing. It expresses itself in the recurrent motifs of margin andwandering. The marginal leitmotiv – peripheral places, paratopical characters – is also articulated with the figure of an author who remains on the fringe of the Argentine literary circle. As for the motif of wandering, it reactivates both the myth of the Wandering Jew,through whom the author takes back his origins, and the Argentine literary topos of the“desert.” At last, wandering is that of the reader, whom Chejfec tries to disorientate through a series of processes that “enstrange” (ostranenie) the form of the texts.The fourth part concentrates on strangerness and the blurring of identities. It analyses the deconstruction of the novelistic character, and the phenomena of splitting, alienating and hybridizing the subjects in Chejfec’s work. The troubled figures of the author, the question of filiation and of origins and strangerness to language are the main threads of this last part

During this period, various cultural, social, political events were severely repressed. There was, during the most repressive years - 1970 - an institutionalized censorship, which bared subversive content to the existing order. It is precisely in this most repressive period that it is focused our research to bring literary materials for analysis of some linguistic phenomena. These materials are some texts, tale genre of Cadeiras proibidas by Ignácio de Loyola Brandão, 1976, and Seminário dos ratos, by Lygia Fagundes Telles, 1977. These materials are the corpora of the research, which were taken sets of textual sequences for analysis, foundations of some theoretical notions from a discursive viewpoint of discourse analysis, more specifically those forged in the polls and French scholars. At first, this research aims an understanding of how such literary constructions created conditions of possibility to produce, incorporate and manage discursive places of meaning, even having as historical conditions of production, a curtailed and oppressive socio-cultural environment. This time also, we want to understand how it was possible the literary creation itself, at a time of censorship and repression, by a hypothesis that is based on the notion concept of paratopia, from Dominique Maingueneau in his studies of literary discourse. To the different axes of paratopic creation, we want to add an axis specifically of political creation, which would explain a certain theoretical and analytical path to literary productions in the historic time when appeared such narratives. In a second step, there is a shift that aims to establish a different theoretical and methodological procedure for, in the same process, comprehend, describe and interpret linguistic elements and discursive elements of these corpora. In this second "time-purpose" it is firmed the hypothesis of scrutinizing the composition and formation of names and descriptions given to characters in their actions in the narrative plot. Moreover, as these names and descriptions are joined also to the composition of stereotypes that ultimately sustain discursive images that the characters give themselves and others, building dialectically, then, a discursive ethos of these texts for the historical and social context in question. These elements, we believe, make up the set of forms for adhesion of speakers to a desired speech by literary texts in any possible worlds. Trying to answer these questions and hypotheses are the objective of this research.<br>Nosso trabalho busca uma compreensão discursiva acerca de algumas manifestações literárias ocorridas durante o último regime ditatorial brasileiro, qual seja, entre 1964 e 1985, período em que militares e civis perpetraram um golpe ao estado democrático político e de direito. Durante este período, diversas manifestações culturais, sociais, políticas foram severamente reprimidas. Havia, durante os anos mais repressivos – década de 1970 – uma censura institucionalizada, que barrava conteúdos ditos subversivos à ordem vigente. É justamente nesse período mais repressivo que incide nossa pesquisa ao trazer materiais literários para análise de alguns fenômenos linguísticos. Estes materiais são alguns textos, do gênero conto, de Cadeiras proibidas, de Ignácio Loyola Brandão, 1976, e Seminário dos ratos, de Lygia Fagundes Telles, de 1977. É desses materiais, os corpora da pesquisa, que retiramos nosso conjunto de sequências textuais para análise e fundamentação de algumas noções teóricas do mirante discursivo da Análise do Discurso, mais especificamente as forjadas nas pesquisas e nos estudiosos franceses. Em um primeiro momento, essa pesquisa visa à compreensão do como tais construções literárias criaram condições de possibilidade para produzir, incorporar e gerir lugares discursivos de sentidos, mesmo tendo, nas condições históricas de produção, um ambiente sociocultural cerceado e opressor. Deste momento ainda, analisamos como foi possível a criação literária em si, em um momento de censuras e repressões, por uma hipótese que se fundamenta na noção-conceito de paratopia política, de Dominique Maingueneau em seus estudos sobre discurso literário. Aos diferentes eixos de criação paratópica, querermos acrescentar um eixo de criação especificamente político, o qual explicaria um certo percurso teórico e analítico para as produções literárias na quadra histórica onde apareceram as narrativas analisadas. Em um segundo momento, há um deslocamento que tem como objetivo estabelecer um outro procedimento teórico-metodológico para, em um mesmo processo, compreender, descrever e interpretar elementos linguísticos e elementos discursivos desses corpora. A este segundo "momento-objetivo" se firma a hipótese de perscrutarmos a composição e formação de nomes e descrições dados a personagens em suas ações na trama narrativa. Mais ainda, como esses nomes e descrições se coadunam também à composição de estereótipos que, em última instância, sustentam imagens discursivas que personagens atribuem a si e a outros, construindo, dialeticamente, então, um ethos discursivo desses textos para a conjuntura histórico-social em questão. Esses elementos, acreditamos, compõem o conjunto de formas de adesão de interlocutores a um discurso almejado pelos textos literários em quaisquer mundos possíveis. Although constrict by institutional censorship, artistic expressions of several kinds have not ceased entirely. There was resistance. Thus, given the importance of these art manifestation during the period as an attempt of escaping from the imposition of discursive silence, between them, literary manifestations, there are some studies which the goal is to reflect on the historical conditions that the works, by chance, carry with them: a background that covers the works; or studies in which structural, regulatory or styles made: an esthetic form that compose them. These researches are given in various theoretical lines, connected to literary theories, or linked to social theories. From another theory viewpoint, our objective with this thesis is to try to analyze what were the conditions of possibility of discursive enunciation, of a collection of short stories, published in Brazil during the historic period referred, analyzing it as an enunciative device of communication basically in two possibilities: a paratopic creation and scenography. For this, we took Cadeiras Proibidas by Ignacio de Loyola Brandão, for analysis, and from the assumptions listed, take it to a set of hypotheses, from authorial position in a literary field to the linguistic-discursive forms in scenography, in which the tales are engendered, making the text of the work its own regulation context. Therefore, mobilizing discursive analytical categories, especially those done by Dominique Maingueneau, but with nuances of other theorists of discourse analysis of French orientation, and try to answer these hypotheses and work analysis.<br>No Brasil, entre 1964 e 1985, liderado por militares, instalou-se no poder do Estado um regime ditatorial que cerceou muitos direitos civis e humanos de indivíduos ou instituições que mantiveram posicionamento político-ideológico contra esse governo repressivo. Embora restritas por censuras institucionais, as manifestações artísticas de diversas ordens não cessaram totalmente. Houve resistência. Assim, dada a importância dessas manifestações artísticas durante o período como uma das tentativas de escapar à imposição do silêncio discursivo e, entre elas, as manifestações literárias, existem alguns estudos cujo objetivo é refletir sobre as condições históricas que as obras, porventura, carregam consigo: um pano de fundo que recobre o conteúdo dessas obras; ou, então, estudos cujo objetivo é refletir as propriedades estruturais, normativas ou de estilos empreendidos: uma forma estética que as compõe. Essas pesquisas se dão em diversas linhas teóricas, sejam ligadas a teorias literárias, sejam ligadas a teorias sociais. De outro mirante teórico, nosso objetivo com esta dissertação é tentar analisar quais foram as condições de possibilidade enunciativo-discursivas de uma coletânea de contos, publicada no Brasil durante o período histórico referido, analisando-a como um dispositivo enunciativo de comunicação em duas possibilidades basicamente: a criação paratópica e a cenografia. Para tanto, tomamos Cadeiras proibidas, do escritor brasileiro Ignácio de Loyola Brandão, como material de análise para, a partir das premissas elencadas, levá-lo a um conjunto de hipóteses que vão desde o posicionamento autoral em um campo literário até as formas linguístico-discursivas da cenografia, com a qual os contos estão engendrados, fazendo dos textos da obra a própria gestão do contexto. Dessa forma, mobilizando categorias analíticas discursivas, sobretudo as forjadas por Dominique Maingueneau, porém com matizes de outros teóricos da análise do discurso de orientação francesa, tentaremos responder a essas hipóteses e análises do trabalho.

Abstract:
Les specificites des deux anticorps anti-cortisol ont ete modifiees par mutagenese. Les genes codant pour les anticorps ont ete introduits dans un vecteur d'expression procaryote sous controle d'un promoteur inductible. Les fragments d'anticorps produits forment des corps d'inclusion insolubles. Une fusion a ete realisee entre un fragment d'anticorps et la mbp (proteine affine du maltose) mais la proteine est egalement insoluble. Par contre, cette fusion est presente dans le milieu de culture sous une forme soluble et active quand elle est exprimee dans une souche dont les proteines periplasmiques sont liberees dans un milieu extra-cellulaire. La mutagenese des paratopes ne peut pas etre envisagee sur l'ensemble des boucles hypervariables (cdr). Des donnees structurales, obtenues par d'autres anticorps ont permis la modelisation des paratopes. Il a ainsi ete possible de reduire le nombre d'acides amines a modifier. Une nouvelle methode a ete mise au point. Elle a permis l'introduction de mutations dans plusieurs cdr en isolant un fragment d'adn simple brin utilise comme amorce. La banque de mutants obtenue a partir des anticorps anti-cortisol a ete introduite dans un vecteur phagemidique et exprimee a la tete des bacteriophages. La banque de phages a ete criblee sur l'estradiol qui n'est pas reconnu par les anticorps initiaux. Il a ete possible d'isoler des clones qui ont acquis la capacite de reconnaitre l'estradiol. L'etude de la structure des nouveaux anticorps est possible grace a la production des proteines de fusion. Ces donnees devraient permettre de mieux comprendre comment une proteine interagit avec un steroide

Abstract:
Au Maroc, comme dans le monde arabe, le règne du despotisme et du holisme nuisent à la démocratie et à la différence. Un régime anti-démocratique ne tolère pas de voix dissonante. Or, l’intellectuel imbu de valeurs universelles, cartésiennes s’oppose à cette manière de gérer la chose publique. Du coup, le système donne la chasse à l’opposant qui ose penser autrement. La tendance groupale interdit au sujet qu’il vive affranchi des autorités traditionnelles. En Europe, la montée des populismes et des logiques identitaires impactent le rapport à l’autre, perçu comme le représentant de son groupe et du coup discriminé et stigmatisé. Fouad Laroui a mis en scène, à travers une écriture autobiographique des personnages qui lui ressemblent, lettrés, francophones, diasporiques en rupture avec leurs groupes. Une catégorie peu fréquente dans le champ littéraire maghrébin : le libre penseur, l’agnostique, voire l’athée. Celui qui vit l’entre deux va donc souffrir car il sera vu à travers une vision identitaire verrouillée qui mutile le francophone de toutes ses appartenances. En réaction à cela Fouad Laroui développe une écriture de la différence, de l’hétérogène. Il fait l’éloge du métissage en accentuant sur la pluralité qui travaille le monde, l’Histoire, la ville, les personnes…<br>In Morocco, as in the Arab world, the reign of despotism and holism hurt democracy and difference. An anti-democratic regime does not tolerate dissonant voices. Now, intellectuals, imbued with universal Cartesian values, are opposed to this way of managing public affairs. As a result, the system chases the opponent who dares to think otherwise. The group tendency forbids the subject to live freed from the traditional authorities. In Europe, the rise of populism and the logic of identity have greatly impacted the relationship with the other, perceived as the representative of his group and therefore discriminated against and stigmatized. Fouad Laroui staged, through autobiographical writing, characters who resemble him, scholars, francophones, diasporas who break with their groups. It is an uncommon category in the Maghreb literary field: the free thinker, the agnostic, even the atheist. The one who lives in between will suffer because one will be seen through a locked identity vision that mutilates the francophone of any sense of belonging. In response to this, Fouad Laroui develops a writing of difference, of heterogeneity. He praises miscegenation by emphasizing the plurality that prevails the world, History, the city, people
