Abstract:
The Lockheed Martin/Northrop Grumman/BAE team was selected as the winning team to build what is considered the last manned combat aircraft, the F-35 Lightning II. The design was based on a commonality of parts between the three variants that will be used by the Air Force, Marines, and Navy. In building these air vehicles there are parts with a criticality rating requiring that they be traced from raw stock to assembly into the air vehicle. This record must be maintained for the life of the air vehicle. The Department of Defense (DoD) requirement, first published in 2003, stipulates that all defense contractors, including Northrop Grumman (NGC), serially label nearly every component and subsystem supplied to the DoD. The last air vehicle delivered to prime contractor Lockheed Martin Fort Worth (LM Aero) was delivered with 64 parts unaccounted for. NGC is currently preparing to ship the next air vehicle to Lockheed and is missing serial numbers on sixteen critical parts. When looking to see what the root cause may be, it became clear that this was not a one-dimensional problem, but actually three: Criticality Traceability, Parts Traceability as a whole, and Identification of each part per DoD mandate. Secondary problems were experienced by the misplaced parts such as: Schedule Delays, Added Resources and Increased Costs. The misplacement caused line personnel and their associated leads to resolve the issue and locate the part(s), thus impacting schedule and driving up costs. This project dealt with identifying and assessing the systems engineering processes required to create a system that would allow 100% traceability of all traceable parts produced and supplied to Northrop Grumman Integrated Systems, through production, and then final delivery to prime. The report examines five key systems engineering principles: Requirements Definition and Management. The system will be required to interface with all current databases and processes to ensure that all parts meeting DoD mandates for Identification and requiring traceability are automatically tracked from raw stock to delivery to prime. System Architecture. The proposed system will be an N-Tiered Web-based system that still retains existing databases using Microsoft Windows and browser-based clients. The ideal system will work by integrating all existing databases under one all encompassing system using API as the interprocess communicator. System/Subsystem Design. There will be three layers in the system, Application, Infrastructure, and Data which will be accessed by use of DAA, IDL, and CORBA with Java and guard technology used to give access rights to all requiring access to the system. System Integration. The system will be capable of interfacing with other applications throughout NGIS' supply chain including the Federal Government for transfer of content, for retrieval of content, and for content management lifecycle processes in which NGIS interacts. Validation and Verification. The test methodology should support a building block process for the integration of the systems. This should be a serial approach with strict entrance and exit criteria building upon each other.

With the cost of microprocessor systems dropping steadily, sensor networks are becoming more and more viable for replacing traditional "dumb" sensors. This presents a host of advantages to the system collecting data, and allows for unprecedented system flexibility. In parallel to these developments, RFID "tagging" has come a long way and has also entered the realm of affordable sensor solutions. The merger of these ideas enables transient, modular, and reconfigurable asset identification and tracking systems. In this paper, we explore the feasibility of implementing such a network with commercial off-the-shelf (COTS) parts for real world applications. Specifically, we create a drop-in system for managing documents and records in a medical triage center, based on identifying assets and personnel by means of RFID. A self-reconfiguring sensor network provides the foundation, allows for the system to be deployed in minutes, both on a battlefield or in a hospital, and once deployed, the hospital beds may move around freely. The advantages of this platform will be discussed alongside the unique requirements that are introduced in this type of project.

L’étude des idiomes pose beaucoup de questions, notamment celles qui concernent leur traitement pour obtenir leurs significations. Le constat est que beaucoup sont en réalité figurés. Ce sont des métaphores qui ont atteint le statut de l’idiomaticité sans pour autant devenir des métaphores mortes. La métaphore constitue, ainsi, un outil de traitement significatif. Il est question dans cette thèse d’un travail portant sur l’identification des idiomes à valeur métaphorique et sur le traitement de cette catégorie dont l’appellation est expression idiomatique se distinguant par la mention de noms de parties corporelles ou de notions abstraites. Comment la métaphore opère t-elle ? La réponse à cette question contredit les idées des théories linguistiques traditionnelles qui ont longtemps considéré que tous les idiomes étaient non-compositionnels. La discussion porte également sur le travail de G. Lakoff (1987) qui a considéré que les expressions idiomatiques sont motivées par des métaphores conceptuelles sous-jacentes de type « A is B ». Ainsi, le traitement des expressions idiomatiques passe nécessairement par l’identification de la métaphore conceptuelle propre à chaque expression. Cependant, ce travail s’avère complexe dans une situation de conversation. De là, l’idée est d’arriver à déchiffrer le sens figuré sans nécessairement passer par la construction de la métaphore conceptuelle. Pour démontrer la motivation des expressions idiomatiques plusieurs arguments ont été proposés. Le choix du lexique (le verbe, la préposition, l’adverbe, etc. ) et l’agencement syntagmatique saillant dans ces expressions sont étudiés. Il est également question de la suffisance ou non du travail linguistique dans le traitement figuré de ce type d’expression. Le recours au domaine conceptuel défini par le savoir conventionnel sur les parties corporelles, les notions abstraites et par leur interaction avec les autres constituants idiomatiques s’avère indispensable pour compléter le travail linguistique
The construction of idiom signification necessitates a special strategy. A large number of idioms prove to be figurative in nature. They were at the beginning mere creative metaphors that survived by frequent use and acquired a new status that of idiomaticity. This figurative character offers a special case of study to demonstrate the motivation of this kind of idioms referred to as idiomatic expressions. This thesis exploits this figure in the case of the idiomatic expressions containing body parts and abstract nouns to show that not all idioms are non-compositional as it was largely assumed by traditional linguistic theories. In this work, the identification of idioms encoding metaphorical analogy comes as a first step before their processing. Can linguistic clues be used to identify this category of idioms? How does metaphor function in the construction of figurative meaning? In addition to this, this work refers to the theory of G. Lakoff (1987) who assumed that idiomatic expressions are motivated by conceptual metaphors of the type “A is B”. In order to construct the figurative meaning of an expression it is necessary to construct the underlying conceptual metaphor that will structure the implicit analogy. However, this work is complex in a conversational situation. Hence, the work consists instead in constructing the figurative meaning without obligatory constructing the conceptual metaphor. The contribution of linguistic clues, namely the verb, the preposition, the aspect, the adverbs, etc. And the typical syntagmatic combinations are subject to study. Significant findings are obtained from the linguistic study. The work consists also in understanding whether the linguistic study is sufficient by it self to account for the figurative status of an expression or that the conceptual knowledge we have on the idiomatic constituents is required

This thesis deals with issues related to the identifiability and traceability of parts manufacturing company engaged in the aviation industry. The paper summarizes the international standard ČSN EN ISO 9001 and ČSN EN 9100, according to which the company is certified, and other in-house standards under which it dealt with the interpretation of international standards. Subsequent to solve the problem is the result of consultation within the company and market research, dealing with the identification of products. The proposed solutions are a self-adhesive label, which allows companies to leverage existing technology of barcodes generated by the SAP system and the simplicity of using the stickers. It is possible their reapplied after each operation, which was performed on the work piece and the staff are on the barcode system used, ie no need for complex training.

A travers diverses sources et particulièrement des sources orales recueillies auprès d’anciens militants communistes simples adhérents ou responsables et d’articles de la presse communiste martiniquaise, cette thèse cherche à éclairer sur la ferveur militante d’hommes et de femmes de la Martinique, d’une famille politique essentielle dans le courant du XXe siècle.Cette étude parcourt une longue période s’étendant principalement de 1920 à 1971, montrant les débuts balbutiants du communisme à la Martinique, son ascension, sa période florissante et les débuts de son lent déclin. La problématique est d’abord de faire découvrir des Martiniquais dans leur vécu de militant communiste, montrer comment ces hommes et ces femmes s’approprièrent une doctrine, la transformèrent peut-être pour l’adapter à leurs besoins, à leur culture. Il s’agit également d’examiner quelle structure communiste fut mise en place en Martinique, cette organisation était-elle à l’image des fédérations métropolitaines ou était-ce une formation politique spécifique au milieu martiniquais?Ensuite pour mieux appréhender les choix et les parcours, il a fallu arriver à l’identification des modèles, des origines de ce militantisme et la formation des militants. Les référents furent tant des figures propres au marxisme et au socialisme international et national que des personnalités contestataires du milieu politique martiniquais.Puis proposer une lecture des pratiques et des stratégies militantes pour évaluer la qualité et la particularité de cette militance ainsi que les conséquences de l’engagement de ces militants dans différentes sphères. Quelles étaient leurs propositions, pourquoi leurs revendications politiques différaient-elles de celles des autres communistes coloniaux ? Comment expliquer leurs choix d’une nouvelle société plus égalitaire dans un État socialiste mais toujours associé à la France ?Ce travail de recherches positionne cette militance au centre d’une analyse qui explore l’histoire politique et sociale d’une population, en s’appuyant sur d’autres disciplines comme la sociobiographie et les sciences politiques. Through various sources, particularly oral sources collected from ancient communists, ordinary members or officials, and articles of martinican Communist press, this thesis shows about the militant fervor of martinican men and women and essential political family in the course of the twentieth century. This study covers a long period extending mainly from 1920 to 1971 showing the early stuttering of communism in Martinique, his ascension, his prosperous period and the beginning of his slow decline. The issue is first to make discover Martinicans in their experience of communist militant, present how these men and women appropriated a doctrine, transformed it perhaps to suit at their needs, at their culture. It is also to consider what communist structure was put in place in Martinique; this organization was it like the metropolitan federations or was it a specific political party? Then to better understand the choices and paths, it’s necessary arrive at identifying mentors, origins of this activism and training of militants. Referents were as figures specific to Marxism and international and national socialism as figures as specific martinican politics. Then offer a reading of practices and activist strategies for assessing the quality and uniqueness of this militancy and the consequences of the commitment of these activists in different spheres. What were their political choices, why their political demands did they differ from others colonial Communists? How to explain their choice of a new society more egalitarian in a socialist state, but always associated with France? This research study positions that militancy in the center of an analysis that explores the political and social history of a population, based on other disciplines such as socio-biography and political-sciences. It examines the shaping of a people by a political significant group between 1920 and 1971, and the proposals of this group face different political and social problems in promoting an approach by the actors

Abstract:
L’anémie de Blackfan-Diamond (ABD) est une érythroblastopénie congénitale rare secondaire à un blocage de la maturation des cellules érythroïdes entre les stades BFU-e (EPO indépendant) et CFU-e (EPO dépendant). La maladie se manifeste par une anémie arégénérative, le plus souvent macrocytaire. Dans 50% des cas, à l’anémie s’associe un syndrome malformatif affectant l’aire céphalique et les extrémités et incluant un retard de croissance. Bien que très hétérogène tant phénotypiquement que génotypiquement, l'ABD est due à des mutations toujours hétérozygotes dans des gènes de protéines ribosomiques (RP) pour 80% des cas. Le premier gène identifié comme impliqué dans l’ABD est le gène de la RPS19 (RPS19), faisant de l’ABD la première ribosomopathie décrite. A ce jour, 20 gènes de RP ont été identifiés. L'haplo-insuffisance en RP entraîne un défaut de maturation des ARN ribosomiques (ARNr) générant un stress nucléolaire, qui lui-même conduit à la stabilisation de la protéine p53. La p53 stabilisée induit l’apoptose et l'arrêt du cycle cellulaire qui sont responsables en grande partie de l’érythroblastopénie. Plus rarement, l’ABD peut être la conséquence de mutations présentes sur le gène GATA-1 (facteur de transcription majeur de l’érythropoïèse), le gène TSR2 (interagissant avec la protéine RPS26 et intervenant dans la biogenèse des ribosomes) ou encore le gène EPO (cytokine clé de l’érythropoïèse). Cependant, 20% des patients atteints d’ABD ne sont toujours pas génotypiquement diagnostiqués : de nouveaux gènes candidats sont encore à découvrir chez ces patients. Dans cette perspective, l’objectif de ma thèse a donc été de caractériser le rôle fonctionnel de nouveaux gènes candidats afin de confirmer leur lien avec l’ABD. Nous avons procédé à un séquençage d’exomes chez 25 familles ce qui a permis d’identifier 8 gènes candidats. Nous présentons ici quatre de ces gènes dont deux gènes codant pour un chaperon ribosomique : HSPA14 et HEATR3, un gène codant pour une RP : RPL9 et un gène codant un facteur de croissance : CECR1.Les protéines chaperons du ribosome représentent un nouveau groupe de gènes pouvant être associé à la maladie. Mes travaux de thèse ont permis d’étudier la prolifération, la division, l’amplification, la différenciation et la viabilité des cellules primaires érythroïdes issues des patients porteurs de variations alléliques dans ces gènes. Ces expériences ont permis de mettre en évidence un défaut de prolifération érythroïde chez l’ensemble des patients testés accompagné d’une diminution de l’amplification et de la division cellulaire ainsi qu’un retard de différenciation. Ces résultats sont objectivés par la persistance des marqueurs d’immaturité CD34 et IL-3R ainsi que d’un retard d’apparition des marqueurs terminaux tels que la BAND3 ou l’alpha4-intégrine. L’étude transcriptionnelle et protéique met en évidence une stabilisation de p53, conduisant à une activation de ces cibles p21 et de bax. Le travail est finalisé pour HEATR3 (manuscrit en préparation) et en cours de finalisation pour HSPA14.L’étude sur le gène RPL9 grâce à un travail collaboratif a permis de mettre en évidence deux phénotypes différents en fonction du variant allélique identifié : un phénotype ABD pour un variant allélique de la 5’UTR ou un phénotype associé à un risque de cancer. L’étude de la différenciation érythroïde montre un impact sur la différenciation et la prolifération pour le variant lié à l’ABD uniquement. Parallèlement, une étude en collaboration sur le gène RPL13, a permis de confirmer le rôle spécifique de certaines protéines RP dans d’autres pathologies que l’ABD et d’ajouter une nouvelle maladie à la liste des ribosomopathies.Le facteur de croissance CECR1, identifié comme muté chez plusieurs familles du consortium EURODBA, représente un nouveau groupe de gènes pouvant être associés à la maladie
Diamond-Blackfan anemia (DBA) is a congenital rare erythroblastopenia due to a blockage in the maturation of erythroid cells between the BFU-e and CFU-e stages. DBA is characterized by an aregenerative, usually macrocytic, anemia, associated with the total absence or less than 5% of erythroid precursors in the bone marrow. In 50% of DBA cases, anemia is associated with congenital malformations affecting the cephalic area and the extremities of the limbs and a growth delay. The DBA phenotype and genotype are heterogeneous, however a mutation in a ribosomal protein (RP) gene, always at heterozygous state, is found in 80% of cases. Up to date, 20 RP genes have been associated with DBA pathophysiology, establishing DBA as the first identified ribosomopathy. Mutations of these RP induce a defect in rRNA maturation. Therefore, for ribosome dysfunction, cell cycle arrest and p53-mediated apoptosis induction are responsible for erythroblastopenia in patients. More rarely, DBA may be the consequence of mutations present on a non-PR gene: the GATA-1 gene (major transcription factor of erythropoiesis), the TSR2 gene (interacting with the RPS26 protein and involved in ribosome biogenesis) or the EPO gene (erythropoiesis key cytokine) have been identified so far. However, 20% of the DBA patients are still not genotypically diagnosed, leaving room for the discovery of new candidate genes. In this perspective, the aim of my PhD was therefore to identify new candidate genes involved in DBA etiology and characterize their functional roles of in order to confirm their link with DBA. For this purpose, we sequenced exomes on 25 families and identified 8 candidate genes. In this manuscript, I will present my work as part of a bigger project to validate four new genes involved in BDA pathophysiology.RPL9 is a RP of the large 60S ribosomal subunit. Mutations in this gene lead to two different phenotypes depending on the allelic variant: a DBA phenotype for an allelic variant of the 5' UTR or a phenotype associated with a cancer risk. As part of a collaborative work that compared the two RPL9 variants, I showed that the DBA variant only has an impact on erythroid differentiation Compared to a healthy individual, patients presenting the DBA variant exhibit a reduced proliferation rate and a delay in the acquisition of erythroid markers. P53-dependent activation of p21 in those cells is most likely responsible for the cell cycle arrest. Activation of caspases sign an induction of apoptosis and is consistent with the reduced viability of erythroid progenitors. A collaborative study on the RPL13 gene confirmed the specific role of certain RP proteins in non-DBA diseases and added a new disease to the list of ribosomopathiesXRibosome chaperone proteins represent a new group of genes that may be associated with DBA. I investigated the proliferation, division, amplification, differentiation and viability of primary erythroid cells from patients with allelic variations in one of these genes: HEATR3. These experiments revealed a lack of erythroid proliferation, with a defect in cell division. The mRNA and protein quantifications showed a stabilization of p53, leading to an activation of its targets: p21, controlling cell cycle, and Bax, involved in apoptosis induction. We also observed a delay in differentiation with the persistence of CD34 and IL-3R immaturity markers and a delay in the appearance of terminal markers such as BAND3 or alpha4-integrin. The role of HSP70 controlling GATA1 localization in early stages of the erythroid differentiation was recently elucidated. In this work, I identified as a new candidate gene for DBA, a HSP70 family member, HSPA14, and I characterized the defects in erythroid differentiation induced by this variant. Furthermore, I was able to identify an association of DBA with a variant in CECR1 gene encoding an adenosine deaminase in several families of the EURODBA consortium

Abstract:
Les éléments transposables (ETs) sont des séquences d’ADN mobiles qui ont la propriété remarquable de se déplacer et se propager dans les génomes. Découverts dans les années 1940 chez le maïs, ils sont présents dans tous les génomes séquencés à ce jour. Bien que le nombre d’études décrivant leur rôle dans l’évolution, la fonction et la structure des génomes n’a cessé d’augmenter depuis, nous sommes encore loin de comprendre toute l’étendue de l’impact biologique des ETs sur les organismes. La distribution des ETs n'est pas aléatoire, phénomène dû en partie aux mécanismes qui orientent l’intégration vers des régions préférentielles du génome. C’est le cas du rétrotransposon Ty1 de la levure S. cerevisiae, qui s’intègre principalement dans une fenêtre d’une kilobase en amont des gènes transcrits par l’ARN polymérase III (Pol III). Cette spécificité résulte d’une interaction entre l’intégrase codée par l’élément et une sous-unité de la Pol III, AC40. En son absence, Ty1 cible préférentiellement les subtélomères, ce qui suggère l’existence d’autres facteurs régulant la sélectivité d’intégration du rétrotransposon. Ce travail a eu pour but d’identifier ces facteurs par deux approches complémentaires. D’une part, nous avons étudié le rôle des marques d’histones corrélant avec les insertions de Ty1 dans le choix du site d’intégration. D’autre part, nous avons recherché de nouveaux partenaires de l’intégrase de Ty1 par une approche protéomique à grande échelle. Si nos résultats n’ont pas permis d’identifier de nouveaux facteurs cellulaires impliqués dans la reconnaissance du site d’intégration de Ty1, ils ont élargi le répertoire des protéines de l’hôte qui régulent la rétrotransposition de Ty1. En effet, nous avons découvert que le variant d’histone H2A.Z et la protéine kinase CK2 répriment la rétrotransposition de Ty1. CK2 agit aux étapes transcriptionnelle et post-transcriptionnelle, interagit avec l’intégrase in vivo et la phosphoryle in vitro sur plusieurs résidus dans le domaine C-terminal, régulant probablement ainsi sa stabilité. Des expériences supplémentaires seront nécessaires pour confirmer cette hypothèse, découvrir par quel mécanisme CK2 régule Ty1, et comprendre le rôle de H2A.Z
Transposable elements (TEs) are mobile DNA sequences, with the extraordinary ability to jump and propagate in genomes. Discovered in the 1940s in maize, they are present in all sequenced genomes. Even if a large amount of data reveals their role in evolution, function and structure of genomes, we are still far from understanding every facet of their biological impact on organisms. TEs are not distributed randomly, partly due to mechanisms that target the integration to preferred genomic regions. This is the case for Ty1, a retrotransposon of S. cerevisiae, that targets a one-kilobase window upstream of RNA Polymerase III (Pol III)-transcribed genes. This specificity is due to an interaction between the element-encoded integrase and AC40, a subunit of Pol III. When this interaction is disrupted, Ty1 insertions are redistributed to subtelomeres, which suggests that other factors that regulate the integration selectivity of the retrotransposon exist. The aim of this work was to determine those factors by two complementary approaches. First, we studied the implication in this process of histone variants that correlate with Ty1 integration sites. Second, we identified new partners of Ty1 integrase by a proteomic screen. Our results did not lead us to identifying new cellular factors involved in the recognition of Ty1 integration site. They however widened the array of host proteins that regulate Ty1 retrotransposition. Indeed, we found that the histone variant H2A.Z and the protein kinase CK2 repress Ty1 retrotransposition. CK2 acts at transcriptional and post-transcriptional steps, interacts with the integrase in vivo and phosphorylates it in vitro on several residues in the C-terminal domain, probably thereby regulating its stability. Additional experiments will be required to confirm this hypothesis, to discover by which mechanisms CK2 regulates Ty1, and to understand the role of H2A.Z

Durant plusieurs décennies, au sein de ce qui s'est appelé la « banlieue rouge », Ivry-sur-Seine faisait figure de « bastion » modèle pour le Parti Communiste Français. Le communisme municipal ivryen avait fait de ses cités ouvrières des espaces laboratoires au service d'un creuset d'affiliation sociopolitique particulièrement efficace. Jusqu'au tournant des années 1980, aux cités Maurice Thorez et Youri Gagarine, les résultats électoraux enregistrés par les différents représentants communistes en ont attesté. Avec la remise en cause du modèle de politisation fondé sur l'écosystème industriel, le renouvellement des générations pose avec acuité la question des conditions de reproduction d'une affiliation sociopolitique favorable aux représentants communistes. Une approche ethnographique sur la longue durée a permis de renseigner cette question. Depuis le milieu des années 1980, la trajectoire sociopolitique contrastée des deux quartiers atteste des ruptures infra-communales touchant ce type de territoire de la banlieue parisienne. Dans le quartier Youri Gagarine, la majorité des anciennes familles ouvrières a été remplacée par les nouveaux milieux populaires essentiellement composés de populations issues de l'immigration. Entretenant une historicité tout à fait différente à l'égard de l'étiquette « communiste(s)», les nouvelles générations participent, parfois activement, d'une contestation de l'ancienne autorité politique locale. À l'inverse, dans le quartier Maurice Thorez, situé au cœur du centre-ville, les descendants des familles ivryennes les plus proches de l'appareil partisan et/ou municipal ont maintenu résidence. Dans ce quartier, autour d'une endocratie politique locale, se maintiennent des liens communautaires fonctionnant de manière relativement indépendante de l'ancien encadrement partisan. Pour de nombreuses familles ivryennes appartenant à la classe moyenne, le maintien d'une certaine autorité communiste facilite leur accompagnement social, politique et électoral des métamorphoses contemporaines du communisme municipal
For decades, Ivry-sur-Seine was seen as a model Communist stronghold within the Paris ‘Red Belt'. The particular brand of communism practiced by Ivry's municipal government had turned its working-class housing estates into laboratories directed towards the production of a singularly efficient political affiliation system. Until the watershed of the 1980s, electoral results for the various communist representatives in the Maurice Thorez and Yuri Gagarin housing estates seemed to corroborate this. The decline of the politicization model born of industrialization as well as the generation gap have radically undermined the conditions in which a socio-political affiliation system favourable to communist representatives can survive, however. The choice of a long-term ethnographic approach can give us insight into this phenomenon. Since the middle of the 1980s, the contrasting socio-political evolution of the Thorez and Gagarin allotments has testified to the intra-municipal disruption that affects this type of suburban Parisian territory. In the Yuri Gagarin area, the majority of older working-class families have been replaced with a new working-class population essentially stemming from immigration. Often unaware of the rich history of communism in their municipality, these new generations are sometimes actively involved in the challenging of the older local political authority. Downtown, on the contrary, the descendants of the families that were closest to the local party machine have maintained residency in the Maurice Thorez area. Community links have survived around a local political “endocracy” that works relatively independently from the older partisan frame. For many middle-class families living in Ivry, the maintenance of a certain communist authority makes it easier to accept the social, political, and electoral transformations of contemporary municipal communism

Abstract:
One of the main goals for political parties is to attract as many voters as possible. The competition for voters among the parties is especially relevant in a multiparty system. A voter in such a system may choose between several parties and can order them in any way he or she pleases. Thus, the voter has an order of party preferences. In this thesis, I argue that differences in a voters’ first and second preference order can be used as a resource to discover unique patterns in the voters’ party identification. To address this conjecture, I have used the Super-Riks-SOM dataset to study such patterns between the years 2005, 2011 and 2013. The results show that along the points of measure, the Swedish voter has become less willing to vote for the middle parties. Hence, the pattern indicates that the party identification in the second preferences is more balanced in 2013 than in 2005.

Les anomalies congénitales des reins et des voies urinaires (CAKUT) regroupent un spectre d’anomalies du développement rénal incluant agénésies, hypoplasies et dysplasies. Elles constituent une cause majeure d’insuffisance rénale chez l’enfant, et d’interruption médicale de grossesse pour les formes les plus graves. Les mécanismes physiopathologiques sont complexes, impliquant des facteurs génétiques et environnementaux. Des mutations de plus de 50 gènes ont été rapportées dans des formes de CAKUT isolées ou syndromiques. Cependant, les causes génétiques de ces anomalies restent largement méconnues. Par séquençage d’exome total ou ciblé nous avons identifié 16 variants hétérozygotes dans le gène GREB1L (Growth Regulation By Estrogen In Breast Cancer 1-Like). Douze d’entre eux étaient présents chez des foetus avec agénésie rénale bilatérale, associée à une agénésie utérine chez les foetus de sexe féminin. GREB1L est un gène de fonction inconnue dont le rôle dans le développement rénal n’avait pas été rapporté précédemment. Nous avons généré un modèle de souris invalidé pour Greb1l par CRISPR-Cas9. Les souris homozygotes meurent in utero. Les embryons Greb1l-/- présentent tous une taille réduite, une exencephalie, des anomalies cardiaques ainsi que l’absence de reins, de canal de Wolff et de canal de Müller. Des études réalisées à des stades embryonnaires précoces nous ont permis de démontrer que Greb1l est nécessaire pour la spécification du mésoderme intermédiaire.En parallèle de ce travail, un autre gène, dans lequel un variant a été identifié dans une famille dont plusieurs membres présentaient des formes kystiques de CAKUT, est été étudié. Ce gène,SRPK1 (Serine/Arginine-Rich Protein Kinase 1) encode pour une kinase spécifique des protéines contenant des domaines riches en Sérine/arginine, notamment les facteurs d’épissage. Nous avons montré que des cellules épithéliales rénales invalidées pour Srpk1 présentent des défauts d’épithélialisation et de polarité apico-basale, des processus important pour le développement rénal. De plus, des études transcriptomiques nous ont permis de montrer que Srpk1 régule l’expression de nombreux gènes impliqués dans le développement rénal. L’ensemble de ces données suggère que SRPK1 est un bon gène candidat pour les CAKUT
Congenital Abnormalities of the Kidney and Urinary Tract (CAKUT) comprise a broad spectrum of renal and urinary tract malformations with a wide range of severity. CAKUT represent major causes of chronic kidney disease in children, and the most severe cases are associated with perinatal lethality and/or termination of pregnancy. CAKUT are due to defects arising during renal development and their pathophysiological mechanisms are complex, involving both environmental and genetic factors. Although disease-causing mutations in more than 50 genes have been reported,mutations in these genes can explain only 20% of the cases, while for the others the genetic causeremains unknown.Using a combined approach of whole-exome and targeted exome sequencing, we identified mutations in a novel gene, GREB1L (Growth Regulation by Estrogen in Breast Cancer 1 Like), in CAKUT-affected cases in 16 independent families, 12 of them with foetuses with bilateral kidneyagenesis (BKA) and uterus agenesis (female foetuses). This gene had never been implicated in kidney development and its function is unknown. We generated a mouse model invalidated for Greb1l usingCRISPR/Cas9. Embryonic lethality was observed in Greb1l knockout mice at the homozygous stateand all Greb1l-/- embryos were smaller and presented exencephaly, BKA and absence of Wolff and Muller ducts, as well as heart defects. Further studies on early embryonic stages demonstrated that Greb1l is necessary for proper intermediate mesoderm specification. In parallel, another candidate gene for CAKUT, SRPK1 (Serine/Arginine-Rich Protein Kinase 1), was studied. A variant in this gene was identified in a family with several cystic CAKUT-affected members. The protein encoded by this gene is a kinase specific for Serine/Arginine rich proteins, especially splicing factors. Invalidation of Srpk1 in kidney epithelial cells led to defects in epithelialisation and establishment of apico-basal polarity, processes important for proper kidney development. Moreover, transcriptomic studies on these cells revealed Srpk1 to be implicated in the regulation of the expression of genes controlling kidney development. Altogether, this suggests thatSRPK1 is a good candidate gene for CAKUT

Chez les eucaryotes, l’organisation de la chromatine est indispensable à la régulation de l’expression des gènes et la stabilité du génome. Les ciliés constituent un excellent modèle d’étude des mécanismes qui permettent de maintenir l’intégrité du génome eucaryote. Notre modèle d’étude, l’eucaryote unicellulaire Paramecium tetraurelia, se distingue par l’élimination massive et reproductible de près de 30% de séquences d’ADN germinal lors du développement du macronoyau somatique après les événements sexuels. Ces séquences sont éliminées grâce à un processus en plusieurs étapes impliquant la formation d’hétérochromatine dirigée par de petits ARN suivi d’excision par la transposase domestiquée Piggy Mac (Pgm) et de réparation. Les mécanismes moléculaires permettant de comprendre la reconnaissance spécifique de ces séquences germinales dans le contexte de la chromatine ainsi que la précision de la coupure demeurent mal compris.Le chaperon d’histone Spt16, associée au partenaire Pob3, fait partie du complexe hétéro-dimérique FACT (FAcilitates Chromatin Transactions). FACT intervient dans de nombreux mécanismes liés au métabolisme de l’ADN tels que la transcription, la réparation, la réplication ou l’accessibilité de la chromatine. Chez Paramecium tetraurelia, on a identifié deux protéines homologues de Spt16 et Pob3 exprimées uniquement durant le développement du macronoyau au moment où se produisent les réarrangements. Les protéines Spt16-1 et Pob3-1 fusionnées à la GFP se localisent dans les macronoyaux en développement. Nous avons montré que la protéine Spt16-1 est essentielle à la production d’une descendance sexuelle viable. Le reséquençage du génome après inactivation de SPT16-1 a montré que Spt16-1 est nécessaire à l‘ensemble des réarrangements du génome et conduit à des défauts semblables à ceux obtenus suite à l’inactivation de PGM. Spt16-1 agit en aval des voies de dépôt des marques d’hétérochromatine guidées par les petits ARNs mais en amont de l’endonucléase Pgm. Nous avons montré que Spt16-1 était nécessaire à la localisation correcte de Pgm responsable de l’introduction des cassures double brin dans les macronoyaux en développement. Nous proposons un modèle dans lequel Spt16-1 favorise l’interaction de la machinerie d’excision avec la chromatine pour rendre accessible les sites de coupure sur l’ADN à Pgm.La méthylation des adénines sur l’ADN (6mA), bien connue chez les bactéries pour son rôle dans le système de restriction modification, a été décrite ces deux dernières années chez plusieurs eucaryotes en faible proportion dans le génome. Cependant son rôle chez les eucaryotes est encore peu compris. D’anciennes analyses par chromatographie à l’aide de nucléotides marqués détectaient de l’ordre de 2,5% des adénines méthylées chez P. tetraurelia mais sa localisation précise n’avait jamais été montrée. Il avait été proposé que cette modification pourrait signaler avec précision les sites de coupures pendant les réarrangements d’ADN où une partie des séquences germinales présentent des bornes AT. L’abondance des adénines méthylées fait de la paramécie un excellent modèle d’étude de cette modification. En combinant des techniques d’Immunofluorescence, d’HPLC-MS et de séquençage, nous avons ainsi décrit la présence et la localisation dans la cellule et au cours du cycle de vie de 6mA chez P. tetraurelia. Ces approches ont également permis de montrer l’absence de cytosine méthylée chez la paramécie. Nous avons montré que la méthylation des adénines était retrouvée majoritairement dans le génome somatique et de manière transitoire dans le génome germinal. Elle est établie au cours du développement du macronoyau somatique quand se produisent les réarrangements du génome. Ces résultats préliminaires permettront de poursuivre l’étude de la méthylation afin d’identifier les enzymes responsables et son rôle dans la cellule
In eukaryotes, chromatin organization is required for the regulation of gene expression and genome stability. Ciliate provide excellent model to study mechanisms involved in maintain of genome integrity. Our study model, the unicellular eukaryote Paramecium tetraurelia, has the particularity to eliminate massively and reproducibly 30% of germinal DNA sequences during the development of the somatic macronucleus after sexual events. Those sequences are eliminated by a multi-step process involving small RNA-directed heterochromatin formation followed by DNA excision by the domesticated transposase Piggy Mac (Pgm) and DNA repair. Molecular mechanisms underlying the specific recognition of those germinal sequences in chromatin context and the precision of the excision, remain elusive. The histone chaperone Spt16, associated to its partner Pob3, is part of the heterodimeric complex FACT (FAcilitates Chromatin Transactions). FACT is implicated in many mechanisms involving DNA metabolism such as transcription, repair, replication or chromatin accessibility. In P. tetraurelia, we identified two homologous proteins to Spt16 and Pob3 expressed only during macronucleus development at the time when genome rearrangements occur. Spt16-1 and Pob3-1 fused to GFP are localized in developing macronuclei. We showed that Spt16-1 is required to obtain a viable sexual progeny. Genome re-sequencing after SPT16-1 inactivation showed that Spt16-1 was required for all DNA elimination events and leads to similar phenotypes and defects to those obtained after PGM inactivation. Spt16-1 acts downstream of small RNA-directed heterochromatin formation and upstream of Pgm. We showed that Spt16-1 was required for the correct localization of Pgm responsible for DNA double strand breaks in developing macronuclei. We proposed a model in which Spt16-1 mediates interaction between chromatin and excision machinery that facilitates access to DNA cleavage sites for the Pgm endonuclease. Adenine DNA methylation, well known in bacteria for its role in restriction modification system, has been described during the last two years in several eukaryotes but in low proportion in the genome. However, its role in eukaryotes remains elusive. Previous analyses by chromatography with radio labelled nucleotides detected around 2,5% of methylated adenine in P. tetraurelia but its precise localization and role have never been analyzed. It has been proposed that this modification could mark with precision the excision sites during genome rearrangements when part of the eliminated sequences carry AT boundaries. The abundance of methylated adenine makes of Paramecium an excellent model to study this modification. Combining immunofluorescence techniques, HPLC-MS and sequencing, we described the presence and the cellular localization during life cycle of 6mA in P. tetraurelia. Those approaches allowed us to show that methylated cytosine are absent in Paramecium. We showed that methylation is mainly found in the somatic genome and transiently in germinal genome. It appears during somatic macronucleus development when genome rearrangements occur. Those preliminary results will allow us to pursue the study of adenine methylation by identifying responsible enzymes and its role in the cell

GREENWOOD, PAUL BRIAN. Sport Fan Team Identification in a Professional Expansion Setting. (Under the direction of Michael A. Kanters.) This study assessed the relationship between team identification of sport fans and a number of different variables including but not limited to: the reasons for initially becoming a fan, specific sport knowledge, and purchasing behavior. A non-random sample of spectators (N = 356) at an Arena Football League (AFL) game completed a survey designed to identify or measure the aforementioned variables. A t-test revealed that males reported significantly higher levels of team identification than females. Analysis using a Pearson correlation matrix showed a positive correlation between team identification and sport knowledge, attendance rate, and purchasing behavior. Likewise, a positive correlation was found between team identification and the following reasons for initially becoming a fan: closest team, born and/or live in the city and/or state, team success, the players and/or coaches, parents and/or family influence, and the tailgating and party atmosphere. The findings have important implications for professional sport franchises in terms of tailoring marketing efforts for potential fans.

L'étude des "Bourgeois de Paris" vise à définir les sens et usages de cette qualification dans la littérature et dans la vie politique, religieuse et sociale parisienne du règne de Louis XIV à la révolution. La littérature du siècle des lumières différencie les "bourgeois" de la noblesse et des classes populaires par leurs pratiques socio-culturelles et leurs mentalités. Il existe une pluralité de groupes de "Bourgeois de Paris" qui se distinguent parce qu'ils rassemblent chacun des individus dont les horizons professionnels, le degré d'intégration dans la capitale. . . Sont différents. Quand ceux-ci ont un statut commun, ils sont associés à des institutions héritées du moyen âge (l'hôtel de ville, la confrérie Notre-Dame aux bourgeois de Paris), ou intégrés dans des hiérarchies juridico-fiscales (les privilèges individuels, la capitation). La définition théorique de ces groupes peut être stable depuis leur création (le corps large est défini comme les habitants demeurant à Paris depuis un an et un jour, nobles et roturiers en font partie), elle a parfois évolué sous la pression des besoins financiers de la monarchie ou bien d'un groupe social (les marchands des six corps monopolisent l'échevinage). Les milieux socio-professionnels dont les membres utilisent concrètement leur statut latent de "Bourgeois de Paris" peuvent également changer (les nobles et les parlementaires se font ainsi plus rares au XVIIIe siècle comme mandés à l'hôtel de ville et comme confrères). Enlightenment's literature distinguishs the "Bourgeois" on one part, nobility and popular classes on the other part, by their socio-cultural practices and their mentalities. There are a lot of groups of "Bourgeois de Paris" which are different because each gather fellows whose profession, degree of integration in the capital. . . Are not similar. When these men have a common status, they are associated to institutions inherited from middle age (municipality, confraternity of notre-dame for the "Bourgeois de Paris"), or they are integrated in legal or fiscal hierarchies (individual privileges, poll-tax). The theorical definition of these groups can be constant since their creation (the wide corporation of "Bourgeois de Paris" has been defined as inhabitants living in Paris since one year and one day, nobles and roturiers take part of it), il has sometimes changes because of financial needings of monarchy or wish of a social group (tradesman of "six corps" monopolize aldermanship). La caractérisation d’une scène radiologique est un enjeu majeur dans de nombreux domaines, comme la sûreté et la sécurité nucléaire, les problématiques d’assainissement-démantèlement ou encore dans le domaine médical. Une telle analyse consiste à répondre à deux questions principales. D’une part, quels sont les radioéléments présents dans la scène et en quelles quantités ? D’autre part, où se situent ces sources radio-émettrices ? Les systèmes conçus pour répondre à ces questions utilisent l’information portée par les photons X et gamma issus de ces sources radioactives, d’où le nom de « caméra gamma » donné à ces systèmes. Leurs performances dépendent naturellement de celles de l’instrument utilisé, mais aussi des performances des algorithmes de traitement appliqués aux données acquises par l’instrument. Dans le cadre de cette thèse, j’étudie le développement d’un système de caméra gamma, basé sur la technologie Caliste, détecteur CdTe de photons de haute-énergie, issu de plus de quinze années de développement, initialement pour des applications en observation astrophysique. Les performances spectrométriques et en imagerie de ce détecteur sont une référence mondiale dans le domaine des détecteurs CdTe, d’où la pertinence de son application pour des caméras gamma. Plus spécifiquement, mon travail de thèse s’articule en trois axes. Dans un premier temps, à travers une modélisation détaillée du détecteur, j’analyse les données spectrométriques issues de Caliste afin de comprendre comment l’information de la nature et de la quantité des radioéléments sont inscrites dans ces données. Cette modélisation et cette analyse me permettent de mettre en place des algorithmes de traitement des données spectrométriques nouveaux et originaux, basés sur les méthodes de deep learning et de réseaux de neurones convolutifs bayésiens, dans le but de répondre à la question de l’identification et de la quantification des radioéléments tout en apportant une estimation de l’incertitude sur les résultats. Dans un second temps, j’étudie l’utilisation des méthodes d’imagerie à masque codé avec le détecteur Caliste, et en particulier, les algorithmes de localisation des sources radioactives. Je montre l’application et les limites des algorithmes de reconstruction habituellement utilisés pour cette problématique puis je démontre le potentiel des algorithmes de deep learning pour surpasser ces limites. Enfin, je m’intéresse à la problématique de la localisation de points chauds radioactifs par imagerie Compton, à travers des algorithmes de traitement adaptés au détecteur Caliste, en comparant les performances des méthodes issues de l’état de l’art à de nouvelles méthodes que je mets en place. Au long de ce travail de thèse, j’effectue les développements de mes algorithmes à partir de données issues de simulation de sorte à pouvoir transposer ces méthodes à d’autres systèmes de détection. Je prends ensuite le soin de les tester sur des données acquises avec le détecteur Caliste, afin d’obtenir une évaluation de leurs performances, fidèle aux conditions réelles et afin d’être confronté aux aléas des mesures, non pris en compte dans les simulations
The characterization of a radiological scene is a major issue in many fields, such as nuclear safety and security, Decommissioning & Dismantling or in the medical field. Such an analysis consists in answering two main questions. On the one hand, what radioelements are present in the scene and in what quantities? On the other hand, where are these radio-emitting sources located? The systems designed to answer these questions use the information carried by the X and gamma photons from these radioactive sources, hence the name "gamma camera" for these systems. Their performances naturally depend on those of the instrument, but also on the performances of the processing algorithms applied to the data acquired by the instrument. In the framework of this thesis, I study the development of a gamma camera, based on the Caliste technology, a CdTe detector for high-energy photons, which results from more than fifteen years of development, initially for applications in astrophysical observation. The spectrometric and imaging performances of this detector are a world reference in the field of CdTe detectors, hence the relevance of its application for gamma cameras. More specifically, my thesis work explores three axes. First, through a detailed modelling of the detector, I analyse the spectrometric data from Caliste in order to understand how they carry the information of the nature and quantity of radioelements. This modelling and this analysis allow me to develop new and original spectrometric data processing algorithms, based on deep learning methods and Bayesian convolutional neural networks, in order to answer the question of identification and quantification of radioelements while providing an estimation of the uncertainty of the results. In a second step, I study the use of coded mask imaging methods with Caliste, and in particular, the algorithms for locating radioactive sources. I show the application and the limits for the reconstruction provided by the algorithms usually used for this problem and then I demonstrate the potential of deep learning algorithms to overcome these limits. Finally, I analyse the problem of localization of radioactive hot spots by Compton imaging, through processing algorithms adapted to Caliste detectors, by comparing the performances of methods resulting from the state of the art to new methods that I develop. Throughout this thesis work, I develop my algorithms from simulation data in order to transpose these methods to other detection systems. Then, I take care to test them on data acquired with Caliste detectors, in order to obtain an evaluation of their performances, faithful to real conditions and in order to confront these algorithms with the hazards of the measurements, not taken into account in the simulations

Cette thèse explore les formes d’identification adoptées par des antillais fréquentant les lieux de divertissements caribéens en région parisienne. Cela, dans une perspective d’études des questions de genre et diasporiques. Mon hypothèse est que la vie nocturne est un espace culturel encourageant les liens transnationaux et diasporiques. Ma méthodologie a été de fréquenter ces lieux selon les méthodes de l’observation participante et de l’entretien en région parisienne ainsi qu’en Martinique. D’abord, j’ai investigué la géographie humaine des lieux de divertissements antillais en banlieue parisienne en enquêtant sur les lieux d’habitation ainsi que sur l’origine sociale de mes informateurs. Par la suite, j’ai lié les pratiques de la vie nocturne aux phénomènes migratoires des antillais de France. Le fort caractère transnational de ces lieux de divertissement témoigne de constructions diasporiques qui n’ont pas été évoquées auparavant. Néanmoins, ma thèse souligne que ces constructions étaient problématiques pour mes informatrices qui devaient négocier leur sorties avec plusieurs contraintes. La deuxième partie de la thèse se concentre sur le caractère transnational et diasporique du zouk, un genre musical de la Caraïbe française. This thesis explores the forms of identification adopted by French Caribbean clubbers in the Parisian region in relation to the issues of gender and diaspora. My hypothesis is that clubbing is a cultural space that fosters diasporic identities and transnational socialities. Methodologically the thesis is the result of fourteen months of participant observation in Paris and one in Martinique. First I have investigated the human geographies of Antillean clubs in the banlieues of Paris by analyzing in detail the residential patterns and sense of class belonging of my informants. Next I have inscribed the night-time leisure practices in the migration patterns of these informants. I argue that the transnational character of Caribbean nightlife is a testimony to relevant diasporic constructions that have not previously been explored. However my thesis underlines how these constructions were not unproblematic for female participants. The second part of the thesis focuses on the specific transnational and diasporic character of zouk, a French Caribbean music genre. I conclude having investigated issues of gender inequality in clubs and the strategies women employ in order to participate in the dancehall scene

Les mécanismes de dégradation de l'ARN représentent un processus cellulaire central. En effet, ils contrôlent la stabilité et la qualité de l'ARN et, par conséquent, régulent l'expression des gènes. D’une part, la régulation de la stabilité des transcrits est un élément essentiel au maintien de l’homéostasie cellulaire mais aussi à l’établissement d’une réponse cellulaire adaptée en cas d’infection virale. D’autre part, le succès de l’infection virale dépend fortement de la capacité du virus à prendre le contrôle des machineries d’expression géniques cellulaires. De ce fait, les virus doivent interagir avec les machineries cellulaires de dégradation de l’ARN afin de contrôler à la fois, l’expression des gènes cellulaires, et celle des gènes viraux. De nombreuses études rapportent l’existence d’une interface majeure d’interaction entre les machineries eucaryotes de dégradation de l’ARN et les protéines virales. Les virus ont non seulement la capacité d’échapper aux voies cellulaires de dégradation, mais ils peuvent également manipuler ces mécanismes cellulaires de dégradation de l’ARN afin de promouvoir leur propre réplication.Les virus influenza de type A (IAV) sont des agents pathogènes majeurs responsables d'épidémies annuelles et de pandémies occasionnelles. Pour leur cycle de réplication, les IAV dépendent de nombreuses protéines cellulaires et établissent ainsi un vaste et complexe réseau d’interactions avec le protéome cellulaire. Par ailleurs, plusieurs études rapportent l’existence de liens étroits entre les IAV et les machineries de dégradation de l’ARN. Ainsi, identifier les interactions mises en jeu lors du cycle viral participe à une meilleure compréhension du cycle viral, nécessaire au développement de stratégies antivirales. Nous avons recherché des interactions entre les protéines virales impliquées dans la réplication des IAV et un ensemble de 75 protéines cellulaires portant des activités exoribonucléases et/ou associées aux mécanismes de dégradation de l'ARN. Au total, 18 protéines ont été identifiées comme interagissant avec au moins une des protéines virales testées. Par ailleurs, l'analyse du réseau d'interaction a mis en évidence un ciblage spécifique et préférentiel des voies de dégradation de l'ARN par les protéines des IAV. Enfin, parmi les interacteurs validés, un criblage par ARN interférence a identifié neuf facteurs comme étant nécessaires à la multiplication virale.Nous avons choisi de nous concentrer sur l’exoribonucléase 1 (ERI1), identifiée comme interacteur de plusieurs composants des RNPv (RiboNucleoProtéine virale) (PB2, PB1 et NP). ERI1, via ses différents rôles dans l’homéostasie des petits ARN régulateurs, dans la maturation des ARN ribosomiques ou dans la maturation et la dégradation des ARNm histones possède un rôle central dans le contrôle de l’expression génique. En explorant l’interaction entre ERI1 et les protéines virales au cours de l’infection, nous avons mis en évidence que i) ERI1 favorise la transcription virale et que, pour ce faire, ses deux activités - liaison à l’ARN et exonucléase - sont nécessaires, ii) ERI1 interagit avec les protéines virales de manière dépendante de l’ARN, iii) ERI1 interagit avec les RNPv, iv) les protéines virales interagissent avec une forme d'ERI1 associée aux ARNm histones. Ainsi, nos données tendent vers un modèle dans lequel ERI1 associée aux ARNm histones est cooptée par la polymérase virale en transcription, favorisant ainsi la multiplication des IAV par un mécanisme qui reste cependant encore à déterminer. Ainsi, le ciblage de ERI1 par les IAV représente un autre exemple du détournement des machineries de dégradation de l’ARN par les virus, visant à créer un environnement cellulaire favorable à la réplication virale
RNA decay is a central cellular process as it regulates RNA stability and quality and thereby gene expression, which is essential to ensure proper cellular physiology and establishment of adapted responses to viral infection. Global takeover of gene expression machineries and rewiring of the cellular environment is key to the success of viral infection. Cellular proteome and viral replication are tightly connected and cellular RNA processing, stability, quality and decay accordingly influence the fate of the viral cycle. Growing evidence points towards the existence of a large interplay between eukaryotic RNA turnover machineries and viral proteins. Viruses not only evolved mechanisms to evade those RNA degradation pathways, but they also manipulate them to promote viral replication.Influenza A viruses (IAV) are major pathogens responsible for yearly epidemics and occasional pandemics. To complete their viral cycle, IAVs rely on many cellular proteins and establish a complex and highly coordinated interplay with the host proteome. Growing evidence supports the existence of a complex interplay between IAV viral proteins and RNA decay machineries. Unraveling such interplay is therefore essential to gain a better understanding of the IAV life cycle, required for the development of antiviral strategies. This led us to systematically screen interactions between viral proteins involved in IAV replication and a selected set of 75 cellular proteins carrying exoribonucleases activities or associated with RNA decay machineries. A total of 18 proteins were identified as interactors of at least one viral protein tested. Analysis of the interaction network highlighted a specific and preferential targeting of RNA degradation pathways by IAV proteins. Among validated interactors, a targeted RNAi screen identified nine factors as required for viral multiplication. We chose to focus on the 3’-5’ exoribonuclease 1 (ERI1), found in our screen as an interactor of several components of the vRNPs (viral RiboNucleoProtein) (PB2, PB1 and NP). The ERI1 protein is a major player in the control of cellular gene expression as it is essential for the maturation and decay of histone mRNA, maturation of 5.8S rRNA and miRNA homeostasis in mammalian cells. Exploring the interplay between ERI1 and viral proteins during the course of IAV infection we found that i) ERI1 promotes viral transcription, and both of its activities – RNA binding and exonuclease – are required, ii) ERI1 interacts with viral proteins in an RNA dependent manner, iii) ERI1 interacts with the transcribing vRNPs, iv) viral proteins interact with a form of ERI1 that is associated to histone mRNA. Ultimately, our data point to a model where ERI1 associated to histone mRNA is co-opted by the transcribing viral polymerase, thereby promoting IAV multiplication, through a mechanism that remains to be precisely determined. Targeting of ERI1 by IAV is another example further supporting the intricate interplay between IAV and RNA decay machineries, used to rewire cellular gene expression in order to create a favorable environment for viral replication

The Macro Polity introduced the study of elections in context and revealed a systematic interplay of voters and parties over time. Adapting the macro polity model to the multiparty system of Germany, the following papers indicate that dynamic representation is not limited to majoritarian systems. Adapting the concepts to the German system and developing time-series for macropartisanship and policy mood, this thesis presents an integrated dynamic model of the German democratic system: Parties are changing their platforms over time, and these changes are found to be driven by gains and losses in their vote shares in the last election. These changes in the vote distribution are linked to the changing public policy mood. As the electorate prefers more liberal or conservative policies, the change in policy mood is reflected in the vote share of the parties of the political left and right. Changes in public policy preferences are tracked back to fundamental economic forces making the electorate prefer more liberal policies depending on the state of the economy. Linking these single parts together over time results in a model of dynamic representation. The state of the economy and confidence in its future influences the partisan distribution of the public and its taste for liberalism. These influence the vote shares parties receive, which in turn makes parties adapt to the changed times by adapting the policies they offer to voters. This dynamic interplay is found to also work in the German multiparty context.

I en enkätundersökning med 125 deltagare i fyra åldersgrupper undersöktes om unga i större utsträckning än äldre identifierar sig med icke-parlamentariska rörelser som för fram sina budskap på Internet. Hos deltagarna mättes graden av parlamentarisk och icke-parlamentarisk identifikation som funktion av ålder och Internettid. Unga hade en signifikant högre Internettid än äldre. Dock påvisades inga andra signifikanta skillnader mellan åldersgrupperna avseende identifikation. Området kring Internetmediet och dess effekt på politisk identifikation förefaller vara relativt begränsad, varför det skulle vara intressant att utveckla denna forskning i Sverige

Traceability systems are capable of both tracking and tracing parts. They offer many benefits to an organisation from assisting with recall applications to monitoring the everyday workings of a production line or supply chain. There are numerous methods able to act as traceability systems but only a few can be regarded as automatic and unique identifiers. Automatic traceability of individual entities is the future. It is already widely used by a number of leading companies throughout different business sectors and wide mass adoption is imminent. At present, they are slightly more expensive than the simpler technologies but once mass produced, the cost will come down. To completely understand how traceability systems are implemented, practical experience is required. When starting a traceability project, there are a lot of different options. The different systems offer their own set of advantages and some don’t work in certain environments. It was for this reason that The Decision Making Model was developed to assist users through the difficult initial stages of traceability implementation (i.e. choosing the system most suitable to a particular environment). This model was programmed in Excel and supplies the user with a number of questions regarding the environment the system would work in as well as the user’s requirements. The answers to these questions help the user work through the different types of traceability options to eliminate unsuitable choices. The result is an easy to use program designed with the ability to be upgraded as the technologies evolve.

North American bats face multiple threats, prompting an increase in bat research and conservation efforts in recent decades. Researchers often use acoustic monitoring, which entails recording bats? echolocation calls and subsequently identifying them to species, typically using automated software. Chapter 1 describes an acoustic monitoring program at eight U.S. national parks that aims to assess changes in bat populations over time. Data collected in 2016-2017 showed that activity levels of the little brown bat (Myotis lucifigus) decreased significantly while other species remained stable. Little brown bats have undergone similar population declines elsewhere due to the disease white-nose syndrome. Chapter 2 investigates whether different versions of bat call identification software are comparable to each other and how accurate they are. For the two software programs tested, agreement among versions was variable and species-dependent. Furthermore, newer versions were more conservative in assigning identifications, though not, on average, more accurate.

Esta tese foca no problema de estimação de estado e de identificação de parâametros para modelos afins por partes. Modelos afins por partes são obtidos quando o domínio do estado ou da entrada do sistema e particionado em regiões e, para cada região, um submodelo linear ou afim e utilizado para descrever a dinâmica do sistema. Propomos um algoritmo para estimação recursiva de estados e um algoritmo de identificação de parâmetros para uma classe de modelos afins por partes. Propomos um estimador de estados Bayesiano que utiliza o filtro de Kalman em cada um dos submodelos. Neste estimador, a função distribuição cumulativa e utilizada para calcular a distribuição a posteriori do estado assim como a probabilidade de cada submodelo. Já o método de identificação proposto utiliza o algoritmo EM (Expectation Maximization algorithm) para identificar os parâmetros do modelo. A função distribuição cumulativa e utilizada para calcular a probabilidade de cada submodelo a partir da medida do sistema. Em seguida, utilizamos o filtro de Kalman suavizado para estimar o estado e calcular uma função substituta da função likelihood. Tal função e então utilizada para identificar os parâmetros do modelo. O estimador proposto foi utilizado para estimar o estado do modelo não linear para vibrações causadas por folgas. Foram realizadas simulações, onde comparamos o método proposto ao filtro de Kalman estendido e o filtro de partículas. O algoritmo de identificação foi utilizado para identificar os parâmetros do modelo do jato JAS 39 Gripen, assim como, o modelos não linear de vibrações causadas por folgas.
This thesis focuses on the state estimation and parameter identi cation problems of piecewise a ne models. Piecewise a ne models are obtained when the state domain or the input domain are partitioned into regions and, for each region, a linear or a ne submodel is used to describe the system dynamics. We propose a recursive state estimation algorithm and a parameter identi cation algorithm to a class of piecewise a ne models. We propose a Bayesian state estimate which uses the Kalman lter in each submodel. In the this estimator, the cumulative distribution is used to compute the posterior distribution of the state as well as the probability of each submodel. On the other hand, the proposed identi cation method uses the Expectation Maximization (EM) algorithm to identify the model parameters. We use the cumulative distribution to compute the probability of each submodel based on the system measurements. Subsequently, we use the Kalman smoother to estimate the state and compute a surrogate function for the likelihood function. This function is used to estimate the model parameters. The proposed estimator was used to estimate the state of the nonlinear model for vibrations caused by clearances. Numerical simulations were performed, where we have compared the proposed method to the extended Kalman lter and the particle lter. The identi cation algorithm was used to identify the model parameters of the JAS 39 Gripen aircraft as well as the nonlinear model for vibrations caused by clearances.

Finalmente, se concluyó que al mejorar la Productividad del “Picking” (Sacado) y del Embalaje, al mejorar la Identificación y Reconocimiento Visual de los Repuestos y de los Espacios y al realizar una mejor Planificación de la Demanda, un adecuado Control del Inventario, una mejor Planificación del Abastecimiento, se reducirán los Reclamos en el Abastecimiento de Repuestos de productos de línea blanca. The present research is to propose a solution to Reduce Claims in supply of spare parts Products Appliances. To do this, we will mainly apply ABC classification methods, diagrams Analysis Activities, Distribution mix of families Methods demand forecast and other tools of industrial engineering. Participation through a political party is declining globally (Dalton & Wattenberg 2000). Instead, new, more horizontal forms of politics have emerged which undermine the mobilization capacity of parties (Cross & Young 2008). In Latin America, along with it, informality and the establishment of direct links between the leader and his followers are elements that further weaken the identification with a party organization. Nevertheless, parties persist, surviving from their precarious institutionality and adapting their strategies to obtain a base of militants that support the organization. How to understand this decision of participating in a political party in a context that discourages it?This article discusses the incentives for young people to choose to be militants in a Peruvian personalist party. For this, it studies the case of young people on Solidaridad Nacional party in Lima; an organization that, under the leadership of José Luna Gálvez in 2014, implemented a youth recruitment strategy that adapted the militant supply to the various motivations they had as altruist goals, a political career or opportunities for study or work. Thus, the study highlights the use of Diplomados Solidarios in charge of Telesup University for the recruitment and identification of militants, as well as the importance of altruistic motivations and social norms that attracts young people during the process of political socialization.
La participación a través de un partido político está en declive a nivel global (Dalton y Wattenberg 2000). En su lugar, han surgido nuevas formas de hacer política más horizontales, las cuales merman la capacidad de movilización de los partidos (Cross y Young 2008). En América Latina, junto a ello, la informalidad y el establecimiento de vínculos directos entre el líder y sus seguidores son elementos que debilitan aún más la identificación con una organización partidaria. No obstante, los partidos persisten, sobreviven desde su precaria institucionalidad, y adaptan sus estrategias para obtener una base de militantes que sostenga la organización. ¿Cómo entender esta decisión por participar dentro de un partido político en un contexto que la desincentiva?El presente artículo discute los incentivos por los que los jóvenes deciden militar en un partido personalista peruano. Para ello, se estudia el caso de los jóvenes del Partido Solidaridad Nacional en Lima; organización que, bajo el liderazgo de José Luna Gálvez en 2014, implementó una estrategia de captación de jóvenes que adecuó la oferta militante a las diversas motivaciones que poseían los militantes. Enre ellos, figuraban fines altruistas, el interés en una carrera política u oportunidades de estudio o trabajo. Así, resalta el empleo de Diplomados Solidarios a cargo de la Universidad Telesup para el reclutamiento e identificación de militantes, además de motivaciones altruistas y normas sociales que atraen a los jóvenes durante el proceso de socialización política.

La première partie est consacrée à l'exposé du modèle d'équilibre des marchés financiers (CAPM) et de la théorie de l'arbitrage (APT) et à la recherche d'une méthode d'analyse factorielle aux tests empiriques de l'APT. - Dans la seconde partie, nous présentons les résultats des tests du CAPM et de l'APT réalisés à la bourse de Paris. Notre conclusion est qu'au-delà du premier facteur, assimilable au marché, les facteurs sont caractérisés par une forte instabilité temporelle (à court terme) et contemporaine (ils n'ont pas la même signification pour les titres individuels et les portefeuilles). Apres avoir constitué huit indices représentatifs des principaux secteurs économiques, nous avons essayé, via l'analyse factorielle, de mettre en évidence des influences sectorielles sur les rendements des actifs financiers. Nous n'en avons identifié aucune, le facteur de marche expliquant seul la quasi-totalité des rendements moyens sectoriels. - La troisième partie s'intitule : application de la théorie des marchés financiers à la gestion de portefeuille ; nous exposons une méthode de constitution d'indice de marche et de sélection de portefeuille dérivée du CAPM. - Notre conclusion générale ouvre une voie de recherche pour la détermination du meilleur ensemble possible de variables réelles et de leur influence respective sur les rendements des actifs financiers.

Abstract:
This dissertation surveys electoral change in Great Britain during the period between 1979 and 1996. It analyzes the long-term factors and the short-term dynamics underlying the evolution of three aspects of the electorate: party identification, voting intentions and party support in inter-election periods. Drawing on cross-sectional and panel data from the British Election Studies and public opinion polls, I investigate the impacts of long-term socialization and short-term perceptions on voters' political decisions. I hypothesize that, over the last four elections, perceptual factors such as evaluations of party leaders and issues, particularly economic concerns, emerged as the major forces that account for the volatility in electoral behavior in Britain. Accordingly, this study is divided into three sections: Part I probes into the evolution in party identification across age cohorts and social classes as illustrated in trends in partisanship. Part II focuses on changes in voting intentions as affected by perceptual factors and party identification. Part III investigates the public's support for governing parties by analyzing the dynamics of aggregate party support during inter-election periods.

The goal of the creative project was to provide recommendations for renovating the Holliday Park Arboretum based upon existing conditions, tree species, open space analysis and interpretive activities.All trees within the Holliday Park Arboretum were mapped, tagged and identified by species. An Indiana native tree species list was developed and compared to the existing tree species in the arboretum. It was then determined how many native and exotic trees existed in the arboretum. Native tree species not existing in the arboretum were placed on a separate list to establish which native trees were needed to have a representative of each native tree in the arboretum.An open space analysis was conducted to determine potential planting areas within the arboretum and the number of additional native trees that could be planted in those spaces. Recommendations were provided for the selection and planting of native trees as part of the renovation process that can be used by the park administration.
