Abstract:

Abstract The bark of Pausinystalia yohimbe [K. Schumann] Pierre (Rubiaceae), long valued as an aphrodisiac in West Africa, recently has been promoted in the United States as a dietary supplement alternative to anabolic steroids for enhancement of athletic performance. As the number of yohimbe products on the retail market increases, concerns about their safety are raised because of the reported toxicity of yohimbine (the major alkaloid of the plant). Although plant materials are usually identified microscopically, we were unable to identify them in many of the products, because as their labels indicated, the products were mixtures of various bo-tanicals or were bark extracts and contained little or no plant material. A method for extraction and capillary gas chromatographic (GC) separation of the alkaloids of P. yohimbe was, therefore, developed and used to analyze a number of commercial yohimbe products. The method involved solvent extraction and partitioning in chloroform–water followed by separation on a methyl silicone capillary GC column (N–P detection). Comparisons of chromatograms of extracts of authentic bark with those of commercial products indicated that, although many products contained measurable quantities of the alkaloid yohimbine, they were largely devoid of the other alkaloids previously reported in this species. Concentrations of yohimbine in the commercial products ranged from <0.1 to 489 ppm, compared with 7089 ppm in the authentic material. Authentic bark has been reported to contain up to 6% total alkaloids, 10–15% of which are yohimbine. The possible presence of undeclared diluents in the products was indicated by peaks in product chromatograms but not in those of authentic bark.

Abstract:

Many substances of secondary plant metabolism have often attracted the attention of scientists and the public because they have certain beneficial effects on human health, although the reason for their biosynthesis in the plant remains unclear. This is also the case for alkaloids. More than 200 years have passed since the discovery of the first alkaloid (morphine), and several thousand substances of this character have been isolated since then. Most often, alkaloid-rich plants are part of folk medicine with centuries-old traditions. What is particularly important to monitor for these herbal products is the spectrum and concentrations of the present active substances, which decide whether the product has a beneficial or toxic effect on human health. In this work, we present a fast, reliable, and robust method for the extraction, preconcentration, and determination of four selected alkaloids with an indole skeleton, i.e., harmine, harmaline, yohimbine, and ajmalicine, by ultra-high performance liquid chromatography coupled with tandem mass spectrometry. The applicability of the method was demonstrated for tobacco and Tribulus terrestris plant tissue, the seeds of Peganum harmala, and extract from the bark of the African tree Pausinystalia johimbe.

Abstract:

Background: The roots of Pausinystalia yohimbe have been traditionally acclaimed as aphrodisiac. In the present study, methanol extract of the root was evaluated for its effect on sexual orientation behaviour and spermatogenesis in albino Wistar rats. Methods: Forty five (45) male albino rats were randomly divided into five groups of nine rats each. Rats in group I (control) were administered 1 mL/kg body weight distilled water (vehicle), group II received 5 mg/kg body weight sildenafil citrate (Viagra), while those in groups III, IV, and V were given 25, 50, and 100 mg/kg body weight, respectively, of methanol extract of Pausinystalia yohimbe root in the same volume. Female albino rats were made receptive by hormonal treatment. Sexual orientation behaviour parameters were monitored on days 1, 7 and 14 by pairing with receptive females. A change in sexual orientation behaviuor was assessed by orientation towards female, towards environment and towards self, while spermatogenic activity was evaluated on day 14. Results: Administration of the extract had pronounced effect on sexual orientation behaviour of male towards the female rats on days 7 and 14. Males treated with the extract displayed more frequent and vigorous anogenital sniffing and mounting as compared to untreated animals. Libido was also at increase in extract treated male rats. The extract had stimulated the spermatogenetic activity and accessory reproductive organs performance in albino rats. The increased spermatogenesis in extract treated groups was confirmed by change in histoarchitecture as evidenced by increase in number of spermatogenic elements and parameters. Conclusion: P. yohimbe exhibited remarkable increase in sexual orientation, libido and spermatogenic activity which are some of the indices that determine the ability of a male to produce viable spermatozoa. These findings support the folk use of this plant as an aphrodisiac.
