Abstract:
Analysoimme artikkelissamme trollauskampanjaa, joka kohdistui nykytaiteen museo Kiasman ARS17-näyttelyssä esillä olleeseen #ALONETOGETHER-taideteokseen keväällä 2017. Poikkeuksellisen suuria tunteita puolesta ja vastaan herättäneen teoksen luona nähtiin kevään aikana innostuneita museokävijöitä, mutta myös teoksen hengen ja normatiivisen galleriakontekstin ulkopuolelle tarkoituksella jättäytyneitä osallistujia. Taideteoksen luona Kiasmassa vieraili useita ihmisiä, jotka pyrkivät häiritsemään pääasiassa yhtä taiteilijoista, Shia LaBeoufia, jonka kanssa pystyi kommunikoimaan teoksen välityksellä. Lisäksi häirintä näkyi verkossa, osin teoksen verkottuneen muodon ja osin trollien verkkokulttuurisen taustan vuoksi. Suomalaiselta Ylilauta-kuvalaudalta sekä sen kansainväliseltä vastineelta 4chanilta käynnistynyt häirintä kytkeytyi osaksi pidempää teoksen taiteilijoiden ja trollien välistä historiaa, jota tarkastelemme sekä transnationaalisen verkkokulttuurin ilmentymänä että yhdysvaltalaisen äärioikeistolaisen poliittisen alt-right-diskurssin suomalaistuneena versiona. Häirintään käytettiin kuvalautakulttuurille ominaista sanastoa ja kuvastoa, joiden eräänlaisena tiivistymänä voidaan pitää Pepe-sammakkoa, sarjakuvamaista symbolia, johon kiinnittyy trollien poliittisia ja retorisia tavoitteita ja joka toisaalta ilmentää myös trollaukselle ominaista trivialisoivaa lähestymistapaa. Analyysimme keskittyy yhtäältä trollauksen ristiriitaiseen rooliin poliittisesti motivoituneena toimintana ja leikkinä, ja toisaalta yleisöä osallistavan taideteoksen kokemiseen leikillisenä tapahtumana. Tarkastelemme myös, miten taiteilijat, trollit ja museokävijät pyrkivät sääntelemään, kaappaamaan ja muokkaamaan teoksen affektiivisia ulottuvuuksia yrittämällä hallita teoksen leikillistä tilaa. Osoitamme, miten trollauksen ymmärtäminen leikkinä auttaa ymmärtämään tapahtumia erityisesti #ALONETOGETHER-taideteoksen yhteydessä, ja miten trollaus voi olla samaan aikaan sekä leikkisää että poliittista.

Teknik pengumpulan data yang digunakan adalah pengamatan, wawancara, catat, dan dokumentasi. Berdasarkan data yang diperoleh menunjukkan bahwa; (1) Tarian Pepe-Pepeka Ri Makka berasal dari dari pengaruh budaya Arab yang masuk ke wilayah Makassar dalam menyebarkan agama Islam. (2) Fase pertunjukan tarian Pepe-Pepeka Ri Makka adalah ragam ridoangnga (ragam berdoa), karenanna(religi permainan), dan ju’julu sulona-Ritongko’ (memberi cahaya api dan ucapan syukur). (3) Fungsi pertunjukan tarian Pepe-pepeka adalah sebagai ritual dalam penyebaran agama Islam pada perkembangan terakhir berfungsi sebagai hiburan. ; (4) terdapat beberapa nilai-nilai budaya yang tergambar dari tarian Pepe-Pepeka Ri Makka yaitu nilai filosofis yang terkait dengan konsep pada masyarakat Makassar yaitu salah satu mitologi appak sulapa (empat persegi) yakni , tana (tanah), jekne (air), angin (angin), dan Pepe (api). Dalam syair tari Pepe-pepeka ri Makka, makna yang terkandung di dalamnya sangat dalam terkait dengan penyebaranagama Islam.

Abstract:
Reports indicate that either temporary calf removal or equine chorionic gonadotropin administration can increase the efficiency (pregnancy rate) of hormonal treatments with progestins during postpartum anestrus. This experiment evaluated effects of TCR and/or eCG administration in a protocol with progesterone that is frequently used for fixed-time artificial insemination (FTAI) in cows during postpartum anestrus. The protocols were tested at three farms in lactating Nelore cows (40 to 70 days post-partum, n = 361) with body condition scores from 2.5 to 3.0 (0- to 5-point scale). At a random stage of the estrous cycle (Day 0), animals received a basic PEPE (progesterone-estrogen-prostaglandin-estrogen) protocol with insertion of an intravaginal device with 1.0 g progesterone (DIB�, Syntex, Buenos Aires, Argentina) and an i.m. injection of 2.5 mg of estradiol benzoate (EB, Estrogin�, Farmavet, S�o Paulo, Brazil). Eight days later (Day 8) cows were treated i.m. with 150 mg D-cloprostenol (PGF2� Prolise�, ARSA S.R.L., Buenos Aires, Argentina), and the DIB was removed. Twenty-four hours after DIB removal, cows received 1.0 mg EB i.m. and 30 to 36 h later all animals were FTAIed without estrus detection. Cows were allocated randomly to four groups: PEPE, PEPE/TCR, PEPE/eCG, and PEPE/TCR/eCG. In Group PEPE/TCR, calves were removed temporarily for 54 h (from DIB removal until FTAI). In Group PEPE/eCG, animals received PEPE treatment plus one dose i.m. of 300 UI eCG (Novormon�, Syntex) following PGF2� administration (Day 8). In Group PEPE/TCR/eCG, animals were treated as in protocol PEPE/TCR plus eCG on D8. All animals were examined by ultrasonography (Aloka SSD 500, 7.5 MHz probe) 10 days before and at the beginning of hormonal treatment in order to detect anestrous cows (absence of corpus lutcum in both exams). Pregnancy diagnosis was performed by ultrasonography 30 days after FTAI. The data were analyzed by logistic regression. The following variables were considered in the model and did not affect pregnancy rates: farms, inseminators, and semen (sire). A total of 75% of the animals were in anestrus (absence of CL) and the pregnancy rates were similar (P > 0.05) among the four groups: PEPE (43/85 = 50.6%), PEPE/TCR (42/98 = 42.9%), PEPE/eCG (41/88 = 46%), and PEPE/TCR/eCG (39/90 = 43.3%). The results indicate that in Nelore cows, in postpartum anestrus and good body condition, TCR and/or eCG administration do not improve the efficiency (pregnancy rate) of the PEPE protocol. Abstract:
The purpose of this study was to explore the hydrolytic ability of Lactobacillus helveticus CICC 22171 with regard to protein and the expression of enzyme genes during protein utilization. The results revealed that the strain hydrolyzed casein from the C-terminal, reached the maximum level in 6 h, and the number of amino acids in the hydrolyzed peptide was 7–33. The molecular weight was 652.4–3432.74 kDa. Hydrophobic peptides produced by hydrolysis were the source of β-casein bitterness. Leucine and glutamine were the preferred cleavage points after 1 h; tyrosine and tryptophan subsequently increased. The first step of hydrolysis was controlled by PrtP and PrtM genes and coordinated with the action of PrtH1 and PrtH2. The transport system consisted of DtpT, OppB, OppD and OppF. The hydrolytic third step endopeptidase system consisted of the aminopeptidases (PepN, PepC, PepM and PepA), the endopeptidases (PepE, PepF and PepO); the dipeptidases (PepV and PepD), the tripeptidase PepT; the proline peptidases (PepX, PepP, PepQ, PepR and PepI). The expression of CEP genes was significantly different, and the expression level of genes related to the transport system significantly increased from 0 to 1 h. The specificity of the substrate and action site of endopeptidase was abundant.

ABSTRACT Genes encoding three putative endopeptidases were identified from a draft-quality genome sequence of Lactobacillus helveticus CNRZ32 and designated pepO3, pepF, and pepE2. The ability of cell extracts from Escherichia coli DH5α derivatives expressing CNRZ32 endopeptidases PepE, PepE2, PepF, PepO, PepO2, and PepO3 to hydrolyze the model bitter peptides, β-casein (β-CN) (f193-209) and αS1-casein (αS1-CN) (f1-9), under cheese-ripening conditions (pH 5.1, 4% NaCl, and 10°C) was examined. CNRZ32 PepO3 was determined to be a functional paralog of PepO2 and hydrolyzed both peptides, while PepE and PepF had unique specificities towards αS1-CN (f1-9) and β-CN (f193-209), respectively. CNRZ32 PepE2 and PepO did not hydrolyze either peptide under these conditions. To demonstrate the utility of these peptidases in cheese, PepE, PepO2, and PepO3 were expressed in Lactococcus lactis, a common cheese starter, using a high-copy vector pTRKH2 and under the control of the pepO3 promoter. Cell extracts of L. lactis derivatives expressing these peptidases were used to hydrolyze β-CN (f193-209) and αS1-CN (f1-9) under cheese-ripening conditions in single-peptide reactions, in a defined peptide mix, and in Cheddar cheese serum. Peptides αS1-CN (f1-9), αS1-CN (f1-13), and αS1-CN (f1-16) were identified from Cheddar cheese serum and included in the defined peptide mix. Our results demonstrate that in all systems examined, PepO2 and PepO3 had the highest activity with β-CN (f193-209) and αS1-CN (f1-9). Cheese-derived peptides were observed to affect the activity of some of the enzymes examined, underscoring the importance of incorporating such peptides in model systems. These data indicate that L. helveticus CNRZ32 endopeptidases PepO2 and PepO3 are likely to play a key role in this strain's ability to reduce bitterness in cheese.

Fragestellung: Die vorliegende Studie überprüft die Wirksamkeit des Musikalischen Konzentrationstrainings mit Pepe (MusiKo mit Pepe) für fünf- bis zehnjährige Kinder mit Aufmerksamkeitsproblemen. Methodik: In einem Prä-Post-Kontrollgruppendesign (N = 108) wurden Veränderungen der Aufmerksamkeitsleistung mittels der Testbatterie zur Aufmerksamkeitsprüfung für Kinder (KiTAP) sowie Veränderungen der kindlichen Lebensqualität mittels des Fragebogens für Kinder (KINDL-R) erfasst. Zusätzlich wurden Fremdbeurteilungsbögen zur Aufmerksamkeitsdefizit-/Hyperaktivitätsstörung (FBB-ADHS) sowie zur Störung des Sozialverhaltens (FBB-SSV) des Diagnostik-Systems für psychische Störungen nach ICD-10 und DSM-IV für Kinder und Jugendliche II und der Eltern- und der Lehrerfragebogen über das Verhalten von Kindern und Jugendlichen (CBCL, TRF) eingesetzt. Ergebnisse: Es zeigen sich für die am Training teilnehmenden Kinder im Vergleich zu der Kontrollgruppe über die Zeit signifikante Verbesserungen der Aufmerksamkeitsleistung sowie der Lebensqualität. Darüber hinaus ergibt sich eine signifikante Reduktion der ADHS-Symptomatik im Eltern- und Lehrerurteil sowie eine Verminderung der Internalisierenden Probleme im Elternurteil. Die Behandlungseffektivität ist unabhängig von Alter, Geschlecht, Intelligenz und Migrationshintergrund der teilnehmenden Kinder. Schlussfolgerung: Das musikbasierte Trainingsprogramm MusiKo mit Pepe stellt eine wirkungsvolle Maßnahme zur Behandlung von Aufmerksamkeitsproblemen dar, sollten sich diese Effekte in Replikationsstudien bestätigen.

Tapa Pepe merupakan sebuah metode penyatuan antara Atman dengan Brahman melalui metode meditasi dengan memusatkan kepada sumber matahari. Tapa Pepe memfokuskan konsentrasinya pada “Sapta Cakra” yang ada dalam tubuh manusia. Untuk mengaktifkan cakra-cakra tersebut dengan tujuan mendekatkan diri dengan Tuhan. Ketika cakra-cakra dalam tubuh manusia terbuka, maka manusia akan banyak mendapatkan manfaat baik secara medis dan non medis, sekala dan niskala. Tapa Pepe merupakan tradisi Hindu yang telah dilaksanakan para leluhur pendahulu kita di tanah Jawa sebagai cara mendekatkan diri dalam proses menyatu dengan alam yang tiada lain adalah Tuhan itu sendiri sesuai dengan tujuan agama Hindu yaitu “mokshartam jagadhita ya ca iti dharma” dimana proses penyatuan diri seseorang melalui laku tertentu ibarat seperti seseorang melaksanakan laku upawasa (puasa) yang juga secara otomatis juga menyucikan dirinya sebagai jalan kepada Tuhan. Ritual Tapa Pepe yang dilakukan oleh Umat Hindu Jawa di Pura Mandira Seta Kraton Surakarta tidak jauh berbeda dengan ritual tapa pepe dibeberapa tempat. Perbedaanya adalah tempat pelaksanaannya dimana pada umumnya dilakukan di tempat terbuka, halaman rumah dan lain-lain. Berbeda dengan Tapa Pepe yang berada di Pura Mandira Seta dilakukan di dalam areal pura, dimana laku tapa pepe ini dilakukan di pelataran mandala tengah pura atau madya mandala, tepatnya di pelataran Arca Bhatari Dhurga Mahisasura Mardini. Tujuan dari Tapa Pepe di Pura Mandira Seta yaitu menyatukan Atman dengan Brahman yang berada dalam diri melalui metode menatap matahari sebagai sarana konsentrasinya. Konsentrasi pada Tapa Pepe atau Yoga Matahari berpusat pada cakra-cakra yang ada dalam tubuh. Tujuh cakra itu yaitu : Cakra Sahasrara (Cakra Mahkota), Ajna Cakra (Cakra Mata Ketiga/kening), Wisudha Cakra (Cakra Tenggorokan), Anahata Cakra (Cakra Jantung), Manipura Cakra (Cakra Pusar), Swadistana Cakra (Cakra Kelamin), Muladara Cakra (Cakra Tulang Ekor). Ketika cakra-cakra itu aktif, maka seseorang yang melakukan Tapa Pepe akan mendapatkan banyak manfaat seperti tidak mudah sakit karena menyerap energi matahari terbukti sekarang dilakukan banyak orang dimasa pendemi covid 19 ini.

Abstract:
Background/Aims: Peperomin E (PepE), a natural secolignan isolated from the whole plant of Peperomia dindygulensis, has been reported by ourselves and others to display potent anti-cancer effects in many types cancer cells, especially gastric cancer. However, the effects of PepE on the metastasis of poorly-differentiated gastric cancer cells and the underlying molecular mechanisms have not been well elucidated. Methods: We evaluated PepE effects on gastric cancer cell invasion and migration in vitro via wound healing and transwell assays and those on growth and metastasis in vivo using an orthotopic xenograft NOD-SCID mouse model. DNA methyltransferase (DNMT) activity was determined using a colorimetric DNMT activity/inhibition assay kit. PepE binding kinetics to DNMTs were determined using the bio-layer interferometry binding assay. Gene and protein levels of DNMTs, AMPKα-Sp1 signaling molecules, and metastatic-suppressor genes in PepE-treated gastric cancer cells were determined using quantitative reverse transcription-PCR arrays and western blotting. The effect of PepE on Sp1 binding to the DNMT promoter was determined by electrophoretic mobility-shift assay. Global DNA methylation levels were determined using liquid chromatography coupled with electrospray ionization tandem mass spectrometry. The methylation status of silenced metastatic-suppressor genes (MSGs) in gastric cancer cells was investigated by methylation-specific PCR. Results: PepE can dose-dependently suppress invasion and migration of poorly-differentiated gastric cancer cells in vitro and in vivo with low toxicity against normal cells. Mechanistically, PepE not only covalently binds to the catalytic domain of DNMT1 and inhibits its activity (IC50 value 3.61 μM) but also down-regulates DNMT1, 3a, and 3b mRNA and protein expression in in gastric cancer cells, by disruption of the physical interaction of Sp1 with the DNMT1, 3a, and 3b promoter and mediation of the AMPKα-Sp1 signaling pathway. The dual inhibition activity of PepE toward DNMTs renders a relative global DNA hypomethylation, which induces MSG promoter hypomethylation (e.g., E-cadherin and TIMP3) and enhances their expression in gastric cancer cells. Conclusion: Collectively, our data indicated that PepE may represent a promising therapeutic lead compound for intervention in gastric cancer metastasis and may also exhibit potential as a DNA methylation inhibitor for use in epigenetic cancer therapy.

Data dianalisis dengan menggunakan teknik deskriptif kualitatif dimana data yang diperoleh akan dianalisa menggunakan analisis SWOT. Hasil penelitian ini menunjukkan bahwa Dusun Pepe memiliki potensi yang rendah dimana dapat dikatakan jika desa tersebut tidak memiliki potensi untuk dijadikan sebagai desa wisata dan selanjutnya dapat dilakukan analisis mengenai usaha pengembangan potensi desa wisata. Kajian usaha pengembangan potensi desa wisata berdasarkan analisis SWOT yaitu: a). Kekuatan yang cukup besar berupa potensi alam. b). Kelemahan potensi wisata Dusun Pepe yaitu beberapa sarana yang belum mendukung dan belum adanya kesenian atau atraksi yang dapat ditampilkan. c). Dusun Pepe belum memiliki prospek dan cinderamata yang mendukung sehingga belum memiliki peluang untuk dijadikan desa wisata. d.) Berdasarkan aspek ancaman, di Dusun Pepe belum ada obyek yang bisa dijadikan sebagai tempat wisata sehingga tidak ada ancaman. Dengan demikian maka dapat diketahui bahwa Dusun Pepe memiliki potensi yang sangat rendah untuk dijadikan sebagai desa wisata.

Der Aufmerksamkeitsdefizit-/Hyperaktivitätsstörung wird in der Forschung zusehends mehr Bedeutung beigemessen, da sie mit einer Prävalenz von 4,8 % zu den häufigsten psychischen Erkrankungen im Kindes- und Jugendalter zählt (KiGGS; Schlack, Hölling, Kurth & Huss, 2007 ). Zudem wächst der Bedarf an alternativen Behandlungsmöglichkeiten neben der Pharmakotherapie. Das hier vorgestellte Musikalische Konzentrationstraining mit Pepe stellt ein innovatives Gruppenprogramm für fünf- bis zehnjährige Kinder mit Konzentrationsproblemen dar. Angelehnt an verhaltenstherapeutische Grundlagen nutzt es Musik als Lern- und Lehrmittel, um die Neugier, Offenheit und Kreativität sowie den Bewegungsdrang der Kinder sinnvoll zu nutzen. Die strukturierten Rhythmus- und Bewegungsübungen zielen auf eine Förderung der fokussierten, geteilten und Daueraufmerksamkeit ab. Weitere Elemente des Trainings wie die Teamarbeit, die Leitfigur Pepe und seine Ampel sollen das Sozialverhalten, die Impulskontrolle sowie die Eigenverantwortlichkeit der Kinder schulen. Die Besonderheiten des Musikalischen Konzentrationstrainings mit Pepe sowie dessen Wirkfaktoren werden im vorliegenden Beitrag präsentiert.

This article conceptualises Pepe Meamea (Samoan concept of infant/toddler) to ground Samoan cultural understandings, values and beliefs of the Samoan child as a collective, holistic and relational being. The Samoan worldview of children and childhood prioritises the whole child, and this has implications when positioned within a dominant Eurocentric early childhood curriculum (ECE) curriculum and pedagogy framework. The semiotics of the Samoan language references a collective position for personhood rather than an individual one which warrants further examination into Samoan conceptualisations of Pepe Meamea pushing back at normative assumptions. As an innovative practice and philosophy for teaching, Pepe Meamea already has deep Samoan cultural knowledges associated with its term. Pepe Meamea is understood and commonly referred to as infants or newborns. This presents initial talanoa (dialogue) with teachers from six Aoga Amata (Samoan early childhood centres) in a two-year research Teaching and Learning Research Initiative (TLRI) project (2021‐2022). Using Samoan language as the conduit for creation and cultural reference ensures Samoan children’s cultural rights are adhered to through an Indigenous Samoan paradigm. As an innovative practice and concept, Pepe Meamea already has deep cultural Indigenous knowledge associated with its term; however, this article presents the possibilities of Pepe Meamea through the development of a pedagogical and relational framework that promotes its implementation in the Aotearoa New Zealand ECE environment. How the New Zealand Te Whāriki early childhood education (ECE) curriculum is responsive to Pepe Meamea is another dimension this paper will consider as the majority of the New Zealand Samoan population are born and raised in a New Zealand context. This research paper reports on the first phase of the project, the process of co-designing a Pepe Meamea pedagogical framework with Samoan teachers.

En la región norte se cultivan variedades criollas en huertos comerciales y en traspatios, predominando la utilización de las variedades Pepe y Negro Santos (variedades locales). Estas variedades se caracterizan por presentar un ciclo intermedio y tardío respectivamente de acuerdo a la época de cosecha, lo cual limita que la producción en esa región se concentre en un período relativamente corto. El objetivo de esta investigación fue caracterizar un grupo de variedades criollas de aguacate en la región norte del estado de Nuevo León mediante caracteres morfológicos de los frutos. Se utilizaron 19 variedades incluyendo las variedades Pepe y Negro Santos (testigos locales), presentes en dos huertas comerciales en los municipios de Sabinas Hidalgo y Bustamante, Nuevo León. Se determinaron sietematerialesconcaracterísticasmorfológicassimilares que las variedades Pepe y Negro Santos, las cuales se clasificaron en cuatro grupos. El primer grupo está formado por una variedad con características del fruto similares a la variedad Pepe y período temprano en la época de cosecha. El segundo grupo está integrado por cuatro variedades con características del fruto similares a las variedades Pepe y Negro Santos y período intermedio en la época de cosecha. El tercer grupo está constituido por una variedad con características del fruto diferente a las variedades Pepe y Negro Santos, período intermedio a la época de cosecha y los frutos son de color verde. El cuarto grupo está integrado por una variedad con características del fruto similares a las variedades Pepe y Negro Santos, con un período tardío a la época de cosecha. Los resultados obtenidos sugieren que existe una amplia variación en los materiales criollos de aguacate cultivados en la región norte del estado de Nuevo León, lo cual se constituye como una alternativa para diversificar la producción y no depender exclusivamente de las variedades Pepe y Negro Santos.

Kali Pepe is the witness of history where culture and trade activities in the rapidly growing city of Surakarta in the past with the ecological function and physical function as transportation trade.Setuping Kali Pepe, according to the Mayor of Surakarta, is directed to serve as recreation/tourism area. Since the Surakarta Mayor initiated the year 2015 that Kali Pepe as a tourist area. The initiated moves the government and society in order to more actively participate in developing the area into a tourist area. This research would like to know how the readiness level of the Kali Pepe area to be developed as a tourist area-based streams. The components of preparedness were seen from aspect of attractions or natural tourist attraction, artificial attractions, acessesiblity, institutional, infrastructure supporting tourism, and the behavior of the flooding of the river. This research is quantitative research in methods of scoring analysis. The result of this research has shown that Kali Pepe less readiness to be developed as a tourist area-based stream. Aspects of accessibility and infrastructure supporting tourism were an aspect which has a readiness. ABSTRACT Salmonella enterica serovar Typhimurium peptidase E (PepE) is an N-terminal Asp-specific dipeptidase. PepE is not inhibited by any of the classical peptidase inhibitors, and its amino acid sequence does not place it in any of the known peptidase structural classes. A comparison of the amino acid sequence of PepE with a number of related sequences has allowed us to define the amino acid residues that are strongly conserved in this family. To ensure the validity of this comparison, we have expressed one of the most distantly related relatives (Xenopus) in Escherichia coli and have shown that it is indeed an Asp-specific dipeptidase with properties very similar to those of serovar Typhimurium PepE. The sequence comparison suggests that PepE is a serine hydrolase. We have used site-directed mutagenesis to change all of the conserved Ser, His, and Asp residues and have found that Ser120, His157, and Asp135 are all required for activity. Conversion of Ser120 to Cys leads to severely reduced (104-fold) but still detectable activity, and this activity but not that of the parent is inhibited by thiol reagents; these results confirm that this residue is likely to be the catalytic nucleophile. These results suggest that PepE is the prototype of a new family of serine peptidases. The phylogenetic distribution of the family is unusual, since representatives are found in eubacteria, an insect (Drosophila), and a vertebrate (Xenopus) but not in the Archaea or in any of the other eukaryotes for which genome sequences are available.

Penelitian ini bertujuan untuk mengetahui prioritas faktor dalam penentuan pola penanganan permukiman kumuh di bantaran Kali Pepe Kota Surakarta. Penanganan permukiman kumuh di bantaran Kali Pepe dilakukan dengan pola pemugaran, peremajaan, dan permukiman kembali. Pendekatan yang digunakan pada penelitian ini adalah pendekatan deduktif. Instrumen survey yang digunakan berupa kuesioner kepada <em>stakeholder </em>terkait penanganan permukiman kumuh bantaran Kali Pepe. Ada delapan faktor dalam pola penanganan permukiman kumuh di bantaran Kali Pepe Kota Surakarta yaitu status legalitas lahan, tingkat kekumuhan, SK Walikota tentang lokasi kumuh, ketersediaan anggaran, luasan kumuh yang ditangani, ketersediaan lahan, peran masyarakat, dan struktur kelembagaan. Teknik analisis yang digunakan untuk mengetahui prioritas dari faktor-faktor tersebut adalah analisis AHP. Berdasarkan hasil analisis AHP, menunjukkan prioritas faktor yang berbeda di masing-masing pola penanganan. Prioritas pada pola pemugaran adalah faktor luasan kumuh yang ditangani, sedangkan prioritas pada pola peremajaan adalah faktor SK Walikota tentang lokasi kumuh, serta prioritas pada pola permukiman kembali adalah faktor status legalitas lahan.</p>

Abstract Mutations that cause loss of acidity in the vacuole (lysosome) of Saccharomyces cerevisiae were identified by screening colonies labeled with the fluorescent, pH-sensitive, vacuolar labeling agent, 6-carboxyfluorescein. Thirty nine vacuolar pH (Vph-) mutants were identified. Four of these contained mutant alleles of the previously described PEP3, PEP5, PEP6 and PEP7 genes. The remaining mutants defined eight complementation groups of vph mutations. No alleles of the VAT2 or TFP1 genes (known to encode subunits of the vacuolar H(+)-ATPase) were identified in the Vph- screen. Strains bearing mutations in any of six of the VPH genes failed to grow on medium buffered at neutral pH; otherwise, none of the vph mutations caused notable growth inhibition on standard yeast media. Expression of the vacuolar protease, carboxypeptidase Y, was defective in strains bearing vph4 mutations but was apparently normal in strains bearing any of the other vph mutations. Defects in vacuolar morphology at the light microscope level were evident in all Vph- mutants. Strains that contained representative mutant alleles of the 17 previously described PEP genes were assayed for vacuolar pH; mutations in seven of the PEP genes (including PEP3, PEP5, PEP6 and PEP7) caused loss of vacuolar acidity.

In this study, the aromatic acetylene compound 4-(phenylethynyl) di(ethylene glycol) phthalate (PEPE) was used as a chain extender, partially replacing 1,4-butanediol. To synthesize an intrinsic flame-retardant thermoplastic polyurethane elastomer (TPU) with an aromatic acetylene structure, PEPE was synthesized by a two-step polymerization. The flame retardancy, thermal stability, and mechanical properties of TPU were studied. The microstructure of TPU char was investigated by scanning electron microscopy to analyze the flame-retardant mechanism. The tensile strength of TPU containing 1.35 wt% PEPE was 39.2 MPa, which was almost twice as much as neat TPU, showed a dramatic decrease in the peak heat release rate and total heat release, and declined by 46.2% and 24.5%, respectively. After the flame-retardant TPU burned, a cross-linked network foaming char structure was formed. The results showed that PEPE improved the mechanical properties of TPU and conferred good stability that promoted the formation of charcoal and reduced heat release during the combustion of TPU.

The antigenic properties of the Zn2+-binding region of two Zn2+-metalloproteases, Leishmania surface protease gp63 and mammalian endopeptidase-24.11 (E-24.11), possessing in their active site the characteristic amino acid sequence HEXXH, were investigated by using oligoclonal antibodies raised against two synthetic peptides, V1VTHEMAHALG11 (pepgp63) and V1IGHEITHGFD11 (pepE-24.11), containing the respective Zn2+-binding sites of the cognate protein. The affinity-purified antibodies, tested on synthetic peptides modelled from the active sites of ten different Zn2+-metalloproteases, showed high selectivity for their respective peptides. However, cross-reactivity was revealed when the antibodies were tested against the gp63 and E-24.11 molecules. A panel of synthetic peptide analogues and peptides of various size was synthesized and used for the fine antigenic characterization of pepgp63 and pepE-24.11. The shortest peptides capable of significant antibody binding were the pentapeptides V1VTHE5 and E5ITHG9 for pepgp63 and pepE-24.11 respectively. His4 and Glu5 were found to be indispensable for anti-pepgp63 binding to pepgp63, whereas in the case of pepE-24.11, Glu5 and His8 were found to be critical. The conformational characteristics of the two peptides correlate well with the observed differences in their antigenicity. 1H-NMR studies showed that pepgp63 adopts a folded structure whereas pepE-24.11 takes up a rather flexible conformation. Moreover, the antigenically critical His4 of pepgp63 contributes to the structural stabilization of the peptide. Similarly, the antigenically critical His8 of pepE-24.11 is involved in partial structural stabilization of its C-terminal region. The generated antibodies may be useful tools for identifying and classifying proteins possessing similar Zn2+-binding motifs and/or environments.

Namun kualitas air Sungai tersebut termasuk dalam golongan IV atau tidak sesuai dengan peruntukannya. Limbah domestik dan industri UKM yang masuk ke sungai serta rendahnya baseflow sungai menjadi penyebabnya. Penelitian ini bertujuan untuk memodelkan kualitas air Sungai Pepe dengan melakukan variasi suplesi debit dari Sungai Anyar sehingga kualitas air Sungai Pepe menjadi lebih baik. Simulasi dilakukan dengan menggunakan model Qual2Kw. Pengukuran kualitas air eksisting dilakukan pada 6 titik pengambilan sampel. Variasi debit suplesi yang digunakan meliputi kondisi eksisting, suplesi 0,5 m3/s dan 1 m3/s. Debit suplesi ke Sungai Pepe Hilir dapat di atur melalui pintu air di Sungai Anyar. Hasil penelitian menunjukkan bahwa kualitas air secara umum menjadi lebih baik sepanjang ruas sungai. Parameter DO dan COD membaik menunjukkan kemampuan areasi yang membaik akibat hidrodinamika aliran sungai. Adanya pencampuran limbah tekstil pada anak sungai masih menjadi parameter yang dominan dalam penurunan kualitas air sungai.

Hasil penelitian ini menunjukkan bahwa prosentase ketuntasan belajar peserta didik kelas IV-C SDN Pepe mengalami kenaikan pada pra siklus prosentasenya adalah 33,33%, pada siklus I naik menjadi 51,72 % dan siklus II menjadi 72,41 %, untuk itu penelitian ini dnyatakan berhasil. Kesimpulan penelitian ini adalah melalui model pembelajaran Kooperatif TGT dapat meningkatkan hasil belajar Matematika pada materi Faktor Persekutuan di kelas IV-C SDN Pepe.

Abstract:
The poems of Pepe Sales (Barcelona, 1954-1994) are marked by his experience as a homosexual, drug addict and AIDS sufferer. He was a key member of the Barcelona counterculture, along with Pau Riba, Pau Malvido and Genís Cano, and has gone down in history as one of the poètes maudits of contemporary Catalan literature. The aim of this article is to analyse the representation of pain and distress in his poems and songs, with some additional references to his paintings. To this end, I draw on the critical work of Susan Sontag, Sara Ahmed, David Le Breton and Joanna Bourke, who conceive pain in general, not as a private and personal experience, but as a cultural and historical event that creates identity and community bonds. The main strategies used by Sales to convey his pain are the scream, animalisation, weapons and wounds and, especially, religious martyrdom. The basic finding is that the poet’s pain is not simply a testimony to the suffering of a lost generation of junkies and misfits that the official account of the transition from Francoism to democracy has stigmatised. On the contrary, it reminds us of an unfair medical, political and financial system that inflicted great suffering on them.

Abstract This study aims to establish an analog prediction model for forecasting daily persistent extreme precipitation (PEP) during a PEP event (PEPE) using the temporal sequences of predictors with different weights applied in the atmospheric spatial field. The predictors are atmospheric variables in areas where the key influential systems of a PEPE are active in the THORPEX Interactive Grand Global Ensemble (TIGGE) dataset. By means of the cosine similarity measure and the cuckoo search technique, a forecast model was established and named the Key Influential Systems Based Analog Model (KISAM). Validations through threat scores (TSs) and root-mean-square errors for PEP during 17–25 June 2010 indicate that KISAM is able to identify the approaching PEP earlier and yield a more accurate forecast for the location and intensity of PEP than direct model output (DMO) at 3-day and longer lead times in the Yangtze–Huai River valley. For the independent PEPE case on 17–19 June 2010, KISAM is able to predict the PEPE about 8 days in advance. That is much earlier than with DMO. In addition, KISAM produces better intensity forecasts and predicts the extent of the PEPE better than DMO at the same lead time of 5 days. In terms of the forecast experiments during June 2010 and 2015, KISAM shows relatively stronger capacity than DMO in predicting the occurrence and intensity of extreme precipitation (EP) and PEP events at lead times of 1 week or even longer. Through validation of more EP, better performance of KISAM compared to DMO on average is further confirmed at 3-day and longer lead times.

Open Street Map (OSM) is a free-editing world map providing web-based spatial information that can be changed in the sense of data updating by everyone, whenever, and wherever. With the data yielded, OSM is very useful in social mapping, disaster mapping, economic mapping, and an area’s terrain mapping, particularly, urban area. In Surakarta, OSM is used for the first time in mapping Kali Pepe (Pepe River) specifically aiming to map the river stream area to cope with flood disaster and generally aiming to provide spatial information with recent data in Surakarta. The utilization of OSM in Kali Pepe mapping was expected to be the beginning of OSM development in Surakarta that would provide digital map data that can be used not only for responding to disaster but also for providing usable data as the reference in policy formulation for Surakarta City Government in economic, cultural and development areas.This study aims to show the result of OSM utilization in the mapping of Kali Pepe in Surakarta for society, stakeholders related to spatial information and also government as the policy maker. The conclusion is that OSM is one of digital mapping that was not popular in Surakarta due to its limited information and accessibility.

<p><em>Flood in Surakarta mostly due to overflowing rivers that flow through the region of Surakarta. The river is part of the urban drainage system. Department of Public Works (DPU) as the local government agency is responsible for the drainage management in Surakarta. One of form of implementation of the river management as part of drainage system is river revitalization program of Pepe River downstream which performed by the DPU. Despite Pepe River downstream revitalization program has been implemented in a variety of activities, the event of flooding due to overflowing Pepe River downstream still occur. Where as the flood is one of the performance indicators of urban drainage system. Thus, from that background it can be defined in research formulation problems, that is how the influence of the river revitalization performed by the DPU Surakarta against Pepe River downstream performance as part of urban drainage system in Surakarta. </em></p><p><em>This research uses quantitative descriptive analysis supported by several analytical techniques including descriptive narrative analysis to identify the characteristics of the revitalization of Pepe River downstream and paired simple t-test analysis to identify the changes of drainage system performance. Next, the relations from the results of both analysis will be identified using explanatory descriptive analysis. Based on the analysis results, it shows that the characteristics of the revitalization of Pepe River downstream is categorized in moderate intensity, because it only includes physical and social activities. Although there are improved changes in the drainage system performance. On the river section I, there are less significant changes with the number of the biggest changes ison the indicator of gabions, that is 11. On the river section II, the drainage system performance changes are significant with the number of the biggest changes is on the indicator of sedimentation, that is 38. In river section III, there are less significant changes with the number of the biggest change is on the indicator of garbage accumulation, with a score of 21.Therefore, it can be argued that the river revitalization program performed by the DPU Surakarta affects the Pepe River downstream performance as the primary drainage channel in the drainage system in Surakarta. The influence can be seen from the reversion of river functions as a drainage channel which is marked by changes in Pepe River downstream drainage system performance from before the river revitalization performed and after the revitalization performed, which is improved.</em></p><p><em> </em></p><p><strong><em>Keywords</em></strong><em>: river revitalization, catchment area, drainage system performence</em></p><p> </p>

Dental decay (Caries) and periodontal disease are globally prevalent diseases with significant clinical need for improved diagnosis. As mediators of dental disease-specific extracellular matrix degradation, proteases are promising analytes. We hypothesized that dysregulation of active proteases can be functionally linked to oral disease status and may be used for diagnosis. To address this, we examined a total of 52 patients with varying oral disease states, including healthy controls. Whole mouth saliva samples and caries biopsies were collected and subjected to analysis. Overall proteolytic and substrate specific activities were assessed using five multiplexed, fluorogenic peptides. Peptide cleavage was further described by inhibitors targeting matrix metalloproteases (MMPs) and cysteine, serine, calpain proteases (CSC). Proteolytic fingerprints, supported by supervised machine-learning analysis, were delineated by total proteolytic activity (PepE) and substrate preference combined with inhibition profiles. Caries and peridontitis showed increased enzymatic activities of MMPs with common (PepA) and divergent substrate cleavage patterns (PepE), suggesting different MMP contribution in particular disease states. Overall, sensitivity and specificity values of 84.6% and 90.0%, respectively, were attained. Thus, a combined analysis of protease derived individual and arrayed substrate cleavage rates in conjunction with inhibitor profiles may represent a sensitive and specific tool for oral disease detection.

Il ritratto dell'assoluta decadenza dell'Italia contemporanea, con la definizione dei suoi abitanti di "polvere d'uomini", indignò gli intellettuali, che avrebbero voluto rispondere nell'Antologia di Vieusseux, il periodico più importante dell'epoca. Pietro Giordani intendeva anche rispondere a Lamartine pubblicando un saggio sulle Operette Morali del giovane (e ancora sconosciuto) Giacomo Leopardi, interpretato come un grande e vivente italiano. La censura ha impedito questa e altre risposte, ma non un aspro riferimento contenuto in un opuscolo dell'esiliiato napoletano Gabriele Pepe. Ferito nell'orgoglio, Lamartine (all'epoca responsabile dell'ambasciata francese a Firenze) sfidò Pepe a duello. La vittoria di Pepe suscitò un grande entusiasmo a Firenze e in tutta Italia. Il tema dell'onore offeso e vendicato con una prova di valore divenne una costante e fu imitato molte altre volte, nella realtà e nella letteratura, alimentando l'immaginazione di diverse generazioni.

e16588 Background: Prostate cancer (PC) is the cancer most commonly diagnosed in men and the second leading cause of death. Multiparametric magnetic resonance imaging (mpMRI) is the benchmark imaging standard for local staging of PC. Patients with pathological extraprostatic tumor (pEPE) have a worse prognosis than those with confined organ disease with a higher risk of biochemical recurrence (BCR) after radical prostatectomy (RP). These factors, in addition to positive lymph nodes (PLN), are pivotal in the decision-making process regarding treatment. However, the subjective MRI analysis has low sensitivity in the detection of EPE and is plagued by the low interobserver agreement. Tumor contact length (TCL) is an objective parameter of mpMRI defined as the length of prostate cancer in contact with the prostate capsule, which may provide additional information about prostate cancer outcomes. We aimed to evaluate TCL as a predictor of pECE, PLN, and BCR in patients undergoing RP. Methods: In this Institutional Review Board approved single-institution prospective study, we update the follow-up of 148 patients and included 31 new patients, with prostate cancer who underwent prostate MRI before radical prostatectomy from March 2014 to november 2018. TCL was measured using T2-weighted magnetic resonance images.Postoperative prostate-specific antigen (PSA) values were obtained every 3 months in the first year, then biannually and annually thereafter. BCR was defined as PSA≥0.2ng/mL. The exclusion criterion was prior treatment. Results: The median follow-up was 44 months. 76/179 (42,4%) patients had pEPE, 12/179 (6,7%) had pLN, and 29/179 (16,2%) had BCR. TCL values were significantly higher in patients with pathological pECE ( 20,6 mm vs 9,9 mm, p < 0.001). Using a cut-off of 11,9 mm we found area under curve (AUC) of 0,71 to predict pathological ECE, with sensitivity 72,5% and specificity 67,5% . Using a cut-off of 16,5 mm we found AUC of 0,64 to predict BCR, with sensitivity 54,2% and specificity 67,8% and with a cut-off of 21,7 mm the AUC to predict PLN was 0,77, with sensitivity 66,7% and specificity 83,6%. Conclusions: We prospectively demonstrated in our population that TCL values were significantly higher in patients with pEPE , PLN and BCR. If validated, this imaging biomarker may facilitate and inform patient counseling and decision-making.

A pesquisa foi conduzida pela abordagem qualitativa e descritiva sobre uma ação realizada pela Fundação Telefônica Vivo, colaboradores e voluntários na instituição OBASE - Organização Batista de Assistência e Ensino, nome fantasia Pepe Bem Querer que tem 12 anos de funcionamento e atende crianças do bairro Bem Querer em Vitória da Conquista, Bahia, com assistência a 180 crianças ano. Por meio das ações realizadas pela Fundação Telefônica, dos voluntários e da contação de histórias foi possível levar as crianças atendidas pelo projeto PEPE um momento de formação para que se inspirem e sonhem em ser o que quiserem quando crescer.

Whether glucagon-like peptide-1 (GLP-1) has insulin-independent effects on glucose disposal in vivo was assessed in conscious dogs by use of tracer and arteriovenous difference techniques. After a basal period, each experiment consisted of three periods (P1, P2, P3) during which somatostatin, glucagon, insulin, and glucose were infused. The control group (C) received saline in P1, P2, and P3, the PePe group received saline in P1 and GLP-1 (7.5 pmol·kg−1·min−1) peripherally (Pe; iv) in P2 and P3, and the PePo group received saline in P1 and GLP-1 peripherally (iv) (P2) and then into the portal vein (Po; P3). Glucose and insulin concentrations increased to two- and fourfold basal, respectively, and glucagon remained basal. GLP-1 levels increased similarly in the PePe and PePo groups during P2 (∼200 pM), whereas portal GLP-1 levels were significantly increased (3-fold) in PePo vs. PePe during P3. In all groups, net hepatic glucose uptake (NHGU) occurred during P1. During P2, NHGU increased slightly but not significantly in all groups. During P3, NHGU increased in PePe and PePo groups to a greater extent than in C, but no significant effect of the route of infusion of GLP-1 was demonstrated (16.61 ± 2.91 and 14.67 ± 2.09 vs. 4.22 ± 1.57 μmol·kg−1·min−1, respectively). In conclusion: GLP-1 increased glucose disposal in the liver independently of insulin secretion; its full action required long-term infusion. The route of infusion did not modify the hepatic response.

Katalan polisiye roman yazarı Manuel Vázquez Montalbán, İspanya'nın transición olarak bilinen demokrasi rejimine geçiş sürecinde polisiye roman türünün önünü açan en önemli yazarlardandır. Montalbán'ın 1972-2004 yılları arasında yazdığı Pepe Carvalho adlı dedektif serisi “whodunit” türündeki klasik polisiye çizgisinden ayrılan romanlardan oluşmasının yanında, Franco sonrası dönemde İspanya'nın geçirdiği toplumsal değişime ışık tutmaktadır. Merkez Komitesinde Cinayet (1981) ise, bu serinin romanlarından biri olarak Montalbán'ın kurguyla gerçeği bir araya getirdiği ve Pepe Carvalho adlı dedektif aracılığıyla İspanya'daki toplum düzenini eleştirdiği bir eserdir. Bu eser aracılığıyla Montalbán'ın, polisiye roman türünü gerçekçi bir yaklaşım ile nasıl yorumladığının bir örneği görülmektedir.
