Full textRhizobial Nod factors (NFs) act as symbiotic signals in the Rhizobium-legume symbiotic interaction. The characterisation of a Medicago truncatula mutant defective for root hair deformation, the induction of a rapid calcium flux, calcium spiking, nodulin gene expression and cortical cell division in response to NFs, led to the identification of a new gene. The gene, called NFP for Nod Factor Perception, is predicted to encode a transmembrane serine/threonine receptor-like kinase containing extracellular LysM domains. The structure of the NFP protein therefore supports the hypothesis that NFP corresponds to a NF receptor. Transcriptomic analysis was performed on wild type plants and the nfp mutant, in response to inoculation by Sinorhizobium meliloti. This led to the identification of a certain number of. The expression of these genes is dependent on the NFP gene, confirming the importance of the NFP-controlled NF signal transduction pathway in the nodulation process

Full textInner hair cells transduce sound stimulation into neurotransmitter release onto the afferent auditory nerve fibers. Here, we studied how cytoskeleton modulates the transduction capabilities of the inner hair cells. Exocytosis at the inner hair cell ribbon synapse is achieved through the coupling between calcium channels and glutamate-filled synaptic vesicles. Using membrane capacitance measurements, we probed whether the actin filament network regulates the exocytosis of synaptic vesicles at the auditory hair cell. Our results suggest that actin network disruption increases exocytosis and that actin filaments may spatially organize a sub-fraction of synaptic vesicles with respect to the calcium channel.The auditory neuropathy 1 (AUNA1) is a form of human deafness, which results from a point mutation in the 5’untranslated region of the Diaphanous homolog 3 (DIAPH3) gene. Strikingly, the DIAPH3 mutation leads to the overexpression of the Diaph3 protein, a formin family member involved in the cytoskeleton nucleation and stabilization. Here, we examined in further details the anatomical, functional and molecular mechanisms that account for AUNA1. We found out that the Diap3-overexpressing transgenic mice show a progressive threshold shift associated to a defect in the inner hair cells. While synaptic function was not affected, Diap3-overexpression results into a selective and early-onset alteration of the inner hair cells cuticular plate, a dense plateform anchoring the stereocilia bundle. Molecular dissection of the apical components revealed that the microtubule meshwork undergoes an aberrant targeting into the cuticular plate of the transgenics’ inner hair cells at early onset, leading to the inabilities of these sensory cells to transduce incoming sound stimulation at later stages

Full textThe DMI2 gene plays a central role for the establishment of the Arbuscular Mycorrhizal and the Legume-Rhizobium symbioses. It is involved in the early steps of perception and transduction of the rhizobial Nod Factor signal. DMI2 encodes a Receptor-Like-Kinase with three LRR and one NSL domain in the extracellular part. DMI2 expression is specific of roots and is induced in nodule primordial and nodule preinfection zone which suggests a role in preparation of the cell to the infection. DMI2 is localised in the plasma membrane and seems to form homodimers and interact with other proteins of the early steps of the signalling pathway, DMI1 and LYK3. Interaction with NFP remains hypothetical. A functional analysis of the NFP, LYK3 and DMI2 RLKs shows autophosphorylation of the LYK3 and DMI2 kinases, contrary to NFP. No evidence of transphosphorylation of NFP by DMI2 and/or LYK3 were obtained. We propose a model of the symbiotic signal transduction

Full textLeguminous plants can establish symbiotic interaction with nitrogen fixing soil-born bacteria collectively referred as rhizobia. Nod factors (NFs) are rhizobia produced molecules essential to the establishment of this interaction. Genetic studies of the NFs perception in M. Truncatula led to the identification of two LysM receptor-like kinases, NFP and LYK3. NFP is necessary for all NFs induced responses while LYK3 specifically controls infection. Signalling events downstream LYK3 are poorly understood. To decipher this signalling pathway, two different yeast two-hybrid screens using M. Truncatula cDNAs and LYK3 kinase as bait were performed. One screen identified an E3 ubiquitin ligase of the U-Box family renamed MtPUB1. MtPUB1 plays a negative role in infection and nodulation, and as for LYK3, this role relies on the NFs structure produced by the rhizobia. In parallel, a second approach based on pairwise interaction assays identified a remorin protein as partner of all three symbiotic receptor-like kinases, NFP, LYK3 and DMI2

Full textPromoting nutrient cycling and tree fitness, ectomycorrhizae (ECM) are mutualistic interactions pivotal in forest ecosystems. However not much is known about the signals and molecular mechanisms underpinning their establishment. Using Populus and Laccaria bicolor as a model system, it was shown that the fungal Mycorrhiza-Induced Small Secreted Protein of 7 kDa (MiSSP7) is secreted upon ECM establishment, penetrates the nuclei of cortical root cells of its host and stabilizes the repressor of jasmonic acid (JA) signaling PtJAZ6, dampening plant responses to JA and promoting mutualism. JA signaling is implied in plant defense and development, but it is currently unknown which part of JA-dependent responses in host roots need to be suppressed for ECM formation. Therefore, the thesis aims to answer the following questions: (i) how does the effector protein MiSSP7 impact the structure of the JA perception complex in poplar roots? (ii) do distinct branches of JA signaling play specific roles during ECM establishment? (iii) does the interplay between JA signaling and other hormone signaling pathways influence symbiotic development? Through protein-protein interaction studies, we deciphered the composition of the PtJAZ6-associated protein complex in poplar roots and showed that MiSSP7 modulates the strength of interactions between PtJAZ6 and its associated transcription factors (TFs). Altered expression of the genes coding for such TFs in poplar roots, through genetic engineering, influences ECM maturation, indicating that PtJAZ6-binding TFs regulate the extent of this mutualistic interaction. In addition, by means of hormonomics coupled with physiologic and transcriptomic analysis of hormone-treated poplar roots and L. bicolor mycelium, we demonstrate that a complex rearrangement of phytohormone biosynthesis and perception takes place in host roots during symbiotic development. In particular, we found that fungus-colonized roots are less sensitive to JA. Altogether, our results suggest a dual role of JA signaling in developing ECM, with one branch functioning to regulate fungal apoplastic penetration to an optimal extent, and another being responsible of fungal exclusion under stress conditions. We propose that finely tuned phytohormone signaling, and in particular JA signaling, is crucial for the integration of fungal and plant-derived signals, in order to reprogram root and mycelial physiology for a successful mutualistic interaction

Full textWe isolated CD36-positive cells from mouse CVP and investigated intracellular signaling events triggered by a polyunsaturated long chain fatty acid, i. E. , linoleic acid (LA). LA induced increases in free intracellular calcium concentrations, [Ca2+]i, by recruiting calcium from endoplasmic reticulum (ER) pool via IP3 production, followed by calcium influx via opening of store operated calcium (SOC) channels. LA also induced phosphorylation of SRC-protein tyrosine kinases (PTKs), particularly of Fyn59 and Yes62. LA-evoked phosphorylation of Fyn59 and Yes62 was implicated in the activation of SOC channels. RT-qPCR revealed that the CD36-positive lipid gustatory cells possessed mRNA of enzymes like tryptophan hydroxylase-1 (TPH-1), L-aromatic amino acid decarboxylase (AADC), tyrosine hydroxylase (TH)), dopamine beta-hydroxylase (DBH) which are involved in the synthesis of monoamine neurotransmitters. Interestingly, addition of LA to these cells induced the release of 5-HT and noradrenalin (NA) to the extracellular environment. The LA-induced release of these neurotransmitters was curtailed by a SOC channel blocker and a SRC-PTK inhibitor. These results altogether demonstrate that LA binds to mouse CD36-positive lipid gustatory cells, triggers calcium signaling and SRC-PTK phosphorylation, and induces the release of 5-HT and NA which, in turn, may be implicated in the downstream signaling to the afferent nerve fibers, thus transmitting the output signal from taste buds to the central nervous system

Full textIn c3 plants, phosphoenolpyruvate carboxylase (pepc) has an anaplerotic function: it is involved in the replenishment of tca cycle intermediates for amino acid synthesis by the gs-gogat cycle. We have shown that arabidopsis leaves contain four pepc isoforms. With pepc2 being the major species. Biochemical and regulatory properties of the enzyme have be en determined : it is sensitive to inhibition by malate, aspartate, and glutamate, this effect is light dependent and it can be reversed by glucose-g-phosphate. Evidence that arabidopsis pepc undergoes a light-dependent phosphorylation process (ph regulation, malate sensitivity, immunological response) was confirmed by an in vivo [32p]-phosphate labelling experiment. The light-transduction pathway was investigated by a pharmacological study using mesophyll protoplasts. It involves photosynthetic electron transfer, a pi-plc activity, transient ip3 production, ip3-dependent tonoplast calcium channels and finally pepc kinase (pepck) synthesis. The arabidopsis genome contains two pepck genes, but only ppck1 is expressed in leaves. The study of a ppck1 knock-out mutant brings the first proof that this pepck is the unique leaf pepc kinase in planta. The mutant displayed no visible phenotype under normal growth conditions. However, when subjected to salt stress (licl, nacl), the mutant showed a significant reduction in growth when compared to the wild type. This result suggests that pepc phosphorylation allows a better tolerance to adverse culture conditions

Full textCan we separate auditory perception and auditory memory ? Intuitively, it seems impossible to remember something that was not perceived. However, there are auditory phenomena for which memory influences perception. The objective of this work is to determine whether the auditory perception and auditory memory are two separable functions, underpinned by two different mechanisms. Two studies using different auditory stimuli (periodic noise, and enhancement) were performed. A population of children and adults with high functioning autism was included. Indeed, they have features in auditory processing. These features can be interpreted in the light of perceptual and / or memory capacity without anything possible to determine whether one or the other or both cognitive functions such as perception and memory, are involved. The combined results of these two studies indicate that the participants with autism have a developmental deficit in processing temporal complexity when their auditory memory is preserved. Two different mechanisms seem to govern the two major cognitive functions such as auditory perception and auditory memory, which would therefore be separated

Full textThe main topic of this study is to discuss the idea according to which perceptual experiences sould be conceive as ways of being in contact with the world and perceptual experiences can give reasons for believing some propositions about the world and can give rise to knowledge. Lots of philosophers countervail this intuitive and naive conception of perception. This rejection is linked to the fact that they adopt an internalist conception of perception and/or justication and/or perceptual knowledge. Perceiving sould be conceive as an purely internal event of subjects which possess perceptual capacities. And so, one may doubt that they can have justifications for their beliefs or that they can know anything. Is it possible to avoid this conclusion ? How to alleviate philosophical fear that this conclusion gives rise ? That is the aim of this study. The proposed philosophical inquiry comes in three forms. In the first place, indirect conception of perception is examined and criticized. Afterwards, an inquiry about the nature of perceptual experience is pursued. The stake for us is to defend a strong form of externalism about perception (metaphysical disjunctivism). Then, in the second part of this work, we get into an ontological reflexion about the nature of properties that perceptual experience seems to acquaintance us with. Color Realism is defended against various antirealist objections. Colors are or can be real properties of things. A realist, non redutive and non relationnal ontology for chromatic properties is sketched (color Primitivism). Lastly, the epistemological stakes of perception are underlined and examined. We defend the idea that perceptual experience is a mean by which certain beings can have justified beliefs although this justification is not reflexive and prima facie. It can be so if it's true that perceptual experience puts us in contact with the world (objects, properties, facts, etc.). We examine and reject different replies to skeptical attacks against the possibility of knowing anything. Then, another strategy is proposed (a morean defense of perceptual knowledge and the questioning of skeptical challenge itself). In fact, from an metaphysical, ontological or epistemological point of view, we don't have to give a negative reponse to the classical and philosophical question "Can mind be really in perceptual and cognitive contact with the world ?" or so we think. Indeed, our study underlines that a positive reply to this question is sustainable, not to say correct, and that the negative reponse is probably wrong

Full textThe perception of the vertical direction is achieved through vestibular, visual and somatosensory information integration. It is studied in the visual (SVV), haptic (SHV) and less often in the visuo-haptic modality (SVHV). The latter raises the question of the integration of visual the information involved in the visual and haptic modalities and of the cognitive model underlying this integration. SVV, SHV and SVHV were compared in the first four studies of this thesis, inhealthy young and older subjects and in right-brain damaged patients with or without visuo-spatial disorders. Performances were closed to the gravity in healthy participants, for SVV as well as forSVHV. VHS, assessed with the right hand, was tilted clockwise in young participants and anticlockwise in older participants. The presence of a visual frame disrupted SVV and SVHV. The right-brain damaged patients had an anti-clockwise deviation of SVV and SVHV and the SHV was even more tilted. SVHV was well predicted from the sum of the SVV and SHV weighted by their relative variances in all conditions and the SVHV variances were lesser. SHV was evaluated in detail in the two last studies of this thesis because of the systematic tilt in the first four studies. The results show that the SHV is tilted clockwise with the right hand and anti-clockwise with the left hand in young healthy subjects. Moreover, deviations reversed in older group and performances are systematically tilted toward the initial positions in the two groups. Taken together, these results show that the way participants integrate visual and haptic information fits the maximum like lihoodmodel with a greater weighting of information available in visual modality and that ageing and right-brain lesions does not alter the multisensory integration. Full textTwo levels may be distinguished in any tonal melody : one corresponding to the physionomical organization (rhythms, melodic contours, dynamics, etc. ) Called "superficial structure", the other corresponding to the underlying organization defined by the network of hierarchical relations associating the phrase notes. The purpose of this study is to specify how musician and non-musician listeners manage to proceed from the perception of physionomical characteristics of the musical surface to the understanding of the underlying network of hierarchical relations. In the first volume, the author considers an important number of experimental studies realized in psychology of music. They inform us of the different perceptual and cognitive processes occuring while listening to music. Their interaction is studied in a detailed review of lerdahl and jackendoff's book : a generativ theory of tonal music (1983). In the second volume the author exposes a series of seven experiments aiming to test the central hypothesis of that modal. The first three show the influence of underlying organization on the memorizing of melodies. The following experiments, proceed by identification tests. They confirm the existence of a processing level in which the phrase is no more represented by its physionomical aspects but by its underlying hierarchical relations. Three general aptitudes are required in order to be able to abstract such a network : one concerning the implicit knowledge of pitch hierarchies set up by the tonal system, another concerning the principles of rhythmic grouping and the last concerning memorizing processes. Every occidental adult listener possesses the three enabling him to get a more or less elaborated understanding of a tonal musical piece. In conclusion, the author considers how those aptitudes appear during the child's growth and what they tell about a more general competence for musical structuring

Full textThe main purpose of this dissertation was to examine the relationship between two kinds of individual variables – New Age beliefs and personality facets – and risk perception. 795 respondents were asked to estimate a) a broad range of hazards reported by Bouyer & al (2001) and evaluated by means of psychometric paradigm (Slovic, Fischhoff & Lichtenstein, 1985a); b) an array of paranormal beliefs that were investigated by Sjöberg & af Wahlberg (2002); And, c) a whole set of personality items extracted from a methodological framework AB5C IPIP (Goldberg, 1999). A eight-factor solution was retained to depict risk perception: Common individual hazards, Pollutants, Energy production, Sex and addictions, Psychotropic drugs, Weapons, Outdoor activities and Medical care. Variables age, sex, and educational level were investigated. Only about 5% of the variance of perceived risk was accounted for by these demographic characteristics. New Age beliefs contributed positively but weakly to the prediction of perceived risk. Energy production was proved to be the most predicted risk factor by New Age beliefs (6% of the variance). Key personality facets pertaining to each of Big Five factors were identified. They were most predictive of risk perception. These facets were Self-disclosure, Tenderness, Rationality, Imperturbability, and Creativity. These facets are effective individual explanatory determinants of perceived risk and should be taken into account for understanding individual risk perception in depth
