Full textDoes exposure to a particular language leave permanent traces in the brain? It is possible for a second language (L2) to replace a first language (L1), under special circumstances? In this thesis we explore the possible remnants of the L1 acquired and "lost" during childhood, as well as competence in the L2 acquired later in life. In order to address these issues, we have chosen to study young adults of Korean origin adopted by French-speaking families during childhood. Our subjects were adopted between the ages of 3 and 10 years and have been severed from the Korean language and culture since their adoption 15 to 30 years earlier. This thesis is comprised of three sections : 1. ) the search for linguistic and other memory traces related to the adoptees'life in Korea; 2. ) the search for remnants of Korean phonology, and 3. ) the evaluation of certain aspects of French. Our findings show that the Korean adoptees have become like native French-speakers in their language processing abilities, suggesting either a loss of Korean, or an important inaccessibility to this language

Full textWith the notice that, today, Gabonese ethnic languages are less spoken by the young, parents become more concern with the necessity to pass on their language to their children. The approach adopted here is based on the analysis of endogroup and exogroup phenomena. We examine, on the one hand the interaction process among members of a linguistic group, and on the other hand the relationships that speakers of different ethnic languages may have one another. Our study is backed upon declared usages stemming from a survey carried out in two Gabonese cities (Libreville and Lambaréné), chosen because of their multiethnic feature. It comes out from this survey that the family still stands as a trustworthy element for the transmission of ethnic languages in Gabon. However, conscious of the fact that the only domestic unit seems not sufficient enough for the maintaining of ethnic languages, we must consider their introduction in the curriculum. But with regard the failure of the various attempts to introduce ethnic languages in the education system, we start wondering about the range of the linguistic policies implemented on the national level. The review of these policies allows us to conclude that the learning of Gabonese ethnic languages needs first a language planning. Full textWe worked on the Vīdēvād, an avestan text whose title is a middle persian word coming from the avestan vidaēuuadāta- "the law that keeps demons away". This anthology contains 22 chapters dealing with religious laws, ritual precepts and measures of purification. These prescriptions aim at rejecting impurity from pure elements in Ahura Mazdā's creation. Our dissertation is an edition and translation with commentary of chapter 19 of this anthology. It relates the temptation of ZaraƟustra by the Bad Evil and how ZaraƟustra overcame him. The text was translated and annotated semantically, phonetically and philologically to highlight linguistic features. Until today, Geldner's edition, which dates back to the end of the 19th century, has been the edition of reference. However, in recent years, scholars have shown that this edition must be revised. Therefore, we prepared an edition of the avestan text by collating 13 manuscripts. This work led us to propose new readings for several words and pursue research on manuscripts filiation. Furthermore, we also studied the middle persian text. This translation is interesting in that it contains glosses that can either be long commentaries or simple synonyms. We also made the edition of the middle persian text on the basis of the two oldest manuscripts (L4 and K1)

Most adoptants resign themselves ; some wish for it, in order to save children in distress, to set up a certain distance, or because they have fantasized a child of another race. The child of international adoption is no ordinary migrant : it occasionnally presents at first, symptoms of distress and phases of regression, then gets adapted on a par with a french child and gives up its original language and culture. Third persons are inclined to intervene greatly in such adoptions and to explain all problems by the uprooting, whereas the main nonspecific difficulty encountered is an affective deficiency. The relation with the original country fluctuates as years go by but it is modeled on the relation with the biological parents

Although the research is oriented to find a possible answer to this question, we propose as a startingpoint, the analysis of the consequences of writing in several languages in modernity. For this reason wedevelop the theoretical concepts of "border", "extraterritoriality", "foreign language" or “mother tongue” that willbe very useful to carry out our analysis. Full textThe aim of this thesis is to study the poetic language of the wind in the poem vents by saint-john perse. In order to identify the linguistic elements which play a part in the elaboration of its poetic field, we approach the poem from a linguistic and stylistic point of view : the poem is considered as a symbolic form; it has a material existence; it is an autonomous entity. The first part consists of doing theoretical research to examine some apparatus which are particular to the language of poetry : the poem as a symbolic form; stylistic activity and poetry; the word as a linguistic unity; the meaning resulting from the complex interactions of different linguistic levels. In the second part, we search for semantic traits in the sentence. It appears that the three semantic traits are cosmic, divine and movement. Regarding discourse, the words force, violence and mouvement function as major factors for the development of the wind. In the third part, we examine the syntactic and morphological aspects of sentences which contain references to the wind. The analysis reveal three aspects : a dialectic movement of being and becoming; the character of ceremonial language; the syntax and morphology revealing the pathos leading to the exclusion of reason. In the last part, we notice that the phonemes (v), (a) contribute to the configuration of the poem at the global level and at the local level. They have architectonic functions and provide the poem with a special structural unity. The phonemes (s), (f), (r) may be looked upon as the expression of the whistling wind

Full textA two-year long internship with the Val-de-Grâce hospital's nephrology services has uncovered, through interviews with patients who have lost their renal functions, certain psychopathological characteristics prevalent among them. With a psychoanalytic and psychosomatic perspective we analyzed the causes and consequences of the loss of renal function, and the ensuing dependency that it brings vis-a-vis the dialysis machine, the hospital staff, and society in general. Following the loss of the kidney we witness the manifestation of a separation that we compare to that that follows the loss of the phantasmatic breast. We refer to the psychosomatic school of Paris' hypothesis to explain that, due to an early trauma that overwhelmed their defense mechanism; these patients could not "mentalize" some amount of anxiety, indispensable to the proper development of the psyche. Hence the test of reality can be perturbed, and "somatization" be made easier. With bodies not "libidinalized" in its whole, patients endow the mechanism of urination with a death pulse, and diverge it of its finality. The loss of the kidney is thus lived like a just punishment to aggressive phantasm toward parents. Dialysis leads to the loss of the patients' sexual functions, and a regression of their body image which recenters itself around their fistula. The repetition of the dialysis, though with a deathlike character, has yet for goal to express, in a body language, their desire to unite at the primordial object, but is also a new attempt to master the boundaries of their bodies

Full textAcknowledged by the end of the 19th century, the loss of chance is nothing else than the prejudice recognizing the Joss of the probability of a positive event occurring. Synonymous of coincidence when used in its singular form but synonymous of probabilities when plural, the chance is in framed by law. The loss of a chance is a repairable prejudice as long as the victim of the harmful attitude shows the gathering of a few substantive conditions. To this effect, the notion of causality ought to be given a particular consideration. The chain of causation must be established between the objectionable behavior and the loss of chance. Hence, the loss of chance prejudice has its own autonomous causation and shall not be mistaken with the entire damage. Furthermore, the chance must be « real and serious » in order to be repairable, thus excluding the low chances on reparation. The victim will then obtain a compensation equal to the loss of chance. Having a legal value, the chance must be lost forever in order to be compensated. Indeed, if the victim claims chance that might still be reached, its compensation will be excluded. Even though it has been rigorously defined, the loss of chance has been brought down to the holdall status, as a legal reaction as if it were the last possibility to obtain compensation. In this perspective, the loss of chance has bypassed the causality, yet essential, in order to award to this prejudice a moral connotation whether than a legal one. The loss of chance undergoes the consequences of the evolution of the civil liability concept and the influence of the precautionary principle. Taking perspective on the loss of chance's definition, this concept has even enable the question of the existence of this legal theory, which encounters a relative success in the European legal systems. Trapped in a legal swirl, the loss of chance has become a compensation reflex, putting all the chances on the same level. By an unexpected revision of the jurisprudence, the Cour de Cassation (Supreme Court), has integrated the reference the "reasonable chance" notion. Thus, the loss of chance finds its original place back, compensating only the substantive chances. This work will aim at demonstrating the impact of the legal revolution on the loss of chance. Whether it is the effectiveness of its legal regime or the stability of its enforcement, the loss of chance has an interest in embracing the legal renewal through strict and pragmatic propositions

Full textExile and mourning, in dialectical relation, are of paramount importance in Linda Lê’s books which are populated by misanthropic characters. The trilogy is nourished by the traumatic loss of the father, the country and the language. The writer is deeply affected by the issue of the language. In fact, she deliberately abandoned her native tongue for that of her adopted country thus generating a feeling of betrayal. From her “exile of language”, Linda Lê constructs a “language of exile”: one of obscurity, of confusion, of crisis, aggravated by the death of her abandoned father. Writing is necessary for her in order to go through mourning and is a pharmakon against madness. Heterogeneous in their form, each book of the trilogy corresponds to one stage of grief. Homogeneous in their substance, they represent the different aspects of the father’s quest. Steeped in the personal experience of the author, the trilogy is nevertheless a work of fiction with a part of extravagance and onirism. The construction of the story also reflects the generic indecision, marked by the coherence between memory and imagination, conscious and unconscious. In addition, the trilogy is a testimony to a cultural duality with the Vietnamese past on the one hand and the European present on the other. Finally, the sentimental duality between love and anger is a mark of the “lê-esque” ideology: paradoxical in appearance but logical and philosophical in reality: life is born from death and death is hidden within life

Full textThis thesis concerns the comparison of acoustic parameters in fluent speech in Tunisian stutterers’ speakers (SB) and non-stutterers (SNB), Tunisians.The acoustic parameters which are compared between SB and SNB include the Voice Onset Time (VOT), the fundamental frequency (F0), the measurements of jitter and shimmer, the scope and direction of the transition of the second formant (F2), and the first three formants (F1, F2 and F3) of the three vowels /a, i, u/. The results are consistent with previous studies regarding the V.O.T, which has been found also be longer for American and French stutterers. Concerning F0values, significant differences between the two groups are obtained for /u/. Measurements of jitter and shimmer show no significant differences, contrary to the results found from American concerning the shimmer. Finally, as regards the study of formant variations, there are tendencies without reaching significance for F1, F2 and F3. F2 values are lower in stutterers with significant results obtained for sequences of C-u. The vowel triangle of the three vowels /a, u, i/ in Tunisian children who stutter, is not centralized, unlike triangles obtained for French and American stuttering speakers, according to the literature

Full textAfter being restricted for a time to limited assumptions, the loss of opportunity loss gradually won over various parts of private law. Praetorian creation of judicial jurisdictions appeared at the end of the nineteenth century and then sanctioned by the administrative judge, the loss of opportunity has recently entered the Civil Code and the Commercial Code. Its success is also demonstrated by its omnipresence in the successive proposals for the reform of the law of obligations that followed in domestic law as well as by its recognition by certain foreign laws, by the European Court of Human Rights and by the Court of Justice of the European Union. Yet, the loss of opportunity shows a deficit of conceptualization that has proved prejudicial. The absence of a rigorously defined framework of use has opened the way to the erroneous extension of the loss of chances as well as the manipulation of the conditions, which are not very constraining to its repair. At the time of impending civil liability reform, the business of conceptualizing loss of opportunity harm was inviting. ln order to effectively grasp the figure of the loss of opportunity, the thesis pays particular attention to the object that characterizes it - yet too little exploited so far - namely the chances of success of the victim preceding the occurrence of the generating event and, more precisely, the hazard that underlies them. The thesis first highlights the fact that this examination makes it possible to justify the principle of the protection of opportunities as an interest protected by civil liability and then to rationalize this protection. The thesis then highlights the fact that it is the examination of the hazard that governs the principles governing the reparation of the harm that is inferred from the injury of this interest

Full textFor several years, loss of chance has interested administrative judges and legal writing in public law. The former, who have widely developed its area in law of administrative liability, have changed the method of compensation of this instrument, by using the example of private law. The second has tried to identify the justifications of the use of loss of chance by using the uncertainty of prejudice and the one of causal relationship. But the analysis of jurisprudence shows that, despite the diversity of risks justifying the use of loss of chance, this is in keeping with a unique scope which is characterized by causal uncertainty. The variety is to be found elsewhere, at the stage of compensation. According to the selected method – the all or nothing approach or proportional compensation – the face of the loss of chance changes. It sometimes forms a presumption of causation, sometimes a prejudice, and so overcomes the causal uncertainty in two different ways. The current hesitations of administrative judges prove that it is difficult to choose between one or the other of two methods of compensation, and therefore, between the two instruments which are linked to them. However, the type of risks which underlies the use of mechanism can give a key to repartition of these two methods of compensation

Full textSurgery as metaphor, aesthetic of looseness, is the study of the staging of surgical procedures and of its metaphors’ interpretation. By proposing an analogy between amorous relationships and those that tie the patient and his surgeon together, and by demonstrating that both are power-based relationships, our research aims to establish a critic of surgical power, as well as to question the status of the operated-on body. In addition, we challenge the visual transfiguration of surgery’s representation. By analysing various masterpieces on the subject, we notices that most of them revolve around two major notions: the reconsideration of the operated-on body’s property – as a subject and object – and the relationship between the surgeon and his patient. Such images also evoke a particular representation in the mind, in that they question the boundaries between medical science and the arts, and also generate their own aesthetics. The metaphors’ analysis potentially conveyed by the subject leads us to establish that surgery is regularly used as a pretext to stage dramatic events. Involving topics such anaesthesia, anxiety, particularly in the case of organ transplantation, that of mourning that may be the outcome, and the theme of surgery itself always evokes the very idea of loss. By exploring surgery as an art media, the very core of the intimate relationship between surgeons and their approach of their patient’s bodies are being questioned in this research

Full textFor a few years, meshes have been conquering a predominant status amongst the different types of computer data structure for geometric objects. More precisely, the meshes composed of simplices – triangles for the modeling of surfaces embedded in 3D, tetrahedral for the modeling volumes – seem to be the most widespread at the present time. The rapid growth of applications using these geometric structures in such various fields as finite elements computing, simulation of surgery, or video games has quickly raised the problem of an efficient coding, well suited for storage or visualization. The advent of the World Wide Web, which needs a compact and progressive representation of the data in order to guarantee the user-friendliness of the exchanges and communications, increases this need. Therefore, since 1995, a large number of algorithms have been proposed for the compression of triangular meshes, using for most of them the following approach : the vertices of the mesh are coded in an order such that is contains partially the topology of the mesh. In the same time, one simple rules attempts to predict the position of the current vertex from the positions of its neighbors that have been previously coded. In this thesis, we have chosen to give the priority to the compression of the vertex positions rather than their connectivity. We describe a set of coding methods which are progressive, lossless, adapted to arbitrary meshes (non necessarily triangular nor manifold, with arbitrary genus), and generalizable to any dimension. Our results are competitive with regards to the best current progressive methods : for instance, for the particular case of triangular surface meshes, about 3. 6 bits per vertex are necessary on average to code the connectivity of usual models

This was the question Georges Bataille encountered during the first years of the war, a question that would drive him to radically change the direction of his past reflections. After having admitted to the impossibility of expenditure, he turned in effect - thanks to Maurice Blanchot - from the " will to loss " to the " will to chance " and sought less to realize expenditure himself than to let it happen on its own. The strange writing of La Somme athéologique, should be understood in relation to this shift in perspective. In order to remain faithful to the " will to chance ", Bataille found himself simultaneously in front of the prohibition and the obligation to write, and therefore similar to the writer described by Blanchot in " De l'angoisse au langage ". The present dissertation, which puts into relief these " dramatic events " experienced by Bataille during World War II, attempts to analyze certain post-atheological and literary texts

Full textIn order to understand the results of experiments on quark-gluon plasma such as ALICE at LHC at CERN, it is important to have accurate models to study the behavior of elementary particles in this kind of exotic medium. This thesis work is incorporated within the framework of radiative energy loss of an ultrarelativistic particle propagating in a dense medium. The radiative energy loss is a process characterized by a time called formation time so one can wonder if this energy loss is modified when the emitted gluon itself interacts in the medium during its formation. From the microscopic viewpoint, we want to look at the situation in which the mean free path in the medium is shorter than the formation time of the primary gluon. Thus, in this window of time, the quark-gluon pair being formed continues to interact in the medium with the possibility of emitting other gluons, called secondary ones, at lower energies and which could have an effect on the energy loss of the quark itself. To investigate this, we propose an effective formalism allowing us to include these secondary radiations in the propagation of the main quark or gluon in an ultrarelativistic limit and for radiations whose energy is negligible compared to the one of the principal pair. Full textThe aim of this thesis is the development of a unified framework to study the regularity of certain characteristics elements of chaotic dynamics (Topological presure/entropy, Gibbs measure, Lyapunov exponents) with respect to the dynamic itself. The main technical issue is the regularity loss occuring from the use of a composition operator, the transfer operator, whose spectral properties are intimately connected to the aformentionned "characteristics elements". To overcome this issue, we developped a regularity theorem for fixed points (with respect to parameter), in the spirit of the implicit function theorem of Nash and Moser. We then apply this "fixed point" approach to the linear response problem (studying the regularity of the system invariant measure w.r.t parameters) for a family of uniformly expanding maps. In a second time, we study the regularity of the top characteristic exponent of a random prduct of expanding maps, building from our regularity theorem and cone contraction theory. We deduce from this regularity w.r.t parameters for the stationanry measure, the variance in the central limit theorem, and other quantities of dynamical interest

corps entier est augmentée. Cependant, les mécanismes biochimiques restent discutés. Nos
résultats montrent que la restriction calorique s'accompagne d'une diminution de l'activité de
la chaîne respiratoire et de la respiration de mitochondries hépatiques, proportionnelle à
l'intensité de la restriction calorique et fortement corrélée au taux plasmatique de leptine. A
l'inverse, chez le rat traité par dexaméthasone (modèle de dénutrition) il y a une augmentation
de la fuite de protons des mitochondries hépatiques et une anomalie in vivo de la
phosphorylation oxydative du muscle squelettique. Si ces modifications peuvent expliquer les
variations du métabolisme de base, l'adaptation à la restriction calorique semble aussi capable
de limiter la production de radicaux libres oxygénés.

Full textThe goal of this document is to verify the feasability of an out-of-step relay using data mining and decision trees. Using EMTP-RV and the Anderson network, 180 simulations were done while changing the place of the short circuit, the length, the type and the load-flow. For these simulations, 39 electrical measures and 8 mechanical measures were made. These simulations were then classified as stable or instable using the center of inertia of angle and speed. With MATLAB, 33 new other variables were created by using the first 39, and then with KNIME, decisions trees such as C4.5, CART, ADABoost, ADTree and random forest were simulated and the sampling time versus the performances were compared. Using Consistency Subset Eval, Symmetrical Uncert Attribute Set Eval and Correlation-based Feature Subset Selection, the features were reduced and the simulations were visualised using the validation set. Results show that with a sampling frequency of 240 [Hz] and 28 variables is enough to obtain a mean area under the curve of 0.9591 for the training and the validation set of the 4 generators.

This paper starts from the assumption that there is a disharmony between speech (meaning) and language (form) in the stuttered discursive functioning. Based on discursive linguistics we understand that in a fluent in position, the speaker slides along the meaning of speech without any attention to the form of language; yet in a stuttered position he reminds occupied in trying to avoid stuttering and by acting like that he remains subdued to the form of language and looses the discursive position that would guarantee his fluency. Objective: To develop the understanding about disharmony between speech and language via the speech of adult stutterers. Method: Prospective qualitative research conducted through semi-structured interviews with six adults, following the key questions: what comes to mind when I say - speech ; to speak with fluency ; public speaking ; talking to myself ; what helps fluency ; what do you makes to not stutter or what helps you not to stutter ; do you know if you will stutter?, and what makes you know that you will stutter . After the speech transcription, the notion of discursive practices and production of meaning (Spink and Medrado, 2004) allowed us to define three categories that organized the results and guided the discussion: Sense Speech composed by the sub-categories: Positive and Negative; Context of Speech, composed by the sub-categories The Effect of Others and The Effect of Being Alone; and Anticipation of Stuttering. Results and Discussion: In Sense Speech-Positive respondents reported how they feel when they flow better in speech and when they allow themselves to stutter. Theirs vision of speech is the one that circulates in common sense. The respondents had an idealized view of fluency as absolute. In Sense of Speech-Negative respondents reported feeling dissimilar and marginalized by his manner of speaking, also reported emotions and feelings related to stuttering. We emphasize the importance of understanding the relationship between the production of stuttering and the subjective position of a stigmatized speaker, to build up therapeutic approaches to overcome this position. In Context of Speech-The Effect of Others, respondents discriminated types of speakers and of discursive contexts that favored fluency or stuttering. They showed negative marks on subjectivity, because when they feel like stutterers they imagine the others monitoring their speech and able to reject or disapprove them. In Context of Speech-The Effect Alone respondents reported that when they are alone they can flow freely, which reiterates the stuttering as a manifestation strongly linked to the presence of others. In Anticipation of Stuttering respondents reveal the ways in which stuttering is expected subjectively before it is materialized in their speech: thoughts or views; emotions or feelings; actions of the body or linguistic strategies. Conclusion: The reports of the respondents indicates that the disharmony between speech and language, in stuttered speech, is related to a speaker who feels stigmatized in his subjectivity in face of certain conditions of discursive production; also indicates the different ways in which this disharmony is materialized in order to avoid the provided stuttering. These characteristics indicate some elements that seem important to the targeting of a therapeutic approach of stutter speech wishing to overcome the subjective/discursive functioning that sustains it
Este trabalho parte da hipótese de que no funcionamento discursivo gaguejante há uma desarmonia entre fala (sentido do dizer) e língua (forma do dizer). Com base na linguística discursiva temos que, na posição fluente, o falante desliza pelo sentido do dizer esquecido da forma; na posição gaguejante, ocupado em evitar essa forma de falar, permanece submetido à forma e perde a posição discursiva que lhe garantiria a fluência. Objetivo: Desenvolver a compreensão sobre a desarmonia entre fala e língua a partir do discurso de pessoas adultas com gagueira. Método: Pesquisa qualitativa prospectiva realizadas por meio de entrevistas semi-estruturadas com 6 adultos, a partir das perguntas-chave: o que vem à sua cabeça quando eu digo - falar ; falar com fluência ; falar em público ; falar sozinho ; o que o ajuda a fluir ; o que faz para não gaguejar ou o que o ajuda a não gaguejar ; se sabe que vai gaguejar ; e o que o faz saber que vai gaguejar . Após a transcrição do discurso a noção de práticas discursivas e produção de sentidos (Spink e Medrado, 2004) permitiu que se definissem três categorias analíticas que organizaram os resultados e guiaram a discussão: Sentido da Fala composta pelas sub-categorias Positivo e Negativo; Contexto de Fala, composta pelas sub-categorias Efeito Outro e Efeito Sozinho e Antecipação da Gagueira. Resultados e Discussão: Em Sentido da Fala Positivo os entrevistados relataram os modos de se sentir relacionados a fluir melhor na fala e a permitir-se gaguejar. Sua visão de fala é a que circula no senso comum e corresponde a uma visão idealizada da fluência como absoluta. Em Sentido da Fala Negativo os entrevistados relataram sentir-se dessemelhantes e marginalizados por seu modo de falar, relataram também emoções e sentimentos relacionados à gagueira. Destaca-se a importância de compreender a relação entre a produção da gagueira e a posição subjetiva de falante estigmatizado, para construir os caminhos terapêuticos de sua superação. Em Contexto de Fala Efeito Outro, os entrevistados discriminaram os tipos de interlocutores e de contextos discursivos que lhes favorecem a fluência ou a gagueira. Revelam marcas negativas na subjetividade, porque ao ocuparem a posição de falantes gagos imaginam o outro na posição daquele que fiscalizará seu dizer e poderá rejeitá-los ou reprová-los. Em Contexto de Fala Efeito Sozinho relataram que na condição sozinhos podem fluir livremente, o que reitera a gagueira como uma manifestação fortemente atrelada à presença do outro. Em Antecipação da Gagueira os entrevistados revelam os modos como a gagueira é prevista subjetivamente antes de materializar-se na fala: pensamentos ou visualizações; emoções ou sentimentos; ações do corpo ou estratégias linguísticas. Conclusão: Os relatos dos entrevistados indicam que a desarmonia entre fala e língua, no discurso gaguejante, relaciona-se a um falante que, na subjetividade, se sente estigmatizado diante de certas condições de produção discursiva; indicam ainda os diferentes modos pelos quais essa desarmonia se materializa com a finalidade de evitar a gagueira prevista. Tais características assinalam alguns elementos que parecem importantes ao direcionamento de uma abordagem terapêutica fonoaudiológica da gagueira que pretenda superar o modo de funcionamento subjetivo/discursivo que a sustenta

Full textIn humans and mice, testicular development in XY gonads involves SRY/SOX9 signaling to promote Sertoli cell differentiation and their formation as testis chords. For ovarian development in XX gonads, RSPO1/WNT/beta-catenin signaling is the main pathway for granulosa cell differentiation and their subsequent assembly into follicles. Indeed, XY Sox9 mutant mice develop ovaries, and XX Rspo1 mutant mice develop ovo-testes, a gonad containing a testicular and an ovarian part. In XX Rspo1 mutant mice, ovo-testicular development involves precocious differentiation of some granulosa cells and their and reprogramming as Sertoli cells. Thus, these single mutant studies demonstrated that SOX9 and RSPO1 are required for testicular and ovarian development respectively, and that SRY is dispensable for testicular development in XX Rspo1 mice. Interestingly, gonad development in XY and XX Rspo1 Sox9 double knockout (DKO) mice has challenged the requirement of SOX9 for testicular development. In XX Rspo1 single mutants, it was assumed that Sertoli cell differentiation was SOX9-dependent, but co-inactivation of Sox9 in DKO mice does not impair the ovo-testicular phenotype. For XY Sox9 single mutant mice developing ovaries, co-inactivation of Rspo1 in XY DKO mice rescues the sex reversal, though the testes are hypo-plastic. Thus, in XY and XX Rspo1 Sox9 DKO mice, SOX9 and/or SRY are dispensable for testicular differentiation, indicating that an alternate testis factor exists. For my research project, we hypothesized that a SOX9-related transcription factor, SOX8, acts redundantly for testicular development in XY and XX Rspo1 Sox9 DKO mice. Thus, to first establish redundancy among the SOX factors, we first analyzed their expression in Rspo1 mutant mice lacking Sox8 or Sox9, and then generated and analyzed gonad development in XY and XX Rspo1 Sox8 DKO mice. Then to test our hypothesis, we studied Rspo1 Sox8 Sox9 triple knockout (TKO) mice. We predicted that a loss of both Sox genes in TKO mice would prevent granulosa cell reprogramming as Sertoli cells and subsequent testicular development. To characterize gonad development and their effects in DKO and TKO mice, we performed analyses in embryonic day 17.5 (E17.5) mice, when granulosa-to-Sertoli cell reprogramming begins in XX Rspo1 single mutants; in juvenile post-natal day 10 (P10) mice, when gonad fate is set; and in young adult P40 mice. We examined a variety of parameters including gonad morphology and secondary sex characteristics, as well as gonad organization and cell population by histological and immunostaining analyses. We report that SOX8 and SOX9 are expressed independently in XY and XX Rspo1 Sox9 DKO and Rspo1 Sox8 DKO gonads in embryonic and juvenile mice. Next, XY and XX Rspo1 Sox8 DKO mice developed testes and ovo-testes, indicating that loss of one SOX factor does not impair testicular differentiation, as in XY and XX Rspo1 Sox9 DKO mice. In XY and XX Rspo1 Sox8 Sox9 TKO mice, granulosa-to-Sertoli cell reprogramming was impaired at E17.5 and post-natal gonads lacked testicular development. Thus, SOX8 can compensate for the loss of SOX9 in Rspo1 Sox9 DKO mice. In addition, gonads in XY and XX TKO mice developed as atrophied ovaries, indicating that ovarian fate is partially maintained.In total, we investigated the etiology of pathophysiological testicular development in RSPO1 loss-of-function mice. Remarkably, though SOX8 is dispensable for male sex determination in mice, it can promote testicular differentiation in the absence of SRY and SOX9 because of functional redundancy with SOX9. Thus, in human cases of sex reversal where testicular development cannot be explained by misexpression of SRY or SOX9, SOX8 could be a causative factor
