Full textENGLISH ABSTRACT: As overpopulation threatens the world’s ability to feed itself, food has become an invaluable resource. Unfortunately, almost a third of the food produced for human consumption is lost annually. Pests including insects, phytopathogens and weeds are responsible for more than a third of the annual major crop losses suffered around the world. The majority of current post-harvest control strategies employ synthetic agents. These compounds, however, have been found to be detrimental to the environment as well as human health, which has led researchers to investigate alternative strategies. Biocontrol agents are environmentally compatible, have a lower toxicity and are biodegradable, making them an attractive alternative to the synthetic control agents. The lipopeptides produced by Bacillus spp. in particular, have shown great potential as biocontrol agents against various post-harvest phytopathogens. Most biocontrol strategies apply the biocontrol organism, for example Bacillus, directly, whereas this study focused on the use of the lipopeptide itself as an antifungal agent. This is advantageous as the lipopeptides are less sensitive to their surroundings, such as temperature and pH, compared to living organisms, allowing for the production of a standardized product. This study investigated the production of the Bacillus lipopeptides surfactin, fengycin and iturin under controlled batch conditions. Parameters increasing lipopeptide production were quantified, focussing on antifungal lipopeptides (iturin and fengycin), and lipopeptide production was optimized. Experiments were performed in a fully instrumented 1.3 L bench-top bioreactor and lipopeptide analyses were performed via high pressure liquid chromatography (HPLC) and liquid chromatography-mass spectroscopy (LC-MS). After screening four Bacillus spp., Bacillus amyloliquefaciens DSM 23117 was found to be the best antifungal candidate. This was based on it outperforming other candidates in terms of maximum antifungals produced, Yp/x,antifungals (yield per cells), and antifungal productivity. Nitrate, in the form of NH4NO3, was critical for lipopeptide production and an optimum concentration was observed above which the CDW (cell dry weight) no longer increased significantly and both μmax (maximum specific growth rate, h-1) and lipopeptide production decreased. For μmax, the optimum NH3NO4 concentration was 10 g/L and for lipopeptides it was 8 g/L. At these respective NH4NO3 concentrations μmax = 0.58 (h-1), the maximum antifungals (fengycin and iturin) were 285.7 mAU*min and the maximum surfactin concentration was 302 mg/L. The lipopeptides produced by B. amyloliquefaciens, the antifungals (fengycin and iturin) and surfactin, are secondary metabolites, regardless of the optimization treatment, i.e. increased NH4NO3 concentrations. Using 30% enriched air extended the nitrate utilization period, suggesting that when increasing supply concentration, more oxygen was available to act as electron acceptors, allowing nitrate to be used for lipopeptide production. The number of iturin and fengycin homologues generally increased with an increase in nitrate concentration. This suggested that process conditions, such as nitrate concentration, can be used to manipulate homologue ratios, allowing for the possibility to tailor-make biocontrol-agent upstream, during the production process, and possibly increase the efficacy of the biocontrol strategy. The lipopeptides produced by B. amyloliquefaciens showed complete inhibition against Botryotinia fuckeliana and diminished the growth capabilities of Botrytis cinerea. No inhibition was observed against Penicillium digitatum. These results indicate potential of the biocontrol strategy, although scale-up and fed-batch studies are recommended, especially when considering commercial implementation. Studies regarding the lipopeptide application method, i.e. a single application or multiple applications, should also be investigated as this will influence the efficacy of the lipopeptides against the target organisms.

Daar word beraam dat een derde van die voedsel wat wêreldwyd geproduseer word vir menslike verbruik verlore gaan elke jaar. Verder is insekte, plantpatogene en onkruide verantwoordelik vir meer as ‘n derde van die verliese rakend jaarlikse oeste. Meeste bestaande na-oes beheermetodes maak gebruik van sintetiese stowwe. Ongelukkig kan hierdie verbindings nadelig wees vir die omgewing sowel as menlike gesondheid. Navorsers het hulsef dus toespits daarop om alternatiewe beheermetodes te ondersoek. Bio-beheermetodes is omgewingsvriendelik sowel as bio-afbreekbaar, wat hulle ideale alternatiewe maak vir die sintetiese stowwe. By 10 g/L NH4NO3 was μmax = 0.58 (h-1) en by 8 g/L was die maksimale antifungiese produksie (fengycin en iturin) 285.7 mAU*min en die maksimale surfactin produksie 302 mg/L onderskeidelik. Die lipopeptide, die antifungiese middels (fengycin en iturin) en surfactin, geproduseer deur B. amyloliquefaciens is sekondêre metaboliete, ongeag van die optimerings-behandelinge wat toegepas word, soos ‘n verhoging in NH4NO3 konsentrasie. Verdere studies met betrekking tot die aanwendingsmetode van die lipopeptiede moet ook verder ondersoek word, m.a.w. enkel teenoor menigte aanwendigs, aangesien dit die effektiwiteit van die lipopeptiede teen die teikenorganismes sal beïnvloed.

Die swam toon belofte vir die beheer van hierdie nematodes en het terselfdertyd nie 'n nadelige effek op nie-teiken, voordelige organismes in die grond nie. Veral in kombinasie met chemiese middels, as deel van geïntegreerde programme, kan dit lei tot verminderde gebruik van hoogs toksiese middels en dus meer omgewingsvriendelike landboupraktyke. Biological control of plant parasitic nematodes on potatoes, citrus and grapevine with the fungus, Paecilomyces liIacinus. Paecilomyces liIacinus, race 251 (registered in terms of act 36 of 1947 as South Africa's first natural nematicide, commercially available as PI Plus) was tested as a biological control agent on potatoes and in integrated control programs on citrus and grapevine against Me/oidogyne species, Ty/enchu/us semipenetrans and various ectoparasitic nematodes respectively. The fungus shows promise for the control of these nematodes, without having a harmful effect on non-target, beneficial organisms in the soil. Especially in combination with chemical products, as part of integrated programs, this can lead to less use of highly toxic compounds and thus to more environmentally friendly agricultural practices.
AFRIKAANSE OPSOMMING: Sedert die ontdekking van die swam, Paeci/omyces Ii/acinus (Thom.) Samson as 'n effektiewe eierparasiet van Meloidogyne incognita acrita en Globodera pal/ida (Jatala et al., 1979) het verdere veldproewe in Peru tot die effektiewe beheer van M. incognita en Tylenchulus semipenetrans gelei. Na verskeie suksesse in Peru is die swam onder verskillende klimaat- en grondkondisies in verskeie ander lande beproef. Die sukses behaal in die Filippyne het gelei tot die kommersiële produksie van die swam onder die handelsnaam Biocon. Full textLa acción insecticida de varios esteres de alilo fue testada en diversos insectos y mediante distintos modos de aplicación. La actividad por aplicación tópica varió en función del éster de alilo. El cinamato de alilo y el naftoato de alilo fueros los compuestos más activos en huevos y larvas neonatas de C. pomonella, G. molesta y L. botrana, mientras que el salicilato de alilo no produjo mortalidad a la dosis más alta testada (10 mg/mL). El cinamato de alilo fue el único éster activo por aplicación tópica en A. pisum mientras que todos los esteres testados lo fueron para T. castaneum. Los esteres de alilo estudiados produjeron pérdida de viabilidad celular en todas las líneas celulares de insectos cuando dicha viabilidad fue analizada mediante dos metodologías distintas (MTT y Azul de Tripano), y siendo ésta debida a la disrupción de la membrana celular. El cinamato de alilo fue el producto más activo, y las células del aparato digestivo de Choristoneura fumiferana (Lepidoptera) las más sensibles. La acción insecticida por ingestión en larvas de S. littoralis y C. pomonella, y en ninfas de A. pisum, fue confirmada y el aparato digestivo fue señalado como principal punto de acción de los esteres de alilo. Los correspondientes ácidos y dicloropropilesteres presentaron una menor o igual acción insecticida que los esteres de alilo siendo dicha acción también debida a un efecto en la membrana celular. Las diferencias en la acción de los distintos compuestos podrían ser debidas a diferencias en las propiedades lipofílicas de los compuestos y su interacción con las membranas celulares. Los esteres de alilo produjeron un efecto en la comunicación química de T. castaneum pero no en A. pisum, lo que podría utilizarse para mantener los productos almacenados libres de T. castaneum. En cuanto a C. pomonella y L. botrana, todos los esteres de alilo probados produjeron una respuesta en las antenas de los machos de C. pomonella, mientras que tan solo el cinamato de alilo la produjo en las antenas de hembras de C. pomonella y en machos y hembras de L. botrana. Esta respuesta no se tradujo en un aumento de la atracción de machos hacia cebos con mezclas de ester de alilo y feromona en ensayos de túnel de viento, pero si aumentó el número de hembras atraídas. Este hecho podría utilizarse par incrementar el número de hembras capturadas en trampas de feromona. Estos resultados, sugieren el papel de los esteres de alilo en el control de plagas, especialmente del cinamato de alilo.

Full textENGLISH ABSTRACT: Phytophthora cactorum (Lebert & Cohn) Schrot. is the primary cause of crown, collar and root rot diseases of apple (Malus domestica Borkh.) trees worldwide. This pathogen is most destructive in commercial apple orchards under waterlogged soil conditions and has recently been identified as causing serious disease in some South African apple orchards. Crown, collar and root diseases are difficult to control because of their unpredictability and catastrophic nature. The use of resistant cultivars and rootstocks is economical and environmentally considerate. Therefore the need to develop screening techniques that will enable the selection of desirable disease resistant traits as part of an apple-breeding program in South Africa was identified. The work undertaken in this study was aimed at optimizing different techniques to test resistance. Using two direct inoculation techniques (excised stem and intact stem) the aggressiveness of lO isolates of P. cactorum on apple rootstocks was determined. The susceptibilities of five apple rootstocks were also compared. Results have shown isolate by rootstock interaction which means isolate aggressiveness was influenced by rootstocks tested. The selectivity of isolates suggests that there may be several strains of the pathogen. Population studies of the pathogen might contribute valuable information that could lead to better interpretation of results. Rootstock susceptibility was monitored in vitro throughout the season by inoculating at monthly intervals for 26-months. It was observed that during winter, rootstock susceptibility was low compared to high susceptibility during summer. These results have revealed new information regarding changes in the relative resistance of the different rootstocks over the growing season, e.g. the susceptibility pattern of rootstock MMl06 occurred 1 to -2 months later than that of other rootstocks. This finding has important implications on the way in which resistance test results are interpreted, and emphasizes the importance of not relying on point sampling. Furthermore, useful information has been acquired regarding the epidemiology of the disease with regard to "windows of susceptibility". The phenomenon of a phase shift in susceptibility of different rootstocks needs to be tested on a broader scale to assess whether it has any practical application on resistance testing. Although different inoculation techniques are applied in breeding programs, up to now there is no consensus on which technique works best for seedling selections. Since large numbers of individuals must be tested to improve the chances of detecting resistant genotypes, mass inoculations of young seedlings is a rapid way of identifying resistant individuals. Two different screening methods were tested during this study. Using the sand-bran technique, seedlings were transplanted onto inoculated soil and the root mass was used as a measure of resistance. In a second method zoospore inoculum was applied to seedlings growing in a sand:bark mixture at different concentrations and the seedlings were subjected either to water drenching or not. In both trials the aggressiveness of isolates differed significantly from each other and only higher inoculum concentrations were effective in causing disease. The age of seedlings used in tests emerged as an important factor. Seedlings under five-months-old should not be used. Drenching inoculated seedlings enhanced disease development but the production of sufficiently high numbers of zoospores was a laborious task. Thus, it is recommended that the sand-bran inoculum technique be tested with the drenching treatment for mass selection. In conclusion this study confirms the importance of both choice of isolate and choice of inoculation intervals in determining susceptibility of rootstocks to infection. In spite of the fact that stem inoculation bioassays have limited resemblance to natural disease situations, these bioassays are useful for obtaining an indication as to whether genotypes have a degree of resistance and merit further testing. For this reason refinement of the stem inoculation bioassay is worthwhile pursuing. With regard to seedling trials, both the sand-bran and the zoospore technique appear promising but refinement of these techniques is necessary in order to present a more practical way of testing large volumes of seedlings.
AFRIKAANSE OPSOMMING: Evaluering van inokulasietegnieke om weerstand teen Phytophthora cactorum in appels te evalueer: Phytophthora cactorum (Lebert & Cohn) Schrot. is die primêre oorsaak van kroon-, kraag en wortelvrot van appelbome (Malus domestica Borkh.). Dit is die mees verwoestende patogeen in kommersiële appelboorde waar daar versuipte toestande grond voorkom. P. cactorum is onlangs identifiseer as die patogeen wat ernstige kroon- en kraag-verotting in Suid Afrikaanse appelboorde veroorsaak. Kroon-, kraag- en wortelvrot is moeilik om te beheer as gevolg van die onvoorspelbaarheid en rampspoedige aard van die siekte. Die gebruik van kultivars en onderstamme wat weerstandbiedend is teen siektes en plae is omgewingsvriendelik en is ekonomies van belang, dus het die behoefte ontstaan om inokulasietegnieke te ontwikkelom weerstandige saailinge te identifiseer en te selekteer as deel van 'n appelteelprogram in Suid Afrika. Die doelwit van hierdie studie is om verskillende inokulasietegnieke te toets en te verfyn om weerstand in appelsaailinge te identifiseer. Deur gebruik te maak van twee inokulasietegnieke (die afgesnyde loot- en intakte loot tegniek), is die relatiewe aggressiwiteit van 10 isolate van P. cactorum en die vatbaarheid van vyf appelonderstamme ondersoek. Resultate het aangetoon dat die aggressiwiteit van die isolate gevarieer het na aanleiding van die onderstam wat getoets is. Die selektiwiteit van die isolate is 'n aanduiding dat daar moontlik verskeie rasse van die patogeen voorkom. Toekomstige studies op die populasiestruktuur van P. cactorum sal 'n belangrike bydrae maak tot die interpretasie van resultate oor weerstand en weerstandsteling. Die vatbaarheid van onderstamme was ook in in vitro proewe ondersoek deur maandelikse inokulasies toe te pas oor 'n tydperk van 26 maande. Dit is opgemerk dat die onderstamvatbaarheid gedurende die winter laag was in vergelyking met die somer. Nie al die onderstamme het dieselfe gereageer gedurende verskillende toetstye nie. Hierdie resultate toon aan dat die relatiewe weerstand van verskillende onderstamme oor die groeiseisoen verskil, byvoorbeeld die vatbare reaksie van die onderstam 'l\.1MI06' het een tot twee maande later voorgekom in vergelyking met ander onderstamme wat getoets is. Hierdie bevinding het belangrike implikasies op die interpretasie van weerstandstoetsing en beklemtoon die moontlike tekortkominge in enkelproefwaarnemings. Bruikbare inligting ten opsigte van die epidemiologie van die siekte is versamel wat beskryf kan word in terme van vensters van vatbaarheid wat verskil van onderstam tot onderstam. Verdere ondersoeke in die verband word aanbeveel. Hoewel verskeie inokulasietegnieke bestaan om jong saailinge vir weerstand te toets, is daar tot op hierdie stadium nog nie ooreenstemming oor die beste tegniek wat toegepas moet word om saailingseleksie te doen nie. In albei hierdie proewe het die aggressiwiteit van die isolate van mekaar verskil. Slegs die hoër inokulumkonsentrasies was effektief in die ontwikkeling van die siekte. Die ouderdom van saailinge is ook uitgewys as 'n belangrike faktor wat 'n rol speel in weerstandstoetsing. Saailinge jonger as 5 maande word nie aanbeveel vir hierdie toetse nie. Verdrenking van saailinge het die voorkoms van die siekte verhoog, maar die produksie van groot getalle soëspore was 'n beperkende faktor in die uitvoering van die proef Dit word aanbeveel dat die sand-semel inokulasietegniek verder evalueer moet word onder verskeie toestande, onder andere deur dit met verdrenkinghte kombineer. Die belang van die keuse van isolaat en inokulasiedatum in bepaling van relatiewe weerstand van onderstamme teen P. cactorum is tydens die studie bevestig. Afgesien van die beperking van die staminokulasietegnieke in soverre dit verwyderd is van natuurlike infeksie, word die tegnieke aanbeveel om 'n indikasie te kry van die relatiewe weerstand van onderstamme. Beide die sand-semel en soëspoor tegnieke kan gebruik word om weerstandige saailinge te identifiseer, maar tegniese verfyning van hierdie tegnieke is nodig om saailinge in massa te evalueer.

Full textÖkat befolkningstryck och större efterfrågan av jordbruksprodukter på marknaden har det senasteseklet lett till stora förändringar i västvärldens jordbrukslandskap. Odlingslandskapet är idaghomogent och monokulturellt där besprutningsmedel används för att ge större avkastning iproduktionen. Skadeinsekten havrebladlus (Rhopalosiphum padi) angriper vårgrödor vilket leder tillstora ekonomiska förluster för bönder runtom i Europa. Pesticider används för att reducera angreppenmen påverkar även miljön negativt samt existerande ekosystemtjänster, såsom naturliga fiender sombiologiskt kan begränsa skadeinsekters angrepp. Tidigare studier visar att heterogena landskap har enhög biologisk mångfald, men ingen studie har undersökt åkerholmars (ÅH) eventuella potential förbiologisk bekämpning av skadeinsekter i säd. Denna studie undersöker därför (i) om ÅH leder tillökad reducering av havrebladlöss i säd, (ii) om större ÅH är mer effektiva än mindre i frågan ompestreducering, samt (iii) om större ÅH begränsar lusangrepp längre ut i fält från ÅH:s kant än mindreholmar. Studiens hypoteser testades genom experiment utförda i nio fält med vårgrödor där lössklistrades på etiketter som placerades ut i omgivande fält. Dessa samlades in efter tjugo fyra timmarför att studera antal löss konsumerade i relation till ÅH. Det insamlade materialet är bearbetatstatistiskt och resultatet visar att ÅH spelar en betydande ekologisk roll för naturliga fiender tillskadeinsekten havrebladlus. Fler löss konsumerades runt större ÅH än de med mindre yta eller i de fältutan ÅH. Däremot har avståndet från ÅH en mindre betydelse, vilket indikerar att pestreduceringen ärmest effektiv nära stora ÅH. Denna studie synliggör värdet av existerande ÅH som inhysernyttoinsekter i jordbrukslandskapet, samt att heterogena landskap har högre potential för en mernaturlig skadedjursbekämpning. Naturliga fiender är därmed en existerande ekosystemtjänst som kanbegränsa bladlusangreppen (R.padi), som i sin tur ger möjligheten att reducera användandet avbekämpningsmedel, vilket för oss närmare miljömålet ”en giftfri miljö”.

Full textENGLISH ABSTRACT: Grapevine leafroll-associated virus-3 is one of ten members of the C/osteroviridae that are known to infect grapevine. Nine of these viruses are associated with grapevine leafroll disease, of which GLRaV-1 and GLRaV-3 are the most important and widespread. Members of the C/osteroviridae are unique amongst the viruses, as it is the only known family whose members encode a heat shock protein 70 kOa homolog (Hsp70h). The Hsp70h is a movement protein (MP) that is required for the active translocation of the virion structure through the plasmodesmata into adjacent cells. Broad-spectrum resistance to unrelated viruses can be obtained by a pathogen-derived resistance (POR) strategy that is based on the expression of a dysfunctional MP in plants. The Hsp70h has two distinct domains. The N-terminal two thirds of the protein is an ATPase domain and shares high homology with the ATPase domains of all Hsp70h proteins from the C/osteroviridae and Hsp70 proteins from the prokaryote and eukaryote kingdoms. Conserved amino acids are found in the ATPase domain and are required for the positioning of the ATP at the catalytic site for ATP hydrolysis. The C-terminal domain is variable and the function of this domain in the Closteroviridae is not known. In prokaryote and eukaryote Hsp70 proteins, the C-terminal domain is required for protein-protein interactions. The American NY-1 isolate of GLRaV-3 has been sequenced and POR strategies have been attempted with the coat protein, divergent coat protein and replicase genes, but not with a dysfunctional form of the hsp70h gene. In this study, double-stranded RNA was isolated from a commercial vineyard with unknown virus status, but with distinct grapevine leafroll symptoms, and from two grapevine sources of known virus status, one with mild and one with severe symptoms. The GLRaV-3 hsp70h gene was amplified by RT-PCR from the dsRNA and the gene sequence was analysed. The hsp70h gene from the three virus sources contained more than 94% nucleotide sequence homology to the NY-1 isolate and the conserved amino acids required for ATPase activity were present. The hsp70h gene isolated from GLRaV-3 from a commercial Stellenbosch vineyard showing clear leafroll symptoms was selected for further work and was subjected to site-directed mutagenesis to engineer four point mutations in the gene. These four mutations resulted in the substitution of Asn for Asp", Gly for Thr1O, Lys for Glu 174 and Asn for Asp 197. The wild type (WT) and mutated (Mut) forms of the hsp 70h genes were cloned into a bacterial expression vector. Expression of both the WT- and Mut-Hsp proteins was achieved, and the protein was expressed in the insoluble inclusion bodies. All attempts to refold and isolate active proteins from the inclusion bodies were unsuccessful. Attempts to increase the concentration of soluble protein within the expressing bacteria were unsuccessful. Due to the lack of active protein, biochemical tests on the ATPase activity of the WT- and Mut-Hsp proteins could not be conducted. The wt- and mut-hsp genes were cloned into a plant expression vector for transformation into tobacco plants. These transformations were successful and gave rise to 22 Km' and 18 Km' plants from the WT- and Mut-Hsp constructs respectively. Two plant lines, M5 and M10, transformed with the mut-hsp transgene construct, appeared to have a high level of resistance to the challenging potato X potexvirus, whereas all the other tested plants were susceptible to the challenging virus. It was thus shown that a dysfunctional form of the GLRaV-3 Hsp70h could provide resistance to an unrelated virus in tobacco.
AFRIKAANSE OPSOMMING: Wingerdrolblaar-geassosieerde virus 3 (GLRaV-3) is een van 10 lede van die Closteroviridae wat wingerd kan infekteer. Nege van die virusse is met wingerdrolblaar geassosieer. Die GLRaV-1 en GLRaV-3 is die belangrikste en mees wyd verspreide lede van die rolblaar-geassosieerde Closteroviridae. Lede van die Closteroviridae is uniek in die opsig dat die virusse vir 'n 70 kDa-homoloë hitteresponsproteïen (Hsp70h) kodeer. Die Hsp70 is 'n bewegingsproteïen (MP) wat belangrik is vir die translokasie van die virus deur die plasmodesmata na die naasliggende sel. Die wt- en mut-hsp-gene is ook in In plantekspressievektor gekloneer vir transformasie in tabakplante. Hierdie transformasies was suksesvol en het aanleiding gegee tot 22 kanamisienbestande (Km') en 18 Km' plante vanaf die WT- en Mut-Hspkonstrukte onderskeidelik. Twee plantlyne, M5 en M10, wat met die mut-hsptransgene getransformeer is, het 'n hoë vlak van weerstand teen die infekterende aartappelvirus X getoon in vergelyking met ander plante wat met die virus geïnfekteer is. Daar is dus bewys gelewer dat 'n disfunksionele vorm van die GLRaV-3 Hsp70h weerstand kan bied teen 'n onverwante virus in tabak.

Biological control agents (BCA) have received great recognition, and their use has contributed as a complement or replacement of agrochemicals. However, there are few studies on the biological control of Black spot of citrus, caused by the fungus Guignardia citricarpa, which prevents the market in natura, besides damaging their export by the intensive use of chemical inputs to control this pathogen. Trichoderma fungi are the most applied worldwide BCA against various relevant plant pathogens. The purpose of this study was to identify extracellular proteins secreted by T. atroviride T17 and of T. harzianum T1A, which are effective for the control of G. citricarpa. Bidimensional electrophoresis (2D) allowed obtaining the secreted protein profiles of Trichoderma grown in glucose medium (control) and in medium containing inactivated mycelium of G. citricicarpa. From the 178 differentially expressed proteins by T. harzianum, 68 were identified, most of them related to biocontrol mechanisms, even in the control medium. In the presence of the pathogen, the expression of proteins related to the metabolism decreases. On the other hand, Trichoderma atroviride showed higher expression of proteins related to biocontrol pathogen when grown in the presence of pathogen mycelium, when compared to the control medium. From the 116 differentially expressed proteins, 59 were identified, mainly proteins related to fungi cell wall degradation such as α-mannosidase, chitinase, mutanase, glycosidase, endochitinase and, some families of glycoside hydrolases. The results indicate that these species have a high potential as biocontrol agents of G. citricicarpa. These results are pioneers in to detail the Trichoderrma interaction with G. citricicarpa through the secretome analysis.

Full textA study was performed to develop a generic pest monitoring system for sampling the main table grape pests in vineyards in the Hex River Valley, Western Cape Province of South Africa. The presence of phytophagous and predatory mites on cover crop plants was also investigated as this may contribute to biological control of the phytophagous mites in vines. Life table studies for Epichoristodes acerbella (Walker), an important phytosanitary pest, were conducted to determine whether or not this pest was sensitive to high temperatures. Information gained from the latter can also be used for breeding purposes in the possible future development of a sterile insect technique (SIT) programme to control this pest. The sampling system consisted of inspecting 20 plots of five vines per plot per one to two hectares. The top fork of each of the five vines per plot was examined for Planococcus ficus (Signoret) to a distance of within 30 cm of the stem, as well as the distal 15 cm of one cane per vine for the presence of P. ficus and damage caused by Phlyctinus callosus Boh. One bunch per vine was examined for insect damage or presence, and one leaf per vine for the presence of leaf infesting arthropods, such as Tetranychus urticae Koch, P. ficus and Frankliniella occidentalis (Pergande). Corrugated cardboard bands, tied around the stem of one vine per plot, were used to monitor activity of P. callosus. Blue sticky traps, at a density of four to five traps per one to two hectares, were used to monitor activity of F. occidentalis. Pheromone traps, at a density of one trap per one to two hectares, were used to monitor activity of P. ficus, E. acerbella and Helicoverpa armigera (Hübner). All the above-mentioned inspections were done at two-weekly intervals, except traps for E. acerbella and H. armigera, which were inspected weekly. In each of the rows in which the sample plots were situated, one leaf of each of the cover crop plant species was examined for the presence of phytophagous mites and their predators. The abundance and distribution of cover crop plants were determined using a co-ordinate sampling system. Cover crop sampling was done at monthly intervals. The current threshold for P. ficus is 2% stem infestation, which is reached when more than 65 males per pheromone trap are recorded. Counting mealybugs on the sticky pads in the pheromone traps is time consuming. However, the number of grid blocks on the sticky pad with males present can be counted. When P. ficus males are found in 27 blocks on the sticky pad, stem inspections should commence. Due to the spatial association between P. ficus bunch and stem infestation, stem infestation could give an indication of where bunch infestation could be expected. The use of blue sticky traps for predicting halo spot damage, caused by F. occidentalis, is not recommended. The presence of thrips on the vine leaves could not give an indication of where to expect bunch damage, since thrips on the leaves and halo spot damage were not spatially associated. A suitable sampling method for F. occidentalis still needs to be developed. The monitoring system described here can only provide information on the infestation status of the vineyard. For E. acerbella, H. armigera and P. callosus, the traps and cardboard bands could be used to identify vineyards where these pests are present and therefore, where phytosanitary problems may arise. The presence of P. callosus under the bands was spatially associated with P. callosus damage and could be used as an indicator of the latter. The presence of drosophilid flies in the bunches could not be used as an indicator of the presence of E. acerbella in the bunches. If 5% bunch damage is used as an economic threshold for E. acerbella and P. callosus, there will be a good chance of not under spraying if control measures are applied at 1% bunch damage. Epichoristodes acerbella favoured more moderate constant temperatures, with constant temperatures of 28°C and above being unfavourable for development. The economic threshold for Tetranychus urticae Koch is six mites per leaf, or if presence-absence sampling is used, 11 to 29% leaf infestation. Three important predatory mites, that kept T. urticae under control, were found in the Hex River Valley, namely Euseius addoensis (Van der Merwe & Ryke), Neoseiulus californicus (McGregor) and an undescribed phytoseiid in the genus Typhlodromus. Various cover crop plants served as hosts for T. urticae and predatory mites. The presence of these plants created suitable conditions for the survival of these mites and may have influenced their presence on the vine leaves. In the case of phytosanitary pests, both field and pack shed inspections can be used to conclude with a 99% degree of certainty that infestation levels in the pack shed will be 10% or less, since similar results for both methods were obtained. However, more than 20 plots will have to be inspected.
