This research is a study of pest management systems in Malmö. Malmö is, as many other cities, growing and a node of people's movement. Occurrences of pests have increased in the last decade; this increase is attributed to travel as well as construction and building.What this paper shows is that the structures of pest management must evolve to be more communicative and collaborative. The thesis also adds to the housing crisis debate, as pests in one’s housing have a physical as well as psychological effect.

Abstract:
Antifeedants and growth regulators have potential as alternatives to conventional methods of insect pest control. As with conventional chemicals it is important to consider their effects on natural enemies of the pests. The aim of this project was to determine whether the bitter-tasting synthetic chemical denatonium benzoate as an antifeedant and compare with neem derivatives. Other aims were to investigate their effect on parasitoids and the field infestation of cabbage pests. Denatonium benzoate is systemic and had no contact effect but acted as an antifeedant on a generalist sap feeder Myzus persicae and reduced the survival, fecundity, and lifespan. Neem derivatives, Azatin EC and Pestistat R were more effective of which Azatin EC was the most effective. Antifeedants further reduced the fecundity and survival when parasitized with its parasitoid, Aphidius matricariae and neem derivatives had adverse effects on both aphid and the parasitoid. Denatonium benzoate and Pestistat R with a specialist leaf feeder, Pieris brassicae and all antifeedants with Plutella xylostella (generalist leaf feeder) and Chrysodeixis eriosoma (specialist brassica leaf feeder) reduced the leaf area consumed and it was inversely proportional to the concentration. The highest leaf area consumption was in denatonium benzoate and the lowest in Pestistat R. Higher azadirachtin concentrations were detrimental but denatonium benzoate was safer to the larval parasitoid, Cotesia plutellae. Denatonium benzoate and Azatin EC reduced field infestation of M. persicae, P. xylostella and C. eriosoma in Peradeniya, Sri Lanka and Azatin EC was the most effective. Denatonium benzoate acts as an antifeedant against cabbage pests and safer towards parasitoids hence can be used in integrated pest management programmes. Neem derivatives are comparatively more effective on pests but harmful to parasitoids. Denatonium benzoate and Azatin EC effectively reduced the pest infestation under field conditions.

Abstract:
The purpose of this study was to assess the adoption response of cotton growers to Integrated Pest Management (IPM) programs, identify the extension strategies used to promote IPM, and provide descriptive comparison of IPM programs between the foremost eleven cotton-producing states in the United States. Integrated pest management is presented as an alternative agricultural production strategy referred to as sustainable agriculture. IPM programs for cotton were evaluated by use of a survey instrument and telephone interviews with the IPM coordinators for the eleven cotton-producing states. The most significant findings were that programs vary considerably between states; the most widely employed IPM techniques were selective use of chemicals, use of economic thresholds and sampling for pests; the Cooperative Extension Service worked closely with private consultants and grower associations; and adoption of a wide variety of IPM techniques has been extensive for cotton producers in the eleven foremost cotton-producing states.

Invasive species are alien to the ecosystem under consideration and cause economic or environmental damage or harm to human health. Two alien insects that fit this description are the brown marmorated stink bug, Halyomorpha halys and the spotted lanternfly, Lycorma delicatula. Both invaders are polyphagous pests that feed on a myriad of plant species and inflict severe crop losses. As sustainable control methods depend on the accurate monitoring of species’ invasion and involve the use of natural enemies, we addressed these two facets by exploring novel monitoring techniques and deciphering host-parasitoid interactions for improved integrated pest management. Thus, we adopted ‘BugMap’, a citizen science initiative that enables students, farmers and everyday citizens to report sightings of H. halys from Italy, with emphasis on Trentino-Alto Adige. Aside from fostering citizen participation in scientific endeavors and the enhanced literacy that ensues, BugMap helped uncover the invasion dynamics of H. halys and forecast its potential distribution in Trentino, all while coordinating technical monitoring and informing management strategies. The most promising agent currently under study for the classical biological control of H. halys is the Asian egg parasitoid Trissolcus japonicus. To assess the wasp’s potential non-target impacts, we investigated its foraging behavior in response to chemical traces ‘footprints’ deposited by its main host H. halys and by a suboptimal predatory species, the spined soldier bug, Podisus maculiventris. Wasps exhibited a ‘motivated searching’ when in contact with footprints originating from both species. However, T. japonicus arrestment was significantly stronger in response to H. halys footprints, compared with P. maculiventris, implying the presence of underlying chemical cues that shape its natural preferences. A series of GC-MS chemical analyses revealed that n-tridecane and (E)-2-decenal were more abundant in H. halys footprints and are probably the key components utilized by the wasp for short range host location. The function of the aforementioned compounds was studied, n-tridecane acted as an arrestant, prolonging T. japonicus residence time, whereas (E)-2-decenal fulfilled its presumed defensive role and repelled the wasp. These results shed new light on the chemical ecology of T. japonicus and help expand the understanding of parasitoid foraging and its implications for classical biological control. Moving to the other invader L. delicatula, an egg parasitoid Anastatus orientalis was reported attacking it at high rates in its native range in Eastern Asia and may play a key role in reducing its populations there. A series of bioassays revealed that wasps responded to footprints deposited by L. delicatula gravid females by initiating a strong searching behavior. Moreover, A. orientalis preferred to oviposit in egg masses with intact oothecae, suggesting that the host’s egg covering functions as a trigger for A. orientalis probing and oviposition. Thus, A. orientalis not only overcomes, but also reverses an important line of host structural defense for its own fitness gains. This dissertation discusses the benefits of combining citizen science with traditional monitoring, and the usefulness of decoding host-parasitoid interactions to design more efficacious management strategies of invasive insect pests.

Abstract:
Dasineura sp. is a gall forming midge that was introduced into South Africa for the biocontrol of the Australian myrtle, Leptospermum laevigatum. It causes galls on both the vegetative and reproductive buds of the plant. Although Dasineura sp. was initially regarded as a potentially successful agent, galling up to 99 percent of the buds of the host plant, it has been preyed on by native opportunistic mites, which caused a decline in the performance of the midge as a biocontrol agent of L. laevigatum. This raised a concern about whether this fly will be able to perform effectively in the presence of its new natural enemies. Therefore, the objectives of this study were to: 1) ascertain whether mite abundance has seasonal variations; 2) determine if plant density and plant size have an effect on midge predation by the mites; and 3) determine if midge predation varies in different locations. The study was conducted at three sites in the Hermanus area, Western Cape Province. Every three weeks for thirteen months, galls were collected and dissected so as to count and record the numbers of midge larvae, pupae, adults and mites that were found. Data collected showed that predation varied with season, and the mites were scarce during the flowering season. Predation also varied among the study sites and plant density had an effect on midge predation. Midges in smaller plants (saplings) were more vulnerable to predation than those in the bigger plants (plants from isolates and thickets). It was concluded that although mites have an effect on midge populations, they do not prevent their establishment on the plant. Therefore, a survey should be done in two to three years time to check if the midges are still persisting on the plant, vi and recommendations are that a new agent should be released to supplement the midges.

There are many commercially available sonic pest devices that claim to be effective.Commercially available sonic pest devices for use in residential applications have not been shown to be effective in scientific studies. For this reason, use of these devices is not advised to treat common pest problems. Although some researchers are developing sonic techniques that illustrate promise for very specific pests, these technologies are yet to be commercially available.

Abstract:
Integrated pest management (IPM) is an approach to pest control which emphasizes the Integration of biological, cultural, and chemical control methods for optimal pest management. The purpose of this thesis is to empirically examine the level and distribution of net economic benefits of Extension IPM, and to assess the relative importance of socioeconomic factors in affecting the adoption of IPM in the states of Indiana, Virginia, Georgia, New York, North Carolina, Texas, Massachusetts, Mississippi and the Northwest region. Budgeting and hypothesis testing procedures are used to conduct a net returns analysis. Consumer-producer surplus analysis is used to assess IPM benefits to producers and consumers. Finally, a polychotomous logit model is used to assess the importance of socioeconomic factors affecting IPM adoption. The results of these analyses show significantly higher returns and less variability of returns per acre for users of IPM as compared to non-users. Moreover, consumers receive significant positive economic gains. However, pesticide cost and the variance of pesticide cost per acre increase with increasing levels of IPM use in several states but decrease in a few others. Gross farm income, percent family income from farming, frequency of contacts with Extension agents, and the education level of respondents are the most important factors related to adoption of IPM. A typical user of IPM is white, male, with at least some college education, has frequent contacts with Extension agents, has a relatively large farm, higher gross farm income, and a higher percent family income from farming.

Abstract:
The biology of key citrus pests Thaumatotibia leucotreta Meyrick (Lepidoptera: Tortricidae), Ceratitis capitata Wiedemann (Diptera: Tephritidae) and Ceratitis rosa Karsch (Diptera: Tephritidae) includes their dropping from host plants to pupate in the soil below citrus trees. Since most EP fungi are soil-borne microorganisms, the development and formulation of alternative control strategies using these fungi as subterranean control agents, targeted at larvae and pupae in the soil, can potentially benefit existing IPM management of citrus in South Africa. Thus, a survey of occurrence of entomopathogenic fungi was undertaken on soils from citrus orchards and natural vegetation (refugia) on conventionally and organically managed farms in the Eastern Cape Province in South Africa. A method for baiting soil samples with citrus pest T. leucotreta and C. capitata larvae, as well as with the standard bait insect, Galleria mellonella Linnaeus (Lepidoptera: Pyralidae), was implemented. Sixty-two potentially useful entomopathogenic fungal isolates belonging to four genera were collected from 288 soil samples, an occurrence frequency of 21.53%. The most frequently isolated entomopathogenic fungal species was Beauveria bassiana (Balsamo) Vuillemin (15.63%), followed by Metarhizium anisopliae var. anisopliae (Metschnikoff) Sorokin (3.82%). Galleria mellonella was the most effective insect used to isolate fungal species (χ2=40.13, df=2, P≤ 0.005), with a total of 45 isolates obtained, followed by C. capitata with 11 isolates, and T. leucotreta with six isolates recovered. There was a significantly (χ2=11.65, df=1, P≤ 0.005) higher occurrence of entomopathogenic fungi in soil samples taken from refugia compared to cultivated orchards of both organically and conventionally managed farms. No significant differences were observed in the recovery of fungal isolates when soil samples from both farming systems were compared. The physiological effects and host range of 21 indigenous fungal isolates obtained in the Eastern Cape were investigated in the laboratory to establish whether these isolates could be effectively used as biological control agents against the subterranean life stages of C. rosa, C. capitata and T. leucotreta. When these pests were treated with a fungal concentration of 1 x 10⁷ conidia ml⁻¹, the percentage of T. leucotreta adults which emerged in fungal treated sand ranged from 5 to 60% (F=33.295; df=21; P=0.0001) depending on fungal isolate and the percentage of pupae with visible signs of mycosis ranged from 21 to 93% (F= 96.436; df=21; P=0.0001). Based on fungal isolates, the percentage adult survival in C. rosa and C. capitata ranged from 30 to 90% and 55 to 86% respectively. The percentage of C. rosa and C. capitata puparia with visible signs of mycosis ranged from 1 to 14% and 1 to 11% respectively. Deferred mortality due to mycosis in C. rosa and C. capitata adult flies ranged from 1 to 58% and 1 to 33% respectively, depending on fungal isolate. Entomopathogenic fungal isolates had a significantly greater effect on the adults of C. rosa and C. capitata than they did on the puparia of these two fruit fly species. Further, C. rosa and C. capitata did not differ significantly in their response to entomopathogenic fungi when adult survival or adult and pupal mycosis were considered. The relative potency of the four most virulent Beauveria isolates as well as the commercially available Beauveria bassiana product, Bb Plus® (Biological Control Products, South Africa), were compared against one another as log-probit regressions of mortality against C. rosa, C. capitata and T. leucotreta which all exhibited a dose-dependent response. Against fruit flies the estimated LC50 values of all five Beauveria isolates ranged from 5.5 x 10¹¹ to 2.8 x 10¹² conidia/ml⁻¹. There were no significant differences between the relative potencies of these five fungal isolates. When T. leucotreta was considered, isolates: G Moss R10 and G 14 2 B5 and Bb Plus® were significantly more pathogenic than G B Ar 23 B3 and FCM 10 13 L1. The estimated LC₅₀ values of the three most pathogenic isolates ranged from 6.8 x 10⁵ to 2.1 x 10⁶ conidia/ml⁻¹, while those of the least pathogenic ranged from 1.6 x 10⁷ to 3.7 x 10⁷ conidia/ml⁻¹. Thaumatotibia leucotreta final instar larvae were exposed to two conidial concentrations, at four different exposure times (12, 48, 72 and 96 hrs) and showed an exposure time-dependant relationship (F=5.43; df=3; P=0.001). At 1 x 10⁷conidia/ml⁻¹ two Beauveria isolates: G Moss R10 and G 14 2 B5 were able to elicit a response in 50% of test insects at 72 hrs (3 days) exposure. Although a limited amount of mycosis was observed in the puparia of both fruit fly species, deferred adult mortality due to mycosis was high. The increased incidence of adult mortality suggests that post emergence mycosis in adult fruit flies may play a more significant role in field suppression than the control of fruit flies at the pupal stage. The increased incidence of pupal mortality, as well as the relatively low concentrations of conidia required to elicit meaningful responses in T. leucotreta pupae may suggest that pre-emergent control of false codling moth will play a more significant role in field suppression than the control of adult life stages using indigenous isolates of entomopathogenic fungi. Various entomopathogenic fungal application techniques targeted at key insect pests within integrated pest management (IPM) systems of citrus are discussed.

Commercially available sonic pest devices for use in residential applications have not been shown to be effective in scientific studies. For this reason, use of these devices is not advised to treat common pest problems. Although some researchers are developing sonic techniques that illustrate promise for very specific pests, these technologies are yet to be commercially available. As our understanding increases of how pest species receive and process sound, more relevant sonic devices may be developed. The allure of sound as a treatment for pests will remain into the future—motivated by the fact that if they are successful they will be more environmentally friendly and safer for humans.

ENGLISH ABSTRACT: As overpopulation threatens the world’s ability to feed itself, food has become an invaluable resource. Unfortunately, almost a third of the food produced for human consumption is lost annually. Pests including insects, phytopathogens and weeds are responsible for more than a third of the annual major crop losses suffered around the world. The majority of current post-harvest control strategies employ synthetic agents. These compounds, however, have been found to be detrimental to the environment as well as human health, which has led researchers to investigate alternative strategies. Biocontrol agents are environmentally compatible, have a lower toxicity and are biodegradable, making them an attractive alternative to the synthetic control agents. The lipopeptides produced by Bacillus spp. in particular, have shown great potential as biocontrol agents against various post-harvest phytopathogens. Most biocontrol strategies apply the biocontrol organism, for example Bacillus, directly, whereas this study focused on the use of the lipopeptide itself as an antifungal agent. This is advantageous as the lipopeptides are less sensitive to their surroundings, such as temperature and pH, compared to living organisms, allowing for the production of a standardized product. This study investigated the production of the Bacillus lipopeptides surfactin, fengycin and iturin under controlled batch conditions. Parameters increasing lipopeptide production were quantified, focussing on antifungal lipopeptides (iturin and fengycin), and lipopeptide production was optimized. Experiments were performed in a fully instrumented 1.3 L bench-top bioreactor and lipopeptide analyses were performed via high pressure liquid chromatography (HPLC) and liquid chromatography-mass spectroscopy (LC-MS). After screening four Bacillus spp., Bacillus amyloliquefaciens DSM 23117 was found to be the best antifungal candidate. This was based on it outperforming other candidates in terms of maximum antifungals produced, Yp/x,antifungals (yield per cells), and antifungal productivity. Nitrate, in the form of NH4NO3, was critical for lipopeptide production and an optimum concentration was observed above which the CDW (cell dry weight) no longer increased significantly and both μmax (maximum specific growth rate, h-1) and lipopeptide production decreased. For μmax, the optimum NH3NO4 concentration was 10 g/L and for lipopeptides it was 8 g/L. At these respective NH4NO3 concentrations μmax = 0.58 (h-1), the maximum antifungals (fengycin and iturin) were 285.7 mAU*min and the maximum surfactin concentration was 302 mg/L. The lipopeptides produced by B. amyloliquefaciens, the antifungals (fengycin and iturin) and surfactin, are secondary metabolites, regardless of the optimization treatment, i.e. increased NH4NO3 concentrations. Using 30% enriched air extended the nitrate utilization period, suggesting that when increasing supply concentration, more oxygen was available to act as electron acceptors, allowing nitrate to be used for lipopeptide production. The number of iturin and fengycin homologues generally increased with an increase in nitrate concentration. This suggested that process conditions, such as nitrate concentration, can be used to manipulate homologue ratios, allowing for the possibility to tailor-make biocontrol-agent upstream, during the production process, and possibly increase the efficacy of the biocontrol strategy. The lipopeptides produced by B. amyloliquefaciens showed complete inhibition against Botryotinia fuckeliana and diminished the growth capabilities of Botrytis cinerea. No inhibition was observed against Penicillium digitatum. These results indicate potential of the biocontrol strategy, although scale-up and fed-batch studies are recommended, especially when considering commercial implementation. Studies regarding the lipopeptide application method, i.e. a single application or multiple applications, should also be investigated as this will influence the efficacy of the lipopeptides against the target organisms.
AFRIKKANSE OPSOMMING: Met oorbevolking wat die wêreld se vermoë om die groeiende bevolking te onderhou belemmer, het dit noodsaaklik geword om huidige voedselbronne te beskerm. Daar word beraam dat een derde van die voedsel wat wêreldwyd geproduseer word vir menslike verbruik verlore gaan elke jaar. Verder is insekte, plantpatogene en onkruide verantwoordelik vir meer as ‘n derde van die verliese rakend jaarlikse oeste. Meeste bestaande na-oes beheermetodes maak gebruik van sintetiese stowwe. Ongelukkig kan hierdie verbindings nadelig wees vir die omgewing sowel as menlike gesondheid. Navorsers het hulsef dus toespits daarop om alternatiewe beheermetodes te ondersoek. Bio-beheermetodes is omgewingsvriendelik sowel as bio-afbreekbaar, wat hulle ideale alternatiewe maak vir die sintetiese stowwe. Bacillus spp. lipopeptiede het veral hoë potensiaal getoon as bio-beheermiddels teen verskeie na-oes plantsiektes. Die optimale NH4NO3 konsentrasie vir μmax was 10 g/L en vir lipopeptiedproduksie was 8 g/L. By 10 g/L NH4NO3 was μmax = 0.58 (h-1) en by 8 g/L was die maksimale antifungiese produksie (fengycin en iturin) 285.7 mAU*min en die maksimale surfactin produksie 302 mg/L onderskeidelik. Die lipopeptide, die antifungiese middels (fengycin en iturin) en surfactin, geproduseer deur B. amyloliquefaciens is sekondêre metaboliete, ongeag van die optimerings-behandelinge wat toegepas word, soos ‘n verhoging in NH4NO3 konsentrasie. Veral in kombinasie met chemiese middels, as deel van geïntegreerde programme, kan dit lei tot verminderde gebruik van hoogs toksiese middels en dus meer omgewingsvriendelike landboupraktyke. Biological control of plant parasitic nematodes on potatoes, citrus and grapevine with the fungus, Paecilomyces liIacinus. Paecilomyces liIacinus, race 251 (registered in terms of act 36 of 1947 as South Africa's first natural nematicide, commercially available as PI Plus) was tested as a biological control agent on potatoes and in integrated control programs on citrus and grapevine against Me/oidogyne species, Ty/enchu/us semipenetrans and various ectoparasitic nematodes respectively. The fungus shows promise for the control of these nematodes, without having a harmful effect on non-target, beneficial organisms in the soil. Especially in combination with chemical products, as part of integrated programs, this can lead to less use of highly toxic compounds and thus to more environmentally friendly agricultural practices.
AFRIKAANSE OPSOMMING: Sedert die ontdekking van die swam, Paeci/omyces Ii/acinus (Thom.) Samson as 'n effektiewe eierparasiet van Meloidogyne incognita acrita en Globodera pal/ida (Jatala et al., 1979) het verdere veldproewe in Peru tot die effektiewe beheer van M. incognita en Tylenchulus semipenetrans gelei. Na verskeie suksesse in Peru is die swam onder verskillende klimaat- en grondkondisies in verskeie ander lande beproef. Die sukses behaal in die Filippyne het gelei tot die kommersiële produksie van die swam onder die handelsnaam Biocon. Anders as met chemiese middels vind die werking van biologiese agente stadig oor tyd plaas. Biologiese beheer sal nie chemiese beheer sonder meer kan vervang nie. Dit behoort egter deel te vorm van geïntegreerde nematode bestuur. Abstract:
Les propietats insecticides d’una sèrie d’esters d’al•lil va ésser avaluada en diferents insectes mitjançant diferents modes d’aplicació. L’acció per aplicació tòpica va variar en funció de l’ester d’al•lil aplicat. El cinamat i el naftoat d’al•lil van ser els compostos més actius front a ous i larves neonates de C. pomonella, G. molesta i L. botrana, mentre que el salicilat d’al•lil no va produir mortalitat a la major dosi assajada (10 mg/mL). El cinamat d’al•lil va ser l’únic ester actiu per aplicació tòpica en adults de A. pisum, mentre que tots els esters d’al•lil testatsvan ser actius en adults de T. castaneum. Els esters d’al•lil assajats van produir una pèrdua de la viabilitat cel•lular en les línees cel•lulars d’insectes quan va èsser mesurda mitjançant dos metodologies diferents (MTT i Blau de tripà), degut a una disrupció de les membranes cel•lulars. El cinamat d’al•lil va ser el compost més actiu i les cèl•lules procedents de l’aparell digestiu de Choristoneura fumiferana (Lepidòpter) les més sensibles. L’acció insecticida per ingestió va ser confirmada en larves de S. littoralis i C. pomonella, i en nimfes de A. pisum, senyalant l’aparell digestiu com a principal punt d’acció dels esters d’al•lil. Els corresponents àcids i dicloropropilesters van mostrar una menor o igual acció insecticida que els corresponents esters d’al•lil, deguda també, a una acció en la membrana cel•lular. Les diferències en l’acció dels compostos seria deguda a diferències en les propietats lipofíliques dels compostos i la seva interacció amb les membranes cel•lulars. Pel que fa a l’efecte dels esters d’al•lil en la comunicació química dels insectes, aquesta va tenir lloc en T. castaneum però no en A. pisum, el que es podria utilitzar per a mantenir els productes enmagatzemats lliures de T. castaneum. Pel que fa a C. pomonella i a L. botrana, tots els esters d’al•lil assajats van produir una resposta en les antenes dels mascles de C. pomonella, però tan sols el cinamat d’al•lil la va provocar en les femelles de C. pomonella i en mascles i femelles de L. botrana. Aquesta acció no es va veure reflexada en un increment de l’atracció de mascles cap a fonts amb l’ester d’al•lil i feromona en assajos en túnel de vent, però si en l’atracció de femelles. Aquest fet podria utilitzar-se per a incrementar el nombre de captures de femelles en trampes de feromona. Aquests resultats suggereixen un paper dels esters d’al•lil en el control de plagues, especialment del cinamat d’al•lil.
La acción insecticida de varios esteres de alilo fue testada en diversos insectos y mediante distintos modos de aplicación. La actividad por aplicación tópica varió en función del éster de alilo. El cinamato de alilo y el naftoato de alilo fueros los compuestos más activos en huevos y larvas neonatas de C. pomonella, G. molesta y L. botrana, mientras que el salicilato de alilo no produjo mortalidad a la dosis más alta testada (10 mg/mL). El cinamato de alilo fue el único éster activo por aplicación tópica en A. pisum mientras que todos los esteres testados lo fueron para T. castaneum. Los esteres de alilo estudiados produjeron pérdida de viabilidad celular en todas las líneas celulares de insectos cuando dicha viabilidad fue analizada mediante dos metodologías distintas (MTT y Azul de Tripano), y siendo ésta debida a la disrupción de la membrana celular. El cinamato de alilo fue el producto más activo, y las células del aparato digestivo de Choristoneura fumiferana (Lepidoptera) las más sensibles. La acción insecticida por ingestión en larvas de S. littoralis y C. pomonella, y en ninfas de A. pisum, fue confirmada y el aparato digestivo fue señalado como principal punto de acción de los esteres de alilo. Los correspondientes ácidos y dicloropropilesteres presentaron una menor o igual acción insecticida que los esteres de alilo siendo dicha acción también debida a un efecto en la membrana celular. Las diferencias en la acción de los distintos compuestos podrían ser debidas a diferencias en las propiedades lipofílicas de los compuestos y su interacción con las membranas celulares. Los esteres de alilo produjeron un efecto en la comunicación química de T. castaneum pero no en A. pisum, lo que podría utilizarse para mantener los productos almacenados libres de T. castaneum. En cuanto a C. pomonella y L. botrana, todos los esteres de alilo probados produjeron una respuesta en las antenas de los machos de C. pomonella, mientras que tan solo el cinamato de alilo la produjo en las antenas de hembras de C. pomonella y en machos y hembras de L. botrana. Esta respuesta no se tradujo en un aumento de la atracción de machos hacia cebos con mezclas de ester de alilo y feromona en ensayos de túnel de viento, pero si aumentó el número de hembras atraídas. Este hecho podría utilizarse par incrementar el número de hembras capturadas en trampas de feromona. Estos resultados, sugieren el papel de los esteres de alilo en el control de plagas, especialmente del cinamato de alilo.

Abstract:
In the management of agricultural insect pests, short-term costs must be balanced against long-term benefits. Controls should be selected to account for both their immediate and downstream effects upon the demography and genetics of the pest, enabling suppression today without threatening suppression tomorrow. The iterative, algorithmic method of dynamic programming can provide optimal solutions to problems of this type, in which actions are taken sequentially and each action may influence those which follow it. However, this approach is fundamentally constrained with regards to the magnitude of the problems it may solve. As questions of insect pest management can be subject to ecological and evolutionary complexities, this may place them beyond the scope of dynamic programming. When it is the intricacies of a problem that are of interest, it may be more productive to utilise approximate dynamic programming (ADP) methods which can attempt problems of arbitrary complexity, although at the expense of no longer guaranteeing optimality. In this thesis I first challenge a dynamic programming algorithm with the management of a hypothetical insect pest feeding upon a transgenic insecticidal crop. The model explores how different realisations of fitness costs to resistance influence the algorithms suggested actions. I then apply a brute-force variant of ADP, a lookahead policy, to the management of a stage-structured, continuously reproducing pest population. This was to explore the extent to which an algorithm with a limited temporal perspective is able to balance the timetable of pest demography against the timescale over which insecticidal sprays and bisex-lethal sterile insect releases unfold. This same decision framework is then applied to a modified problem in which resistance to insecticidal toxins may evolve and releases are now male-selecting. This was used to assess the efficacy with which simple lookahead policies utilise a control with delayed benefits (the male-selecting releases) and possible constraints on their capacity to respond to resistance evolution. Dynamic programming and ADP methods offer a versatile toolbox for accounting for the potential impacts of the evolutionary and ecological peculiarities of particular pests upon control decisions.

The results presented in this dissertation have been partially presented at scientific meetings. Chapter 2 is an exact copy of the technical contents of a paper submitted to a scientific journal. The format of this paper however has been changed to comply with the format used in this dissertation. The results of this paper were partially presented at the 36th National Congress of the South African Society of Animal Science, 5 – 8 April 1998 held in Stellenbosch. In this chapter errors occurred in the execution of the trial, but these were corrected in the follow-up trial after consulting with the manufacturers. It is discussed in the paper. Results of chapter 3 were partially presented at the 5th International Sheep Veterinary Congress 21 – 25 January 2001 held in Stellenbosch and at an International Congress (The FLICS (Flystrike & Lice IPM Control Strategies) Conference, 25 – 27 June 2001, Launceston) in Tasmania by Mr. S.W.P. Cloete in 2001. The updated results are given in this chapter. The results of chapter 4 were presented at a combined congress between the Grasslands and the Animal Production Societies of South Africa (The GSSA/SASAS Joint Congress 2002. 13 – 16 May) in Christiana in 2002. The results in this chapter represent the same experimental period as was reported upon at that congress, but additional statistical analyses were executed on the same data set. Copies of the abstracts of the contributions to these meetings are attached in chapter 6. The major objective of this study initially was to evaluate the Lucitrap® system under South African conditions. During the experimental period I had an opportunity to visit Australia and meet many role players. It came under my attention that the control of the blowfly is a rather complex problem with many facets. A current issue worldwide on the use of chemicals as a control method against insects and its hazardous influence on the environment is forcing producers to minimize chemical residues in textiles and other agricultural products. An Integrated Pest Management (IPM) approach seems to be the only option. This study has been executed to try and understand some aspects of the blowfly problem for future application in such a blowfly control program.

ENGLISH ABSTRACT: Phytophthora cactorum (Lebert & Cohn) Schrot. is the primary cause of crown, collar and root rot diseases of apple (Malus domestica Borkh.) trees worldwide. This pathogen is most destructive in commercial apple orchards under waterlogged soil conditions and has recently been identified as causing serious disease in some South African apple orchards. Crown, collar and root diseases are difficult to control because of their unpredictability and catastrophic nature. The use of resistant cultivars and rootstocks is economical and environmentally considerate. Therefore the need to develop screening techniques that will enable the selection of desirable disease resistant traits as part of an apple-breeding program in South Africa was identified. The work undertaken in this study was aimed at optimizing different techniques to test resistance. Using two direct inoculation techniques (excised stem and intact stem) the aggressiveness of lO isolates of P. cactorum on apple rootstocks was determined. The susceptibilities of five apple rootstocks were also compared. Results have shown isolate by rootstock interaction which means isolate aggressiveness was influenced by rootstocks tested. The selectivity of isolates suggests that there may be several strains of the pathogen. Population studies of the pathogen might contribute valuable information that could lead to better interpretation of results. Rootstock susceptibility was monitored in vitro throughout the season by inoculating at monthly intervals for 26-months. It was observed that during winter, rootstock susceptibility was low compared to high susceptibility during summer. These results have revealed new information regarding changes in the relative resistance of the different rootstocks over the growing season, e.g. the susceptibility pattern of rootstock MMl06 occurred 1 to -2 months later than that of other rootstocks. This finding has important implications on the way in which resistance test results are interpreted, and emphasizes the importance of not relying on point sampling. Furthermore, useful information has been acquired regarding the epidemiology of the disease with regard to "windows of susceptibility". The phenomenon of a phase shift in susceptibility of different rootstocks needs to be tested on a broader scale to assess whether it has any practical application on resistance testing. Although different inoculation techniques are applied in breeding programs, up to now there is no consensus on which technique works best for seedling selections. Since large numbers of individuals must be tested to improve the chances of detecting resistant genotypes, mass inoculations of young seedlings is a rapid way of identifying resistant individuals. Two different screening methods were tested during this study. Using the sand-bran technique, seedlings were transplanted onto inoculated soil and the root mass was used as a measure of resistance. In a second method zoospore inoculum was applied to seedlings growing in a sand:bark mixture at different concentrations and the seedlings were subjected either to water drenching or not. In both trials the aggressiveness of isolates differed significantly from each other and only higher inoculum concentrations were effective in causing disease. The age of seedlings used in tests emerged as an important factor. Seedlings under five-months-old should not be used. Drenching inoculated seedlings enhanced disease development but the production of sufficiently high numbers of zoospores was a laborious task. Thus, it is recommended that the sand-bran inoculum technique be tested with the drenching treatment for mass selection. In conclusion this study confirms the importance of both choice of isolate and choice of inoculation intervals in determining susceptibility of rootstocks to infection. In spite of the fact that stem inoculation bioassays have limited resemblance to natural disease situations, these bioassays are useful for obtaining an indication as to whether genotypes have a degree of resistance and merit further testing. For this reason refinement of the stem inoculation bioassay is worthwhile pursuing. With regard to seedling trials, both the sand-bran and the zoospore technique appear promising but refinement of these techniques is necessary in order to present a more practical way of testing large volumes of seedlings.
AFRIKAANSE OPSOMMING: Evaluering van inokulasietegnieke om weerstand teen Phytophthora cactorum in appels te evalueer: Phytophthora cactorum (Lebert & Cohn) Schrot. is die primêre oorsaak van kroon-, kraag en wortelvrot van appelbome (Malus domestica Borkh.). Dit is die mees verwoestende patogeen in kommersiële appelboorde waar daar versuipte toestande grond voorkom. P. cactorum is onlangs identifiseer as die patogeen wat ernstige kroon- en kraag-verotting in Suid Afrikaanse appelboorde veroorsaak. Kroon-, kraag- en wortelvrot is moeilik om te beheer as gevolg van die onvoorspelbaarheid en rampspoedige aard van die siekte. Die gebruik van kultivars en onderstamme wat weerstandbiedend is teen siektes en plae is omgewingsvriendelik en is ekonomies van belang, dus het die behoefte ontstaan om inokulasietegnieke te ontwikkelom weerstandige saailinge te identifiseer en te selekteer as deel van 'n appelteelprogram in Suid Afrika. Die doelwit van hierdie studie is om verskillende inokulasietegnieke te toets en te verfyn om weerstand in appelsaailinge te identifiseer. Deur gebruik te maak van twee inokulasietegnieke (die afgesnyde loot- en intakte loot tegniek), is die relatiewe aggressiwiteit van 10 isolate van P. cactorum en die vatbaarheid van vyf appelonderstamme ondersoek. Resultate het aangetoon dat die aggressiwiteit van die isolate gevarieer het na aanleiding van die onderstam wat getoets is. Die selektiwiteit van die isolate is 'n aanduiding dat daar moontlik verskeie rasse van die patogeen voorkom. Toekomstige studies op die populasiestruktuur van P. cactorum sal 'n belangrike bydrae maak tot die interpretasie van resultate oor weerstand en weerstandsteling. Die vatbaarheid van onderstamme was ook in in vitro proewe ondersoek deur maandelikse inokulasies toe te pas oor 'n tydperk van 26 maande. Dit is opgemerk dat die onderstamvatbaarheid gedurende die winter laag was in vergelyking met die somer. Nie al die onderstamme het dieselfe gereageer gedurende verskillende toetstye nie. Verdrenking van saailinge het die voorkoms van die siekte verhoog, maar die produksie van groot getalle soëspore was 'n beperkende faktor in die uitvoering van die proef Dit word aanbeveel dat die sand-semel inokulasietegniek verder evalueer moet word onder verskeie toestande, onder andere deur dit met verdrenkinghte kombineer. Die belang van die keuse van isolaat en inokulasiedatum in bepaling van relatiewe weerstand van onderstamme teen P. cactorum is tydens die studie bevestig. Afgesien van die beperking van die staminokulasietegnieke in soverre dit verwyderd is van natuurlike infeksie, word die tegnieke aanbeveel om 'n indikasie te kry van die relatiewe weerstand van onderstamme. Beide die sand-semel en soëspoor tegnieke kan gebruik word om weerstandige saailinge te identifiseer, maar tegniese verfyning van hierdie tegnieke is nodig om saailinge in massa te evalueer.

Control of the citrus pest, false codling moth (FCM), Thaumatotibia leucotreta Meyrick (Lepidoptera: Tortricidae) is crucial for the South African citrus industry. The economic losses and phytosanitary status of this pest, coupled with increased consumer awareness and demands, has created a need for effective, IPM-compatible control measures for use against the soil-dwelling life stages of FCM. Promising developments in the field of microbial control through the use of entomopathogenic fungi (EPF) and entomopathogenic nematodes (EPNs) have highlighted the need for research regarding pupation biology, behaviour and survival of FCM, as a good understanding of biology of the target organism is an important component of any biological control programme. The aim of this study was to improve the current understanding of FCM pupation habits through the manipulation of soil texture class, ground cover, shading, soil compaction, air temperature, and soil moisture in the laboratory. These findings would then be used to aid the biological control programmes using EPF and EPNs against FCM in the soil. Three soil texture classes (sandy loam, silt loam and silty clay loam) were obtained from orchards for use in the study. FCM larvae were allowed to drop into the soil of their own accord and the pupation behaviour that followed was then captured on film with pupae formed in the soil being kept in order to measure adult eclosion. In general, very few abiotic factors had a clear influence on FCM pupation. Larval wandering time and distance was short, but also variable between individuals. Distance did increase when soils were moist. Pupation depth was shallow, with pupal cocoons generally being formed on the soil surface. Depth of pupation was less than one centimetre for all abiotic conditions, with little burrowing into soil. Eclosion success was higher for sandier soils when these were dry and uncompacted, but the addition of both moisture and soil compaction increased FCM eclosion success. FCM was sensitive to desiccation when the soils were dry and temperature limits of 15 °C and 32 °C had a strongly negative impact on eclosion success. Preferences for particular abiotic conditions were limited to only certain moisture conditions when interacting with soil texture class and a preference for pupating in soil when it is available. Limited preference was found for particular soil textures despite this having a strong influence on eclosion success, but individuals did appear to pupate in close proximity to one another. Viable direct habitat manipulation for FCM control could not be identified. These results and all of the abiotic variables measured have important implications for EPF and EPN application, survival and persistence in the soil in order to improve the ability of these biological control agents to control FCM. These are discussed in each chapter.

Abstract:
False codling moth (FCM), Thaumatotibia leucotreta is an extremely important pest of citrus in South Africa and with the shift away from the use of chemicals, alternate control options are needed. One avenue of control which has only recently been investigated against the soil-borne life stages of FCM is the use of entomopathogenic fungi (EPF). In 2009, 12 entomopathogenic fungal isolates collected from South African citrus orchards showed good control potential during laboratory conducted bioassays. The aim of this study was to further analyse the potential of these isolates through concentration-dose and exposure-time response bioassays. After initial re-screening, concentration-dose response and exposure-time response sandconidial bioassays, three isolates were identified as exhibiting the greatest control potential against FCM in soil, Metarhizium anisopliae var. anisopliae (G 11 3 L6 and FCM Ar 23 B3) and Beauveria bassiana (G Ar 17 B3). Percentage mycosis was found to be directly related to fungal concentration as well as the amount of time FCM 5th instar larvae were exposed to the fungal conidia. LC50 values for the three isolates were not greater than 1.92 x 10⁶ conidia.ml⁻ₑ and at the LC₅₀, FCM 5th instar larvae would need to be exposed to the fungus for a maximum of 13 days to ensure a high mortality level. These isolates along with two commercially available EPF products were subjected to field persistence trials whereby net bags filled with a mixture of autoclaved sand and formulated fungal product were buried in an Eastern Cape citrus orchard. The viability of each isolate was measured on a monthly basis for a period of six months. All isolates were capable of persisting in the soil for six months with the collected isolates persisting far better than the commercially used isolates. Two of the isolates, G 11 3 L6 and G Ar 17 B3, were subjected to small scale laboratory application trials. Two formulations were investigated at two concentrations. For each isolate, each formulation and each concentration, FCM 5th instar larvae were applied and allowed to burrow into the soil to pupate before fungal application or after fungal application. Contact between fungi and FCM host is essential as, in contrast to pre-larval treatments, percentage mortality in post-larval treatments was low for both formulations and both isolates. For isolate G Ar 17 B3, a conidial suspension applied as a spray at a concentration of 1 x 10⁷ conidia.ml⁻ₑ obtained the highest percentage mortality (80 %). For isolate G 11 3 L6 however, both formulations performed equally well at a high, 1 x10⁷ conidia.ml⁻ₑ concentration (conidial suspension: 60 %; granular: 65 %) The results obtained thus far are promising for the control of FCM in citrus, but if these EPFs are to successfully integrate into current FCM control practices more research, some of which is discussed, is essential

During last century rising populations and changed market preferences have led to large structuralchanges in agriculture in the developed world. Most conventional cultivation methods involve theproduction of few monoculture crops within a homogenous landscape, where pesticides and inorganicfertilizers are used to increase yields. The cereal aphid Rhopalosiphum padi (L.) is a pest in cerealsresulting in large economic losses for farmers throughout Europe, and are currently removed withchemical pest control. However, the pesticides also affect existing ecosystem services like potentiallypest-controlling insects as well as the surrounding environment negatively. According to previousstudies higher landscape diversity leads to a higher diversity of natural enemies to pest insects, but nostudy has investigated if the pest control is enhanced around non-crop remnants such as midfield islets(MI). This thesis investigates (i) if MI will reduce the number of aphids in crop field, (ii) if larger MIwill be more effective than smaller MI by having higher aphid predation rate, and (iii) if larger MI willprovide a more effective reduction of pest insects over a longer distance from edge. To assess thepotential for pest control, aphids were glued on papers and placed at ground level in crop fields andaround MI in nine fields. The collected data was processed statistically, and the result demonstratesthat MI play an important role by providing habitats for natural enemies to pests. The increased size ofMI have a positive effect of number of aphids consumed around MI, however the distance from edgewas not significance. This study highlights the value of MI in crop fields as an existing ecosystemservice in biological pest control. The naturally occurring predators are able to reduce R.padi andthereby minimize the need for insecticide applications.
Ökat befolkningstryck och större efterfrågan av jordbruksprodukter på marknaden har det senasteseklet lett till stora förändringar i västvärldens jordbrukslandskap. Odlingslandskapet är idaghomogent och monokulturellt där besprutningsmedel används för att ge större avkastning iproduktionen. Skadeinsekten havrebladlus (Rhopalosiphum padi) angriper vårgrödor vilket leder tillstora ekonomiska förluster för bönder runtom i Europa. Pesticider används för att reducera angreppenmen påverkar även miljön negativt samt existerande ekosystemtjänster, såsom naturliga fiender sombiologiskt kan begränsa skadeinsekters angrepp. Tidigare studier visar att heterogena landskap har enhög biologisk mångfald, men ingen studie har undersökt åkerholmars (ÅH) eventuella potential förbiologisk bekämpning av skadeinsekter i säd. Denna studie undersöker därför (i) om ÅH leder tillökad reducering av havrebladlöss i säd, (ii) om större ÅH är mer effektiva än mindre i frågan ompestreducering, samt (iii) om större ÅH begränsar lusangrepp längre ut i fält från ÅH:s kant än mindreholmar. Studiens hypoteser testades genom experiment utförda i nio fält med vårgrödor där lössklistrades på etiketter som placerades ut i omgivande fält. Dessa samlades in efter tjugo fyra timmarför att studera antal löss konsumerade i relation till ÅH. Det insamlade materialet är bearbetatstatistiskt och resultatet visar att ÅH spelar en betydande ekologisk roll för naturliga fiender tillskadeinsekten havrebladlus. Fler löss konsumerades runt större ÅH än de med mindre yta eller i de fältutan ÅH. Däremot har avståndet från ÅH en mindre betydelse, vilket indikerar att pestreduceringen ärmest effektiv nära stora ÅH. Denna studie synliggör värdet av existerande ÅH som inhysernyttoinsekter i jordbrukslandskapet, samt att heterogena landskap har högre potential för en mernaturlig skadedjursbekämpning. Naturliga fiender är därmed en existerande ekosystemtjänst som kanbegränsa bladlusangreppen (R.padi), som i sin tur ger möjligheten att reducera användandet avbekämpningsmedel, vilket för oss närmare miljömålet ”en giftfri miljö”.

Maize contributes substantially to food security in the Eastern Cape province of South Africa. It is a staple food to many of the province’s rural and urban inhabitants. Insect pests are one of the factors that hamper its productivity and as a result, deprive farmers of good yields. The adverse effects of insecticides and the high cost associated with them and the cost of transgenic seeds are some of the challenges faced by small-scale farmers in rural areas. Alternative control methods which include the use of indigenous techniques to control pests are now sought. A study to identify and document ethnobiological means used by rural farmers to manage insect pests of maize was conducted in the rural areas of the Eastern Cape Province. A total of 217 participants were interviewed on the matter, using semi-structured but detailed questionnaires. Rural farmers due to their linkage to agriculture activities and the fact that they are considered as custodians of agricultural indigenous knowledge were selected as respondents. Only maize producing and IsiXhosa speaking people were chosen to contribute. Main focus was on the demography of respondents, crop production activities and insect pest control. Pretesting of the questionnaire in order to assess the appropriateness of questions and comprehension by both farmers and enumerators was done. Data was analysed using descriptive statistics. Fifty five percent (55 %) of the respondents were females and the highest number of participants was from the Chris Hani District Municipality. Majority of the people were unemployed or pensioners. Most had only attended primary school and the mean age was 59 years. Apart from maize, respondents were cultivating other crops such as cabbage, Swiss chard, potatoes etc. Stalk borers followed by cutworms were the main pests of maize in these areas. Respondents used mainly insecticides, followed by alternative substances, which also included cultural control methods such as planting date manipulation. Few respondents used plants as control agents for insect pests. Some people did not control pests even though they were a problem in their fields. The most used plant was Chenopodium ambrosiodes L, while the most used substance was Madubula (a detergent). The most used insecticide was carbaryl from the carbamite family. Respondents listed different preparation techniques for all the control methods mentioned. These techniques revealed different times of preparation, quantities of ingredients, amounts applied on plants, modes of application and intervals of application. Rural farmers in the study areas used different atypical methods which may play a significant role in pest management today. Some of the products may have a positive influence on agriculture, while some are dangerous to humans and environmental health. Further research which will investigate their potential use in pest control needs to be done.

Abstract:
The olive fly, Bactrocera oleae, is the single most important pest in olive plantations. Currently, control of olive fly relies on the heavy use of chemical pesticides. The sterile insect technique (SIT) is a highly effective, species-specific and environmentally non-polluting method of pest control that involves the mass-release of sterilised insects. SIT is considered a potentially valuable method for the control of olive fly. Previous olive fly SIT attempts failed due to an inability to produce large numbers of flies, low egg production rates and lack of a method to separate the sexes. RIDL (Release of Insects carrying a Dominant Lethal) is a biotechnology-based variant of SIT. This could potentially overcome several problems of classical SIT, including the radiation damage to insects. To develop fly male sterility, we have identified and tested several different germline specific promoters and several potential effector genes. These have been linked to the ‘tet-off’ expression system, which is suppressed by dietary tetracycline, and were initially tested in the Mediterranean fruit fly (Ceratitis capitata) for practicality. In the absence of tetracycline, tTAV binds to its target sequence, tetO, and activates expression of downstream genes. Flies carrying a promoter construct (topi-tTAV or β2-tubulin-tTAV) in medfly were crossed to flies carrying effector constructs (tetO-I-ppoI, tetO-3zincfinger or tetO-ProtamineFokI). A combination of β2-tubulin-tTAV and tetO-ProtamineFokI gave the best male sterility in medfly. A construct containing both elements was designed, and transposon-based germline transformation was used to generate and test ten olive fly strains. Progeny assessment off tetracycline indicates high penetrance of the male-sterile phenotype in all strains, with only 0.0-2.4% viable progeny; this sterile phenotype appears to be completely suppressed by provision of dietary tetracycline.

ENGLISH ABSTRACT: Grapevine leafroll-associated virus-3 is one of ten members of the C/osteroviridae that are known to infect grapevine. Nine of these viruses are associated with grapevine leafroll disease, of which GLRaV-1 and GLRaV-3 are the most important and widespread. Members of the C/osteroviridae are unique amongst the viruses, as it is the only known family whose members encode a heat shock protein 70 kOa homolog (Hsp70h). The Hsp70h is a movement protein (MP) that is required for the active translocation of the virion structure through the plasmodesmata into adjacent cells. Broad-spectrum resistance to unrelated viruses can be obtained by a pathogen-derived resistance (POR) strategy that is based on the expression of a dysfunctional MP in plants. The Hsp70h has two distinct domains. The N-terminal two thirds of the protein is an ATPase domain and shares high homology with the ATPase domains of all Hsp70h proteins from the C/osteroviridae and Hsp70 proteins from the prokaryote and eukaryote kingdoms. Conserved amino acids are found in the ATPase domain and are required for the positioning of the ATP at the catalytic site for ATP hydrolysis. The C-terminal domain is variable and the function of this domain in the Closteroviridae is not known. In prokaryote and eukaryote Hsp70 proteins, the C-terminal domain is required for protein-protein interactions. The American NY-1 isolate of GLRaV-3 has been sequenced and POR strategies have been attempted with the coat protein, divergent coat protein and replicase genes, but not with a dysfunctional form of the hsp70h gene. In this study, double-stranded RNA was isolated from a commercial vineyard with unknown virus status, but with distinct grapevine leafroll symptoms, and from two grapevine sources of known virus status, one with mild and one with severe symptoms. The GLRaV-3 hsp70h gene was amplified by RT-PCR from the dsRNA and the gene sequence was analysed. The hsp70h gene from the three virus sources contained more than 94% nucleotide sequence homology to the NY-1 isolate and the conserved amino acids required for ATPase activity were present. The hsp70h gene isolated from GLRaV-3 from a commercial Stellenbosch vineyard showing clear leafroll symptoms was selected for further work and was subjected to site-directed mutagenesis to engineer four point mutations in the gene. These four mutations resulted in the substitution of Asn for Asp", Gly for Thr1O, Lys for Glu 174 and Asn for Asp 197. The wild type (WT) and mutated (Mut) forms of the hsp 70h genes were cloned into a bacterial expression vector. Expression of both the WT- and Mut-Hsp proteins was achieved, and the protein was expressed in the insoluble inclusion bodies. All attempts to refold and isolate active proteins from the inclusion bodies were unsuccessful. Attempts to increase the concentration of soluble protein within the expressing bacteria were unsuccessful. Due to the lack of active protein, biochemical tests on the ATPase activity of the WT- and Mut-Hsp proteins could not be conducted. The wt- and mut-hsp genes were cloned into a plant expression vector for transformation into tobacco plants. These transformations were successful and gave rise to 22 Km' and 18 Km' plants from the WT- and Mut-Hsp constructs respectively. Two plant lines, M5 and M10, transformed with the mut-hsp transgene construct, appeared to have a high level of resistance to the challenging potato X potexvirus, whereas all the other tested plants were susceptible to the challenging virus. It was thus shown that a dysfunctional form of the GLRaV-3 Hsp70h could provide resistance to an unrelated virus in tobacco.
AFRIKAANSE OPSOMMING: Wingerdrolblaar-geassosieerde virus 3 (GLRaV-3) is een van 10 lede van die Closteroviridae wat wingerd kan infekteer. Nege van die virusse is met wingerdrolblaar geassosieer. Die GLRaV-1 en GLRaV-3 is die belangrikste en mees wyd verspreide lede van die rolblaar-geassosieerde Closteroviridae. Lede van die Closteroviridae is uniek in die opsig dat die virusse vir 'n 70 kDa-homoloë hitteresponsproteïen (Hsp70h) kodeer. Die Hsp70 is 'n bewegingsproteïen (MP) wat belangrik is vir die translokasie van die virus deur die plasmodesmata na die naasliggende sel. Breë-spektrum weerstand teen onverwante virusse kan behaal word deur 'n patogeen-afgeleide weerstandstrategie (POR), wat op die uitdrukking van 'n disfunksionele MP wat in plante uitgedruk word, gebaseer is. Die Hsp70hproteïen het twee gebiede. Die N-terminale gebied is In ATPase-gebied en toon hoë homologie met ander ATPase-gebiede van Hsp70h-proteïene van die Closteroviridae, asook die prokariotiese en eukariotiese koninkryke. Gekonserveerde aminosure wat belangrik is vir die posisionering van ATP in die katalitiese domein vir ATP-hidrolise is in die ATPase-gebied gevind. Die C-terminale gebied is variërend en die funksie van die gebied in die Closteroviridae is onbekend. In prokariotiese en eukariotiese Hsp70h-proteïene is die C-terminale gebied belangrik vir proteïenproteïen interaksies. Die nukleotiedvolgorde van die Amerikaanse NY-1-isolaat van GLRaV-3 is al bepaal en POR-strategieë is ook op die kapsiedproteïen, uiteenlopende kapsiedproteïen en die replikasie-proteïen uitgevoer, maar nog nie op 'n disfunksionele vorm van die Hsp70h-geen nie. In hierdie studie is dubbelstring-RNA (dsRNA) van 'n kommersiële wingerd met onbekende virusstatus wat rolblaarsimptome toon, geïsoleer, asook van twee wingerde met 'n bekende virusstatus, een met ligte en een met strawwe simptome. Die GLRaV-3 hsp70h-geen is met hulp van die polimerasekettingreaksie-metode (PKR) vanaf die dsRNA geamplifiseer en die geen se nukleotiedvolgorde is bepaal. Die hsp 70-gene van drie verskillende wingerde het meer as 94% homologie met die NY-1-isolaat getoon. Die gekonserveerde aminosure wat vir ATPase-aktiwiteit belangrik is, was teenwoordig. Die hsp70h-geen van GLRaV-3, wat uit 'n kommersiële wingerd met duidelike rolblaarsimptome in die Stellenbosch-gebied geïsoleer is, is vir verdere navorsing gekies en dit is aan setel-gerigte mutagenese blootgestelom vier mutasies van die geen te bewerkstellig. Die gevolg van hierdie vier mutasies was die verandering van Asn na Asp", Gly na Thr1o, Lys na Glu174 en Asn na Asp197. Die wilde (WT) en veranderde (Mut) vorms van die hsp-gene is in 'n bakteriese uitdrukkingsvektor gekloneer. Uitdrukking van beide die WT- en die Mut-Hspproteïene is behaal, maar die proteïene was in die onoplosbare fraksie geleë. Pogings om die onoplosbare proteïene te isoleer en in 'n aktiewe oplosbare vorm te verkry, was onsuksesvol. Verdere pogings om die proteïene in die oplosbare fraksie van die bakteriese ekspressiesisteem uit te druk, was ook onsuksesvol. As gevolg van die gebrek aan aktiewe proteïen kon biochemiese toetse nie op die ATPaseaktiwiteit van die WT- en Mut-Hsp proteïne gedoen word nie. Die wt- en mut-hsp-gene is ook in In plantekspressievektor gekloneer vir transformasie in tabakplante. Hierdie transformasies was suksesvol en het aanleiding gegee tot 22 kanamisienbestande (Km') en 18 Km' plante vanaf die WT- en Mut-Hspkonstrukte onderskeidelik. Twee plantlyne, M5 en M10, wat met die mut-hsptransgene getransformeer is, het 'n hoë vlak van weerstand teen die infekterende aartappelvirus X getoon in vergelyking met ander plante wat met die virus geïnfekteer is. Daar is dus bewys gelewer dat 'n disfunksionele vorm van die GLRaV-3 Hsp70h weerstand kan bied teen 'n onverwante virus in tabak.

Abstract:
The common maize weevil, Sitophilus zeamais Motschulsky (Coleoptera: Curculionidae), a well known pest of stored-maize grain in most parts of the world, was identified as one of the major constraints of harvested maize grains in the Eastern Cape Province, South Africa. The use of plants or their products is one of the recent methods being investigated for insect pest control worldwide. Therefore, the main aim of the present study was to identify available plants in the Eastern Cape that could be used to combat the problem of Sitophilus zeamais in stored-maize grains. For the realization of the aims of this research, the following studies were carried out; a preliminary survey was conducted to obtain baseline information on the farmers’ knowledge and experience of indigenous insect pest control methods in the Eastern Cape. Also, studies on the insecticidal potential of the essential oils of some plants were investigated against the maize weevil. The quality parameters of maize grains treated with the essential oils was also studied and, using a rat model, the toxicity of the essential oils was investigated. The outcome from this study revealed that there is awareness amongst the farmers in the Eastern Cape on the use of plants or their products to control insect pests. Unfortunately, such methods are currently being neglected and the knowledge of their application was found to be eroding. Among the various essential oils screened were those from Mentha longifolia L. and Tagetes minuta L. which evoked an appreciable level of contact, fumigant and repellent toxicity on the maize weevil. Further work done to determine the effects of the oils on maize stored over a period of three months revealed that the two oils had no adverse effect on the proximate compositions and some quality parameters of the stored maize. However, the toxicological study conducted on rats showed that the oils at tested concentrations exhibited some level of toxicity. It is, therefore, suggested that the essential oils of M. longifolia and T. minuta should not be used to treat maize grains intended for human consumption.

Os agentes de controle biológico têm recebido grande reconhecimento, e o seu uso tem contribuído como um complemento ou substituição de agroquímicos. No entanto, existem poucos estudos sobre o controle biológico da Pinta Preta dos Citros, causada pelo fungo Guignardia citricarpa, o que impede o seu mercado in natura, além de prejudicar a sua exportação pelo uso intensivo de insumos químicos para controlar este patógeno. Os fungos do gênero Trichoderma são agentes de controle utilizados em todo o mundo contra vários fitopatógenos relevantes. Este estudo visou identificar proteínas extracelulares secretadas por T. atroviride T17 e por T.harzianum T1A, eficazes para o controle de G. citricarpa. Por eletroforese bidimensional (2D) foram obtidos perfis de proteínas secretadas por Trichoderma em meio de glicose (controle) e em meio suplementado com micélio desativado de G. citricarpa. As proteínas foram identificadas por LC-MS/MS mostrando que ambas as espécies secretam proteínas diferentes. Foram identificadas 68 proteínas das 178 diferencialmente expressas por T. harzianum, sendo a maioria relacionada aos mecanismos de biocontrole, mesmo no meio controle. Foi verificado que em contato com o patógeno a expressão de proteínas relacionadas com o metabolismo primário diminui. Por outro lado, Trichoderma atroviride mostrou uma maior expressão de proteínas relacionadas com biocontrole na presença de micélio do patógeno. Nesta espécie identificamos 59 proteínas de 116 diferencialmente expressas, principalmente proteínas relacionadas com a degradação da parede celular: α- manosidase, quitinase, mutanase, glicosidase, endoquitinase e, algumas famílias de glicoside hidrolases. Os resultados indicam que estas espécies apresentam um elevado potencial como agentes de controle de G. citricarpa. Os resultados são pioneiros em detalhar a interação de Trichoderrma com G. citricarpa, por meio da análise do secretoma.

Biological control agents (BCA) have received great recognition, and their use has contributed as a complement or replacement of agrochemicals. However, there are few studies on the biological control of Black spot of citrus, caused by the fungus Guignardia citricarpa, which prevents the market in natura, besides damaging their export by the intensive use of chemical inputs to control this pathogen. Trichoderma fungi are the most applied worldwide BCA against various relevant plant pathogens. The purpose of this study was to identify extracellular proteins secreted by T. atroviride T17 and of T. harzianum T1A, which are effective for the control of G. citricarpa. Bidimensional electrophoresis (2D) allowed obtaining the secreted protein profiles of Trichoderma grown in glucose medium (control) and in medium containing inactivated mycelium of G. citricicarpa. From the 178 differentially expressed proteins by T. harzianum, 68 were identified, most of them related to biocontrol mechanisms, even in the control medium. In the presence of the pathogen, the expression of proteins related to the metabolism decreases. On the other hand, Trichoderma atroviride showed higher expression of proteins related to biocontrol pathogen when grown in the presence of pathogen mycelium, when compared to the control medium. From the 116 differentially expressed proteins, 59 were identified, mainly proteins related to fungi cell wall degradation such as α-mannosidase, chitinase, mutanase, glycosidase, endochitinase and, some families of glycoside hydrolases. The results indicate that these species have a high potential as biocontrol agents of G. citricicarpa. These results are pioneers in to detail the Trichoderrma interaction with G. citricicarpa through the secretome analysis.

Abstract:
The Wheeler Orchid Collection is home to some of the most endangered species of orchids in the world. This fantastic reservoir of endangered species has been enhanced and broadened by its function as a plant rescue station for the U.S. customs service. Unfortunately, this responsibility increases the risk of bringing orchids, which harbor contageous diseases, into the greenhouse where sap transmitted diseases such as the Tobacco Mosaic Virus (TMV), can run rampant. Although manipulation of orchid characteristics is typically done by classical plant breeding techniques, genetic engineering is emerging as a useful technique for the introduction of desirable traits into the orchid genome. Through the use of genetic engineering techniques it may be possible to mitigate the symptoms associated with this destructive virus. Virus resistance may be achieved through the expression of either the sense or antisense viral coat protein gene in orchid tissues if an efficient means of orchid transformation is developed. In this research two transformation protocols were examined for their ability to efficiently transform orchid tissue. The first transformation protocol explored utilized the native ability of Aq bacterium tumefaciens to incorporate DNA into host plants to achieve transformation. The second mechanism explored was particle bombardment transformation.Many strains of A. tumefaciens were employed using direct exposure of Cattleya_ orchid protocorm and callus tissue. Particle bombardment using DNA coated 0.5 um diameter tungsten particles and high pressure helium tank acceleration was employed. The particle bombardment procedure employed the pG35barB plasmid which confers herbicide resistance to the herbicide basta when integrated and expressed in plant tissues.GUS fluorescence assays and PCR analysis indicate that T-DNA is present in orchid tissues, while Southern blot analysis was unable to display that integration had occurred. Particle bombardment yielded herbicide resistant orchid tissues which have yet to be analyzed by Southern blot analysis to confirm integration due to limited tissue quantities.

A study was performed to develop a generic pest monitoring system for sampling the main table grape pests in vineyards in the Hex River Valley, Western Cape Province of South Africa. The presence of phytophagous and predatory mites on cover crop plants was also investigated as this may contribute to biological control of the phytophagous mites in vines. Life table studies for Epichoristodes acerbella (Walker), an important phytosanitary pest, were conducted to determine whether or not this pest was sensitive to high temperatures. Information gained from the latter can also be used for breeding purposes in the possible future development of a sterile insect technique (SIT) programme to control this pest. The sampling system consisted of inspecting 20 plots of five vines per plot per one to two hectares. The top fork of each of the five vines per plot was examined for Planococcus ficus (Signoret) to a distance of within 30 cm of the stem, as well as the distal 15 cm of one cane per vine for the presence of P. ficus and damage caused by Phlyctinus callosus Boh. One bunch per vine was examined for insect damage or presence, and one leaf per vine for the presence of leaf infesting arthropods, such as Tetranychus urticae Koch, P. ficus and Frankliniella occidentalis (Pergande). Corrugated cardboard bands, tied around the stem of one vine per plot, were used to monitor activity of P. callosus. Blue sticky traps, at a density of four to five traps per one to two hectares, were used to monitor activity of F. occidentalis. Pheromone traps, at a density of one trap per one to two hectares, were used to monitor activity of P. ficus, E. acerbella and Helicoverpa armigera (Hübner). All the above-mentioned inspections were done at two-weekly intervals, except traps for E. acerbella and H. armigera, which were inspected weekly. In each of the rows in which the sample plots were situated, one leaf of each of the cover crop plant species was examined for the presence of phytophagous mites and their predators. The abundance and distribution of cover crop plants were determined using a co-ordinate sampling system. Cover crop sampling was done at monthly intervals. The current threshold for P. ficus is 2% stem infestation, which is reached when more than 65 males per pheromone trap are recorded. Counting mealybugs on the sticky pads in the pheromone traps is time consuming. However, the number of grid blocks on the sticky pad with males present can be counted. When P. ficus males are found in 27 blocks on the sticky pad, stem inspections should commence. Due to the spatial association between P. ficus bunch and stem infestation, stem infestation could give an indication of where bunch infestation could be expected. The use of blue sticky traps for predicting halo spot damage, caused by F. occidentalis, is not recommended. The presence of thrips on the vine leaves could not give an indication of where to expect bunch damage, since thrips on the leaves and halo spot damage were not spatially associated. A suitable sampling method for F. occidentalis still needs to be developed. The monitoring system described here can only provide information on the infestation status of the vineyard. For E. acerbella, H. armigera and P. callosus, the traps and cardboard bands could be used to identify vineyards where these pests are present and therefore, where phytosanitary problems may arise. The presence of P. callosus under the bands was spatially associated with P. callosus damage and could be used as an indicator of the latter. The presence of drosophilid flies in the bunches could not be used as an indicator of the presence of E. acerbella in the bunches. If 5% bunch damage is used as an economic threshold for E. acerbella and P. callosus, there will be a good chance of not under spraying if control measures are applied at 1% bunch damage. Epichoristodes acerbella favoured more moderate constant temperatures, with constant temperatures of 28°C and above being unfavourable for development. The economic threshold for Tetranychus urticae Koch is six mites per leaf, or if presence-absence sampling is used, 11 to 29% leaf infestation. Three important predatory mites, that kept T. urticae under control, were found in the Hex River Valley, namely Euseius addoensis (Van der Merwe & Ryke), Neoseiulus californicus (McGregor) and an undescribed phytoseiid in the genus Typhlodromus. Various cover crop plants served as hosts for T. urticae and predatory mites. The presence of these plants created suitable conditions for the survival of these mites and may have influenced their presence on the vine leaves. In the case of phytosanitary pests, both field and pack shed inspections can be used to conclude with a 99% degree of certainty that infestation levels in the pack shed will be 10% or less, since similar results for both methods were obtained. However, more than 20 plots will have to be inspected.
