Abstract:
<p> This research utilized qualitative methods to explore how students and faculty perceive the contextual engagement of the PhD program at three select theological institutions in the Majority World. This research has value, as such engagement is both an explicit and implicit value of doctoral programs located in the Majority World. Over the last decade, the number of PhD programs established to serve the church in Africa, Asia, Latin America, Eastern Europe and the Middle East and the number of students in them has risen rapidly. The research is descriptive in nature and not an attempt to evaluate the level of success or merit of such programs. Rather, it assumed that each program engages its context and sought to identify common themes as well as areas of convergence and divergence as indicated by faculty and student stakeholders. </p><p> The study consisted of thirty-six interviews. Sets of six faulty members and six doctoral students at each of three seminaries located in the Majority World were asked to share from their experiences in the doctoral program. The participating schools were Africa International University in Nairobi, Kenya; South Asian Institute for Advanced Christian Studies in Bangalore, India and Seminario Teol&oacute;gico Centroamericano, Guatemala City, Guatemala. The study asked participants to describe how the PhD program engages the context including classroom teaching and learning tasks, course outputs, including dissertations, and other non-formal aspects of the doctoral experience. </p><p> The findings revealed three primary categories of contextual engagement described by the faculty and students across the three contexts. The participants first described how the intentional design of the programs including explicit and implicit components of the curriculum engage the context. Secondly, students and faculty addressed issues of worldview and cultural values. Contextual insiders provide a depth of understanding not available outside of the context. Intracultural critique ensures contextually engaged aspects are improved through peer interactions. Finally, student and faculty participants found adult learning practices very helpful to overcome conditioned dependency on traditional education models. Participants perceived these practices as particularly helpful for engaging the local context.</p>

<p> The purpose of this study was to contribute to the debate over the value of the Ph.D. versus the Ed.D. in higher education. A mixed methods design was used to explore why some institutions simultaneously offer two tracks to the higher education doctorate (Ph.D. and Ed.D.) and the difference between programs at institutions where only one track to the doctorate was offered. Specifically, I aimed to identify the level of distinction between these two types of doctoral degrees. The differences were examined between the Ph.D. and the Ed.D. in higher education in terms of programs&rsquo; rationale, mission, admission requirements, curricula, and dissertations. The research study relies on the inputs-environment-outcomes model proposed by Astin (1993). In the first phase of this study qualitative data were collected from five doctoral programs in higher education that offered both Ph.D. and Ed.D. tracks to the doctorate in higher education. The results from the qualitative phase were used for the development of a questionnaire and a dissertation analysis form to be used in the second quantitative phase of the study.</p><p> The quantitative phase of the study consisted of a survey and a curriculum analysis of more with more than 2,600 courses from 125 doctoral programs in higher education. Fife&rsquo;s (1991) classification of higher education courses was used to categorize and compare the curriculum of Ph.D. and Ed.D. programs. Based on the results of this study, a profile of Ph.D. and Ed.D. programs in higher education was developed.</p>

Abstract:
U.S. doctoral program completion rates have remained persistently low in the humanities and biomedical sciences despite educators' efforts. A variety of factors, including stress and dissitation advisor-related issues, were associated with high attrition rates and extended time-to-degree for PhD candidates. The purpose of this correlational study was to examine relationships among life stressors, advisor-related factors, and time-to-degree for a convenience sample of 74 online social sciences doctoral degree holders. Holmes and Rahe's work on stress and Tinto's framework for education program attrition provided the framework for the study. Linear regression and Pearson's correlation statistics were used to examine the relationships between Social Readjustment Rating Scale (SRRS) scores, Advisor-Related Factor scores, and time-to-degree after controlling for covariates of age, ethnicity, and gender. Key findings included: a) SRRS significantly (p < .01) predicted time-to-degree after controlling for age, ethnicity, and gender; and b) no significant relationship was found between advisor-related factors. By identifying at-risk students, early intervention could reduce the time need to complete a PhD program and reduce financial and university resources required to finish. Doctoral program administrators could provide closer supervision with PhD candidates and make adjustments based on an accumulation of extraordinary stressors to help PhD candidiates adjust and finish their programs.

Abstract:
Our initial studies reported on the cloning and characterization of FoxP3 (T regulatory cell specific) for the first time in any teleost species along with T-bet and STAT6 in zebrafish, which are specific to Th1 and Th2 cells respectively and looked at their expression in the zebrafish.  Our next studies focused on CD4 expression by T cells and addresses the question whether teleost Th cells are functionally similar to their mammalian counterpart.  A CD4 specific antibody was developed and using molecular techniques have been able to characterise for the first time CD4+ cells within the zebrafish. In addition, to the CD4L gene, three other novel CD4 related genes have also been discovered which need to be investigated, which could mean in fish different T helper cell populations exist expressing CD4 related molecules.  Another FoxP3 gene was discovered, a factor important in controlling the differentiation and function of T regulatory (Treg) cells, and so we shifted our investigations to studying these FoxP3 molecules within the zebrafish model.  These two orthologs of mammalian FoxP3 were only found in zebrafish and it appears that higher vertebrates including modern fish only have one copy of the FoxP3 gene.  Through the development of an antibody we were also able to look at the expression of the zebrafish genes within cells and determine where they were being expressed.  Lastly, having all the molecular tools that were developed during the course of this thesis, allowed us to use <i>in vivo </i>zebrafish models of <i>Mycobacterium marinum </i>and <i>Salmonella typhimurium </i>infection, and attempt to determine the types of adaptive immune responses that were being developed. Understanding the regulatory mechanisms behind the teleost adaptive immune response will help to establish the zebrafish model for vertebrate immune function.

Abstract:
Outdoor education offers a promising method for developing life skills, however this field is undermined by ad hoc theory and limited research. This thesis offers a critical synthesis of theoretical and empirical outdoor education literature, develops instrumentation to measure life effectiveness, and reports on a large, longitudinal study of outdoor education program life effectiveness outcomes. Outdoor education was reviewed as consisting of seven theoretically interactive domains (participant, environment, program, activity, group, instructor, and culture) within a dynamic, experiential milieu. A theoretical systems framework is proposed, drawing on Dewey’s theory of experience, to illustrate how participants’ experiences of outdoor education might be understood as arising within a “complex system”. A critical review of traditional and meta-analytic reviews of empirical outdoor education research indicated small-moderate positive impacts on typically measured outcomes (e.g. self-concept, locus of control, and social skills; ES ~ 0.35). However, this research has been limited by a lack of appropriate dependent measures, low statistical power, over-reliance on inferential statistics, a lack of control and comparison groups, a lack of longitudinal data, and a lack of investigation of independent variables. To address such issues, Study 1 developed new measurement instrumentation and Study 2 conducted a large, longitudinal study. “Life effectiveness” was proposed to refer to generic life skills which facilitate surviving and thriving across a variety of situations. Life effectiveness skills were further proposed to be enhanceable through intervention. Study 1 investigated the psychometrics of the 11-factor, 64-item Life Effectiveness Questionnaire version G (LEQ-G) through congeneric and confirmatory factor analyses (N = 1,164). Three problematic factors and several weaker items were removed, leading to an 8-factor, 24-item model (LEQ-H) with an excellent fit (TLI = .984; N = 1,892). A global second-order model also provided an excellent fit (TLI = .980), with evidence for factorial invariance of the second-order factor across gender and age, and promising evidence for factorial invariance of the first-order model. Further development of the LEQ was recommended to consider construct validity, other life effectiveness factors, and ongoing item development (e.g. to reduce skewness).

<p> The Information Science doctoral program at the University at Albany, State University of New York, faces many of the same challenges found in highly interdisciplinary programs across educational institutions worldwide such as complex curricula development, abundant discipline languages and cultures, and stakeholders clinging to the traditional, single-discipline university system. In 2006, the University at Albany Information Science Ph.D. program faculty redefined the program's structure in hopes of addressing the challenges it was facing. Program structure is a social process shaped by community participation and is influenced by many factors including students, faculty members, and both informal and formal knowledge production. </p><p> Drawing on data collected with both students and faculty present before, during and after the transition to the new program structure, a mixed-method research strategy was performed to examine student retention rates and time to degree, and to explore the experiences of program faculty members' and doctoral students' sense of community and connectedness. Drawing on Wenger's (1998) Community of Practice model and Tinto's model of Institutional Departure, this study occurs in three stages: [1] program and participants' content analysis, [2] surveying of student and faculty members, and [3] select interviews with student and faculty members. </p><p> The data presented here highlights the unique challenges of doctoral interdisciplinary programs and supports the need for collaboration among faculty, and calls for the unquestionable patronage of the institution and the diverse departments involved. Seeing that interdisciplinary programs work across different disciplines, students and faculty alike often find it difficult to assimilate the diverse ways of teaching and methods of research thus calling for unique organizational and pedagogical strategies addressed in the curricula. Successful interdisciplinary programs need faculty who are broad-minded and willing to embrace and learn new methodologies, and respect sometimes conflicting viewpoints. Departments need to develop new models of organizational structure and funding sources to facilitate interdisciplinary research and interdisciplinary community. University leadership needs to move away from rigid hierarchical structures, add more flexibility to allow faculty members to have some movement between disciplinary departments, and needs to provide physical spaces to pull the diverse faculty and student communities together.</p>

Abstract:
The strength of program static analysis techniques lies on its ability to detect faulty behaviors prior to the execution. This ability requires that the analysis process foresees any possible runtime scenario. A task which is even more complex in the case of concurrent programs, because of the number of alternatives introduced by the usual nondeterminism. In this particular case, some of the most common faulty behaviors are those about erroneous usage of resources, presence of deadlocks and data race conflicts. Behavioral type systems for programming languages provide a strong mechanism for reasoning on programs actions at static time. In this thesis we discuss two static analysis techniques based on this approach. The first one, targets the resource usage in an ad-hoc language with full-fledged operations for acquiring and releasing virtual machines. The second one, targets the deadlock analysis of Java programs. In both cases we provide a formal proof of correctness, along with prototype implementations that allow practically to test the feasibility of these solutions. These prototypes have also allowed assessing these techniques against others existing in the literature obtaining very encouraging results.

Abstract:
This mixed-method study explored how students in a doctoral educational leadership cohort at one university used the cohort structure as a learning community or as a method of social support. Survey data were collected from 45 past and present cohort students and qualitative data were collected from three focus groups of 15 participants total. The survey measured four factors: General Cohort Experience, Trust Within the Cohort, Network, and Community of Learners. Quantitatively, one cohort was found to be significantly different from the others in terms of Trust Within the Cohort; and the 60 hour cohorts were found to be more satisfied with the cohort experience than the 48 hour doctoral cohorts at the university studied. The theme of trust and support from and to fellow members both during and after the cohort had dissolved was a strong recurrent theme in this study. Cohort members felt that they developed and strengthened their skills professionally as a result of participating in the cohort; however, they first had to develop a sense of community and trust with their fellow members in order to learn from them.

Abstract:
Hypertension affects 1 billion individuals worldwide and is the major contributing factor to cardiovascular disease. However, the WHO considers current antihypertensive drug therapies inadequate, highlighting a need for a novel approach to hypertension management. Baroreceptors are a promising drug target, and express an unusual glutamate receptor coupled to phospholipase D (PLD-GluR). The PLD-GluR has not been isolated and characterised, which is an important step towards its use as a drug target. A good source of the PLD-GluR is muscle spindle primary afferent nerve terminals, the largest mechanoreceptor in the body. This study thus focuses upon the identification and progress towards isolation of the PLD-GluR from muscle spindle primary afferent nerve terminals. A novel dissection method for high yield extraction of muscle spindles from a high density source, the rat deep masseter muscle, was developed for Western blotting and mass spectrometry screens of all GluRs. Western blots showed spindle homogenate contained a low molecular weight mGluR5 isoform and GluK2. Immunofluorescence showed mGluR5 was expressed on putative nociceptors, not mechanosensory nerve terminals. However, spindle mechanosensory nerve terminals labelled brightly for GluK2, as did baroreceptor nerve terminals. Furthermore, GluK2 appears to be the only GluR subunit on these mechanoreceptors, although mass spectrometry and affinity chromatography could not successfully isolate this receptor. Finally, piezo2 has recently been suggested as the major mechanotransducer protein. However, no evidence was found for piezo2 expression in adult spindle mechanosensory nerve terminals in adult rats or mice. As previous studies have largely focussed on adolescent mice, this could represent a developmental difference. Conversely, a number of candidate mechanosensory proteins, such as TRPs, were identified by a targeted mass spectrometry approach. This provides good candidates for future research. Collectively, this study indicates both spindle and baroreceptor mechanosensory nerve terminals express GluK2, suggesting it is at least a component of the PLD-GluR, and therefore potentially represents a novel drug target for treating hypertension.

Estudos com tumores sólidos mostram que a hipóxia é um importante fator preditor de resposta prognóstica e as vias de hipóxia, estresse oxidativo e imunossupressão podem atuar na progressão tumoral, influenciar a resposta ao tratamento e a resposta imunológica. Uma vez que estas vias podem ter a expressão de seus genes afetada tanto pela exposição à fumaça do cigarro quanto à radioterapia, o presente trabalho buscou investigar a influencia destes fatores de exposição na expressão das proteínas PHD3, HIF-1α, VEGF, RA2A e Foxp3 nos infiltrados intra e peritumorais de câncer colorretal induzido. Para tanto, foram utilizados 53 ratos Wistar, 5 com o intestino saudável como controle negativo (G0) e os 48 ratos restantes foram induzidos à tumorigênese colorretal com 1,2-dimetilhidrazina (DMH) e divididos em 4 grupos, Grupo DMH (G1), Grupo DMH/Radioterapia (G2), Grupo DMH/Fumaça (G3) e Grupo DMH/Fumaça/Radioterapia (G4). A exposição à fumaça do cigarro dos grupos G3 e G4 ocorreu em câmara de inalação e correspondeu a 12 cigarros por dia/grupo durante 20 semanas. Na 21ª semana, os animais do grupo G2 e G4 foram submetidos a três sessões de radioterapia na dose de 700 cGy cada, totalizando 2500 cGy. Na 22ª semana, os animais foram eutanasiados e as lesões fixadas, processadas e coradas com hematoxilina e eosina para diagnóstico. As lesões classificadas como Adenocarcinoma Tubular foram submetidas à Imunohistoquímica para as proteínas PHD3, HIF-1α, VEGF, da via de hipóxia, SOD-1 da via de estresse oxidativo e RA2A e Foxp3 da via de imunossupressão. Todas as amostras tumorais e controles foram analisadas nos infiltrados intra e peritumoral de forma semi-quantitativa e avaliados quanto à exposição à fumaça do cigarro e radioterapia na modulação da expressão destas proteínas. A resposta à radioterapia também foi avaliada pelo índice apoptótico através do anticorpo da caspase-3 clivada nas amostras pertencentes aos grupos G2 e G4. Os resultados mostraram uma relação entre a exposição à fumaça do cigarro e radioterapia com alteração da expressão das proteínas nos infiltrados inflamatórios intra e peritumorais. A expressão das proteínas pode diferir entre os tipos de infiltrado inflamatório, bem como da expressão em células tumorais, mostrando a importância da função diferencial destas células no microambiente tumoral. Além disso, o grupo exposto à fumaça e radioterapia apresentou melhores características histopatológicas em relação à malignidade e melhor resposta terapêutica. Assim concluímos que os ratos expostos à fumaça do cigarro e tratados com radioterapia apresentaram melhores parâmetros histoquímicos dos marcadores de morte celular, inflamação, progressão tumoral, estresse oxidativo e resposta à radioterapia do que os não expostos à fumaça do cigarro. Uma vez que a fumaça é consagrada como indutora de tumor em vários estágios da tumorigênese, e que nossos resultados mostraram características opostas, sugerimos a necessidade de novos estudos que confirmem o real impacto do tabagismo nos processos de tumorigênese, inflamação e na resposta ao tratamento radioterápico.

Abstract:
The study is concerned with improving the spelling performance of below average spellers in an upper primary mainstream classroom setting. The main target group is children who do not qualify for learning support resources yet have difficulty thriving in the literacy area of spelling. It was expected that lessons designed to expand students’ knowledge about words and the English language system would generate a greater student facility with learning spellings. A quasi-experimental methodological approach was taken which also has features of a controlled trials study. Four spelling intervention lesson programs were designed and each has a different pedagogic emphasis: 1) Metacognitive and multisensory learning strategies 2) Teacher-Best-Practice 3) Teacher designed inquiry-based group work 4) Learning with a computer-based Team Learning System. The study design is both practical and pragmatic in that only resources already available to the schools were utilised; lesson formats and teaching practices can be easily replicated; and the spelling programs are compatible with NSW curriculum directives. The intervention lesson program took place for one hour per week over a two-term period and involved four schools situated in the outer areas of Sydney, Australia. Students and teachers in nine upper primary classrooms took part in the study, five of which were intervention classes and four were non-intervention classes. The classroom teachers were an important factor in the program presentation and lessons with an explicit focus on words were conducted from a constructivist perspective. Results indicate that when spelling becomes an instructional focus, substantial progress in performance levels can be made in both intervention and non-intervention classrooms. Discussion of the findings examines how pedagogic factors influenced learning outcomes for all ability groups in the literacy area of spelling. Particular attention is given to the progress in spelling age made by below average performers, as determined on pre-testing, and students with lower than normal cognitive ability. It is considered that the more interactive approach to teaching spelling, plus a high level of congruence with the Quality Teaching model (NSW DET, 2003) elements of Engagement, High-order Thinking, and Substantive Communication, assisted these target groups of students in the intervention classes. The principal conclusion that can be drawn from the findings of the study is that minimal modifications to teaching practices can bring about improvement in students’ spelling performance levels. It illustrates the capacity of children to advance their learning outcomes when lessons are designed and delivered in a manner that encourages engagement and interactive support. Such a conclusion directs the responsibility for students’ learning outcomes to their classroom teachers. The study identifies that there are implications for the providers of in-service and pre-service professional development of teachers situated in upper primary mainstream classrooms, and the pedagogic aspirations of the primary school community.

Abstract:
In spite of the substantial repository of literature about respirators, little is known about the practicalities of their use. The focus of this research was about the practical aspects of using respirators in New South Wales workplaces. Two self-administered postal surveys were used to assess the level of implementation of respirator programs in 1996 and 2001. After five years, six elements improved. The most improvement was seen in the area of documentation including written procedures, keeping training records, recording respirator tasks, and maintenance records. The second survey investigated respirator maintenance and found little automated cleaning. Thorough washing was scarce with more than 50% of organisations relying on moist towelettes. For the third portion of the research methodology 485 used, half facepiece reusable respirators from 36 different sites were examined to determine the most common respirator defects. Maintenance and cleaning procedures were primitive and probably inadequate. Disinfection or sanitisation was common practice indicating concern about infectious diseases. The 2001 survey found that physical inspection of respirators in the workplace usually checked for the common types of defects found in the examination of used respirators. Weight, breathing resistance, heat and tightness were reported as causes of discomfort. The key outcomes from the research were that respirator programs were poorly implemented in a group of organisations that were expected to have more expertise than most and that the most common defects could be corrected by good respirator cleaning programs.

Estudos com tumores sólidos mostram que a hipóxia é um importante fator preditor de resposta prognóstica e as vias de hipóxia, estresse oxidativo e imunossupressão podem atuar na progressão tumoral, influenciar a resposta ao tratamento e a resposta imunológica. Uma vez que estas vias podem ter a expressão de seus genes afetada tanto pela exposição à fumaça do cigarro quanto à radioterapia, o presente trabalho buscou investigar a influencia destes fatores de exposição na expressão das proteínas PHD3, HIF-1&#945;, VEGF, RA2A e Foxp3 nos infiltrados intra e peritumorais de câncer colorretal induzido. Para tanto, foram utilizados 53 ratos Wistar, 5 com o intestino saudável como controle negativo (G0) e os 48 ratos restantes foram induzidos à tumorigênese colorretal com 1,2-dimetilhidrazina (DMH) e divididos em 4 grupos, Grupo DMH (G1), Grupo DMH/Radioterapia (G2), Grupo DMH/Fumaça (G3) e Grupo DMH/Fumaça/Radioterapia (G4). A exposição à fumaça do cigarro dos grupos G3 e G4 ocorreu em câmara de inalação e correspondeu a 12 cigarros por dia/grupo durante 20 semanas. Na 21ª semana, os animais do grupo G2 e G4 foram submetidos a três sessões de radioterapia na dose de 700 cGy cada, totalizando 2500 cGy. Na 22ª semana, os animais foram eutanasiados e as lesões fixadas, processadas e coradas com hematoxilina e eosina para diagnóstico. As lesões classificadas como Adenocarcinoma Tubular foram submetidas à Imunohistoquímica para as proteínas PHD3, HIF-1&#945;, VEGF, da via de hipóxia, SOD-1 da via de estresse oxidativo e RA2A e Foxp3 da via de imunossupressão. Todas as amostras tumorais e controles foram analisadas nos infiltrados intra e peritumoral de forma semi-quantitativa e avaliados quanto à exposição à fumaça do cigarro e radioterapia na modulação da expressão destas proteínas. A resposta à radioterapia também foi avaliada pelo índice apoptótico através do anticorpo da caspase-3 clivada nas amostras pertencentes aos grupos G2 e G4. Os resultados mostraram uma relação entre a exposição à fumaça do cigarro e radioterapia com alteração da expressão das proteínas nos infiltrados inflamatórios intra e peritumorais. A expressão das proteínas pode diferir entre os tipos de infiltrado inflamatório, bem como da expressão em células tumorais, mostrando a importância da função diferencial destas células no microambiente tumoral. Além disso, o grupo exposto à fumaça e radioterapia apresentou melhores características histopatológicas em relação à malignidade e melhor resposta terapêutica. Assim concluímos que os ratos expostos à fumaça do cigarro e tratados com radioterapia apresentaram melhores parâmetros histoquímicos dos marcadores de morte celular, inflamação, progressão tumoral, estresse oxidativo e resposta à radioterapia do que os não expostos à fumaça do cigarro. Uma vez que a fumaça é consagrada como indutora de tumor em vários estágios da tumorigênese, e que nossos resultados mostraram características opostas, sugerimos a necessidade de novos estudos que confirmem o real impacto do tabagismo nos processos de tumorigênese, inflamação e na resposta ao tratamento radioterápico.

This thesis is an analysis of the nature of scientific history in the late Scottish Enlightenment. Using Adam Ferguson's History of the Progress and the Termination of the Roman Republic (1799) as the main analytical framework, the thesis explores two fundamental questions: the consolidation of a scientific epistemology in the field of history and its role in shaping British politics during the Age of Revolutions. Still trying to navigate the new political realities of the Union of 1707, for philosophers of the Scottish Enlightenment such as Adam Ferguson the French Revolution posed a direct challenge to the commercial civilisation epitomised by modern Britain. Aiming to mirror the successful discoveries that empiricist methodologies had prompted in the field of natural philosophy, Ferguson set out to uncover the moral and political laws underpinning society. He turned to the history of the rise and fall of Roman Republic as his source material and elevated it to the status of universal tragedy. Ultimately, what Ferguson wanted was to provide the political class of modern Britain with a scientific-based model for discussion. Moreover, he claimed that Progress and Termination was intended as an impartial narrative for the wider civic community to articulate its national identity through a fact-based public debate and in the face of a new political era for modern Europe. This thesis examines the theory and practice behind this approach and demonstrates that the practice of philosophical history during the late Scottish Enlightenment cannot be understood without the development of a modern scientific epistemology. The thesis is divided into four chapters which focus on context, method, structure, and content of Progress and Termination. The first two chapters analyse the figure of the writer and that of the reader of history. They situate Ferguson's persona and work within the political and cultural contexts of late eighteenth-century Scotland. In doing so, they show how Ferguson's theory of history shaped his views of the civic function of historians and their readers within the modern political nation. Chapter three examines the structure of Progress and Termination through Ferguson's use of three different categories of historical time; it illustrates how Ferguson's use of time informed the political and moral message of his history. Finally, chapter four studies how the narrative of Progress and Termination allowed Ferguson to engage with the convulsive political landscape of post-1789 Europe and why he believed that his methodology could provide Britain with a solution to the challenges of modernity.

The thesis extends the use of systemic functional linguistics (SFL) to describe and analyse the semiotic systems beyond language by providing a detailed and systematic approach to the description of multimodal hypertext systems. The thesis uses a social semiotic approach to the text in order to develop an analytical framework for the description of hypertext through the two dimensions of rank and metafunction. This approach is employed to describe, assess and evaluate the pathways that student user groups construct using hypertext resources during a task-based information search session. The resources realised at the ranks of element, screen and pathway are described across four metafunctions: Representational, Interactive, Compositional and Logical. The data which forms the basis of the thesis was collected from a Year 4-5-6 classroom in a primary school in Sydney, Australia. The substantive contributions of this thesis detail the resources of the hypertext analytical framework.
