Abstract:
AbstractThe field of design research has been expanding into a wide diverse range of multidisciplinary topics. It takes substantial time for young researchers to attain a cumulative overview of state of the art on ever more complex methodologies. Teaching doctoral candidates in summer schools is an approach being taken by the design society to support them attaining an immersed understanding of a chosen research field as well as to help them formulate their own line of research. The aim for a new researcher is to form exchanges and collaborations with other researchers. The 'International Summer School on Product Architecture Design - PAD 2018' was such an effort, where 17 international PhD researchers and three international faculties met for a week and explored research in product architecture through hands-on exercises. We surveyed the researchers for effectiveness of the summer school and found that structure and concept of the summer school was effective for providing a background baseline of state of the art. We found there was a significant but less impact on individual participant´s research. We have yet to understand if the creation of collaborations among participants will occur.

Abstract:
Purpose This study aims to examine how post-PhD researchers construct their identities through significant work experiences as they endeavour to develop their research independence and a distinct scholarly profile. The authors were especially interested in how they made meaning of their important work experiences, the ones that were emotionally salient. Design/methodology/approach Using a narrative approach, the analysis was conducted on a data subset from a large cross-national mixed-methods research project about early-career researchers’ identity development. The sample included 71 post-PhD researchers from the UK who completed an online survey. Ten of whom were also interviewed through a semi-structured protocol. Findings Post-PhD researchers considered work experiences to be significant when those experiences helped them to gauge whether their self-representation as researchers was coherent and a further research career was practicable. The same type of significant event (e.g. publishing in a prestigious journal) could hold different meanings depending on who experienced it. Positive experiences helped to maintain their motivation and made them feel that they were consolidating their identities. Negative experiences tended to challenge their sense of identity and their sense of belonging to academia. Whereas positive feelings towards a significant experience appeared to persist over time, negative feelings seemed to fade or evolve through self-reflection, but ultimately had greater saliency. Originality/value Few previous studies have been conducted on how emotionally powerful work experiences influence post-PhD researchers’ identity development. Besides highlighting how emotions and feelings, often-neglected aspects of identity development, influence the process, this study offers a constructive – and, in some ways, alternative – view of the impact that negative experiences have on their identity development.

Abstract:
The aim of this study was to identify factors contributing to the low participation rates of women in research-based postgraduate study. Although it was acknowledged that there were increases in the participation of women in postgraduate studies, this was primarily at the Graduate Diploma and Graduate Certificate level. As these courses do not provide a direct entry point to Masters and PhD programs, a large potential source of research degree candidates is being lost. In recognition of the value of successful completion of an Honours degree as a foundation for research-based higher degree qualifications, the parameters of this project were narrowed in order to examine the specific factors influencing the enrolment of students in Honours programs.

Abstract:
Novice researchers face challenges in applying grounded theory and choosing between its two historical approaches—Glaserian and Straussian. Although much has been discussed regarding the differences between the Glaserian and Straussian approaches, these differences can confuse early researchers, leading to the flawed use of grounded theory in management and organizational research. Using three case studies (a PhD graduate, a PhD candidate, and a PhD supervisor) in a management and organizational research context, this article illustrates these key differences and provides guidance for researchers in choosing between them. By providing examples and commentary, this article aims to help researchers to choose and apply the most appropriate form of grounded theory within the field of management and organizational research.

Abstract:
OBJECTIVE: This study evaluated the profile and scientific production of researchers in oral pathology who received grants in the area of Dentistry from the Brazilian National Research and Development Council. MATERIAL AND METHOD : The standardized online curriculum vitae (Curriculum Lattes) of 34 researchers in oral pathology who received grants in the years 2008-2010 were analyzed. The variables were: gender, affiliation, time from completion of the PhD program, scientific production, and supervision of undergraduate students and Master's and PhD programs. RESULT: The States of São Paulo (52.94%) and Minas Gerais (26.47%) were responsible for 79.41% of the researchers. Regarding affiliation, three institutions accounted for approximately 44.11% of the researchers: UNICAMP (17.64%); USP (14.70%); and UFMG (11.76%). The researchers published a total of 906 full-text articles, with a median of 26.64 articles per researcher in the triennium and 8.88 articles per year. It was found that of 906 articles published, 366 (40.39%) were published in strata A (qualis-CAPES). Oral pathology researchers supervised 437 scientific initiation and post-graduate students. Of these, 138 (31.57%) were scientific initiation, 169 (38.67%) were Master and 130 (29.74%) were PhD students. CONCLUSION: An important scientific production of oral pathology researchers in the 2008-2010 triennium was observed. By knowing the profile of researchers in oral pathology, more effective strategies for encouraging scientific production and demanding resources to finance research projects can be defined.

Abstract:
Purpose This paper aims to consider the realities and problematics of applying a grounded theory (GT) approach to research, as a novice, within a mixed methods study during post graduate research. Its intention is to provide the novice user with a framework of considerations and greater awareness of the issues that GT can expose during research activity. Design/methodology/approach Using empirical evidence and a comparative approach, the paper compares the efficacy of both the classic Glaserian and Straussian models. It observes the effects of a positivist academic environment upon the choice of approach and its application. This study was specific to design education; however, its reliance upon a social science epistemology results in findings beneficial to research novices across broader disciplines. Findings GT presents the novice researcher with several potential pitfalls. Most problematic were the immutable, positivist institutional requirements, researcher a priori knowledge, the reliance upon literature for the research proposal and structure of the proposal itself. These include suspension of the notion that the purist use of either model can be applied in the current academic environment, the need for a close relationship with the data and toleration of a non-linear process with unexpected results. Originality/value The practicalities of GT research are often reflected upon by the academy, but use by novice researchers is little considered. The findings from this study provide a novel set of guidelines for use by those embarking on GT research and particularly where the requirements of formal education may cause a conflict.

Abstract:
This doctoral dissertation is concerned with an exploratory study on how emerging learning ecologies enabled by Web 2.0 and social web are affecting the self-organized practices and dispositions in the digital settings of individual PhD students. The research endorses a constructivist grounded theory approach, where data collection has been undertaken across three Italian and one UK universities and has included a sequence of online questionnaires, individual interviews and focus groups. The findings being generated provide a repertoire of social media practices for research purposes; a framework conceptualizing the trajectories in the digital, in terms of Space, Time, Socialization, Digital identity, Stance and Tensions; the forms of resilience and the tensions underlying the PhD researchers’ digital engagement. The affordances of PhD e-researchers’ emerging ecologies are therefore understood as multi-dimensional and transitional trajectories intentionally undertaken by the individuals and generating a range of reactions toward the opportunities provided by the open Web.

Nausa, Ricardo. "The language of oral presentations given by PhD researchers in an EAP class : level of performance and disciplinary differences." Thesis, University of Birmingham, 2019. http://etheses.bham.ac.uk//id/eprint/8883/.

Full text
Abstract:
This thesis explores PhD-researcher oral presentations (OPs) in five studies on engagement and clarification strategies in a parallel corpus of 88 OP transcription-essay pairs (n=128228 tokens). Corpus and statistical significance procedures identify features that discriminate among researchers' levels of oral achievement and disciplines: gestural-verbal deixis, audience and impersonal identity projection, code glosses, and transformations of written into oral content. Features analyses include distribution across the levels and disciplines subcorpora, recurrent patterns, discourse functions, and pragmatic appropriacy and grammatical variety. The studies reveal that levels differ in the way that presenters mark stance authorship, anticipate the audience need for help, and vary their strategies grammatically. Disciplinary differences re-present the ways in which disciplines (re)produce knowledge. Hard-fields focus on research methods and outcomes is observed in interaction with images, academic identity projection, and technical terms explanation. Soft-field OPs focus on interpretations is observed in opinions towards existing knowledge and use of folk examples. Language choices also reflect the non-expert character of the audience. This thesis contributes to the study of oral academic genres by demonstrating the importance of multimodal, across modes, non-deficiency analyses; confirming disciplinary differences; and proposing ways of understanding levels of achievement based on pragmatic success rather than grammatical accuracy.

Thesis, Uppsala universitet, Institutionen för informatik och media, 2017. http://urn.kb.se/resolve?urn=urn:nbn:se:uu:diva-332028.

Abstract:
This research aims to understand the different practices and strategies early career researchers and PhD students from the social sciences have in Social Networking Sites (SNSs) for science communication in one particular country: Brazil. Following this purpose, the central research question is which are the motives and rationale of the researchers for using social networking sites for science communication. Two sub-questions arise from this general research question: How do practices and strategies relate to the academic system of this country? And How do the traditional science communication practices translate into the use of Social Networking Sites (SNS)? This research is empirically oriented building up on case studies in Brazil. This study makes use of the adaptation that Van Dijck (2013) made of the Actor-Network Theory (ANT) and the review of affordances of social media platforms (Bucher & Helmond, 2016) to apply it to the study of social media as the theoretical approach. The methodological approach of this research is qualitative, using both interviews and netnography as research methods. The primary motivations for using different Social Networking Sites are all related to connectivity: communication with peers, to the public and research subjects, updating themselves about their research issue, dissemination of research, availability of papers, self-branding and participation in interest groups are the most mentioned. These motivations translate into cross-posting practices and integrated communication strategies -combining online and offline elements- on the different Social Networking Sites. These motivations translate into perceived affordances all related to social affordances, therefore, social capital processes: availability, scalability, visibility and multimediality. The academic system of the country has remained unchanged as it privileges traditional scholarly academic formats; therefore, early career researchers and PhD students from the social sciences only use the different Social Networking Sites (SNS) as a side aid but not as a primary means of communication. Social media is underused as a means of public science communication, even though these platforms offer a lot of advantages for pursuing such issue. Traditional science communication practices translate into the use of Social Networking Sites (SNSs). The most important issue that came out in this report was the fact that social affordances provided by Social Networking (SNSs) are still required to be endorsed by real life meeting to start further collaboration and the fact that English is the preferred language for such issues.

Abstract:
We examine two questions: what factors influence the engagement of researchers in public engagement activities ? what effects, for himself, can a researcher derive from popularization (or public communication of sciences and technologies : PCST)? After a synthetic history brushing different paradigms of the PCST, we investigate 20 studies carried out between 1967 and 2014. These interrogate the researchers. We criticize them one by one and clear some trends. PCST has generally "good press" and the commitment of researchers seems weak but robust. Opinion-type studies tend to allow the diagnosis and recommendations to be declared to the researchers themselves, others use less declarative theoretical models, and divergences appear around the influence of the "gaze of others" especially. The question of effects is not analyzed. To explore our two questions further, we use two approaches. On the one hand adapting a theoretical model, the theory of planned behavior, to the corpus of PhD students of the University of Burgundy; on the other hand, to take advantage of my experience as head of the Experimentarium, an extension program which, for 17 years, has been hiring doctoral students. The survey using the theory of planned behavior determines the main factors of engagement: past behavior, attitude towards extension, the opinion of colleagues. Beyond general trends, multifactorial analyzes make it possible to better identify, on a case by case basis, the factors that influence the commitment of each one. Each doctoral student has its own history. The observation of the Experimentarium supports the influence of attitude, pleasure to popularize, as well as the importance of the consequent socialization of actions of popularisation. These criteria are both factors of commitment and effects for the researcher. ...These reflections invite thinking about popularisation, not as a task or a duty to be fulfilled, but as a situation which - if prepared, observed, adjusted and therefore reflexive - can be a source of emancipation for the public but also for the searcher

Abstract:
Para alcançar o objetivo proposto, realizou-se um inquérito online, e várias entrevistas aos vários atores do processo político de modo a perceber as suas posições em relação à problemática em estudo.<br>Portugal, with regard to human resources (HR) policies in science and technology (S&T), during the 1990s several measures were implemented to increase HR in S&T, through doctoral and postdoctoral fellowship programs. Later, during the 2000s, with the aim of strengthening the working relationships of researchers, policies were implemented to promote scientific employment, such as the Ciência Programme and the FCT Investigator Programme. However, during the duration of these two programmes, the problem of the precariousness and lack of future perspectives of the investigators became more evident, leading to their denouncement on the part of researchers and representative associations. It is the intent of this work, to study and understand the problem of the precariousness of PhD researchers, more precisely, observing the different views and perceptions of those involved in the process, namely the directors of the host institutions, the representatives of political authority, the researchers themselves and the representatives of researcher associations and also seeks to understand the impact of scientific employment policies on the development of the Portuguese Scientific Technological System and the obstacles to the development of research careers in most institutions. To achieve the proposed objective, an online survey was carried out, and several interviews were carried out with the several actors of the political process in order to understand their positions in relation to the problem under study.

Abstract:
Yes<br>Background Pilot studies can play an important role in qualitative studies. Methodological and practical issues can be shaped and refined by undertaking pilots. Personal development and researchers’ competence are enhanced and lessons learned can inform the development and quality of the main study. However, pilot studies are rarely published, despite their potential to improve knowledge and understanding of the research. Aim To present the main lessons learned from undertaking a pilot in a qualitative PhD study. Discussion This paper draws together lessons learned when undertaking a pilot as part of a qualitative research project. Important methodological and practical issues identified during the pilot study are discussed including access, recruitment, data collection and the personal development of the researcher. The resulting changes to the final study are also highlighted. Conclusion Sharing experiences of and lessons learned in a pilot study enhances personal development, improves researchers’ confidence and competence, and contributes to the understanding of research.

This study collects 28 domestic doctoral theses, using Hedges and Olkin (1985) meta-analysis to conduct analysis, the application of Comprehensive Meta-analysis software version 2.0 is for statistical analysis, after conversing into the effect of volume, and it will be merged later, and the total calculation explain the average effect of volume. Based on the findings and analysis the summarized are as follows: 1. A male educators tend to perceive a higher dree about a higher degree about the principals' leadership than the female educators do. And the subjects on the study are the moderator variables. They affect on different gender educators about education leadership. 2. There is no significant difference for perceiving the principals’ leadership between the principals with the different years of experiece. And the subjects on the study, the sample area, and school level are the moderator variables. They affect educators’ perception about education leadership. 3. There is no striking difference for perceiving the principals’ leadership between the principals with the different education background. And the subjects on the study, the sample area are the moderator variables. They affect on different gender educators about education leadership. 4. The teachers that also moonlight as the administrators tend to have higher perception about the principals’ leadership than the teachers whose sole responsibility is to teach. And the subjects on the study, the sample area, and school level are not the moderator variables. They won’t affect educators’ perception about education leadership. 5. The educators in smaller school tent to perceive a higher degree of the principals’ leadership than those who teacher in lager school did. And the subjects on the study, the sample area, and school level are the moderator variables. They affect educator’s perception about education leadership. Based on the above-mentioned conclusion, I suggest few points of view for government authoritites on Educational Administration, Elementary and Secondary school, and ongoing studies for practical reference.

The non-motile and opportunistic pathogenic bacteria are ubiquitous and distributed widely in the nature. Because of the A. baumannii easily mutate to antibiotic resistant strain and the antibiotics were not probably used, more and more pandrug-resistant strains are found and are without suitable medicine to administrate. There are many bacteriophage applied to bacterial infection in the foreign, but very few reports are concerned about phage therapy on A. baumannii. We have isolated several lytic bacteriophages to multidrug resistant A. baumannii and also have the data to characterize the relations between the phages with hosts. In this study, the animal model was used to investigate the therapeutic efficacy of the best lytic efficiency phage ?km18p. First, the great results showed that the phage ?km18p has the well protection on macrophage RAW 264.7 (from mouse) that was infected with A. baumannii. Then the lethal dose (2 ~ 3 × 10^8 CFU) on mice was determined. The phage phi-km18p with different MOI concentrations of 10, 1, 0.1 and the bacterium were simultaneously injected to the individual mice and more than 90% mice were survival, but only 60% survived when phage was administrated 1 hrs later after bacteria challenged. After phage administration 3 hrs later, the whole blood specimens showed that the bacteremia concentration decreased from 2.5 × 10^8 CFU to 10^4 CFU/ml. Detection of the inflammatory cytokine of infected mice after phage treatment, the data showed that the concentration of tumor necrosis factor-alpha (TNF-alpha) decrease from 3000 pg/ml to 250 pg/ml; and also the lnterlukin-6 (IL-6) decrease from 35000 pg/ml to 1000 pg/ml. In this study, some wild type mutated to phage-resistant A. baumannii, by Southern blotting detection revealed that the phage genome did not integrate into the host genome. The phage resistant strains all have the 20 kb size of plasmid, but the genome size of extra plasmid is different from phage genome and plasmid dose not contain any phage genes in it. In vitro experiment, we occasionally found that the phage resistant strains were sensitive to the fetal bovine serum contained the complements. When incubated in the human whole blood, the phage resistant strains were totally removed, but the wild type strains were still survival. From the Western blotting, there were significant different proteins expressed between wilt type and phage resistant strains.

Full text
Abstract:
In her detailed retelling of three iconic movements in India, Professor Emerita Krishna Mallick, PhD, gives hope to grassroots activists working toward environmental justice. Each movement deals with a different crisis and affected population: Chipko, famed for tree-hugging women in the Himalayan forest; Narmada, for villagers displaced by a massive dam; and Navdanya, for hundreds of thousands of farmers whose livelihoods were lost to a compact made by the Indian government and neoliberal purveyors of genetically modified organisms (GMOs). Relentlessly researched, the book presents these movements in a framework that explores Hindu Vedic wisdom, as well as Development Ethics, Global Environment Ethics, Feminist Care Ethics, and the Capability Approach. At a moment when the climate threatens populations who live closest to nature--and depend upon its fodder for heat, its water for life, and its seeds for food--Mallick shows how nonviolent action can give poor people an effective voice.

Abstract:
In the last years, there has been a large increase in high-educated and high-skilled people’s mobility as a consequence of the internationalization and globalization, the weakening of research and university systems of sending countries (the “brain drain” process), the increase in skilled demand and improvements in higher education of host countries (the “brain gain” process). At the micro-level, academic mobility has positive consequences on occupational prospects and careers of researchers, both in the short- and long- run. Nevertheless, numerous research studies have demonstrated the challenges of engaging in international academic mobility for people with caring responsibilities, particularly women. Using Italian data on occupational conditions of PhDs collected in 2018 by Istat and modelling multinomial logistic regression analyses, we intend to verify if female researchers are associated with a lower international mobility irrespective their field of study, and the extent to which gender interacts differently in the various fields of study in affecting the probability of moving abroad after PhD qualification. Also, the distinction between long-term and short-term mobility, which has been mainly neglected in the literature concentrating on longer stays, has taken into account. In this respect, short-term mobility is a potentially high-value investment that may be pursued also by those researchers and scientists who cannot move for longer periods, such as women with caring responsibilities. In the literature, it is acknowledged that an experience abroad during early career may have positive effects on future occupational prospects. With our work, we intend to shed light on potential disparities on moving abroad that may exist among researchers in their early career by gender, and which could contribute to leave behind women in academia.

AbstractThe difficulties in estimating the Worst-Case Execution Time (WCET) of applications make the use of modern computing architectures limited in real-time systems. Critical embedded systems require the tasks of hard real-time applications to meet their deadlines, and formal proofs on the validity of this condition are usually required by certification authorities. In the last decade, researchers proposed the use of probabilistic measurement-based methods to estimate the WCET instead of traditional static methods. In this chapter, we summarize recent theoretical and quantitative results on the use of probabilistic approaches to estimate the WCET presented in the PhD thesis of the author, including possible exploitation scenarios, open challenges, and future directions.

Abstract:
It has been found that most PhD graduates (&amp;gt;85%) do not achieve a long-term academic career and thus there is a growing need to re-imagine PhD education that incentivizes doctoral students to engage with research consumers, not only within their discipline, but also, across other disciplines and sectors to have real social impact for an improved society. The aim of this work is to identify intersectoral/interdisciplinary courses that are considered to broaden student career outside and inside academia. For this purpose, a survey was designed to identify modules which lead to the improvement of students' skills while an analysis of their attributes was also performed. Two target groups have been considered: (a) young researchers and (b) program directors each of which can provide different information regarding the courses of interest. 52 students and 11 directors from 5 European Universities, participated in the study. An absence of such courses in the standard PhD program was observed, while any intersectoral/interdisciplinary activities were conducted outside the PhD program, and organized by collaboration of academia and other organizations. The survey findings reveal the need to restructure the PhD programs.

Following the national and international trends in mid-2020 the Klebelsberg Kuno Library of the University of Szeged has also started to deal with the issue of research data management. After thorough self-training the library staff studied the Hungarian and international best practices of managing research data. We tried to assess the needs of the institutional research data management habits and the opinion of the researchers of SZTE with a comprehensive questionnaire. We compiled a comprehensive questionnaire to assess the needs of our researchers, learn what they’re thinking about RDM and what kind of practices regarding RDM already exist in the research community. By evaluating the questionnaire we have determined the areas in which the library could provide professional assistance where there was a real need among researchers. Keeping in mind the needs of the research community of University of Szeged we have decided to develop the following services: copyright consulting, RDM trainings for PhD students, theoretical and methodological assistance for RDM, write institutional FAIR data management recommendations. The last four services have been successfully implemented. We also wrote a feasibility study to assess the possibilities of developing our own institutional data repository.

Agency for Healthcare Research and Quality (AHRQ), 2020. http://dx.doi.org/10.23970/ahrqepcregistryoutcome.

Full text
Abstract:
Background: Major depressive disorder is a common mental disorder. Many pressing questions regarding depression treatment and outcomes exist, and new, efficient research approaches are necessary to address them. The primary objective of this project is to demonstrate the feasibility and value of capturing the harmonized depression outcome measures in the clinical workflow and submitting these data to different registries. Secondary objectives include demonstrating the feasibility of using these data for patient-centered outcomes research and developing a toolkit to support registries interested in sharing data with external researchers. Methods: The harmonized outcome measures for depression were developed through a multi-stakeholder, consensus-based process supported by AHRQ. For this implementation effort, the PRIME Registry, sponsored by the American Board of Family Medicine, and PsychPRO, sponsored by the American Psychiatric Association, each recruited 10 pilot sites from existing registry sites, added the harmonized measures to the registry platform, and submitted the project for institutional review board review Results: The process of preparing each registry to calculate the harmonized measures produced three major findings. First, some clarifications were necessary to make the harmonized definitions operational. Second, some data necessary for the measures are not routinely captured in structured form (e.g., PHQ-9 item 9, adverse events, suicide ideation and behavior, and mortality data). Finally, capture of the PHQ-9 requires operational and technical modifications. The next phase of this project will focus collection of the baseline and follow-up PHQ-9s, as well as other supporting clinical documentation. In parallel to the data collection process, the project team will examine the feasibility of using natural language processing to extract information on PHQ-9 scores, adverse events, and suicidal behaviors from unstructured data. Conclusion: This pilot project represents the first practical implementation of the harmonized outcome measures for depression. Initial results indicate that it is feasible to calculate the measures within the two patient registries, although some challenges were encountered related to the harmonized definition specifications, the availability of the necessary data, and the clinical workflow for collecting the PHQ-9. The ongoing data collection period, combined with an evaluation of the utility of natural language processing for these measures, will produce more information about the practical challenges, value, and burden of using the harmonized measures in the primary care and mental health setting. These findings will be useful to inform future implementations of the harmonized depression outcome measures.

Full text
Abstract:
Two years into implementation, this policy brief examines how California’s Local Control Funding Formula (LCFF) and its accompanying Local Control Accountability Plan (LCAP) meet the needs of English Learners (ELs). Researchers seek to understand district administrator perspectives on the impact of LCFF for ELs through interviews and focus groups with administrators that represent districts from Northern, Central, and Southern California. Findings reveal that although the LCAP serves as a mechanism to increase personnel and PD efforts to address EL needs, it is still largely viewed as a compliance document that requires alignment with other strategic documents and is sensitive to changes in leadership. The following policy recommendations are made as a result of these findings: 1) re-design the LCAP to support districts in specifying EL learning goals, services, assessments and expected outcomes; 2) differentiate support for district administrators; and 3) invest (long-term) in district-level and site-level professional development with a focus on EL success.

United States Department of Agriculture, 2010. http://dx.doi.org/10.32747/2010.7592122.bard.

Full text
Abstract:
Chickpea is the third most important pulse crop in the world and ranks first in the Middle East; however, it has been subjected to only limited research in modern genomics. In the first period of this project (US-3034-98R) we constructed two large-insert BAC and BIBAC libraries, developed 325 SSR markers and mapped QTLs controlling ascochyta blight resistance (ABR) and days to first flower (DTF). Nevertheless, the utilities of these tools and results in gene discovery and marker-assisted breeding are limited due to the absence of an essential platform. The goals of this period of the project were to use the resources and tools developed in the first period of the project to develop a BAC/BIBAC physical map for chickpea and using it to identify BAC/BIBACcontigs containing agronomic genes of interest, with an emphasis on ABR and DTF, and develop DNA markers suitable for marker-assisted breeding. Toward these goals, we proposed: 1) Fingerprint ~50,000 (10x) BACs from the BAC and BIBAC libraries, assemble the clones into a genome-wide BAC/BIBAC physical map, and integrate the BAC/BIBAC map with the existing chickpea genetic maps (Zhang, USA); 2) fine-map ABR and DTFQTLs and enhance molecular tools for chickpea genetics and breeding (Shahal, Sherman and DaniShtienberg, Israel; Chen and Muehlbauer; USA); and 3) integrate the BAC/BIBAC map with the existing chickpea genetic maps (Sherman, Israel; Zhang and Chen, USA). For these objectives, a total of $460,000 was requested originally, but a total of $300,000 was awarded to the project. We first developed two new BAC and BIBAC libraries, Chickpea-CME and Chickpea- CHV. The chickpea-CMEBAC library contains 22,272 clones, with an average insert size of 130 kb and equivalent to 4.0 fold of the chickpea genome. The chickpea-CHVBIBAC library contains 38,400 clones, with an average insert size of 140 kb and equivalent to 7.5 fold of the chickpea genome. The two new libraries (11.5 x), along with the two BAC (Chickpea-CHI) and BIBAC (Chickpea-CBV) libraries (7.1 x) constructed in the first period of the project, provide libraries essential for chickpea genome physical mapping and many other genomics researches. Using these four libraries we then developed the proposed BAC/BIBAC physical map of chickpea. A total of 67,584 clones were fingerprinted, and 64,211 (~11.6 x) of the fingerprints validated and used in the physical map assembly. The physical map consists of 1,945 BAC/BIBACcontigs, with each containing an average of 39.2 clones and having an average physical length of 559 kb. The contigs collectively span ~1,088 Mb, being 1.49 fold of the 740- Mb chickpea genome. Third, we integrated the physical map with the two existing chickpea genetic maps using a total of 172 (124 + 48) SSR markers. Fourth, we identified tightly linked markers for ABR-QTL1, increased marker density at ABR-QTL2 and studied the genetic basis of resistance to pod abortion, a major problem in the east Mediterranean, caused by heat stress. Finally, we, using the integrated map, isolated the BAC/BIBACcontigs containing or closely linked to QTL4.1, QTL4.2 and QTL8 for ABR and QTL8 for DTF. The integrated BAC/BIBAC map resulted from the project will provide a powerful platform and tools essential for many aspects of advanced genomics and genetics research of this crop and related species. These includes, but are not limited to, targeted development of SNP, InDel and SSR markers, high-resolution mapping of the chickpea genome and its agronomic genes and QTLs, sequencing and decoding of all genes of the genome using the next-generation sequencing technology, and comparative genome analysis of chickpea versus other legumes. The DNA markers and BAC/BIBACcontigs containing or closely linked to ABR and DTF provide essential tools to develop SSR and SNP markers well-suited for marker-assisted breeding of the traits and clone their corresponding genes. The development of the tools and knowledge will thus promote enhanced and substantial genetic improvement of the crop and related legumes.
