Full textThe present work concerns the study of an amorphous hydrated calcium and sodium mixed ortho- and pyrophosphate materials which present an interest for biomaterial application in the case of bone regeneration due to the presence of orthophosphate ions (elementary brick of biological apatite formation) and pyrophosphate ions (hydrolysis phenomenon at low pH or by enzymatic activities in bone tissue). These materials are synthesis using soft chemistry protocol including a solution-gel transition and then to a solid state. The objective of this thesis is to study the effect of different synthesis parameters in order to determine the formation mechanism of the original materials and to describe their structure. In fact, we studied the effect of relative composition of initial solution on phosphate precursor (various ranges of molar ratios of ortho- and pyrophosphates) and the drying temperature effect during the last step of the synthesis protocol on the composition of the composition and the structure of the final material by using several complementary characterizations and at various scales (XRD, SEM, TEM, Raman spectroscopy, GTA/DTA, solid-state NMR and chemical analysis). Firstly, we showed that it is possible to control the composition of the final material using the composition of the initial phosphate precursor solution. The extreme compositions (100% pyro- and 100% ortho-) lead to a more or less crystallized phase such as a-canaphite and nanocrystalline apatite, respectively. On the other hand, all materials synthesised with an intermediate composition of phosphate precursor solution (mixed ortho- and pyro-) are amorphous and their compositions were used to propose a general chemical formula for theses amorphous hydrated calcium and sodium mixed ortho- and pyrophosphate materials. It has been possible to clarify certain hypotheses, such as the inhibitory effect of the pyrophosphate ions on the calcium orthophosphate crystallization (and vice-versa), which control the final structure of the material (amorphous or crystalline) and this control can be extended to a large range of composition. The solid-state NMR and the TEM highlight the colloidal formation of the materials and it can be relevant to compare them with Posner cluster model. In a second part, we showed that the drying temperature is a key parameter to control the hydration state of these materials but we can also use this parameter to understanding the internal hydrolysis reaction of the pyrophosphate ions into orthophosphate ions in the final material.[...]

Full textO principal objectivo deste estudo foi o desenvolvimento de cimentos à base de fosfatos de cálcio dopados com Mg, Sr e Zn, para aplicações clínicas. A síntese dos pós foi obtida através de reacções de precipitação, seguido de tratamento térmico de forma a obter as fases mais apropriadas, a (alpha) e b (beta)-TCP. A caracterização dos pós envolveu a quantificação de fases e o refinamento estrutural de fases através da análise de difracção de raios-X por refinamento de Rietveld, bem como, análise da área superficial por BET, e respectivos tamanhos de partícula. Os cimentos foram preparados através da mistura dos pós com meios líquidos diferentes, usando ácido cítrico como acelerador de presa, e o polietilenoglicol (PEG) e o hidroxil propilmetilcelulose (HMPC) como agentes gelificantes. A formação da brushite foi um dos produtos resultantes obtidos da hidratação dos cimentos. Do ponto de vista de aplicação clínica, os cimentos foram caracterizados em termos de presa, injectabilidade, análise calorimétrica e resistência mecânica. Os presentes resultados demonstraram que a incorporação de iões nos cimentos levou a uma melhoria significativa das propriedades destes quando comparados com TCP puro. Os resultados obtidos demonstraram ainda que o tempo inicial de presa tende a decrescer na presença de modificadores reológicos, uma vez que estes aumentam a viscosidade das pastas, e aumenta com o acréscimo da razão L/P, tendo sido considerada a gama de 0.30-0.34 mL g-1 como aceitável para manusear as pastas. As pastas cimentícias apresentaram uma boa injectabilidade, nomeadamente o seu comportamento após extrusão, com aplicação de uma força máxima de 100 N. Investigou-se ainda, com estes testes, a ausência do efeito de “filter-pressing” e que as pastas foram totalmente expelidas para uma razão L/P de 0.36 mLg-1. Os testes de calorimetria isotérmica demonstraram que as pastas apresentam reacções exotérmicas, referentes à dissolução dos pós de partida e à formação de fases intermédias; e à nucleação e crescimento da brushite. Resistências à compressão dos cimentos estudados, após imersão numa solução de PBS durante 48h situam-se entre 1-30 MPa, valores reportados para o osso trabecular. Testes de citotoxicidade, bioactividade e biocompatibilidade dos cimentos foram obtidos através de testes de culturas celulares, mostrando a nãotoxicidade destes. A biocompatibilidade in vivo e a reabsorção dos cimentos foram avaliadas em estudos histológicos e histomorfométricos de secções descalcificadas obtidas através de ensaios de experimentação animal, usando o porco como modelo. Os resultados mostraram que os cimentos implantados são biocompatíveis e osteocondutivos, sem evidência de reacções infecciosas, e portanto, bons candidatos para aplicação como substitutos ósseos.
The main objective of this study was the development of cements based on calcium phosphates doped with Mg, Sr and Zn, for clinical applications. Powder synthesis was obtained through precipitation reactions, followed by heat treatment in order to obtain appropriate phases, a (alpha) and b (beta)-TCP. The cements were prepared through mixing the powders with different liquids, using citric acid as setting accelerator, and polyethyleneglycol and hydroxyl propylmethylcellulose as gelling agents. Brushite was the end product of the hydration reaction. Injectability and setting behaviour were accessed through rheological measurements, extrusion, calorimetric analysis, Vicat and Gilmore needles. Phase quantification and the structural refinement of powders and cements were determined through X-ray diffraction with Rietveld refinement, as well as, BET specific surface area and particle size analysis. Mechanical strengths of wet hardened cements were evaluated. The results obtained showed that the incorporation of ions into cements led to a significant improvement of their overall properties. Initial setting time increased in the presence of rheological modifiers due to their specific roles at the solid/liquid interface and with increasing L/P ratio. Acceptable workability pastes were obtained for L/P ratios in the range of 0.30-0.34 mL g-1. The cement pastes presented good injectability even under a maximum applied force of 100 N. Filter pressing effects were absent, and all cement pastes could be fully injected for LPR > 0.36 mL g-1. Isothermal calorimetry revealed that hydration reactions produce exothermic effects due to: (i) dissolution of the starting powders and formation of intermediate phases; and (ii) nucleation and growth of brushite crystals. The intensity of the exothermic effects depended on doping element, being stronger in the case of Sr. Wet compressive strength of the cement specimens (after immersion in PBS solution for 48 h) was in the range of values reported for trabecular bone (10-30 MPa). Cell cultures used to evaluate citotoxicity, bioactivity and biocompatibility of cements revealed no toxic effects. The biocompatibility in vivo and cements resorption were evaluated using a pig model through histological and histomorphometric studies of decalcified sections. The results show that the implanted cements are biocompatible and osteoconductive, without foreign body reaction. These properties make them good candidates for applications as bone substitutes.

Full textOur strategy is to associate NMR high resolution results and first-principles calculation in order to better understand the influence of 17O local environment onto the NMR parameters (chemical shift, quadrupolar parameters). The calculations were performed using the GIPAW algorithm, developed by Pickard and Mauri, which gives access to the NMR magnetic shielding for all nuclei of a given structure. This method improved previous approaches based on first-principles calculation by considering the symmetrical translation properties in crystals. The document will include data on crystalline as well as glass systems. Indeed, we report data obtained on cristalline sodium phosphates and from which general trends were deduced between the NMR parameters and the structural/local 17Oenvironments. In a following step, molecular dynamics has been used inorder to generate a number of glass structures that would model our real glass systems. The calculated NMR parameters are discussed and compared to those obtained experimentally. Finally, the distribution of NMR parameters usually observed on high resolution spectra will be interpreted using a new distribution model, which is an extension of the so-called Czjzek model

Full textInorganic phosphate (Pi) acts as a signalling molecule in skeletal cells, affecting cell functions and gene expression. With respect to the presence of elevated extracellular calcium and phosphate levels during bone remodelling or carious processes, we questioned whether calcium may play a role in the differenciation of osteoblasts and odontoblasts. In a fist part, as evidenced by real-time PCR and Western blot, we show mat Pi stimulates the matrix gla protein (MGP) expression in osteoblasts through the ERKl/2-Fra-l pathway. We next show that calcium is required in these effects of Pi and that the formation of calcium phosphate precipitates might also be involved in these events. In a second part, we showed that Pi stimulates the type III sodium dependant phosphate transporters in odontoblasts through the ERK1/2 pathway and that Pi is involved in the apoptosis of these cells. These data highlight the importance of Pi in the differentiation of skeletal cells that are localized in a phosphate-rich environment

Full textIn order to avoid the release of 129I (long-lived intermediate-level waste) in the environment, we describe a novel material incorporating iodate in a calcium phosphate based hydroxyapatite. This material is prepared by two synthetic processes: a wet precipitation route followed by a spark plasma sintering and a cementitious route. A high iodine content (with a maximum incorporation rate of 10 wt.%) is reached for both processes, by incorporation of the iodate in the apatitic structure. A monolith with relative density of 88.6% was obtained after shaping of the precipitated powders by spark plasma sintering. This material reveals satisfactory leaching properties, with an initial leaching rate in pure water at 50 °C of 10-2 g.m-2.j-1, and a residual leaching rate at 50 °C of 10-5 g.m-2.j-1 in underground water of potential geological repositories. All in all, this material is a potential candidate for the conditioning of radioactive iodine

Full textThis report concerns the study of the synthesis and characterization of calcium phosphates biomaterials. In the first part, the Ca3 (PO4)2 -Mg3 (PO4)2 system is described. We have shown that the Ca²+/Mg²+ substitution in the β-tricalcium orthophosphate lattice occured according to successive occupation of the Ca(5) then Ca(4) sites of the structure up to the limit of solubility of 14 %. Above this concentration appeared a phase, isostructural with stanfieldite, which corresponded to the chemical formula Ca8Mg9 (Ca, Mg) (PO4)12. The second part concerns the description of a continuous precipitation method which allowed to prepare well crystallized apatites. The atomic Ca/P ratio of these apatites was found to vary from that of pure hydroxylapatite (1,67) to that of pure tricalcium phosphate (1,50). In the third part, we present a study of plasma sprayed coatings of hydroxylapatite , tricalcium phosphate and tricalcium phosphate containing 8 % of Mg²+ ions, onto a titanium alloy substrate. We have shown that the chemical composition of these calcium phosphates might change during spraying. Finally, we report a study of high temperature reactions that occured between titanium oxyde and β-tricalcium phosphate

In general, the micro hardness and diametral tensile strength tests revealed that Mg2+ ions in large amounts (7.5 % mole) had negative effects on the mechanical properties of the samples, while substitution of the F- ions had a positive effect on their mechanical properties.

Full textL'os és el teixit més trasplantats desprès de la sang. L'organització Mundial de la Salut ha posat de relleu l'increment de les malalties musculoesquelètiques, les quals esdevindran la quarta causa mundial de discapacitat en el següents anys. Malgrat la capacitat natural de l'ós per autoregenerar-se, els defectes ossis de grans dimensions necessiten l'ajuda de materials per restaurar-se completament. Actualment, l'ús d'empelts naturals és l'alternativa més emprada clínicament. Tot i això, els autoempelts comporten certes limitacions que requereixen ser adreçades per tal de fer front a les elevades demandes d'una població mundial amb un grau d'envelliment creixent. Els empelts basats en fosfats càlcics (CaPs) són coneguts des de la dècada del 1970 i són uns excel·lents candidats per a la regeneració òssia donada la seva composició, similar a la fase mineral de l'os, que consisteix en aproximadament un 70% d'hidroxiapatita (HA). Els CaPs, en particular l'HA, posseeixen unes propietats intrínseques excepcionals com ara la biocompatibilitat, bioactivitat o la capacitat de suportar el creixement de nou os. Malgrat la seva semblança amb l'os, l'HA és massa estable químicament, i un cop implantada es degrada molt lentament. L'empelt ossi sintètic idealment s'hauria d'integrar en el cicle de remodelació òssia, el que requereix d'un balanç entre la seva reabsorció i la progressiva substitució per os nou. Aquesta capacitat es pot modular mitjançant propietats inherents del material o per mitjà de la combinació de substrats amb molècules capaces d'orquestrar adequadament les respostes de les diverses cèl·lules implicades en la restauració o regeneració òssia. La present tesis cerca explorar, per una banda, la possibilitat de modular les propietats físico-químiques dels CaPs per tal de millorar la seva degradació, així com investigar el potencial de funcionalitzar els CaPs amb heparina, amb la finalitat de millorar les interaccions biològiques a les diferents etapes de la restauració òssia, tant durant la primera etapa inflamatòria, com durant la resorció i el creixement d'os nou. Concretament, la primera part de la present tesi explora la manera en què la modificació de propietats com la porositat, la superfície específica, la microestructura o la composició dels CaPs pot ser emprada per regular la degradació d'aquests materials en una solució acídica similar a l'emprada pels osteoclasts durant la resorció òssia. La interrelació de totes aquestes propietats emmascara de vegades la importància relativa de les propietats texturals, molt lligades a les composicionals, dificultant la predicció dels nivells de degradació dels materials depenent de cada propietat. Per tal de mimetitzar encara més la composició de l'os, s'estudiarà també diferents estratègies per incorporar ions carbonat en l'estructura cristal·lina de l'HA. El segon bloc de la tesi explora la funcionalització dels CaPs amb heparina, un tipus de glicosaminoglicà altament sulfonat present en la matriu extracel·lular de l'os. L'afinitat de l'heparina per captar factors de creixement fan d'aquesta molècula un candidat excel·lent per capturar factors de creixement endògens capaços de regular la resposta cel·lular. Així doncs, partint de les propietats anti-inflamatòries de l'heparina, i de la seva implicació en els processos d'osteogènesi i osteoclastogènesi s'ha formulat la hipòtesi de que aquesta biomolècula podria contribuir a millorar les prestacions dels empelts ossis sintètics. Amb aquest objectiu, s'ha posat a punt un procés d'heparinització de CaPs i s'ha avaluat el seu efecte sobre la resposta de cèl·lules humanes del sistema immune, cèl·lules osteogèniques i osteoclàstiques, per tal d'escatir les possibles sinèrgies de tots dos sistemes, l'immune i l'ossi en la regeneració.

Full textWe aimed at investigating the mechanisms of bacterial formation of Ca and Fe-phosphates and determining traces left by bacterial phosphogenesis in the geological record. Tools were developed allowing a submicrometer-scale characterization of Ça-phosphate (bio)minerals using STXM at the C K-, Ca L2,3- and P L2,3-edges. Experiments of Ca-phosphate precipitation by engineered E. Coli overexpressing the alkaline phosphatase PHO A showed that the amount of this enzyme produced by the cells is a major parameter controlling biomineralization. The mineralogy of the phase formed during these experiments, a Ca-deficient hydroxyapatite, was characterized by SEM, TEM, IR spectroscopy, STXM and NMR. The texture) patterns of the minerais formed in association with bacteria were described, and the chemical evolution of the solution during the course of mineralization was modeled. The modern formation of Fe-phosphates in the watercolumn of the meromictic Lake Pavin was shown to be linked with microbial activities. The oxidation of Fe2+ by Fe-oxidizing bacteria in the presence of high dissolved inorganic phosphate (Pi) concentrations may explain the formation of Fe(II)-Fe(III)-phosphates under the redoxcline. Polyphosphate-accumulating bacteria may participate as well in this process of phosphogenesis. The characterization at the nanometer-scale of microfossils present in a recent phosphorite frorr the Peruvian sheif and a Paleocene («60 Ma) phosphatic coprolite from Morocco shows textural and chemical traces of past calcified bacteria in these rocks, based on comparisons with bacterial fossilization experiments

Full textThe aim of this work was the synthesis and the study of the pharmacological properties of adenophostin mimes Ins(1,4,5)P3 analogues. The biological function and targets of Ins(1,4,5)P3 and adenophostin as well as their structure/activity relationships are reported. The therapeutic potential linked to the inositol phosphates cycle is mentioned. The use of a myo-inositol orthoester, inverting the inositol chair conformation, permitted the activation of the position 2 and the introduction of the lateral chains. This work describes an innovative synthetic pathway of adenophostin mimes Ins(1,4,5)P3 analogues possessing a purinylated butylic arm. The pharmacological properties of our analogues are matching those of the Ins(1,4,5)P3. This is more noticeable if we consider the fact that our compounds are racemic. These results support those of previous studies indicating that the position 2 tolerates some substitution, as indicated by the commonly admitted pharmacophore model
