Full textThe plastid terminal oxidase PTOX is encoded by higher plants, algae and some cyanobacteria. PTOX is a plastid-localized plastoquinol (PQH2) oxygen oxidoreductase. PTOX was shown to be implicated in plant carotenoid biosynthesis, photosynthetic electron transport and chlororespiration and may act as a safety valve protecting plants against photo-oxidative stress. PTOX protein levels increase during abiotic stress indicating a function in stress acclimation. But overexpression of PTOX in Arabidopsis did not attenuate the severity of photoinhibition or, when overexpressed in tobacco, even increased the production of reactive oxygen species (ROS) and exacerbated photoinhibition.Biochemical analysis of recombinant purified PTOX (PTOX from rice fused to the maltose-binding protein) showed that the enzyme exists mainly as a tetramer, which dissociated to a certain extent during electrophoresis, mainly into a dimeric form. The PTOX activity was 320 electrons s−1 PTOX−1. It was also shown that PTOX generates ROS in a side reaction in a substrate (decylPQH2) and pH-dependent manner when liposomes were used: at the basic stromal pH of photosynthetically active chloroplasts, PTOX was antioxidant at low decylPQH2 gaining prooxidant properties with increasing quinol concentrations. It is concluded that PTOX can act as a safety valve when the steady state [PQH2] is low while a certain amount of ROS is formed at high light intensities.It was shown by chlorophyll a fluorescence that recombinant purified PTOX is active when added to photosystem II (PSII)-enriched membrane fragments. PTOX attached tightly to the PSII-enriched membrane fragments. The amount of PTOX attaching to the membrane depended on pH and salts: an alkaline pH and monovalent compared to divalent cations increased PTOX attachment.PTOX activity in planta and its effect on photosynthetic electron transport were investigated using Arabidopsis expressing bacterial phytoene desaturase and tobacco expressing PTOX1 from Chlamydomonas. Arabidopsis expressing bacterial phytoene desaturase (CRTI lines) showed a higher PTOX content and increased PTOX related ROS generation. Furthermore, cyclic electron flow was suppressed in these lines. This implicates that PTOX competes efficiently with cyclic electron flow for PQH2 in the CRTI-expressing lines and that it plays a crucial role in the control of the reduction state of the plastoquinone pool. Using tobacco expressing PTOX1 from Chlamydomonas, it was shown that PTOX competes efficiently with photosynthetic electron flow, but gets inactive when the stromal pH is neutral. Based on the in vitro and in vivo results, a model is proposed, where the association of PTOX to the membrane is controlled by the stromal pH: When the stromal pH is neutral, PTOX exists as a soluble form and is enzymatically inactive avoiding the interference of PTOX with linear electron flow. When the stromal pH is alkaline and the photosynthetic electron chain is highly reduced under stress conditions as high light, PTOX binds to the membrane, gets enzymatically active and can serve as safety valve

Full textOxygen deficiency stress in plant cells leads through inhibition of mitochondrial respiration to an accumulation of NADH and a decrease in ATP content. Rice plants can survive oxygen deficiency better than wheat or potato plants. The aim of this PhD-work was to examine, which biochemical processes are responsible for plant tolerance against low oxygen stress. The studies were focused on the analysis of ethanolic fermentation, photosynthesis and the function of pyrophosphate (PPi) as an alternative energy source. By using ethanolic fermentation, plants can regenerate NAD and maintain ATP formation during glycolysis. Tolerant rice plants produced much higher amounts of ethanol during anoxia in darkness compared to sensitive wheat plants. The high fermentation rate mainly occurred in the shoots as a result of high activities of fermentative enzymes as well as high availability of carbohydrates. Nevertheless, rice plants could not survive more than 24 h of anoxia in the dark because of carbohydrate depletion. Illumination during anoxia extended survival of plants. Ethanolic fermentation rate was reduced during light exposure of plants, indicating that other energy-producing processes can compensate. However, it could be shown that the complete photosynthesis was slowed down during oxygen deficiency due to CO2 deficiency. It is likely that cyclic electron transport could at least partially contribute to ATP production during these conditions. In the past, it was speculated that PPi could replace ATP for phosphorylating processes during low oxygen stress. With transgenic potato plants expressing E. coli pyrophosphatase and therefore containing less PPi it was demonstrated that PPi is a significant alternative energy donor for sucrose cleavage during hypoxia. However, in transgenic potato plants with a reduction of synthesis and activity of PPi-dependent phosphofructokinase it could not be demonstrated that these plants suffer more from oxygen deficiency than the wildtype.

Full textDiese Arbeit beinhaltet quantenchemische und molekularmechanische Modelle zum Isotopeneffekt des Enzyms RuBisCO, der spektralen Charakterisierung des Blaulicht-Rezeptors BLUF und dem Lichtsammelkomplex II (LHCII). Es wurden vor allem die Einflüsse der Umgebung auf die entsprechenden Systeme untersucht. Für RuBisCO wurde gefunden, dass der Isotopeneffekt nur marginal von der Umgebung abhängt. Im Falle der BLUF Domäne wurde eine Aminosäure charakterisiert (Ser41), die bis dato experimentell noch nicht beschrieben war. Es wurde festgestellt, dass Ser41 hochmobil ist und einen systematischen Einfluss auf die spektrale Verschiebung des BLUF Chromophors (Flavin) hat. Schließlich wurde bei LHCII festgestellt, dass kleine Veränderungen in der Geometrie eines Chlorophyll b/Violaxanthin Chromophorenpaares bereits massive Einflüsse auf den Mechanismus des Lichtsammelprozesses haben können. Insbesondere hier zeigt sich, wie kritisch die genaue Beschreibung der Umgebung ist. Zusammenfassend wurde beobachtet, dass sich die Umgebung in oft unerwarteter Weise auf die molekularen Eigenschaften auswirken kann und es daher nicht möglich zu sein scheint, die entsprechenden Effekte vorher abzuschätzen.

Full textAfin de déterminer le rôle de PTOX, nous avons généré des plantes de tabac surexprimant cette protéine. Des mesures de fluorescence de la chlorophylle nous ont permis de démontrer l'implication de PTOX dans l'oxydation non photochimique du pool de PQ. Nous avons conclu que le complexe Ndh et PTOX, tous deux localisés dans les thylacoïdes lamellaires, constituent avec le pool de plastoquinones les éléments d'une chaîne de transfert d'électrons de type chlororespiratoire. Nous avons proposé un mécanisme par lequel l'activité du transfert cyclique des électrons autour du PS I serait régulée par l'activité de la chlororespiration, à travers un contrôle fin du niveau rédox de certains transporteurs de la chaîne de transfert d'électrons.

Full textPrimary plastids derive from a cyanobacterium that entered into an endosymbioticrelationship with a eukaryotic host. This event gave rise to the supergroup Archaeplastida whichcomprises Viridiplantae (green algae and land plants), Rhodophyta (red algae) and Glaucophyta. Afterprimary endosymbiosis, red and green algae spread the ability to photosynthesize to other eukaryoticlineages via secondary endosymbioses. Although considerable progress has been made in theunderstanding of the evolution of photosynthetic eukaryotes, important questions remained debatedsuch as the present-day closest cyanobacterial lineage to primary plastids as well as the number andidentity of partners in secondary endosymbioses.The main objectives of my PhD were to study the origin and evolution of plastid-bearing eukaryotesusing phylogenetic and phylogenomic approaches to shed some light on how primary and secondaryendosymbioses occurred. In this work, I show that primary plastids evolved from a close relative ofGloeomargarita lithophora, a recently sequenced early-branching cyanobacterium that has been onlydetected in terrestrial environments. This result provide interesting hints on the ecological setting whereprimary endosymbiosis likely took place. Regarding the evolution of eukaryotic lineages with secondaryplastids, I show that the nuclear genomes of chlorarachniophytes and euglenids, two photosyntheticlineages with green alga-derived plastids, encode for a large number of genes acquired by transfersfrom red algae. Finally, I highlight that SELMA, the translocation machinery putatively used to importproteins across the second outermost membrane of secondary red plastids with four membranes, has asurprisingly complex history with strong evolutionary implications: cryptophytes have recruited a set ofSELMA components different from those present in haptophytes, stramenopiles and alveolates.In conclusion, during my PhD I identified for the first time the closest living cyanobacterium to primaryplastids and provided new insights on the complex evolution that have undergone secondary plastid-bearing eukaryotes
