The synthesis of isoflavonoid phytoalexins in legumes is relatively well understood, but far less is known about how these phytotoxic compounds are metabolised by the plant when no longer required. In this project medicarpin, the major isoflavonoid phytoalexin in alfalfa, was prepared in radiolabelled form and fed to cell cultures and seedlings of alfalfa. The metabolism of the radioactive phytoalexin was then studied by characterising the radiolabelled metabolites formed. Uptake of radiolabelled phytoalexin by cells was faster in elicitor-treated cultures than in untreated cultures. However,

3-Hydroxy-3-methylglutaryl CoA reductase (HMGR, EC 1.1.1.34) catalyzes the reaction from hydroxymethylglutaryl CoA to mevalonate in the isoprenoid pathway. In solanaceous plants, one class of endproducts of the pathway are sesquiterpenoid phytoalexins, antibiotic compounds produced by plants in response to pathogens. We are interested in the role of the defense-inducible isoforms of HMGR in phytoalexin production and disease resistance. Botrytis cinerea in contact with mature grape berries encounters an environment particularly rich in polyphenols and PR proteins, where the stilbenic phytoalexin trans-resveratrol may accumulate. To mimic conditions similar to those found in grape berries, B. cinerea was grown in vitro with grape PR proteins and polyphenols extracted from mature grapes and with trans-resveratrol. The pathogenicity of two isolates of the fungus <i>Verticillium albo-atrum</i> obtained either from lucerne (V1) or tomato(V2) towards four cultivars of lucerne (<i>Medicago sativa</i>.L), namely Maris Kabul, Europe, Vela, and Euver was investigated. The results confirmed that only the V1 isolate is capable of causing wilt disease in these four cultivars.
