Full textIn order to elucidate what are the phenotypic characteristics controlled by Cx43 in glioblastoma cells, we inhibited its expression, by using the shRNA strategy, in human glioblastoma U251 cells. This approach confirmed that Cx43 is involved in various processes associated to glioma development. Indeed, if its sbsence favors cell proliferation under some conditions (growth in soft agar), it increase angiogenesis while decreasing the adhesion to extracellular matrix proteins, the invasion capacity in Boyden chamber and ex vivo tissue invasion. Moreover, since the invasive capacities of glioblastoma cells are responsible for the tumor recurrence, we were interested to see how Cx43 regulates such capacities. First, a proteomic analysis by mass spectrometry of the secretome of rat glioma cells permitted to consider that such a capacity which is induced by the expression of Cx43 is at least partly mediated by a different pool of secreted proteins [citokines (MCP-1, TGF-β binding protein 1, galectin-1), proteases (MMP3, cathepsins B and L1) and extracellular matrix compounds (fibronectin, SPARC, collagen- α-1) are notably oversecreted by rat C6 glioma cells expressing Cx43]. Second, our study demonstrates that lipid rafts are involved in the invasion process controlled by Cx43. Indeed, the chemically-induced disorganization of these membrane microdomains leads to a drastic decrease of the invasive capacities of U251 cells associated with the inhibition of communication capacities between themselves (homocellular communication) or between them and the astrocytes in primary cultures (heterocellular communication). In conclusion, our results suggest that Cx43 play a complex and even contradictory role in the control of the phenotype of glioblastoma cells. Indeed, while Cx43 inhibits the proliferation of U251 cells, it favors their invasive capacities in association with the presence of lipid rafts

Full textThis thesis deals with the characteristics of the theatre of the world metaphor, as they appear in several contemporary french-speaking plays. Seven plays have been selected as images of the world, since they summon various emblematic elements of life on earth: humans, animals and flora, and emphasize their interaction in a universal surroundings. Firstly, the principal academic contributions concerning the historical notion of theatre of the world are gathered and commented. The main part of the research consists of the demonstration of the metaphor's contemporary extensions, through the study of the corpus. The analysis of each play is preceded by a presentation of its author's writing motivations and motives. Then, the play itself is examined carefully with the help of a reading grid which focuses successively on the dramatic categories : characters, space and time, actions, stage directions. Thus, the thesis's body is built as an exploration of seven variations of the theatre of the world, beginning with the most abstract one and leading to the most organic one. A synthesis ends this journey, pointing out that the plays match three composition patterns, which induces many combinations of the dramatic categories. This classification unveils the ambition at the origin of each writing project. In conclusion, it appears that the theatre of the world, generally considered as an old topos which refers to the theatrality of human life, is a framework strong enough to stand up to the poetics of comtemporary drama, and vast enough to house the most singular dramaturgic inventions

Full textNous avons analysé les articles de Pierre-Joseph-Olivier Chauveau publiés dans le Courrier des États-Unis de New York (1828-1938) pendant la décennie 1840. Le journal rejoignait de nombreuses communautés francophones dispersées dans les Amériques. Chauveau est le seul Canadien français à y collaborer au milieu du XIXe siècle et se révèle un correspondant étranger particulièrement prolifique. Afin de comprendre les poétiques d’écriture du correspondant canadien, nous avons comparé ses articles avec ceux des autres correspondants, principalement parisiens. Nous avons observé que les correspondances, autant celles de Chauveau que celles de ses collègues d’outre-mer, répondent aux mêmes logiques d’écriture que les autres genres journalistiques de l’époque, en particulier la chronique; aussi conservent-elles des traces d’un fort héritage épistolaire. De surcroît, pour pallier le manque de balises dans la manière d’écrire le journal, les correspondants convoquent des formes « canoniques et livresques » : l’influence de la matrice littéraire est tangible. L’étude du contenu des correspondances de Chauveau révèle qu’il utilise sa tribune dans l’espoir de désenclaver sa nation afin de l’arrimer au reste des Français d’Amérique. Pour y parvenir, il exalte la nostalgie pour la Nouvelle-France en misant sur l’identité de « Français d’autrefois » du Canadien, critique de manière récurrente l’opposant commun, l’Angleterre, et fait ressortir les multiples impacts du pouvoir de la religion catholique, rappelant ainsi la France prérévolutionnaire.

The newspaper was produced in New York and reached many French-speaking communities scattered throughout the Americas. We discovered that his articles are a means to convince his readers that French Canadians should be perceived as a legitimate part of French America. In order to do so, Chauveau exacerbated the nostalgy for New France. He also tried to prove that French Canadians are actually pre-revolutionary French citizens, namely by highlighting the extent of the Catholic Church’s powers. Moreover, by constantly criticizing a common opponent, England, Chauveau aspired to strengthen his readers’ sympathy for his nation

Création: A.M.I.S. : La première partie concerne la création d’une web série pour adolescents, A.M.I.S., qui relate un drame survenu durant une soirée dans un cimetière. Sous les effets de l’alcool, de la drogue et la colère, Michaël bat Amanda violemment en plus de poignarder Jade et Éloi, venus l’aider. Samuel, fils d’un avocat, comprend que la présence d’alcool et de drogue pourrait leur nuire et tente de convaincre ses amis de se liguer contre Michaël pour qu’il soit le seul reconnu coupable. Au poste de police, les adolescents créent un scénario différent des événements sans se douter qu’ils avaient été observés par Elsa, une fille rejetée par le groupe. La mort d’Éloi rend le groupe encore plus uni pour punir Michaël et déjouer les plans d’Elsa. Réflexion critique: La violence dans les séries télévisées pour adolescents. La seconde partie traite de la violence mise en scène dans les séries télévisées dans le but de comprendre son fonctionnement et les raisons qui motivent les scénaristes de l’utiliser dans certaines scènes. D’abord, une définition sommaire met en lumière les notions «d’intention» et de «motifs» pour saisir les nuances du phénomène de la violence. Ensuite, nous appuyant sur les théories de Dominique Sipière, nous tentons de saisir le fonctionnement du personnage du «méchant.» En appliquant ces théories aux notions de violence, il est possible de constater un parallèle qui justifie l’utilisation de la violence dans certaines scènes. Pour finir, un aperçu de certaines thématiques présentes dans les séries montre un univers plus sombre et plus complexe.

This study focuses on three understudied aspects of this major modern figure drawing from neglected primary sources in the French National Archives, the Mediathèque of the Paris Conservatory, the Sacher Stiftung, and private collections, notably 1º Messiaen’s eductation at the Paris Conservatory during the 1920s and the relationship of his music to the institutions pedagogical traditions, 2º the development of Messiaen’s technique of borrowing, transforming and juxtaposing pre-existing music in his compositions during the 1930s and 1940s, and finally, 3º Messiaen’s experimentation in the domains of microtonal music and musique concrète.
