Abstract:
Exome sequencing of two sisters with congenital cataracts, short stature, and white matter changes identified compound heterozygous variants in the PISD gene, encoding the phosphatidylserine decarboxylase enzyme that converts phosphatidylserine to phosphatidylethanolamine (PE) in the inner mitochondrial membrane (IMM). Decreased conversion of phosphatidylserine to PE in patient fibroblasts is consistent with impaired phosphatidylserine decarboxylase (PISD) enzyme activity. Meanwhile, as evidence for mitochondrial dysfunction, patient fibroblasts exhibited more fragmented mitochondrial networks

AbstractAs in radiation-induced cancer, post-irradiation sudden deafness (PISD) is defined as sudden onset deafness in patients post-irradiation. Fifteen nasopharyngeal carcinoma (NPC) patients with PISD were enrolled in this study. The mean interval from the completion of irradiation to the occurrence of PISD was 12 years. Seven had total deafness, seven had profound hearing loss, and one had moderate hearing loss. Nine out of 15 (60 per cent) patients experienced hearing recovery within three months after treatment. In conclusion, PISD is a late complication in NPC patients post-irradiation.

Mitochondrial transcription factor A (TFAM) is required for mitochondrial DNA replication and transcription, which are essential for mitochondrial biogenesis. Previous studies reported that depleting mitochondrial functions by genetic deletion of TFAM impaired autophagic activities. However, the underlying mechanisms remain largely unknown.
