Vasoactive intestinal polypeptide was shown to be present in the chicken anterior pituitary gland using high performance liquid chromatography (HPLC), radioimmunoassay (RIA) and immunocytochemistry. The VIP antibodies used for both RIA and immunocytochemistry showed no cross reaction with any known VIP-like peptides, including pituitary adenylate cyclase activating polypeptide (PACAP). The mouse AtT-20/D16-16 anterior pituitary tumour cell line was employed as a model system for the study of the mechanisms mediating calcium ion-stimulated adrenocorticotropin (ACTH) secretion. The present study indicates that calcium ion-stimulated ACTH secretion from AtT-20 cells is mediated by a GTP-binding protein which is present in a variety of other cell types and has been dubbed Ge (for reviews see Gomperts, 1990; Lindau & Gomperts, 1991). This thesis describes molecular cloning and characterisation studies of two G-protein coupled receptors (GPRs), the thyrotrophin-releasing hormone receptor (TRH-R) and the dopamine D2 receptor. Sequence analysis identified the clone as a TRH-R encoding a putative protein of 398 amino acids. The human pituitary TRH-R had high sequence homology with the rat and mouse TRH-Rs, except at the 3' end of the carboxy (COOH) tail. The Wnt and Notch signal transduction pathways are two important regulators of pituitary organogenesis. Wnt signalling regulates maintenance, proliferation and differentiation of the progenitor pool during early pituitary development, whilst Notch signalling prevents the premature specification of endocrine cell-types. Spring-born ram lambs were used to examine: (1) sexual maturational changes in LH, FSH and prolactin (PRL) secretion, testicular gonadotropin receptors, and testicular size and function; (2) predictability of yearling ram reproductive function from juvenile testicular size and reproductive hormone measurements. Despite continuous increases in testis size, serum LH-profile characteristics became greatest between 2 and 4 months and declined thereafter.
