The objective of this work was to evalue the effect of bio-stimulant Stimulate applicated or not in differents development stages of cotton plant cv Delta Opal, cultivated in a vegetation house and submitted or not to hydric stress. There were applicated 25, 60 and 100% of total volume of pores (VTP) filled out with water and Stimulate (registrated mark) applicated on the seed and on the flower botton stage and without biostimulant application 69 days after sowing. The hydric stress period of time was 15, 30 and 45 days after hydric stress application. The experimental delineation was wholly randomized in a 3x3x3 factorial scheme, with four repetitions of five plants each one. The cotton plant cv Delta Opal did not support hydric eack of 25% VTP on the flower botton beginning, being observed the minor heights, lap diameters, number of flowers, and endangering the formation of reproductive frames. The bio-stimulant dose and application form did not offer improvement on plants development when submitted to eack of water, but on plants submitted to water excess, a lap diameter increase occurred
O objetivo do trabalho foi avaliar o efeito do bioestimulante Stimulate (marca registrada), aplicado ou não em diferentes fases do desenvolvimento do algodoeiro cv. Delta Opal, cultivado em casa de vegetação e submetido ou não ao estresse hídrico. Foram aplicados 25, 60 e 100% do volume total de poros (VTP) preenchidos com água.e o Stimulate® aplicado na semente, na semente e na fase de botão floral e sem a aplicação do bioestimulante, aos 69 dias após a semeadura. O período de duração do estresse hídrico foi de 15, 30 e 45 dias após a aplicação do estresse hídrico. O delineamento experimental empregado foi inteiramente casualizado em esquema fatorial 3 x 3 x 3, com quatro repetições de cinco plantas cada. O algodoeiro cv Delta Opal não tolerou deficiência hídrica de 25% VTP iniciando na fase de botão floral, sendo observadas as menores alturas, diâmetros de colo, número de folhas, e comprometendo também a formação de estruturas reprodutivas. O bioestimulante na dose e na forma aplicada não proporcionou melhoras no desenvolvimento das plantas quando submetidas à falta de água, já em plantas submetidas ao excesso de água, houve aumento do diâmetro do colo das plantas

A previous study indicates OPEL is a novel elicitor, which induce cell death, reactive oxygen species (ROS) accumulation, callose deposition, PAMP-triggered immunity (PTI), and systemic acquired resistance (SAR) on Nicotiana tabacum. In this study, we focus on OPEL homologous genes to verify whether OPEL is a pathogen-associated molecular pattern (PAMP) or not. Phylogenetic analysis OPEL homologous genes by tblastn search on NCBI, JGI and Broad institute genome database has shown homologs of OPEL have gene duplication in oomycetes. Besides, oomycetes plant pathogens have the most copy number of OPEL homologous genes. Analysis domain conformation of OPEL homologs indicates thaumatin-like domain is a distinct domain in oomycetes plant pathogen. Phylogenetic analysis glycoside hydrolase family 16 (GH16) in Phytophthora parasitica demonstrated OPEL homologous genes are different from other GH16 genes. These study imply OPEL homologs are unique genes in oomycetes. To characterize the interaction between OPEL homologs and plants, we overexpress OPEL homologs of P. parasitica by using agroinfiltration. PPTG_17496 induces necrosis on N. benthamiana, and PPTG_16550, PPTG_17497, and PPTG_17498 induce small region cell death on N. tabacum. Both results indicate OPEL homologs are PAMPs. However, PPTG_17496 consume 10 days to establish necrosis symptoms on N. benthamiana, which may be caused by DAMP-triggered immunity (DTI) but PTI. We need further research to find out OPEL homologs induce PTI or DTI on plants.

To combat invasion by pathogens, plants have developed different defense mechanisms, including pattern-triggered immunity (PTI), which is elicited upon the perception of pathogen-associated molecular patterns (PAMPs) or danger/ damage-associated molecular patterns (DAMPs) by plant pattern recognition receptors (PRRs). Previously, a novel elicitor protein named OPEL was identified in the cultural fluid of Phytophthora parasitica, a notorious pathogen of a wide variety of economically important crop species. It is characterized by the presence of thaumatin-like domain, glycine-rich domain and glycoside hydrolase 16 (GH16) domain. Infiltration of OPEL recombinant protein into the leaves of tobacco (Nicotiana tabacum cv. Samsun-NN) resulted in PTI responses such as cell death, callose deposition, reactive oxygen species (ROS) production, and induced expression of salicylic acid-responsive defense genes, and the GH16 domain is indispensable for OPEL to induce PTI. Search of NCBI databases identified in total 25 GH16-containing genes in the genome of P. parasitica. Conserved sequence analysis showed that PPTG_16550, PPTG_17496, PPTG_17497, and PPTG_17498 are highly conserved with OPEL, thus they are very likely close homologs of OPEL. The purpose of this study is to investigate the role of OPEL homologs in plant basal defense response. Analysis by quantitative reverse transcriptase-PCR showed that all four genes showed different expression patterns. Especially, the expression of PPTG-17497 and PPTG_17498 was significantly induced at 12 hours post pathogen inoculation. Of the four putative OPEL homologs, PPTG_16550 is unique in that it lacks the thaumatin-like domain. To know its role in plant defense, PPTG_16550 recombinant protein was expressed in Escherichia coli, which caused cell death, reactive oxygen species (ROS) production, and callose deposition upon infiltration on the leaves of tobacco. Nevertheless, this protein shows no activity to induce systemic acquired resistance against Tobacco mosaic virus (TMV). These results indicate that PPTG-16550 is an elicitor, which elicits PTI response in tobacco. Its role in the process of plant infection by P. parasitica awaits further investigation.
