Tissue-cultured shoots and plantlets usually have leaves with non-functional, open stomata and little epicuticular and cuticular wax, resulting in excess evapotranspiration after transplantation. Various strategies were evaluated to decrease ex vitro acclimatization difficulties for 'Silvan' blackberry, including transplanting unrooted shoots, increasing the medium agar concentration from 6 to 9 or 12 g/l and diluting the basal medium. A novel technique was developed to immobilize plant cells. The cells are deposited on a surface of man-made fibrous material which provides for strong binding of the plant tissue biomass growing in the submerged culture. Two methods of vegetative propagation, stem cuttings under mist and air layering, were examined for the 'Desert Museum' hybrid palo verde and species of Cercidium, Parkinsonia, and Prosopis. Basal cuttings of 'Desert Museum' gave higher rates of rooting than apical cuttings and showed better rooting in April than September. The propagation methods of achene germination, stem cuttings, air layering and micropropagation were investigated to obtain an efficient propagation method. Smoke water dilutions of 1:100, 1:50 and 1:10 significantly increased germination, with the 1:100 smoke water dilutions showing the highest germination percentage (22.4 percent). Morphological and growth observations made on landraces of African yam bean (AYB) used in this study confirm that this species is the most morphologically variable in the genus (Potter, 1992). Morphological characters such as seed colour, stem colour, internode length, leaf size and number of leaves per plant were found to vary between landraces. Growth and development was controlled by both genotype and environment. Flowering was observed only when plants were grown at 25°C, rather than at 30°C, with a 12 h photoperiod.
