Full textIn this thesis I present four chapters, and in all of them I discuss how dwarf shrubs interact with co‐occurring plants under varying regimes of stress. This research involved ecosystems of great environmental, nature conservation and ecological value, yet highly sensitive to environmental changes, in four contrasting cold regions at high altitude or high latitude. Following a latitudinal order, the selected ecosystems were: 1) a temperate alpine treeline in the Central Pyrenees; 2) a primary succession gradient in a boreal ecosystem in Finland; 3) a subarctic alpine treeline in Lapland; and 4) a high‐arctic tundra in north‐eastern Greenland. The first two chapters are based on an experimental approach and focus on how shrubs commonly found near the treeline interact with tree seedlings of treelineforming species in two contrasting environments, i.e. in a subarctic forest‐tundra ecotone in Lapland, northern Sweden, and in a more southern, temperate forestalpine pasture ecotone in the Central Pyrenees. In addition, since trees living near their limit of distribution are very sensitive to environmental changes, especially to increased temperature during the growing season (Körner 2003), we also assessed how distinct environmental change scenarios may affect tree seedling growth and survival across the ecotone. The first chapter is entitled ‘Shrub‐tree interactions and environmental changes drive treeline dynamics in the Subarctic’, where we explain the three‐year‐long experiment performed in the Abisko National Park, in the subarctic Scandes, Northern Sweden; the species studied were Betula pubescens tree seedlings and the shrub Vaccinium myrtillus. In this study we showed the importance of facilitative and competitive shrub‐tree interactions as drivers of subarctic treeline dynamics. Furthermore, we demonstrated that the great sensitivity of tree seedlings to warming had strong implications for treeline dynamics under the predicted warmer scenario at high latitudes, and we identified that complex interactions between shrubs and herbivores are critical to predicting future changes. The second chapter is entitled ‘Similar tree seedling responses to shrubs and to simulated environmental changes at Pyrenean and subarctic treelines’. Here we presented a comparison between the results obtained in the experiment presented in the first chapter and those obtained in a parallel experiment performed during a similar period near the treeline in the Alt Pirineu Natural Park, in the Central Pyrenees, Catalonia. This experiment was based on the same factorial design but with different species (i.e. Pinus uncinata tree seedlings and the shrub Rhododendron ferrugineum). To our knowledge, it is the first study which experimentally tests the responses of plants to distinct environmental scenarios in a high mountain ecosystem in the Pyrenees. In this chapter we presented some mechanisms for understanding the recently observed variability of local responses of both subarctic and alpine treelines to currently changing climate while identifying some commonalities that can be used to generalise large scale response of treelines to climate warming. The third chapter focuses on the effects of a dominant dwarf shrub (i.e. Empetrum nigrum) on Pinus sylvestris tree seedlings along a primary succession within a boreal ecosystem on an uplifting island in Bothnian Bay, Finland. This chapter is called ‘An ericoid shrub plays a dual role in recruiting both pines and their fungal symbionts along primary succession gradients’. Here we showed that facilitative and competitive effects of shrubs markedly determined tree seedling establishment and their fungal colonisation along this succession gradient, but in this chapter we did not relate these findings to any environmental changes. As far as we know, we presented the first finding that an ericoid mycorrhizal shrub may enhance both the performance of the ectomycorrhizal host tree and the tree’s fungal symbionts. The study presented in the fourth chapter was performed along a snow‐depth gradient in an extreme arctic tundra ecosystem in the Northeast Greenland National Park, the largest national park in the world. The chapter is entitled ‘Plant interactions and higharctic vegetation composition along a snow‐depth gradient in NE Greenland’. This ecosystem is probably the most sensitive and fragile among the ecosystems studied in this thesis as the eastern coast of Greenland is expected to experience substantial changes in climate due to marked changes in snow precipitation and temperature regimes (Brown and Mote 2009). Here we assessed plant species richness, establishment and composition patterns in distinct growth forms occurring in common arctic plant communities associated with varying snow‐depth during the winter season. This study will help to predict potential diversity and vegetation changes in the high Arctic if snow precipitation regime changes in the future as anticipated.

Para tanto, as plantas foram cultivadas durante seu desenvolvimento em solução nutritiva no 2 de Hoagland & Arnon contendo 160 mg L-1 de cálcio e modificada para fornecimento de 200, 120, 80 e 40 mg L-1. Although calcium has been extensively studied, the metabolism of this macronutrient needs further evaluation since this element acts as a secondary messenger in signal transduction pathways in plant cells and, due to variations in its cell level, acts by means of modulator proteins and their target molecules, regulating several cell processes, from ion transportation control to gene expression. Thus, there are doubts about the effect of calcium on plant development, gas exchanges and essential oil production route, especially concerning yield and chemical composition. This study aimed to expand the knowledge of calcium metabolism aspects in Mentha piperita, an aromatic medicinal species, grown under nutrient solution. The influence of calcium level variation on physiological indexes, gas exchanges and essential oil yield and chemical composition was assessed. The plants were grown during their development in Hoagland & Arnon nutrient solution number 2 containing 160 mg L-1 and modified to supply 200, 120, 80 and 40 mg L-1. At 45, 65, 85, 105 and 140 days after transplant (DAT) of seedlings to the culture solution, the assessed physiological indexes were leaf area, leaf blade and total dry matter, and essential oil yield and chemical composition. To analyze gas exchange variables, an open portable photosynthesis system (LI-6400, Li-Cor Inc., NE, USA) was used to determine, at 65, 85 and 105 DAT, CO2 assimilation, transpiration, stomatal conductance, intercellular CO2 concentration and water use efficiency. Full textPara la presente investigación de los Paisajes Culturales en la Región, se decidió trabajar sobre la actividad ganadera, ya que es una actividad que dentro de una geografía y clima peculiar dio como respuesta a una interesante cultura (la cultura del ovejero), que más allá de una forma de vida, tanto patrimonio intangible, destacó por ser un soporte tangible de expresión a diferentes escalas, tales como: la forma de habitar el territorio, la arquitectura, las materialidades, etc. Además cabe destacar la importancia económica que esta actividad significa para la región, ya que es una de las principales actividades que le dan sustento, y condicionan otras actividades complementarias.

Full textThe main purpose of this work was to study an alternative way to control the Passionfruit woodiness virus - PWV through the production of transgenic plants which contained the Passionfruit woodness virus coat protein gene. The binary vector was built by using pCambia 2300 and pCambia 2301 plasmids, which contain the selection gene nptII. The pCambia 2301 plasmid also contains the reporter gene uidA (GUS). The plasmids were introduced into Agrobacterium tumefaciens, EHA 105 and LBA 4404 strains, via thermal shock method. The explants for the genetic transformation were young leaf disks (6 mm of diameter) of IAC 275 and IAC 277 varietys, extracted from plants kept under 16 h photoperiod, at 27 °C. The explants were inoculated with a bacterial suspension (5x108 UFC/mL) for 20 min and then transferred to Petri dishes containing cocolture medium MS + thidiazuron (TDZ - 0,25 mg/L) + silver nitrate (AgNO3 - 4 mg/L) + acetosyringone (1 µM/L). The co-culture was performed at 24 °C t, in the dark, for a three-day period. For the selection and regeneration of plants, the explants were transferred to the selection culture medium MS + TDZ (0,25 mg/L) + AgNO3 (4 mg/L) + kanamycin (100 mg/L) + cefotaxime (500 mg/L). The incubation was performed at 27 °C, in dark, for 4 - 6 weeks. The adventitious buds developed were then transferred to the culture medium MSM + 10% coconut water and kept incubated under 16 h photoperiod. The genetic transformation was identified through GUS and PCR tests. There were 22 PCR positive plants. Out of those, 8 were Southern blot analyzed for the confirmation of transgenc integration. The transgene transcription and expression were determined by Northern and Western blot respectively. The transgenic plants were then multiplied and inoculated with 3 different strains of PWV, and the line 2 showed resistance to the three strains used.

Resumo: Foram estudadas as características fisico-químicas e teores de macro e micronutrientes das espécies das plantas medicinais Arctium lappa minor, Plantago major e Porophyllum ruderale. Através de aplicação de extratos aquosos das referidas plantas, efetuou-se testes de cicatrização em lesões nos dorsos de ratos Wistar, na dosagem de 1 gota ao dia, por quatorze dias. Usou-se os mesmos animais para controle com aplicação de água destilada em lesões-controle. Efetuou-se o sacrifício dos animais aos 3º, 7º, 11º e 14º dias e retirou-se os recidos tratados com os extratos e os tratados-controle para realizar-se os cortes histológicos, os quais foram histometrados em lentes Zeiss Kpl-W-10X, com 25 hits. Contou-se os fibroblastos, fibras colágenas e vasos sanguíneos para averiguação dos efeitos de cicatrização, confirmando que de fato as plantas em estudos atuam como cicatrizantes. Os dados levantados então, foram analisados estatisticamente, obtendo-se os resultados da eficácia do tratamento com os extratos comparados aos tratados-controle. Com respeito a atividade antimicrobiana, utilizamos os extratos hidroalcoólicos por maceração a frio, e constatamos a eficácia dos mesmos em relação a Staphylococcus aureus, Streptococcus pyogenes tipo A e Streptococcus pyogenes tipo B, cedidos pelo Laboratório Prev Lab de Piracicaba. Tais testes foram efetuados de acordo com o método de difusão em discos (Método Kirbi-Bauer) pela técnica da diluição em tubos. Full textLepidium peruvianum Chacón, is a Cruciferae native from the Andes. It grows between 3,500 and 4500 m. Original from the Bombón plateau, located at the Peruvian localities Junín and Pasco. It became in a crop with a high economical value, due its medicinal and nutritional properties. Actually, it is extended to other regions of the country. The main objective of this research is to study the tissue culture ability of the crop to use in vitro tissues as a tool for secondary metabolite production. Leaves, petioles, roots and hypocotils of L. peruvianum were tested as explants to induce calli. Different concentrations of 2,4-D and Kinetin, in MS basic medium were tested. Calli were induced in most of the media tested, the most efficient hormone ratio auxin/citokinin was 1. It was evaluated the presence of glucosinolates and alkaloids in the callus induced and compared to maca hypocotils as control sample. Two glucosinolates fractions were obtained from calli analyzed. It was found one or two fractions according to the callus and in most of the cases the concentration was higher in callus than in control. In the other hand, it was observed a high variability in the alkaloid fractions and other unidentified metabolites extracted from the calli evaluated in this work. It was also evaluated the presence of myrosinases in the calli studied, and it was found positive bands either in callus as in maca hypocotils.

Full textEl servicio de secado ofrece una interesante oportunidad de negocio, por la relevancia que tiene en la cadena de valor de la madera dimensionada que se exporta a mercados norteamericano, europeo y nipón preferencialmente. La provincia de Arauco en la Octava Región, acusa una fracción del mercado del secado que no está atendido. Para satisfacerla, la locación de Curanilahue ofrece una ubicación estratégica por la cercanía con los principales demandantes, la proximidad a los puertos de embarque para exportación, la infraestructura industrial implementada e incentivos a la inversión para los que allí se instalen. El servicio, de características genéricas, se realizará mediante tecnología de punta con personal de amplia experiencia e incluirá el despacho a punto de embarque dentro de la octava región. El horizonte de evaluación de proyecto es de 5 años, equivalente al 60% de vida útil de los principales activos. Estos generarán un servicio de secado de 26.400 m3/año a través de ciclos convencionales de 96 horas, con ingresos anuales de US$ 660.000 y costo operacional de US$ 174.760. La generación de ingresos requiere de una inversión de US$ 470.592 más impuestos que contemplan la inversión en activos dentro de los cuales destacan por el monto, la caldera y las cámaras de secado. En base a estos antecedentes, el estudio de costos operacionales y factibilidad técnica del proyecto, se determinaron flujos de los estados financieros, con el fin de evaluar la viabilidad del proyecto. La evaluación arroja un VAN de US$ 334.881, descontados a una tasa de un 18.18% y una TIR de un 52%, corroborando el atractivo de invertir en este negocio. Esta inversión se financiará a través de privados, pagadera en tres cuotas anuales iguales y sucesivas a contar del primer año, con garantía prendaría sobre los activos. La oferta a estos inversionistas es el 72% de las utilidades como rentabilidad, la alternativa de capitalización de las mismas transformándola en participación de la empresa y derecho a la administración si su inversión supera del 50%.
