Abstract:
Malaria remains one of the most serious tropical infectious diseases affecting mankind. The prevention of the disease is hampered by the increasing resistance of the parasite to existing chemotherapies. The need for novel therapeutic targets and drugs is therefore of the utmost importance and detailed knowledge of the biochemistry of the parasite is imperative. This study was directed at the biochemical characterisation of the polyamine metabolic pathway of P. falciparum in order to elucidate differences between the parasite and its human host that can be exploited in the design of novel antim

The objective of this work has been to investigate the frequency of cross-fertilisation between gametes of genetically distinct clones of the human malaria parasite Plasmodium falciparum. Previous genetic experiments involving both rodent malaria parasites in vivo and human malaria parasites in vitro have demonstrated higher than expected numbers of recombinants among the progeny of crosses. It has been suggested that this could be due to a favouring of cross-fertilisation over self-fertilisation in the mosquito phase of the life-cycle. The work has involved examining the

The main thrust of this thesis was to characterize P.falciparum genetic diversity in northern Ghana. To do this, I used simple techniques to purify P. falciparum DNA from clinical samples across a rural setting for whole-genome sequencing. The goal was to provide a framework for exploring host-parasite genetic interactions. Utilizing Illumina deep sequencing data for 277 isolates I analyzed P. falciparum genetic diversity and described within-host diversity across this area. I observed random mating (ie no population structure) in the local parasite population, and a high genetic diversity ind

A new era of malaria eradication programs relies on increased knowledge of the parasite through sequencing of the Plasmodium genome. The way malaria parasites are transmitted in space will have an influence on their genetic relationships. It can be expected that parasites collected within close geographic distances of each other would be more closely related than those across large geographic distances. Further to this, because malaria transmission is focal and heterogeneous in space, then the genetic relatedness between malaria parasites in these foci of malaria transmission would be greater within tightly clustered regions. Thus, using the level of genetic relatedness of these parasites would reveal how they are transmitt

Plasmodium falciparum is a human intracellular parasite that is the causative agent of a deadly form of malaria. This species alone is responsible for 200 million cases of malaria annually resulting in over 1 million deaths worldwide. The excessive mortality due to P. falciparum infection is due to its ability to cause severe pathogenesis through hyperparasitemia and cytoadherence defined as the ability of infected red blood cells to adhere to host vasculature. Cytoadherence is mediated through the export of parasite proteins to the surface of the infected red blood cell (RBC). Exported protei

The malaria parasite, Plasmodium falciparum, has a demonstrated history of adaptation to antimalarials and host immune pressure. This ability unraveled global eradication programs fifty years ago and seriously threatens renewed efforts today. Despite the magnitude of the global health problem, little is known about the genetic mechanisms by which the parasite evades control efforts. Population genomic methods provide a new way to identify the mutations and genes responsible for drug resistance and other clinically important traits.
