The research contained within this dissertation aims to shed light onto the basic biology of one of Australia’s unique native wildlife species, the platypus. In particular, it aims to broaden our knowledge of mammalian venom, by bioinformatic identification of genes encoding known platypus venom components, phylogenetic analyses of platypus and other animal venom toxins, and investigation of tissue expression patterns of platypus venom and related non venom genes. Finally, this research culminates in the use of a platypus venom transcriptome to identify previously unknown platypus venom toxins

This study characterised the pathological and immunopathological features of selected infectious diseases in the koala and the platypus. Originally, lymphosarcoma, cryptococcosis and chlamydiosis in the koala were chosen. Lymphosarcoma was included because of the putative retroviral involvement. Another infectious disease of Australian wildlife, mucormycosis of the platypus, was also included in the study. Serine proteases are a family of enzymes with a wide array of functions across both eukaryotes and prokaryotes. Here we have attempted to produce the serine proteases rat mast cell protease 2 and mouse mast cell protease 5 in a culture of HEK 293 cells; and mouse mast cell protease 4, platypus granzyme B-like protease and platypus hypothetical protease in a baculovirus expression system. Following production we wanted to analyse these serine proteases using a phage display assay and a battery of chromogenic substrates.

Platypuses ( Ornithorhynchus anatinus) in the north of the island state of Tasmania suffer from a serious disease called ulcerative mycosis, which is responsible for high morbidity and, presumably, mortality rates in areas where the disease occurs. The disease is caused by the fungus Mucor amphibiorum, which is also found in Queensland and New South. However, it does not cause disease in platypuses in those states. Information on the life cycle and associations of the mountain pinhole borer, Platypus subgranosus Schedl (Coleoptera : Curculionidae : Platypodinae) is reviewed. Existing unpublished data combined with data collected in this study are used to provide quantitative descriptions of aspects of P.subgranosus biology.
