Old age comes coupled with frailty and disease and, thus, the ageing of the World’s population have spurred the interest on the causes and mechanisms of senescence. Senescence has long been a mystery, with no single universally accepted theory accounting for its ultimate evolutionary causes (if indeed these causes exist). Perhaps two of the most popular evolutionary explanations proposed so far are the Mutation Accumulation Theory of Senescence proposed by Peter Medawar in 1951 and the Antagonistic Pleiotropy Theory of Senescence, suggested by George C. Genome-wide association studies (GWAS) have become one of the most common types of genetic studies, due to their success at finding markers associated with diseases and other traits of interest. With thousands of these studies performed in the last decade, a variety of methods have been developed to meta-analyze their results to search for variants that affect multiple traits. This thesis introduces one such method, based on approximate Bayes factors (ABFs), which relies only on effect size estimates and standard errors from GWAS. This thesis discusses the pleiotropy theory of ageing in evolutionary biology in some of its most important aspects. In short, the theory says that natural selection fosters the early-life survival and reproductive performances of an organism even if this would necessarily impose a cost on later-life survivorship and reproduction thereby leading to the evolution of that peculiar pattern of age-correlated decline in biological functioning that characterizes ageing. The theory predicts that the level of external/environmental mortality should positively correlate with the rate of ageing in a pop

Abstract:
This thesis focuses on the role of the forkhead box factor Foxg1 in the development of the vertebrate telencephalon. Mice mutant for Foxg1 exhibit multiple telencephalic defects demonstrative of an important pleiotropic role of rFoxg1 in telencephalic organogenesis. Severe hypoplasia of the null mutant telencephalon demonstrates that Foxg1 is required for the regulation of telencephalic growth. Experiments descried in this thesis provide the most systematic study to date of the mechanisms by which Foxg1 regulates this process. Broadly, the results demonstrate that Foxg1 is required to m

Abstract:
Matrin3 (MATR3) is an RNA binding protein involved in many roles in the nucleus, such as chromatin architecture and gene expression regulation, modulating transcriptional and post-transcriptional processes as RNA splicing and mRNA stabilization. Nevertheless, some functions of MATR3 within the cells are not entirely clear. MATR3 has been associated with Amyotrophic Lateral Sclerosis (ALS), a neurodegenerative disease that damages motor neuron (MN) cells and leads to progressive muscle paralysis and respiratory failure. A better understanding of MATR3 activity within cell physiology could repre

Full text
Abstract:
In this thesis, the pleiotropic effects of statins on the disruption of prenylation and membrane rafts were investigated in immune effector cells. T cells and NK cells were extracted from various groups of patients treated with statins and proliferation and cytotoxicity respectively, measured ex vivo. In a study of patients with cardiovascular disease, a 23% reduction in T cell proliferation and 43% reduction in NK cytotoxicity were observed. In a normal volunteer study where healthy subjects received simvastatin (40mg per day) for 4 weeks, a comparable reduction in T cell proliferation was no

Abstract:
Vitamin D insufficiency is seasonally endemic in populations north of the 40th parallel, and epidemiological data show an association with colorectal cancer risk and prognosis. Molecular mechanisms that underpin the relationship are not well established. Vitamin D status is shown to be associated with age-related silencing of tumour suppressors in colonic stem cells via aberrant DNA methylation, suggesting that insufficiency contributes to transformation. In this text, vitamin D’s ability to promote bowel health via modification of DNA methylation patterns has been investigated. Chronic inflam

Abstract:
Serratia sp. ATCC 39006 (S39006) is known for producing carbapenem and prodiginine antibiotics; 1-carbapen-2-em-3-carboxylic acid (car) and prodigiosin. It displays different motility mechanisms, such as swimming and swarming aided by flagellar rotation and biosurfactant production. In addition, S39006 produces gas vesicles to float in aqueous environments and enable colonization of air-liquid interfaces. Gas vesicles are thought to be constructed solely from proteins expressed from a gene cluster composed of two contiguous operons, gvpA1-gvpY and gvrA-gvrC. Prior to this study, three cognate
