Full textA major issue in aquaculture is improving knowledge about sexual differentiation and oogenesis in fish. Their regulation particularly involves the synthesis and action of sexual steroids, especially estrogens. Shbg is a sex hormone-binding globulin whose major functions may be protection and transport of steroids and modulation of steroids signaling. Two paralogs have been identified in rainbow trout, Oncorhynchus mykiss, Shbga, majorly expressed by the liver and secreted into the blood, and Shbgb, expressed in the ovary by granulosa cells. Our results confirm a higher binding specificity of recombinant Shbgb for estradiol and testosterone. Shbgb expression in granulosa cells and its link with aromatase at early stages of gonadogenesis have been demonstrated. Shbgb has been shown to be implicated in testosterone protection against its aromatization and that could be a way to maintain high testosterone levels in female salmonids. In order to study Shbgb functions and regulations, primary cultures of granulosa cells were developed. Shbgb expression regulation was shown to evolve during oogenesis and the role of sexual steroids would be worth to be further studied. . Finally, several transfections methods for these cells, maintained at 12°C, were tested. Electroporation was clearly more efficient than chemical transfection. Nonetheless, electroporation can be deleterious for the cells steroidogenic properties. These last findings provide information for choosing wisely transfection methods to be used on fish cells primary culture and show that the accurate analysis of the effects of these methods by themselves is crucial for a relevant data interpretation

Full textRainbow trout is an economically important species and genomic studies have explored gene regulation in key processes such as growth, adaptation or reproduction. Nevertheless, the regulation of gene expression by non-coding RNAs is far from being understood in this species. The present study characterizes the miRNA repertoire of rainbow trout with the identification of 164 miRNAs belonging to 84 families. The tissue distribution of those miRNAs in 16 different tissues or organs was also characterized and we confirmed that miRNAs are expressed in a tissue-dependent manner. Then, we showed that miRNA duplicates are highly conserved after whole genome duplication, twice more than proteins. Moreover, we characterized miRNA expression profiles during oogenesis and sex differentiation. During oogenesis, we identified twelve miRNA candidates as potential regulators and study the function of one of them: miR-202-5p. MiR-202-5p expression is restricted to the gonad only and localized to granulosa cells in female rainbow trout. By inhibiting and overexpressing miR-202-5p, we show that it targets pfn2l and pgam1a in granulosa cells. This work not only provides a repertoire of rainbow trout miRNAs but also new insights into regulation of gene expression in oogenesis and sex differentiation

Full textNew Guinea Island is an immense, rugged landscape and remains one of the last unexplored places. This is particularly true for its ichthyological fauna. With freshwater Gudgeon and Gobiids, the Austro-New Guinean rainbowfishes are the most diverse groups with important endemism. Rainbowfish taxonomy is ancient and recently attracted much attention for various domains interest. Despite a large number of species already described, their taxonomy remains incomplete and somewhat confused. Data on their phylogenetic relatedness or biogeographical history are scarce. The results presented here give a contribution to diversification processes of Rainbowfishes from West Papua and adjacent biogeographic region and provide a complete taxonomic revision of the Western New Guinean species. A total of 716 specimens belonging to 3 genera have been studied in morphology and molecular relatedness. Molecular phylogenies were inferred from 3 mitochondrial regions (Cox-1, Cyt-b, D-loop) and 1 nuclear intron (S7). The dataset includes more than 5,000 base pairs with a portion of DNA Barcodes. The choice of both mitochondrial and nuclear molecular markers was driven by the multiple objectives to resolve deeper and shallow radiations and to evidence possible hybridization events. The phylogenies revealed an unexpected cryptic diversity within the genus Melanotaenia and strongly supported the polyphyly of the genera Glossolepis and Chilatherina. The results also revealed a strong congruence between phylogenetic relatedness, clade composition and geography and confirmed that the center of origin of Melanotaenia sensu lato was dated to the Mid-Miocene in the Bird's Head Peninsula. The eological context of the area revealed that the formation of the Lengguru fold-and-thrust Belt together with the ridge of the Central Dividing Range were the main events responsible of the basal diversification processes within this group of obligate freshwater fishes

Full textFish eggs, especially those of the rainbow trout (Oncorhynchus mykiss) in the present study, are an interesting nutritional aquatic source. They contain proteins of high value, as well as an oil rich in polyunsaturated fatty acids (PUFA) with a large percentage of phospholipids. However, they exhibit a high auto-protection capacity against environmental constraints and thus, the degree of hydrolysis of rainbow trout eggs by Alcalase®, Neutrase® and Protamex® proteases varied solely within 3-7 %. This value was low compared with the 20 % obtained in most animal proteins. The phospholipid content was high (53 % of total lipids) and PUFA accounted for 42 % of total fatty acids. Among PUFA, DHA was found preferably at the sn-2 position of the glycerol backbone, which is of special interest about nutritional properties. The oil release by enzymatic hydrolysis was found limited compared with chemical methods, probably because of the strong interactions engaged with the incomplete destructured protein network. The oxidative stability of the oil was studied through several methods in which the infrared Fourier transform appeared as the best tool for structural analysis along the oxidation process. As a conclusion, lipids from fish eggs, especially from rainbow trout, could be a nutritional breakthrough, as far the enzymatic hydrolysis of the vitellus and of the chorion proteins is achieved

Full textThe aim of the thesis was to characterise the differences in digestive and metabolic utilisation of dietary carbohydrates between two lines of rainbow trout divergently selected for muscle fat content (Fat-F and Lean-L), when fed diets with (20%) or without gelatinised starch. In both lines, starch intake did not adversely affect growth, resulted in a moderate postprandial hyperglycemia, enhanced protein/lipid retention and a distinct intracellular signalling pattern in the liver involving the energy sensor AMPK and nutrient sensor TOR-S6. No difference between the two lines was observed for the apparent digestibility of starch, the mRNA levels of genes encoding glucose transporters in the intestine, regulation of postprandial glycemia and utilisation of glucose in the peripheral tissues (muscle and adipose tissue). However when compared to the L line, the F line was characterised by lower growth and feed efficiency; better capacity to store excess glucose as indicated by higher glycogen levels and mRNA levels of lipogenic markers in the liver (with no impact on glucose homeostasis); and a specific lipid metabolism (a greater potential to synthesise chylomicrons and to bioconvert fatty acids, coupled with a lower potential to oxidise fatty acids). In a separate study, replacement of fish oil in the diet with a blend of vegetable oils was found to suppress most of the molecular differences previously observed between the two genotypes. Starch intake under the vegetable oil diet regime led to different metabolic changes in the intestine and liver, for example, elicited a higher transcriptional response of key desaturase and elongase enzymes in both lines. Overall, our data demonstrated the existence of significant metabolic differences between the two trout lines, but it did not lead to better utilisation of dietary carbohydrates. In addition, our work highlights the importance of diet composition in the growth and metabolic response of different genotypes of fish

Full textFlavobacterium psychrophilum is a Gram negative bacteria responsible for fish infection. We used a proteomic approach to identify some outer membrane components such as putative adhesins, proteins involved in iron acquisition or in efflux systems, a HtrA homologue and some other molecules with unknown function. Several major antigens have been identified in the outer membrane including the two components OmpH/P18 and OmpA/P60. They are surface-exposed since they were completely digested by in situ proteinase K treatment and the two monospecific sera were bacteriostatic/bactericidal. Vaccination trials showed that both proteins can induce a high titter of specific antibodies which are protective. Collectively, these results indicate that these two proteins could be used in future vaccine development as promising candidate antigens
